The presented research aims to define a parametric modelling methodology that allows, in short time and at a sustainable cost, the digital acquisition, modelling and semantic structuring of urban city blocks to facilitate 3D city modelling applied to historic centres. The methodology is based on field surveying and derives 3D data for the realisation of a parametric City Information Model (CIM). This is pursued through the adoption of parametric modelling as main method combined with AI procedures like supervised machine learning. In particular, the Visual Programming Language (VPL) Grasshopper is adopted as main working environment. The methodology proposed, called Scan-to-CIM, is developed to automate the cognitive operations of interpretation and input of surveying data performed in the field in order to create LoD4 city block models in a semi-automatic way. The proposed Scan-to-CIM methodology is applied to a city block located in the historic centre of Catania, Italy.

Long uphill stretches of single-carriageway rural roads with one lane per travel direction may reduce the Level of Service (LoS), due to the decreased speed of heavy vehicles. In those circumstances, a slowdown of traffic, resulting in the formation of platoons, may be generated due to the difficulty of performing overtaking maneuvers safely. To solve this critical issue, an additional (climbing) lane for slow vehicles may be included in the road platform. This study aims to evaluate the effectiveness of such climbing lanes in a real case in Italy (National Road n. 4 “Via Salaria”—around 44+000 km). Using a microsimulation model implemented in VISSIM, the study analyzes speeds and travel times, delays, and queuing waiting times, comparing the Actual Scenario (AS) without climbing lanes, with two counterfactual scenarios: the first one (CS1) with three stretches of climbing lanes, and the second one (CS2), with just two stretches, in which the first two additional lanes of CS1 are merged together. The obtained results confirm the effectiveness of installing climbing lanes on road sections with the described characteristics, and the potential of microsimulation models also to carry out such kind of evaluations.

Sandy barrier systems are highly dynamic, with the most significant natural morphological changes to these systems occurring during high-energy storm conditions. These systems provide a range of economic and ecosystem benefits and protect inland areas from flooding and storm impacts, but the persistence of many coastal barriers is threatened by storms and sea-level rise (SLR). This study employed observations and modeling to examine recent and potential future influences of storms on a sandy coastal barrier system in Nauset Beach, MA. Drone-derived imagery and digital elevation models (DEMs) of the study area collected throughout the 2023–2024 winter revealed significant alongshore variability in the geomorphic response to storms. Severe, highly localized erosion (i.e., an erosional "hotspot") occurred immediately south of the Nauset Bay spit as the result of a group of storms in December and January. Modeling results demonstrated that the location of the hotspot was largely controlled by the location of a break in a nearshore sandbar system, which induced larger waves and stronger currents that affected the foreshore, backshore and dune. Additionally, model simulations of the December and January storms assuming 0.3 m (1 ft) of SLR showed the system to be relatively resistant to major geomorphic changes in response to an isolated storm event, but more susceptible to significant overwash and breaching in response to consecutive storms. This research suggests that both very strong isolated storm events and sequential moderate storms pose an enhanced risk of major overwash, breaching, and possibly inlet formation today and into the future, raising concern for adjacent communities and resource managers. Gamma-oryzanol (ORY), found in rice (Oryza sativa L.), is a mixture of ferulic acid esters with triterpene alcohols, well-known for its antioxidant and anti-inflammatory properties. Our past research demonstrated its positive impact on cognitive function in adult mice, influencing synaptic plasticity and neuroprotection. In this study, we explored whether ORY can exert neuro-differentiating effects by using different experimental models. For this purpose, chemical characterization identified four components that are most abundant in ORY. In human neuroblastoma cells, we showed ORY’s ability to stimulate neurite outgrowth, upregulating the expression of GAP43, BDNF, and TrkB genes. In addition, ORY was found to guide adult mouse hippocampal neural progenitor cells (NPCs) toward a neuronal commitment. Microinjection of ORY in zebrafish Tg (-3.1 neurog1:GFP) amplified neurog1-GFP signal, islet1, and bdnf mRNA levels. Zebrafish nrf2a and nrf2b morphants (MOs) were utilized to assess ORY effects in the presence or absence of Nrf2. Notably, ORY’s ability to activate bdnf was nullified in nrf2a-MO and nrf2b-MO. Furthermore, computational analysis suggested ORY’s single components have different affinities for the Keap1-Kelch domain. In conclusion, although more in-depth studies are needed, our findings position ORY as a potential source of bioactive molecules with neuro-differentiating potential involving the Nrf2 pathway.

Turkey Hemorrhagic Enteritis (THE) is an acute disease caused by a Siadenovirus that affects 4 week-aged and older turkeys, characterized by acute depression, bloody droppings, and a high mortality rate. The immunosuppressive attributes of THE can protract disease progression and create a predisposition in birds towards subsequent bacterial infectiodoralns involving Escherichia coli and Clostridium perfringens (necrotic enteritis). Turkey Hemorrhagic Enteritis Virus (THEV) predominantly affects turkeys and carries substantial economic implications for this industry. Macrophages and B lymphocytes are recognized as the predominant target cells for the virus, while the spleen is the principal site of viral replication. Infected cells have also been observed in various other tissues, including the intestines, bursa of Fabricius, cecal tonsils, thymus, liver, kidney, peripheral blood leukocytes, and lungs. The economic relevance of this disease is derived both from the high mortality rate, which can reach 60% depending on the virulence of the strain, and from subclinical disease responsible for poor performance in vaccinated animals. This review aims to provide a comprehensive overview of THE, spanning etiology, epidemiology clinical signs and gross lesions, prevention, and management.

This paper describes the selection and validation of supporting measures (SMs) aimed at enhancing biosecurity compliance within Italian poultry farms. A tailored methodology, based on a stakeholders’ survey involving farmers and advisors, included a virtual farm tour, group discussion, and farmer coaching. Virtual farm tours and group discussions were delivered during two meetings targeting meat and egg production stakeholders, separately. Coaching was validated in 26 pilot farms (PFs) by assessing farmers’ attitudes towards change (i.e., ADKAR®) and farms’ biosecurity score (i.e., Biocheck.UgentTM) before and after a minimum six-month period. A total of 20 out of 26 farmers agreed to implement at least one action plan (AP). Full implementation of the agreed APs was observed in ten farms, while others only partially implemented (n = 7) or did not implement (n = 3) the improvement. Most APs focused on enhancing house hygiene locks (n = 7), followed by bacterial auto-control after cleaning and disinfection (n = 4). Scoring tools indicated minimal or no variations in farmers’ attitudes towards change and farm biosecurity. Virtual farm tours and group discussions were found to be effective in fostering interaction and facilitating the exchange of experiences and knowledge among farmers and stakeholders of poultry production. Coaching indicated that farmers might prefer implementing minor changes possibly influenced by time and cost constraints associated with structural interventions. These limitations could have also impacted the scores of the farmer/farm. The findings of this study provide a foundation for further application of SMs to improve biosecurity in Italian poultry farms.

The ability of Salmonella species to adhere to surfaces and form biofilms, leading to persistent environmental reservoirs, might represent a direct link between environmental contamination and food processing contamination. The purpose of this study was to investigate the biofilm-forming ability of 80 multidrug-resistant (MDR) and extended-spectrum beta-lactamase (ESBL) producing Salmonella enterica serovar Infantis strains isolated from the broiler food chain production through whole genome sequencing (WGS), PCR, and morphotype association assays. Biofilm formation was quantified by testing the strains at two different temperatures, using 96-well polystyrene plates. The rough and dry colony (rdar) morphotype was assessed visually on Congo red agar (CRA) plates. Based on our results, all tested S. Infantis strains produced biofilm at 22 °C with an rdar morphotype, while at 37 °C, all the isolates tested negative, except one positive. Most isolates (58.75%) exhibited strong biofilm production, while 36.25% showed moderate production. Only 5 out of 80 (6.25%) were weak biofilm producers. WGS analysis showed the presence of the fim cluster (fimADF) and the csg cluster (csgBAC and csgDEFG), also described in S. Typhimurium, which are responsible for fimbriae production. PCR demonstrated the presence of csgD, csgB, and fimA in all 80 S. Infantis strains. To our knowledge, this is the first study comparing the effects of two different temperatures on the biofilm formation capacity of ESBL producing S. Infantis from the broiler production chain. This study highlights that the initial biofilm components, such as curli and cellulose, are specifically expressed at lower temperatures. It is important to emphasize that within the broiler farm, the environmental temperature ranges between 18–22 °C, which is the optimum temperature for in vitro biofilm formation by Salmonella spp. This temperature range facilitates the expression of biofilm-associated genes, contributing to the persistence of S. Infantis in the environment. This complicates biosecurity measures and makes disinfection protocols on the farm and in the production chain more difficult, posing serious public health concerns.

In this update to the laserbeamFoam toolbox, a multi-component version of the solver is included with pair-wise Fickian Diffusion between selected component pairs. This new solver, multiComponentlaserbeamFoam, captures the multi-component nature of mixing between alloys, and as-such could be used to simulate dissimilar laser joining processes, and dissimilar laser-powder-bed additive manufacturing, among others. Additionally a multi-laser version of the original laserbeamFoam solver is included (arraylaserbeamFoam) that can simulate N incident laser sources on a metallic substrate, where the direction, velocity, and other laser characteristics can be specified independently. In both these solvers, as in the original laserbeamFoam solver, the phenomenological recoil pressure treatment is used, as opposed to the full volumetric dilation treatment, for computational tractability.

The structure and diversity of microbial communities are intrinsically hierarchical due to the shared evolutionary history of their constituents. This history is typically captured through taxonomic assignment and phylogenetic reconstruction, sources of information that are frequently used to group microbes into higher levels of organization in experimental and natural communities. Connecting community diversity to the joint ecological dynamics of the abundances of these groups is a central problem of community ecology. However, how microbial diversity depends on the scale of observation at which groups are defined has never been systematically examined. Here, we used a macroecological approach to quantitatively characterize the structure and diversity of microbial communities among disparate environments across taxonomic and phylogenetic scales. We found that measures of biodiversity at a given scale can be consistently predicted using a minimal model of ecology, the Stochastic Logistic Model of growth (SLM). This result suggests that the SLM is a more appropriate null-model for microbial biodiversity than alternatives such as the Unified Neutral Theory of Biodiversity. Extending these within-scale results, we examined the relationship between measures of biodiversity calculated at different scales (e.g. genus vs. family), an empirical pattern previously evaluated in the context of the Diversity Begets Diversity (DBD) hypothesis (Madi et al., 2020). We found that the relationship between richness estimates at different scales can be quantitatively predicted assuming independence among community members, demonstrating that the DBD can be sufficiently explained using the SLM as a null model of ecology. Contrastingly, only by including correlations between the abundances of community members (e.g. as the consequence of interactions) can we predict the relationship between estimates of diversity at different scales. The results of this study characterize novel microbial patterns across scales of organization and establish a sharp demarcation between recently proposed macroecological patterns that are not and are affected by ecological interactions.

After the second Intergovernmental Negotiating Committee (INC) for the Plastics Treaty meeting in Paris in June 2023, a Zero Draft of the Treaty was released for comment. Member states involved in the treaty negotiations were to respond to the Zero Draft before the third INC meeting in November 2023 in Nairobi. In this paper, we analyse the content and structure of the Zero Draft. We identify parts of the Zero Draft that work and others that will need further attention. These include applicability to different regions; atmospheric input; recycling and waste management; labelling and standards; harmful chemicals; scientific backing; the circular economy; just transition and climate change. We argue that this draft was a useful starting point for further negotiations for member states and the Revised version of the Zero Draft has resolved some of its shortcomings but not all. These are likely to contribute to further debate during future INC negotiations. We conclude with an overview of INC-3 and INC-4 and how the Zero Draft and Revised Zero Draft were received.

To address the global challenge of plastic waste, 175 UN Member States are negotiating a legally binding instrument, the Plastic Treaty, aimed at ending plastic pollution. This ambitious framework, targeting both terrestrial and marine sources, is being developed through Intergovernmental Negotiating Committee meetings scheduled to conclude by 2025. Amidst uncertainties and power dynamics influencing state positions, this research identifies 10 critical, yet unresolved, points within the treaty negotiations (i.e., the scope of the treaty, equity and differentiation, involvement of non-state actors, integration with existing agreements, standards and specifications, trade implications, monitoring and reporting, responsibility and historical contributions, binding vs. non-binding commitments, and financing and technology transfer). The findings suggest potential compromises in the treaty’s environmental provisions, influenced by domestic interests and international power disparities. As negotiations progress, the imperative for cooperation and decisive action against plastic pollution becomes increasingly pressing, challenging member states to prioritize global environmental integrity over national interests.
