Objective To investigate the effect of GLP-1 receptor agonist, exenatide, on the proliferation and osteoblastic differentiation by the human osteosarcoma MG63 cells. Methods Different concentrations of exenatide (0 mol/L, 10- 9mol/L, 10- 8mol/L, and 10- 7mol/L, respectively) were additioned to MG63 cells cultured in vitro. Cell proliferation was measured with cell counting kit-8 (CCK-8) after 24 h. Moreover,the supernatants of exenatide-treated MG63 cells were collected. AKP activity was tested using AKP kit. Results (1) Exenatide promoted proliferation of MG63 cells (P < 0. 05). With the increase of the concentration,the proliferation rate of MG63 cells declined (P < 0. 05). The most stimulation effect was achieved at the concentration of 10- 9mol/L. (2) The AKP activity of MG63 cells increased after treatment with exenatide, and the effect was the most obvious at the lower the concentration of exenatide (P < 0. 05). Conclusion Exenatide promotes proliferation and osteoblastic differentiation by human osteosarcoma MG63 cells. The falling-off cause of exhaust valve seats in a strengthened diesel engine is analyzed in terms of material selection and design. Three materials, cobalt, nickel and powder based alloy, are tested on their strengths, thermal expansion properties, friction properties and volume stabilities. The assembly interference is considered through FEM simulation, and the engine test is carried out to validate the reliability of the new material valve seat. The results show that nickel-based alloy has the lowest friction property. The volume stability of powder-based alloy is lower than that of cobalt-based alloy. Cobalt- based alloy has the highest temperature compressive and friction properties, and presents a good ability to defend the abnormal operations. Moreover, with the increase in the material strength, the assembly interference could be enhanced to 0.081~0.113 mm, with a safe coefficient of 1.4 for both cylinder head and valve seats. Medical caregivers as auxiliary medical care personnel have become an indispensable labor supplement in nursing work. Given the public's rising awareness of health and rights protection, the quality of patients' nursing staff has also improved accordingly. In order to strengthen life care skills and professional quality, it's required to standardize professional training. However, there are still many problems in the training of medical nursing staff and content in China. This article analyzed the current employment conditions, management and training needs of medical caregivers, aiming to provide reference for further establishment of standardized training curriculums.
