Abstract

Particularly in the later stages of his career, Glenn Gould hoped to create large-scale structural coherence in multi-movement works through his choice of tempi. In a scripted conversation with music critic Tim Page, Gould described the “rhythmic reference point” in his 1981 recording of Bach’s “Goldberg Variations.” Gould’s process in the recording studio did not involve comparison of discrete sections of music; Gould listened to or played along with the end of the previous variation to set the tempo of the following variation. Empirical analysis of Gould’s 1981 recording shows how the processual logic of Gould’s execution contrasts with the architectural emphasis of the theoretical literature on proportional tempo. In this recording, proportional tempo relationships depend on large-scale slowing across entire movements. Despite being common in Gould’s recordings, these tempo shapes have not previously been described in the scholarly or popular literature; I argue that listeners do not perceive them as tempo per se, but as structure. This heard sense of structure is created in an area underdetermined by Bach’s notation and is based almost entirely on Gould’s decisions as a performer. 225. Evaluation of drug-drug interactions in hospitalized patients on medications for OUD

Abstract

Introduction: Medications used to treat OUD have common metabolic pathways and pharmacodynamic properties that can lead to drug-drug interactions (DDIs) that may go unnoticed in the inpatient setting. The purpose of this study was to identify the frequency of DDIs between medications prescribed for OUD and commonly used inpatient medications. Methods: This was a retrospective review of orders for buprenorphine, buprenorphine-naloxone, and methadone to identify potential DDIs. Adult inpatients with an order for one of these medications for OUD were included. Medication regimens were evaluated throughout the inpatient stay and on day of discharge for DDIs. DDIs were classified by severity and type of interaction (increased risk of QT prolongation, additive CNS effects/respiratory depression, and opioid withdrawal). The primary endpoint was the number of potential DDIs. Other endpoints included number of each classification/severity of DDI, duration of therapy of interacting medications, and modifications made to OUD medications because of DDIs. Results: A total of 102 patients were included, with 215 inpatient interactions and 83 interactions at discharge identified. While inpatient, 85% of patients were on an interacting medication, and 46% of patients were on an interacting medication at discharge. The most common classification of DDI was additive CNS effects/respiratory depression (68.8% inpatient, 50.6% discharge), followed by QT prolongation (24.2% inpatient, 45.8% discharge). The majority of DDIs were classified as requiring close monitoring rather than contraindicated. Discussion: There are opportunities to optimize the prescribing practices surrounding OUD medications in both the inpatient setting and at discharge to ensure patient safety.

Abstract

Conventional methods for histopathologic tissue diagnosis are labor- and time-intensive and can delay decision-making during diagnostic and therapeutic procedures. We report the development of an automated and biocompatible handheld mass spectrometry device for rapid and nondestructive diagnosis of human cancer tissues. The device, named MasSpec Pen, enables controlled and automated delivery of a discrete water droplet to a tissue surface for efficient extraction of biomolecules. We used the MasSpec Pen for ex vivo molecular analysis of 20 human cancer thin tissue sections and 253 human patient tissue samples including normal and cancerous tissues from breast, lung, thyroid, and ovary. The mass spectra obtained presented rich molecular profiles characterized by a variety of potential cancer biomarkers identified as metabolites, lipids, and proteins. Statistical classifiers built from the histologically validated molecular database allowed cancer prediction with high sensitivity (96.4%), specificity (96.2%), and overall accuracy (96.3%), as well as prediction of benign and malignant thyroid tumors and different histologic subtypes of lung cancer. Notably, our classifier allowed accurate diagnosis of cancer in marginal tumor regions presenting mixed histologic composition. Last, we demonstrate that the MasSpec Pen is suited for in vivo cancer diagnosis during surgery performed in tumor-bearing mouse models, without causing any observable tissue harm or stress to the animal. Our results provide evidence that the MasSpec Pen could potentially be used as a clinical and intraoperative technology for ex vivo and in vivo cancer diagnosis.

Abstract

We have applied Bayesian inference for simultaneous inversion of multiple microseismic data to obtain event locations along with the subsurface velocity model. The traditional method of using a predetermined velocity model for event location may be subject to large uncertainties, particularly if the prior velocity model is poor. Our study indicated that microseismic data can help to construct the velocity model, which is usually a major source of uncertainty in microseismic event locations. The simultaneous inversion eliminates the requirement for an accurate predetermined velocity model in microseismic event location estimation.We estimate the posterior probability density of the velocity model and microseismic event locations with the maximum a posteriori estimation, and the posterior covariance approximation under the Gaussian assumption. This provides an efficient and effective way to quantify the uncertainty of the microseismic location estimation and capture the correlation between the velocity model and microseismic event locations. We have developed successful applications on both synthetic examples and real data from the Newberry enhanced geothermal system. Comparisons with location results based on a traditional predetermined velocity model method demonstrated that we can construct a reliable effective velocity model using only microseismic data and determine microseismic event locations without prior knowledge of the velocity model.

Abstract

El treball explora l'accessibilitat de les dades de recerca generades o usades pel personal investigador del Campus Nord de la UPC a partir de l'anàlisi de la informació indicada als articles publicats el 2022. L'anàlisi mostra que al 23,4% dels articles s'informa de la publicació de dades a través d'algun mitjà, mentre que en un 55% dels articles no s'ofereix cap informació sobre la disponibilitat de les dades. Es detallen els dipòsits i plataformes emprades per publicar dades i codi informàtic. The analysis shows that 23.4% of the articles are informed about the publication of data through some means, while in 55% of the articles, no information is provided about the availability of the data. The repositories and platforms used to publish data and computer code are detailed. El treball mostra els resultats de l'enquesta adreçada als doctorands del Campus Nord de la UPC, en relació amb dos temes: 1) L'ús i la gestió de les dades de recerca generades en l'activitat de recerca. 2) Les aplicacions que es fan servir com a suport a l'elaboració de la tesi doctoral. Els resultats de l'enquesta es van complementar amb els comentaris recollits en un focus group. L'anàlisi de les dades obtingudes mostra com només un 21% dels enquestats es consideren informats sobre procediments específics per a la gestió de dades dins del seu entorn de recerca. Les aplicacions majoritàriament usades pels enquestats són, els gestors de referències bibliogràfics, Overleaf i Grammarly. El 35% de los encuestados son usuarios de ChatGPT o de otras aplicaciones basadas en la inteligencia artificial generativa., The work shows the results of the survey addressed to the doctoral students of the North Campus of the UPC, about two topics: 1) The use and management of the research data generated in the research activity. 2) The applications that are used as support for the preparation of the doctoral thesis. The results of the survey were supplemented by comments collected in a focus group. The analysis of the data obtained shows how only 21% of respondents consider themselves informed about specific procedures for data management within their research environment. Abstract

White adipose tissue (WAT) dysfunction independently predicts cardiometabolic disease, yet there is a lack of effective adipocyte-targeting therapeutics. B3AR agonists enhance adipocyte mitochondrial function and hold potential in this regard. Based on enhanced sensitivity to B3AR-mediated browning in estrogen receptor (ER)alpha-null mice, we hypothesized that ERβ may enhance the WAT response to the B3AR ligand, CL316,243 (CL).Methods: Male and female wild-type (WT) and ERβ DNA binding domain knock-out (ERβDBDKO) mice fed high-fat diet (HFD) to induce obesity were administered CL (1 mg/kg) daily for 2 weeks. Systemic physiological assessments of body composition (EchoMRI), bioenergetics (metabolic chambers), adipocyte mitochondrial respiration (oroboros) and glucose tolerance were performed, alongside perigonadal (PGAT), subcutaneous (SQAT) and brown adipose tissue (BAT) protein expression assessment (Western blot). Mechanisms were tested in vitro using primary adipocytes isolated from WT mice, and from Esr2-floxed mice in which ERβ was knocked down. Statistical analyses were performed using 2 × 2 analysis of variance (ANOVA) for main effects of genotype (G) and treatment (T), as well as GxT interactions; t-tests were used to determine differences between in vitro treatment conditions (SPSS V24).Results: There were no genotype differences in HFD-induced obesity or systemic rescue effects of CL, yet ERβDBDKO females were more sensitive to CL-induced increases in energy expenditure and WAT UCP1 induction (GxT, p < 0.05), which coincided with greater WAT B3AR protein content among the KO (G, p < 0.05). Among males, who were more insulin resistant to begin with (no genotype differences before treatment), tended to be more sensitive to CL-mediated reduction in insulin resistance. With sexes combined, basal WAT mitochondrial respiration trended toward being lower in the ERβDBDKO mice, but this was completely rescued by CL (p < 0.05). Confirming prior work, CL increased adipose tissue ERβ protein (T, p < 0.05, all), an effect that was enhanced in WAT and BAT the female KO (GxT, p < 0.01). In vitro experiments indicated that an inhibitor of ERβ genomic function (PHTPP) synergized with CL to further increase UCP1 mRNA (p = 0.043), whereas full ERβ protein was required for UCP1 expression (p = 0.042).Conclusion: Full ERβ activity appears requisite and stimulatory for UCP1 expression via a mechanism involving non-classical ERβ signaling. This novel discovery about the role of ERβ in adipocyte metabolism may have important clinical applications.

Abstract

In this paper we develop algorithms to solve generalized weighted Fermat-Torricelli problems with positive and negative weights and multifacility location problems involving distances generated by Minkowski gauges. We also introduce a new model of clustering based on squared distances to convex sets. Using the Nesterov smoothing technique and an algorithm for minimizing differences of convex functions called the DCA introduced by Tao and An, we develop effective algorithms for solving these problems.

Abstract

The Herschel Space Observatory was the fourth cornerstone mission in the European Space Agency (ESA) science programme with excellent broad band imaging capabilities in the sub-mm and far-infrared part of the spectrum. Although the spacecraft finished its observations in 2013, it left a large legacy dataset that is far from having been fully scrutinised and still has a large potential for new scientific discoveries. This is specifically true for the photometric observations of the PACS and SPIRE instruments. Some source catalogues have already been produced by individual observing programs, but there are many observations that risk to remain unexplored. To maximise the science return of the SPIRE and PACS data sets, we are in the process of building the Herschel Point Source Catalogue (HPSC) from all primary and parallel mode observations. Our homogeneous source extraction enables a systematic and unbiased comparison of sensitivity across the different Herschel fields that single programs will generally not be able to provide. Abstract

Human respiratory syncytial virus (HRSV) is the leading viral cause of serious pediatric respiratory disease, and lifelong reinfections are common. Its 2 major subgroups, A and B, exhibit some antigenic variability, enabling HRSV to circulate annually. Globally, research has increased the number of HRSV genomic sequences available. To ensure accurate molecular epidemiology analyses, we propose a uniform nomenclature for HRSV-positive samples and isolates, and HRSV sequences, namely: HRSV/subgroup identifier/geographic identifier/unique sequence identifier/year of sampling. We also propose a template for submitting associated metadata. Universal nomenclature would help researchers retrieve and analyze sequence data to better understand the evolution of this virus.

Abstract

Studies of early 20th-century performance practice tend to focus on features that are alien to late 20th- and early 21st-century ears. Empirical analysis of timing in recordings of Chopin's Etude, Op. 25 no. 1—a piece for which performance style has remained relatively static—suggests how some foundational rules of phrasing and expressive nuance have changed over the history of recorded music. Melody note onsets were marked manually in 127 commercial recordings dating from 1909 to 2016. Overall, the data do not show an increase or decrease over time in the amount of tempo fluctuation. Independently of a tendency to use slower tempi, pianists changed the way they employ rubato. Several factors contribute to a trend whereby the fourth beat is lengthened at the expense of the second and third beats: an increase in phrase-final lengthening, an increase in the use of tempo arching for shorter groups of measures, and a tendency to delay the arrival of an accented dissonance or change of harmony instead of lengthening the melody inter-onset interval that contains it. The data illustrate nearly imperceptible shifts in interpretation and suggest that some practices thought to be the bedrock of expressive performance may be historically conditioned.
