In mammals, the extraocular muscle fibers can be categorized in singly-innervated and multiply-innervated muscle fibers. In the monkey oculomotor, trochlear and abducens nucleus the motoneurons of multiply-innervated muscle fibers lie separated from those innervating singly-innervated muscle fibers and show different histochemical properties. In order to discover, if this organization is a general feature of the oculomotor system, we investigated the location of singly-innervated muscle fiber and multiply-innervated muscle fiber motoneurons in the rat using combined tract-tracing and immunohistochemical techniques. The singly-innervated muscle fiber and multiply-innervated muscle fiber motoneurons of the medial and lateral rectus muscle were identified by retrograde tracer injections into the muscle belly or the distal myotendinous junction. The belly injections labeled the medial rectus muscle subgroup of the oculomotor nucleus or the greatest part of abducens nucleus, including some cells outside the medial border of abducens nucleus. In contrast, the distal injections labeled only a subset of the medial rectus muscle motoneurons and exclusively cells outside the medial border of abducens nucleus. The tracer detection was combined with immunolabeling using antibodies for perineuronal nets (chondroitin sulfate proteoglycan) and non-phosphorylated neurofilaments. In monkeys both antibodies permit a distinction between singly-innervated muscle fiber and multiply-innervated muscle fiber motoneurons. The experiments revealed that neurons labeled from a distal injection lack both markers and are assumed to represent multiply-innervated muscle fiber motoneurons, whereas those labeled from a belly injection are chondroitin sulfate proteoglycan- and non-phosphorylated neurofilament-immunopositive and assumed to represent singly-innervated muscle fiber motoneurons. The overall identification of multiply-innervated muscle fiber and singly-innervated muscle fiber motoneurons within the rat oculomotor nucleus, trochlear nucleus, and abducens nucleus revealed that the smaller multiply-innervated muscle fiber motoneurons tend to lie separate from the larger diameter singly-innervated muscle fiber motoneurons. Our data provide evidence that rat extraocular muscles are innervated by two sets of motoneurons that differ in their molecular, morphological, and anatomical properties.

Medaka (Oryzias latipes) is one of the few vertebrate experimental animals in which inbred lines have been established. It is also a species that has advanced in genetic studies in a manner comparable to zebrafish. This fish is therefore a good model for studying functional organization of the nervous system, but anatomical analysis of its nervous system has been limited to embryonic stages. In the present study, we investigated anatomy of cranial nerves in adult fish focusing on the visual function, using an inbred strain of medaka. Cranial nerves of medaka were labeled using biocytin, revealing a central distribution of retinofugal terminals, retinopetal neurons, and oculomotor, trochlear and abducens motor neurons. The optic nerve of the adult medaka was of a complete decussation type. Retinofugal terminals were located in 8 brain nuclei, the suprachiasmatic nucleus, nucleus pretectalis superficialis, nucleus dorsolateralis thalami, area pretectalis pars dorsalis (APd), area pretectalis pars ventralis (APv), nucleus of the posterior commissure (NPC), accessory optic nucleus, and the tectum opticum. Retinopetal neurons were identified in 6 brain nuclei, the ganglion of the terminal nerve, preoptic retinopetal nucleus, nucleus dorsolateralis thalami, APd, APv, and NPC. The oculomotor neurons were mostly labeled ipsilaterally and were located dorsomedially, abutting the fasciculus longitudinalis medialis in the mesencephalon. The trochlear nucleus was located contralaterally and dorsolaterally adjacent to the fasciculus longitudinalis medialis in the mesencephalon. The abducens nucleus was located ipsilaterally in a ventrolateral part of the rhombencephalic reticular formation. These results, generally similar to those in other teleosts, provide the basis for future behavioral and genetic studies in medaka.

"Eight-and-a-half" syndrome is "one-and-a-half" syndrome (conjugated horizontal gaze palsy and internuclear ophthalmoplegia) plus ipsilateral fascicular cranial nerve seventh palsy. This rare condition, particularly when isolated, is caused by circumscribed lesions of the pontine tegmentum involving the abducens nucleus, the ipsilateral medial longitudinal fasciculus, and the adjacent facial colliculus. Its recognition is therefore of considerable diagnostic value. We report a 71-year-old man who presented with eight and a half syndrome associated with contralateral hemiparesis and hemihypesthesia, in which brain magnetic resonance imaging scans revealed a lacunar pontine infarction also involving the corticospinal tract and medial lemniscus. These features could widen the spectrum of pontine infarctions, configuring a possible "nine" syndrome.

Two adult rhesus monkeys that had undergone 2 years of electrode penetrations into their abducens and vestibular nuclei, for chronic eye movement studies, were examined histologically. An analysis of their VIth nucleus neurons and lateral rectus muscles revealed the following. Twenty-two percent of the large neurons (approximately 30 microm in diameter), on average, were missing and extensive neuropil disruption and gliosis was evident in the experimental side abducens nuclei as compared with the control side in each animal. While the lateral rectus muscles showed small, but inconsistent, changes in total fiber number, the muscle fiber diameters were altered, leading to a more homogenous muscle and making the typical orbital and global subdivisions of the muscle less distinct. Eye movement records from before and after the electrophysiological studies were comparable. We discuss how the complex architecture of the extraocular muscles as well as the possibility of polyneuronal innervation of single muscle fibers could explain our results.

In the last decades, there have been many efforts directed to gain a better understanding on adult neuron-target cell relationships. Embryonic grafts have been used for the study of neural circuit rewiring. Thus, using several donor neuronal tissues, such as cerebellum or striatum, developing grafted cells have been shown to have the capability of substituting neural cell populations and establishing reciprocal connections with the host. In addition, different lesion paradigms have also led to a better understanding of target dependence in neuronal cells. Thus, for example, axotomy induces profound morphofunctional changes in adult neurons, including the loss of synaptic inputs and discharge alterations. These alterations are probably due to trophic factor loss in response to target disconnection. In this review, we summarize the different strategies performed to disconnect neurons from their targets, and the effects of target substitution, performed by tissue grafting, upon neural properties. Using the oculomotor system-and more precisely the abducens internuclear neurons-as a model, we describe herein the effects of disconnecting a population of central neurons from its natural target (i.e., the medial rectus motoneurons at the mesencephalic oculomotor nucleus). We also analyze target-derived influences in the structure and physiology of these neurons by using cerebellar embryonic grafts as a new target for the axotomized abducens internuclear neurons.

The loss of afferent synaptic boutons is a prominent alteration induced by axotomy on adult central neurons. In this work we attempted to prove whether synapse loss could be reverted by reconnection with a new target. We severed the medial longitudinal fascicle of adult cats and then transplanted embryonic cerebellar primordia at the lesion site immediately after lesion. As previously shown, the transected axons from abducens internuclear neurons penetrate and reinnervate the graft [J Comp Neurol 444 (2002) 324]. By immunocytochemistry and electron microscopy we studied the synaptology of abducens internuclear neurons under three conditions: control, axotomy and transplant (2 months of survival time). Semithin sections of the abducens nucleus were immunostained against calretinin, to identify abducens internuclear neurons, and either synaptophysin (SF), to label synaptic terminals, or glial fibrillary acidic protein (GFAP) to detect the astrocytic reaction. Optical and linear density of SF and GFAP immunostaining were measured. Data revealed a significant decrease in the density of SF-labeled terminals with a parallel increase in GFAP-immunoreactive elements after axotomy. On the contrary, in the transplant group, the density of SF-labeled terminals was found similar to control, and the astrocytic reaction induced by lesion was significantly reduced. At the ultrastructural level, synaptic coverage and linear density of boutons were measured around the somata of abducens internuclear neurons. Whereas a significant reduction in both parameters was found after axotomy, cells of the transplant group received a normal density of synaptic endings. The ratio between F- and S-type boutons was found similar in the three groups. Therefore, these findings indicate that the grafting of a new target can prevent the loss of afferent synaptic boutons produced by the axotomy.

In this report, we provide the first characterization of abducens nucleus neuron (ABN) discharge dynamics during horizontal disjunctive saccades. These movements function to rapidly transfer the visual axes between targets located at different eccentricities and depths. Our primary objective was to determine whether the signals carried by ABNs during these movements are appropriate to drive the motion of the eye to which they project. We also asked whether ABNs encode eye movements similarly during disjunctive saccades and disjunctive fixation. To address the first objective we 1) assessed whether we could predict the discharge dynamics of individual neurons during disjunctive saccades based on their discharge properties during conjugate saccades and 2) directly estimated the sensitivity of individual neurons to either the ipsilateral/contralateral eye or the conjugate/vergence position and velocity using bootstrap statistics. Our main finding was that during disjunctive saccades in the direction ipsilateral to the recording site (on-direction), the majority of ABNs preferentially encoded the velocity and the position of the ipsilateral eye. The remaining neurons predominantly encoded the conjugate motion of the eyes (i.e., were equally sensitive to the motion of both eyes). Generally, ipsilateral/contralateral eye based models better described neuronal discharges than conjugate/vergence based models, yet both model structures yielded similar conclusions. Moreover, the preferred eye of individual neurons based on their position and velocity sensitivities were generally well matched. We also found that for saccades in the off-direction, the pausing behavior of ABNs was similar during conjugate and disjunctive saccades, with the exception that for movements of small amplitudes, more ABNs paused during conjugate saccades. Finally, we found that putative motoneurons and internuclear neurons encoded on- and off-direction disjunctive saccades in a similar manner. To address our second objective, we compared the discharge properties of individual ABNs during disjunctive saccades and disjunctive fixation. Good coherence was observed between the preferred eye of individual ABNs during the two behaviors. Taken together, our results indicate that although individual ABNs can encode the motion of both eyes to various degrees, the population drive of ABNs accounts for most of the movement of the ipsilateral eye during disjunctive saccades and disjunctive fixation.

We evaluated the two-dimensional distribution of superior colliculus (SC) neurons visualized after retrograde transneuronal transport of rabies virus injected into the lateral rectus muscle of rhesus monkeys to test whether the density of projection neurons might play a role in the spatiotemporal transformation and vector decomposition. If this were the case, the number of horizontal eye movement-related SC neurons should increase with their distance from the rostral pole of the SC and decrease with their distance from the representation of the horizontal meridian. Labeled neurons of the intermediate SC layers were counted inside a 1-mm-wide band that matched the horizontal meridian of the collicular motor map. Local areal densities were plotted against distance from the rostral SC pole. At 2.5 days after inoculation, there was no labeling in the SC. At 3 days, moderate labeling appeared on both sides, mostly in the intermediate layers. At 3.5 days, cell numbers substantially increased and the laminar distribution changed as cells appeared in the superficial SC layers. At 3 days, rostrocaudal density profiles were unimodal, with peaks at locations near 50 degrees (contralateral SC) and 25-30 degrees (ipsilateral SC) horizontal eccentricity. At 3.5 days, distributions were bimodal due to the appearance of a second high-density region near the rostral pole of the SC. The distribution of SC neurons influencing the abducens nucleus, thus, was nonuniform. Caudal sites contained more neurons, but the experimentally observed density gradients were shallower than the theoretically predicted ones that would be necessary to fully account for the spatiotemporal transformation. Similarly, we studied the distributions of cell densities in the intermediate SC layers along an isoamplitude line (representing saccades of equal amplitudes but different directions). Consistent with theoretical estimates of the density gradients required for vector decomposition, we found that the concentrations of labeled cells were highest in the vicinity of the horizontal meridian but their decrease toward the periphery of the motor map was steeper than predicted. We conclude that SC cell density gradients cannot fully account for the spatiotemporal transformation and vector decomposition in the absence of an additional mechanism such as the previously demonstrated (Grantyn et al., [1997] Soc. Neurosci. Abstr. 23:1295; Moschovakis et al., [1998] J. Neurosci. 18:10219-10229) locus-dependent weighting of the strength of efferent projections to the saccade generators. © 2002 Wiley-Liss, Inc.

In three patients, acute horizontal gaze pareses developed that could not be overcome with the oculocephalic maneuver, indicating a putative lesion of the ipsilateral abducens nerve nucleus. None of the patients had a facial nerve paresis or evidence of a trigeminal sensory neuropathy. Although most lesions that affect the abducens nerve nucleus also damage the ipsilateral fasciculus of the facial nerve, small lesions in this region can produce an isolated horizontal gaze paresis.

Anterograde tracer injections in the rat sensory trigeminal complex are shown here to demonstrate projections to the contralateral facial motor (VII) and accessory abducens (VIacc) nuclei. Most of the trigeminal fibres originated within the pars oralis (5o) and contacted neurones in the medial and intermediate VII. Moderate projections from the pars caudalis (5c) and interpolaris (5i) reached the lateral and dorsolateral VII. Rare projections from the principal nucleus (5P) were found. Trigeminal projections to the contralateral VIacc originated mainly from the 5P and 5o. Few projections from the 5i and 5c to the contralateral VIacc were found. Retrograde tracer injections in the VII showed premotor neurones to the contralateral VII scattered throughout the 5o and in the ventromedial portion of the caudal 5i and the 5c. Double retrograde tracing experiments provide evidence that neurones in the 5o and 5c project to both the ipsi- and contralateral VII. Such collateralization would play a significant role in the co-ordination of the musculature of the face.

Mime artists often use movement to imply the breaking of physical laws: suitcases levitate, air becomes as impermeable as glass. We present the simulated (and actual) violation of a physiologic principle, Hering's Law of Equal Innervation. To provide stereopsis while avoiding double vision, the movements of forward-facing primate eyes must be closely coordinated. In a 19th-century debate, Helmholtz contended that this coordination is a learned process, taming naturally independent eyes. Hering, meanwhile, believed the eyes to be inherently reined to each other and driven by a single control signal.1 Hering's view predominated and this final common pathway for horizontal eye movement came to be accepted as a unilateral signal to the abducens nucleus. This directly controls the lateral rectus of the ipsilateral, abducting, eye but also necessarily adducts the other eye, via interneurons to the contralateral oculomotor nucleus. More recent neurophysiologic evidence, however, indicates that primate saccadic commands can be encoded monocularly,2 in common with the laterally mounted, independently moving eyes of our distant evolutionary forebears. Binocular coordination might therefore not arise as a necessary consequence of neuroanatomic connectivity. If so, it might be possible to execute voluntary monocular saccades. Duane retraction syndrome (DRS) consists of deficient horizontal eye movements, eyelid retraction, palpebral fissure narrowing, and abnormal vertical eye movements. Most cases are sporadic and unilateral (usually left side) with a slight female predominance. Several associated ocular and systemic conditions have been described in DRS patients. In most cases, the abducens nucleus and nerve are absent or hypoplastic, and the lateral rectus muscle is innervated by a branch of the oculomotor nerve. However, there may be contributing mechanical abnormalities. Type I DRS (primary gaze position esotropia with limitation of abduction) comprises the majority of cases. Approximately 50% of type I DRS patients are orthophoric in primary gaze. Esotropia is the most common type of strabismus encountered, and characteristic up shoots and down shoots occur in adduction. Surgical intervention has gradually become more popular in order to improve the primary gaze alignment and mitigate some of the associated abnormalities in ocular motility. However, patients are rarely rendered clinically normal, and limited expectations are appropriate.

Hypothesis: The goal of this study was to map the VIIIth cranial nerve by electrical stimulation. Specifically, the authors sought to 1) characterize auditory and vestibular evoked responses elicited by electrical stimuli delivered directly to the exposed surface of the VIIIth cranial nerve and 2) compare electrically evoked responses elicited in brainstem nuclei with extracranially recorded far-field potentials. Background: Intraoperative monitoring of auditory brainstem responses is useful during cerebellopontine angle surgery. Identification of the vestibular portion of the VIIIth cranial nerve, which traditionally has been performed by physical characteristics and some electrophysiologic properties, is important because the vestibular subdivision in humans is indistinct in approximately 25% of cases. Positive identification of evoked responses emanating from the vestibular nerve would constitute a marked improvement over existing intraoperative techniques that use acoustic stimuli only. Methods: Experiments were performed on 12 anesthetized cats. Electrical pulse stimuli were delivered using a bipolar electrode placed directly on the surface of the exposed VIIIth cranial nerve at several sites. Computer-averaged evoked responses were recorded from far-field electrodes placed on the scalp and from near-field electrodes stereotaxically positioned in or near the inferior colliculus and abducens nucleus. Results: Latencies and morphologies of waves recorded in brainstem nuclei were compared with those of waves recorded extracranially. Direct electrical stimulation of the cochlear nerve elicited a four-wave, auditory brainstem response-like extracranial response, strong activity in the inferior colliculus, and weak activity in the abducens nucleus. Direct stimulation of the vestibular nerve produced a two-wave extracranial response, weak inferior colliculus activity, and strong abducens activation. Stimulation at the border of the cochlear and vestibular nerves produced intermediate responses that possessed both cochlear and vestibular characteristics. Conclusion: Direct electrical stimulation of the cochlear and vestibular subdivisions elicits evoked responses with distinctly different wave morphologies. Obtaining electrically evoked responses intraoperatively is feasible and may be of substantial value in the unambiguous identification of VIIIth cranial nerve subdivisions.

Tetanus neurotoxin (TeNT) is a blocker of synaptic vesicle exocytosis in central synapses with preferential affinity for inhibitory neurotransmission. Following its intramuscular injection, TeNT is retrogradely and trans-synaptically transported towards the premotor terminals. Therefore, we have used TeNT as a tool to study the consequences of functional deafferentation on motoneurons following its peripheral administration. For this, we injected the toxin into the lateral rectus muscle at doses of 5 or 0.5 ng/kg and recorded the discharge activity of abducens motoneurons and internuclear neurons in the alert cat. Our results showed that: (i) TeNT blocked selectively the afferent inhibitory signals on abducens neurons only when used at a low dose, whereas both excitatory and inhibitory synaptic drive was lost after the high dose treatment; (ii) all effects were reversible within one month; and (iii) strikingly, the internuclear neurons of the abducens nucleus showed similar discharge alterations to the motoneurons, suggesting a TeNT action on shared common afferences.

The goal of this study was to examine and compare the number and size of motoneurons in the cat and squirrel monkey abducens nucleus. We also examined medial rectus muscle motoneuron compartmentalization in the squirrel monkey oculomotor nucleus and compared those cells to abducens nucleus motoneurons. Retrograde labeling of the motoneurons, using cholera toxin conjugate of horseradish peroxidase (CTHRP) injected into cat and monkey lateral or medial rectus muscles, was observed after 24 h. The CTHRP was histochemically localized with tetramethylbenzidine. The slide-mounted sections were analyzed using a computerized imaging system. Cat abducens nucleus motoneurons showed a wide range of cell sizes (26.0-66.0 microm, mean = 37.2 +/- 6.2 microm), four or more dendrites per cell and an average of 1,418 cells within a relatively loosely packed nucleus. Squirrel monkey abducens nucleus motoneurons were significantly smaller than those in the cat with a narrower range of cell sizes (20.0-44.0 microm, mean = 31.7 +/- 3.8 microm), four or more dendrites per cell and an average of 2,473 cells densely packed within the nucleus. Squirrel monkey medial rectus muscle motoneurons were organized into MRa, MRb and MRc subgroups. MRa motoneurons comprise the primary innervation for the medial rectus muscle and were similar in size to abducens nucleus motoneurons while the MRc subgroup cells were significantly smaller in size. Similar relationships among medial rectus motoneurons have been seen in rhesus monkeys. The relationship of these anatomical findings to previous physiological results regarding the generation of extraocular muscle force in the squirrel monkey is discussed.

The occurrence of systematic interocular differences in the time of initiation of saccades in various directions was investigated in normal human subjects (n=4). Saccades were recorded binocularly with scleral sensor coils on each of the eyes with a temporal resolution of 0.1 ms (sampling frequency 10 000 Hz). Analysis was done in the velocity domain after digital differentiation. It was found that, in the initial phase of horizontal saccades, the nasalward moving eye lagged the temporalward moving eye consistently by slightly less than 1 ms. No such difference was found in vertical (upward or downward) saccades. In oblique saccades, the systematic initial lag of the nasalward moving eye was similarly present in the horizontal component of the saccade, but absent in the vertical component. It is postulated that this interocular timing difference in horizontal saccades is due to the additional synaptic delay caused by the abducens internuclear neuron in the pathway to the medial rectus muscle of the eye.

Olfactory bulb ensheathing cell (OBEC) transplants promoted axonal regeneration in the spinal cord dorsal root entry zone and in the corticospinal tract. However, OBECs failed to promote abducens internuclear neuron axon regeneration when transplanted at the site of nerve fibre transection. In experiments performed in both cats and rats, OBECs survived for up to 2 months, lining themselves up along the portion of the regrowing axons proximal to the interneuron cell body. However, OBECs migrated preferentially towards abducens somata, in the direction opposite to the oculomotor nucleus target. OBECs seem to promote nerve fibre regeneration only where preferred direction of glial migration coincides with the direction of axonal growth towards its target.

The presence of internuclear neurons in the abducens and oculomotor nuclei of lampreys [Gonzalez, M.J., Pombal, M.A., Rodicio, M.C. and Anadon, R., Internuclear neurons of the ocular motor system of the larval sea lamprey, J. Comp. Neurol. 401 (1998) 1-15] indicates that coordination of eye movements by internuclear neurons appeared early during the evolution of vertebrates. In order to investigate the possible involvement of inhibitory neurotransmitters in internuclear circuits, the distribution of γ- aminobutyric acid (GABA) in the extraocular motor nuclei of the lamprey was studied using immunocytochemical techniques. Small GABA-immunoreactive (GABAir) neurons were observed in the three ocular motor nuclei. Numerous GABAir neurons were observed in the group of internuclear neurons of the dorsal rectus oculomotor subnucleus. A second group of GABAir neurons was observed among and below the trochlear motoneurons. Two further groups of GABAir interneurons, periventricular and lateral, were located in the abducens nucleus among the cells of the caudal rectus and the ventral rectus motor subnuclei, respectively. In addition to the presence of GABAir neurons, in all the ocular motor nuclei the motoneurons were contacted by numerous GABAir boutons. Taken together, these results suggest that GABA is involved as a neurotransmitter in internuclear pathways of the ocular motor system of lampreys. (C) 2000 Elsevier Science B.V.

The topography and localization of motoneurons innervating the six extraocular muscles in the chameleon (Chamaeleo chameleon) was studied following HRP injection in each of these individual muscles. Four muscles were innervated ipsilaterally: medial rectus, inferior rectus, inferior oblique and lateral rectus. The medial rectus muscle was innervated by the dorsomedial part of the oculomotor nucleus. The innervation to the inferior rectus muscle arose from the lateral part of the intermediate oculomotor subnucleus, which extended to the lateral part of the dorsal subdivision. The lateral rectus muscle was innervated by the abducens nucleus, which was composed by two subgroups of labeled cells, respectively observed in the principal and accessory abducens subnuclei, whereas efferents to the inferior oblique muscle originated from both the ventral and intermediate oculomotor subnuclei. The contralateral pattern consisted of motoneurons innervating the superior rectus and the superior oblique that were located respectively in the caudal portion of the ventral oculomotor nucleus and in the trochlear nucleus. These results confirmed data reported in most vertebrate species, and were discussed from a comparative and functional point of view.

Objectives/Hypothesis: The impact of childhood conductive HL (CHL) on development of auditory function has long been debated. The present study was conducted to define and compare the consequences of CHL and cochlear ablation (CA) in young and adult animals, using 2-deoxyglucose (2-DG) uptake as a measure of metabolic activity. It was hypothesized that, for both ages, CHL would result in a decrease in activity in the major ascending central auditory system pathway of the manipulated ear, but that this decrease would be significantly less than that observed with CA. Study Design: Shamcontrolled study of metabolic effects of CHL during sound stimulation. Methods: Gerbils (aged 21 days or adult), underwent malleus removal, CA, or a sham procedure. Young animals survived either 48 hours or 3 weeks; adults survived 3 weeks. Each age/survival CHL group contained eight animals; otherwise, each group (CA and sham) contained five animals, for a total number of 54. At the appropriate survival time, animals were given an intracardiac injection of 14 C-2-DG, and sacrificed under anesthesia after 45 minutes of exposure to normal laboratory sounds. Tissue sections were prepared for exposure to x-ray film for optical density measurements, and alternate sections stained for identification of nuclei. Measurements from auditory nuclei of experimental animals were corrected against an unaffected control area (abducens nucleus) and compared with measurements taken from animals in the sham group. Auditory evoked potential thresholds to both air- and bone-conducted stimuli were obtained in a second group of neonatal and adult animals. Results: Both CHL and CA resulted in a marked decrease in 2-DG uptake in the major ascending projection of the manipulated ear, in both the neonatal and adult animals. In young animals, effects of CHL and CA were similar. Effects of CHL in adult animals were less marked and significantly different from either effects of CHL in young animals or effects of CA in adult animals. HL following malleus removal only was purely conductive and ranged from 38 to 55 dB across frequency. Conclusions : Results suggest that, particularly in young animals, a unilateral CHL may have profound effects on metabolic activity in the central auditory system.

The modulation of kainate-induced responses by pentobarbitone and the 2,3-benzodiazepine GYKI-53784 (LY303070), a potent non-competitive AMPA antagonist, was studied in vivo using both extracellular recordings of antidromic field potentials and intracellular recordings from abducens motoneurons in ketamine/diazepam-anesthetized rats. In previous studies on pentobarbitone-anesthetized rats [M. Ouardouz, J. Durand, GYKI-52466 antagonizes glutamate responses but not NMDA and kainate responses in rat abducens motoneurons, Neurosci. Lett. 125 (1991) 5-8; M. Ouardouz, J. Durand, Involvement of AMPA receptors in trigeminal postsynaptic potentials recorded in rat abducens motoneurons in vivo, Eur. J. Neurosci. 6 (1994) 1662-1668; A. Ruiz, J. Durand, Blocking the trigeminal EPSPs in rat abducens motoneurons in vivo with the AMPA antagonists, NBQX and GYKI-53655, J. Neurophysiol. (1998) submitted], we showed that 2,3-benzodiazepines do not affect kainate-induced depolarizations in abducens motoneurons. Here, we tested whether pentobarbitone is involved in the pharmacological discrimination by 2,3-benzodiazepines between AMPA- and kainate-induced responses. Kainate-induced depolarizations were reversibly depressed after application of either GYKI-53784 and pentobarbitone. However, kainate-induced depolarizations were not inhibited by GYKI-53784 with pentobarbitone; they were even potentiated sometimes. Using extracellular recordings, we confirmed that in the presence of pentobarbitone, GYKI-53784 counteracts the effects of AMPA but not of kainate on antidromic field potentials in the abducens nucleus. Blockade of kainate-induced responses by GYKI-53784 was reversed with pentobarbitone, which appears relevant to the discrimination between AMPA- and kainate receptor-mediated responses in vivo. In the presence of pentobarbitone, kainate would depolarize motoneurons mainly via kainate receptors since kainate-induced responses were not depressed by 2,3-benzodiazepines. This finding strongly favors the existence of kainate receptors in adult motoneurons but their role is still unknown.

The expression pattern of the human nephroblastoma overexpressed (novH) gene in the fetal human central nervous system was examined by in situ hybridization using digoxigenin-labeled novH-specific riboprobes. In the spinal cord, the nov-expressing neurons were first detected both in the ventral region at 16 weeks of gestation (G16W) and in the dorsal region at G38W. In the medulla, nov-expressing neurons were detected in the principal nucleus of the inferior olive, the hypoglossal nucleus and the dorsal motor nucleus of vagus at G16W. Nov-positive neurons were detected at G28W in the nucleus of the spinal tract of the trigeminal and cuneate nucleus, and at G38W in the abducens nucleus of pons, the red nucleus and the substantia nigra of the midbrain, the ventral posterolateral and the mediodorsal thalamic nucleus. A strong labeling was also detected in the striatum of the cerebrum and the cerebral cortex of the parietal lobe. These data established that novH is mainly expressed in somato-motor neurons in the lower central nervous system at early developmental stages and in the higher central nervous system at later stages, suggesting that nov may play an important role in neuronal differentiation.

Binocular coordination of eye movements is essential for stereopsis (depth perception) and to prevent double vision. More than a century ago, Hering and Helmholtz debated the neural basis of binocular coordination. Helmholtz1 believed that each eye is controlled independently and that binocular coordination is learned. Hering2 believed that both eyes are innervated by common command signals that yoke the eye movements (Hering's law of equal innervation). Here we provide evidence that Hering's law is unlikely to be correct. We show that premotor neurons in the paramedian pontine reticular formation that were thought to encode conjugate3,4,5,6 velocity commands for saccades (rapid eye movements) actually encode monocular commands for either right or left eye saccades. However, 66% of the abducens motor neurons, which innervate the ipsilateral lateral rectus muscle, fire as a result of movements of either eye. The distribution of sensitivity to ipsilateral and contralateral eye movements across the abducens motor neuron pool may provide a basis for learning binocular coordination in infancy and adapting it throughout life.

The synaptic alterations induced in abducens motoneurons by the injection of 3 ng/kg of botulinum neurotoxin type A into the lateral rectus muscle were studied using ultrastructural and electrophysiological techniques. Motoneurons identified by the retrograde transport of horseradish peroxidase showed a progressive synaptic stripping already noticeable by four days post-injection which increased over the study period. By 35 days post-injection, the normal coverage of motoneurons by synaptic boutons (66.4 +/- 4.0%) significantly decreased to 27.2 +/- 4.0%. Synaptic boutons detached by a widening of the subsynaptic space but remained apposed by synaptic contacts and desmosomes to the motoneuron. Detachment did not affect equally flat and round vesicle-containing boutons. The control motoneuron had almost equal numbers of both types of boutons, but after 35 days post-injection the ratio of round to flat vesicle-containing boutons was 1.20 +/- 0.01. Synaptic boutons impinging on motoneurons showed signs of alterations in membrane turnover, as indicated by an increase in the number of synaptic vesicles and a decrease in the number of coated vesicles and synaptic vesicles near the active zone. Abducens motoneurons had a transient increase in soma size by 15 days that returned to normal at 35 days, but no signs of chromatolysis or organelle degeneration were seen. Accompanying the swelling of motoneurons, a 15-fold increase in the number of spines, very infrequent in controls, was observed. Spines located in the soma and proximal dendritic trunk received synaptic contacts from both flat and round vesicle-containing boutons that could be either partly detached or completely attached to the motoneuron. An increased turnover of the plasmatic membrane of the motoneuron was observed, as indicated by a four-fold increase in the number of somatic coated vesicles. Animals were implanted with bipolar electrodes in the ampulla of both horizontal semicircular canals for evoking contralateral excitatory and ipsilateral inhibitory postsynaptic potentials. Motoneurons were antidromically identified from the lateral rectus muscle. Synaptic potentials of vestibular origin were recorded in abducens motoneurons. In the period between two and six days post-injection, a complete abolition of inhibitory synaptic potentials was observed. By contrast, excitatory synaptic potentials remained, but were reduced by 82%. The imbalance between excitatory and inhibitory inputs to motoneurons induced a progressive increase of firing frequency within a few stimuli applied to the contralateral canal. Between 7 and 15 days post-injection, both excitatory and inhibitory postsynaptic potentials were virtually abolished and remained so up to the longest time checked (105 days). Some motoneurons recorded beyond 60 days post-injection showed signs of recovery of excitatory postsynaptic potentials. During the whole time-span studied, presynaptic wavelets were present, indicating no affecting of the conduction of afferent volleys to the abducens nucleus. Taken together, these data indicate that botulinum neurotoxin at high doses causes profound synaptic alterations in motoneurons responsible for the effects seen in the behavior of motoneurons recorded in alert animals.

We have reinvestigated the embryonic development of the vestibulocochlear system in mice using anterograde and retrograde tracing techniques. Our studies reveal that rhombomeres 4 and 5 include five motor neuron populations. One of these, the abducens nucleus, will not be dealt with here. Rhombomere 4 gives rise to three of the remaining populations: the facial branchial motor neurons; the vestibular efferents; and the cochlear efferents. The migration of the facial branchial motor neurons away from the otic efferents is completed by 13.5 days post coitum (dpc). Subsequently the otic efferents separate into the vestibular and cochlear efferents, and complete their migration by 14.5 dpc. In addition to their common origin, all three populations have perikarya that migrate via translocation through secondary processes, form a continuous column upon completion of their migrations, and form axonal tracts that run in the internal facial genu. Some otic efferent axons travel with the facial branchial motor nerve from the internal facial genu and exit the brain with that nerve. These data suggest that facial branchial motor neurons and otic efferents are derived from a common precursor population and use similar cues for pathway recognition within the brain. In contrast, rhombomere 5 gives rise to the fourth population to be considered here, the superior salivatory nucleus, a visceral motor neuron group. Other differences between this group and those derived from rhombomere 4 include perikaryal migration as a result of translocation first through primary processes and only then through secondary processes, a final location lateral to the branchial motor/otic efferent column, and axonal tracts that are completely segregated from those of the facial branchial and otic efferents throughout their course inside the brain. Analysis of the peripheral distribution of the cochlear efferents and afferents show that efferents reach the spiral ganglion at 12.5 dpc when postmitotic ganglion cells are migrating away from the cochlear anlage. The efferents begin to form the intraganglionic spiral bundle by 14.5 dpc and the inner spiral bundle by 16.5 dpc in the basal turn. They have extensive collaterals among supporting cells of the greater epithelial ridge from 16.5 dpc onwards. Afferents and efferents in the basal turn of the cochlea extend through all three rows of outer hair cells by 18.5 dpc. Selective labeling of afferent fibers at 20.5 dpc (postnatal day 1) shows that although some afferents are still in early developmental stages, some type II spiral ganglion cells already extend for long distances along the outer hair cells, and some type I spiral ganglion cells end on a single inner hair cell. These data support previous evidence that in mice the early outgrowth of afferent and efferent fibers is essentially achieved by birth.

Brain-stem afferents to the n. isthmo-opticus (NIO) and ectopic neurons (EN) of the centrifugal visual system (CVS) were determined in the pigeon using the retrograde transneuronal transport of the fluorescent dye Rhodamine β-isothiocyanate (RITC) after its intraocular injection. In other experiments, either RITC was injected into various periocular tissues (controls) or the retrograde tracer Fluoro-gold (FG) was injected stereotaxically in the NIO. Following intraocular injections, the RITC retrograde labeling of cell bodies was observed contralaterally in the NIO and EN and transneuronally in layers 9/10 of the optic tectum, area ventralis-Tsai, zona peri-NIII, mesencephalic and pontine reticular formation (PRF), n. linearis caudalis-raphe, and bilaterally within a region referred to as zona peri-n.NVI (Zp-n.NVI) immediately underlying the abducens nerve nucleus. None of the above structures were labeled after RITC periocular injections. The FG labeling revealed that the tectal efferent neurons were mainly medium-sized, multipolar cells whose dendrites extended superficially to retino-recipient tectal layers 6 and 5. Quantitative measurements of the distribution of layers 9/10 RITC-labeled neurons indicated the highest densities to be localized within the ventral tectum corresponding to the representation of the dorsal retina and inferior visual field. We suggest that visual and nonvisual brain-stem afferents upon NIO and EN may play a role in the proposed mechanism of the avian CVS in attention, ground-feeding behavior, and modulation of retinal sensitivity.

45-year-old man with multiple sclerosis presented with worsening weakness in his right leg and double vision. Neurologic examination revealed horizontal diplopia during lateral gaze in both eyes. The patient had an adduction deficit in the right eye and nystagmus in the left eye on leftward gaze (Panel A). He also had an adduction deficit in the left eye and nystagmus in the right eye on rightward gaze (Panel B). Upward gaze (Panel C), downward gaze (Panel D), and normal primary position (Panel E) were unremarkable (video). Internuclear ophthalmoplegia is characterized by impaired horizontal eye movement that is caused by a lesion in the medial longitudinal fasciculus, a fiber tract that rises from the abducens nucleus in the pons to the contralateral oculomotor nucleus in the midbrain. Lesions in the medial longitudinal fasciculus result in the failure of adduction on attempted lateral gaze. Any brain-stem syndrome can interrupt the medial longitudinal fasciculus and result in impaired horizontal eye movement, but the most frequent underlying cause is multiple sclerosis. This patient had internuclear ophthalmoplegia in both eyes due to demyelinating lesions. Glucocorticoids were administered intravenously, but the deficits did not resolve. On follow-up at 2 months, the patient’s gait had improved, but the internuclear ophthalmoplegia remained.

Intracellular recording of abducens motoneurons in vivo has shown that ionophoretic applications of N-methyl-D-aspartate produced long-lasting membrane potential oscillations including a slow depolarization plateau with a burst of fast action potentials. This complex N-methyl-D-aspartate pattern was reproduced in the model of abducens motoneuron in vivo identified, intracellularly stained with horseradish peroxidase and reconstructed at high spatial resolution. The excitable soma of the simulated cell contained voltage-gated Ca, Na and K conductances, N-methyl-D-aspartate-gated voltage-sensitive Ca-Na-K conductance and Ca-dependent K conductance. The dendrite was passive either completely or with the exception of branching nodes containing N-methyl-D-aspartate conductances of the same slow kinetics but of lower values than at the soma. In the completely passive case, the N-methyl-D-aspartate pattern decayed with different rates along different dendritic paths depending on the geometry and topology of the reconstructed dendrite. The branches formed four clusters discriminated in somatofugal attenuations of steady voltages, and were correspondingly discriminated in attenuation of the complex N-methyl-D-aspartate pattern. Fast spikes decayed more than the slow depolarization plateau so that the prevalence of slow over fast components in the transformed pattern increased with somatofugal path distance. As a consequence, the lower the electrotonic effectiveness of a branch in the cluster or in the whole arborization, the lower both the voltage level and the frequency range of its voltage modulation by N-methyl-D-aspartate oscillations. In the case of active branching points, the somatic pattern changed depending on the level of activation of dendritic N-methyl-D-aspartate conductances with slow kinetics of voltage sensitivity. The higher this level, the longer the plateau and burst, and the greater the discharge rate; and the spikes in the burst were smaller. When the pattern spread in the dendrite, the fast spikes decayed and the slow plateau was boosted, with a greater effect along the somatofugal path containing more branching points. These results show how the somatofugal back-invasion along the dendrites by activity patterns generated at the soma can tune voltage-sensitive dendritic conductances. The dendritic back-invasion is geometry- and topology-dependent. It is proposed as a subtle feedback mechanism for the neuron to control its own synaptic inputs.

The synaptic organization of the saccade-related neuronal circuit between the superior colliculus (SC) and the brainstem saccade generator was examined in an awake monkey using a saccadic, midflight electrical-stimulation method. When microstimulation (50-100 microA, single pulse) was applied to the SC during a saccade, a small, conjugate contraversive eye movement was evoked with latencies much shorter than those obtained by conventional stimulation. Our results may be explained by the tonic inhibition of premotor burst neurons (BNs) by omnipause neurons that ceases during saccades to allow BNs to burst. Thus, during saccades, signals originating from the SC can be transmitted to motoneurons and seen in the saccade trajectory. Based on this hypothesis, we estimated the number of synapses intervening between the SC and motoneurons by applying midflight stimulation to the SC, the BN area, and the abducens nucleus. Eye position signals were electronically differentiated to produce eye velocity to aid in detecting small changes. The mean latencies of the stimulus-evoked eye movements were: 7.9 +/- 1.0 ms (SD; ipsilateral eye) and 7.8 +/- 0.9 ms (SD; contralateral eye) for SC stimulation; 4.8 +/- 0.5 ms (SD; ipsilateral eye) and 5.1 +/- 0.7 ms (SD; contralateral eye) for BN stimulation; and 3.6 +/- 0.4 ms (SD; ipsilateral eye) and 5.2 +/- 0.8 ms (SD; contralateral eye) for abducens nucleus stimulation. The time difference between SC- and BN-evoked eye movements (about 3 ms) was consistent with a disynaptic connection from the SC to the premotor BNs.

✓ The authors reviewed the results of continuous intraoperative electromyographic (EMG) monitoring of muscles innervated by cranial nerves in 17 children whose preoperative imaging studies showed compression or infiltration of the fourth ventricular floor by tumor to determine how intraoperative EMG activity correlated with postoperative cranial nerve morbidity. Bilateral lateral rectus (sixth) and facial (seventh) nerve musculatures were monitored in all children. Cranial nerve function was documented immediately postoperatively and at 1 year. Of the 68 nerves monitored, nine new neuropathies occurred in six children (sixth nerve in four children and seventh nerve in five). In five new neuropathies, intraoperative EMG activity could be correlated in one of four sixth nerve injuries and four of five seventh nerve injuries. Electromyographic activity could not be correlated in four children with new neuropathies. Of 59 cranial nerves monitored that remained unchanged, 47 had no EMG activity. Twelve cranial nerves (three sixth nerves and nine seventh nerves) had EMG activity but no deficit. Of four children with lateral rectus EMG activity, three had new seventh nerve injuries. Lateral rectus EMG activity did not predict postoperative abducens injury. The absence of lateral rectus EMG activity did not assure preserved abducens function postoperatively. Likely because of the close apposition of the intrapontine facial nerve to the abducens nucleus, lateral rectus EMG activity was highly predictive of seventh nerve injury. Although facial muscle EMG activity was not an absolute predictor of postoperative facial nerve dysfunction, the presence of facial muscle EMG activity was associated statistically with postoperative facial paresis. The absence of facial muscle EMG activity was rarely associated with facial nerve injury. The authors speculate that EMG activity in the facial muscles may have provided important intraoperative information to the surgeon so as to avoid facial nerve injury.

The projection from the nucleus prepositus hypoglossi (PH) to the superior colliculus (SC) has been proposed to provide a feedback control of collicular saccadic activities. The present study aimed to identify the functional properties of PH neurones projecting to the SC relative to eye movement parameters. Preposito-collicular neurones were identified in alert cats by antidromic invasion and collision tests following electrical stimulations of the contralateral SC. Their discharges were then correlated with the horizontal component of eye movements. Particular attention was given to the timing of discharges relative to saccade onsets. Most preposito-collicular neurones (12/14) displayed transient activities associated to eye velocity, and onsets preceded the saccade onset by 9.4-19.9 ms. The mean eye velocity sensitivity of these "early" preposito-collicular neurones (1.46 +/- 0.53 spikes/s per degree per second) was quite similar to that calculated from a sample of putative motoneurones or interneurones that have been recorded within abducens nucleus and quantified in the same conditions. The remaining two preposito-collicular neurones exhibited transient activity related to saccades, but this followed the transient putative motoneuronal discharge. These "delayed" neurones also had lower eye velocity sensitivities (0.38 sp/s per degree per second and 0.58 sp/s per degree per second, respectively) compared with early neurones. Both classes of preposito-collicular neurones also displayed a subsequent tonic activity correlated with the eye position. Taken together, these results demonstrate that preposito-collicular neurones code both eye position and eye velocity just like ocular motoneurones, but in a predictive manner. The anticipatory discharge of early neurones makes them likely candidates for the control of peak activities of saccade-related collicular neurones, particularly in the caudal colliculus. Delayed preposito-collicular neurones may also participate in the control of collicular activities, but probably in more rostral SC, where peak activities occur later during eye movements together with smaller motor error coding.

1. The discharge patterns and axonal projections of saccadic long-lead burst neurons (LLBNs) with somata in the pontine reticular formation were studied in alert squirrel monkeys with the use of the method of intraaxonal recording and horseradish peroxidase injection. 2. The largest population of stained neurons were afferents to the cerebellum. They originated in the dorsomedial nucleus reticularis tegmenti pontis (NRTP) including its dorsal cell group (N = 5), the preabducens intrafascicular nucleus (N = 5), and the raphe pontis (N = 1). Axons of all neurons coursed under NRTP and entered brachium pontis without having synapsed in the brain stem. Three axons sent collaterals to the floccular lobe, but other more distant targets of these and the other cerebellar afferents could not be determined. Movement fields of these neurons were intermediate between vectorial and directional types. 3. Four neurons had their somata in nucleus reticularis pontis oralis and terminations in the brain stem reticular formation. Each neuron was different, but all terminated in the region containing excitatory burst neurons, and most terminated in the region containing inhibitory burst neurons. Other targets include nucleus reticularis pontis oralis and caudalis, NRTP, raphe interpositus, and the spinal cord. Discharge patterns included both vectorial and directional types. 4. Two reticulospinal neurons had large multipolar somata either just rostral or ventral to the abducens nucleus. These neurons also projected to the medullary reticular formation, caudal nucleus prepositus hypoglossi, and dorsal and ventral paramedian reticular nucleus. 5. The functional implications of the connections of these LLBNs and those reported in the companion paper are extensively discussed. The fact that the efferents of the superior colliculus target the regions containing medium-lead saccadic burst neurons confirms the role of the colliculus in saccade generation. However, the finding that many other neurons project to these regions and the finding that superior colliculus efferents project more heavily to areas containing reticulospinal neurons argue for a diminished role of the superior colliculus in saccade generation but an augmented role in head movement control.

Orphan transporter v7-3 is a member of a new subfamily of Na+, Cl− dependent neurotransmitter transporters with two large extracellular loops. Distribution of v7-3 mRNA was investigated in the rat brain. In situ hybridization study revealed that v7-3 mRNA was widely distributed in the rat central nervous system, including the olfactory bulb, the hypothalamus, the cerebral cortex, the hippocampus, and the cerebellum. In addition, intense v7-3 mRNA expression was found in the motor nuclei including the oculomotor nucleus, abducens nucleus, trigeminal motor nucleus, facial nucleus, hypoglossal nucleus and ventral horn of spinal cord. Intense hybridization signals were also observed in the nuclei containing monoaminergic neurons, such as locus coeruleus, the substantia nigra pars compacta, the ventral tegmental area, the dorsal raphe nucleus and the median raphe nucleus. This multifocal and broad nature of the v7-3 distribution suggests widespread roles for this gene product in neurons mediating several important brain function.

The coexistence of gamma-aminobutyric acid (GABA) and glycine in axon terminals impinging on rat abducens motoneurones was investigated using a double staining procedure combining retrograde labelling of the motoneurones with HRP and post-embedding immunocytochemical staining of axon terminals. Adjacent ultrathin sections of cell bodies of identified motoneurones were individually treated with GABA or glycine antibodies. The terminals single labelled for GABA represented 11.4% of the terminals analyzed, while 8% of them were glycine immunoreactive and 9% were both GABA and glycine immunoreactive. All the labelled terminals contained pleomorphic vesicles. The mean length of apposition of the double labelled terminals was statistically larger (2.20 +/- 0.97 microns) than the GABA (1.65 +/- 0.57 microns) or glycine immunoreactive ones (1.37 +/- 0.35 microns).

Effects of stimulation of the superior colliculus on saccade-related brain stem neurons were studied in the alert cat. Extracellular recordings were made from medium-lead burst neurons (MLBNs), omnipause neurons (OPNs) and burster-driving neurons (BDNs) in the paramedian pontomedullary region rostral and caudal to the abducens nucleus. MLBNs were activated from the contralateral superior colliculus with monosynaptic latencies when single-pulse stimulation was given during saccades or ipsilateral head rotation, although this activation was not observed during fixation periods. The caudal SC was more effective than the rostral SC in monosynaptic activation of MLBNs. Most OPNs were also activated monosynaptically from the SC. In constrast to MLBNs, the activation of OPNs was more frequently induced from the rostral SC than from the caudal SC. Stimulation of the caudal SC often induced suppression of spikes in OPNs. BDNs received excitation from the ipsilateral SC through a di- or trisynaptic pathway. Like MLBNs, BDNs tended to receive stronger input from the caudal SC than the rostral SC. Results indicate the existence of tectofugal excitatory pathways to MLBNs and BDNs and an inhibitory pathway to OPNs. It seems likely that these pathways originate from saccade-related burst cells in the SC. Since excitation of BDNs and inhibition of OPNs increase the excitability of MLBNs, all of these pathways may contribute to burst activity in MLBNs and thereby saccade generation. Results also support the current idea that cells in the rostral SC may participate in fixation by activating OPNs.

The pattern of axonal terminations of individual premotoneuronal medium lead burst neurons (MLBs) has been elucidated with the help of the intraaxonal recording and horseradish peroxidase injection technique in alert behaving monkeys. These findings indicate that individual MLBs do not influence individual muscles; instead they influence groups of muscles. Horizontal medium lead burst neurons (EBNs) project to ipsilateral lateral rectus motoneurons directly, and could contact contralateral medial rectus motoneurons indirectly, through the internuclear neurons of the ipsilateral abducens nucleus. Also, upward MLBs project to motoneurons innervating both the superior rectus and the inferior oblique muscles of both eyes. Finally, downward MLBs project to ipsilateral motoneurons innervating the ipsilateral inferior rectus muscle and the contralateral superior oblique muscle. All in all, the pattern of oculomotor terminations of MLBs provides experimental support for Hering's law of equal innervation. EBNs also project to the nucleus prepositus hypoglossi while vertical MLBs also project to the interstitial nucleus of Cajal (NIC). These structures are thought to participate in the process of “velocity to position integration”, in the horizontal and vertical planes respectively. Intracellular recording from individual neurons of the NIC in alert behaving monkeys followed by biocytin or horseradish peroxidase injections demonstrates that their axons pass through the posterior commissure on their way to vertical extraocular motoneurons. It also demonstrates that these fibers carry a phasic signal related to saccades and a tonic signal related to eye position in a precise quantitative manner. Work is in progress to determine whether their pattern of oculomotor terminations is also appropriate for Hering's law.

1. We recorded single vestibular nucleus neurons shown by electrical stimulation to receive floccular inhibition [flocculus receiving neurons (FRNs)] and/or to project toward midbrain motoneuronal pools [midbrain projecting neurons (MPNs)] in awake, head-fixed rabbits during compensatory eye movements. Stimuli included head rotation in the light, head rotation in the dark, and rotation of an optokinetic drum about the animal. We employed sinusoidal and triangular position profiles in the 0.05- to 0.8-Hz frequency band. We also examined transient responses to step changes in eye position. 2. We found identified vestibular nucleus cells (i.e., FRN/non-MPNs, FRN/MPNs, and non-FRN/MPNs) in the parvocellular and magnocellular portions of the medial vestibular nucleus, at the rostrocaudal level of the dorsal acoustic stria. 3. All identified vestibular nucleus neurons were excited during ipsilateral (relative to side of recording) head rotation and contralateral eye rotation. 4. The neuronal firing rates could be related to eye position and its time derivatives, and that relationship could be approximated by a two-pole, one-zero linear transfer function. As with abducens neurons, a more detailed approximation requires inclusion of two nonlinearities-a hysteresis and a variable sensitivity term that increases as eye movement amplitude decreases. 5. When the vestibuloocular reflex is suppressed by a conflicting full-field visual stimulus [visual vestibular conflict condition (VVC)], vestibular nucleus neuron modulation is largely suppressed. The remaining modulation is motoric in nature, because it can be related to the residual eye movements. Cells with "sensory vestibular signals," i.e., cells whose modulation during VVC correlates better with head rotation than eye movement, were not encountered. 6. We examined the dependence of firing rate parameters on stimulus modality. All neurons exhibited increased phase lead with respect to abducens nucleus neurons during stimuli involving head rotation. This finding could indicate that vestibular-derived inputs are inhomogeneously distributed on premotor neurons and that the studied premotor population receives a stronger vestibular input than another premotor group, not recorded in the current experiments. 7. FRNs and non-FRNs were similar in their qualitative response to the fast phases, the applicability of the two-pole, one-zero transfer function, hysteresis, and the amplitude nonlinearity. 8. FRNs differed from non-FRNs in having a phase advanced firing rate at all stimulus frequencies during visual and vestibular stimuli. The phase difference suggests that one role of the rabbit flocculus is to regulate phase of the net premotor signal.

1. We recorded abducens neurons, identified by electrical stimulation as internuclear neurons or motoneurons, in awake rabbits. The relationship of firing rate to eye movement was determined from responses during stable fixations, sinusoidal rotation in the light (0.05-0.8 Hz), and triangular optokinetic stimulation at 0.1 Hz. 2. All abducens neurons were excited during temporal movement of the ipsilateral eye. Temporal and nasal saccades were associated with bursts or pauses, respectively, in the firing rate. 3. Motoneurons and internuclear neurons are qualitatively indistinguishable. There was no significant quantitative difference between the phase and sensitivity of the two groups for 0.2-Hz sinusoidal rotation in the light. 4. On the basis of the response to stable eye positions, we determined static eye position sensitivity of the abducens neuron pool to be 8.2 +/- 2.5 (SD) spikes.s-1/0, with a static hysteresis of 8.9 spikes/s (1.14 +/- 0.37 degrees). 5. We determined apparent eye position sensitivity (k) and apparent eye velocity sensitivity (r) from the responses to sinusoidal rotation in the light. k increases and r decreases with stimulus frequency, which indicates that the simplest transfer function mediating conversion of abducens nucleus (VI) firing rate to eye position (E) has two poles and one zero. 6. The VI-->E relationship has an "amplitude nonlinearity," manifest as a tendency for k, r, and firing rate phase lead to decrease as eye movement amplitude increases at a fixed frequency. On a percentage basis, phase is less affected than are the sensitivities. The nonlinearity becomes less pronounced for stimulus amplitudes > 2.5 degrees, and consequently a linear model of the VI-->E transformation remains useful, provided that consideration is restricted to the appropriate range of stimulus/response amplitudes. 7. We determined time constants of the linear two-pole, one-zero transfer function from the variation of r/k versus stimulus frequency. The pole time constants were T1 = 3.4 s and T2 = 0.28 s, and the zero time constant (Tz) = 1.6 s. The magnitude of Tz was corroborated by measuring the time constant of the exponential decay in firing rate after step changes in eye position. This transient method yielded a Tz of 1.1 s. 8. The time constants of the VI-->E transfer function are roughly 10 times larger than those reported for the rhesus macaque. The difference is attributable to the reported 10-fold lower stiffness of the rabbit oculomotor plant, which may in turn relate to rabbits postulated lower degree of activation of extraocular muscles at any given position.(ABSTRACT TRUNCATED AT 400 WORDS)

Studies of the pathways involved in the vestibulo-ocular reflex have suggested that the projection from the superior vestibular nucleus to the ipsilateral oculomotor nucleus is inhibitory, whereas the medial vestibular nucleus, the abducens nucleus and the contralateral superior vestivular nucleus most likely exert excitatory effects on oculomotor neurons. In order to determine directly the termination pattern and the neurotransmitter of these afferents, we studied their input to the oculomotor nucleus in the rabbit at the light microscopic level with the use of anterograde tracing of Phaseolus vulgaris -leucoagglutinin combined with retrograde tracing of horseradish peroxidase from the extraocular muscles, and at the ultrastructural level with the use of anterograde tracing of wheatgerm-agglutinated horseradish peroxidase combined with GABA and glycine postembedding immunocytochemistry. The general ultrastructural characteristics of the neuropil and the types of boutons observed in the rabbit oculomotor nuclei are in general agreement with the descriptions for the oculomotor complex of other mammals. The superior vestibular nucleus projected bilaterally to the superior rectus and inferior oblique subdivisions, and ipsilaterally to the inferior rectus and medial rectus subdivision; the medial vestibular nucleus projected bilaterally to the medial rectus, inferior oblique, inferior rectus and superior rectus subdivisions with a strong contralateral predominance. The abducens nucleus projected contralaterally to the medial rectus subdivision. More than 90% of all the anterogradely labled terminals from the ipsilateral superior vestibular nucleus were GABAergic. These terminals were characterized by flattened vesicles and symmetric synapses, and they contacted somata, as well as proximal and distal dendrites of motoneurons. All terminals derived from the medial vestibular nucleus the abducens nucleus and the contralateral superior vestibular nucleus were non-GABAergic. These non-GABAergic terminals showed spherical vesicles and asymmetric synapses, and they contacted predominantly distal dendrites. None of the anterogradely labeled terminals from the studied vestibular nuclei or abducens nucleus were glycinergic. The present study provides the first direct anatomical evidence that most, if not all, of the synaptic input from the superior vestibular nucleus to the ipsilateral oculomotor nucleus is GABAergic, and that the medial rectus subdivision is included in the termination area. Furthermore, it confirms that the projections from the medial vestibular nucleus, the abducens nucleus and the contralateral superior vestibular nucleus are exclusively non-GABAergic.

The horizontal vestibulo-ocular reflex (HVOR) and vestibulo-collic reflex (HVCR) contribute to gaze stabilization during head movements in the horizontal plane (Baker et al., 1981; Berthoz, 1989). During head rotations, sensory inputs from the horizontal semicircular canals of the labyrinth modulate the discharge of the first-order vestibular neurons in proportion to head velocity (Curthoys, 1982; Fernandez and Goldberg, 1971). These primary afferents contact monosynaptically the second-order vestibular neurons located in the central vestibular nuclei, which in turn project to the prepositus hypoglossi neurons, and to the appropriate extraocular and spinal motoneuron pools in order to implement the vestibuloocular and vestibulo-spinal reflexes. Whereas part of the second-order vestibular neurons are inhibitory and project through ipsilateral pathways (reviewed in Uchino and Isu, 1992a), another subgroup of second-order vestibular neurons are excitatory and project through contralateral pathways (Berthoz et al., 1989; Iwamoto et al., 1990; McCrea et al., 1987; Uchino et al., 1981,1982; Uchino and Isu, 1992b). According to their axonal projections to the extraocular and neck motor nuclei, the excitatory second-order vestibular neurons have been classified into three distinct groups (reviewed in Uchino and Isu, 1992b): the vestibuloocular neurons, which project exclusively towards cells within the contralateral abducens nucleus, the vestibulo-collic neurons, which project exclusively towards contralateral neck and spinal motoneurons and, finally, the vestibulo-oculo-collic neurons, which project contralaterally towards both the abducens and neck motor nuclei (Isu and Yokota, 1983;McCrea et al.,1987;Uchino et al.,1982).

1. In the companion article, a variety of head movements were elicited by stimulation in, and adjacent to, the gigantocellular reticular nucleus (Cowie and Robinson 1994). We refer to this area, caudal to the abducens nucleus, as the gigantocellular head movement region. In the present paper, the anatomical connections of this region, as determined by injections of wheat-germ agglutinin conjugated horseradish peroxidase (WGA-HRP), are reported. The majority of efferent and afferent connections were with areas related to head movements. 2. Efferent fibers from the region projected via two paths to the caudal medulla and upper cervical spinal cord. Labeled fibers descended in the anterolateral funiculus of the ipsilateral spinal cord to terminate in lateral parts of the ventral horn. A second pathway descended bilaterally in the medial longitudinal fasciculus to the anterior funiculi and medial portions of the ventral gray. These efferents paralleled the head-movement topography demonstrated physiologically. Other projections included efferents to the interstitial nucleus of Cajal, caudal field H of Forel, paramedian pontine reticular formation, and caudal vestibular nuclei. Other efferent fibers projected to the trigeminal, facial, and hypoglossal nuclei, as well as to the parvocellular reticular field, which contains interneurons for these motor groups. However, no efferent or afferent labeling involved the ocular motor nuclei. 3. Afferents to the gigantocellular head movement region arose mainly from head-movement areas. In all animals, labeled cells were found in the intermediate and deep layers of the caudal superior colliculus. Labeled neurons also were found in the caudal field H of Forel, interstitial nucleus of Cajal, pontine medial tegmentum including the pontine paramedian reticular formation, nucleus subcoeruleus, and vestibular nuclear complex. Caudally, filled cells were located in the parvocellular, magnocellular, dorsal, and ventral reticular nuclei, the supraspinal nucleus, and the upper cervical ventral horn. 4. In one animal, the ipsilateral frontal cortex contained retrogradely labeled neurons. These cells were found in layer V of cortical areas 4 and 6. Other afferent cells were found consistently in the periventricular and periaqueductal gray matter. 5. A control injection into the caudal vestibular nuclear complex showed projections to the gigantocellular reticular formation and labeled cells in the vestibular and parvocellular reticular nuclei. These observations show that the connections of the gigantocellular region are not typical of all head movement sites. 6. These data indicate that the gigantocellular head-movement region has the requisite efferent and afferent connections to function in the subcortical control of head, but not eye, movements.(ABSTRACT TRUNCATED AT 400 WORDS)

In elasmobranch fishes, including the Atlantic stingray, the medial rectus muscle is innervated by the contralateral oculomotor nucleus. This is different from most vertebrates, in which the medial rectus is innervated by the ipsilateral oculomotor nucleus. This observation led to the prediction that the excitatory vestibulo-extraocular motoneuron projections connecting each semicircular canal to the appropriate muscle should use a contralateral projection from the vestibular nuclei to the motoneurons. This hypothesis was examined in the Atlantic stingray by injecting horseradish peroxidase unilaterally into the oculomotor nucleus. It was found that vestibulo-oculomotor projections arise from the ipsilateral anterior octaval nucleus and the contralateral descending octaval nucleus. The same pattern was observed when the trochlear nucleus was involved in the injection. There were no cells labeled in the region of the abducens nucleus, and no candidate for a nucleus prepositus hypoglossus was identified. The presence of compensatory eye movements, the directional sensitivity of the semicircular canals, the location of the motoneurons innervating each eye muscle, and our results indicate that the excitatory input to the extraocular motoneurons is derived from the contralateral descending octaval nucleus, and the inhibitory input is derived from the ipsilateral anterior octaval nucleus. The absence of both abducens internuclear interneurons and a nucleus prepositus hypoglossus suggests that eye movements, particularly those in the horizontal plane, are controlled differently in elasmobranchs than in other vertebrates examined to date.

The physiological signals and patterns of synaptic connectivity that CNS neurons display after the loss of their target cells were evaluated in adult cats for one year. Abducens internuclear neurons were chosen as the experimental model because of their highly specific projection onto the medial rectus motoneurons of the oculomotor nucleus. Selective death of medial rectus motoneurons was induced by the injection into the medial rectus muscle of ricin, a potent cytotoxic lectin that leaves the presynaptic axons intact. The electrical activity of antidromically identified abducens internuclear neurons was recorded in chronic alert animals, during both spontaneous and vestibularly induced eye movements, before and after target removal. During the three weeks that followed ricin injection, abducens internuclear neurons exhibited several firing-related abnormal properties. There was an overall reduction in firing rate with a corresponding increase in the eye position threshold for recruitment. In addition, neuronal sensitivities to eye position and velocity were significantly decreased with respect to control data. Bursting activity was also altered since low-frequency delayed burst accompanied the saccades in the on-direction and, occasionally, internuclear neurons exhibited low-frequency discharges associated with off-directed saccades. Intracellular recordings carried out seven and 15 days after ricin injection demonstrated no significant changes in their electrical properties, although a marked depression of synaptic transmission was evident. The amplitude of both excitatory and inhibitory postsynaptic potentials of vestibular origin was reduced by 60–85% with respect to controls. However, postsynaptic potentials recorded one month after ricin injection showed normal amplitude values which persisted unaltered one year after target loss. Recovery of synaptic transmission occurred at the same time as the re-establishment of normal eye-related signals in the discharge pattern of abducens internuclear neurons recorded in alert cats from days 25–30 post lesion. The functional restoration of firing properties was maintained in the long term (one year). Conversely, abducens motoneurons showed normal firing and synaptic patterns at all time intervals analysed. These results demonstrate that, after an initial period of altered physiological properties, abducens internuclear neurons survive the loss of their target motoneurons and regain a normal discharge pattern and afferent synaptic connections.

The twitch fibers, orSIFs, are the type of muscle fibers that constitute allskeletal muscles, i.e., they respond to electrical excita-tion with an all-or-nothing response that propagatesalong the whole length of the fiber. They are innervatedby large en plaque motor endplates in an endplate zoneoccupying the central third of the muscle. The MIFs arehighly unusual in mammals, occurring only in eyemuscles, the larynx, and muscles of the middle ear. Thefibers are fatigue-resistant and respond to electricalstimulation with a slow tonic contraction, which is notpropagated along the muscle fiber (they are non-twitch).The motor endplates are typically small and are distrib-uted all along the length of the fiber. At the distal tip ofthe eye muscle fiber, as it inserts into the tendon, theglobal layer MIFs are capped by a tangle of nerve ter-minals called palisade endings, or myotendinous cylin-ders.

1. In order adequately to control eye movements, oculomotoneurones have to be supplied with both an eye-velocity signal and an eye-position signal. However, all the command signals of the oculomotor system are velocity signals. Nowadays, there is general agreement about the existence of a brainstem network that would convert velocity command-signals into an eye-position signal. This circuit, because of its function, is called the oculomotor neural integrator. The most obvious symptom of its eventual failure is a gaze-holding deficit: in this case, saccades are followed by a centripetal post-saccadic drift. Although the oculomotor neural integrator is central in oculomotor theory, its precise location is still a matter for debate. 2. Previously, microinjections of kainic acid (KA) into the region of the nucleus prepositus hypoglossi (NPH) and of the medial vestibular nucleus (MVN) were found to induce a horizontal gaze-holding failure both in the cat and in the monkey. However, the relatively large volumes (1-3 microliters) and concentrations (2-4 micrograms microliters-1) used in these injections made it difficult to know if the observed deficit was due to a disturbance of the NPH or of the nearby MVN. These considerations led us to inject very small amounts of kainic acid (50 nl, 0.1 microgram microliter-1) either into the rostral part of the MVN or into different sites along the NPH of the cat. 3. The search coil technique was used to record (1) spontaneous eye movements (2) the vestibulo-ocular reflex (VOR) induced by a constant-velocity rotation (50 deg s-1 for 40 s) and the optokinetic nystagmus (OKN) elicited by rotating an optokinetic drum at 30 deg s-1 for 40 s. 4. In each injection experiment, the location of the abducens nucleus of the alert cat was mapped out by recording the antidromic field potentials evoked by the stimulation of the abducens nerve. Two micropipettes were then glued together in such a way that when the tip of the recording micropipette was in the centre of the abducens nucleus the tip of the injection micropipette was in a target area. The twin pipettes were then lowered in the brainstem until the recording micropipette reached the centre of the abducens nucleus. Kainic acid was then injected into the brainstem of the alert cat through the injection micropipette by an air pressure system. 5. Carried out according to such a protocol, KA injections into the NPH or the rostral part of the MVN consistently led to specific eye-movement changes.(ABSTRACT TRUNCATED AT 400 WORDS)

Rat abducens motoneurons were intracellularly recorded in vivo during synaptic excitation and extracellular microionophoretic application of N-methyl-d-aspartate (NMDA). Trigeminal excitatory post-synaptic potentials (EPSPs) evoked in abducens motoneurons were studied during intracellular current injection. They were not sensitive to hyperpolarization or depolarization of the membrane potential in the range of –75 mV to –55 mV using current pulse intensities between –3 nA and + 1 nA. Microionophoretic applications of aminophosphonovalerate (APV), MK801 and i.v. injections of MK801 (1 – 3 mg/kg) or ketamine (10 mg/kg) did not modify trigeminal EPSPs, suggesting that NMDA receptors are not involved in this synaptic transmission. However, microionophoretic applications of NMDA on abducens motoneurons enhanced trigeminal EPSPs and gave rise to regenerative oscillations. The co-activation of NMDA receptors and trigeminal synapses induced these oscillations. The trigeminal EPSP may delay and reset the oscillations depending on where it was evoked in the oscillatory cycle. Depolarizing current pulses intracellularly applied to abducens motoneurons could trigger a post-hyperpolarization followed by rebound depolarization during NMDA application, confirming the activation of active membrane properties. However, depolarizing current pulses could not trigger oscillations similar to those entrained by the EPSPs. The importance of the location of trigeminal synapses in relation to those of NMDA receptors in the dendritic arborization of abducens motoneurons is discussed. Our results show that the same sensory stimulus may have different post-synaptic effects on abducens motoneurons during the co-activation of NMDA receptors. A complete modification of the motor output during NMDA receptor activation strongly supports an active role of abducens motoneurons provided that NMDA receptors are physiologically activated during motor pattern generation.

1. As far as horizontal eye movements are concerned, the well-known hypothesis of a common neural integrator states that the eye-position signal is generated by a common network, regardless of the type of versional movement. The aim of this study was to evaluate the validity of this hypothesis by analysing the behaviour of the abducens motoneurones, the system into which the horizontal neural integrator(s) project(s). If there were a common neural integrator, the different motoneurones would receive the eye position signal through the same pathway and the sensitivity to eye position would be the same regardless of the type of versional movement. If there were multiple integrators, the sensitivity to eye position in one type of versional movement might be different from the sensitivity to eye position in another type of versional movement, at least for occasional motoneurones. 2. The discharge of thirty-one antidromically identified abducens motoneurones was recorded in the alert cat during spontaneous eye movements made in the light and in response to sinusoidal rotations of the head in complete darkness. 3. All of the abducens motoneurones exhibited a burst of action potentials for lateral saccades. During fixation between saccades, they maintained a steady firing rate that increased as the cat fixated increasingly lateral eye positions. 4. For each abducens motoneurone, the sensitivity to eye position (Kf) was determined from measurements carried out during intersaccadic fixations. Kf was calculated from the slope of the firing rate-eye position linear regression line. 5. The discharge rate of the identified motoneurones was observed during four sinusoidal vestibular stimulations (+/- 10 deg, 0.10 Hz; +/- 20 deg, 0.10 Hz; +/- 30 deg, 0.10 Hz; +/- 40 deg, 0.10 Hz). The motoneurones exhibited a burst of activity during fast phases in the lateral direction and paused during fast phases in the opposite direction. During slow phases, motoneurones modulated their activity as a function of the vestibularly induced eye movements except for slow phases that occurred in position ranges below their recruitment threshold. In these cases their activity was cut off. 6. A new method was developed to measure the sensitivity to eye position of neurones during vestibular slow phases. The difficulty came from the fact that, during slow phases, eye velocity and eye position changed simultaneously and that each of those two variables could influence neuronal activity.(ABSTRACT TRUNCATED AT 400 WORDS)

The retrograde tracer wheat germ agglutinin-conjugated horseradish peroxidase was used to establish the organization of the extraocular muscle motoneuron pools in a prosimian, Galago senegalensis, for comparison with the organization in monkeys and non-primates. Medial rectus motoneurons were distributed in three subgroups in the ipsilateral oculomotor nucleus, a pattern similar to that of the monkey. Furthermore, the other component of the near response system, the preganglionic parasympathetic motoneurons, were confined within the Edinger-Westphal nucleus, as in the monkey. In contrast, the distribution of the levator palpebrae and superior rectus motoneurons was similar to that of the cat. Specifically, the majority of levator palpebrae motoneurons were located contralaterally, in the caudal central subdivision of the oculomotor nucleus, and the superior rectus motoneurons had a dorsocaudal location in the contralateral oculomotor nucleus. The distributions of motoneurons supplying the superior oblique and lateral rectus muscles were similar to those of other mammals. Unlike previously studied species, the galago was found to have two accessory muscles, that lie beneath the medial and lateral rectus muscles. Motoneurons supplying the accessory rectus muscles were found ventrolateral to the main abducens nucleus, in a position similar to that occupied by the cat accessory abducens nucleus; although others may be present in the main nuclei. Taken together, these results suggest that the organization of extraocular and preganglionic motoneurons in the galago exhibits both monkey and non-primate features. These observations are consistent with the notion that the galago is a primate species whose oculomotor organization is more similar to the general mammalian scheme. © 1993 Wiley-Liss, Inc.

The electrical activity of antidromically identified abducens internuclear neurons selectively deprived of their target motoneurons was recorded in chronic alert cats. Target motoneurons were killed by the injection of the cytotoxic lectin of Ricinus communis into the medial rectus muscle. Following target removal, the discharge pattern of abducens internuclear neurons showed an overall decrease in firing rate, a significant reduction in their sensitivity to eye position and velocity, and the presence of anomalous responses such as bursts of spikes associated with off-directed saccades. The decreased excitability of abducens internuclear neurons correlated well with a marked reduction in the synaptic efficacy of their inputs. Thus, both excitatory and inhibitory synaptic potentials of vestibular origin showed a noticeable decrease in amplitude. The alterations in firing properties and synaptic transmission were only observed during an initial period of 3 weeks following ricin injection. Within 1 month the electrophysiological parameters returned to control values and remained unaltered for 1 year. Retrograde labelling of abducens internuclear neurons revealed that no cell death occurred after target loss. The anterograde axonal labelling of these neurons showed a progressive decrease in the density of their axonal terminals, and no sign of redistribution to other areas was found. These findings indicate that abducens internuclear neurons are not dependent on the presence of their natural target cells, either for the survival or for the maintenance of appropriate physiological signals.
