Abstract: Endocrine cell distribution within the islets of Langerhans may vary both between species and under different energetically demanding conditions such as cold acclimation. The naked mole-rat, Heterocephalus glaber, lacking an effective insulatory pelage, is effectively a poikilotherm, yet it shows a typical mammalian cold-acclimation response by substantially increasing food intake to meet higher energy requirements when housed at lower temperatures. The endocrine component of the pancreas of thermoneutral and cold-acclimated naked mole-rats was thus characterized using immunocytochemistry and ultrastructural analyses. Four distinct endocrine cells were identified: α (glucagon-producing), β (insulin-producing), δ (somatostatin-producing), and PP (pancreatic polypeptide-producing) cells. Distribution of these cells differed from that of other rodents, in that β cells formed the mantle while α cells formed the core of the islets. This distribution may contribute to the observed insulin insensitivity of this species, as indicated in abnormal responses to glucose tolerance tests. Insulin-producing cells, however, were more numerous than glucagon-producing cells. This ratio was unchanged with cold acclimation. Immunoreactivity of α and β cells was more intense in cold-acclimated than in thermoneutral animals, possibly indicative of a change in hormonal production in animals housed at a lower temperature. We studied the retinal projections, the distribution of cytochrome oxidase activity and the cyto- and myeloarchitecture of the subcortical visual system in the subterranean Ansell's mole-rat Cryptomys anselli. The optic nerve contained 1500 myelinated and a similar number of unmyelinated fibres. The retina projected to all the visual structures described in surface-dwelling sighted rodents. The suprachiasmatic nucleus was large and received bilateral retinal input. All other visual nuclei were reduced in size, were cytoarchitecturally poorly developed and received almost exclusively contralateral retinal projections. The dorsal and ventral lateral geniculate nuclei were moderately reduced and heavily innervated. The intergeniculate leaflet could be identified between these two nuclei. Pretectal nuclei were also relatively well-developed. The nucleus of the optic tract, the olivary pretectal nucleus, and the anterior and posterior pretectal nuclei were innervated by the retina. By contrast, the superficial, retinorecipient layers of the superior colliculus showed extreme reduction. The strata zonale, griseum superficiale and opticum were collapsed to a single layer 40 µm thick. The accessory optic system was vestigial. These findings indicate that the functional subsystems involved in photoperiod perception, form and brightness discrimination, and movement analysis are anatomically rather well developed, whereas those involved in coordination of visuomotor reflexes are severely reduced. Thus, the visual system of C. anselli is much better developed than that of the blind mole-rat Spalax ehrenbergi. We suggest that Cryptomys anselli has retained basic visual capabilities. Colonies of co-operatively breeding African mole-rats have traditionally been thought to be composed of a single breeding female, one or two breeding males, and their offspring. In the naked mole-rat ( Heterocephalus glaber), the occurrence of facultative inbreeding means incest avoidance cannot prevent reproduction in subordinate group members, and physiological suppression of reproductive function by the breeding female occurs in both sexes. In contrast, previous studies of captive colonies of the Damaraland mole-rat ( Cryptomys damarensis) suggest that breeding within a colony is restricted to a single breeding pair, simply because all other colony members are highly related (first- or second-order relatives) and this species is an obligate outbreeder. Using microsatellite markers, we investigated parentage and colony composition in 18 wild Damaraland mole-rat colonies to determine whether inbreeding avoidance alone can explain the high levels of reproductive skew in this species. Multiple and unidentified paternity was widespread within colonies and immigrants of both sexes were regularly identified. Unrelated, opposite-sex nonbreeders were found coexisting in two colonies. These results suggest that, in the wild, conditions exist where nonreproductive females can come into contact with unrelated males, even when they do not disperse from their natal colony. Inbreeding avoidance alone is therefore insufficient to maintain the high levels of reproductive skew identified in this species suggesting that the breeding female somehow suppresses the reproductive function in nonbreeding females. Cryptomys represents the most speciose and widely distributed genus of the Bathyergidae (Mammalia; Rodentia), a family of mole-rats endemic to sub-Saharan Africa. Throughout its range in southern, central, and western Africa, Cryptomys displays diversity in terms of morphology, patterns of behavior, and chromosome number, thus complicating the systematics of the group. A molecular phylogeny was obtained by separate and combined analyses of the mitochondrial 12S rRNA and intron I of the nuclear transthyretin gene for chromosomally and geographically diverse populations of Cryptomys. Our results show that Cryptomys sensu lato is comprised of two distinct and divergent monophyletic clades: hottentotus and mechowi. Based on our analyses, we propose the elevation of the mechowi clade to the genus Coetomys, retaining Cryptomys as the generic epithet of the South African hottentotus clade. This is supported by: (1) reciprocal monophyly of the two lineages based on independent datasets, (2) the level of sequence divergence observed between these lineages relative to other genera (i.e., Bathyergus and Georychus), and (3) different patterns of chromosomal evolution, with Cryptomys sensu stricto being karyotypically conserved (2N=54) and Coetomys exhibiting high karyotypic diversity (2N=40–78). We have determined the presence of spectral cone types, and the population densities of cones and rods, in subterranean mole-rats of the rodent family Bathyergidae, for which light and vision seems of little importance. Most mammals have two spectral cone types, a majority of middle- to long-wave-sensitive (L-) cones, and a minority of short-wave-sensitive (S-)cones. We were interested to see whether the subterranean bathyergids show the same pattern. In three species, Ansell's mole-rat Cryptomys anselli, the giant mole-rat Cryptomys mechowi and the naked mole-rat Heterocephalus glaber, spectral cone types and rods were assessed immunocytochemically with opsin-specific antibodies. All three species had rod-dominated retinae but possessed significant cone populations. A quantitative assessment in C. anselli and C. mechowi revealed surprisingly low photoreceptor densities of 100 000–150 000/mm2, and high cone proportions, ≈ 10% (8000–15 000/mm2). In all three species, the vast majority of the cones were strongly S-opsin-immunoreactive; L-opsin immunoreactivity was much fainter. In C. anselli, ≈ 20% of the cones showed exclusive S-opsin label, ≈ 10% exclusive L-opsin label and ≈ 70% strong S-opsin and faint L-opsin double label (potential dual-pigment cones). This is the first observation in any mammal of an S-opsin dominance and low levels of L-opsin across the entire retina. It contrasts starkly with the situation in the muroid blind mole-rat Spalax ehrenbergi, which has been reported to possess L-opsin but no S-opsin. African mole-rats are subterranean Hystricomorph rodents, distributed widely throughout sub-Saharan Africa, and displaying a range of social and reproductive strategies from solitary dwelling to the ‘insect-like’ sociality of the naked mole-rat, Heterocephalus glaber. Both molecular systematic studies of Rodentia and the fossil record of bathyergids indicate an ancient origin for the family. This study uses an extensive molecular phylogeny and mitochondrial cytochrome b and 12s rRNA molecular clocks to examine in detail the divergence times, and patterns of speciation of the five extant genera in the context of rift valley formation in Africa. Based on a value of 40–48 million years ago (Myr) for the basal divergence of the family ( Heterocephalus), we estimate divergence times of 32–40 Myr for Heliophobius, 20–26 Myr for Georychus/ Bathyergus and 12–17 Myr for Cryptomys, the most speciose genus. While early divergences may have been independent of rifting, patterns of distribution of later lineages may have been influenced directly by physical barriers imposed by the formation of the Kenya and Western Rift, and indirectly by accompanying climatic and vegetative changes. Rates of chromosomal evolution and speciation appear to vary markedly within the family. In particular, the genus Cryptomys appears to have undergone an extensive radiation and shows the widest geographical distribution. Of the two distinct clades within this genus, one exhibits considerable karyotypic variation while the other does not, despite comparatively high levels of sequence divergence between some taxa. Mole-rats are strictly subterranean and hardly, if ever, come into contact with external light. As a result, their classical visual system is severely regressed and the circadian system proportionally expanded. The family Bathyergidae presents a unique opportunity to study the circadian system in the absence of the classical visual system in a range of species. Daily patterns of activity were studied in the laboratory under constant temperature but variable lighting regimes in individually housed animals from 3 species of mole-rat exhibiting markedly different degrees of sociality. All 3 species possessed individuals that exhibited endogenous circadian rhythms under constant darkness that entrained to a light-dark cycle. In the solitary species, Georychus capensis, 9 animals exhibited greater activity during the dark phase of the light cycle, while 2 individuals expressed more activity in the light phase of the light cycle. In the social, Cryptomys hottentotus pretoriae, 5 animals displayed the majority of their activity during the dark phase of the light cycle and the remaining 2 exhibited more activity during the light phase of the light cycle. Finally in the eusocial Cryptomys damarensis, 6 animals displayed more activity during the light phase of the light cycle, and the other 2 animals displayed more activity during the dark phase of the light cycle. Since all three mole-rat species are able to entrain their locomotor activity to an external light source, light must reach the SCN, suggesting a functional circadian clock. In comparison to the solitary species, the 2 social species display a markedly poorer response to light in all aspects. Group living is an important component of the social biology of numerous rodent species. For the past 2 decades, efforts to understand the ecological factors associated with group living in rodents have focused on subterranean taxa, in particular African mole-rats of the family Bathyergidae. Comparative analyses of the habitats occupied by solitary and social bathyergids suggest that group living occurs when the combined effects of sporadic rainfall, hard soils, and patchily distributed food resources render energetic costs of burrow excavation prohibitive for lone individuals. To determine whether these variables were associated with group living in other subterranean taxa, we characterized ecological differences between a solitary and a social member of a phylogenetically independent lineage of subterranean rodents, the Ctenomyidae. Specifically, we quantified soil conditions and spatial distributions of food resources for 1 population each of the solitary Patagonian tuco-tuco (Ctenomys haigi) and the group-living colonial tuco-tuco (C. sociabilis), both of which occur in the Limay Valley of southwestern Argentina. Our analyses revealed that while members of both species occurred in relatively mesic mallin habitats, only C. haigi also occurred in arid steppe habitat. No differences in spatial distributions of food resources were found between study sites. Significant differences in soil conditions, however, were detected; soils from steppe habitat were significantly harder to penetrate than soils from mallin areas, suggesting that costs of burrow excavation may be higher for the solitary C. haigi. Thus, data from ctenomyids in the Limay Valley are not consistent with explanations for group living developed for bathyergid mole-rats. Although ecological comparisons of ctenomyids over larger spatial scales are required, our data suggest that interactions between ecology and group living in subterranean rodents may be more diverse than previously realized. African naked mole-rats are subterranean rodents that have a robust orienting response to stimulation of unique vibrissa-like body hairs that are widely spaced over an otherwise hairless skin. To determine whether these large body hairs have a specialized organization similar to facial vibrissae, the structure and innervation of facial vibrissa follicles, body hair follicles, and intervening skin in naked mole-rats was compared with that in rats and a furred African mole-rat species (the common mole-rat). Immunofluorescence and lectin-binding analyses revealed that the body hair follicles in naked mole-rats were exceptionally large and well innervated, similar to guard hairs of furred species. However, these body vibrissae lacked the anatomic specializations and unique types of innervation affiliated with follicle sinus complexes of facial vibrissae. In contrast to the furred species, naked mole-rats had a paucity of Aβ-fiber Merkel endings at all peripheral locations. Naked mole-rats also were completely lacking in cutaneous C-fibers immunoreactive for substance P and calcitonin gene-related peptide. In contrast, the hairless skin of the naked mole-rats had an exceptional abundance of presumptive Aδ-fibers. The unusual features of the cutaneous innervation in naked mole-rats are presumably adaptations to their subterranean environment and that they are the only known poikilothermic mammal. The African mole-rats, Bathyergidae are a model taxon on which to test evolutionary theories of the origin and maintenance of mammalian social behavior, since species within the family range in social complexity from solitary to eusocial. The aridity food-distribution hypothesis suggests that sociality within the Bathyergidae has evolved as a consequence of habitat aridity, and the subsequent pattern of geophyte abundance and distribution. Twenty-five burrows of seven species are examined within the Bathyergidae and a simple box-counting method is used to determine their fractal dimension, which is proposed as a measure of burrow architecture that allows explicit testing of differences in African mole-rat burrows within the theoretical framework of the aridity food-distribution hypothesis. This habitat classification may be oversimplified, but it reflects the difficulty of obtaining accurate rainfall data for specific burrows, since rainfall may vary considerably even over short distances, and the two classes of site—arid and mesic—are likely to represent real differences in rainfall pattern.

We report 11 microsatellite loci for the African mole-rat genus Cryptomys (Rodentia: Bathyergidae), isolated from the Damaraland mole-rat ( Cryptomys damarensis) and the Common mole-rat ( C. hottentotus hottentotus). All loci are highly polymorphic in the source species, with between six and 13 alleles and observed heterozygosity ranging from 0.54 to 0.86. Two C. damarensis loci (DMR1 and 6) may be located on the X-chromosome. Cross-species amplification was investigated in 10 Bathyergid species, representative of the five genera. The majority of loci amplified polymorphic product in all Cryptomys species tested. Amplification was also widespread in the other genera, except in Heterocephalus. [ABSTRACT FROM AUTHOR]

Eusociality in mammals is defined in the present paper by the following criteria: reproductive altruism (which involves reproductive division of labor and cooperative alloparental brood care), overlap of adult generations, and permanent (lifelong) philopatry. We argue that additional criteria such as the existence of castes, colony size, reproductive skew, and social cohesion are not pertinent to the definition of eusociality in mammals. According to our definition of mammalian eusociality, several rodent species of the African family Bathyergidae can be considered eusocial, including the naked mole-rat ( Heterocephalus glaber), Damaraland mole-rat ( Cryptomys damarensis), and several additional, if not all, species in the genus Cryptomys. Furthermore, some species of social voles (like Microtus ochrogaster) may also fulfill criteria of mammalian eusociality. Understanding the evolution of eusociality in mole-rats requires answers to two primary questions: (1) What are the preconditions for the development of their eusocial systems? (2) Why do offspring remain in the natal group rather than dispersing and reproducing? Eusociality in mammals is by definition a special case of monogamy (more specifically: monogyny one female breeding), involving prolonged pair bonding for more than one breeding period. We argue that eusociality in mole-rats evolved from a monogamous mating system where cooperative brood care was already established. A tendency for group living is considered to be an ancestral (plesiomorph) trait among African bathyergid mole-rats , linking them to other hystricognath rodents. A solitary lifestyle seen in some genera, such as Bathyergus, Georychus, and Heliophobius, is assumed to be a derived trait that arose independently in different lineages of bathyergids, possibly as a consequence of selective constraints associated with the subterranean environment. In proximate terms, in eusocial mole-rats either puberty is assumed to be developmentally delayed so that under natural conditions most animals die before dispersal is triggered (e.g., in the case of Heterocephalus) or dispersal is induced only by an incidental encounter with an unfamiliar, yet adequate sexual partner (e.g., in the case of Cryptomys). Ultimately, a combination of strategies involving either dispersal and/or philopatry can be beneficial, especially in a highly unpredictable environment. If genetic relatedness among siblings is high (e.g., a coefficient of relatedness of 0.5 or more), then philopatry would not invoke an appreciable loss of fitness, especially if the cost of dispersing is higher than staying within the natal group. High genetic relatedness is more likely in a monogamous mating system or a highly inbred population. In this paper, we argue that the preconditions for eusociality in bathyergid mole-rats were a monogamous mating system and high genetic relatedness among individuals. We argue against the aridity food-distribution hypothesis (AFDH) that suggests a causal relationship between cooperative foraging for patchily distributed resources and the origin of eusociality. The AFDH may explain group size dynamics of social mole-rats as a function of the distribution and availability of resources but it is inadequate to explain the formation of eusocial societies of mole-rats, especially with respect to providing preconditions conducive for the emergence of eusociality. The ecological constraints prevalent in arid environments have promoted the evolution of social groups with a high reproductive skew in mole rat species occurring there. Outbred social bathyergids face conflict between maintaining colony integrity to enhance personal foraging success and hence survival, and dispersal to maximum individual lifetime reproductive success (LRS). This conflict will be a crucial determinant of the response of colony members to the presence of foreign conspecifics. We investigated how ecological constraints, sex, and reproductive status influence the outcome of meetings between foreign common mole rats (Cryptomys hottentotus hottentotus) in a series of dyadic encounters. Individuals from two localities, at the extremes of an aridity gradient, were used to assess the effects of aridity. The effects of sex and reproductive status were investigated by trials between individuals of differing sex and status. The arid population revealed substantially higher levels of rejection than the mesic population. Sex and status played a significant role in moderating individual response. For both populations, encounters between different-sexed individuals produced lower levels of rejection than those between same-sexed individuals. For the mesic site, rejection was greatest for encounters between reproductive animals. Conversely, for the arid site, the levels of rejection were comparatively high and comparable for all combinations of reproductive status. Ecological constraints, sex, and reproductive status are significant factors in interactions between foreign common mole rats, ultimately influencing individual survival, colony integrity and the differential LRS of all colony members. We investigated some of the ecological determinants of sociality in the Damaraland mole-rat, including the spatial distribution and biomass of resources (geophytes) available to foraging Damaraland mole-rats in partly vegetated sand dunes in the Kalahari and in grasslands near Dordabis, Namibia, and the foraging behaviour and residency characteristics of colonies at Dordabis. In both study areas, the geophytes had a clumped distribution, but the highest coefficients of dispersion and mean biomass occurred in the Kalahari where the principal food was the gemsbok cucumber. However, because the coefficient of digestibility was lower in geophytes from the Kalahari than from Dordabis, and the mole-rats only ate about half of a gemsbok cucumber, there was less energy available to mole-rats in the Kalahari. At Dordabis, large established colonies occur in the areas with the richest resources and remain resident in the same area for many years; within this area they search (blindly) for food during brief periods when the soil, at burrow depth, is moist and easily worked. Initially, long straight burrows are dug and few bulbs are taken; once the soil dries, minor changes are made to the burrow system as the mole-rats exploit the food patches they located immediately after the rain. Our results show that the characteristics of the resources, and the short time interval during which location of new resources is possible, favour group living; however, the constraints imposed by these features affect large and small colonies in different ways. Small colonies are more likely to fail than large ones and some crucial factors in the survival of these newly formed colonies are the richness of the area in which their burrows are located, and the size of the colony work force available to locate the food. Abstract: Trib2 is a member of the Trib family of serine/threonine kinase-like proteins (Trib1, Trib2, Trib3). Mice reconstituted with hematopoietic stem cells (HSC) retrovirally expressing Trib2 uniformly developed fatal transplantable acute myelogenous leukemia (AML). Trib2-induced AML was clonal and we sought to identify cooperating genes in Trib2-induced AML. Using Splinkerette PCR techniques, we identified proviral insertion near HoxA9 in a Trib2 monoclonal tumor, which resulted in greatly elevated HoxA9 expression. Mice reconstituted with HSC cotransduced with HoxA9 and Trib2 had accelerated onset of AML compared to either gene alone. These data identify Trib2 and HoxA9 as cooperating genes in AML. Eusociality, which occurs among mammals only in two species of African mole-rat, is characterized by division of labour between morphologically distinct ‘castes’. In Damaraland mole-rats (Cryptomys damarensis), colony labour is divided between ‘infrequent worker’ and ‘frequent worker’ castes. Frequent workers are active year-round and together perform more than 95% of the total work of the colony, whereas infrequent workers typically perform less than 5% of the total work. Anecdotal evidence suggests that infrequent workers may act as dispersers, with dispersal being limited to comparatively rare periods when the soil is softened by moisture. Here we show that infrequent workers and queens increase their daily energy expenditure after rainfall whereas frequent workers do not. Infrequent workers are also fatter than frequent workers. We suggest that infrequent workers constitute a physiologically distinct dispersing caste, the members of which, instead of contributing to the work of the colony and helping the queen to reproduce, build up their own body reserves in preparation for dispersal and reproduction when environmental conditions are suitable. I examined defense against conspecific intruders in colonies of the Damaraland mole-rat. I introduced foreign animals to captive colonies and examined the effect of the sex and breeding status of the intruder on the sex and breeding status of those that defended. Intruding mole-rats were almost always attacked. Colony defense against intruders was carried out almost exclusively by dominant, reproductive individuals and was directed primarily toward individuals of the same sex. Nonbreeding, subordinate mole-rats participated only rarely. These results contrast sharply with colony defense in the related naked mole-rat and suggest that investment in defense activities in Damaraland mole-rats primarily reflects repulsion of potential competitors for breeding status. Subordinate mole-rats may lack incentive to participate in potentially costly defense, as they are unlikely to gain reproductive opportunities within the colony and probably gain fitness primarily through dispersing to breed. Giant mole-rats ( Fukomys mechowii ) are remarkably long-lived subterranean rodents (maximum recorded lifespan as reported here greater than 26 years) that live in families with one reproductive pair (breeders) and their non-reproductive offspring (non-breeders). Previous studies have shown that breeders live on average approximately twice as long as non-breeders, a finding contradicting the classic trade-off between reproduction and lifespan. Because recent evidence points to the hypothalamic-pituitary-adrenal axis as playing an important role in shaping the pace of ageing in mole-rats, we analysed the influence of the social environment of giant mole-rats on intrafamilial aggression levels, indicators of long-term stress, and, ultimately, mortality. Behavioural data indicated that family constellation, especially the presence or the absence of parents, influences agonistic behaviour. As a measure of long-term stress, we established a non-invasive method of extracting and measuring cortisol from hair of giant mole-rats. Interestingly, orphaned non-breeders exhibited significantly lower levels of cortisol and lower mortality rates than did non-breeders living with both parents. Because hypercortisolism is harmful in the long-term, intrafamilial stress could help explain the earlier onset of senescence in non-breeders, resulting in a shorter lifespan. Our findings suggest that the social environment should be considered as a further factor in ageing studies involving group-living animals. Naked mole-rats express many unusual traits for such a small rodent. Their morphology, social behaviour, physiology, and ageing have been well studied over the past half-century. Many early findings and speculations about this subterranean species persist in the literature, although some have been repeatedly questioned or refuted. While the popularity of this species as a natural-history curiosity, and oversimplified story-telling in science journalism, might have fuelled the perpetuation of such misconceptions, an accurate understanding of their biology is especially important for this new biomedical model organism. We review 28 of these persistent myths about naked mole-rat sensory abilities, ecophysiology, social behaviour, development and ageing, and where possible we explain how these misunderstandings came about., (© 2020 The Authors. The African mole-rat family (Bathyergidae) includes the first mammalian species identified as eusocial: naked mole-rats. Comparative studies of eusocial and solitary mole-rat species have identified differences in neuropeptidergic systems that may underlie the phenomenon of eusociality. These differences are found in the oxytocin, vasopressin and corticotrophin-releasing factor (CRF) systems within the nucleus accumbens, amygdala, bed nucleus of the stria terminalis and lateral septal nucleus. As a corollary of their eusociality, most naked mole-rats remain pre-pubertal throughout life because of the presence of the colony's only reproductive female, the queen. To elucidate the neuroendocrine mechanisms that mediate this social regulation of reproduction, research on the hypothalamo-pituitary-gonadal axis in naked mole-rats has identified differences between the many individuals that are reproductively suppressed and the few that are reproductively mature: the queen and her male consorts. These differences involve gonadal steroids, gonadotrophin-releasing hormone-1 (GnRH-1), kisspeptin, gonadotrophin-inhibitory hormone/RFamide-related peptide-3 (GnIH/RFRP-3) and prolactin. Naked mole-rats Heterocephalus glaber (NMRs) are the longest-lived rodent and also resist the normal signs of senescence. In a number of species, cellular ageing has been correlated with a reduction in telomere length, yet relatively little is known about telomeres and their age-related dynamics in NMRs and other African mole-rats. Here, we apply fluorescence in situ hybridisation (FISH) to quantify telomeric repeat sequences in the NMR, the Damaraland mole-rat, Fukomys damarensis (DMR) and the Mahali mole-rat, Cryptomys hottentotus mahali (MMR). Both terminal and non-terminal telomeric sequences were identified in chromosomes of the NMR and DMR, whilst the MMR displayed only terminal telomeric repeats. Measurements of tooth wear and eruption patterns in wild caught DMRs and MMRs, and known ages in captive bred NMRs, were used to place individuals into relative age classes and compared with a quantitative measure of telomeric fluorescence (as a proxy for telomere size). While NMRs and MMRs failed to show an age-related decline in telomeric fluorescence, the DMR had a significant decrease in fluorescence with age, suggesting a decrease in telomere size in older animals.
