This study set out to evaluate the wound healing properties of brittle star extracts in vitro and in vivo. Due to the great arm regeneration potential of the brittle star, Ophiocoma cynthiae, the present study aimed to evaluate the wound healing effect of hydroalcoholic extracts of brittle star undergoing arm regeneration in wound healing models. The brittle star samples were collected from Nayband Bay, Bushehr, Iran. After wound induction in the arm of brittle stars, hydroalcoholic extracts relating to different times of arm regeneration were prepared. The GC-MS analysis, in vitro MTT cell viability and cell migration, Western blot, and computational analysis tests were performed. Based on the in vitro findings, two BSEs were chosen for in vivo testing. Macroscopic, histopathological and biochemical evaluations were performed after treatments. The results showed positive proliferative effects of BSEs. Specifically, forty-two compounds were detected in all groups of BSEs using GC-MS analysis, and their biological activities were assessed. The MTT assay showed that the 14 d BSE had a higher proliferative effect on HFF cells than 7 d BSE. The cell migration assay showed that the wound area in 7 d and 14 d BSEs was significantly lower than in the control group. Western blot analysis demonstrated an increase in the expression of proliferation-related proteins. Upon the computational analysis, a strong affinity of some compounds with proteins was observed. The in vivo analysis showed that the evaluation of wound changes and the percentage of wound healing in cell migration assay in the 7 d BSE group was better than in the other groups. Histopathological scores of the 7 d BSE and 14 d BSE groups were significantly higher than in the other groups. In conclusion, the hydroalcoholic extract of O. cynthiae undergoing arm regeneration after 7 and 14 days promoted the wound healing process in the cell and rat skin wound healing model due to their proliferative and migratory biological activity.

Background: This study aims to investigate the effects of carob (Ceratonia siliqua L.) pod extract (CPE) on the viability of human endometrial mesenchymal stromal/stem cells (EnMSCs) and its impact on mRNA and protein expressions of DNA methyltransferases (DNMT1, DNMT3A, and DNMT3B), histone deacetylase 1 (HDAC1), matrix metalloproteinase-2 (MMP2), and cyclooxygenase-2 (COX-2) in endometriotic patients. Materials and Methods: In this experimental study, EnMSCs were derived from endometrium of patients with ovarian endometrioma (OMA-EnMSCs group) and deep infiltrative endometriosis (DIE) samples of 10 endometriosis-associated infertility (EAI) women (E-EnMSCs group) and compared to EnMSCs derived from the endometrium of an endometriosis-free, normal woman as the control group (C-EnMSCs). The metabolic activity of the control and case groups were evaluated by treating them with different concentrations of CPE. Cell viability was analysed by MTT. Real-time reverse transcription-polymerase chain reaction (RT-PCR) and Western blot were used to evaluate the expression of specific genes at the mRNA and protein levels, respectively. Results: Treatment with 0.8 and 2 μg/mL of CPE downregulated COX-2 and HDAC1 in the E-EnMSC group compared to the C-EnMSCs group. Treatment with 0.8 μg/mL of CPE also decreased MMP2 and DNMT3B gene expressions. The COX-2 and DNMT3A genes were significantly upregulated after treatment with 2 μg/mL of CPE. Expressions of the COX-2, HDAC1, DNMT1, DNMT3A, and DNMT3B peptides decreased in the all three groups after treatment with 0.8 and 2 μg/mL of CPE. Gas chromatography-mass spectroscopy (GC-MS) analysis of CPE identified 14 bioactive compounds. Molecular docking showed the best position of each bioactive compound on the different target proteins that are involved in the process of apoptosis in EnMSCs. Conclusion: In vitro and in silico analyses of CPE bioactive compounds show that they may downregulate the cell inflammatory pathway involved in the pathophysiology of endometriosis. Much of the complexity and diversity found in nature is driven by nonlinear phenomena, and this holds true for the brain. Nonlinear dynamics theory has been successfully utilized in explaining brain functions from a biophysics standpoint, and the field of statistical physics continues to make substantial progress in understanding brain connectivity and function. This study delves into complex brain functional connectivity using biophysical nonlinear dynamics approaches. We aim to uncover hidden information in high-dimensional and nonlinear neural signals, with the hope of providing a useful tool for analyzing information transitions in functionally complex networks. By utilizing phase portraits and fuzzy recurrence plots, we investigated the latent information in the functional connectivity of complex brain networks. Our numerical experiments, which include synthetic linear dynamics neural time series and a biophysically realistic neural mass model, showed that phase portraits and fuzzy recurrence plots are highly sensitive to changes in neural dynamics and can also be used to predict functional connectivity based on structural connectivity. Furthermore, the results showed that phase trajectories of neuronal activity encode low-dimensional dynamics, and the geometric properties of the limit-cycle attractor formed by the phase portraits can be used to explain the neurodynamics. Additionally, our results showed that the phase portrait and fuzzy recurrence plots can be used as functional connectivity descriptors, and both metrics were able to capture and explain nonlinear dynamics behavior during specific cognitive tasks. In conclusion, our findings suggest that phase portraits and fuzzy recurrence plots could be highly effective as functional connectivity descriptors, providing valuable insights into nonlinear dynamics in the brain. In this research, rock mass deformation modulus (Emass), classification of rock mass, cohesion (c) and internal friction angle (Φ), and intact rock properties of the Khersan 2 dam site were investigated. Factors affecting mechanical properties were investigated and compared with the results of previous studies. To assess the shear strength and the Hoek and Brown failure criteria parameters, the triaxial test was performed on samples under saturated conditions. The Q-system and rock mass rating (RMR) were calculated for five-metre sections of boreholes. Results of the Mohr–Coulomb failure criterion showed that the limestones have a mean Φ of 40 degrees and a mean cohesion of 1 MPa, marl limestones have an average of 30 degrees and cohesion of 0.50 MPa, and anhydrite and gypsum showed an average of 26 degrees and 0.10 MPa. The results showed that c and Φ increase with the increase in the shear box size. Based on the RMR and Q-system the site was classified in good class. The mean of Emass showed that the dilatometry test gives the lowest value. Linear and logarithmic relationships were established between RMR89 with compressional wave velocity (Vp) (R2 = 0.95 and RMSE = 1.63) and Q-system (R2 = 0.95 and RMSE = 2.05), respectively. In this paper, the plasma parameters of hydrogen glow discharge were measured using Langmuir probe system. To measure these parameters in Damavand tokamak, different parts of the Langmuir probe were designed and fabricated. The electrode used in this system is a tungsten wire with diameter and length of 0.1 and 6 mm, respectively. The electrical circuit constructed for this system is capable of supplying voltage and current in the order of 400 V and 1 A, and can produce triangular pulses in the frequency range of 10 Hz to 10 kHz, respectively. Glow discharge hydrogen plasma, which is used for vacuum vessel cleaning and conditioning, is produced by two electrodes located in the lower part of the Damavand tokamak with the voltage of 450 V, current of 1.5 A and pressure of 10-3 Torr. By applying voltage to the electrode of probe, in the range of 200 to 350 V, the current drawn from the probe was measured. By interpreting the current-voltage characteristic, the plasma parameters were determined. The electron temperature, plasma density, plasma potential, and Debye length of the hydrogen glow discharge measured using this system are 5.8e V, 1.17×1015 m-3, 236 V and 523 μm, respectively.
