Abstract In buffalo (Bubalus bubalis) reproductive seasonality, causing cycles of milk production, is one of the major factors affecting farming profitability. Follicular fluid (FF) contains extracellular vesicles (EVs) playing an important role in modulating oocyte developmental competence and carrying microRNAs (miRNAs) essential for in vitro fertilization outcomes. The aim of this work was to characterize the FF-EVs-miRNA cargo of antral (An) and preovulatory (pO) follicles collected in the breeding (BS) and non-breeding (NBS) seasons, to unravel the molecular causes of the reduced oocyte competence recorded in buffalo during the NBS. The increasing number of objects in Earth orbit has encouraged the development of space surveillance and tracking (SST) applications. A critical aspect of SST is the identification and characterization of close encounters between pairs of space objects. The present work introduces a tool for the analysis of conjunctions, consisting of several modules. The first module, which has been shown to greatly speed up the process, employs a series of geometric and temporal filters to shorten the list of potential colliding pairs. The remaining objects are then propagated to compute important parameters such as time of closest approach (TCA), miss distance (MD), and probability of collision (PoC), the latter using three different methods. When a conjunction assessment returns an MD or a PoC that exceeds predefined alert thresholds, the algorithm enables the planning of an impulsive collision avoidance maneuver (CAM) at specific maneuver epochs. CAM candidates are determined using an analytical Keplerian approach, with the goal of achieving the desired PoC or MD. The user can then verify the performance of a specific candidate through perturbed propagation, and the MD and PoC are recalculated after the maneuver to ensure that they meet the desired thresholds. In conclusion, this paper evaluates the performance of the tool using synthetic and real data, providing valuable insights into its effectiveness.

In this study, the effect of increasing exposure time on the microstructures, porosity, mechanical properties and corrosion behavior of selective laser melted sample Al-Si10-Mg powder was investigated. The samples were processed at the same power (375 W) and scan speed (2000 mm/s), but with increasing exposure time. Exposure time equal to 40, 50 and 60 µs was applied. The features of the analyzed samples show that with increasing exposure time, greater efficiency of the heat input was obtained, with a larger size of the melt pool and Si particles and lower porosity. Specifically, at the highest exposure time the melt pool showed an increase of 19% in width and 48% in depth, while the volume percentage of the voids decreased by 50% with respect to the lowest exposure time. Moreover, with the coarser microstructure being associated with a lower level of voids, the average hardness is similar for the analyzed samples. Corrosion resistance was evaluated, being one of the most important properties that may affect the service performance of Al-Si10-Mg alloy in the aerospace, marine and automotive industries. The potentiodynamic curves of the samples show that the voids occurrence is more significant with respect to the scale of the microstructure on corrosion behavior, with the sample processed at the highest exposure time being the more resistant to corrosion. The experimental techniques used in the present study were Optical Microscope (OM), Scanning Electron Microscope (SEM), hardness and X-Ray Computed Tomography.

Environmental monitoring for SARS-CoV-2 has become a useful adjunct to clinical testing because it is widely available and relatively inexpensive. During the period May–December 2022 (spring–summer: May–September–autumn: October–December), we assessed the presence and viability of the virus on surfaces in university settings in the Apulia region (Southern Italy) after the resumption of face-to-face teaching activities and evaluated surface monitoring as an early warning system. The sampling plan provided for the selection of 75% of the surface types (e.g., student and teacher desks, computer, handrail) in different materials (plasticized wood, wood, metal, plastic) present in different environments. Overall, 5.4% of surfaces (all students’ desks) resulted in positive with RT-PCR and negative with viral culture. Greater contamination was found in the spring–summer period than in the autumn (χ2 test with Yates correction = 7.6003; p-value = 0.006). The Poisson regression model showed a direct association between the average number of COVID-19 cases among university students in the seven days following sampling and the percentage of SARS-CoV-2 positive swabs on sampling day and (Intercept = 5.32498; β = 0.01847; p < 0.001). Our results show that environmental monitoring for SARS-CoV-2, especially in crowded settings such as universities, could be a useful tool for early warning, even after the end of the COVID-19 emergency.

Adolescent idiopathic scoliosis (AIS) is a lateral, rotated curvature of the spine. It is a 3-dimensional deformity that arises in otherwise healthy children at or around puberty. AIS is the most common form of scoliosis in the pediatric population. The etiology is multifactorial, including genetic and environmental factors. The incidence is roughly equal between males and females, while there is a higher risk of progression in females. Guidelines for AIS treatment identify three levels of treatment: observation, physiotherapy scoliosis-specific exercises, and braces. In this paper, we carried out a review of the scientific literature about the indication and success rates of the braces provided for free by the National Health Service in Italy (SSN). Despite a general consensus on the efficacy of rigid bracing treatment and its use in AIS, an important heterogeneity about the treatment is present in the scientific literature, demonstrating a high degree of variability. The overall success rate of the braces provided by the SSN is high, suggesting an important therapeutic role in the treatment of AIS. Robust guidelines are needed to ensure uniform and effective treatments.

Abstract Background Tanimilast is a novel and selective inhaled inhibitor of phosphodiesterase-4 in advanced clinical development for chronic obstructive pulmonary disease (COPD). Tanimilast is known to exert prominent anti-inflammatory activity when tested in preclinical experimental models as well as in human clinical studies. Recently, we have demonstrated that it also finely tunes, rather than suppressing, the cytokine network secreted by activated dendritic cells (DCs). This study was designed to characterize the effects of tanimilast on T-cell polarizing properties of DCs and to investigate additional functional and phenotypical features induced by tanimilast. Methods DCs at day 6 of culture were stimulated with LPS in the presence or absence of tanimilast or the control drug budesonide. After 24 h, DCs were analyzed for the expression of surface markers of maturation and activation by flow cytometry and cocultured with T cells to investigate cell proliferation and activation/polarization. The regulation of type 2-skewing mediators was investigated by real-time PCR in DCs and compared to results obtained in vivo in a randomized placebo-controlled trial on COPD patients treated with tanimilast. Results Our results show that both tanimilast and budesonide reduced the production of the immunostimulatory cytokine IFN-γ by CD4+ T cells. However, the two drugs acted at different levels since budesonide mainly blocked T cell proliferation, while tanimilast skewed T cells towards a Th2 phenotype without affecting cell proliferation. In addition, only DCs matured in the presence of tanimilast displayed increased CD86/CD80 ratio and CD141 expression, which correlated with Th2 T cell induction and dead cell uptake respectively. These cells also upregulated cAMP-dependent immunosuppressive molecules such as IDO1, TSP1, VEGF-A and Amphiregulin. Notably, the translational value of these data was confirmed by the finding that these same genes were upregulated also in sputum cells of COPD patients treated with tanimilast as add-on to inhaled glucocorticoids and bronchodilators. Conclusion Taken together, these findings demonstrate distinct immunomodulatory properties of tanimilast associated with a type 2 endotype and CD141 upregulation in DCs and provide a mechanistic rationale for the administration of tanimilast on top of inhaled corticosteroids.
