Abstract

Abstract Background An environment of gestational diabetes mellitus (GDM) can modify the phenotype of stem cell populations differentially according to their placental localization, which can be useful to study the consequences for the fetus. We sought to explore the effect of intrauterine GDM exposure on the angiogenic properties of human amniotic membrane stem cells (hAMSCs). Methods We comprehensively characterized the angiogenic phenotype of hAMSCs isolated from 14 patients with GDM and 14 controls with normal glucose tolerance (NGT). Maternal and fetal parameters were also recorded. Hyperglycemia, hyperinsulinemia and palmitic acid were used to in vitro mimic a GDM-like pathology. Pharmacological and genetic inhibition of protein function was used to investigate the molecular pathways underlying the angiogenic properties of hAMSCs isolated from women with GDM. Abstract

Abstract Background Many genes and molecular pathways are associated with obesity, but the mechanisms from genes to obesity are less well known. Eating behaviors represent a plausible pathway, but because the relationships of eating behaviors and obesity may be bi-directional, it remains challenging to resolve the underlying pathways. A longitudinal approach is needed to assess the contribution of genetic risk during the development of obesity in childhood. In this study we aim to examine the relationships between the polygenic risk score for body mass index (PRS-BMI), parental concern of overeating and obesity indices during childhood. Methods The IDEFICS/I.Family study is a school-based multicenter pan-European cohort of children observed for 6 years (mean ± SD follow-up 5.8 ± 0.4). Children examined in 2007/2008 (wave 1) (mean ± SD age: 4.4 ± 1.1, range: 2–9 years), in 2009/2010 (wave 2) and in 2013/2014 (wave 3) were included. A total of 5112 children (49% girls) participated at waves 1, 2 and 3. For 2656 children with genome-wide data we constructed a PRS based on 2.1 million single nucleotide polymorphisms. Z-score BMI and z-score waist circumference (WC) were assessed and eating behaviors and relevant confounders were reported by parents via questionnaires. Parental concern of overeating was derived from principal component analyses from an eating behavior questionnaire. Results In cross-lagged models, the prospective associations between z-score obesity indices and parental concern of overeating were bi-directional. In mediation models, the association between the PRS-BMI and parental concern of overeating at wave 3 was mediated by baseline z-BMI (β = 0.16, 95% CI: 0.10, 0.21) and baseline z-WC (β = 0.17, 95% CI: 0.11, 0.23). To a lesser extent, baseline parental concern of overeating also mediated the association between the PRS-BMI and z-BMI at wave 3 (β = 0.10, 95% CI: 0.07, 0.13) and z-WC at wave 3 (β = 0.09, 95% CI: 0.07, 0.12). Conclusions The findings suggest that the prospective associations between obesity indices and parental concern of overeating are likely bi-directional, but obesity indices have a stronger association with future parental concern of overeating than vice versa. The findings suggest parental concern of overeating as a possible mediator in the genetic susceptibility to obesity and further highlight that other pathways are also involved. A better understanding of the genetic pathways that lead to childhood obesity can help to prevent weight gain. Abstract

Fog computing has emerged as a promising technology that can bring the cloud applications closer to the physical IoT devices at the network edge. While it is widely known what cloud computing is, and how data centers can build the cloud infrastructure and how applications can make use of this infrastructure, there is no common picture on what fog computing and a fog node, as its main building block, really is. One of the first attempts to define a fog node was made by Cisco, qualifying a fog computing system as a mini-cloud, located at the edge of the network and implemented through a variety of edge devices, interconnected by a variety, mostly wireless, communication technologies. Thus, a fog node would be the infrastructure implementing the said mini-cloud. Other proposals have their own definition of what a fog node is, usually in relation to a specific edge device, a specific use case or an application. In this paper, we first survey the state of the art in technologies for fog computing nodes as building blocks of fog computing, paying special attention to the contributions that analyze the role edge devices play in the fog node definition. We summarize and compare the concepts, lessons learned from their implementation, and show how a conceptual framework is emerging towards a unifying fog node definition. We focus on core functionalities of a fog node as well as in the accompanying opportunities and challenges towards their practical realization in the near future.

236. Diffuse flux of galactic neutrinos and gamma rays

Abstract

We calculate the fluxes of neutrinos and gamma rays from interactions of cosmic rays with interstellar matter in our galaxy. We use EPOS-LHC, SIBYLL and GHEISHA to obtain the yield of these particles in proton, helium and iron collisions at kinetic energies between 1 and 10^8 GeV, and we correlate the cosmic ray density with the mean magnetic field strength in the disk and the halo of our galaxy. We find that at E>1 PeV the fluxes depend very strongly on the cosmic-ray composition, whereas at 1-5 GeV the main source of uncertainty is the cosmic-ray spectrum out of the heliosphere. We show that the diffuse flux of galactic neutrinos becomes larger than the conventional atmospheric one at E>1 PeV, but that at all IceCube energies it is 4 times smaller than the atmospheric flux from forward-charm decays., Comment: 14 pages, version to appear in JCAP

Abstract

We find that the ratio r_{mu e} of the muon to the electromagnetic component of an extended air shower at the ground level provides an indirect measure of the depth X_{max} of the shower maximum. This result, obtained with the air-shower code AIRES, is independent of the hadronic model used in the simulation. We show that the value of r_{mu e} in a particular shower discriminates its proton or iron nature with a 98% efficiency. We also show that the eventual production of forward heavy quarks inside the shower may introduce anomalous values of r_{mu e} in isolated events., Comment: 10 pages, version to appear in Astroparticle Physics

Abstract

Animals play a profoundly important and intricate role in our lives today. Dogs have been human companions for thousands of years, but they now work closely with us to assist the disabled, and in combat and search and rescue situations. Farm animals are a critical part of the global food supply chain, and there is increasing consumer interest in organically fed and humanely raised livestock, and how it impacts our health and environmental footprint. Wild animals are threatened with extinction by human induced factors, and shrinking and compromised habitat. This review sets the goal to systematically survey the existing literature in smart computing and sensing technologies for domestic, farm and wild animal welfare. We use the notion of \emph{animal welfare} in broad terms, to review the technologies for assessing whether animals are healthy, free of pain and suffering, and also positively stimulated in their environment. Also the notion of \emph{smart computing and sensing} is used in broad terms, to refer to computing and sensing systems that are not isolated but interconnected with communication networks, and capable of remote data collection, processing, exchange and analysis. We review smart technologies for domestic animals, indoor and outdoor animal farming, as well as animals in the wild and zoos. The findings of this review are expected to motivate future research and contribute to data, information and communication management as well as policy for animal welfare.

BackgroundCoronavirus-19 (COVID-19) disease is driven by an unchecked immune response to the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) virus which alters host mitochondrial-associated mechanisms. Compromised mitochondrial health results in abnormal reprogramming of glucose metabolism, which can disrupt extracellular signalling. We hypothesized that examining mitochondrial energy-related signalling metabolites implicated in host immune response to SARS-CoV-2 infection would provide potential biomarkers for predicting the risk of severe COVID-19 illness.MethodsWe used a semi-targeted serum metabolomics approach in 273 patients with different severity grades of COVID-19 recruited at the acute phase of the infection to determine the relative abundance of tricarboxylic acid (Krebs) cycle-related metabolites with known extracellular signaling properties (pyruvate, lactate, succinate and α-ketoglutarate). 247. Effects of stem cells from inducible brown adipose tissue on diet-induced obesity in mice

Abstract

Abstract Adipose-derived mesenchymal stem cells (ASCs) are a promising option for the treatment of obesity and its metabolic co-morbidities. Despite the recent identification of brown adipose tissue (BAT) as a potential target in the management of obesity, the use of ASCs isolated from BAT as a therapy for patients with obesity has not yet been explored. Metabolic activation of BAT has been shown to have not only thermogenic effects, but it also triggers the secretion of factors that confer protection against obesity. Herein, we isolated and characterized ASCs from the visceral adipose tissue surrounding a pheochromocytoma (IB-hASCs), a model of inducible BAT in humans. We then compared the anti-obesity properties of IB-hASCs and human ASCs isolated from visceral white adipose tissue (W-hASCs) in a murine model of diet-induced obesity. We found that both ASC therapies mitigated the metabolic abnormalities of obesity to a similar extent, including reducing weight gain and improving glucose tolerance. However, infusion of IB-hASCs was superior to W-hASCs in suppressing lipogenic and inflammatory markers, as well as preserving insulin secretion. Our findings provide evidence for the metabolic benefits of visceral ASC infusion and support further studies on IB-hASCs as a therapeutic option for obesity-related comorbidities.
