There is currently a lack of research investigating the effectiveness of commercial broiler enrichments, and in particular the ability of these additions to create opportunities for positive welfare. One aim of this study was to investigate whether offering broiler chickens enrichments that have recently been found to be preferred leads to increased levels of activity. A second aim was to investigate the emotional effects of provision of these enrichments by assessing levels of fearfulness and play-like activity. Commercially housed broilers were assigned to treatment houses containing either: (1) platform perches, (2) platform perches+peat dust baths, (3) no enrichment (control). Activity levels and play behaviours in unenriched areas of the house were measured in weeks 3, 4 and 5. Levels of active behaviours, such as foraging and locomotion, were determined from video recordings of undisturbed birds in unenriched areas of the house. To stimulate play-like behaviours an observer walked through the birds, displacing them and creating a space. The broilers using the space were then filmed for 5 min and the occurrences of frolicking, sparring and food-running were recorded. Fearfulness of broilers in unenriched areas of the house, and also when using enrichments, was measured using observer avoidance tests in week 5. We found that creating space among the broilers was a successful method of stimulating play (largely sparring and frolicking), with play being observed in 93% of videos, however the presence of enrichments did not have an effect on the level of play recorded (P>0.05). There was also no treatment effect on activity levels of broilers in unenriched areas (P>0.05), however levels of overall activity decreased as broilers aged. Compared with the control, flight distances in unenriched areas were significantly lower in the perches+dust bath treatment (P=0.026), and were numerically lower in the perches treatment. This suggests a reduction in fearfulness with increased environmental complexity, and thus possible welfare benefits. It is suggested that further research should investigate whether increasing the level of provision of these enrichments leads to more marked improvements in welfare.

Introduction Optimal immunisation programme service delivery and childhood vaccine coverage remains an ongoing challenge in South Africa. Previous health systems approaches have made recommendations on how to address identified barriers but detailed local implementation studies are lacking. This study aimed to improve immunisation service delivery in children under 24 months in Khayelitsha, Western Cape Province using an adaptive, co-design approach to assess and improve childhood immunisation service delivery at the clinic level.Methods A rapid, adaptive approach to identification of barriers and assessment of current childhood immunisation service delivery was developed with three clinics in Khayelitsha, Western Cape Province. This informed a short co-design process with key stakeholders and service providers to develop local interventions targeted at high priority barriers. Interventions were implemented for 4–6 months and evaluated using theory-based evaluation tools. Clinic service delivery, satisfaction and changes to clinic processes and parent engagement and knowledge were measured.Results Interventions developed included weekly community immunisation education radio sessions, daily clinic health talks, immunisation education and promotion materials and service provider and parent quality checklists. Evaluation post-intervention showed improvement in parents’/guardians’ knowledge about immunisation, parent engagement and service provider commitment to improvement in service quality. Radio sessions and immunisation education and communication materials were deemed most useful by parents and providers.Conclusion Immunisation service delivery can be strengthened using an adaptive, clinic-led assessment process which can effectively identify barriers, inform co-designed interventions and be evaluated over a short period. This approach provides a framework to guide future local participatory action research to more effectively improve childhood immunisation service delivery and other child health services in under-resourced settings.

As teachers are being viewed as critical agents of educational change, conceptions and approaches of how to prepare teachers are changing. Among these changing conceptions of teacher preparation, the kinds of knowledge prospective teachers need to develop so as to be effective at teaching are often at the center of the debates. Pedagogical content knowledge (PCK) is considered a crucial part of the knowledge base for teaching and was anticipated to have the greatest impact on teachers' classroom actions. The purpose of this study was to investigate the extent to which science methods courses help prospective teachers to develop PCK. A content analysis method was used. Course modules and assessment tools were the sources of data for this study. The objectives and the subtitles of the sections of modules and items of course assignments and final exams were analyzed. The observations were coded against the 3 knowledge bases of teaching. The observations that were coded as PCK were further coded in terms of the 5 components of PCK along with important descriptors that were later used for analysis. The results of this study showed that the courses did not adequately foster preservice teachers' PCK. Despite an awareness of the purposes of the courses that was evident in the formulation of objectives and the preliminary notes in the introductory parts of the course modules, the elements of the courses as a whole were very far from helping preservice teachers develop PCK for teaching secondary school science subjects. Moreover, of those elements of the courses that were identified as fostering PCK, the knowledge of instructional strategies component was given due emphasis and the knowledge of assessment in science component was seldom addressed. The findings imply a need to rethink ways of preparing curriculum materials in light of PCK and also rethink the pathways of becoming a teacher educator.

Within the military, cognitive readiness is essential to ensure the warfighter can return to highly demanding combat training and deployment operations. The warfighter must be able to make split second decisions and adapt to new tools and environments. After a traumatic brain injury, clinicians helping the warfighter must have techniques that address warfighter cognitive readiness. Current rehabilitation for cognitive complaints used in military medicine are modeled after civilian therapies which focus on remediating moderate to severe impairment through building compensatory strategies. This traditional approach to cognitive rehabilitation does not translate well to mild traumatic brain injury (mTBI) where impairments are subtle, nor does it meet the needs of our warfighters in deployed and combat training environments. Challenging our current methods is critical in adapting to the needs of this highly valued population to ensure that our warfighters are able to carry out mission critical decision making. Here we present a review of our best current practices for cognitive rehabilitation, describe the limitations our traditional approaches impose for mTBI in military personnel, and present an alternative treatment called Strategic Memory Advanced Reasoning Training (SMART) that can be adopted through a randomized clinical trial design. We propose directly comparing traditional treatment approaches with a novel cognitive rehabilitation strategy which has been well validated outside of the military setting. Procedures were developed to execute this clinical trial in a way that is most relevant to the study population by establishing ecologically valid outcome metrics.

Despite the addition of several new agents to the armamentarium for the treatment of multiple myeloma (MM) in the last decade and improvements in outcomes, the refractory and relapsing disease continues to take a great toll, limiting overall survival. Therefore, additional novel approaches are needed to improve outcomes for MM patients. The oncogenic transcription factor MYC drives cell growth, differentiation and tumor development in many cancers. MYC protein levels are tightly regulated by the proteasome and an increase in MYC protein expression is found in more than 70% of all human cancers, including MM. In addition to the ubiquitin-dependent degradation of MYC by the 26S proteasome, MYC levels are also regulated in a ubiquitin-independent manner through the REGγ activation of the 20S proteasome. Here, we demonstrate that a small molecule activator of the 20S proteasome, TCH-165, decreases MYC protein levels, in a manner that parallels REGγ protein-mediated MYC degradation. TCH-165 enhances MYC degradation and reduces cancer cell growth in vitro and in vivo models of multiple myeloma by enhancing apoptotic signaling, as assessed by targeted gene expression analysis of cancer pathways. Furthermore, 20S proteasome enhancement is well tolerated in mice and dogs. These data support the therapeutic potential of small molecule-driven 20S proteasome activation for the treatments of MYC-driven cancers, especially MM.

Provision of an appropriate dustbathing substrate may allow broiler chickens to satisfy a natural motivation and give them an opportunity to exercise. The main aim of this study was to evaluate the extent to which different substrates promote dustbathing behaviour in broilers. The trial was replicated over three production cycles in one commercial broiler house, with ~22 000 Ross broilers housed per cycle. The birds were provided with access to five experimental substrates from day 10 of the 6-week production cycle. The substrates included the following: (1) peat (P), (2) oat hulls (OH), (3) straw pellets (SP), (4) clean wood shavings (WS), and (5) litter control (C). The substrates were provided in 15 steel rings (1.1 m in diameter, three rings per substrate) dispersed throughout the house. The level of occupancy of the rings, behaviours performed in each substrate, and the effect of ring position (central or edge of house) were assessed in weeks 3, 4, 5 and 6 using scan sampling from video footage. Where substrates successfully promoted dustbathing, the length and components of the bouts (including number of vertical wing shakes and ground pecks) were also assessed. Results showed that birds used P significantly more than the remaining substrates for dustbathing (P0.05). The use of OH is likely to be more environmentally sustainable than that of P, and our results suggest that this substrate is relatively successful in promoting dustbathing. However, a preference was still observed for P and further work should investigate whether other suitable substrates could better reflect its qualities.
