We review 69 studies of the diet of Badgers Meles meles L. in the former Soviet Union, using information from 58 Russian-language papers. Animal foods occurred more frequently in the diet than vegetal foods and constituted, on average, 62% of all intake by volume. The most important individual food categories were insects (30% by volume) and small mammals (20% by volume), which appear to be staple foods of Badgers over virtually the whole of the former Soviet Union. Earthworms were mentioned in only a few reports and never exceeded 5% of the diet by volume. Other animal foods included birds, reptiles, amphibians, fish, molluscs and carrion. Fruits were seasonally important in some areas while roots, tubers, bulbs, cereals and nuts were also eaten; but in general, the Soviet literature lacks detailed information about vegetal foods. Many authors comment on seasonal variation in the diet and there was statistically significant seasonal variation in the frequency with which small mammals, reptiles, insects and vegetal foods were eaten. No largescale geographical trends were apparent in the composition of the diet, but foods which were of minor importance overall, such as reptiles, amphibians and molluscs, were sometimes eaten with high frequency in particular localities. The results support the view that Badgers are opportunistic, generalist foragers, with a preference for animal foods when these are available.

We examine phytoplankton community structure and how it is influenced by commercial tilapia (Oreochromis niloticus L.) cage farming in three tropical lakes in eastern Brazil. Sampling occurred during both the wet and dry seasons for two treatments – within tilapia cages and outside the cages in the lake. Total ammonia, total phosphorus, chlorophyll a , phycocyanin and phytoplankton were measured in all water samples. In the phytoplankton community, we estimated species richness and total abundance. Thirty-three genera of algae distributed in 8 classes were identified. The most abundant classes were Chlorophyceae and Cyanophyceae (the latter of which was the most species-rich class). In Palmas Lake, chlorophyll a was constant over time (summer and winter) and location (within and outside cage farms), which may reflect the fact that fish farming is more recent in this lake. The richness and abundance of species were similar among the lakes, but Palminhas Lake tended to have more extreme values, apparently associated with the longer time fish have been farmed there. Fish farming clearly influences water quality as measured by algae species richness and abundance but is also moderated by the unique conditions in each lake and the amount of time over which the fish have been farmed. Understanding and preventing the impacts and detrimental consequences of tilapia cage farming on both the farming yield and the water quality in tropical lakes require continual monitoring.

Trisomy 21 (Ts21) affects craniofacial precursors in individuals with Down syndrome (DS). The resultant craniofacial features in all individuals with Ts21 may significantly affect breathing, eating and speaking. Using mouse models of DS, we have traced the origin of DS-associated craniofacial abnormalities to deficiencies in neural crest cell (NCC) craniofacial precursors early in development. Hypothetically, three copies of Dyrk1a (dual-specificity tyrosine-(Y)-phosphorylation regulated kinase 1A), a trisomic gene found in most humans with DS and mouse models of DS, may significantly affect craniofacial structure. We hypothesized that we could improve DS-related craniofacial abnormalities in mouse models using a Dyrk1a inhibitor or by normalizing Dyrk1a gene dosage. In vitro and in vivo treatment with Epigallocatechin-3-gallate (EGCG), a Dyrk1a inhibitor, modulated trisomic NCC deficiencies at embryonic time points. Furthermore, prenatal EGCG treatment normalized some craniofacial phenotypes, including cranial vault in adult Ts65Dn mice. Normalization of Dyrk1a copy number in an otherwise trisomic Ts65Dn mice normalized many dimensions of the cranial vault, but did not correct all craniofacial anatomy. These data underscore the complexity of the gene–phenotype relationship in trisomy and suggest that changes in Dyrk1a expression play an important role in morphogenesis and growth of the cranial vault. These results suggest that a temporally specific prenatal therapy may be an effective way to ameliorate some craniofacial anatomical changes associated with DS.

Vaccination is potentially one of the most valuable methods of control against the caligid copepod Lepeophtheirus salmonis , the salmon louse, a major pathogen of farmed salmonids in the Northern hemisphere. A search for suitable potential antigens from this complex metazoan parasite forms the basis of this study. Salmon louse homogenate was separated by column chromatography into specific fractions which were assayed for a range of enzyme activities. Fractions found to be associated with enzymic activity or peaks of UV absorbance were used to raise antisera in 44 Wistar rats. An immunocytochemical investigation of the midgut of the adult louse using these specific antisera revealed immunostaining of the lumenal and basal surfaces of the louse gut. It is suggested that the proteins in the chromatographic fractions may represent digestive enzymes and/or structural or functional components of the salmon louse gut.

Multiresistant Klebsiella pneumoniae strains with plasmid-borne extended-spectrum beta-lactamases (ESBL) are increasingly frequent nosocomial pathogens. A major outbreak of clinical infections, mainly involving patients in the Newborn Services Unit with limited spread to adult patients, occurred at our hospital. This epidemic was investigated by typing the isolates phenotypically and with random amplified polymorphic DNA analysis (RAPD) and plasmid analysis. Forty-eight isolates, consisting of 44 consecutive clinical isolates and 4 selected surveillance isolates, were studied. A single decamer primer was used for the RAPD, and this was effective in demonstrating that the majority of isolates (45 of 48) had the same profile. Three other isolates had different RAPD patterns identifying them as nonepidemic strains. Plasmids were extracted by alkaline lysis with Magic-miniprep kits from 10 isolates selected to represent the epidemic and nonepidemic strains. This method produced small (< 20-kb) plasmids; larger ESBL-carrying plasmids were not produced, but the small plasmids nonetheless allowed strain differentiation. Antibiotic susceptibility patterns alone were not reliable as strain indicators, since some isolates with the RAPD pattern characteristic of the epidemic strains did not express ESBL and therefore were susceptible to extended-spectrum cephalosporins. The investigation showed the predominance of a single epidemic strain that was transmitted between patients in the Newborn Services Unit. RAPD was the best of the methods used for detecting strain differences, and its speed and ability to type a wide variety of species suggest that it will be an increasingly useful molecular epidemiologic tool.

The olfactory systems of birds resemble those of amphibians, reptiles and mammals (for reviews see Kare & Mason 1986, Wenzel 1987) but the importance of olfaction in the daily lives of birds remains a matter of controversy (Roper 1999). Nevertheless, there is now good behavioural evidence of olfactory capability in birds of a variety of species and of its use in several different functional contexts including orientation, foraging and reproduction (Jones & Roper 1997, Roper 1999). Thus, it seems increasingly likely that some degree of olfactory sensitivity is widespread in birds. The present study tests ability to discriminate odour cues in two captive Yellow-backed Chattering Lories Lorius garrulus flavopalliatus. I chose this species because it feeds on nectar, flowers and fruit and so may use olfaction for foraging purposes. This is the first behavioural investigation of olfactory capability in any parrot species.

Down syndrome (DS) and Fetal Alcohol Syndrome (FAS) are two leading causes of birth defects with phenotypes ranging from craniofacial abnormalities to cognitive impairment. Despite different origins, we report that in addition to sharing many phenotypes, DS and FAS may have common underlying mechanisms of development.Literature was surveyed for DS and FAS as well as mouse models. Gene expression and apoptosis were compared in embryonic mouse models of DS and FAS by qPCR, immunohistochemical and immunoflurorescence analyses. The craniometry was examined using MicroCT at postnatal day 21.A literature survey revealed over 20 comparable craniofacial and structural deficits in both humans with DS and FAS and corresponding mouse models. Similar phenotypes were experimentally found in pre- and postnatal craniofacial and neurological tissues of DS and FAS mice. Dysregulation of two genes, Dyrk1a and Rcan1, key to craniofacial and neurological precursors of DS, was shared in craniofacial precursors of DS and FAS embryos. Increased cleaved caspase 3 expression was also discovered in comparable regions of the craniofacial and brain precursors of DS and FAS embryos. Further mechanistic studies suggested overexpression of trisomic Ttc3 in DS embyros may influence nuclear pAkt localization and cell survival.This first and initial study indicates that DS and FAS share common dysmorphologies in humans and animal models. This work also suggests common mechanisms at cellular and molecular levels that are disrupted by trisomy or alcohol consumption during pregnancy and lead to craniofacial and neurological phenotypes associated with DS or FAS.

The possibility of arsenic release and the potential role of Geobacter in arsenic biogeochemistry during in situ uranium bioremediation was investigated because increased availability of organic matter has been associated with substantial releases of arsenic in other subsurface environments. In a field experiment conducted at the Rifle, CO study site, groundwater arsenic concentrations increased when acetate was added. The number of transcripts from arrA, which codes for the α-subunit of dissimilatory As(V) reductase, and acr3, which codes for the arsenic pump protein Acr3, were determined with quantitative reverse transcription-PCR. Most of the arrA (>60%) and acr3-1 (>90%) sequences that were recovered were most similar to Geobacter species, while the majority of acr3-2 (>50%) sequences were most closely related to Rhodoferax ferrireducens. Analysis of transcript abundance demonstrated that transcription of acr3-1 by the subsurface Geobacter community was correlated with arsenic concentrations in the groundwater. In contrast, Geobacter arrA transcript numbers lagged behind the major arsenic release and remained high even after arsenic concentrations declined. This suggested that factors other than As(V) availability regulated the transcription of arrA in situ, even though the presence of As(V) increased the transcription of arrA in cultures of Geobacter lovleyi, which was capable of As(V) reduction. These results demonstrate that subsurface Geobacter species can tightly regulate their physiological response to changes in groundwater arsenic concentrations. The transcriptomic approach developed here should be useful for the study of a diversity of other environments in which Geobacter species are considered to have an important influence on arsenic biogeochemistry.

Recent work has demonstrated that attention can be configured to multiple potential targets in spatial search. However, this previous work relied on a fixed set of targets across multiple trials, allowing observers to offload attentional control settings to longer-term representations. In the present experiments, we demonstrate multiple attentional control settings that operate independently of space (Experiments 1 and 2). More important, we show that observers can be cued to different control settings on a trial-by-trial basis (Experiment 3). The latter result suggests that observers were capable of maintaining multiple control settings when the demands of the task required an attentional search for specific feature values. Attention can be configured to extract multiple feature values in a goal-directed manner, and this configuration can be can be dynamically engaged on a trial-by-trial basis. These results support recent findings that reveal the high precision, complexity, and flexibility of attentional control settings.

The possibility of assessing endogenous adrenal activity in the collared peccary (Pecari tajacu) was tested by using an adrenocorticotropic hormone (ACTH) challenge in a fecal glucocorticoid metabolite (FGM) assay. Feces were collected from 12 captive adult male peccaries beginning 48 hr prior to challenge; six of these animals received the challenge as an ACTH injection and the other six were injected with saline solution. Feces collection ended 120 hr after injections. As a control, feces were collected for eight consecutive days from another six adult male peccaries that remained in their original mixed-sex herds in semiconfined paddocks. All feces samples were freeze-dried, extracted by an ethanol vortex method, and assayed for glucocorticoids by means of an enzyme immunoassay. FGM concentrations were compared between the treatments by a repeated measures analysis of variance (ANOVA) followed by a post hoc Tukey test. The assay is reliable but, instead of the usual proportion of 1:50 in ethanol (fecal mass:solvent), 1:10 is recommended for best extraction of FGM. Baseline FGM concentrations were similar among the ACTH, saline, and control treatments (29.7 +/- 11.2 ng/g(-1) dry feces) during the 48 hr before the challenge. The ACTH group reached an FGM excretion peak at 24 hr post-treatment, followed by a decline, while in the control and saline groups FGM levels remained relatively constant. Therefore, the fecal glucocorticoid metabolite assay reflects endogenous adrenal activity in the collared peccary and is a powerful tool for noninvasive stress monitoring in peccaries.

The effect of gentamicin upon planktonic cultures of Pseudomonas aeruginosa, Escherichia coli, Staphylococcus epidermidis, and Staphylococcus aureus was measured with and without application of 67-kHz ultrasonic stimulation. The ultrasound was applied at levels that had no inhibitory or bactericidal activity against the bacteria. Measurements of the MIC and bactericidal activity of gentamicin against planktonic cultures of P. aeruginosa and E. coli demonstrated that simultaneous application of 67-kHz ultrasound enhanced the effectiveness of the antibiotic. A synergistic effect was observed and bacterial viability was reduced several orders of magnitude when gentamicin concentrations and ultrasonic levels which by themselves did not reduce viability were combined. As the age of the culture increased, the bacteria became more resistant to the effect of the antibiotic alone. Application of ultrasound appeared to reverse this resistance. The ultrasonic treatment-enhanced activity was evident with cultures of P. aeruginosa and E. coli but was not observed with cultures of gram-positive S. epidermidis and S. aureus. These results may have application in the treatment of bacterial biofilm infections on implant devices, which infections are usually more resistant to antibiotic therapy.

Although most authors characterize the badger Meles meles as an omnivore, the species is sometimes described as a food specialist and in particular as a specialist predator on earthworms Lumbricus terrestris. The validity of the‘food specialist’hypothesis is examined, using data from 11 quantitative studies of badger diet to test two predictions: 1) any one population of badgers relies to a disproportionate extent on a single type of food; and 2) consumption of the predominate food is independent of its availability. As regards the first prediction, when data on year-round consumption were corrected for seasonal variation in total intake, no study showed earthworms to comprise more than 50% of the diet and in three studies they accounted for no more than 5% of intake. In two studies, fruit comprised more than 50% of the diet and fruit and insects together accounted for 95–97% of total consumption. Compared across different studies, consumption of any one food, including earthworms, varied more or less continuously across a wide spectrum of values, providing no support for the idea that a given food can be characterized as being of either major or minor importance. With respect to the second prediction, there was no evidence that consumption of either earthworms or any other food was consistently invariant across seasons; and in any case, the idea that badger diet contains a constant proportion of any one food does not make functional sense. Taken together with information about the morphology of the digestive system and a priori arguments about the conditions in which food specialization is likely to be adaptive, studies of diet confirm that the badger is best viewed as a generalist or opportunist feeder.

Porphyrins with two fused dihydronaphtho units on the opposite or adjacent pyrrole rings have been prepared by the MacDonald condensation or by the cyclization of suitably substituted a,c,-biladienes; dehydrogenation with 2 equivalents of DDQ afforded the corresponding dinaphthoporphyrins in excellent yields.

Eight badger (Meles meles L.) Setts (two main setts, four subsidiaries, one annex and one outlier) were surveyed by means of soil resistivity and magnetometry techniques, of a type normally used for the detection of buried archaeological remains. The results were compared with data concerning the underground sleeping locations adopted by radio-collared badgers in the same setts. Of the eight resistivity surveys, six gave sufficiently detailed results for the overall size of the setts in question to be determined and for some features of the constituent tunnel systems to be discerned. Areas of low resistivity matched locations where radio-collared badgers were found to sleep, so presumably corresponded with underground tunnels and chambers. Magnetometry surveys were not successful, probably because tunnels and chambers were too deep to be detected by this method.

1.1 Trisomy 21 causes phenotypes associated with Down syndrome Down syndrome (DS) occurs in approximately 1 out of 700 live births and most commonly results from three copies of human chromosome 21 (Hsa21) (Christianson, 2006). DS is a multifaceted disorder with over 80 clinically defined phenotypes including those affecting the central nervous system, heart, gastrointestinal tract, skeleton and immune system (Epstein, 2001; Van Cleve and Cohen, 2006). Phenotypes associated with individuals with DS vary in both incidence and severity, leading to a vast array of phenotypic combinations among those with Trisomy 21 (Ts21) (Deutsch et al., 2005; Epstein, 2001; Van Cleve et al., 2006; Van Cleve and Cohen, 2006). For example, cognitive impairment, hypotonia and craniofacial features are universal phenotypes, whereas cardiac abnormalities only affect approximately half of individuals with DS. Individual phenotypes similar to those seen in DS have been documented in individuals without Ts21, but a general higher incidence and severity of these phenotypes in individuals with DS suggests that trisomy plays an important role in initiating or modifying these features (Epstein, 2001; Roper and Reeves, 2006). Although it has been suggested that common mechanisms may be involved in similar phenotypes seen in individuals with and without Ts21, common pathophysiology must be proven for each individual phenotype. A chromosomal basis for DS was postulated as early as 1932 (Patterson and Costa, 2005). After Jerome Lejeune established that DS was caused by an extra Hsa21, he studied the metabolic pathways associated with Ts21 phenotypes in an attempt to cure DS (Neri and Opitz, 2009). Alternate causes of DS, including translocations and mosaicism of extra material from Hsa21, were established shortly after the chromosomal basis of DS had been discovered (Patterson and Costa, 2005). These early genetic findings set the stage for current paradigms and research surrounding the gene-phenotype relationships in DS. Although ~95% of the incidence of DS is due to trisomy of the entire Hsa21, DS also results from extra genetic material from Hsa21 translocated to other chromosomes. Additionally, some individuals with DS phenotypes have Ts21 in only a portion of their cells (mosaicism). Differences in genetic and cellular composition of the trisomy may lead to the observed differences in DS phenotypic incidence and severity. Genotypic and phenotypic variations have also been used to hypothesize about trisomic genes or chromosomal regions that may cause or significantly alter DS phenotypes. Such genotype-phenotype correlations are important in defining the etiology of traits associated with DS as well as suggesting possible therapeutic mechanisms to overcome deficits seen in individuals with DS.

Ts65Dn is a mouse model of Down syndrome; a syndrome that results from Chromosome (Chr) 21 trisomy and is associated with congenital defects, cognitive impairment, and ultimately Alzheimer’s Disease. Ts65Dn mice have segmental trisomy for distal mouse Chr 16, a region sharing conserved synteny with human Chr 21. As a result, this strain harbors three copies of over half of the human Chr 21 orthologs. The trisomic segment of Chr 16 is present as a translocation chromosome (Mmu 1716), with breakpoints that have not been defined previously. To molecularly characterize the Chr 16 and Chr 17 breakpoints on the translocation chromosome in Ts65Dn mice, we used a selective enrichment and high-throughput, paired end sequencing approach. Analysis of paired end reads flanking the Chr 16, Chr 17 junction on Mmu1716 and de-novo assembly of the reads directly spanning the junction provided the precise locations of the Chr 16 and Chr 17 breakpoints at 84,351,351 bp and 9,426,822 bp, respectively. These data provide the basis for low cost, highly efficient genotyping of Ts65Dn mice. More importantly, these data provide, for the first time, complete characterization of gene dosage in Ts65Dn mice.

The mammalian fetus represents a semiallograft within the maternal uterus yet is not rejected. This situation is particularly pronounced in species with a hemochorial type of placentation, such as humans and rodents, where maternal tissues and blood are in direct contact with fetal trophoblast and thus potentially with paternal antigens. The main polymorphic antigens responsible for graft rejection are MHC antigens. In humans the trophoblast cells invading into the decidua have a unique pattern of MHC class I expression characterized by both classical (HLA-C) and nonclassical (HLA-G and HLA-E) molecules. Whether such an unusual MHC repertoire on the surface of trophoblast is a conserved feature between species with hemochorial placentation has not been resolved. Here we demonstrate, using a range of methods, that C57BL/6 mouse trophoblast predominantly expresses only one MHC class I antigen, H2-K, at the cell surface of giant cells but lacks expression of nonclassical MHC molecules. Antigenic disparity between parental MHCs affects trophoblast-induced transformation of the uterine vasculature and, consequently, placental and fetal gowth. Maternal uterine blood vessels were more dilated, allowing for increased blood supply, in certain combinations of maternal and paternal MHC haplotypes, and these allogeneic fetuses and placentas were heavier at term compared with syngeneic controls. Thus, maternal–fetal immune interactions are instrumental to optimize reproductive success. This cross-talk has important implications for human disorders of pregnancy, such as preeclampsia and fetal growth restriction.

Abstract Badgers (Meles meles) defecate, urinate and scent mark at latrines which seem to have a territorial function. The main aim of the present study was to compare defecation patterns at boundary and hinterland latrines, in order to test the hypothesis that these two types of latrine have a similar function. We investigated latrine use by means of a year-round survey of all the latrines in 7 badger territories, by bait-marking of 15 territories, and by monitoring latrine use in 6 radio-collared badgers belonging to three social groups. The spatial distribution of latrines within a territory was bimodal, with the greatest densities oflatrines close to the outside, and close to the centre, of the territory respectively. Boundary latrines were larger and more consistently used than hinterland latrines, but these differences could be accounted for by the fact that boundary latrines are visited by the members of more than one social group. Defecation at latrines was subject to seasonal variation, with a major peak in latrine use in spring and a minor peak in autumn. The spring peak was largely attributable to an increase in the use of hinterland latrines, the autumn peak to an increase in the use of boundary latrines. Males visited boundary latrines considerably more often than did females, but both sexes visited hinterland latrines equally often. Overmarking occurred equally often at both types of latrine and involved animals from the same as well as from different groups, but there was a significant tendency for more between-group than within-group overmarking. Overmarking occurred mainly on fresh, as opposed to old, faeces deposits. The sex and seasonal differences in use of boundary latrines suggest that these function at least partly as a form of mate-guarding, to deter neighbouring males from entering a territory for mating purposes. It is less clear why females mark at hinterland latrines. One possibility, consistent with the observed spatial distribution of hinterland latrines, is that they function to defend the main burrow system, which is used for breeding; another is that they carry information about social status. Overmarking probably serves to obliterate the marks of competitors, which are members of neighbouring social groups in the case of boundary latrines, but may be members of the same social group in the case of hinterland latrines. We conclude that previous ideas about the function of territoriality in badgers, and about the information conveyed by latrines, are oversimplified.

Five experiments were conducted to investigate the effect of novelty on visually-mediated taste-avoidance learning in domestic chicks. In experiments l a and b, chicks were reared with either uncoloured or blue fluid in their home cages, and then required to discriminate between blue and uncoloured fluids that were either palatable or unpalatable (quinine-adulterated). For some chicks the distasteful fluid was novel in appearance, for others it was familiar. In both experiments chicks readily discriminated between a novel unpalatable fluid and a familiar palatable one, but failed to discriminate between a familiar unpalatable fluid and a novel palatable one. This failure to discriminate resulted from avoidance of the palatable fluid. In neither experiment did novelty enhance the rate of avoidance learning. Experiment 2 tested more directly the effect of novelty on speed of avoidance learning. Chicks were reared on either red or blue palatable fluid, then tested with either red or blue distasteful fluid. Avoidance learning was more rapid when the distasteful fluid was novel in colour, in both red-reared and blue-reared chicks. Experiment 3 investigated the inability of chicks to discriminate between a familiar unpalatable fluid and a novel palatable one, demonstrated in experiment 1. Chicks were required to discriminate between different-coloured palatable and unpalatable fluids when both were familiar in appearance (experiment 3a) or when both were novel (experiment 3b). Discrimination occurred in the first case but not in the second. In addition, avoidance learning was slower when both unpalatable fluids were familiar. I conclude that (a) novelty faciliates visually-mediated taste-avoidance learning in chicks and (b) the failure of chicks to discriminate a novel palatable fluid from a familiar unpalatable one depends on the relative novelty of the palatable fluid and not on the relative familiarity of the unpalatable one. The results are discussed in the context of warning coloration and are explained in terms of an interaction between unlearned and learned avoidance tendencies.

In the Australian context, bismuth minerals are important constituents, associated with Mo, W and Sn, of highly acidic and very rich, low tonnage, deposits of the eastern ranges of Australia. Bi has been used extensively as a pathfinder element in geochemical exploration for a variety of such deposits and others and therefore it is important to understand its behaviour[1-3]. The behaviour of Bi has been described in the terms of its solution speciation as a function of pH which is well-established [4-7]and provides a first order, but simplistic, assessment of its mobility in pore solution in the regolith. Recent reviews have highlighted a lack of understanding in regards to bismuth in oxidising environments. Bismuth is estimated to have an average crustal abundance of 48 ppb [8], but is highly fractionated across different rock types and sediments with reported concentrations from 30 ppb to greater than 20 ppm an average Bi concentration in soils of 0.2 ppm but this is difficult to reconcile with the data mentioned above. A review of bismuth values [9] concluded similar concerns and further highlighted that a background ground water concentration could not be honestly concluded especially in uncontaminated environments. This is due to no independent focused study on Bi concentrations in the environment and that other factors such as sampling, testing methods and protocols were not consistent. To draw any conclusion on bismuth values, a proper understanding of the dispersion of Bi in oxidising environments must be made. This requires understanding of its low temperature aqueous chemistry and knowledge of the secondary Bi minerals that serve to buffer the element between the solid and solution states. This study used numerous natural and synthetic bismuth mineral samples and whole rock specimens. Full spectrum quantitative X-ray Mapping (XRM) was performed on a JEOL JXA 840 SEM with a Moran Scientific energy dispersive spectroscopy (EDS) Microanalysis system operated at 20kV. A JEOL 8600 EM Probe with a combined EDS and three JEOL wavelength dispersive spectrometers (WDS’s) were also used to perform trace and quantitative analysis. The analysis was undertaken at 15 kV and 20 nA. Drill core samples from the Mineral Hill mine site in regional NSW have been studied to determine the primary bismuth phases at this site which have been associated with a localised Au resource and then related to the known secondary expressions (Figure 1). Experiments have been carried out on a series of synthetic secondary bismuth minerals (Figure 2) to determine the chemical behaviour of these minerals by High Temperature X-ray Diffraction (HTXRD) and Thermogravimetric Analysis coupled with a Differential Scanning Calorimeter (TGA-DSC). These experiments have provided data which can be combined with low temperature solubility studies to give an in-depth understanding into the geochemical roles of secondary bismuth minerals in the supergene environment. For the first time a series of thermodynamic data sets for major and important bismuth mineral phases have been derived. As such a series of useful geochemical models have been

EMSY interacts directly with BRCA2 and links the BRCA2 pathway to sporadic breast and ovarian cancer. It also interacts with BS69 and HP1b, both of which are involved in chromatin remodelling, and with NIF-1 and DBC-1 in the regulation of nuclear receptor-mediated transcription. Here we investigate the function of EMSY during amphibian development, and in doing so provide the first loss-of-function analysis of this protein. Injection of Xenopus tropicalis embryos with antisense morpholino oligonucleotides targeting XtEMSY impairs gastrulation movements, disrupts dorsal structures, and kills embryos by tailbud stages. Consistent with these observations, regional markers such as Xbra, Chd, Gsc, Shh, Sox3 and Sox17 are downregulated. In contrast to these regional markers, expression of p53 is upregulated in such embryos, and at later stages Bax expression is elevated and apoptotic cells can be detected. Our results demonstrate that EMSY has an essential role in development and they provide an in vivo loss-of-function model that might be used to explore the biochemical functions of this protein in more detail.

Down syndrome (DS) is a genetic disorder resulting from trisomy 21 that causes cognitive impairment, low muscle tone and craniofacial alterations. Morphometric studies of the craniofacial and appendicular skeleton in individuals with DS suggest that bone development and homeostasis are affected by trisomy. The Ts65Dn mouse model has three copies of approximately half the genes found on human chromosome 21 and exhibits craniofacial skeletal and size differences similar to those observed in humans with DS. We hypothesized that Ts65Dn and euploid mice have distinct differences in bone development and homeostasis influencing both the craniofacial and appendicular skeletal phenotypes. Quantitative assessment of structural and mechanical properties of the femur in Ts65Dn and control mice at 6 and 16 weeks of age revealed significant deficiencies in trabecular and cortical bone architecture, bone mineral density, bone formation, and bone strength in trisomic bone. Furthermore, bone mineral density and dynamic dentin formation rate of the skull and incisor, respectively, were also reduced in Ts65Dn mice, demonstrating that trisomy significantly affects both the craniofacial and appendicular skeleton.

Trisomy 21 results in Down syndrome (DS) and causes phenotypes that may result from alterations of developmental processes. The Ts65Dn mouse is the most widely used genetic and phenotypic model for DS. We used over 1,500 offspring from Ts65Dn and two nontrisomic genetically similar control strains to investigate the influence of trisomy on developmental alterations and number of offspring. For the first time, we demonstrate gross developmental attenuation of Ts65Dn trisomic offspring at embryonic day (E) 9.5 and E13.5 and show that the major determinant of the developmental changes is segmental trisomy of the embryo and not the trisomic maternal uterine environment. Maternal alleles of nontrisomic genes linked to Pde6b may also influence the development of Ts65Dn offspring. Both developmental attenuation and the contribution of trisomic and nontrisomic genes are important components in the genesis of DS phenotypes.

Substances that bind calcium are given to determine where and how fast bone is forming. Several vital dyes are used (tetracycline, calcein, alizarin, xylenol), but it is not known whether the histomorphometric results they provide are equivalent. This work tests whether different fluorochrome labels give the same results when they are quantitatively measured. Twelve-week-old rats (n = 58) were divided into six groups and given double labels IP of calcein, tetracycline HCl, alizarin complexone, or xylenol using a 1-7-1 scheme. Two other groups received either calcein followed by tetracycline, or tetracycline followed by calcein. Our results show that (a) tetracycline hydrochloride leads to a significant underestimation of mineralizing surface when given as the second label, probably because of its weak fluorescence; (b) there were no differences among any of the non-tetracycline labels for any of the histomorphometric measurements; (c) there was no evidence of osteoblast suppression with any label; and (d) there was no evidence that tetracycline caused increased osteoblast resting periods.

Badger setts vary considerably in size, ranging from simple single-entrance burrows to complex tunnel systems hundreds of metres long with multiple entrances and underground chambers. Data from 19 excavated setts show that main setts are larger than other setts in terms of area and volume, and contain more chambers, nests and latrines; but setts of different sizes and types are built according to the same basic architectural principles. Little is known about the environmental conditions within setts, other than that temperature and humidity are constant in parts of a sett that are at least 7 m from the nearest entrance. Setts are used for breeding and as sleeping places and refuges, but a question remains as to the functional value of large setts. It is suggested that large main setts allow members of a social group to avoid one another underground, especially when breeding. Little is known about the use of other types of sett.

Summary: Many online sources of gene interaction networks supply rich visual data regarding gene pathways that can aid in the study of biological processes, disease research and drug discovery. PathGen incorporates data from several sources to create transitive connections that span multiple gene interaction databases. Results are displayed in a comprehensible graphical format, showing gene interaction type and strength, database source and microarray expression data. These features make PathGen a valuable tool for in silico discovery of novel gene interaction pathways, which can be experimentally tested and verified. The usefulness of PathGen interaction analyses was validated using genes connected to the altered facial development related to Down syndrome. Este trabalho teve por objetivo acompanhar e analisar as interações inter e intra-específicas de beija-flores e como estas interações mudaram ao longo do ano. O estudo foi realizado em um remanescente de floresta no município de Piraquara, Paraná. Beija-flores foram filmados mensalmente entre março de 2008 e abril de 2009. Foi possível identificar indivíduos focais, medir intervalos de tempo durante o qual o indivíduo focal ficou no bebedouro e anotar o momento e a espécie envolvida em uma interação. A espécie mais comum foi Leucochloris albicollis que foi observado em 2.300 visitas, seguido por Colibri serrirostris, Amazilia versicolor, Florisuga fusca e Thalurania glaucopis. Há variações entre as espécies no tempo de exploração do recurso: L. albicollis tende a ficar 21s em cada visita, em contraste com F. fusca que tende a ficar somente 7s. As conseqüências destas interações complexas podem estruturar a comunidade com uma hierarquia flutuante, que varia ao longo do ano.

The molecular processes that govern the first cell lineage decisions after fertilization also dictate the developmental potency of stem cells derived from the early mouse embryo. Our understanding of these mechanisms is therefore instrumental for stem cell biology and regenerative medicine. A number of transcription factors are known that determine a cell's fate towards either the embryonic or extraembryonic trophoblast lineages. Recent insights have shown that the definitive fixation of cell lineage fate is achieved by an epigenetic restriction through DNA methylation of the transcription factor Elf5. Lineage crossover can be induced, however, by manipulation of lineage determinants and gatekeepers, or their epigenetic regulation. Here we summarize the accumulating number of experimental conditions where such 'transdifferentiation' is observed that shed light onto the genetic and epigenetic pathways involved in lineage separation and the developmental potential of stem cells.

Background Pseudomonas aeruginosa encodes a putative topoisomerase with sequence similarity to the eukaryotic type IB topoisomerase from Vaccinia virus. Residues in the active site are conserved, notably Tyr292 which would be predicted to form the transient covalent bond to DNA. Results The gene encoding the P. aeruginosa topoisomerase I was cloned and expressed in E. coli. The enzyme relaxes supercoiled DNA, while a mutant containing a Tyr292 to Phe substitution at the active site was found to be catalytically inert. This is consistent with the role of Tyr in forming the covalent intermediate. Like Vaccinia topoisomerase, the P. aeruginosa topoisomerase relaxes DNA in the absence of ATP, but unlike Vaccinia topoisomerase, P. aeruginosa topoisomerase does not relax supercoiled DNA without MgCl2 present. In addition, high concentration of NaCl is not able to substitute for MgCl2 as seen for Vaccinia topoisomerase. A truncated derivative of the topoisomerase lacking residues 1–98 relaxes DNA, with both full length and truncated enzyme exhibiting equivalent requirements for divalent cations and the ability to relax DNA to completion, suggesting a shared domain organization. DNA-binding assays suggest an only modest preference for the CCCTT pentameric sequence required for transesterification by Vaccinia topoisomerase IB. Conclusion P. aeruginosa encodes a functional topoisomerase with significant similarity to the type IB enzyme encoded by poxviruses. In contrast to the Vaccinia-encoded homolog, the P. aeruginosa-encoded enzyme requires divalent cations for catalytic activity, relaxes DNA to completion, and does not exhibit a strong preference for the pentameric sequence stringently required by the Vaccinia-encoded homolog. A comparison with the structure of poxviral topoisomerase in complex with DNA suggests that bacterial homologs of the eukaryotic type IB topoisomerase may exhibit a relaxed sequence preference due to the lack of conservation of certain residues involved in sequence-specific DNA contacts, and that interaction with an only modestly preferred sequence may result in suboptimal positioning of catalytic residues.

Three badger (Meles meles L.) setts in the south of England, which formed a single sett complex belonging to one social group of badgers, were excavated prior to being destroyed by construction of a new road. Setts 1 and 2, classified as annexes, were excavated completely; sett 3, classified as a main sett, was only excavated partially, but its total size was estimated from the excavated portion. The setts consisted of tunnels totalling 16 m, 140 m and 879 m, respectively; contained one, nine and 50 chambers; and had five, 42 and 178 entrances. The total volume of the three setts was about 45 m3, and their construction was estimated to have required the removal of about 70 tonnes of soil. In the two smaller setts tunnels ran on a single level with an average depth of 99 cm; in the larger sett they ran on two levels with modal depths of 50 cm and 110–120 cm, respectively. All three setts contained bedding material (dry grass and plastic bags) but only the main sett contained latrines. None of the setts contained badger bones and the interiors of all three setts were remarkably clean and orderly. We discuss hypotheses as to why badgers sometimes continue to extend even large well-established setts but conclude that the survival value of very large setts remains problematical.

Computer generated discharge summaries are gaining in popularity. A simple and flexible method for generating neonatal summaries using commercially available software is described.

Social animals regularly face consensus decisions whereby they choose, collectively, between mutually exclusive actions. Such decisions often involve conflicts of interest between group members with respect to preferred action. Conflicts could, in principle, be resolved, either by sharing decisions between members (‘shared decisions’) or by one ‘dominant’ member making decisions on behalf of the whole group (‘unshared decisions’). Both, shared and unshared decisions, have been observed. However, it is unclear as to what favours the evolution of either decision type. Here, after a brief literature review, we present a novel method, involving a combination of self-organizing system and game theory modelling, of investigating the evolution of shared and unshared decisions. We apply the method to decisions on movement direction. We find that both, shared and unshared, decisions can evolve without individuals having a global overview of the group's behaviour or any knowledge about other members' preferences or intentions. Selection favours unshared over shared decisions when conflicts are high relative to grouping benefits, and vice versa. These results differ from those of group decision models relating to activity timings. We attribute this to fundamental differences between collective decisions about modalities that are disjunct (here, space) or continuous (here, time) with respect to costs/benefits.

1. Urban and rural populations of animals can differ in their behaviour, both in order to meet their ecological requirements and due to the constraints imposed by different environments. The study of urban populations can therefore offer useful insights into the behavioural flexibility of a species as a whole, as well as indicating how the species in question adapts to a specifically urban environment. 2. The genetic structure of a population can provide information about social structure and movement patterns that is difficult to obtain by other means. Using non-invasively collected hair samples, we estimated the population size of Eurasian badgers Meles meles in the city of Brighton, England, and calculated population-specific parameters of genetic variability and sex-specific rates of outbreeding and dispersal. 3. Population density was high in the context of badger densities reported throughout their range. This was due to a high density of social groups rather than large numbers of individuals per group. 4. The allelic richness of the population was low compared with other British populations. However, the rate of extra-group paternity and the relatively frequent (mainly temporary) intergroup movements suggest that, on a local scale, the population was outbred. Although members of both sexes visited other groups, there was a trend for more females to make intergroup movements. 5. The results reveal that urban badgers can achieve high densities and suggest that while some population parameters are similar between urban and rural populations, the frequency of intergroup movements is higher among urban badgers. In a wider context, these results demonstrate the ability of non-invasive genetic sampling to provide information about the population density, social structure and behaviour of urban wildlife.

Ports are important locations for the introduction of marine species, while marinas and pontoons often serve as secondary habitats for these species. In a marina near Paranagua Port, a major international port in southern Brazil, the encrusting community was studied to (i) identify possibly introduced species, and (ii) examine the use of artificial substrata by these species. Samples (20 x 20 cm) were taken from fibreglass floats (boardwalks and boat hulls) and concrete columns. A total of 85 species were found of which 50 were classified into three categories: four introduced, 33 cryptogenic and 13 native. The introduced species were the hydrozoan Garveia franciscana (on floats, boats and submerged concrete), the polychaete Polydora cornuta (more abundant on floats and submerged concrete) and the barnacles Amphibalanus reticulatus (equally abundant on the three substrata) and Striatobalanus amaryllis (only on floats and submerged concrete). Organisms were most abundant on floating boardwalks, while species richness and composition were similar to that on boat hulls (32 and 37, respectively), which are an important vector for intraregional transport. All substrata supported at least three of the four introduced, and many of the cryptogenic species. The proportion of introduced to the total number of species was greater than observed in other ports. This demonstrates that the introduction potential is great in Paranagua Bay, especially considering that this study was restricted to one site and sampled only hard substrata. None of the introduced species has yet been identified as invasive, but all are generalists with respect to substratum, indicating their invasive potential. The ability to colonise stable concrete walls shows that they could also colonise the natural granite rocky substrata in the bay, and the ability to colonise floating surfaces indicate their capability of spreading in the region on the hulls of recreational boats.

We report a case of angioedema caused by angiotensin-converting enzyme inhibitor and topical lignocaine spray, administered during nasendoscopy.Angioedema induced by angiotensin-converting enzyme inhibitors is a rare but well known entity. Allergy to topical lignocaine has been acknowledged as a rare phenomenon when used for dental surgery and for skin anaesthesia, but it has not previously been reported after topical administration prior to nasendoscopy. In the reported case, our patient was unfortunate enough to be allergic to both lisinopril and lignocaine. The result was life-threatening airway obstruction, and the continued use of lignocaine spray sustained the laryngeal oedema. We advise that patients are asked about any and every allergy – specifically, any previous problems with dental procedures – before administration of local anaesthetic spray to the upper aerodigestive tract.

We observed meadow brown (Maniola jurtina) and gatekeeper (Pyronia tithonus) butterflies at habitat boundaries and observed spontaneous movements out of suitable habitat in order to investigate such movements in relation to dispersal. We found that butterflies of both species were aware of the position of a highly permeable habitat boundary without needing to cross it. Nevertheless, a considerable proportion of butterflies close to the boundary left their habitat (25-43%). Butterflies that crossed the boundary, and moved substantial distances into unsuitable habitat (up to 350 m in M. jurtina and 70 m in P. tithonus), usually returned to their original habitat patch (98-100%). Movement trajectories, at least in M. jurtina, were significantly different from, and more directed and systematic than, a correlated random walk. Approximately 70-80% of spontaneous movements into unsuitable habitat in both species were "foray" loops comparable to those described in mammals and birds. We conclude that, since migrants seemed to have considerable control over leaving their patch and over their subsequent movement trajectories, chance encounter rates with habitat boundaries, and indeed habitat leaving rates, might be less crucial in determining dispersal rates than is usually assumed. In addition, random dispersal trajectories should not be taken for granted in population or evolution models.

Ts65Dn mice inherit a marker chromosome, T(1716)65Dn, producing segmental trisomy for orthologs of about half of the genes on human chromosome 21. These mice display a number of phenotypes that are directly comparable to those in humans with trisomy 21 and are the most widely used animal model of Down syndrome (DS). However, the husbandry of Ts65Dn mice is complicated. Males are sterile, and only 20–40% of the offspring of Ts65Dn mothers are trisomic at weaning. The lower-than-expected frequency of trisomic offspring has been attributed to losses at meiosis, during gestation and at postnatal stages, but no systematic studies support any of these suppositions. We show that the T(1716)65Dn marker chromosome is inherited at expected frequency and is fully compatible with development to midgestation. Disproportional loss of trisomic offspring occurs in late gestation and continues through birth to weaning. Different maternal H2 haplotypes are significantly associated with the frequency of trisomy at weaning in patterns different from those reported previously. The proportion of trisomic mice per litter decreases with age of the Ts65Dn mother. These results provide the first statistical and numerical evidence supporting the prenatal and perinatal pattern of loss in the Ts65Dn mouse model of DS.

Bacterial cultures produce subpopulations of cells termed ‘persisters’, reputedly known for high tolerance to killing by antibiotics. Ecologically, antibiotics produced by competing microflora are only one potential stress encountered by bacteria. Another pressure in the environment is toxic metals that are distributed ubiquitously by human pollution, volcanic activity and the weathering of minerals. This study evaluated the time- and concentration-dependent killing of Escherichia coli planktonic and biofilm cultures by the water-soluble metal(loid) oxyanions chromate (), arsenate (), arsenite (), selenite (), tellurate () and tellurite (). Correlative to previous reports in the literature, control antibiotic assays indicated that a small proportion of E. coli biofilm populations remained recalcitrant to killing by antibiotics (even with 24 h exposure). In contrast, metal oxyanions presented a slow, bactericidal action that eradicated biofilms. When exposed for 2 h, biofilms were up to 310 times more tolerant to killing by metal oxyanions than corresponding planktonic cultures. However, by 24 h, planktonic cells and biofilms were eradicated at approximately the same concentration in all instances. Coloured complexes of metals and chelators could not be generated in biofilms exposed to or , suggesting that the extracellular polymeric matrix of E. coli may have a low binding affinity for metal oxyanions. Viable cell counts at 2 and 24 h exposure revealed that, at high concentrations, all of the metal oxyanions had killed 99 % (or a greater proportion) of the bacterial cells in biofilm populations. It is suggested here that the short-term survival of E. coli biofilms to high concentrations of metal oxyanions.
