Understanding past and present hydrosystem feedbacks to global ocean–atmospheric interactions represents one of the main challenges to preventing droughts, extreme events, and related human catastrophes in the face of global warming, especially in arid and semiarid environments. In eastern Africa, the El Niño–Southern Oscillation (ENSO) was identified as one of the primary drivers of precipitation variability affecting water availability. However, the northern East African Rift System (EARS) still suffers from the underrepresentation of predictive and ENSO teleconnection models because of the scarcity of local to regional historical or palaeo-data. In this paper, we provide a 50-year seasonal flood and drought chronicle of the Awash River catchment from the study of laminated sediment from Gemeri and Afambo lakes (central Afar region, Ethiopia) with the aim of reconstructing the magnitude of regional hydroclimatic events. Pluricentimetric micro-laminated lithogenic facies alternating with plurimillimetric carbonate-enriched facies are investigated in both lakes. We couple dating methods including radiocarbon, short-lived radionuclides, palaeomagnetic field variations, and varve counting on both lake deposits to build a high-resolution age model and to discuss the regional hydrosedimentary dynamics of the Awash River over the last ∼ 700 years with a focus on the last 50 years. Using a multiproxy approach, we observe that following a multicentennial enhanced hydrological period, the two lakes have experienced a gradual decrease in river load inflow since 1979 CE, attaining extreme drought and high evaporative conditions between 1991 and 1997 CE. In 2014, the construction of a dam and increased agricultural water management in the lower Awash River plain impacted the erodibility of local soils and the hydrosedimentary balance of the lake basins, as evidenced by a disproportionate sediment accumulation rate. Comparison of our quantitative reconstruction with (i) lake water surface evolution, (ii) the interannual Awash River flow rates, and (iii) the El Niño 3.4 model highlights the intermittent connections between ENSO sea surface temperature anomalies, regional droughts, and hydrological conditions in the northern EARS.

Abstract Sperm whales (Physeter macrocephalus) have been studied for decades, but the development of their clicks during the animal growth is not yet well known. The click they emit during socialization and echolocation contains information about the length of their acoustic organs and, therefore the length of the body through the interpulse interval (IPI). This paper provides the first IPI/age relationship for juvenile male and female sperm whales (Physeter macrocephalus) based on field recordings of individuals whose age is largely known. Across 9 years, audiovisual recordings of a Mauritian sperm whale social unit were carried out. Adult female and juvenile sperm whales were identified and aged. The dataset made from those recordings is publicly available. The interpulse interval was measured for individuals whose ages ranged from 7 days to around 38 years. The growth of the acoustic organ of juveniles showed an early inter-individual variability as well as sexual dimorphism. Usual growth models were also fitted, predicting a mean $$IPI_\infty$$ I P I ∞ of 3.5 ms for adults and a physical maturity reached at around 30 years old. The use of passive acoustic monitoring (PAM) is one of the main tools used to study sperm whales. This IPI-age relationship may aid demographic studies on sperm whales by enabling PAM to assess the ages of recorded sperm whales.

Abstract As major terrestrial carbon sinks, forests play an important role in mitigating climate change. The relationship between the seasonal uptake of carbon and its allocation to woody biomass remains poorly understood, leaving a significant gap in our capacity to predict carbon sequestration by forests. Here, we compare the intra-annual dynamics of carbon fluxes and wood formation across the Northern hemisphere, from carbon assimilation and the formation of non-structural carbon compounds to their incorporation in woody tissues. We show temporally coupled seasonal peaks of carbon assimilation (GPP) and wood cell differentiation, while the two processes are substantially decoupled during off-peak periods. Peaks of cambial activity occur substantially earlier compared to GPP, suggesting the buffer role of non-structural carbohydrates between the processes of carbon assimilation and allocation to wood. Our findings suggest that high-resolution seasonal data of ecosystem carbon fluxes, wood formation and the associated physiological processes may reduce uncertainties in carbon source-sink relationships at different spatial scales, from stand to ecosystem levels.

Abstract Psoriasis is a multifactorial, chronic inflammatory skin disease with unresolved questions on its primary events. Iron overload has been described in the epidermis of psoriasis patients, but its relevance remains unknown. We found that the key iron regulatory hormone hepcidin was highly expressed in the epidermis of psoriasis patients, especially the pustular variants resistant to treatments. In a murine model of acute skin inflammation, keratinocyte-derived hepcidin was required for iron retention in keratinocytes, leading to hyperproliferation of the epidermal layer and neutrophil recruitment, two main features of psoriatic skin lesions. Keratinocytes overexpressing hepcidin were sufficient to elicit these psoriasiform features in a transgenic mouse model. Furthermore, transcriptome analysis of these keratinocytes revealed canonical pathways found in human psoriasis, pointing to a causal role for hepcidin in the pathogenesis of the disease. Altogether, our data suggest that hepcidin could be an actionable target for skin psoriasis treatment, in addition to current therapeutics, or targeted as maintenance therapy during remission to prevent recurrence.

Abstract The demographical history of France remains largely understudied despite its central role toward understanding modern population structure across Western Europe. Here, by exploring publicly available Europe-wide genotype datasets together with the genomes of 3234 present-day and six newly sequenced medieval individuals from Northern France, we found extensive fine-scale population structure across Brittany and the downstream Loire basin and increased population differentiation between the northern and southern sides of the river Loire, associated with higher proportions of steppe vs. Neolithic-related ancestry. We also found increased allele sharing between individuals from Western Brittany and those associated with the Bell Beaker complex. Our results emphasise the need for investigating local populations to better understand the distribution of rare (putatively deleterious) variants across space and the importance of common genetic legacy in understanding the sharing of disease-related alleles between Brittany and people from western Britain and Ireland.

Abstract While significant advances have been made in understanding renal pathophysiology, less is known about the role of glycosphingolipid (GSL) metabolism in driving organ dysfunction. Here, we used a small molecule inhibitor of glucosylceramide synthase to modulate GSL levels in three mouse models of distinct renal pathologies: Alport syndrome (Col4a3 KO), polycystic kidney disease (Nek8 jck ), and steroid-resistant nephrotic syndrome (Nphs2 cKO). At the tissue level, we identified a core immune-enriched transcriptional signature that was shared across models and enriched in human polycystic kidney disease. Single nuclei analysis identified robust transcriptional changes across multiple kidney cell types, including epithelial and immune lineages. To further explore the role of GSL modulation in macrophage biology, we performed in vitro studies with homeostatic and inflammatory bone marrow-derived macrophages. Cumulatively, this study provides a comprehensive overview of renal dysfunction and the effect of GSL modulation on kidney-derived cells in the setting of renal dysfunction.

Influenza D virus was isolated from pigs on a mixed pig and beef farm in France. Investigation suggested bull-to-pig transmission and spread among pigs. The swine influenza D virus recovered was a reassortant of D/660 and D/OK lineages. Reported mutations in the receptor binding site might be related to swine host adaptation.

In December 2023, we observed through hospital-based surveillance a severe outbreak of enterovirus D68 infection in pediatric inpatients in Dakar, Senegal. Molecular characterization revealed that subclade B3, the dominant lineage in outbreaks worldwide, was responsible for the outbreak. Enhanced surveillance in inpatient settings, including among patients with neurologic illnesses, is needed.

Abstract Background Families and significant others of people with borderline personality disorder (BPD) show increased levels of psychological distress. Family Connections®, a 12-week group intervention based on the principles of Dialectical Behavior Therapy, was designed to provide families with both information about the disorder and emotion regulation skills. It has been progressively implemented in French-speaking European countries. Methods We conducted an observational, multicenter study in France and Switzerland. In total, 149 participants of the Family Connections program were included among five centers. Burden, depression, coping, and emotion regulation were assessed before and after the intervention. Abstract Bed bugs are pervasive global pests that have reemerged in the last 20 years as a significant public health concern, especially in densely populated urban areas. Beyond financial losses, expenses, inconvenience, and psychological distress, bed bug infestations often necessitate chemical management, posing poisoning risks to those with an infestation. The French Poison Control Centers recorded 1056 cases of exposure to bed bug insecticide products between 1999 and 2021. This study followed cases over 2007–2021, with a notable surge in reports of adverse reactions from 2016 onwards. Data revealed an increased recurrent misuse of insecticides, including substances banned or not approved for this use. Our findings underscore the growing public reliance on chemical insecticides for home bed bug management. With this is the concern of increased poisoning risks, and potential long-term health consequences from non-professional efforts by the public to manage bed bugs in their homes. This escalating trend emphasizes the need for safer and more sustainable pest management strategies in urban environments.

Abstract Background POINCARE-2 trial aimed to assess the effectiveness of a strategy designed to tackle fluid overload through daily weighing and subsequent administration of treatments in critically ill patients. Even in highly standardized care settings, such as intensive care units, effectiveness of such a complex intervention depends on its actual efficacy but also on the extent of its implementation. Using a process evaluation, we aimed to provide understanding of the implementation, context, and mechanisms of change of POINCARE-2 strategy during the trial, to gain insight on its effectiveness and inform the decision regarding the dissemination of the intervention. Methods We conducted a mixed-method process evaluation following the Medical Research Council guideline. Both quantitative data derived from the trial, and qualitative data from semi-structured interviews with professionals were used to explain implementation, mechanisms of change of the POINCARE-2 strategy, as well as contextual factors potentially influencing implementation of the strategy. Results Score of actual exposure to the strategy ranged from 29.1 to 68.2% during the control period, and from 61.9 to 92.3% during the intervention period, suggesting both potential contamination and suboptimal fidelity to the strategy. Lack of appropriate weighing devices, lack of human resources dedicated to research, pre-trial rooted prescription habits, and anticipated knowledge of the strategy have been identified as the main barriers to optimal implementation of the strategy in the trial context. Conclusions Both contamination and suboptimal fidelity to POINCARE-2 strategy raised concerns about a potential bias towards the null of intention-to-treat (ITT) analyses. However, optimal fidelity seemed reachable. Consequently, a clinical strategy should not be rejected solely on the basis of the negativity of ITT analyses’ results. Our findings showed that, even in highly standardized care conditions, the implementation of clinical strategies may be hindered by numerous contextual factors, which demonstrates the critical importance of assessing the viability of an intervention, prior to any evaluation of its effectiveness. Abstract Bacteria often thrive in surface-attached communities, where they can form biofilms affording them multiple advantages. In this sessile form, fluid flow is a key component of their environments, renewing nutrients and transporting metabolic products and signaling molecules. It also controls colonization patterns and growth rates on surfaces, through bacteria transport, attachment and detachment. However, the current understanding of bacterial growth on surfaces neglects the possibility that bacteria may modulate their division behavior as a response to flow. Here, we employed single-cell imaging in microfluidic experiments to demonstrate that attached Escherichia coli cells can enter a growth arrest state while simultaneously enhancing their adhesion underflow. Despite utilizing clonal populations, we observed a non-uniform response characterized by bistable dynamics, with co-existing subpopulations of non-dividing and actively dividing bacteria. As the proportion of non-dividing bacteria increased with the applied flow rate, it resulted in a reduction in the average growth rate of bacterial populations on flow-exposed surfaces. Dividing bacteria exhibited asymmetric attachment, whereas non-dividing counterparts adhered to the surface via both cell poles. Hence, this phenotypic diversity allows bacterial colonies to combine enhanced attachment with sustained growth, although at a reduced rate, which may be a significant advantage in fluctuating flow conditions.

Abstract Key Message Understanding the impact of extreme drought on the canopy fuel moisture content (CFMC) is crucial to anticipate the effects of climate change on wildfires. Our study demonstrates that foliage mortality, caused by leaf embolism, can substantially diminish CFMC during drought on Pinus halepensis Mill. and Quercus ilex L. It emphasizes the importance of considering plant hydraulics to improve wildfire predictions. Context Canopy fuel moisture content (CFMC), which represents the water-to-dry mass ratio in leaves and fine twigs within the canopy, is a major factor of fire danger across ecosystems worldwide. CFMC results from the fuel moisture content of living foliage (live fuel moisture content, LFMC) and dead foliage (dead fuel moisture content, DFMC) weighted by the proportion of foliage mortality in the canopy (α Dead ). Understanding how LFMC, α Dead , and ultimately CFMC are affected during extreme drought is essential for effective wildfire planning. Aims We aimed to understand how plant hydraulics affect CFMC for different levels of soil water deficit, examining its influence on both LFMC and α Dead . Methods We conducted a drought experiment on seedlings of two Mediterranean species: Aleppo pine (Pinus halepensis Mill.) and Holm oak (Quercus ilex L.). Throughout the drought experiment and after rewatering, we monitored CFMC, LFMC, and α Dead along with other ecophysiological variables. Results LFMC exhibited a significant decrease during drought, and as leaf water potentials reached low levels, α Dead increased in both species, thereby reducing CFMC. Distinct water use strategies resulted in species-specific variations in dehydration dynamics. Conclusion Our findings demonstrate that as drought conditions intensify, foliage mortality might become a critical physiological factor driving the decline in CFMC.

Abstract The nanoscopic magnetic texture forming in a monolayer of iron on the (111) surface of iridium, Fe/Ir(111), is spatially modulated and uniaxially incommensurate with respect to the crystallographic periodicities. As a consequence, a low-energy magnetic excitation is expected that corresponds to the sliding of the texture along the incommensurate direction, i.e., a phason mode, which we explicitly confirm with atomistic spin simulations. Using scanning tunneling microscopy (STM), we succeed to observe this phason mode experimentally. It can be excited by the STM tip, which leads to a random telegraph noise in the tunneling current that we attribute to the presence of two minima in the phason potential due to the presence of disorder in our sample. This provides the prospect of a floating phase in cleaner samples and, potentially, a commensurate-incommensurate transition as a function of external control parameters.

Abstract Veterinary school is intense and represents a challenging time for students, and increasingly, veterinary students are being diagnosed with a variety of concerning mental health issues including stress, anxiety, and depression, often resulting in poor academic performance. Because of the disproportionately high risk of mental health problems in the veterinary field, it is important that students learn, understand, and utilize simple and effective techniques to cope with stress, and demands of the profession. Spending time in natural environments such as outdoor parks, green spaces, and urban forests is an effective strategy for improving mental health. In this study, we examined the self-reported effects of repeated, structured time in a nature-based intervention, vs a control group of veterinary students. Participants reported significantly lower perceived stress levels compared to a control group. Students overwhelmingly reported that they would recommend this stress reduction technique to their classmates, and they stated that they planned to continue spending time in nature to improve their mindset and reduce stress in the future.

En arrivant dans un pays dont on ne connaît ni la culture, ni la langue, les mots manquent pour communiquer. Comment alors exprimer ses besoins et ses ressentis, dans un contexte qui renforce précisément les opportunités d’expression de besoins et de ressentis ? En français, les mots savoir et saveur ont la même étymologie. Or, pour savoir une langue, le meilleur moyen n’est-il pas d’avoir justement accès à sa saveur ? Ce texte, à travers le compte rendu de diverses expériences artistiques avec des publics allophones, propose une réflexion sur l’importance de la sensibilité, du plaisir et du ludique pour aider à un apprentissage de la langue et des codes culturels du pays d’accueil, dont la nécessité exige a priori tout le contraire : rationalité, labeur et sérieux. Il développe enfin les nombreux et étonnants points de liaison entre la pratique pragmatique des artistes et intervenants avec leurs publics et la didactique structurée de l’enseignement des arts à l’école (notamment primaire) française.

Abstract Accurate mapping and monitoring of tropical forests aboveground biomass (AGB) is crucial to design effective carbon emission reduction strategies and improving our understanding of Earth’s carbon cycle. However, existing large-scale maps of tropical forest AGB generated through combinations of Earth Observation (EO) and forest inventory data show markedly divergent estimates, even after accounting for reported uncertainties. To address this, a network of high-quality reference data is needed to calibrate and validate mapping algorithms. This study aims to generate reference AGB datasets using field inventory plots and airborne LiDAR data for eight sites in Central Africa and five sites in South Asia, two regions largely underrepresented in global reference AGB datasets. The study provides access to these reference AGB maps, including uncertainty maps, at 100 m and 40 m spatial resolutions covering a total LiDAR footprint of 1,11,650 ha [ranging from 150 to 40,000 ha at site level]. These maps serve as calibration/validation datasets to improve the accuracy and reliability of AGB mapping for current and upcoming EO missions (viz., GEDI, BIOMASS, and NISAR).

There is a prevalent consensus among science education scholars that teaching socioscientific issues (SSI) has notable importance for promoting functional scientific literacy and raising responsible citizens. However, teachers encounter numerous problems when teaching SSI, so they may not prefer to teach them. Today, we still do not know many specific particulars about how teachers' own opinions, beliefs, or experiences influence the way in which they present controversies and which teacher's role is best suited for teaching SSI. This paper aims to review the empirical studies that include teachers' stances during teaching SSI and, if stated, their rationales about preferred stances. We employed the Web of Science and Education Resources Information Center (ERIC) databases and Google Scholar to find the relevant literature. Then, we independently screened the titles, abstracts, and full texts of the selected studies for eligibility. Based on the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement checklist, eighteen empirical studies were included in this review. Three main profiles of teachers' positions while addressing SSI were identified: excluders, neutrals, and disclosers. Two subprofiles were identified for both neutrals and disclosers. Neutral teaching was found to be the most adopted and preferred teacher position for introducing SSI. It was followed by the avoidance of SSI teaching because the teachers viewed value-free education as important. Teachers' own opinions, beliefs, epistemological orientations, concerns of indoctrination, and external factors were reported as the main underlying reasons for their preferred positions. Obviously, the studies conducted with in-service teachers yielded different reasoning patterns for their neutrality. We concluded that, given the pedagogical and epistemological challenges of teaching SSI, more research on teachers' positions in addressing SSI in science classrooms is needed.

Pancreatic ductal adenocarcinoma (PDAC) is a lethal cancer with a low survival rate. Recently, new drugs that target KRASG12D, a common mutation in PDAC, have been developed. We studied one of these compounds, MRTX1133, and found it was specific and effective at low nanomolar concentrations in patient-derived organoid models and cell lines harboring KRASG12D mutations. Treatment with MRTX1133 upregulated the expression and phosphorylation of EGFR and HER2, indicating that inhibition of ERBB signaling may potentiate MRTX1133 antitumor activity. Indeed, the irreversible pan-ERBB inhibitor, afatinib, potently synergized with MRTX1133 in vitro, and cancer cells with acquired resistance to MRTX1133 in vitro remained sensitive to this combination therapy. Finally, the combination of MRTX1133 and afatinib led to tumor regression and longer survival in orthotopic PDAC mouse models. These results suggest that dual inhibition of ERBB and KRAS signaling may be synergistic and circumvent the rapid development of acquired resistance in patients with KRAS mutant pancreatic cancer.SignificanceKRAS-mutant pancreatic cancer models, including KRAS inhibitor-resistant models, show exquisite sensitivity to combined pan-ERBB and KRAS targeting, which provides the rationale for testing this drug combination in clinical trials.

Silicon quantum dot qubits show great promise but suffer from charge noise with a 1/fα spectrum, where f is frequency and α≲1. It has recently been proposed that 1/fα noise spectra can emerge from a few thermally activated two-level fluctuators in the presence of sub-bath temperature fluctuations associated with a two-dimensional electron gas (2DEG). We investigate this proposal by performing Monte Carlo simulations of a single Ising spin in a bath with a fluctuating temperature. We find that to obtain noise with a 1/fα spectrum with α≲1 down to low frequencies, the duration of temperature fluctuations must be comparable to the inverse of the lowest frequency at which the noise is measured. This result is consistent with an analytic calculation in which the fluctuator is a two-state system with dynamics governed by time-dependent switching rates. In this case we find that the noise spectrum follows a Lorentzian at frequencies lower than the inverse of the average duration of the lowest switching rate. We then estimate relaxation times of thermal fluctuations by considering thermal diffusion in an electron gas in a confined geometry. We conclude that temperature fluctuations in a 2DEG sub-bath would require unphysically long durations to be consistent with experimental measurements of 1/f-like charge noise in quantum dots at frequencies extending well below 1 Hz.

Membrane tension is thought to be a long-range integrator of cell physiology. Membrane tension has been proposed to enable cell polarity during migration through front-back coordination and long-range protrusion competition. These roles necessitate effective tension transmission across the cell. However, conflicting observations have left the field divided as to whether cell membranes support or resist tension propagation. This discrepancy likely originates from the use of exogenous forces that may not accurately mimic endogenous forces. We overcome this complication by leveraging optogenetics to directly control localized actin-based protrusions or actomyosin contractions while simultaneously monitoring the propagation of membrane tension using dual-trap optical tweezers. Surprisingly, actin-driven protrusions and actomyosin contractions both elicit rapid global membrane tension propagation, whereas forces applied to cell membranes alone do not. We present a simple unifying mechanical model in which mechanical forces that engage the actin cortex drive rapid, robust membrane tension propagation through long-range membrane flows.

The massive galaxy cluster El Gordo (z = 0.87) imprints multitudes of gravitationally lensed arcs onto James Webb Space Telescope Near-Infrared Camera (NIRCam) images. Eight bands of NIRCam imaging were obtained in the “Prime Extragalactic Areas for Reionization and Lensing Science” (“PEARLS”) program. Point-spread function-matched photometry across Hubble Space Telescope and NIRCam filters supplies new photometric redshifts. A new light-traces-mass lens model based on 56 image multiplicities identifies the two mass peaks and yields a mass estimate within 500 kpc of (7.0 ± 0.30) × 1014 M ⊙. A search for substructure in the 140 cluster members with spectroscopic redshifts confirms the two main mass components. The southeastern mass peak that contains the brightest cluster galaxy is more tightly bound than the northwestern one. The virial mass within 1.7 Mpc is (5.1 ± 0.60)×1014 M ⊙, lower than the lensing mass. A significant transverse velocity component could mean the virial mass is underestimated. We contribute one new member to the previously known z = 4.32 galaxy group. Intrinsic (delensed) positions of the five secure group members span a physical extent of ∼60 kpc. 13 additional candidates selected by spectroscopic/photometric constraints are small and faint, with a mean intrinsic luminosity ∼2.2 mag fainter than L *. NIRCam imaging admits a fairly wide range of brightnesses and morphologies for the group members, suggesting a more diverse galaxy population in this galaxy overdensity.

Zinc is an essential micronutrient that promotes normal growth, development and immune function. In the context of persistent dietary zinc inadequacies, large-scale food fortification can help fill the gap between intake and requirements. Burkina Faso mandates wheat flour fortification with iron and folic acid. We used activity-based cost modelling to estimate the cost of adding zinc to the country's wheat flour fortification standard assuming (1) no change in compliance with the national standard, and (2) a substantial improvement in compliance. We used household food consumption data to model effective coverage, that is, the number of women of reproductive age (WRA) predicted to achieve adequate zinc density (zinc intake/1000 kcal) with the addition of fortification to diets. Without interventions, the prevalence of inadequate dietary zinc density was ~35.5%. With no change in compliance, the annual average incremental cost of adding zinc to fortified wheat flour was $10,347, which would effectively cover
