Background: Cell-free DNA (cfDNA) is present in healthy individuals but is elevated in those undergoing physical exertion, trauma, sepsis, and certain cancers. Maintaining cfDNA concentrations is vital for immune homeostasis and preventing inflammatory responses. Understanding cfDNA release and clearance is essential for using cfDNA as a biomarker in clinical diagnostics. We focused on the fragment size of cfDNA and investigated cfDNA dynamics and half-life, particularly the 100–250 base pair fragments. Methods: Healthy, adult men (n = 5; age 40 ± 4.1 years) were subjected to a 30 min treadmill exercise. Blood samples were collected at 0, 5, 10, 15, 30, and 60 min post-exercise using PAXgene® Blood ccfDNA tubes to stabilize and prevent nuclease-mediated cfDNA degradation and minimize genomic DNA contamination risk. The cfDNA concentration was measured using an electrophoresis-based technique (4150 TapeStation system) to quantify the concentration based on cfDNA fragment size. Results: The results showed a cfDNA half-life of 24.2 min, with a transient increase in 100–250 base pair cfDNA fragments post-exercise, likely due to nuclease activity. These levels rapidly reverted to the baseline within an hour. Conclusions: The rapid clearance of cfDNA underscores its potential as a biomarker for real-time disease monitoring and the evaluation of treatment efficacy. This study is expected to standardize cfDNA investigations, enhancing diagnosis and treatment monitoring across various disease conditions.

This study investigated the effects of ankle dorsiflexion angle adjustments in ankle–foot orthoses (AFOs) on the gait of healthy individuals. Fifteen healthy participants engaged in treadmill walking tasks while wearing AFOs with dorsiflexion angles set at 0°, 5°, 10°, and 15°. Three-dimensional treadmill gait analysis was used to collect data during treadmill walking. The analysis focused on toe clearance and the contribution of the vertical component of limb shortening (LS), and compared them across different dorsiflexion angles. The results indicated a significant increase in toe clearance at 10° (median [interquartile change]: 5.03 [0.90] vs. 5.98 [1.18], p < 0.01) and 15° (5.03 [0.90] vs. 5.82 [1.11], p < 0.01) dorsiflexion angle conditions compared to the 0° condition. Similarly, LS demonstrated significant increases at 10° (4.89 [1.97] vs. 5.87 [1.31], p < 0.01) and 15° (4.89 [1.97] vs. 5.61 [1.65], p < 0.01) conditions when compared with the 0° condition. These findings support the notion that higher dorsiflexion angles in AFOs lead to increased toe clearance and LS, suggesting the effectiveness of dorsiflexion angle adjustments as a strategy to address reduced toe clearance.

We report the results of a search for MeV-scale astrophysical neutrinos in KamLAND presented as an excess in the number of coincident neutrino interactions associated with the publicly available high-energy neutrino datasets from the IceCube Neutrino Observatory. We find no statistically significant excess in the number of observed low-energy electron antineutrinos in KamLAND, given a coincidence time window of ±500 s, ±1,000 s, ±3,600 s, and ±10,000 s around each of the IceCube neutrinos. We use this observation to present limits from 1.8 MeV to 100 MeV on the electron antineutrino fluence, assuming a mono-energetic flux. We then compare the results to several astrophysical measurements performed by IceCube and place a limit at the 90% confidence level on the electron antineutrino isotropic thermal luminosity from the TXS 0506+056 blazar.

We report the case of a 66-year-old woman who presented with diarrhea and weight loss approximately 14 months after unrelated allogeneic bone marrow transplantation for acute myeloid leukemia. Her early post-transplant course was notable for mild acute skin graft-versus-host disease (GVHD) and biopsy-proven upper gastrointestinal (GI) acute GVHD, both of which resolved with treatment. She then developed weight loss and diarrhea treated with prednisolone for what was thought to be GI late acute GVHD. However, her diarrhea and weight loss persisted. Colonoscopy showed a grossly intact mucosa, and stool studies only confirmed steatorrhea. However, an atrophic pancreas was found on an abdominal computed tomography (CT) scan. Exocrine pancreatic enzymes, such as lipase and pancreatic amylase, were markedly decreased, yet pancreatic endocrine function remained intact. The patient’s diarrhea and weight loss improved upon treatment with pancrelipase. Therefore, we suggest that her exocrine pancreatic insufficiency was likely partly caused by atypical chronic GVHD.

Abstract We report an Arase‐all sky imager (ASI) conjugate event in which the pulsating aurora (PsA) has a one‐to‐one correspondence with chorus bursts. Wavelet analysis displayed three peaks at ∼0.3 Hz, 4 Hz, and >10 Hz, corresponding to the main pulsation, internal modulation, and fast modulation, respectively. These correspond to the old terms of ∼5–15 s pulsations, chorus risers/elements and subelements/subpackets, respectively. Electron “microbursts” correspond to the 4‐Hz peak. The internal and fast modulations are further verified by the analysis based on fast Fourier transform analyses. Moreover, the spatial distributions of the Fourier spectral amplitude show that the internal and fast modulations are well‐structured within auroral patches. The above results indicate a paradigm shift away from quasilinear theory which implicitly assumes diffuse wave generation. The three time‐scale modulations are consistent with coherent chorus which has been theoretically argued to lead to pitch angle transport three orders of magnitude faster.

IntroductionElucidating the enduring effects of childhood social capital is vital for shaping future well-being. Here, we investigate the impact of childhood social capital (SC) and parental engagement on adult psychological well-being and cognitive performance.MethodsUtilizing a cohort of 292 Japanese university students, we assessed the influences of SC and parental time during childhood on adult outcomes. Participants retrospectively reported their early childhood experiences, focusing on social interactions and parental involvement.ResultsOur findings reveal a significant positive correlation between childhood SC and adult subjective well-being, particularly in its positive dimensions. Additionally, maternal involvement during childhood is associated with cognitive abilities in adulthood, as measured by Raven’s Advanced Progressive Matrices. Hierarchical multiple linear regression analysis highlights the substantial impact of childhood SC on adult well-being, elucidating the intricate interplay between social and parental contributions to developmental trajectories.DiscussionThese results highlight the crucial roles of both parental and non-parental figures in fostering social, emotional, and cognitive development from childhood to adulthood, underscoring the importance of nurturing supportive relationships throughout early life to promote positive developmental outcomes.

Introduction Reducing neonatal deaths in premature infants in low- and middle-income countries is key to reducing global neonatal mortality. International neonatal networks, along with patient registries of premature infants, have contributed to improving the quality of neonatal care; however, the involvement of low-to-middle-income countries was limited. This project aims to form an international collaboration among neonatal networks in Asia (AsianNeo), including low-, middle- and high-income countries (or regions). Specifically, it aims to determine outcomes in sick newborn infants, especially very low birth weight (VLBW) infants or very preterm infants, with a view to improving the quality of care for such infants.Methods and analysis Currently, AsianNeo comprises nine neonatal networks from Indonesia, Japan, Malaysia, Philippines, Singapore, South Korea, Sri Lanka, Taiwan and Thailand. AsianNeo will undertake the following four studies: (1) institutional questionnaire surveys investigating neonatal intensive care unit resources and the clinical management of sick newborn infants, with a focus on VLBW infants (nine countries/regions); (2) a retrospective cohort study to describe and compare the outcomes of VLBW infants among Asian countries and regions (four countries/regions); (3) a prospective cohort study to develop the AsianNeo registry of VLBW infants (six countries/regions); and (4) implementation and evaluation of educational and quality improvement projects in AsianNeo countries and regions (nine countries/regions).Ethics and dissemination The study protocol was approved by the Research Ethics Board of the National Center for Child Health and Development, Tokyo, Japan (reference number 2020–244, 2022–156). The study findings will be disseminated through educational programmes, quality improvement activities, conference presentations and medical journal publications.

IntroductionThis study examined whether visuospatial perspective uses the character perspective during narrative comprehension.MethodParticipants read narrative stimuli depicting the spatial positional relationships between characters and objects and judged whether the objects were on the left or right from the character's perspective. We manipulated whether the spatial positional relationships between characters depicted in the narrative stimuli resulted in a visuospatial perspective. We hypothesized that the high-load perspective-taking condition would indicate longer reaction times compared to the low-load perspective-taking condition, as shifting perspectives between characters in the high-load condition require more time for visuospatial perspective-taking.ResultsAs predicted, the reaction time was longer for high-load perspective-taking than for low-load perspective-taking.DiscussionDuring narrative comprehension, the reaction time for visuospatial perspective-taking must move virtually within the representation from the main character's perspective to that of another character. Visuospatial perspective-taking is involved in narrative comprehension.

Amidst increasing water resource instability in various regions, the necessity for water demand-side management (WDM) to ensure sustainable water use has grown. Recent advancements in technology have revealed the potential of various methodologies. This study examined the effect of financial incentives in the form of rebates for water conservation and nudges using a public goods approach, as well as a combination of the two, on water-saving behaviors, assuming a temporary water resource shortage. We utilized visual nudges as an intrinsic motivation recognizing water as a public good. The findings revealed that financial incentives reduced water usage in low-consumption households but had no discernible effect in high-consumption households. Although financial incentive and nudge were not synergistic, they showed negative interaction effects in high-consumption households. Participants from low-consumption households conserved water to save money on their water bills, with their already low water consumption likely due to their desire to keep their bills low. Consequently, a financial incentive in the form of a rebate for water conservation might further decrease water use. In a follow-up experiment to determine the cognitive mechanism, economic interventions like raising water tariffs significantly increased the motivation of low-consumption households to reduce their future water use. Recognizing the complexity of effective WDM, further accumulation of knowledge is needed for effective WDM.

Summary: The mechanisms governing autophagy of proteins and organelles have been well studied, but how other cytoplasmic components such as RNA and polysaccharides are degraded remains largely unknown. In this study, we examine autophagy of glycogen, a storage form of glucose. We find that cells accumulate glycogen in the cytoplasm during nitrogen starvation and that this carbohydrate is rarely observed within autophagosomes and autophagic bodies. However, sequestration of glycogen by autophagy is observed following prolonged nitrogen starvation. We identify a yet-uncharacterized open reading frame, Yil024c (herein Atg45), as encoding a cytosolic receptor protein that mediates autophagy of glycogen (glycophagy). Furthermore, we show that, during sporulation, Atg45 is highly expressed and is associated with an increase in glycophagy. Our results suggest that cells regulate glycophagic activity by controlling the expression level of Atg45.

The decay of the primordial isotopes 238U, 235U, 232Th, and 40K has contributed to the terrestrial heat budget throughout the Earth's history. Hence, the individual abundance of those isotopes are key parameters in reconstructing contemporary Earth models. The geoneutrinos produced by the radioactive decays of uranium and thorium have been observed with the Kamioka Liquid-Scintillator Antineutrino Detector (KamLAND). Those measurements have been improved with more than 18-year observation time, and improvement in detector background levels mainly with an 8-year nearly reactor-free period, which now permit spectroscopy with geoneutrinos. Our results yield the first constraint on both uranium and thorium heat contributions. The KamLAND result is consistent with geochemical estimations based on elemental abundances of chondritic meteorites and mantle peridotites. The High-Q model is disfavored at 99.76% C.L. and a fully radiogenic model is excluded at 5.2σ assuming a homogeneous heat producing element distribution in the mantle.

We present the results of a search for core-collapse supernova neutrinos, using long-term KamLAND data from 2002 March 9 to 2020 April 25. We focus on the electron antineutrinos emitted from supernovae in the energy range of 1.8-111 MeV. Supernovae will make a neutrino event cluster with the duration of ∼10 s in the KamLAND data. We find no neutrino clusters and give the upper limit on the supernova rate to be 0.15 yr−1 with a 90% confidence level. The detectable range, which corresponds to a >95% detection probability, is 40-59 kpc and 65-81 kpc for core-collapse supernovae and failed core-collapse supernovae, respectively. This paper proposes to convert the supernova rate obtained by the neutrino observation to the Galactic star formation rate. Abstract 5′ adenosine monophosphate–activated protein kinase–related kinase 5 (ARK5) is involved in mitochondrial ATP production and associated with poor prognosis of multiple myeloma (MM). However, the molecular mechanisms of ARK5 in MM remain largely unknown. This study examined the pathogenic role of ARK5 in mitochondria by using genetically modified isogenic cell clones with or without ARK5 in human myeloma cell lines, KMS-11 and Sachi, which overexpress ARK5. The biallelic knockout of ARK5 (ARK5-KO) inhibited cell proliferation, colony formation, and migration with increased apoptosis. Mitochondrial fusion was enhanced in ARK5-KO cells, unlike in ARK5 wild-type (ARK5-WT) cells, which exhibited increased mitochondrial fission. Furthermore, ARK5-KO cells demonstrated a lower phosphorylated dynamin–related protein 1 at serine 616, higher protein expression of mitofusin-1 (MFN1) and MFN2, optic atrophy 1 with a lower level of ATP, and higher levels of lactate and reactive oxygen species than ARK5-WT cells. Our findings suggest that ARK5-enhanced myeloma cells can survive associated mitochondrial fission and activity. This study first revealed the relationship between ARK5 and mitochondrial morphological dynamics. Thus, our outcomes show novel aspects of mitochondrial biology of ARK5, which can afford a more advanced treatment approach for unfavorable MM expressing ARK5.

We investigated imaging conditions for the distribution of lutetium oxodotreotide (Lu-177 DOTATATE) in the body during peptide receptor radionuclide therapy for neuroendocrine tumor (NET). We investigated imaging conditions using gamma rays emitted from the radionuclide. The gamma rays had energy peaks at 113 and 208 keV and characteristic X-rays at 56 keV. Image quality was compared by utilizing a combination of low–medium-energy general-purpose (LMEGP) and medium-energy general-purpose (MEGP) collimators. This study included the measurement of total spatial resolution (Full Width at Half Maximum) using a line source phantom. We compared the image quality of static images using a plane phantom and SPECT images using a cylindrical phantom. This comparison involved assessing recovery coefficient curves, count ratio, and %CV. Imaging evaluation was also performed on one NET patient. In phantom studies and the clinical study, comparing the combination of the three energy peaks (56 + 113 + 208 keV) using the LMEGP collimator and the conventional combination (113 + 208 keV) using the MEGP collimator revealed a count ratio of 1.9 times the maximum, stable %CV, and the best image quality.
