The egg parasitoids Trichogramma minutum Riley and Trichogramma platneri Nagarkatti are commonly used in North America for the control of Lepidoptera in orchards, These species are morphologically indistinguishable, but reproductively incompatible. An easy identification technique is highly desirable, because release of the wrong species in the native area of the other species may result in a depression of intended biological control. Here we show that the DNA sequences of the mitochondrial cytochrome oxidase II (COII) gene differ between the species. These differences can be visualized by restricting the COII PCR product with the restriction enzyme; VspI. An emerging body of evidence indicates a role for plant genotype as a determinant of the species and genetic composition of the saprophytic microbial community resident to the rhizosphere. In this study, experiments were conducted to determine the capacity of five different wheat cultivars to enhance resident populations and support introduced strains of 2,4-diacetylphloroglucinol (2,4-DAPG)-producing fluorescent pseudomonads, a group of bacteria known to provide biological control of several soilborne diseases. When soils were cropped with three successive 28-day growth cycles of wheat, the 2,4-DAPG-producing strains were consistently recovered from the rhizosphere of the cultivar Lewjain, and commonly were present at populations higher than those recovered from other wheat cultivars. Based on restriction fragment length polymorphism and sequence analyses of phlD, a key gene involved in 2,4-DAPG production, two previously undefined phlD+ genotypes, referred to as genotypes PfZ and PfY, were discovered. Wheat cultivar Lewjain was the primary source of genotype PfY while cultivar Penawawa yielded the majority of genotype PfZ. Based on 16S rDNA sequence analysis, both new phlD genotypes were classified as P. fluorescens. Comparison of the rhizosphere competence of 2,4-DAPG-producing P. fluorescens Q2-87 (genotype B) and P. fluorescens LR3-A28 (genotype PfY) showed that both strains persisted at similar populations in the rhizosphere of all cultivars tested over a 30 day period when introduced as a seed inoculant. However, when strain LR3-A28 was applied as a soil inoculant, this strain was recovered at higher populations from the rhizosphere of wheat cultivar Lewjain than from the rhizospheres of two other cultivars. No cultivar effects were shown for strain Q2-87. Collectively, these results add further to evidence indicating a degree of specificity in interactions between plant cultivars and specific members of the saprophytic microbial community. Furthermore, as 2,4-DAPG-producing fluorescent Pseudomonas spp. have a central role in the spontaneous reduction in severity of take-all disease of wheat in response to continuous wheat monoculture, we postulate that the use of specific cultivars, such as Lewjain, which possess a superior capacity to enhance resident soil populations of these bacteria may have potential to reduce the length of the monoculture period required to induce natural suppressiveness of soils toward this disease.

Species of Prosopis introduced into South Africa from the Americas for fuel wood, shade and fodder, have become naturalized and widespread in the dry northwestern areas of this country. Invasive Prosopis species have been the target of a biological control programme in South Africa since 1985. During a survey for potential fungal biological control agents in Mexico and Texas in 2001, a pod disease was recorded on Prosopis glandulosa in both countries. The disease is characterized by black/grey pycnidia, flattening of the pods, and seed decay. Morphological investigations of the causal organism showed it to be a Coniothyrium-like, coelomycete. However, based on conidiogenous cell morphology and proliferation, we concluded that the organism is not congeneric with Coniothyrium s. str. Phylogenetic analysis of the SSU gene placed this fungus in the Diaporthales. Parsimony analysis of the ITS region (ITS-1, 5.8S, ITS-2) revealed it to group closely to Cryphonectria and Endothia. Consequently, a new genus, Prosopidicola, with type species Prosopidicola mexicana, is proposed.

The lady beetle Coccinella septempunctata L. is an important predator of aphids in arable crops, but depends on noncrop landscape elements, such as hedgerows, for hibernation. We studied the effect of the shape, area, and fragmentation of noncrop landscape elements on the control of aphids by C. septempunctata using a spatially explicit simulation model. The model is based on a description of the phenology and population dynamics of aphids and C. septempunctata, as well as the predation dynamics and dispersal characteristics of C. septempunctata. The study compares biocontrol in 12 landscapes consisting of field crops and hedgerows that differ in the shape, proportion (1, 4, 9, and 16%), and fragmentation of the hedgerow elements (hedgerow area divided into 1, 4, or 16 elements) in the landscape. Linear hedgerow elements provided better control than square elements in 3 of the 12 simulated landscapes and resulted in similar levels of control in the other nine landscapes. The total area of hedgerow habitat was the key factor for the control of aphid populations. Landscapes with 9% and 16% noncrop habitat had large enough local populations of C. septempunctata to control aphid infestations, whereas landscapes with only 1% or 4% of hibernation area had no potential for improved aphid control. In landscapes with sufficient noncrop habitat, the best control was achieved when small hedgerow elements were evenly distributed over the landscape.

The antibiotic 2,4-diacetylphloroglucinol (2,4-DAPG) plays an important role in the suppression of plant pathogens by several strains of Pseudomonas spp. Based on the results of this study, there is variation within and among Pythium spp. to 2,4-DAPG. Also, various propagules of Pythium ultimum var. sporangiiferum, that are part of the asexual stage of the life cycle, differ considerably in their sensitivity to 2,4-DAPG. Mycelium was the most resistant structure, followed by zoosporangia, zoospore cysts, and zoospores. Additionally, we report for the first time that pH has a significant effect on the activity of 2,4-DAPG, with a higher activity at low pH. Furthermore, the level of acetylation of phloroglucinols is also a major determinant of their activity. Transmission electron microscopy studies revealed that 2,4-DAPG causes different stages of disorganization in hyphal tips of Pythium ultimum var. sporangiiferum, including alteration (proliferation, retraction, and disruption) of the plasma membrane, vacuolization, and cell content disintegration. The implications of these results for the efficacy and consistency of biological control of plant-pathogenic Pythium spp. by 2,4-DAPG-producing Pseudomonas spp. are discussed.

Compost amended soil has been found to be suppressive against plant diseases in various cropping systems. The level and reproducibility of disease suppressive properties of compost might be increased by the addition of antagonists. In the present study, the establishment and suppressive activity of two fungal antagonists of soil-borne diseases was evaluated after their inoculation in potting soil and in compost produced from different types of organic waste and at different maturation stages. The fungal antagonists Verticillium biguttatum, a mycoparasite of Rhizoctonia solani, and a non-pathogenic isolate of Fusarium oxysporum antagonistic to fusarium wilt, survived at high levels (103–105 CFU g–1) after 3 months incubation at room temperature in green waste compost and in potting soil. Their populations faded out in the organic household waste compost, especially in the matured product. In bioassays with R. solani on sugar beet and potato, the disease suppressiveness of compost increased or was similar after enrichment with V. biguttatum. The largest effects, however, were present in potting soil, which was very conducive for the disease as well as the antagonist. Similar results were found in the bioassay with F. oxysporum in carnation where enrichment with the antagonistic F. oxysporum had a positive or neutral effect. We foresee great potential for the application of antagonists in agriculture and horticulture through enrichment of compost or potting soil with antagonists or other beneficial micro-organisms.

Fusarium wilt, caused by Fusarium oxysporum f. sp. lycopersici, and Verticillium wilt, caused by either Verticillium albo-atrum or Verticillium dahliae, are devastating diseases of tomato (Lycopersicon esculentum) found worldwide. Monitoring is the cornerstone of integrated pest management of any disease. The lack of rapid, accurate, and reliable means by which plant pathogens can be detected and identified is one of the main limitations in integrated disease management. In this paper, we describe the development of a molecular detection system, based on DNA array technology, for rapid and efficient detection of these vascular wilt pathogens. We show the utility of this array for the sensitive detection of these pathogens from complex substrates like soil, plant tissues and irrigation water, and samples that are collected by tomato growers in their greenhouses.

White mould disease leads to production of sclerotia, which subsequently survive in soil and may be responsible for future epidemics. The effect of the mycoparasite Coniothyrium minitans in decreasing survival of sclerotia of Sclerotinia sclerotiorum was studied. Infection of sclerotia of S. sclerotiorum by C. minitans can be achieved by a single conidium. Under optimal conditions, 2 conidia per sclerotium produced 63% of the maximum infection (ca. 90%) of sclerotia produced by up to 1000 conidia. Similar results were observed on the infection of stem pieces infected by S. sclerotiorum. In field trials, application of conidial suspensions of C. minitans to a bean crop soon after white mould outbreak led to a higher percentage of sclerotial infection than later applications. Ninety per cent infection of sclerotia was obtained within 3 weeks of application by C. minitans suspensions in the range of 5 × 105 and 5 × 106 conidia ml¿1 at 1000 l ha¿1. The concentration of the conidial suspensions and the isolate used were of less importance. The result was marginally affected by the germinability of the conidia (75% against 61% infected sclerotia at 91% and 16% viability of isolate IVT1, respectively). Less apothecia of S. sclerotiorum developed in soil samples collected after 2 months from plots sprayed immediately after disease outbreak than from those treated 11¿18 days later. It is concluded that a suspension of 106 conidia ml¿1 in 1000 l ha¿1 (= 1012 conidia ha¿1) sprayed immediately after the first symptoms of disease are observed, results in > 90% infection of sclerotia of S. sclerotiorum. The infection of sclerotia, which prevents their carry-over, occurs within a broad range of inoculum quality

Carnivorous arthropods exhibit complex intraspecific and interspecific behaviour among themselves when they share the same niche or habitat and food resources. They should simultaneously search for adequate food for themselves and their offspring and in the meantime avoid becoming food for other organisms. This behaviour is of great ecological interest in conditions of low prey availability. We examined by means of an olfactometer, how volatile chemicals from prey patches with conspecific or heterospecific predators might contribute to shaping the structure of predator guilds. To test this, we used the exotic predatory mites Typhlodromalus manihoti and T. aripo, and the native predatory mite Euseius fustis, with Mononychellus tanajoa as the common prey species for the three predatory mite species. We used as odour sources M. tanajoa-infested cassava leaves or apices with or without predators. T. manihoti avoided patches inhabited by the heterospecifics T. aripo and E. fustis or by conspecifics when tested against a patch without predators. Similarly, both T. aripo and E. fustis females avoided patches with con- or heterospecifics when tested against a patch without predators. When one patch contained T. aripo and the other T. manihoti, females of the latter preferred the patch with T. aripo. Thus, T. manihoti is able to discriminate between odours from patches with con- and heterospecifics. Our results show that the three predatory mite species are able to assess prey patch profitability using volatiles. Under natural conditions, particularly when their food sources are scarce, the three predatory mite species might be involved in interspecific and/or intraspecific interactions that can substantially affect population dynamics of the predators and their prey.

The foraging behavior of Amitus fuscipennis MacGown & Nebeker and Encarsia formosa Gahan was studied on tomato leaflets with 20 Trialeurodes vaporariorum (Westwood) larvae in the first or third stage. Ten of the whitefly larvae were previously parasitized and contained a conspecific or a heterospecific parasitoid egg or larva. The host type (host stage and/or previous parasitization) did not influence the foraging behavior of either parasitoid species. The residence time on these tomato leaflets was about 0.9 h for A. fuscipennis and 1.9 h for E. formosa. Amitus fuscipennis hardly stood still and fed little, while E. formosa showed extensive standing still and feeding. As a result, the time walking while drumming was similar for both parasitoid species. The numbers of host encounters and ovipositions per leaflet were similar for both parasitoid species. However, the residence time of A. fuscipennis was half as long as that of E. formosa so the rate of encounters and ovipositions was higher for A. fuscipennis. Amitus fuscipennis is more efficient in finding and parasitizing hosts under these conditions. The walking activity and host acceptance of the synovigenic E. formosa diminished with the number of ovipositions, but not those of the proovigenic A. fuscipennis. Encarsia formosa is egg limited, while A. fuscipennis is time limited because of its short life span and high egg load. Both parasitoid species discriminated well between unparasitized larvae and self-parasitized larvae, but discriminated poorly those larvae parasitized by a conspecific and did not discriminate larvae parasitized by a heterospecific. Self-superparasitism, conspecific superparasitism, and multiparasitism were observed for both parasitoid species. Superparasitism always resulted in the emergence of one parasitoid and multiparasitism resulted in a higher emergence of one parasitoid of the species that had parasitized first. The data suggest that A. fuscipennis is a good candidate for use in biological control of high-density spots of T. vaporariorum when we consider its high encounter and oviposition rate.

Studies on the protective effect of botanical products against pest insects have infrequently been extended to side-effects on natural enemies. Indirect effects of botanicals on the storability of seeds could occur through their possible negative impact on biological control agents. Four plant powders and six plant oils with a known effect on the cowpea beetle Callosobruchus maculatus (Fabr.) (Coleoptera: Bruchidae) were investigated for their effect on the beetles' egg parasitoid Uscana lariophaga (Steffan) (Hymenoptera: Trichogrammatidae) and the larval parasitoid Dinarmus basalis (Rondani) (Hymenoptera: Pteromalidae). All treatments caused a decrease in parasitisation by U. lariophaga, and developing parasitoids of this species were affected by powders of Nicotiana tabacum (L.) and Tephrosia vogelii (Hook. f). In a two-choice situation using a linear olfactometer, U. lariophaga was repelled by most of the oils. In a no-choice situation, parasitisation by D. basalis was hampered by treatment with plant powders, but eggs that were laid developed normally. In a Y-tube olfactometer, this parasitoid did not discriminate between odours of untreated and plant-powder-treated beans. The powders of N. tabacum and T. vogelii had stronger negative effects on the two parasitoids than the powders of Azadirachta indica (Juss.) or Blumea aurita (DC). In samples collected from untreated traditional storage facilities, subsequently treated with plant powders in the laboratory, none of the treatments could prevent the increase in beetle numbers. At 24 days after treatment, most beetles had emerged from beans treated with powders of N. tabacum and T. vogelii. Parasitoids were affected by the botanical insecticides tested here, but the powders of A. indica and B. aurita may be compatible with biological control by D. basalis.

To determine the influence of pesticide treatments on the population dynamics of the whiteflies Trialeurodes vaporariorum (Westwood) and Bemisia tabaci Gennadius (Homoptera: Aleyrodidae) and their naturally occurring parasitoids, we performed field experiments on insecticide sprayed and unsprayed fields during a cropping season of snap bean in Pradera, Valle del Cauca, Colombia. Substantially larger populations of whitefly nymphs occurred in the unsprayed field than in the sprayed field. Parasitoids were more frequent in unsprayed than in sprayed fields with Encarsia nigricephala Dozier being more prevalent than Amitus fuscipennis MacGown & Nebeker. In sprayed fields the nymphs parasitized by E. nigricephala exceeded the unparasitized whitefly nymphs at the end of the cropping season. Our results suggest that while under current agricultural practices whiteflies on snap beans cannot be exclusively controlled by naturally occurring parasitoids, parasitoids may be integrated with chemical control in order to reduce crop damage.

Epidemics of onion leaf spot caused by Botrytis spp. depend on necrotic leaf tissue for inoculum build up in the crop. Ulocladium atrum Preuss. is a strong competitor on necrotic above-ground plant tissues. The potential of the antagonist to reduce colonisation of necrotic leaf tissue by Botrytis spp. and subsequent sporulation was studied in two field experiments. U. atrum colonised necrotic tissues and consistently reduced the sporulation of fungal competitors. Although incidence of Botrytis spp. was low, significantly lower spore loads of Botrytis spp. were found on spore traps located within U. atrum treated onion plots as compared to untreated control plots. The number of leaf spots caused by Botrytis spp. was low in both field experiments but was significantly reduced by 60 fter U. atrum applications in the second experiment. Spraying of the fungicide Ronilan resulted in the same control level. Results show that U. atrum has a potential for biological control against diseases caused by Botrytis spp. in onions.

In a previous paper, we proposed a fungal growth model (Lamour et al., 2001 IMA J. Math. Appl. Med. Biol., 17, 329-346), describing the colonization and decomposition of substrate, subsequent uptake of nutrients, and incorporation into fungal biomass, and performed an overall-steady-state analysis. In this paper we assume that where nutrient dynamics are much faster than the dynamics of fungal biomass and substrate, the system will reach a quasi-steady-state relatively quickly. We show how the quasi-steady-state approximation is a simplification of the full fungal growth model. We then derive an explicit fungal invasion criterion, which was not possible for the full model, and characterize parameter domains for invasion and extinction. Importantly, the fungal invasion criterion takes two forms: one for systems where carbon is limiting, another for systems where nitrogen is limiting. We focus attention on what happens in the short term immediately following the introduction of a fungus to a fungal-free system by analysing the stability of the trivial steady state, and then check numerically whether the fungus is able to persist. The derived invasion criterion was found to be valid also for the full model. Knowledge of the factors that determine invasion is essential to an understanding of fungal dynamics. The simplified model allows the invasion criterion to be tested with experimental data.

East Africa's Lake Victoria provides resources and services to millions of people on the lake's shores and abroad. In particular, the lake's fisheries are an important source of protein, employment, and international economic connections for the whole region. Nonetheless, stock dynamics are poorly understood and currently unpredictable. Furthermore, fishery dynamics are intricately connected to other supporting services of the lake as well as to lakeshore societies and economies. Much research has been carried out piecemeal on different aspects of Lake Victoria's system; e.g., societies, biodiversity, fisheries, and eutrophication. However, to disentangle drivers and dynamics of change in this complex system, we need to put these pieces together and analyze the system as a whole. We did so by first building a qualitative model of the lake's social-ecological system. We then investigated the model system through a qualitative loop analysis, and finally examined effects of changes on the system state and structure. The model and its contextual analysis allowed us to investigate system-wide chain reactions resulting from disturbances. Importantly, we built a tool that can be used to analyze the cascading effects of management options and establish the requirements for their success. We found that high connectedness of the system at the exploitation level, through fisheries having multiple target stocks, can increase the stocks' vulnerability to exploitation but reduce society's vulnerability to variability in individual stocks. We describe how there are multiple pathways to any change in the system, which makes it difficult to identify the root cause of changes but also broadens the management toolkit. Also, we illustrate how nutrient enrichment is not a self-regulating process, and that explicit management is necessary to halt or reverse eutrophication. This model is simple and usable to assess system-wide effects of management policies, and can serve as a paving stone for future quantitative analyses of system dynamics at local scales.

AimInvasion biologists have made an extensive exploration of ways to identify the characteristic traits of invasive plants based on the assumption that plant attributes remain similar when plants become invasive. However, this assumption needs to be critically evaluated when predicting successful future introductions.LocationGlobal.MethodsUsing a global database of plant functional traits (i.e. specific leaf area, maximum canopy height and individual seed mass) encompassing 129 different species three questions were evaluated using a meta-analytical approach. (1) Do traits of introduced aliens change between native and introduced areas? (2) Do the responses show directionality, indicating that traits of aliens are either consistently higher or lower in their introduced range? (3) Are there smaller differences in species between native and introduced areas (within-species multitrait variation) than between two random species (between-species multitrait variability)?ResultsMean trait differences (measured as log-response ratios) between native and introduced (invasive+naturalized), native and naturalized, or native and invasive areas showed no significant differences across evaluated species, even after controlling for invasion status and growth form. This pattern was the result of aliens showing both higher (Alien(area)>Native(area)) and lower (Alien(area)Main conclusionsThe similarity of evaluated traits between areas, in combination with a smaller within-species than among-species trait variability, are fundamental results for conservation and nature management efforts. The results presented in this study validate the assumption that mean trait measurements in the native areas are likely to be reasonably representative of trait mean values in the invaded areas, and support the use of trait-based prediction methods to evaluate the potential of an introduced plant to become established.

BACKGROUND Polygodial, a sesquiterpene dialdehyde of the drimane family, has been shown to have deterrent and antifeedant effects on various insect species, including Myzus persicae (Sulzer), Spodoptera spp. and Leptinotarsa decemlineata (Say). This compound may have potential as a broad-spectrum biocontrol agent, similar to pyrethrins, given that it was previously reported to improve yield when sprayed on barley fields. RESULTS This study compares the deterrent effect of polygodial and pyrethrins against the silverleaf whitefly Bemisia tabaci (Gennadius) and the green peach aphid M. persicae in dual-choice assays using compound-coated tomato leaf discs. B. tabaci adults were deterred by polygodial at an ED50 (effective dose at which 50% of the insects are deterred) of about 25 µg g-1 fresh weight (FW), and green peach aphids at about 54 µg g-1 FW. Bioassays were benchmarked with pyrethrins that had a 20-fold lower ED50 of approximately 1.4 µg g-1 FW against whiteflies, but only a twofold lower ED50 (about 28 µg g-1 FW) against peach aphids. Polygodial showed moderate phytotoxic effects (score of 2 on a scale of 1–5) on tomato leaves at concentrations above the ED50 concentrations (=90 µg g-1 FW). CONCLUSION The sesquiterpene dialdehyde polygodial is 2–20 times less deterrent than pyrethrins, depending on the insect species, but it could provide a useful complement to pyrethrin sprays as it has a different mode of action, is food grade and has low volatility. However, a formulation that reduces the risks of phytotoxic effects should be developed. © 2013 Society of Chemical Industry

Understanding and disentangling different processes underlying the assembly and diversity of communities remains a key challenge in ecology. Species can assemble into communities either randomly or due to deterministic processes. Deterministic assembly leads to species being more similar (underdispersed) or more different (overdispersed) in certain traits than would be expected by chance. However, the relative importance of those processes is not well understood for many organisms, including terrestrial invertebrates. Based on knowledge of a broad range of species traits, we tested for the presence of trait underdispersion (indicating dispersal or environmental filtering) and trait overdispersion (indicating niche partitioning) and their relative importance in explaining land snail community composition on lake islands. The analysis of community assembly was performed using a functional diversity index (Rao's quadratic entropy) in combination with a null model approach. Regression analysis with the effect sizes of the assembly tests and environmental variables gave information on the strength of under- and overdispersion along environmental gradients. Additionally, we examined the link between community weighted mean trait values and environmental variables using a CWM-RDA. We found both trait underdispersion and trait overdispersion, but underdispersion (eight traits) was more frequently detected than overdispersion (two traits). Underdispersion was related to four environmental variables (tree cover, habitat diversity, productivity of ground vegetation, and location on an esker ridge). Our results show clear evidence for underdispersion in traits driven by environmental filtering, but no clear evidence for dispersal filtering. We did not find evidence for overdispersion of traits due to diet or body size, but overdispersion in shell shape may indicate niche differentiation between snail species driven by small-scale habitat heterogeneity. The use of species traits enabled us to identify key traits involved in snail community assembly and to detect the simultaneous occurrence of trait underdispersion and overdispersion. © 2014 The Authors. Habitat bioaugmentation and introduction of protective microbiota have been proposed as potential conservation strategies to rescue endangered mammals and amphibians from emerging diseases. For both strategies, insight into the microbiomes of the endangered species and their habitats is essential. Here, we sampled nests of the endangered sea turtle species Eretmochelys imbricata that were infected with the fungal pathogen Fusarium falciforme. Metagenomic analysis of the bacterial communities associated with the shells of the sea turtle eggs revealed approximately 16,664 operational taxonomic units, with Proteobacteria, Actinobacteria, Firmicutes and Bacteroidetes as the most dominant phyla. Subsequent isolation of Actinobacteria from the eggshells led to the identification of several genera ( Streptomyces, Amycolaptosis, Micromomospora Plantactinospora and Solwaraspora) that inhibit hyphal growth of the pathogen F. falciforme. These bacterial genera constitute a first set of microbial indicators to evaluate the potential role of microbiota in conservation of endangered sea turtle species.

Agricultural intensification may result in important shifts in insect community composition and function, but this remains poorly explored. Studying how groups of species with shared traits respond to local and landscape scale land-use management can reveal mechanisms behind such observed impacts. We tested if ground beetles (Coleoptera: Carabidae) divided into trait groups based on body sizes, wing morphologies and dietary preferences respond differently to farming practise (organic and conventional), farming intensity (measured as yield) and landscape complexity (measured as the proportion of arable land within a 1,000 m radius) across Europe. We used data from 143 farms in five regions in northern and central Europe. Organic farms did not differ in abundance or richness of any trait group compared to conventional farms. As farm scale intensity (yield) increased, overall abundance of beetles decreased, but abundances of small and medium sized beetles, as well as that of wingless beetles, were unaffected. Overall species richness was not affected by yield, whereas consideration of traits revealed that phytophagous and omnivorous beetles were less species rich on farms with high yields. Increasing the proportion of arable land in the landscape increased overall beetle abundance. This was driven by an increase in omnivorous beetles. The total species richness was not affected by an increase in the proportion arable land, although the richness of wingless beetles was found to increase. Potential effects on ecosystem functioning need to be taken into account when designing schemes to maintain agricultural biodiversity, because species with different ecological traits respond differently to local management and landscape changes.

The combined release of species of generalist predators can enhance multiple pest control when the predators feed on different prey, but, in theory, predators may be excluded through predation on each other. This study evaluated the co-occurrence of the generalist predators Macrolophus pygmaeus Rambur and Orius laevigatus (Fieber) and their control of two pests in a sweet pepper crop. Both predators consume pollen and nectar in sweet pepper flowers, prey on thrips and aphids, and O. laevigatus is an intraguild predator of M. pygmaeus. Observations in a commercial sweet pepper crop in a greenhouse with low densities of pests showed that the two predator species coexisted for 8 months. Moreover, their distributions in flowers suggested that they were neither attracted to each other, nor avoided or excluded each other. A greenhouse experiment showed that the predators together clearly controlled thrips and aphids better than each of them separately. Thrips control was significantly better in the presence of O. laevigatus and aphid control was significantly better in the presence of M. pygmaeus. Hence, combined inoculative releases of M. pygmaeus and O. laevigatus seem to be a good solution for controlling both thrips and aphids in greenhouse-grown sweet pepper. The predators are able to persist in one crop for a sufficiently long period and they complement each other in the control of both pests. This study also provides further evidence that intraguild predation does not necessarily have negative effects on biological control.

Bacteria are believed to play an important role in the fitness and biochemistry of sponges (Porifera). Pseudomonas species (Gammaproteobacteria, Pseudomonadales) are capable of colonizing a broad range of eukaryotic hosts, but knowledge of their diversity and function in freshwater invertebrates is rudimentary. We assessed the diversity, structure and antimicrobial activities of Pseudomonas spp. in the freshwater sponge Ephydatia fluviatilis. Polymerase Chain Reaction - Denaturing Gradient Gel Electrophoresis (PCR-DGGE) fingerprints of the global regulator gene gacA revealed distinct structures between sponge-associated and free-living Pseudomonas communities, unveiling previously unsuspected diversity of these assemblages in freshwater. Community structures varied across E. fluviatilis specimens, yet specific gacA phylotypes could be detected by PCR-DGGE in almost all sponge individuals sampled over two consecutive years. By means of whole-genome fingerprinting, 39 distinct genotypes were found within 90 fluorescent Pseudomonas isolates retrieved from E. fluviatilis. High frequency of in vitro antibacterial (49%), antiprotozoan (35%) and anti-oomycetal (32%) activities was found among these isolates, contrasting less-pronounced basidiomycetal (17%) and ascomycetal (8%) antagonism. Culture extracts of highly predation-resistant isolates rapidly caused complete immobility or lysis of cells of the protozoan Colpoda steinii. Isolates tentatively identified as P. jessenii, P. protegens and P. oryzihabitans showed conspicuous inhibitory traits and correspondence with dominant sponge-associated phylotypes registered by cultivation-independent analysis. Our findings suggest that E. fluviatilis hosts both transient and persistent Pseudomonas symbionts displaying antimicrobial activities of potential ecological and biotechnological value. While the area of organic crop production increases at a global scale, the potential interactions between pest management in organic and conventionally managed systems have so far received little attention. Here, we evaluate the landscape-level co-dependence of insecticide-based and natural enemy-based pest management using a simulation model for parasitoid–host interactions in landscapes consisting of conventionally and organically managed fields. In our simulations conventional management consists of broad-spectrum or selective insecticide application, while organic management involves no insecticides. Simulations indicate that insecticide use can easily result in lose–lose scenarios whereby both organically and conventionally managed fields suffer from increased pest loads as compared to a scenario where no insecticides are used, but that under some conditions insecticide use can be compatible with biocontrol. Simulations also suggest that the pathway to achieve the insecticide reduction without triggering additional pest pressure is not straightforward, because increasing the proportion of organically managed fields or reducing the spray frequency in conventional fields can potentially give rise to dramatic increases in pest load. The disruptive effect of insecticide use, however, can be mitigated by spatially clustering organic fields and using selective insecticides, although the effectiveness of this mitigation depends on the behavioral traits of the biocontrol agents. Poorly dispersing parasitoids and parasitoids with high attack rates required a lower amount of organically managed fields for effective pest suppression. Our findings show that the transition from a landscape dominated by conventionally managed crops to organic management has potential pitfalls; intermediate levels of organic management may lead to higher pest burdens than either low or high adoption of organic management.

International audience; Several parasitoids of the genus Psyttalia have been repeatedly introduced as biological control agents against the principal pest of olive, the fly Bactrocera oleae. However, few of the parasitoids released have become established and proved effective against B. oleae. It may however still be possible to find effective biological control agents adapted to local environmental conditions among the highly diverse Psyttalia species and populations infesting B. oleae worldwide. For this purpose, we have developed a rapid, sensitive molecular method based on the polymerase chain reaction (PCR) for estimating and comparing the parasitism success of Psyttalia parasitoids through the detection of eggs and larvae within the host. This method was tested and shown to be appropriate for two Psyttalia species (Psyttalia concolor and Psyttalia lounsburyi). The possible detection of DNA was also demonstrated for several populations of these species and for other Psyttalia species, namely Psyttalia humilis and Psyttalia ponerophaga. For P. concolor and P. lounsburyi, a strong correlation was observed between the parasitism rates estimated by PCR, host larva dissection and counts of emerging parasitoids. No significant difference was found between the rates of parasitism estimated by host larva dissection and PCR, whereas the rates of parasitism estimated by PCR were significantly higher than those estimated from emergence, suggesting occurrence of mortality during the parasitoid development. This PCR method is thus highly reliable and provides an objective criterion for estimating the efficacy of biological control agent candidates from diverse taxa and populations of Psyttalia. ⇑ Corresponding author.

In this study, the efficacy of bacterial antagonists was tested in both pre- and post-harvest stages against Botrytis cinerea, which causes one of the major diseases of strawberries. In total, 219 bacterial antagonists were obtained from various parts of strawberry plants, and their efficacies against B. cinerea were determined. Two isolated bacteria, which were identified as Bacillus megaterium and Pseudomonas vesicularis, were found to be effective and otherwise suitable as biological control agents. Moreover, an isolate of Pseudomonas fluorescens from pea plants was successfully used against B. cinerea in the experiments. These three antagonist isolates were field- tested in both the pre- and post-harvest periods in two consecutive years. The field studies demonstrated that all three isolates were as effective as a synthetic fungicide. In the first year field tests, the isolates reduced the decay incidence to 41.08–43.03 % compared with 55.48 % in the control. Similar results were obtained in the second year. In the post-harvest stage, all the bacterial isolates also controlled the disease effectively. The bacterial population was monitored in both the pre- and post-harvest periods using a spontaneous antibiotic-resistant mutant and the RAPD-PCR method. The RAPD-PCR technique alone proved insufficient to determine the population levels of the bacterial antagonists.

Since typically there are many predators feeding on most herbivores in natural communities, understanding multiple predator effects is critical for both community and applied ecology. Experiments of multiple predator effects on prey populations are extremely demanding, as the number of treatments and the amount of labour associated with these experiments increases exponentially with the number of species in question. Therefore, researchers tend to vary only presence/absence of the species and use only one (supposedly realistic) combination of their numbers in experiments. However, nonlinearities in density dependence, functional responses, interactions between natural enemies etc. are typical for such systems, and nonlinear models of population dynamics generally predict qualitatively different results, if initial absolute densities of the species studied differ, even if their relative densities are maintained. Therefore, testing combinations of natural enemies without varying their densities may not be sufficient. Here we test this prediction experimentally. We show that the population dynamics of a system consisting of 2 natural enemies (aphid predator Adalia bipunctata (L.), and aphid parasitoid, Aphidius colemani Viereck) and their shared prey (peach aphid, Myzus persicae Sulzer) are strongly affected by the absolute initial densities of the species in question. Even if their relative densities are kept constant, the natural enemy species or combination thereof that most effectively suppresses the prey may depend on the absolute initial densities used in the experiment. Future empirical studies of multiple predator – one prey interactions should therefore use a two-dimensional array of initial densities of the studied species. Varying only combinations of natural enemies without varying their densities is not sufficient and can lead to misleading results.

Orius insidiosus is a generalist predator for which diet influences important biological traits like reproduction and predation. We tested the effects of different diets alone or in combination on reproduction, longevity and predation capacity of this predator. The diets tested were: no food (control); pollen; Frankliniella occidentalis (Pergande) nymphs and adults; F. occidentalis nymphs and adults and pollen; Anagasta kuehniella (Zeller) eggs; A. kuehniella eggs and pollen; A. kuehniella eggs; F. occidentalis nymphs and adults and A. kuehniella eggs, F. occidentalis nymphs and adults and pollen. The pre-oviposition period was shortest when the diet consisted of pollen and prey (A. kuehniella and/or F. occidentalis), of the two prey species together or of only A. kuehniella eggs. The highest values for the length of the oviposition period (50.1, 48.0, 46.3 and 46.1 days), daily fecundity (3.8, 3.9, 4.0 and 4.2 eggs/female/day), total fecundity (190.3, 187.7 185.2 and 193.6 eggs/female) and longevity (52.1, 49.9, 48.7 and 48.0 days) were found with diets consisting of only A. kuehniella eggs. Pollen did not improve any of the performance parameters of the predator when offered exclusively or as a complementary food. The results show that selection of a proper diet can strongly improve reproduction, longevity and predation capacity of O. insidiosus. An important finding is that A. kuehniella eggs, which can easily be produced in large quantities, are an excellent factitious prey for mass production of O. insidiosus.

International audience; Several bacterial strains of the Pseudomonas genus provide plant growth stimulation, plant protection against pests or bioremediation. Among these bacteria, P.fluorescensPf29Arp reduces the severity of take-all, a disease caused by the pathogenic fungus Gaeumannomyces graminis var. tritici (Ggt) on wheat roots. In this study, we obtained a draft genome of Pf29Arp and subsequent comparative genomic analyses have revealed that this bacterial strain is closely related to strains of the P.brassicacearum-like' subgroup including P.brassicacearum ssp. brassicacearumNFM421 and P.fluorescensF113. Despite an overall chromosomal organization similar to these strains, a number of features including antibiotic synthesis gene clusters from secondary metabolism are not found in the Pf29Arp genome. But Pf29Arp possesses different protein secretion systems including type III (T3SS) and type VI (T6SS) secretion systems. Pf29Arp is the first Pseudomonas sp. strain described with four T6SS clusters (cluster I, II, III and IV). In addition, some protein-coding genes involved in the assembly of these secretion systems are basally expressed during Pf29Arp colonization of healthy wheat roots and display different expression patterns on necrotized roots caused by Ggt. These data suggest a role of T3SS and T6SS in the Pf29Arp adaptation to different root environments.

Immature development of parasitoid wasps is restricted to resources found in a single host that is often similar in size to the adult parasitoid. When two or more parasitoids of the same or different species attack the same host, there is competition for monopolization of host resources. The success of intrinsic competition differs between parasitoids attacking growing hosts and parasitoids attacking paralyzed hosts. Furthermore, the evolution of gregarious development in parasitoids reflects differences in various developmental and behavioral traits, as these influence antagonistic encounters among immature parasitoids. Fitness-related costs (or benefits) of competition for the winning parasitoid reveal that time lags between successive attacks influence the outcome of competition. Physiological mechanisms used to exclude competitors include physical and biochemical factors that originate with the ovipositing female wasp or her progeny. In a broader multitrophic framework, indirect factors, such as plant quality, may affect parasitoids through effects on immunity and nutrition.

International audience; Pest control that results from the activity of naturally occurring parasitoids is an important service that could help reduce pesticide use. We analyzed parasitism in codling moth diapausing larvae from a total of 122 apple orchards in southeastern France during five consecutive years (2006-2010) in relation to the agronomic and land cover characteristics at both the local and landscape levels. Three species of hymenoptera parasitoids were observed, including two primary (Braconidae and Ichneumonidae) and one hyperparasitoid (Perilampidae) wasps. Parasitoid community compositions differed according to the year (in term of the relative abundance of the primary parasitoid species) and the presence of windbreaks and spontaneous hedgerows around the orchards (in terms of the relative abundance of primary vs. hyperparasitoid species). Invasive soilborne plant pathogens cause substantial damage to crops and natural populations, but our understanding of how to prevent their epidemics or reduce their damage is limited. A key and experimentally-tested concept in the epidemiology of soilborne plant diseases is that of a threshold spacing between hosts below which epidemics (invasive spread) can occur. We extend this paradigm by examining how plant-root growth may alter the conditions for occurrence of soilborne pathogen epidemics in plant populations. We hypothesise that host-root growth can 1) increase the probability of pathogen transmission between neighbouring plants and, consequently, 2) decrease the threshold spacing for epidemics to occur. We predict that, in systems initially below their threshold conditions, root growth can trigger soilborne pathogen epidemics through a switch from non-invasive to invasive behaviour, while in systems above threshold conditions root growth can enhance epidemic development. As an example pathosystem, we studied the fungus Rhizoctonia solani on sugar beet in field experiments. To address hypothesis 1, we recorded infections within inoculum-donor and host-recipient pairs of plants with differing spacing. We translated these observations into the individual-level concept of pathozone, a host-centred form of dispersal kernel. To test hypothesis 2 and our prediction, we used the pathozone to parameterise a stochastic model of pathogen spread in a host population, contrasting scenarios of spread with and without host growth. Our results support our hypotheses and prediction. We suggest that practitioners of agriculture and arboriculture account for root system expansion in order to reduce the risk of soilborne-disease epidemics. We discuss changes in crop design, including increasing plant spacing and using crop mixtures, for boosting crop resilience to invasion and damage by soilborne pathogens. We speculate that the disease-induced root growth observed in some pathosystems could be a pathogen strategy to increase its population through host manipulation. © 2013 Leclerc et al.

During seed production, Brassica seed may become infected with Xanthomonas campestris pv. campestris after systemic colonization of plants upon leaf infection, or alternatively, after flower infection. Polytunnel experiments were conducted in 2007 and 2008 to study the relative importance of these colonization routes resulting in seed infection. Cauliflower plants (Brassica oleracea) were spray-inoculated at the 8-leaf stage, after formation of cauliflowers or during flowering, at which stage leaves or blossoms were inoculated. Inoculation at all stages resulted in a relatively high percentage of systemic infection; the average estimated infection incidences for stem base and peduncle infections were 16 % and 19 %, respectively. When seed samples were examined by dilution plating for deep-seated infection following hot water treatment, Xcc was detected in 61 % of the 23 seed samples harvested from plants with inoculated flowers. However, symptom development in seedlings raised from the seeds could not be confirmed in a grow-out test under favourable conditions for Xcc infection at a high RH (>95 %) and a relative high temperature (28 °C). Xcc was not detected in 59 seed samples harvested from leaf-inoculated plants with the exception of one sample from plants inoculated at peduncle formation. In a third polytunnel experiment carried out in 2009, the population dynamics of Xcc on inoculated flowers was investigated. Following spray-inoculation of flowers, 52 % of the flowers were infected with Xcc. During development of siliques, infection incidence decreased slowly and at 56 dpi, 20 % of the superficially disinfected siliques were infected with Xcc. It was estimated that 0.18 % of the seeds was infected and that 1–10 % of the infected siliques contained infected seeds. The implications of these results for control of Xcc in a seed production crop are discussed.

International audience; Resource enrichment can potentially destabilize predator-prey dynamics. This phenomenon historically referred as the "paradox of enrichment" has mostly been explored in spatially homogenous environments. However, many predator-prey communities exchange organisms within spatially heterogeneous networks called metacommunities. This heterogeneity can result from uneven distribution of resources among communities and thus can lead to the spreading of local enrichment within metacommunities. Here, we adapted the original Rosenzweig-MacArthur predator-prey model, built to study the paradox of enrichment, to investigate the effect of regional enrichment and of its spatial distribution on predator-prey dynamics in metacommunities. We found that the potential for destabilization was depending on the connectivity among communities and the spatial distribution of enrichment. In one hand, we found that at low dispersal regional enrichment led to the destabilization of predator-prey dynamics. This destabilizing effect was more pronounced when the enrichment was uneven among communities. In the other hand, we found that high dispersal could stabilize the predator-prey dynamics when the enrichment was spatially heterogeneous. Our results illustrate that the destabilizing effect of enrichment can be dampened when the spatial scale of resource enrichment is lower than that of organismss movements (heterogeneous enrichment). From a conservation perspective, our results illustrate that spatial heterogeneity could decrease the regional extinction risk of species involved in specialized trophic interactions. From the perspective of biological control, our results show that the heterogeneous distribution of pest resource could favor or dampen outbreaks of pests and of their natural enemies, depending on the spatial scale of heterogeneity.

Climatic niche models based on native-range climatic data accurately predict invasive-range distributions in the majority of species. However, these models often do not account for ecological and evolutionary processes, which limit the ability to predict future range expansion. This might be particularly problematic in the case of invaders that occupy environments that would be considered marginal relative to the climatic niche in the native range of the species. Here, we assess the potential for future range expansion in the shrub Chromolaena odorata that is currently invading mesic savannas (>650 mm MAP) in South Africa that are colder and drier than most habitats in its native range. In a greenhouse experiment we tested whether its current distribution in South Africa can be explained by increased competitive ability and/or differentiation in drought tolerance relative to the native population. We compared aboveground biomass, biomass allocation, water use efficiency and relative yields of native and invasive C. odorata and the resident grass Panicum maximum in wet and dry conditions. Surprisingly, we found little differentiation between ranges. Invasive C. odorata showed no increased competitive ability or superior drought tolerance compared to native C. odorata. Moreover we found that P. maximum was a better competitor than either native or invasive C. odorata. These results imply that C. odorata is unlikely to expand its future range towards more extreme, drier, habitats beyond the limits of its current climatic niche and that the species' invasiveness most likely depends on superior light interception when temporarily released from competition by disturbance. Our study highlights the fact that species can successfully invade habitats that are at the extreme end of their ranges and thereby contributes towards a better understanding of range expansion during species invasions.

We conducted laboratory tests to evaluate the suitability of extrafloral nectar (EFN) from Vicia faba as a potential food source for female Diaeretiella rapae in an intercropping context, and how EFN affects the survivorship and progeny production in this species. Extrafloral and floral nectar have a similar sugar composition, a highly nutritious food for parasitoids. Under controlled conditions, intercropping V. faba with Brassica oleracea had no impact on V. faba EFN sugar composition. Female parasitoids lived significantly longer when provided with EFN (14 days) than when provided with water (4 days), honeydew (4 days), and honey and water (9 days) or when they were without food (3 days). The longevity of females provided with EFN did not differ in the absence or presence of aphids. The numbers of hosts parasitised and offspring produced per female over the course of her lifespan were significantly increased by placing a V. faba plant next to a B. oleracea plant. However, this increase was observed for both nectary-bearing and nectary-less V. faba plants. Feeding on EFN resulted in an increase in the length of the oviposition period, which was primarily concentrated during the first four days of the life of the female. No impacts of V. faba EFN provision were observed on offspring larval survival, sex ratio or size. A long-term experiment on the effect of chitin addition to soil on the suppression of soil-borne pathogens was set up and monitored for eight years in an experimental field, Vredepeel, the Netherlands. Chitinous matter obtained from shrimps was added to soil top layers at two different occasions and the suppressiveness of soil towards Verticillium dahliae as well as plant-pathogenic nematodes was assessed, in addition to analyses of the abundances and community structures of members of the soil microbiota. The data revealed that chitin amendment had raised the suppressiveness of soil, in particular towards Verticillium dahliae, 9 months after the (second) treatment, extending to two years following treatment. Moreover, major effects of the added chitin on the soil microbial communities were detected. First, shifts in both the abundances and structures of the chitin-treated soil microbial communities were found, both of total soil bacteria and fungi. In addition, the abundances and structures of soil actinobacteria and Oxalobacteriaceae were affected by chitin. At the functional gene level, the abundance of specific (family-18 glycoside hydrolase) chitinase genes carried by the soil bacteriota also revealed upshifts as a result of the added chitin. The effects of chitin noted for the Oxalobacteriaceae were specifically related to significant upshifts in the abundances of the species Duganella violaceinigra and Massilia plicata. These effects of chitin persisted over the time of the experiment.

BACKGROUND: Most insect-resistant transgenic crops employ toxins to control pests. A novel approach is to enhance the effectiveness of natural enemies by genetic engineering of the biosynthesis of volatile organic compounds (VOCs). Before the commercialisation of such transgenic plants can be pursued, detailed fundamental studies of their effects on herbivores and their natural enemies are necessary. The linalool/nerolidol synthase gene FaNES1 was constitutively expressed from strawberry in three Arabidopsis thaliana accessions, and the behaviour of the aphid Brevicoryne brassicae L., the parasitoid Diaeretiella rapae McIntosh and the predator Episyrphus balteatus de Geer was studied. RESULTS: Transgenic FaNES1-expressing plants emitted (E)-nerolidol and larger amounts of (E)-DMNT and linalool. Brevicoryne brassicae was repelled by the transgenic lines of two of the accessions, whereas its performance was not affected. Diaeretiella rapae preferred aphid-infested transgenic plants over aphid-infested wild-type plants for two of the accessions. In contrast, female E. balteatus predators did not differentiate between aphid-infested transgenic or wild-type plants. CONCLUSION: The results indicate that the genetic engineering of plants to modify their emission of VOCs holds considerable promise for facilitating biological control of herbivores. Validation for crop plants is a necessary next step to assess the usefulness of modified volatile emission in integrated pest management.

In the light of ongoing land use changes, it is important to understand how multitrophic communities perform at different land use intensities. The paradox of enrichment predicts that fertilization leads to destabilization and extinction of predator-prey systems. We tested this prediction for a land use intensity gradient from natural to highly fertilized agricultural ecosystems. We included multiple aboveground and belowground trophic levels and land use-dependent searching efficiencies of insects. To overcome logistic constraints of field experiments, we used a successfully validated simulation model to investigate plant responses to removal of herbivores and their enemies. Consistent with our predictions, instability measured by herbivore-induced plant mortality increased with increasing land use intensity. Simultaneously, the balance between herbivores and natural enemies turned increasingly towards herbivore dominance and natural enemy failure. Under natural conditions, there were more frequently significant effects of belowground herbivores and their natural enemies on plant performance, whereas there were more aboveground effects in agroecosystems. This result was partly due to the "boom-bust'' behavior of the shoot herbivore population. Plant responses to herbivore or natural enemy removal were much more abrupt than the imposed smooth land use intensity gradient. This may be due to the presence of multiple trophic levels aboveground and belowground. Our model suggests that destabilization and extinction are more likely to occur in agroecosystems than in natural communities, but the shape of the relationship is nonlinear under the influence of multiple trophic interactions. The biocontrol effect of Clonostachys rosea (strains 016 and 1457) on Fusarium graminearum, F. avenaceum, F. verticillioides, F. langsethiae, F. poae, F. sporotrichioides, F. culmorum and Microdochium nivale was evaluated on naturally infected wheat stalks exposed to field conditions for 180 days. Experiments were conducted at two locations in Argentina, Marcos Juarez and Rı´o Cuarto. Antagonists were applied as conidial suspensions at two inoculum levels. Pathogens were quantified by TaqMan real-time qPCR. During the first year at Marcos Juarez, biocontrol was observed in one antagonist treatment for F. graminearum after 90 days (73% reduction) but after 180 days, the pathogen decreased to undetectable levels. During the second year, biocontrol was observed in three antagonist treatments for F. graminearum and F. avenaceum (68 3% and 98 9% DNA reduction, respectively, after 90 days). Fusarium verticillioides was not controlled at Marcos Juarez. At Rı´o Cuarto, biocontrol effects were observed in several treatments at different intervals, with a mean DNA reduction of 88 7% for F. graminearum and F. avenaceum, and 100% reduction for F. verticillioides in two treatments after 180 days. Populations of F. avenaceum and F. verticillioides were stable; meanwhile, F. graminearum population levels varied during the first 90 days, and low levels were observed after 180 days. The other pathogens were not detected. The study showed that wheat stalks were important reservoirs for F. avenaceum and F. verticillioides populations but less favourable for F. graminearum survival. International audience; Molecular methods are increasingly used to detect and identify parasites in their hosts. However, existing methods are generally not appropriate for studying complex host-parasite interactions because they require prior knowledge of species composition. DNA barcoding is a molecular method that allows identifying species using DNA sequences as an identification key. We used DNA amplification with primers common to aphid parasitoids and sequencing of the amplified fragment to detect and identify parasitoids in their hosts, without prior knowledge on the species potentially present. To implement this approach, we developed a method based on 16S rRNA mitochondrial gene and LWRh nuclear gene. First, we designed two primer pairs specific to Aphidiinae (Hymenoptera), the main group of aphid parasitoids. Second, we tested whether the amplified regions could correctly identify Aphidiinae species and found that 61 species were accurately identified of 75 tested. We then determined the ability of each primer pair to detect immature parasitoids inside their aphid host. Detection was earlier for 16S than for LWRh, with parasitoids detected, respectively, 24 and 48 h after egg injection. Finally, we applied this method to assess parasitism rate in field populations of several aphid species. The interest of this tool for analysing aphid-parasitoid food webs is discussed.

International audience; Over the past 16 years, vast plantings of transgenic crops producing insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) have helped to control several major insect pests(1-5) and reduce the need for insecticide sprays(1,5,6). Because broad-spectrum insecticides kill arthropod natural enemies that provide biological control of pests, the decrease in use of insecticide sprays associated with Bt crops could enhance biocontrol services(7-12). However, this hypothesis has not been tested in terms of long-term landscape-level impacts(10). On the basis of data from 1990 to 2010 at 36 sites in six provinces of northern China, we show here a marked increase in abundance of three types of generalist arthropod predators (ladybirds, lacewings and spiders) and a decreased abundance of aphid pests associated with widespread adoption of Bt cotton and reduced insecticide sprays in this crop. We also found evidence that the predators might provide additional biocontrol services spilling over from Bt cotton fields onto neighbouring crops (maize, peanut and soybean). Our work extends results from general studies evaluating ecological effects of Bt crops(1-4,6,12,13) by demonstrating that such crops can promote biocontrol services in agricultural landscapes.

Aims: To determine the role of phenazines (PHZ) and lipopeptide surfactants (LPs) produced by Pseudomonas in suppression of stem rot disease of groundnut, caused by the fungal pathogen Sclerotium rolfsii. Methods and Results: In vitro assays showed that PHZ-producing Pseudomonas chlororaphis strain Phz24 significantly inhibited hyphal growth of S. rolfsii and suppressed stem rot disease of groundnut under field conditions. Biosynthesis and regulatory mutants of Phz24 deficient in PHZ production were less effective in pathogen suppression. Pseudomonas strains SS101, SBW25, and 267, producing viscosin or putisolvin-like LPs, only marginally inhibited hyphal growth of S. rolfsii and did not suppress stem rot disease. In contrast, Pseudomonas strain SH-C52, producing the chlorinated LP thanamycin, inhibited hyphal growth of S. rolfsii and significantly reduced stem rot disease of groundnut in nethouse and field experiments, whereas its thanamycin-deficient mutant was less effective. Conclusions: Phenazines and specific lipopeptides play an important role in suppression of stem rot disease of groundnut by root-colonizing Pseudomonas strains. Significance and Impact of the Study: Pseudomonas strains Phz24 and SH-C52 showed significant control of stem rot disease. Treatment of seeds or soil with these strains provides a promising supplementary strategy to control stem rot disease of groundnut

International audience; Expansion and intensification of human land use represents the major cause of habitat fragmentation. Such fragmentation can have dramatic consequences on species richness and trophic interactions within food webs. Although the associated ecological consequences have been studied by several authors, the evolutionary effects on interacting species have received little research attention. Using a genetic algorithm, we quantified how habitat fragmentation and environmental variability affect the optimal reproductive strategies of parasitic wasps foraging for hosts. As observed in real animal species, the model is based on the existence of a negative trade-off between survival and reproduction resulting from competitive allocation of resources to either somatic maintenance or egg production. We also asked to what degree plasticity along this trade-off would be optimal, when plasticity is costly. We found that habitat fragmentation can indeed have strong effects on the reproductive strategies adopted by parasitoids. With increasing habitat fragmentation animals should invest in greater longevity with lower fecundity; yet, especially in unpredictable environments, some level of phenotypic plasticity should be selected for. Other consequences in terms of learning ability of foraging animals were also observed. The evolutionary consequences of these results are discussed.

The effect of moisture source and diet on the development and reproduction of the pirate bugs, Orius thripoborus (Hesse) and Orius naivashae (Poppius) (Hemiptera: Anthocoridae) was examined in the laboratory. Both species had been collected in and around sugarcane fields in South Africa. Supplementing eggs of the flour moth Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae) with a green bean pod as a moisture source yielded better nymphal survival and faster development, as compared with free water encapsulated in Parafilm, suggesting that the predators may extract extra nutrients from the bean pod. The impact of two factitious foods and moist honey bee pollen on developmental and reproductive parameters of both predators was also investigated. The overall performance of both Orius species on E. kuehniella eggs and cysts of brine shrimp, Artemia franciscana Kellogg (Crustacea: Artemiidae) was better than on pollen. Nonetheless, a pollen diet alone allowed 66 and 78% of the nymphs of O. thripoborus and O. naivashae, respectively, to reach adulthood. Overall, developmental and reproductive performance of O. thripoborus on the tested diets was superior to that of O. naivashae. The implications of these findings for the mass production of these predators and their potential role in biological control programs in southern Africa are discussed.

The area under organic farming is increasing in many countries. The effect of a significant increase in the proportion of organic agriculture on pest (sensu lato) populations at the landscape scale is unknown and will depend on both the production of propagules in organic fields and the risk of pest dispersal between fields. In this study, we observed the dynamics of four foliar diseases, aphids, and weeds in 216 wheat fields over 2 years in northern France. We used the survey data to estimate the local effect of how a field was managed (organic or conventional) and the presence or absence of adjacent organic fields (neighbourhood effect) on pest abundance in that field. Because conventional and organic may be considered extremes along a continuum of management practices, a large survey was undertaken of management practices to ensure that the fields were classified according to the actual cropping practices. The presence or absence of organic certification was determined to be the only relevant criterion for classifying cropping practices. The results of proportional odds mixed models showed that some pests responded to local crop management: leaf blotch incidence and aphid density were significantly lower while weed diversity and abundance were higher in organic fields. Only aphids and leaf blotch responded to the neighbourhood effect: the presence of organic fields in the neighbourhood decreased the number of aphids in both organic and conventional fields and decreased leaf blotch incidence but only in conventional fields. These results indicate that the increase in organic acreage in landscapes will not increase pest problems in the short term under the conditions of the study (low disease pressure). International audience; Bacillus subtilis strain 14B was used to produce a novel antimicrobial peptide (bacteriocin) called Bac 14B. Pure bacteriocin was obtained after heat and acidic treatments (80A degrees C and pH 4), precipitation by ammonium sulfate, and chromatography on Sephadex G-50 and Mono Q Sepharose columns. Based on MALDI-TOF mass spectrometry analysis, purified Bac 14B is a monomer protein with a molecular mass of 20110.13 Da. N-terminal sequencing allowed for the straightforward identification of its first 12 residues, which were of a pure bacteriocin. It also revealed that this bacteriocin contained a unique sequence, namely M-L-K-A-N-L-Q-N-P-L-N-A, suggesting the identification of a novel compound. Bac 14B was stable for 1 h at temperatures up to 80A degrees C and pH of 4 similar to 8. It also proved sensitive to various proteases, which demonstrated its protein nature. Bac 14B displayed a bacteriolytical mode of action and a broad range of inhibitory spectra toward Gram-positive and -negative pathogens. Interestingly, based on conventional agronomic seed vigor parameters, the application of Bac 14B (500 activity units/mL) to various crops revealed that this bacteriocin was a potent exogenous enhancer of growth that stimulated the seedling vigor of tomatoes and muskmelons. Compared to those of the control, the germination percentage, shoot weight, shoot height, and root length were all significantly enhanced in Bac 14B-treated plant seeds. Bac 14B also exhibited effective disinfectant properties against a wide range of seedborne diseases and significant effects on the control of damping off diseases, particularly at the pregermination stage. It also proved to be effective against root rot diseases caused by Alternaria solani and other bacterial seedborne pathogens such as wilt diseases. The findings indicate that Bac 14B is the first B. subtilis-produced bacteriocin ever reported to exhibit such promising biological properties.

To assess their potential to control poultry red mites (Dermanyssus gallinae), we tested selected predaceous mites (Androlaelaps casalis and Stratiolaelaps scimitus) that occur naturally in wild bird nests or sometimes spontaneously invade poultry houses. This was done under laboratory conditions in cages, each with 2–3 laying hens, initially 300 poultry red mites and later the release of 1,000 predators. These small-scale tests were designed to prevent mite escape from the cages and they were carried out in three replicates at each of three temperature regimes: 26, 30 (constant day and night) and 33–25 °C (day-night cycle). After 6 weeks total population sizes of poultry red mites and predatory mites were assessed. For the temperature regimes of 26 and 33/25 °C S. scimitus reduced the poultry red mite population relative to the control experiments by a factor 3 and 30, respectively, and A. casalis by a factor of 18 and 55, respectively. At 30 °C the predators had less effect on red mites, with a reduction of 1.3-fold for S. scimitus and 5.6-fold for A. casalis. This possibly reflected hen manure condition or an effect of other invertebrates in the hen feed. Poultry red mite control was not negatively affected by temperatures as high as 33 °C and was always better in trials with A. casalis than in those with S. scimitus. In none of the experiments predators managed to eradicate the population of poultry red mites. This may be due to a prey refuge effect since most predatory mites were found in and around the manure tray at the bottom of the cage, whereas most poultry red mites were found higher up in the cage (i.e. on the walls, the cover, the perch, the nest box and the food box). The efficacy of applying predatory mites in the poultry industry may be promoted by reducing this refuge effect, boosting predatory mite populations using alternative prey and prolonged predator release devices. Biocontrol success, however, will strongly depend on how the poultry is housed in practice (free range, cage or aviary systems) and on which chemicals are applied to disinfect poultry houses and to control other pests.
