203. Complete cDNA sequences of the DRB6 gene from humans and chimpanzees: a possible model of a stop codon readingthrough mechanism in primates

Abstract: The defective major histocompatibility complex (MHC) DRB6 gene is transcribed into mRNA in human [peripheral blood lymphocytes, transfected and Epstein-Barr virus (EBV)] and chimpanzee EBV cell lines. MHC-DRB6 presents several anomalies, which include stop codons in exon 2, lack of the usual polyadenilation signal of other MHC-DRB genes, and a promoter region and exon 1 taken from a locally inserted retrovirus. The complete cDNA sequences from human DRB6*0201 and three common chimpanzee alleles (Patr-DRB6*0108, Patr-DRB6*0109, Patr-DRB6*0111) have been obtained; two exon 1-exon 2 cDNA sequences from bonobos (Papa-DRB6*0101 and Papa-DRB6*0102) are also shown. In contrast to chimpanzee DRB6 transcripts, the human ones: (1) present an exon 1-exon 2 splicing site that includes the transcription of the first 141 nucleotides of intron 1, rendering a longer exon 1, and (2) show a duplication of exon 6, which would render a longer cytoplasmic tail in a putative DRB6 protein. These two characteristics are found in all the human sequences obtained, regardless of the cellular type tested, and they are not present in any of the chimpanzee alleles reported; consequently, they are human-specific. All the alleles reported here bear stop codons in the three possible reading frames; however, a certain level of expression of DRB6 has been observed by cytofluorometry. This could be due to the presence of a selenocysteine insertion sequence (SECIS) stem-loop structure located at the 3 untranslated region of the DRB6 mRNA, which directs selenocysteine incorporation at UGA codons. DRB6 transcription and translation would be the first gene model of a readingthrough stop codon mechanism in primate MHC. It is also feasible that the DRB6 gene might generate a population of short polypeptides, bound to plasmatic membranes, having non-antigen-presenting functions or which are presented by other MHC molecules as HLA-E presents HLA-G and -B leader sequence-derived peptides.

The present work reports the DNA sequence of the polymorphic region from the uncommon complement allele C4B93. It shows a new combination of Chido and Rodgers antigenicities and expresses reverse antigenicity because it carries Rodgers 1, 2, and 3. C4B93 could have arisen from an ancient, non-homologous recombination between C4A3 (or C4A6) and C4B1a or from a homologous recombination between C4B1a and C4B5. These events would be enhanced by the presence of recombination promoting Escherichia coliχ-like signals in the fragment between positions 1157 and 1186. The generation of the C4 polymorphism by recombination would explain the concerted evolution of C4 genes in primates. [ABSTRACT FROM AUTHOR]

It is experimentally shown that the phase acquisition time can be reduced relative to the non-injection locked VCO situation. At the same time, the phase relationship between an injection-locked oscillator and the reference oscillator can also be fixed. These techniques can be applied to the local oscillators synthesis in a coherent receiver for the 12.5, 20 and 30 GHz beacons of the Olympus satellite.

The direct broadcast satellite receiver in the 12 GHz band developed in the Electrophysics Department in cooperation with the R & D Departments of Mier Allende, S.A. and Tagra, S.A. is described. It features a low-noise amplifier with 0.3 and 0.5 mm gate width- GaAs MESFET, balanced mixer and monolithic IF amplifier along with filters, polarizer and feeder. The system has been integrated in a module installed in the focus a 0. 4 - f/D parabolic reflector. Noise figures lesser than 3 dB have been obtained leading to G/T factors greater than 24.5 db/K when using a 3 meterantenna.

Microstrip coupled lines technology has been used to design and built low noise amplifiers in 20 and 30 GHz bands. Active devices are GaAs MESFET and HEMT in chip form. The amplifiers have input and output waveguide interfaces, probe transitions between microstrip line and rectangular waveguide are described. A digital radio-relay link in the band 10.7- 11.7 GHz developed for the remote control system of ENHER is presented. The objectives were the construction of a reliable and yet low-cost transmitter-receiver. To accomplish this, the RF unit has been constructed in microstrip on plastic substrate, and the FI from commercially available TV subsystems. Experimental results for S-parameters measurement of a GaAs FET chip in the 3-22 GHz frequency range are presented. The authors present the 20/30-GHz front-end of the TMS-7 station for performing the measurement of the flux density of one or more signal carriers received from the OLYMPUS satellite. The front-end is composed of two receiving channels and a pilot generator. The 26-36 GHz receiver for a radio astronomy radiometer, to characterize the polarization of the Cosmic Microwave Background, is presented. This receiver is based on very low noise cryogenically cooled amplifiers in the front-end module, while in the back-end module a broadband amplification and direct detection chain is operated at room temperature. Noise temperature of the low-noise amplifier (LNA) is 19 K when cooled at 18 K. Experimental results show a good sensitivity of the complete receiver with an effective bandwidth around 8 GHz. Moreover, output detected voltages agree with estimated values. The ESA Planck satellite is designed to achieve precision imaging of the Cosmic Microwave Background with an unprecedented combination of angular resolution, sensitivity, spectral range and sky coverage. The Low Frequency Instrument is one of two complementary instruments, and covers 30, 44, and 70 GHz with an array of wideband pseudo-correlation, cryogenic radiometers. Advanced qualification models of the radiometer chains and of the instrument electronics have been manufactured, tested and integrated into the LFI Qualification Model. The main radiometer calibration, RF tuning and performance characterization is carried out at a single radiometer chain level, and then verified at instrument integrated level in dedicated cryofacilities. Here we describe the main requirements and instrument design, and we summarize the radiometer calibration strategy optimised during the qualification activity in view of the LFI Flight Model campaign.

Microstrip coupled lines technology has been used to design and built low noise amplifiers in 20 and 30 GHz bands. Active devices are GaAs MESFET and HEMT in chip form. The amplifiers have input and output waveguide interfaces, probe transitions between microstrip line and rectangular waveguide are described. Two three-stage low noise amplifiers, at 20 and 30 GHz respectively, are presented.

The QUIJOTE (Q-U-I JOint Tenerife) CMB Experiment is designed to observe the polarization of the Cosmic Microwave Background and other Galactic and extragalactic signals at medium and large angular scales in the frequency range of 10-40 GHz. The first of the two QUIJOTE telescopes and the multi-frequency (10-20 GHz) instrument have been in operation since November 2012. In 2014 a second telescope and a new instrument at 30GHz will be ready for commissioning, and an additional instrument at 40 GHz is in its final design stages. After three years of effective observations, the data obtained by these telescopes and instruments will have the required sensitivity to detect a primordial gravitational-wave component if the tensor-to-scalar ratio is larger than r = 0.05. At the moment, we have completed half of the wide Galactic survey with the multi-frequency instrument covering 18 000 square degrees of the Northern hemisphere. - Abstract
We present the current status of the QUIJOTE (Q-U-I JOint TEnerife) experiment, a new polarimeter with the aim of characterizing the polarization of the Cosmic Microwave Background, and other galactic or extra-galactic physical processes that emit in microwaves in the frequency range 10–42 GHz, and at large angular scales (around 1 degree resolution). The experiment has been designed to reach the required sensitivity to detect a primordial gravitational wave component in the CMB, provided its tensor-to-scalar ratio is larger than r ∼ 0.05. The project consists of two telescopes and three instruments which will survey a large sky area from the Teide Observatory to provide I, Q and U maps of high sensitivity. The first QUIJOTE instrument, known as Multi-Frequency Instrument (MFI), has been surveying the northern sky in four individual frequencies between 10 and 20 GHz since November 2012, providing data with an average sensitivity of 80 µK beam−1 in Q and U in a region of 20, 000 square-degrees. The second instrument, or Thirty-GHz Instrument (TGI), is currently undergoing the commissioning phase, and the third instrument, or Forty-GHz Instrument (FGI), is in the final fabrication phase. Finally, we describe the first scientific results obtained with the MFI. Some specific regions, mainly along the Galactic plane, have been surveyed to a deeper depth, reaching sensitivities of around 40 µK beam−1. We present new upper limits on the polarization of the anomalous dust emission, resulting from these data, in the Perseus molecular complex and in the W43 molecular complex.

- Abstract
The QUIJOTE-CMB project has been described in previous publications. Here we present the current status of the QUIJOTE multi-frequency instrument (MFI) with five separate polarimeters (providing 5 independent sky pixels): two which operate at 10-14 GHz, two which operate at 16-20 GHz, and a central polarimeter at 30 GHz. The optical arrangement includes 5 conical corrugated feedhorns staring into a dual reflector crossed-draconian system, which provides optimal cross-polarization properties (designed to be < −35 dB) and symmetric beams. Each horn feeds a novel cryogenic on-axis rotating polar modulator which can rotate at a speed of up to 1 Hz. The science driver for this first instrument is the characterization of the galactic emission. The polarimeters use the polar modulator to derive linear polar parameters Q, U and I and switch out various systematics. The detection system provides optimum sensitivity through 2 correlated and 2 total power channels. The system is calibrated using bright polarized celestial sources and through a secondary calibration source and antenna. The acquisition system, telescope control and housekeeping are all linked through a real-time gigabit Ethernet network. All communication, power and helium gas are passed through a central rotary joint. The time stamp is synchronized to a GPS time signal. The acquisition software is based on PLCs written in Beckhoffs TwinCat and ethercat. The user interface is written in LABVIEW. The status of the QUIJOTE MFI will be presented including pre-commissioning results and laboratory testing.

The Andromeda Galaxy (M31) is the Local Group galaxy that is most similar to the Milky Way (MW). The similarities between the two galaxies make M31 useful for studying integrated properties common to spiral galaxies. We use the data from the recent QUIJOTE-MFI Wide Survey, together with new raster observations focused on M31, to study its integrated emission. The addition of raster data improves the sensitivity of QUIJOTE-MFI maps by almost a factor 3. Our main interest is to confirm if anomalous microwave emission (AME) is present in M31, as previous studies have suggested. To do so, we built the integrated spectral energy distribution of M31 between 0.408 and 3000 GHz. We then performed a component separation analysis taking into account synchrotron, free–free, AME, and thermal dust components. AME in M31 is modelled as a log-normal distribution with maximum amplitude, A AME, equal to 1.03 ± 0.32 Jy. It peaks at |${\nu _{\rm AME}}=17.2\pm 3.2{\rm \, GHz}{}$| with a width of W AME = 0.58 ± 0.16. Both the Akaike and Bayesian information criteria find the model without AME to be less than 1 per cent as probable as the one taking AME into consideration. We find that the AME emissivity per 100 |$\mu$| m intensity in M31 is |${\epsilon _{\rm AME}^{\rm 28.4\, GHz}}=9.6\pm 3.1\,\mu$| K MJy−1 sr, similar to that of the MW. We also provide the first upper limits for the AME polarization fraction in an extragalactic object. M31 remains the only galaxy where an AME measurement has been made of its integrated spectrum. We use the new QUIJOTE-MFI wide survey (11, 13, 17, and 19 GHz) to produce spectral energy distributions (SEDs), on an angular scale of 1°, of the supernova remnants (SNRs) CTB 80, Cygnus Loop, HB 21, CTA 1, Tycho, and HB 9. We provide new measurements of the polarized synchrotron radiation in the microwave range. The intensity and polarization SEDs are obtained and modelled by combining QUIJOTE-MFI maps with ancillary data. In intensity, we confirm the curved spectra of CTB 80 and HB 21 with a break frequency νb at 2.0 |$^{+1.2}_{-0.5}$| and 5.0 |$^{+1.2}_{-1.0}$| GHz, respectively; and spectral indices above the break of |$-0.6^{+0.04}_{-0.05}$| and |$-0.86^{+0.04}_{-0.05}$|. We provide constraints on the Anomalous Microwave Emission, suggesting that it is negligible towards these SNRs. From a simultaneous intensity and polarization fit, we recover synchrotron spectral indices as flat as −0.24, and the whole sample has a mean and scatter of −0.44 ± 0.12. The polarization fractions have a mean and scatter of 6.1 ± 1.9 per cent. When combining our results with the measurements from other QUIJOTE (Q-U-I JOint TEnerife CMB experiment) studies of SNRs, we find that radio spectral indices are flatter for mature SNRs, and particularly flatter for CTB 80 (|$-0.24^{+0.07}_{-0.06}$|) and HB 21 (|$-0.34^{+0.04}_{-0.03}$|). In addition, the evolution of the spectral indices against the SNRs age is modelled with a power-law function, providing an exponent −0.07 ± 0.03 and amplitude −0.49 ± 0.02 (at 10 kyr), which are conservative with respect to previous studies of our Galaxy and the Large Magellanic Cloud. Anomalous microwave emission (AME) is an important emission component between 10 and 60 GHz that is not yet fully understood. It seems to be ubiquitous in our Galaxy and is observed at a broad range of angular scales. Here we use the new QUIJOTE-MFI wide survey data at 11, 13, 17, and 19 GHz to constrain the AME in the Galactic plane (| b | < 10°) on degree scales. We built the spectral energy distribution between 0.408 and 3000 GHz for each of the 5309 0.9° pixels in the Galactic plane, and fitted a parametric model by considering five emission components: synchrotron, free–free, AME, thermal dust and CMB anisotropies. We show that not including QUIJOTE-MFI data points leads to the underestimation (up to 50 per cent) of the AME signal in favour of free–free emission. The parameters describing these components are then intercompared, looking for relations that help to understand AME physical processes. We find median values for the AME width, W AME, and for its peak frequency, νAME, respectively of |$0.560^{+0.059}_{-0.050}$| and |$20.7^{+2.0}_{-1.9}$| GHz, slightly in tension with current theoretical models. We find spatial variations throughout the Galactic plane for νAME, but only with reduced statistical significance. We report correlations of AME parameters with certain ISM properties, such as that between the AME emissivity (which shows variations with the Galactic longitude) and the interstellar radiation field, and that between the AME peak frequency and dust temperature. A new octave bandwidth high selectivity compact microstrip diplexer working in the band from 10 to 20 GHz is presented, intended to be a part of an electro-optical interferometer covering the whole frequency band. The circuit is based on the combination of hybrid couplers and bandpass filters both on the microstrip technology. The diplexer provides two output frequency bands which cover the 10–14 GHz (relative bandwidth of 33%) and the 16–20 GHz (relative bandwidth of 22%) ranges, respectively, with a stop band in between from 14 to 16 GHz. Measured results show a insertion loss level of 3.3 dB for both output bands and high selectivity performance, exhibiting a rejection level between output bands higher than 20 dB and return loss better than 10 dB in all ports.
