An accurate investigation of the landslide dam breach process is crucial for the understanding the breach mechanism and disaster prediction. However, the numerical research on the landslide dam breach process to date is rarely reported, especially regarding the soil-water flow coupling effect incorporated in the erosion process. This paper presents a numerical investigation on the longitudinal breach process of landslide dams via a coupled discrete element method (DEM) and computational fluid dynamics (CFD) with the volume of fluid (VOF). Moreover, a virtual sphere model is proposed to overcome the computational instability caused by the particle size approaching the mesh size. The accuracy and validity of the improved coupled method are verified using a series of single particle sedimentation cases. By employing this method, the longitudinal breach process of landslide dams featuring different materials and hydrodynamic conditions has been simulated. It is found to satisfactorily reproduce the longitudinal breach process of landslide dams including surface flow erosion, backward erosion, head-cut erosion, and water and sediment rebalance or complete breach. The effects of the inflow discharges and dam materials on the erosion process are systematically resolved. The breach flow can cause the rotation trend of particles and lead to the increase of tangential contact force at the initial stage of the dam breaching. During the breach process, both the strength and density of the force chain continue to attenuate. The results obtained from the improved coupled DEM-CFD simulations can reasonably explain the particle-fluid interaction mechanisms, physical and morphological evolution and breach process at both macroscopic and mesoscopic scales.

Background: The Ommaya reservoir implantation technique allows for bypass of the blood-brain barrier. It can be continuously administered locally and be used to repeatedly flush the intracranial cavity to achieve the purpose of treatment. Accurate, fast, and minimally invasive placement of the drainage tube is essential during the Ommaya reservoir implantation technique, which can be achieved with the assistance of robots.Methods: We retrospectively analyzed a total of 100 patients undergoing Ommaya reservoir implantation, of which 50 were implanted using a robot, and the remaining 50 were implanted using conventional surgical methods. We then compared the data related to surgery between the two groups and calculated the accuracy of the drainage tube of the robot-assisted group.Results: The average operation time of robot-assisted surgery groups was 41.17 ± 11.09 min, the bone hole diameter was 4.1 ± 0.5 mm, the intraoperative blood loss was 11.1 ± 3.08 ml, and the average hospitalization time was 3.9 ± 1.2 days. All of the Ommaya reservoirs were successful in one pass, and there were no complications such as infection or incorrect placement of the tube. In the conventional Ommaya reservoir implantation group, the average operation time was 65 ± 14.32 min, the bone hole diameter was 11.3 ± 0.3 mm, the intraoperative blood loss was 19.9 ± 3.98 ml, and the average hospitalization time was 4.1 ± 0.5 days. In the robot-assisted surgery group, the radial error was 2.14 ± 0.99 mm and the axial error was 1.69 ± 1.24 mm.Conclusions: Robot-assisted stereotactic Ommaya reservoir implantation is quick, effective, and minimally invasive. The technique effectively negates the inefficiencies of craniotomy and provides a novel treatment for intracranial lesions.

Abscisic acid (ABA) is an important phytohormone regulating plant growth, development and stress responses. A multitude of key factors implicated in ABA signaling have been identified; however, the regulation network of these factors needs for further information. AtS40.4, a plant-specific DUF584 domain-containing protein, was identified previously as a senescence regulator in Arabidopsis. In this study, our finding showed that AtS40.4 was negatively involved in ABA signaling during seed germination and early seedling growth. AtS40.4 was highly expressed in seeds and seedlings, and the expression level was promoted by ABA. AtS40.4 was localized both in the nucleus and the cytoplasm. Moreover, the subcellular localization pattern of AtS40.4 was affected by ABA. The knockdown mutants of AtS40.4 exhibited an increased sensitivity to ABA, whereas the overexpression of AtS40.4 decreased the ABA response during seed germination and seedling growth of Arabidopsis. Furthermore, AtS40.4 was involved in ABRE-dependent ABA signaling and influenced the expression levels of ABA INSENTIVE (ABI)1-5 and SnRK2.6. Further genetic evidence demonstrated that AtS40.4 functioned upstream of ABI4. These findings support the notion that AtS40.4 is a novel negative regulator of the ABA response network during seed germination and early seedling growth.

Background and Aims. Simethicone (SIM), as an antifoaming agent, has been shown to improve bowel preparation during colonoscopy. However, the optimal timing of SIM addition remained undetermined. We aimed to investigate the optimal timing of SIM addition to polyethylene glycol (PEG) to improve bowel preparation. Methods. Eligible patients were randomly assigned to two groups: the SIM evening group (SIM addition to PEG in the evening of the day prior to colonoscopy) and the SIM morning group (SIM addition to PEG in the morning of colonoscopy). The primary outcome was Bubble Scale (BS). The secondary outcomes were Boston Bowel Preparation Scale (BBPS) and adenoma detection rate (ADR). Results. A total of 419 patients were enrolled in this study. The baseline characteristics of the patients were similar in both groups. No significant differences were observed in terms of BS (8.76 ± 0.90 vs. 8.65 ± 1.16, P = 0.81), ADR (34.1% vs. 30.8%, P = 0.47), Boston Bowel Preparation Scale (BBPS) (8.59 ± 0.94 vs. 8.45 ± 1.00, P = 0.15), and withdrawal time (8.22 ± 2.04 vs. 8.01 ± 2.51, P = 0.094) between the two groups. Background. Submucosal tunneling endoscopic resection (STER) has effectively removed esophageal submucosal tumors (SMTs) originating from the muscularis propria (MP) layer. However, clinical failure and adverse events of STER remain concerned. In this study, we described a mark-guided STER (markings before creating entry point) and evaluated its feasibility and safety for esophageal SMTs originating from MP. Methods. Patients receiving the mark-guided STER from October 2017 to July 2020 were included and followed up (ranged from 3 to 30 months). The primary outcomes included complete resection, en bloc resection, and R0 resection rates. The secondary outcomes included procedure duration, main complication, and residual lesions. Results. A total of 242 patients with 242 SMTs (median diameter of 22 mm, ranging from 7 mm to 40 mm) received the mark-guided STER. The median procedure duration was 55 min (ranging from 35 min to 115 min). The complete resection, en bloc resection, and R0 resection rates were 100%, 98.3%, and 97.5%, respectively. The adverse event rate was 4.5%. However, there was no severe complication. No residual SMTs were detected during the follow-up period. Logistic regression demonstrated that the SMT size and procedure duration were independent factors associated with en bloc resection (P=0.02 and P=0.04, respectively). Moreover, logistic regression demonstrated that the SMT size was an independent risk factor for main complications (P=0.02). Conclusion. Mark-guided STER was feasible and safe to remove esophageal SMTs ≦40 mm. However, it is necessary to further verify the feasibility and safety for the esophageal SMTs >40 mm.

Streptococcus agalactiae infection has become a serious bacterial disease of tilapia for its damage. In order to effectively prevent the outbreak of streptococcicosis, a recombinant Lactococcus lactis expressing surface immunogenicity protein (Sip) of S. agalactiae was constructed as probiotic vector oral vaccine in this study. The immunogenicity of the recombinant L. lactis was evaluated through detecting the level of specific serum IgM antibody, expression level of immune-related genes and relative percent survival (RPS). The results showed that the recombinant L. lactis could express a 45.5 kDa protein consistent with the expected size of Sip. The recombinant Sip mainly expressed in the form of inclusion body and the concentration of purified Sip could reach 7.65 mg / mL. Nile tilapia were orally immunized with different concentrations (2.24 × 109, 2.24 × 1010 and 2.24 × 1011 CFU / mL) of recombinant L. lactis (NZ9000-pNZ8148-sip). The analysis results of specific serum antibody showed that the level of antibody of tilapia which were vaccinated with medium and high concentration of recombinant L. lactis (2.24 × 1010 CFU / mL and 2.24 × 1011 CFU / mL) were significantly higher than that of control group. The relative percentage of survival (RPS) of tilapia vaccinated with medium concentration of NZ9000-pNZ8148-sip reached 61.6 % at 21 days post-immunization. Quantitative real-time PCR (qRT-PCR) results revealed that the immune-related genes of IgT, IgM, CD8a and C3 were significantly upregulated expression in thymus, liver, spleen and hindgut. In conclusion, the recombinant L. lactis vaccine induced both humoral and cellular responses of tilapia. This study demonstrates the potential of using L. lactis as a delivery system to develop an oral vaccine against S. agalactiae.

Photo-fermentative hydrogen production, the new energy production alternative, was greatly enhanced by formed biofilm. Experimental results indicated that a possible and effective altered system could transfer light to hydrogen. Proteins were significantly regulated, for example those related with nitrogenase, flagellin, EPS transportation and DNA duplication were up-regulated while those concerned photosystem were down-regulated. It revealed these changes of proteins contributed to positive activity of key enzymes, improved communication system and increased total light utilization efficiency thus leading to enhanced capacity of hydrogen production. Besides above metabolic changes inside the cells, EPS secreted by the bacteria played an important role in hydrogen production and its yield decided the release of hydrogen. When EPS descended to a lower concentration during biofilm formation, it meant carbon source for EPS synthesis was reduced, and more energy and reducing power could be transferred into hydrogen energy. More importantly, this work found that composition and structure of EPS were efficiently influenced by the formation of biofilm, such as benzene and O-H structure, secondary protein structure and the kinds of protein, which were important to stable biofilm and efficient hydrogen production. Therefore, final hydrogen yield was improved by altered protein and EPS resulted from biofilm formation. This study demonstrated that formation of biofilm is an efficient, ecological and attracting way to the future bio-hydrogen production.

Nitrate/nitrite-dependent anaerobic methane oxidation (n-DAMO) coupling to Anaerobic ammonium oxidation (Anammox) provides an opportunity for simultaneous nitrogen removal and methane emissions mitigation from wastewater. However, to achieve high nitrogen removal rate in such a process remains a critical challenge in practical application. This work investigated the interactions between n-DAMO and Anammox in membrane biofilm reactor (MBfR) and then developed operational strategies of MBfR for high rate nitrogen removal from landfill leachate. Initially, influent containing nitrate and ammonium facilitated the development of n-DAMO and Anammox microorganisms in MBfR, but nitrogen removal performance is hard to be further improved even deteriorated. Detailed investigations of interactions among n-DAMO and Anammox microorganisms confirmed that extra addition of nitrite into MBfR fed with nitrate and ammonium not only stimulated the activities of Anammox bacteria, but also enhanced the activities of n-DAMO archaea from 172.3 to 356.9 mg NO3−-N L−1 d−1. Functional gene analysis also indicated that mcrA and hzsA genes increased after nitrite addition. Based on this finding, influent containing NO3−, NO2− and NH4+ enabled nitrogen removal rates of MBfR increase from 224.9 to 888.2 mg N L−1 d−1. Finally, nitrate in the influent was gradually replaced with nitrite to mimic the effluent from partial nitriation of landfill leachate, but maintain the nitrate availability for n-DAMO archaea through increasing nitrate production from Anammox. These operation strategies enabled MBfR achieve the steady state with a nitrogen removal rate of 6.1 kg N m−3 d−1. Microbial community analysis revealed n-DAMO archaea, n-DAMO bacteria and Anammox bacteria jointly dominated the biofilm, and their relative abundance dynamically shifted with feeding regime. This work provides promising operational strategies for high rate of nitrogen removal from landfill leachate through integrating n-DAMO and Anammox process.

Methane emissions and plastic pollution are critical global challenges. The biological conversion of methane to poly-β-hydroxybutyrate (PHB) not only mitigates methane emissions but also provides biodegradable polymer substitutes for petroleum-based materials used in plastics production. This work provides an early overview of the methane-based PHB advances and discusses challenges and related strategies. Recent advances of PHB, including PHB biosynthetic pathways, methanotrophs, bioreactors, and the performances of PHB materials are introduced. Major challenges of methane-based PHB production are discussed in detail; these include low efficiency of methanotrophs, low gas-liquid mass transfer efficiency, and poor material properties. To overcome these limitations, various approaches are also explored, such as feast-famine regimes, engineered microorganisms, gas-permeable membrane bioreactors, two-phase partitioning bioreactors, poly-β-hydroxybutyrate-co-hydroxyvalerate synthesis, and molecular weight manipulation.

Objective. To explore the feasibility and safety of the active retrograde backup (ARB) for treatment of chronic total occlusion (CTO) during retrograde percutaneous coronary intervention (PCI). Background. Guiding support plays an important role in guidewire and microcatheter coronary channel (CC) tracking in retrograde PCI therapy for patients with CTO. However, the feasibility and safety of retrograde active use of a mother-and-child catheter are still unclear. Patients and Methods. A total of 271 consecutive patients with CTO who underwent retrograde PCI between January 2015 and January 2020 were retrospectively analyzed. Clinical data of two groups were compared to evaluate the feasibility and safety of ARB. Results. Of the 271 patients, 69.0% (187/271) underwent therapy through the septal branch, 31.0% (84/271) through the epicardial collateral channel, and 47.6% (129/271) through active retrograde extra backup with a mother-and-child catheter to facilitate retrograde microcatheter collateral CC tracking. The time of wire CC tracking was shorter in the ARB group than that in the non-ARB group (25.4 ± 8.5 vs 26.4 ± 9.7, p=0.348), but there was no significant difference. The duration of the retrograde microcatheter tracking (10.2 ± 3.8 vs 15.5 ± 6.8, p=0.012) and the retrograde approach (62.8 ± 20.3 vs 70.4 ± 24.3, p=0.026) in the ARB group was significantly shorter than that in the non-ARB group. The radiation dose (223.6 ± 112.7 vs. 295.2 ± 129.3, p=0.028), fluoroscopy time (50.6 ± 21.3 vs 62.3 ± 32.1, p=0.030), and contrast volume (301.8 ± 146.7 vs 352.2 ± 179.5, p=0.032) in the ARB group were significantly lower than that in the non-ARB group. There were no life-threatening procedural complications in either group. Complications unrelated to ARB included two cases of donor-vessel dissection, one case of CC perforation, and two cases of target-vessel perforation. There was no statistically significant difference in major adverse cardiac and cerebrovascular events between the groups during hospitalization p>0.05. Conclusion. ARB is feasible, safe, and conducive to guidewire and microcatheter CC tracking in the recanalization of coronary CTO. It improves procedural efficiency and is worthy of further promotion.

Abstract Background Emerging evidence indicate that miRNAs play an important role on gastric cancer (GC) progression via regulating several downstream targets, but it is still partially uncovered. This study aimed to explore the molecular mechanisms of GC by comprehensive analysis of mRNAs and miRNA expression profiles. Methods The mRNA and miRNA expression profiles of GSE79973 and GSE67354 downloaded from Gene Expression Omnibus were used to analyze the differentially expressed genes (DEGs) and DE-miRNAs among GC tissues and normal tissues. Then, targets genes of DE-miRNAs were predicted and the DE-miRNA–DEG regulatory network was constructed. Next, function enrichment analysis of the overlapped genes between the predicted DE-miRNAs targets and DEGs was performed and a protein–protein interactions network of overlapped genes was constructed. Finally, RT-PCR analysis was performed to detect the expression levels of several key DEGs and DE-miRNAs. Results A set of 703 upregulated and 600 downregulated DEGs, as well as 8 upregulated DE-miRNAs and 27 downregulated DE-miRNAs were identified in GC tissue. hsa-miR-193b-3p and hsa-miR-148a-3p, which targeted most DEGs, were highlighted in the DE-miRNA–DEG regulatory network, as well as hsa-miR-1179, which targeted KNL1, was newly predicted to be associated with GC. In addition, NCAPG, which is targeted by miR-193b-3p, and KNL1, which is targeted by hsa-miR-1179, had higher degrees in the PPI network. RT-qPCR results showed that hsa-miR-148a-3p, hsa-miR-193b-3p, and hsa-miR-1179 were downregulated, and NCAPG and KNL1 were upregulated in GC tissues; this is consistent with our bioinformatics-predicted results. Conclusions The downregulation of miR-193b-3p might contribute to GC cell proliferation by mediating the upregulation of NCAPG; as additionally, the downregulation of miR-193b-3p might contribute to the mitotic nuclear division of GC cells by mediating the upregulation of KNL1.

Background: A beneficial memory effect of acute fornix deep brain stimulation (DBS) has been reported in clinical studies. The aim of this study was to investigate the acute changes in glucose metabolism induced by fornix DBS. Methods: First, the Morris water maze test and novel object recognition memory test were used to confirm declined memory in aged mice (C57BL/6, 20–22 months old). Then, four groups of mice were used as follows: aged mice with stimulation (n = 12), aged mice with sham-stimulation (n = 8), adult mice (3–4 months old) with stimulation (n = 12), and adult mice with sham-stimulation (n = 8). Ipsilateral hippocampal glucose metabolism and glutamate levels were measured in vivo by microdialysis before, during, and after fornix DBS treatment. Histological staining was used to verify the localization of electrodes and mice with inaccurate placement were excluded from subsequent analyses. The effects of fornix DBS on extracellular glucose, lactate, pyruvate, and glutamate levels over time were analyzed by repeated-measures analysis of variance followed by Fisher's least significant difference post hoc test. Results: The aged mice had a higher basal lactate/pyruvate ratio (LPR) and lactate/glucose ratio (LGR) than the adult mice (LPR: 0.34 ± 0.04 vs. 0.13 ± 0.02, t = 4.626, P < 0.0001; LGR: 6.06 ± 0.59 vs. 4.14 ± 0.36, t = 2.823, P < 0.01). Fornix DBS decreased the ipsilateral hippocampal pyruvate and lactate levels (P < 0.05), but the glucose levels were not obviously changed in aged mice. Similarly, the LGR and LPR also decreased in aged mice after fornix DBS treatment (P < 0.05). Glucose metabolism in adult mice was not significantly influenced by fornix DBS. In addition, fornix DBS significantly decreased the ipsilateral hippocampal extracellular levels of glutamate in aged mice (P < 0.05), while significant alterations were not found in the adult mice. Conclusions: The present study provides experimental evidence that fornix DBS could significantly improve hippocampal glucose metabolism in aged mice by promoting cellular aerobic respiration activity.

This experiment was designed to evaluate the potential for dietary alfalfa saponins (AS) to influence cholesterol (Ch) concentrations and the mRNA expressions of the ATP-binding cassette (ABC) that concludes transporters G5 (ABCG5) and G8 (ABCG8) in the livers of laying hens. Hundred and fifty laying hens (27 weeks old) were divided into five treatment groups randomly. Five replicates were assigned to each treatment group consisting of six birds per replicate. The same basic diet was used for all groups with supplementation of 0 (control), 60, 120, 240, and 480 mg/kg of AS, which was given ad libitum to hens for 60 days. The results showed that AS in the diets of laying hens could effectively reduce egg Ch concentrations, and increase the excretion of bile acid by the liver with the expression levels of ABCG5 and ABCG8 improving. Our study found that dietary AS could effect cholesterol metabolism by up-regulating the expression of G5 and G8 in hens, and that AS is a potential agent for reducing Ch concentrations and 120 mg/kg of AS in the diet was suitable and effective for laying hens.

In this study, a statistical model was established to estimate the groundwater table using precipitation, evaporation, the river stage of the Liangduo River, and the tide level of the Yellow Sea, as well as to predict the groundwater table with easily measurable climate data in a coastal plain in eastern China. To achieve these objectives, groundwater table data from twelve wells in a farmland covering an area of 50 m × 150 m were measured over a 12-month period in 2013 in Dongtai City, Jiangsu Province. Trend analysis and correlation analysis were conducted to study the patterns of changes in the groundwater table. In addition, a linear regression model was established and regression analysis was conducted to understand the relationships between precipitation, evaporation, river stage, tide level, and groundwater table. Half-bridge modules with integrated GaN high electron mobility transistors (HEMTs) and driver dies were designed and fabricated in this research. Our design uses flip-chip technology for fabrication, instead of more generally applied wire bonding, to reduce parasitic inductance in both the driver-gate and drain-source loops. Modules were prepared using both methods and the double-pulse test was applied to evaluate and compare their switching characteristics. The gate voltage (Vgs) waveform of the flip-chip module showed no overshoot during the turn-on period, and a small oscillation during the turn-off period. The probabilities of gate damage and false turn-on were greatly reduced. The inductance in the drain-source loop of the module was measured to be 3.4 nH. The rise and fall times of the drain voltage (Vds) were 12.9 and 5.8 ns, respectively, with an overshoot of only 4.8 V during the turn-off period under Vdc = 100 V. These results indicate that the use of flip-chip technology along with the integration of GaN HEMTs with driver dies can effectively reduce the parasitic inductance and improve the switching performance of GaN half-bridge modules compared to wire bonding.

Abstract Background The light/dark cycle is one of the most important factors affecting the microalgal growth and lipid accumulation. Biomass concentration and lipid productivity could be enhanced by optimization of light/dark cycles, and this is considered an effective control strategy for microalgal cultivation. Currently, most research on effects of light/dark cycles on algae is carried out under autotrophic conditions and little information is about the effects under mixotrophic cultivation. At the same time, many studies related to mixotrophic cultivation of microalgal strains, even at large scale, have been performed to obtain satisfactory biomass and lipid production. Therefore, it is necessary to investigate cellular metabolism under autotrophic and mixotrophic conditions at different light/dark cycles. Even though microalgal lipid production under optimal environmental factors has been reported by some researchers, the light/dark cycle and temperature are regarded as separate parameters in their studies. In practical cases, light/dark cycling and temperature variation during the day occur simultaneously. Therefore, studies about the combined effects of light/dark cycles and temperature variation on microalgal lipid production are of practical value, potentially providing significant guidelines for large-scale microalgal cultivation under natural conditions. Results In this work, cell growth and lipid accumulation of an oleaginous microalgal mutant, Scenedesmus sp. Z-4, were investigated at five light/dark cycles (0 h/24 h, 8 h/16 h, 12 h/12 h, 16 h/8 h, and 24 h/0 h) in batch culture. The results showed that the optimal light/dark cycle was 12 h/12 h, when maximum lipid productivity rates of 56.8 and 182.6 mg L−1 day−1 were obtained under autotrophic and mixotrophic cultivation, respectively. Poor microalgal growth and lipid accumulation appeared in the light/dark cycles of 0 h/24 h and 24 h/0 h under autotrophic condition. Prolonging the light duration was unfavorable to the production of chlorophyll a and b, which was mainly due to photooxidation effect. Polysaccharide was converted into lipid and protein when the light irradiation time increased from 0 to 12 h; however, further increasing irradiation time had a negative effect on lipid accumulation. Due to the dependence of autotrophically cultured cells on light energy, the light/dark cycle has a more remarkable influence on cellular metabolism under autotrophic conditions. Furthermore, the combined effects of temperature variation and light/dark cycle of 12 h/12 h on cell growth and lipid accumulation of microalgal mutant Z-4 were investigated under mixotrophic cultivation, and the results showed that biomass was mainly produced at higher temperatures during the day, and a portion of biomass was converted into lipid under dark condition. Conclusions The extension of irradiation time was beneficial to biomass accumulation, but not in favor of lipid production. Even though effects of light/dark cycles on autotrophic and mixotrophic cells were not exactly the same, the optimal lipid productivities of Scenedesmus sp. Z-4 under both cultivation conditions were achieved at the light/dark of 12 h/12 h. This may be attributed to its long-term acclimation in natural environment. By combining temperature variation with optimal light/dark cycle of 12 h/12 h, this study will be of great significance for practical microalgae-biodiesel production in the outdoor conditions.
