Ecosystem modelling has predicted that fishing for western rock lobster Panulirus cygnus in deep water (50–80m) habitats will result in increased abundance of their macroinvertebrate prey, which would, in turn, support greater production of small-bodied invertivore fish species. To investigate the potential ecosystem effects of rock lobster fishing, a targeted fisheries closure was proposed in deep water habitats along the coast of Western Australia. Data on small-bodied invertivore fish abundance, from baited video, were used to investigate differences across habitats and simulate the likely power of any study to detect change. In general, small-bodied invertivore fish were more abundant at shallower macroalgae-dominated sites, whereas the most abundant single species, the western king wrasse Coris auricularis, was abundant across all habitats and sites. Power simulation of a mixed-model before–after–control–impact (BACI) design found that a 40–50% decrease in either Coris auricularis or small-bodied invertivore fish in general would be very likely to be detected (power ~0.8). Based on the power simulation, we suggest a general sampling design to investigate change before and after the establishment of the fishery closure and make suggestions for further ecological studies to investigate the predicted ecosystem effects of rock lobster fishing.

To assess the characteristics of hybrid receptors composed of one kinase-inactive alpha beta-insulin half-receptor and one endogenous alpha beta-insulin-like growth factor 1 (IGF-1) or insulin half-receptor, a cell line expressing an insulin receptor truncated by 365 amino acids (HIR delta 978) was studied, which lacks most of the cytoplasmic beta-subunit. Analysis by sodium dodecyl sulfate polyacrylamide gel electrophoresis under nonreducing conditions revealed four distinct receptor species: endogenous receptors, the more rapidly migrating HIR delta 978 homodimer, and two intermediate species representing HIR delta 978/IGF-1 hybrid receptors and HIR delta 978/IR hybrid receptors. In vivo ligand-binding affinity of the hybrid receptors was studied by receptor-ligand cross-linking, and the delta 978/IGF-1R hybrid receptor was found to have a high affinity for IGF-1, whereas its affinity for insulin was low. Autophosphorylation studies of lectin-purified receptors revealed that neither the HIR delta 978 holoreceptor nor the hybrid receptors underwent autophosphorylation in response to either ligand, despite the presence of intact IGF-1 or insulin half-receptors in the hybrids. Neither hybrid receptor underwent ligand-induced endocytosis, as assessed with the bioactive photoaffinity probes B2(2-nitro-4-azidophenylacetyl)-des-PheB1-insulin and N-epsilon B28-monoazidobenzoyl-IGF-1. In conclusion, the HIR delta 978/IGF-1R hybrid receptor has a high in vivo affinity for IGF-1 but not for insulin. Neither the delta 978/IGF-1R nor the delta 978/IR hybrids undergo autophosphorylation or ligand-induced endocytosis in response to either ligand, indicating that intramolecular trans-, rather than cis-, signal transduction is important in mediating autophosphorylation and endocytosis.

Epidermal growth factor (EGF) treatment causes autophosphorylation of the epidermal growth factor receptor (EGFR) leading to increased guanine nucleotide exchange factor (GEF; Sos) activity and enhanced formation of p21ras-GTP. The connection of the EGFR to p21ras activation can occur through binding of Grb2.Sos complexes to the EGFR or through the adaptor protein Shc via EGFR.Shc.Grb2.Sos multimeric complexes. Therefore, we investigated the importance of Shc in coupling the EGFR to activation of ras GEF (Sos). EGF treatment led to rapid tyrosine phosphorylation of Shc. Although phosphorylated EGFR can bind to both Shc and Grb2, the predominant linkage was observed between EGFR and Shc. Similarly, more Grb2 was associated with Shc than with EGFR after EGF stimulation. Immunoprecipitation of Shc from EGF-stimulated cells removed almost all EGFR-associated Grb2. Furthermore, immunodepletion of Shc proteins from membrane fractions of EGF-stimulated cells removed 93% of the ras GEF activity, whereas, precipitation of EGFR had only a small effect on ras GEF activity. These data indicate that coupling to Shc provides the major pathway linking activated EGFRs to Grb2.Sos and stimulation of the p21ras pathway.

Insulin stimulates tyrosine phosphorylation of insulin receptor substrate-1 (IRS-1) and She in Rat1 fibroblasts overexpressing wild type insulin receptors. We investigated the relative role of IRS-1 and She in insulin activation of guanine nucleotide releasing factor (GNRF) and p21ras-GTP formation. The time course of insulin-stimulated tyrosine phosphorylation of IRS-1 was rapid, whereas Shc phosphorylation was relatively slow. Growth factor receptor bound protein-2 (Grb2) associated with IRS-1 rapidly and gradually dissociated after 5 min, whereas Grb2 association with Shc was slower and reached a maximum at 10 min after insulin stimulation. Thus, the kinetics of Grb2 association with IRS-1 and She corresponded closely to the time course of tyrosine phosphorylation of IRS-1 and Shc, respectively. Importantly, 3-13-fold more Grb2 was associated with Shc than with IRS-1. In addition, the kinetics of insulin-stimulated GNRF activity and p21ras-GTP formation corresponded more closely to the time course of Shc phosphorylation than to the kinetics of IRS-1 phosphorylation. Furthermore, immunoprecipitation of Shc proteins from cell lysates of insulin-stimulated cells removed 67% of the GNRF activity, whereas precipitation of IRS-1 had a negligible effect on GNRF activity. Thus, although both IRS-1 and Shc associate with Grb2, the current results indicate that Shc plays a more important role than IRS-1 in insulin stimulation of GNRF activity and subsequent p21ras-GTP formation.

The differential cross sections for the Kr (e, 3e) Kr++ reaction are calculated (by using correlated wave function) in the case of high incident energy (5 keV) for the three terms of the final Kr++ (4p 4) ion. We have performed an ab initio calculation on the basis of the first Born approximation using correlated wave functions for the target. The agreement with the first available (e, 3e) experimental data is fairly good. Other typical experimental situations are proposed and discussed.

We have tested synthetic peptides composed of Th (T1) and V3 loop B cell neutralizing determinants [SP10 MN(A)] of HIVMN gp120 and the fusogenic (F) domain of gp41 as immunogens in rhesus monkeys. After two immunizations with either HIV env peptide T1-SP10 MN(A) or F-T1-SP10 MN(A), rhesus monkey serum neutralization titers against the HIVMN isolate ranged from 1:160 to 1:1400, and in cell-cell syncytium inhibition assay ranged from 1:20 to 1:80. However, in contrast to animals immunized with T1-SP10 MN(A), animals immunized twice with F-T1-SP10 MN(A) had no rise in anti-gp120 and neutralizing antibodies with an additional immunization with F-T1-SP10 MN(A) peptide. One of 4 rhesus monkeys (18987) had anti-HIVMN antibodies that cross-neutralized divergent HIV isolates HIVIIIB and HIVRF. Serum from animal 18987 neutralized 5 of 10 HIV isolates tested, and neutralizing activity against HIVIIIB of 18987 serum was absorbed with the conserved gp120 loop V3 sequence IGPGRAF. Anti-HIV neutralizing antibodies were boosted after a 6-mo rest by 500 micrograms of T1-SP10 MN(A) in 4 of 4 animals previously immunized with T1-SP10 MN(A) and in 2 of 2 animals previously immunized with F-T1-SP10 MN(A). However, immunization after 6-mo rest of animal 18987 with 500 micrograms of T1-SP10 MN(A) peptide, although boosting anti-HIVMN neutralizing antibodies, selectively did not boost cross-neutralizing anti-HIVIIIB antibodies. Thus, synthetic peptides containing T and B cell epitopes of HIV gp120 can induce high levels of anti-HIVMN neutralizing antibodies in primates.

To assess the function of the cytoplasmic domain of the insulin receptor (IR) beta subunit, we have studied a mutant IR truncated by 365 aa (HIR delta 978), thereby deleting > 90% of the cytoplasmic domain. HIR delta 978 receptors were processed normally to homodimers that were expressed at the cell surface where they bind insulin with normal affinity. Although these truncated IRs were inactive with respect to ligand-induced internalization and autophosphorylation, insulin stimulated endogenous substrate (pp185) phosphorylation significantly more in HIR delta 978 cells than in untransfected Rat1 cells. Importantly, despite absence of the beta-subunit cytoplasmic domain, fibroblasts expressing HIR delta 978 receptors displayed enhanced sensitivity to insulin for stimulation of glucose incorporation into glycogen, alpha-aminoisobutyric acid uptake, thymidine incorporation, and S6 kinase activity compared with parental fibroblasts. Insulin also induced the expression of the protooncogene c-fos and the early growth response gene Egr-1 in HIR delta 978 cells far greater than in parental Rat1 fibroblasts. Furthermore, an agonistic monoclonal antibody specific for the human IR stimulated insulin action in fibroblasts expressing wild-type human IR but had no effect on HIR delta 978 cells. In conclusion, the HIR delta 978 truncated IRs appear to confer enhanced insulin sensitivity by augmenting the signaling properties of the endogenous rodent IRs.

The fusogenic (F) domain of human immunodeficiency virus (HIV) gp41 envelope (env) protein has sequence similarities to many virus and mediates the fusion of HIV-infected cells. During a survey of the immunogenicity of HIV env peptides in chimpanzees, we have observed that HIV peptide immunogenicity was dramatically altered by the NH2-terminal synthesis of the gp41 F domain to an otherwise immunogenic peptide. We compared two hybrid peptide types comprised of T helper (Th) and B cell epitopes of HIV gp120 env protein for their immunogenicity in chimpanzees. The Th-B epitope hybrid peptides contained the HIV gp120 Th cell determinant, T1 (amino acids [aa] 428-440)-synthesized NH2 terminal to gp120 V3 loop peptides, which contain B cell epitopes that induce anti-HIV-neutralizing antibodies (SP10IIIB [aa 303-321] and SP10IIIB [A] [aa 303-327]). The F-Th-B peptide contained the HIV gp41 F domain of HIVIIIB gp41 (aa 519-530)-synthesized NH2 terminal to the Th-B peptide. Whereas Th-B peptides were potent immunogens for chimpanzee antibody and T cell-proliferative responses, the F-Th-B peptide induced lower anti-HIV gp120 T and B cell responses. Moreover, immunization of chimpanzees with F-Th-B peptide but not Th-B peptides induced a significant decrease in peripheral blood T lymphocytes (mean decrease during immunization, 52%; p < 0.02). Chimpanzees previously immunized with F-Th-B peptide did not respond well to immunization with Th-B peptide with T or B cell responses to HIV peptides, demonstrating that the F-Th-B peptide induced immune hyporesponsiveness to Th and B HIV gp120 env determinants. These observations raise the hypothesis that the HIV gp41 env F domain may be a biologically active immunoregulatory peptide in vivo, and by an as yet uncharacterized mechanism, promotes primate immune system hyporesponsiveness to otherwise immunogenic peptides.

Two Macaca fascicularis with preexisting immunity to vaccinia virus were immunized twice with recombinant vaccinia virus expressing SIVmne gp160. Their SIV-specific antibody responses were lower than that of vaccinia-naive animals immunized similarly. Upon repeated boosting with gp160, the SIV-specific antibody titers in vaccinia-primed animals reached similar levels as vaccinia-naive animals and with comparable neutralizing titers. Both animals were protected against repeated intravenous challenge with low-dose SIVmne E11S. These results are significant because SIVmne E11S infection in M. fascicularis is pathogenic and leads to AIDS-like diseases.

A method is proposed for approximately computing the post collisional interaction of the two escaping electrons in otherwise standard first order ionisation calculations. At intermediate energies and in asymmetric kinematics the effects are found to be very small. The interactions of these electrons with the residual ion are considered through the use of distorted waves. For the slow one the latter are used in the calculations but for the fast one their effect is investigated using Fourier transforms. These interactions play a significant role.

Electron-impact double ionization of noble gases is investigated theoretically for the case of high incident energies (5 keV). An ab initio calculation is made including partial correlation in the initial state as well as in the final state. The results of the calculations are compared with those of other theories and with the first available (e, 3e) experimental data on krypton 4p 6.

Vaccines prepared from the envelope glycoprotein, gp120, of the common laboratory isolate of human immunodeficiency virus type 1 (HIV-1) (IIIB/LAV-1) elicit antibodies that neutralize the homologous virus but show little if any cross-neutralizing activity. This may be because the principal neutralizing determinant (PND) of gp120 is highly unusual in the IIIB/LAV-1 strain and is not representative of those found in the majority of field isolates. We have now examined the immunogenicity of recombinant gp120 prepared from the MN strain of HIV-1 (MN-rgp120), whose PND is thought to be representative of approximately 60% of the isolates in North America. Our results show that MN-rgp120 is a potent immunogen and elicits anti-gp120 titers comparable to those found in HIV-1-infected individuals. While both MN-rgp120 and IIIB-rgp120 induced antibodies able to block gp120 binding to CD4, strain-specific and type-common blocking antibodies were detected. Finally, antibodies to MN-rgp120 but not to IIIB-rgp120 were effective in neutralizing a broad range of laboratory and clinical isolates of HIV-1. These studies demonstrate that susceptibility or resistance to neutralization by antibodies to gp120 correlates with the PND sequence and suggest that the problem of antigenic variation may not be insurmountable in the development of an effective AIDS vaccine.

Simultaneous exposure to chemicals which can oxidize the haemoglobin of the red blood cell to methaemoglobin is common. Although the effects of some of these agents have been documented individually, little research considers the interactive effects. In-vitro experiments on the treated blood of female Dorset sheep assessed the interactive capacity of chlorite, copper and nitrite to affect methaemoglobin formation. All combinations of doses which produced 2.5, 5, 10% methaemoglobin were tested in all possible combinations (a total of 80), as were the controls. This included data on each chemical alone, each two-way combination and the three-way combination. The response is largely additive (the sum of the individual effects) except for one of the two-way interactions, chlorite/nitrite (P < . 01), which showed antagonism. Chlorite may oxidize nitrite which could explain the less-than-additive response. Overall, the result of combining these agents on methaemoglobin was additive.

To identify the principal neutralization determinant (PND) of simian immunodeficiency virus (SIV), antisera were generated using recombinant gp110 [the SIV analog of the human immunodeficiency virus type 1 (HIV-1) external envelope glycoprotein, gp120], gp140, several large recombinant and proteolytic envelope fragments, and synthetic peptides of the SIVmac251 isolate. When purified under conditions that retain its native structure, gp110 bound CD4 and elicited antisera that neutralized SIVmac251 with high titer. Native gp110 also completely inhibited neutralizing antibody in sera from SIVmac251-infected macaques. In contrast, denatured gp110 and gp140, large envelope fragments, and synthetic peptides (including peptides analogous to the HIV-1 PND) elicited very low or undetectable neutralizing antibody titers and did not inhibit neutralizing antibody in infected macaque sera. Enzymatically deglycosylated gp110 efficiently absorbed neutralizing antibodies from macaque sera, showing that neutralizing antibodies primarily bind the protein backbone. A 45-kDa protease digest product, mapping to the carboxyl-terminal third of gp110, also completely absorbed neutralizing antibodies from infected macaque sera. These results show that the PND(s) of this SIV isolate depends on the native conformation and that linear peptides corresponding to the V3 loop of SIV envelope, in contrast to that of HIV-1, do not elicit neutralizing antibody. This may affect the usefulness of SIVmac for evaluating HIV-1 envelope vaccine approaches that rely on eliciting neutralizing antibody.

Simian immunodeficiency virus (SIV) was used as a model to study the protective efficacy of an immunization regimen currently being evaluated as candidate vaccines against HIV in human subjects. Four Macaca fascicularis were first immunized with recombinant vaccinia virus expressing the envelope glycoprotein gp160 of SIVmne and then boosted with subunit gp160. Both cell-mediated and humoral immune responses against SIV, including neutralizing antibodies, were elicited. The macaques were shown to be protected from a homologous virus infection as determined by serology, lymphocyte cocultivation, polymerase chain reactions and in vivo transmission analyses. Four unimmunized control animals were readily infected. However, viremia in infected control animals could decrease substantially following the initial phase of infection so that persistent infection might not be readily detectable.

Simian immunodeficiency virus (SIV) is a primate lentivirus related to human immunodeficiency viruses and is an etiologic agent for acquired immunodeficiency syndrome (AIDS)-like diseases in macaques. To date, only inactivated whole virus vaccines have been shown to protect macaques against SIV infection. Protective immunity was elicited by recombinant subunit vaccines. Four Macaca fascicularis were immunized with recombinant vaccinia virus expressing SIVmne gp160 and were boosted with gp160 produced in baculovirus-infected cells. All four animals were protected against an intravenous challenge of the homologous virus at one to nine animal-infectious doses. These results indicate that immunization with viral envelope antigens alone is sufficient to elicit protective immunity against a primate immunodeficiency virus. The combination immunization regimen, similar to one now being evaluated in humans as candidate human immunodeficiency virus (HIV)-1 vaccines, appears to be an effective way to elicit such immune responses.

Calculations of triple differential cross sections for the process where impact of a moderately energetic electron on a two-electron target simultaneously ejects one of the electrons and excites the other are made in the framework of a first Born approximation without exchange. Results obtained using ejected electron wavefunctions calculated with Coulomb potentials of different effective charge, with the static potential due to the unperturbed residual ion, or with Klapisch-type parametric potentials, are compared with experimental data. Strong dependence on the final state wavefunction is found, notably for the ratio between contributions from excitation of the 2s0, 2p0 and 2p+or-1 residual ion states. The cross sections, both globally and for the individual contributions, are however insensitive to the amount of correlation in the initial state wavefunctions.

The influence of human anti-human immunodeficiency virus type 1 (HIV-1) antibody on HIV-1 infection of freshly isolated normal human peritoneal macrophages and blood monocytes was examined. Each of 14 HIV antibody-positive human serum samples was found to block the infection of four virus isolates (human T-cell lymphotropic virus type IIIBa-L [HTLV-IIIBa-L], HTLV-IIIB, D.U. 6587-7, and D.U. 7887-8) at serum dilutions ranging from 10(-1) to 10(-2). Three of these isolates (HTLV-IIIBa-L, D.U. 6587-7, and D.U. 7887-8) infected cultures of monocytes and macrophages rapidly and produced high levels of virus reverse transcriptase and p24 antigen. A fourth virus isolate (HTLV-IIIB) infected the monocytes and macrophages more slowly and produced low levels of viral protein. More dilute HIV antibody-positive sera had no significant effect on the overall level or rate of virus infection or expression. Complement did not appear to influence the course of infection by any combination of antisera or virus examined. Successful HIV-1 infection of the peritoneal macrophages and blood monocytes under the conditions tested showed strict dependence on CD4 since a recombinant CD4 polypeptide and an anti-CD4 monoclonal antibody effectively blocked the process.

Sensitive and reproducible assays for SIV infection and syncytium formation have been developed in which high titers of neutralizing and fusion-inhibiting antibodies can be recorded. These assays will contribute toward our understanding of the role of humoral responses in SIV vaccine strategies.

The possibility of constructing a distorted-wave second Born approximation model for electron-helium collisions at energies above 30 eV is examined. It is proposed to calculate the second Born term by analytic integration over the intermediate momentum of the colliding electron and by replacing the infinite sum over intermediate target states by a finite sum over a pseudostate expansion. A method is proposed for obtaining pseudostate bases which approximately reproduce known bound-state contributions to the oscillator strength sum rules for n3,1 S to n3,1 P transitions and polarizabilities of the 11 S, 21 S and 23 S states. These bases are composed of 'exact' n3,1 S, n3,1 P and n3,1 D states up to n=3 and of a restricted number of 3,1 S, 3,1 P and 3,1 D L2 -type pseudostates. Certain of the latter lie in the continuum and their energies are adjusted to correspond to quadrature points for Gauss-Laguerre integration, this in order to easily evaluate the continuum contributions to oscillator strength sums, polarizabilities and second Born terms.

An intravenous solution of 99% pure globulin (hyperimmune IgG, HIVIG) was obtained from pooled plasma of selected human immunodeficiency virus (HIV-1)-seropositive asymptomatic donors with greater than 400 CD4+/microliters cells per microliter and a high titer of antibody to HIV-1 p24 protein. HIVIG had high titers of antibody to p24, glycoprotein 41 (gp41), and gp120, group-specific neutralizing activity, and binding to the gp120 hypervariable loop region. It inhibited syncytia formation. At low concentration, it enhanced viral production of HIV-1 in infected peripheral blood monocytes but was inhibitory at higher concentration. HIVIG directed group-specific antibody-dependent cellular cytotoxicity against HIV-infected targets. For a period of 6 to 28 months, plasma donors kept stable antibody titers and had a 1.0% decrease in CD4+ cells per month. One gram per kilogram HIVIG injected in two juvenile chimpanzees was well tolerated and did not transmit HIV, as measured by negative cell culture, IgM immune response to HIV proteins, and polymerase chain reaction. The mean half-life of HIV-1 p24 antibody was 15 days. These preliminary data suggest that HIVIG is a safe product suitable for clinical trial in HIV-1-infected individuals.

Various recent findings in experiment, theory and simulation, although not yet entirely reconciled, combine to suggest that an adequate continuum theory of two‐dimensional foam rheology can be formulated.

A semiclassical method used previously for high-energy proton-atom collisions is adapted to the case of electron-impact ionisation of atomic targets at intermediate and high energies. Incident electron trajectories are calculated classically using the spherically averaged field of the target. Scattered and ejected electron trajectories are calculated using the averaged field of the residual ion, their starting conditions being determined by taking approximate account of classical momentum conservation at the instant of transition. Scattering angles and related ejected electron angular distributions are thus obtained for particular impact parameters and collision times. The corresponding ionisation probability is determined quantum mechanically using the time-dependent perturbation of the target system calculated from the incident trajectory.

We have previously described a synthetic peptide (T1-SP10) derived from two noncontiguous regions of HTLVIIIB envelope gp120 (T1, amino acids 428-443; SP10, amino acids 303-321) that induced type-specific anti-HIV neutralizing antibodies and T cell proliferative responses against native HIV gp120 when used as a carrier-free immunogen in goats. In this study, HTLVIIIB T1-SP10 synthetic peptides were used to immunize rhesus monkeys to determine if the peptides were capable of eliciting HIV-specific neutralizing antibody and proliferative responses in primates. Four compounds (alum, polyA:polyU, threonyl-muramyldipeptide (MDP) and IFA) were also compared for efficacy as adjuvants in this system. Rhesus monkeys immunized with T1-SP10 peptides generated high titers of antibodies against the immunogens and also against HTLVIIIB gp120. Sera from all four animals given T1-SP10 in IFA or threonyl-MDP neutralized infection by HTLVIIIB and blocked virus-dependent cell fusion events. A peak neutralization titer of 1:940 was seen in one animal given IFA (19600) and a titer of 1:900 was seen in one of the monkeys (17371) given threonyl-MDP. Proliferative responses of immune rhesus PBMC to T1-SP10 appeared after the first injection. After eight immunizations, two of eight monkeys (one injected with peptides in threonyl-MDP and one given peptides in IFA) had PBMC proliferative responses to native HTLVIIIB gp120. These data demonstrate that the carrier-free T1-SP10 synthetic peptide construct can induce high titers of neutralizing anti-HIV antibody responses and PBMC proliferative responses to HIV in primates.

summary Changes in carbohydrate content and composition of ryegrass (Lolium perenne L.) cv. Reveille field-grown in Normandy were investigated from May 1987 to May 1988. During the winter period, the concentrations of high DP (degree of polymerization) fructan were the lowest whilst those of ethanol-soluble carbohydrates were the highest. Ethanol-soluble carbohydrates also accounted for most of the non-structural carbohydrates of four cultivars of Lolium perenne field-grown in central France in January 1989. The accumulation of osmotically active carbohydrates during winter leads only to a small depression of freezing point and consequently, these sugars cannot be considered as efficient cryoprotectants in ryegrass.

Several infaunal bivalve taxa show patterns of decreased biomass in areas with higher densities of adjacent reef-associated predators (the snapper, Pagrus auratus and rock lobster, Jasus edwardsii). A caging experiment was used to test the hypothesis that patterns observed were caused by predation, using plots seeded with a known initial density of the bivalve Dosinia subrosea to estimate survivorship. The caging experiment was replicated at several sites inside and outside two highly protected marine reserves: predators are significantly more abundant inside these reserves. Survivorship in fully caged, partially caged and open plots were then compared at sites having either low (non reserve) or high (reserve) predator density. The highest rates of survivorship of the bivalve were found in caged plots inside reserves and in all treatments outside reserves. However, inside reserves, open and partially caged treatments exhibited low survivorship. It was possible to specifically attribute much of this mortality to predation by large rock lobsters, due to distinctive marks on the valves of dead D. subrosea. This suggests that predation by large rock lobster could indeed account for the distributional patterns previously documented for certain bivalve populations. Our results illustrate that protection afforded by marine reserves is necessary to investigate how depletion through fishing pressure can change the role of upper-level predators and trophic processes between habitats.

In 2003, asplenia had involved 250000 patients in France. These patients are at risk of severe infection, mostly with capsulated bacteria as pneumococci, meningococci and Haemophilus. The higher mortality and morbidity due to infection in asplenic patient led in June 2003 a French expert committee to propose preventive management based on vaccination and antibioprophylaxis.Update article.For vaccination, two vaccines against pneumococci are available. The first one, the antipolysaccharide (Pneumo 23) is recommended for adults. It is effective for the majority of the serotypes even if its efficacy can be variable. The second one a conjugated pneumococcal vaccine (Prenevar) is used for children under two years because it has higher activity on antibiotic resistant strains therefore increasing antibiotic prophylaxis efficiency. When splenectomy is required, vaccination against pneumococci, Haemophilus (b type) and C meningococci must be performed at least 15 days before surgery, in order to get better immune stimulation. In case of emergency, vaccines have to be administrated within 30 days after surgery. Antibioprophylaxis is based on cefazolin injection before splenectomy and by postoperative intravenous amoxicillin administration. As soon as oral intake is allowed, antibioprophylaxis is continued for at least two years in adults and five years in children. Both antibiotic and vaccination have been reported to reduce pneumococcus infections.

Clinical equipoise exists regarding whether relatives of individuals with venous thromboembolism (VTE) and thrombophilia should be screened for thrombophilia. There have been no systematic attempts to summarize studies that have assessed the incidence of VTE in relatives. The purpose of this review was to systematically identify and review observational studies with thrombophilic relatives and to summarize their findings with respect to their risk of VTE. We conducted a systematic literature review and included nine observational studies meeting a priori inclusion criteria. Potentially eligible studies evaluated VTE incidence in relatives of index patients (probands) with symptomatic thrombophilia. In the four prospective studies, the incidence of VTE for asymptomatic family members with factor V Leiden ranged from 0.58-0.67% per year, 1.0-2.5% for protein C deficiency, 0.7-2.2% for protein S deficiency, and 4% for antithrombin deficiency. About half of all VTEs occurred during well-known risk periods but incidence rates were decreased by use of prophylaxis. No deaths from pulmonary embolism or fatal hemorrhages from anticoagulants were reported. The incidence of VTE was generally lower in the retrospective studies. The pooled relative risk from four retrospective studies for factor V Leiden carriers was 3.69 (CI 2.27, 6.00) and from two studies the pooled relative risk for deficiencies of protein C, protein S, and antithrombin was 10.58 (CI 5.38, 20.81). In conclusion, the risk of VTE events in asymptomatic relatives is low, but this may be an underestimate. Anticoagulant prophylaxis during risk periods appears to be of benefit but further research in this area is required.

This paper examines how to create consistent, portable, usable UUT requirement specifications utilizing COTS computer languages such as Basic, Java, C or C++. It goes on to describe the benefits of using standard, common building blocks that represent signals, and identifies the importance of allowing users to extend their own signal components. It concludes by showing several examples of the same, real requirement captured in different languages, and how these can be ported from one to another.

Quantum Defect theory is a well established theoretical concept in modern spectroscopy that was found particularly powerful in the study of Rydberg states and photoionization of various atomic and molecular species. We show that this approach can also be useful in electron impact ionization problems where state of the art theoretical methods are presently restricted mostly to simple atomic targets. We found that this approach leads to significant improvements over previous calculations for the well documented case of the ionization of argon in equal energy sharing geometry.

Introduction La distension pulmonaire est un mecanisme majeur de lesions induites par la ventilation mecanique pouvant entrainer un syndrome de detresse respiratoire de l’adulte (SDRA). Or, il a ete demontre par tomodensitometrie, que meme lors d’une ventilation dite « protectrice », pouvaient persister des zones distendues [1] . La distension peut etre aussi etre mesuree sur le degre convexite ou de concavite de la courbe pression/temps lors de la phase a debit constant aussi appele le stress index (SI) [2] . L’objectif de notre etude est de depister la distension (SI > 1) et identifier les facteurs de risque associes. Materiel et methodes Etude observationnelle, retrospective, en reanimation chirurgicale du CHRU de Lille de mai 2013 a fevrier 2014. Recueil dans 30 dossiers, apres information et consentement des proches de patients consecutirifs ventiles en volume assiste controle avec un ventilateur comportant la mesure automatisee du SI (Servo i™, Maquet GmbH), sans efforts de respiration spontanee. Recueil de donnees anamnestiques, cliniques, d’imagerie, parametres de ventilation dont le SI, et gazometriques des 1eres 24 h. Comparaison des patients distendus (SI > 1) ou non (SI≤1) au sein de l’effectif global et de sous-groupes par analyse des donnees du dossier, notamment l’imagerie pulmonaire tomodensitometrique et/ou ultrasonographique (groupes SDRA ou non selon les definitions de l’American-European Consensus Conference ou de Berlin [3] , groupe epanchements et/ou atelectasies, et groupe de patients sans atteinte respiratoire aigue). Comparaisons effectuees avec les tests non-parametrique de Mann-Whitney, Chi 2 et Fisher exact lorsqu’approprie (logiciel SPSS v18, IBM Corp). Resultats Soixante-douze patients ont ete ventiles sur les 4 mois du recueil, dont 30 ventiles en VAC avec mesure automatisee du SI dans des conditions de validite. Le SI recueilli revele 30 % de distension (9/30) avec comme facteurs de risque : volume courant (Vt) bas ( p = 0,028), FiO2 elevee ( p = 0,040), et PEP elevee ( p = 0,040). Les patients en SDRA etaient distendus dans 64 % des cas (9/14) avec comme facteurs de risque : Vt bas ( p = 0,048) et pression de plateau (Pplat) plus elevee ( p = 0,036). Les patients atteints d’atelectasies et/ou d’epanchement etaient distendus dans 63 % des cas (5/8) avec comme facteurs de risque: Vt bas ( p = 0,043) et PEP plus elevee ( p = 0,018). Aucune distension n’etait relevee dans le groupe sans atteinte respiratoire Discussion Ce premier recueil du SI en routine clinique revele jusqu’a 64 % de distension chez des patients atteints de SDRA malgre ventilation protectrice et limites de Pplat respectees, confirmant l’impossibilite en routine d’avoir une certitude de ventilation « protectrice ». D’autre part, le groupe de patients ayant des epanchements et/ou atelectasies, individualises dans la definition de Berlin, sont a haut risque de distension par la PEP. Enfin, les patients n’ayant pas d’atteinte respiratoire n’ont pas de distension lorsque ventiles a faible volume courant. En conclusion, le SI pourrait etre un outil de titration et de surveillance de la PEP chez les patients a risque de distension.

Understanding the development of the idea of universal human rights is the first step in reconceptualising human rights. Only by facing the historical and contingent nature of the concept can we make use of it in a way that is both just and philosophically sound. This approach is taken to be just because it avoids two a priori assumptions. First, that intellectual constructs developed by one dominant culture emerging from its own historically and philosophically contingent milieu are necessarily applicable to all societies and cultures. This assumption ignores and disdains the possibility that in and from other less dominant societies and cultures equally valuable constructs might be found and used. Second, a historically aware approach cautions against the opposite assumption: that it is not possible for an intellectual construct from one specific culture and society to emerge as the most effective tool for achieving a particular goal across all cultures. This approach is also taken to be philosophically sound, because the assumption that the concept of universal human rights is ahistorical and evident in all cultures and societies is demonstrably false. Any argument supporting the use of the concept of universal human rights premised on assumptions that can be empirically proven false will not be sustainable and may subvert its own cause. The argument developed in the remainder of the book is an attempt to construct a theoretical understanding of human rights that might act as a common framework in the context of pluralism. While the argument is premised on the recognition of the historicity and non-universality of concepts such as human rights, it is not an argument for cultural or philosophical relativism. Rather, it seeks to provide a framework within which competing non-relativist systems of thought and being can come to a place of agreement about how they live together.
