Mammalian odorant-binding proteins (OBPs) are soluble lipocalins produced in the nasal mucosa and in other epithelial tissues of several animal species, where they are supposed to serve as scavengers for small structurally unrelated hydrophobic molecules. These would include odorants and toxic aldehydes like 4-hydroxy-2-nonenal (HNE), which are end products of lipid peroxidation; therefore OBP might physiologically contribute to preserve the integrity of epithelial tissues under oxidative stress conditions by removing toxic compounds from the environment and, eventually, driving them to the appropriate degradative pathways. With the aim of developing a biological model based on a living organism for the investigation of the antioxidant properties of OBP, here we asked whether the overexpression of the protein could confer protection from chemical-induced oxidative stress in Escherichia coli. To this aim, bacteria were made to overexpress either GCC-bOBP, a redesigned monomeric mutant of bovine OBP, or its amino-terminal 6-histidine-tagged version 6H-GCC-bOBP. After inducing overexpression for 4 h, bacterial cells were diluted in fresh culture media, and their growth curves were followed in the presence of hydrogen peroxide (H2O2) and tert-Butyl hydroperoxide (tBuOOH), two reactive oxygen species whose toxicity is mainly due to lipid peroxidation, and menadione, a redox-cycling drug producing the superoxide ion. GCC-bOBP and 6H-GCC-bOBP were found to protect bacterial cells from the insulting agents H2O2 and tBuOOH but not from menadione. The obtained data led us to hypothesize that the presence of overexpressed OBP may contribute to protect bacterial cells against oxidative stress probably by sequestering toxic compounds locally produced during the first replication cycles by lipid peroxidation, before bacteria activate their appropriate enzyme-based antioxidative mechanisms. Background: A variable percentage of patients with systemic mast cell (MC) activation symptoms meet criteria for systemic mastocytosis (SM). We prospectively evaluated the clinical utility of the REMA score versus serum baseline tryptase (sBt) levels for predicting MC clonality and SM in 158 patients with systemic MC activation symptoms in the absence of mastocytosis in the skin (MIS). Methods: World Health Organization criteria for SM were applied in all cases. MC clonality was defined as the presence of KIT-mutated MC or by a clonal HUMARA test. The REMA score consisted of the assignment of positive or negative points as follows: male (+1), female (-1), sBt <15 μg/l (-1) or >25 μg/l (+2), presence (-2) or absence (+1) of pruritus, hives or angioedema and presence (+3) of presyncope or syncope. Efficiency of the REMA score for predicting MC clonality and SM was assessed by receiver operating characteristic (ROC) curve analyses and compared to those obtained by means of sBt levels alone. Results: Molecular studies revealed the presence of clonal MC in 68/80 SM cases and in 11/78 patients who did not meet the criteria for SM. ROC curve analyses confirmed the greater sensitivity and a similar specificity of the REMA score versus sBt levels (84 vs. 59% and 74 vs. 70% for MC clonality and 87 vs. 62% and 73 vs. 71% for SM, respectively). Conclusions: Our results confirm the clinical utility of the REMA score to predict MC clonality and SM in patients suffering from systemic MC activation symptoms without MIS. Copyright © 2011 S. Luminex bead-based assays are routinely used in the study of anti-human leukocyte antigen (HLA) donor-specific antibodies (DSA). Single antigen (SA) assays use beads coated with recombinant antigens whereas Luminex crossmatch (Xm-DSA) tests consist of beads coated with isolated donor-specific HLA molecules. The aim of this study was to compare these techniques used to detect DSA. A total of 24 sera recognizing different HLA class I (seven anti-HLA-A and seven anti-HLA-B) as well as class II (seven anti-HLA-DR and three anti-HLA DQ) specificities by complement dependent cytotoxicity were included in the study. These sera were used undiluted and in serial dilutions to perform both class I and II SA and Xm-DSA assays. In the case of Xm-DSA the same serum was checked with different lysates. A total of 42 lysates were used to perform a total of 61 crossmatches: 42 to detect anti-class I and 19 to detect anti-class II antibodies. The maximum positive dilution was higher for SA in 76% of the class I and in 90% of the class II crossmatches. Those cases with a higher sensitivity of the Xm-DSA could not be explained by a larger number of antigen targets. [ABSTRACT FROM AUTHOR]

Identification and genetic mapping of loci conferring resistance to polycyclic pathogens such as the rust fungi depends on accurate measurement of disease resistance. We converted an absolute quantification assay of Puccinia coronata DNA to a relative assay by adding a TaqMan® primers/probe set specific to the oat β-actin gene to simplify and improve quantification of fungal infection. The new multiplex assay estimates the amount of fungal DNA in a sample relative to the amount of host DNA and requires fewer and less labour-intensive steps than previous assays. The relative fungal DNA assay (RFDNA) reliably detected and quantified both host and pathogen DNA over five orders of magnitude and was at least as sensitive as either digital image analysis (DLA) or absolute estimation of fungal DNA (AFDNA) in repeated greenhouse studies using 12 oat cultivars with different resistance responses to P. coronata isolate LGCG. Measuring crown rust resistance to LGCG using DLA, AFDNA and RFDNA in a P8669/P94163 recombinant inbred line population produced segregation ratios that did not differ from the 1:1 Mendelian ratio expected for a single gene. Compared to the AFDNA assessment method, the RFDNA assay is equally sensitive, yet faster and much easier to use than the AFDNA method for precise quantification of the crown rust pathogen in oat leaves. The method will be especially useful for streamlining measurement of partial resistance, since uncovering small differences in resistance requires phenotypic evaluation of large populations. The mechanization of fields that is taking place in some Mediterranean areas is generating soils with poor structure and lack of nutrients. The application of organic wastes for improving soil properties is becoming a frequent practice, but with the risk of producing a negative impact, because of the presence of metals. This application is carried out without any additional checks using the same rates in disturbed and in non-disturbed soils. In the present study, the influence of composted cattle manure application in vineyard soils from the NE of Spain was evaluated. The study focuses on the changes of soil total Zn content in both disturbed and undisturbed soils. The same treatments and practices were applied in both plots. The area has a Mediterranean climate and the main type of soil is Typic Calcixerept. Total levels (digestion with aqua regia) and plant available fractions (extraction with (NH4)OAc-EDTA and CaCl2-DTPA) of Zn, the main metal component of the applied organic wastes, were analysed and compared with those existing in the untreated soils. Zinc levels rose to 130 mg kg−1, which implies increases ranging between 60 and 100% in some parts of the plots. Significant increases were observed in the treated soil samples. The available fraction represents, on average, 30% for Zn extracted with EDTA and 10.8% when it was extracted using DTPA. Significant differences were found between disturbed and undisturbed soils. The organic matter content was the soil characteristic that showed a higher influence on the Zn total levels and on the available fraction, although it was different for the two methods used. The analysis points out the importance of an analysis previous to the application of any waste at general scale. This practice, which is common in the area and repeated in each plot every 3 or 4 years implies a risk of soil pollution in a very short time period. [Copyright &y& Elsevier]

Mitochondria from a patient diagnosed with Kearns-Sayre syndrome (KSS) exhibited severely diminished cytochrome c oxidase activity and at least four mitochondrial DNA (mtDNA) species: 9%-11% of the fulllength mtDNA (16.6 kb), 70%-75% of a ll.7-kb population (harboring the 4,977-bp common deletion), 2%-3% of a 10.5-kb population, and 12%-17% of a 8.9-kb population. The 8.9-kb mtDNA exhibited a secondary deletion that extended 7,704 bp from nucleotide 7,979 in the cox2 gene to nucleotide 15,683 in the cytb gene. The article reports on the chemical ratios and groundwater contamination in east Texas. Multiple sources of contamination often impact groundwater quality. These sources may be anthropogenic, natural, or a combination of both. Chemical ratios can be useful for identifying probable sources of groundwater contamination. The purpose of this study was to map and interpret chemical ratios in groundwater of east Texas. The studies conducted in various regions has shown that certain chemical ratios may be useful for identifying salt domes, lignite deposits, or oilfield brine as sources of groundwater contamination.

This investigation considers the association between patterns of emotional reactivity and reliance on mother in infancy and cognitive and language developments at age 2. Low-income women (N = 518) and their firstborn infants participated in (1) a lab-based assessment where emotion challenges were presented when the infants were 6 to 9 months old, and (2) an assessment of language and cognitive skills at age 2. After controlling for birthweight, early sensorimotor delay, and age at testing, infants who displayed a pattern of combined high reactivity and high reliance on mother in response to positive, anger, and fear emotion challenges had higher cognitive and language skills at age 2 compared with infants who displayed patterns of low reactivity and low reliance on mother. Children who showed high fearful distress and low reliance on mother and whose mothers had low psychological resources had especially poor developmental outcomes. We present griz photometric light curves for the full 5 yr of the Dark Energy Survey Supernova (DES-SN) program, obtained with both forced point-spread function photometry on difference images (DiffImg) performed during survey operations, and scene modelling photometry (SMP) on search images processed after the survey. This release contains 31,636 DiffImg and 19,706 high-quality SMP light curves, the latter of which contain 1635 photometrically classified SNe that pass cosmology quality cuts. This sample spans the largest redshift (z) range ever covered by a single SN survey (0.1 < z < 1.13) and is the largest single sample from a single instrument of SNe ever used for cosmological constraints. We describe in detail the improvements made to obtain the final DES-SN photometry and provide a comparison to what was used in the 3 yr DES-SN spectroscopically confirmed Type Ia SN sample. We also include a comparative analysis of the performance of the SMP photometry with respect to the real-time DiffImg forced photometry and find that SMP photometry is more precise, more accurate, and less sensitive to the host-galaxy surface brightness anomaly. The charge particle behavior in the electron cyclotron resonance plasma confined in a magnetic field of mirror type is studied through the particle-in-cell method. The energy which is transmitted to the plasma is of 2.45 GHz microwaves of 2.0 kV/cm amplitude. It is shown that when the electron cyclotron resonance surface have a form of a one sheet hypeboloid is formed a driven plasma ring localized in the median plane of the hyperboloid. Bariatric surgery is the most effective treatment for severe obesity and its related comorbidities. Roux-en-Y Gastric Bypass (RYGB) and Sleeve Gastrectomy (SG) are currently the most popular weight-loss surgeries used worldwide. Following these surgeries, many patients self-report changes in taste perception and decreased preference for unhealthy foods. These reported changes might account for increased adherence to healthier diets and successful weight loss after surgeries. However, researchers have used a variety of methodologies to assess patients' reported changes andresults are discrepant. The goal of this review is to summarize the literature regarding changes to taste function and ingestive behavior following RYGB and SG to examine differences in findings by methodology (indirect vs. direct measurements). We focused our review around changes in sweets, fats, and alcohol because most of the documented changes in ingestive behavior post-surgery are related to changes in these dietary items. We found that studies using surveys and questionnaires generally find that subjects self-report changes in taste and decrease their preference and cravings for energy-dense foods (particularly, sweets and high-fats). However, studies using validated sensory techniques that include oral sampling or by using direct food intake measurements find little to no change in subjects' ability to perceive taste or their preference for energy-dense foods. Therefore, reported changes in taste and food preferences are unlikely to be explained by alterations in taste intensity and diet selection, and are rather related to changes in the rewarding value of food. Further, that RYGB, and likely SG, is associated with increased alcohol consumption and arisk to develop an alcohol use disorder) supports the notion that these surgeries alter central circuits of reward that are critical in the regulation of ingestive behavior.
