Faced with the challenges of global warming, which are likely to have a strong impact on the generation currently in school, the role of the latter is questioned by teachers supporting youth movements, such as the climate strike. A research-training project aimed at teachers wishing to develop and share their practices to integrate the climate emergency is described and the speeches produced in this context are analyzed. Several strong themes emerge from this analysis, particularly in the form of concerns and projected avenues. The teachers' comments are discussed in light of trends aimed at making the environment a school subject that has existed since the 1970s. The results highlight the diversity and richness of the activities offered and the strong commitment offered by these teachers to integrate the climate emergency into their practices. They also highlight the paradoxical role given to students, moving from the role of a driving force to that of a brake on commitment. Growing concerns regarding health and eco-safety have led to a resurgence in the recognition of natural products. Although plant colorants are often mentioned in ethnobotanical studies in northern Thailand, they have not been comprehensively examined. This study aims to (i) investigate the colorant plants used by ethnic minorities in northern Thailand and (ii) compare colorant plants used among the groups in northern Thailand with other regions worldwide. Data on colorant plants used by ten ethnic groups across 142 villages in northern Thailand were extracted in a systematic review of 42 published references. The importance of the colorant plant species was evaluated using the ethnobotanical index "use value" (UV), and the homogeneity of knowledge was assessed using the "information consensus factor" (ICF). The similarity of colorant plants used among the ethnic minority groups and beyond was evaluated using Jaccard's index. The 42 published references included information about 104 colorant plant species belonging to 85 genera and 43 families from seven provinces in northern Thailand. This represents nearly half of the 212 colorant plant species reported across all 72 provinces in Thailand. The most important species were Strobilanthes cusia and Morinda angustifolia, which are sources of blue and red colors. Fabaceae, Rubiaceae, and Lamiaceae were the plant families with most species used as colorants. The colorant plant species yielded ten different color shades, with blue being the most prevalent. The Jaccard's index varied from 0 to 0.27 among the northern Thai ethnic minorities and from 0 to 0.13 for regions outside of northern Thailand. The regions located closest to northern Thailand exhibited the highest degree of similarity. This study provides valuable insights into the traditional knowledge and usage of dye plants in northern Thailand, contributing to preserving the cultural heritage and providing basic knowledge for the sustainable use of natural colorants for modern applications. Background: Persistent cognitive impairment is frequent across bipolar disorder (BD) and major depressive disorder (MDD), highlighting an urgent need for pro-cognitive treatments. Aim: This study investigated effects of erythropoietin (EPO) on cognitive impairment and dorsal prefrontal cortex (dPFC) activity in affective disorders. Methods: In this randomized, double-blinded, placebo-controlled trial, cognitively impaired patients with remitted BD or MDD received 1 weekly recombinant human EPO (40,000 IU/mL) or saline infusion for a 12-week period. Assessments were conducted at baseline, after 2 weeks of treatment (week 3), immediately after treatment (week 13) and at 6-months follow-up. Participants underwent functional MRI during performance on a n-back working memory (WM) task at baseline and week 3, and for a subgroup 6 weeks post-treatment (week 18). The primary outcome was a cognitive composite score at week 13, whereas secondary outcomes comprised sustained attention and functioning. WM-related dPFC activity was a tertiary outcome. Results: Data were analysed for 101 of the 103 included patients (EPO, n = 58; saline, n = 43). There were no effects of EPO over saline on any cognitive or functional outcomes or on WM-related dPFC activity. Conclusions: The absence of treatment-related changes in cognition and neural activity was unexpected and contrasts with multiple previous preclinical and clinical studies. It is possible that the lack of effects resulted from a recent change in the manufacturing process for EPO. Nevertheless, the findings support the validity of dPFC target engagement as a biomarker model for pro-cognitive effects, according to which treatments that do not improve cognition should not modulate dPFC activity. The authors investigated the frequency and determinants of long-term use and risk of dose escalation of benzodiazepines and benzodiazepine-related drugs (benzodiazepine receptor agonists, or BZRAs). All adults ages 20–80 years living in Denmark on January 1, 2000 (N=4,297,045) were followed for redeemed prescriptions of BZRAs in the Danish National Prescription Registry from January 1, 2000, to December 31, 2020. For each drug class, we calculated long-term use for more than 1 or 7 years, and dose escalation measured as increase in dose to a level above the recommended level. Associations were examined using logistic regression. The authors identified 950,767 incident BZRA users, of whom 15% and 3% became long-term users for more than 1 or 7 years, respectively. These percentages were highest for individuals who initiated Z-drugs (17.8% and 4%). Among the 5% of BZRA users who had at least 3 years of continuous use, there was no indication of dose escalation, as the median dose remained relatively stable. However, 7% (N=3,545) of BZRA users escalated to doses above the recommended level. Psychiatric comorbidity, especially substance use disorder, was associated with higher risk of long-term use and dose escalation. A limited portion of the population that received BZRA prescriptions were classified as continuous users, and only a small proportion of this group escalated to doses higher than those recommended in clinical guidelines. Los bosques tropicales del Chocó biogeográfico presentan una de las mayores concentraciones de especies de plantas del mundo. Las palmas (Arecaceae) son una de las familias más importantes en las tierras bajas del Chocó, pero su ecología está poco estudiada. Investigamos la diversidad de comunidades de palmas en el Chocó y su relación con la precipitación en 48 transectos distribuidos en un gradiente de 2000–10 000 mm de precipitación. En cada transecto (5×500 m) se identificaron todas las especies de palmas y se contabilizaron todos los individuos. Registramos un total de 57 especies de palmas (promedio ± DS: 17 ± 8 por transecto) y 37 955 individuos (791 ± 446). Encontramos que la riqueza y abundancia de palmas están significativamente relacionadas con la precipitación (r² = 0,86; P > 0,001). Se registró una fuerte correlación positiva entre la composición florística de las comunidades de palmas y la distancia geográfica (rm = 0,63; P = 0,001). La riqueza registrada es una de las más altas para el Neotrópico. La relación entre la precipitación, riqueza y abundancia de palmas por primera vez se documenta a escala regional en este punto caliente de biodiversidad.

Abstract Background Treatment options in metastatic breast cancer are limited. New therapies preferable with predictive biomarkers are needed. The aim of these trials was to investigate if gene copy number of the topoisomerase 1 gene was predictive of response to the topoisomerase inhibitor irinotecan. Methods Two open-label, single-arm phase II studies including HER2 positive and negative patients were conducted. Patients were eligible for inclusion if the primary tumor or a metastatic lesion had increased expression of the topoisomerase 1 gene defined as a TOP1 gene copy number of ≥4 or a TOP1/CEN20 ratio of ≥2. Patients were treated with irinotecan +/− trastuzumab weekly for 4 weeks following 2 weeks break, until progression or unacceptable toxicities. Evaluation scans were performed every 6 weeks. Primary endpoint was clinical benefit rate defined as the fraction of patients with stable disease for ≥4 months. Results The pre-planned number of 18 patients in each trial was not reached, thus no formal statistical analysis could be performed. Nine patients with HER2 negative disease and three patients with HER2 positive disease were included. Three patients obtained a partial remission and two patients had SD. Conclusions The trials did not include the planned number of patients. No association between gene copy number of the topoisomerase 1 gene and response to irinotecan could be proved, however a clinical benefit was found in 5/12 patients and in 2/3 patients with HER2 positive disease. This could call for further investigation of the drug in the metastatic setting, especially in HER2 positive BC. Trial registration Eudract registration numbers 2012–002348-26 and 2012–002347-23. We studied how range sizes and shifts in species ranges depend on niche breadth in European dragonflies. We measured range sizes and shifts over a 22-year period (1988–2010) and grouped species into those reproducing in permanent running (perennial lotic) water, permanent standing (perennial lentic) water, and temporary (running or standing) water. Running water species are more specialized and have narrower niches with a more fixed niche position than standing water species. Temporary water species are more generalist and have broader niches without a fixed niche position as clear as permanent water species because they may utilize both temporary and permanent habitats. Running water species have smaller ranges, and some of them have contracted their ranges more than species reproducing in standing or temporary waters; that is, they are especially at risk of habitat loss and climate change because of the joint effects of their narrow niches and small range sizes. Temporary water species track climate changes better than permanent water species. This suggests that ecological specialization may cause contemporary range shifts to lag behind changes in climate and resources. Furthermore, it indicates that recent changes in climate and human land use cause biotic homogenization, where specialists are outperformed and replaced by generalists.

Ethnomedicinal studies have demonstrated that Fabaceae include the highest number of medicinal species of all plant families in Thailand. To evaluate which legumes hold the greatest potential for development of public herbal medicines, we used ethnobotanical indices to screen traditional medicinal species from 152 literature sources published from 1986–2019. The homogeneity of each use category was evaluated using the informant consensus factor (ICF). The popularity of the used species was evaluated by percentage of fidelity level (FL) and use value (UV). A total of 261 species from 98 genera of legumes were recorded for their traditional medicinal uses. The uses included 5387 use reports that covered treatments of 420 disorders. The ethnobotanical indices pointed to 15 species with high use values (e.g., Biancaea sappan (L.) Tod., Cassia fistula L., Senna siamea (Lam.) H.S.Irwin & Barneby). Nine of these species are already used in public herbal medicine. The other six species should be studied in greater depth, especially to identify their bioactive compounds so they can be developed for use in public herbal medicine.

Background Cannabinoids; tetrahydrocannabinol (THC), cannabidiol (CBD) and cannabinol (CBN), might show antibacterial activity. Trema orientalis is a species in the Cannabaceae that is closely related to Cannabis through plastome phylogenetic evidence. This species is widely distributed throughout tropical Asia and is used as traditional medicine, particularly for the treatment of infectious diseases. However, no studies on the antibacterial activity of cannabinoid-containing inflorescences extracts are available. Thus, the aim of this study was to determine cannabinoid content and antibacterial activity of inflorescences fractions from T. orientalis native to Thailand. Methods We hypothesized that inflorescences from T. orientalis might display cannabinoids similar to Cannabis because of their close taxonomic relationship. We extracted the mature inflorescences and infructescence of T. orientalis in three disparate populations from different Thailand floristic regions. Extractions were subsequently partitioned into hydrophilic and lipophilic fractions using distilled water and chloroform. The lipophilic extracts were further fractionated by the column chromatography with gradient elution and analyzed by gas chromatography-mass spectrometry (GC-MS). Characterized cannabinoids were used in bioassays with multidrug-resistance bacteria. Results Lipophilic extracts and fractions of inflorescences from all Thailand floristic regions consistently displayed cannabinoids (THC, CBD and CBN) in various quantities. These extracts exhibited inhibitory activity for Staphylococcus aureus, Pseudomonas aeruginosa, and Acinetobacter baumannii strains with minimum inhibitory concentration values varying from 31.25 to 125 µg/mL. Conclusion Our study is the first to report cannabinoid detection in extracts from inflorescences of T. orientalis, a species in the Cannabaceae. These extracts and their fractions containing cannabinoids showed pronounced antibacterial activity. The use of analytic methods also demonstrated reproducible cannabinoid extraction.

Højmoser repræsenterer de største tørvedækkede områder i Danmark, og en undersøgelse af disse områder kan belyse menneskelig indflydelse på de danske tørvejorder. Vi har studeret den menneskelige indflydelse ved Store Vildmose og anvendt arealanvendelsen fra topografiske kort af forskellig alder (høje målebordsblade, lave målebordsblade og 4-cm-kort), punktopmålt højdeinformation fra bymålingerne, en moderne LiDAR (Light Detection and Radar) baseret højdemodel samt boredata, som giver information om tørvens tykkelse. Formålet med undersøgelsen har været at kvantificere ændringer i Store Vildmoses overfladehøjde forårsaget af både tørvedannelse, tørvesætning (material-isering og afvanding) og tørvegravning fra 1880 til 2010.

Synaptotagmins confer calcium-dependence to the exocytosis of secretory vesicles, but how coexpressed synaptotagmins interact remains unclear. We find that synaptotagmin-1 and synaptotagmin-7 when present alone act as standalone fast and slow Ca2+-sensors for vesicle fusion in mouse chromaffin cells. When present together, synaptotagmin-1 and synaptotagmin-7 are found in largely non-overlapping clusters on dense-core vesicles. Synaptotagmin-7 stimulates Ca2+-dependent vesicle priming and inhibits depriming, and it promotes ubMunc13-2- and phorbolester-dependent priming, especially at low resting calcium concentrations. The priming effect of synaptotagmin-7 increases the number of vesicles fusing via synaptotagmin-1, while negatively affecting their fusion speed, indicating both synergistic and competitive interactions between synaptotagmins. BackgroundWeight loss after bariatric surgery varies widely between individuals, partly due to genetic differences. In addition, genetic determinants of abdominal obesity have been shown to attenuate weight loss after dietary intervention with special attention paid to the rs1358980-T risk allele in the VEGFA locus. Here we aimed to test if updated genetic risk scores (GRSs) for adiposity measures and the rs1358980-T risk allele are linked with weight loss following gastric bypass surgery.MethodsFive hundred seventy six patients with morbid obesity underwent Roux-en-Y gastric bypass. A GRS for BMI and a GRS for waist-hip-ratio adjusted for BMI (proxy for abdominal obesity), respectively, were constructed. All patients were genotyped for the rs1358980-T risk allele. Associations between the genetic determinants and weight loss after bariatric surgery were evaluated.ResultsThe GRS for BMI was not associated with weight loss (β = -2.0 kg/100 risk alleles, 95% CI -7.5 to 3.3, p = 0.45). Even though the GRS for abdominal obesity was associated with an attenuated weight loss response adjusted for age, sex and center (β = -14.6 kg/100 risk alleles, 95% CI -25.4 to -3.8, p = 0.008), it was not significantly associated with weight loss after adjustment for baseline BMI (β = -7.9 kg/100 risk alleles, 95% CI -17.5 to 1.6, p = 0.11). Similarly, the rs1358980-T risk allele was not significantly associated with weight loss (β = -0.8 kg/risk allele, 95% CI -2.2 to 0.6, p = 0.25).DiscussionGRSs for adiposity derived from large meta-analyses and the rs1358980-T risk allele in the VEGFA locus did not predict weight loss after gastric bypass surgery. The association between a GRS for abdominal obesity and the response to bariatric surgery may be dependent on the association between the GRS and baseline BMI.

Preclinical studies suggest that some effects of conventional chemotherapy, and in particular, gemcitabine, are mediated through enhanced antitumor immune responses. The objective of this study was to use material from a randomized clinical trial to evaluate whether patients with preexisting immune infiltrates responded better to treatment with gemcitabine + docetaxel (GD) compared to docetaxel alone. Formalin fixed, paraffin-embedded breast cancer tissues from SBG0102 phase 3 trial patients randomly assigned to treatment with GD or docetaxel were used. Immunohistochemical staining for CD8, FOXP3, LAG3, PD-1, PD-L1 and CD163 was performed. Tumor infiltrating lymphocytes (TILs) and tumor associated macrophages were evaluated. Prespecified statistical analyses were performed in a formal prospective-retrospective design. Time to progression was primary endpoint and overall survival secondary endpoint. Correlations between biomarker status and endpoints were evaluated using the Kaplan–Meier method and Cox proportional hazards models. Biomarker data was obtained for 237 patients. There was no difference in treatment effect according to biomarker status for the whole cohort. In planned subgroup analysis by PAM50 subtype, in non-luminal (basal-like and HER2E) breast cancers FOXP3 was a significant predictor of treatment effect with GD compared to docetaxel, with a HR of 0.22 (0.09–0.52) for tumors with low FOXP3 compared to HR 0.92 (0.47–1.80) for high FOXP3 TILs (Pinteraction = 0.01). Immune biomarkers were not predictive of added benefit of gemcitabine in a cohort of mixed breast cancer subtypes. However, in non-luminal breast cancers, patients with low FOXP3+ TILs may have significant benefit from added gemcitabine.

Depression is a mental illness affecting more than 260 million people worldwide. In Greenland, the prevalence of patients treated with antidepressant medicine (antidepressants) has not previously been investigated. The aim of this study was to estimate and compare the age- and gender-specific prevalence of patients treated with antidepressants in Greenland and Denmark. The study was a cross-sectional register study using data from population and medical registers in Greenland and Denmark. Included was 1,573 Greenlandic patients and 419,151 Danish patients treated with antidepressants, respectively. The aim of this study was to estimate the age- and gender-specific prevalence and quality of care among patients using medication targeting obstructive lung disease in the five regions of Greenland. The study was designed as a cross-sectional study. Data on patients using medication targeting obstructive lung disease was obtained from the electronically medical record used in Greenland. The prevalence was calculated using the population of Greenland as background population. The quality of care was determined using indicators proposed by international literature and the Steno Diabetes Center Greenland guidelines. The total prevalence of patients using medication targeting obstructive lung disease was 7.5%. The prevalence was significantly higher among women compared to men and differed significantly between the five regions. Smoking status, blood pressure and spirometry were registered within one/two years for 29.8%/43.2%, 29.2%/41.1% and 15.9%/26.0% of the patients, respectively. Regional differences were observed for all indicators. The use of medication targeting obstructive lung disease is common in Greenland. Yet, the quality of care was low and interventions improving the quality of care is recommended.
