Water and nutrients, particularly phosphorus (P), are the two most limiting factors for dwarf bamboo growth in tropical and subtropical areas. Dwarf bamboo is highly sensitive to water stress and often causes severe P deficiency in its growing soils due to the characteristics of shallower roots and expeditious growth. However, little is known about its photoprotective response to soil water deficit and the underlying mechanisms regulated by P application. In this study, a completely randomized design with two factors of two water regimes (well-watered and water-stressed) and two P levels (with and without P application) was arranged to investigate this issue in dwarf bamboo ( Fargesia rufa ) plants. Water stress not only decreased water status and photochemical activity but also increased lipid peroxidation due to reactive oxygen species (ROS) accumulation irrespective of P application. In this case, thermal dissipation and antioxidative defense were promoted. Moreover, the role of the water−water cycle under this stress still could not be ignored because it accounted for a large proportion of total energy ( J PSII ). P application significantly enhanced photochemical activity accompanied by increased chlorophyll content in water-stressed plants. Meanwhile, P application remarkably reduced thermal dissipation and hardly affected photorespiration and the water−water cycle under water stress. Although P application only enhanced ascorbate (AsA) level, ROS, particularly hydrogen peroxide (H 2 O 2 ), and lipid peroxidation were significantly reduced in water-stressed plants. Therefore, P application can improve the photosynthetic capacity by regulating the redistribution of energy absorbed by PSII antennae and independently activating of the H 2 O 2 -scavenging function of AsA to alleviate oxidative damage in F. rufa plants, thereby improving their survival under water stress conditions. In this study, we have compared the photosynthetic characteristics of two contrasting species of Tradescantia plants, T. fluminensis (shade-tolerant species), and T. sillamontana (light-resistant species), grown under the low light (LL, 50-125 µmol photons m s) or high light (HL, 875-1000 µmol photons m s) conditions during their entire growth period. For monitoring the functional state of photosynthetic apparatus (PSA), we measured chlorophyll (Chl) a emission fluorescence spectra and kinetics of light-induced changes in the heights of fluorescence peaks at 685 and 740 nm ( F and F ). We also compared the light-induced oxidation of P and assayed the composition of carotenoids in Tradescantia leaves grown under the LL and HL conditions. The analyses of slow induction of Chl a fluorescence (SIF) uncovered different traits in the LL- and HL-grown plants of ecologically contrasting Tradescantia species, which may have potential ecophysiological significance with respect to their tolerance to HL stress. The fluorometry and EPR studies of induction events in chloroplasts in situ demonstrated that acclimation of both Tradescantia species to HL conditions promoted faster responses of their PSA as compared to LL-grown plants. Acclimation of both species to HL also caused marked changes in the leaf anatomy and carotenoid composition (an increase in Violaxanthin + Antheraxantin + Zeaxanthin and Lutein pools), suggesting enhanced photoprotective capacity of the carotenoids in the plants grown in nature under high irradiance. Collectively, the results of the present work suggest that the mechanisms of long-term PSA photoprotection in Tradescantia are based predominantly on the light-induced remodeling of pigment-protein complexes in chloroplasts. Nannochloropsis spp. are algae with high potential for biotechnological applications due to their capacity to accumulate lipids. However, little is known about their photosynthetic apparatus and acclimation/photoprotective strategies. In this work, we studied the mechanisms of non-photochemical quenching (NPQ), the fast response to high light stress, in Nannochloropsis gaditana by “locking” the cells in six different states during quenching activation and relaxation. Combining biochemical analysis with time-resolved fluorescence spectroscopy, we correlated each NPQ state with the presence of two well-known NPQ components: de-epoxidized xanthophylls and stress-related antenna proteins (LHCXs). We demonstrated that after exposure to strong light, the rapid quenching that takes place in the antennas of both photosystems was associated with the presence of LHCXs. At later stages, quenching occurs mainly in the antennas of PSII and correlates with the amount of de-epoxidised xanthophylls. We also observed changes in the distribution of excitation energy between photosystems, which suggests redistribution of excitation between photosystems as part of the photo-protective strategy. Light adaptive strategies were studied in a comparative analysis of the congeneric seagrass species Zostera muelleri and Zostera marina at two case study areas in New Zealand and Germany. Surveys in intertidal seagrass meadows were conducted from pre-dawn until sunset on days when either low or high tide coincided with noon. The results show marked fluctuations of photophysiology (optimum and effective quantum yield, non-photochemical quenching, cycling of xanthophyll cycle (XC) pigments) over daily and tidal cycles. At both locations, we observed a near complete conversion (de-epoxidation) of violaxanthin to zeaxanthin at times with high irradiance and a rapid and complete re-epoxidation under subsequent lower light conditions. At the New Zealand site we also observed significantly larger XC-pigment pool sizes in seagrass leaves sampled in a week when low tide coincided with noon (larger daily fluence and higher maximum irradiance), compared with leaves sampled in a week when high tide was at noon. This dynamic adjustment of xanthophyll pool size has not been previously reported for intertidal seagrasses. Diatoms contain a highly flexible capacity to dissipate excessively absorbed light by nonphotochemical fluorescence quenching ( NPQ) based on the light-induced conversion of diadinoxanthin (Dd) into diatoxanthin (Dt) and the presence of Lhcx proteins. Their NPQ fine regulation on the molecular level upon a shift to dynamic light conditions is unknown., We investigated the regulation of Dd + Dt amount, Lhcx gene and protein synthesis and NPQ capacity in the diatom Phaeodactylum tricornutum after a change from continuous low light to 3 d of sine ( SL) or fluctuating ( FL) light conditions. Four P. tricornutum strains with different NPQ capacities due to different expression of Lhcx1 were included., All strains responded to dynamic light comparably, independently of initial NPQ capacity. During SL, NPQ capacity was strongly enhanced due to a gradual increase of Lhcx2 and Dd + Dt amount. During FL, cells enhanced their NPQ capacity on the first day due to increased Dd + Dt, Lhcx2 and Lhcx3; already by the second day light acclimation was accomplished. While quenching efficiency of Dt was strongly lowered during SL conditions, it remained high throughout the whole FL exposure., Our results highlight a more balanced and cost-effective photoacclimation strategy of P. tricornutum under FL than under SL conditions. Main conclusion: A special domain of the thylakoid membrane of higher plants has been isolated which carries out the de-epoxidation of the xanthophyll cycle pigment violaxanthin to zeaxanthin. Recent models indicate that in the chloroplast of higher plants, the violaxanthin (V) cycle takes place within specialized domains in the thylakoid membrane. Here, we describe a new procedure to directly isolate such a domain in functional state. The procedure consists of a thylakoid membrane isolation at a pH value of 5.2 which realizes the binding of the enzyme V de-epoxidase (VDE) to the membrane throughout the preparation process. Isolated thylakoid membranes are then solubilized with the very mild detergent n-dodecyl α- d-maltoside and the pigment-protein complexes are separated by sucrose gradient ultracentrifugation. The upper main fraction of the sucrose gradient represents a V cycle domain which consists of the major light-harvesting complex of photosystem II (LHCII), a special lipid composition with an enrichment of the galactolipid monogalactosyldiacylglycerol (MGDG) and the VDE. The domain is isolated in functional state as evidenced by the ability to convert the LHCII-associated V to zeaxanthin. The direct isolation of a V cycle domain proves the most important hypotheses concerning the de-epoxidation reaction in intact thylakoid membranes. It shows that the VDE binds to the thylakoid membrane at low pH values of the thylakoid lumen, that it binds to membrane regions enriched in LHCII, and that the domain contains high amounts of MGDG. In this study, the effect of low light (LL, 340–360 μmol m −2 s −1 ) on thylakoid membrane activity, photosystem I and II (PSI and PSII) activities, transient quantum yields, reactive oxygen species (ROS), cyclic electron flow (CEF) and proton motive force of tomato leaves was investigated. Results indicated that LL treatment led to low integrity of the thylakoid membrane, ATPase activity, and photoinhibition of PSII and PSI. The treatment also yielded low electron transport rate [ETR(II) and ETR(I)], high PSI donor side limitation [Y(ND)] and efficient electron transfer between the intermediate carriers to the final acceptors of PSI (δRo). Hence, the possible inhibition sites include Q A -Q B and PSI-F d . Moreover, LL increased the excitation pressure, ROS scavenge enzyme activities, CEF/Y(II) radio, formation of proton gradient and decreased chlorophyll a/b ratio in the thylakoid membrane, thereby alleviating inhibition of PSII and PSI to a certain extent. The goal of this study was to explore the relationships between the photochemical reflectance index (PRI) at the leaf level and pigment pools, focusing on the constitutive role of pigments in influencing PRI over seasonal or ontogenetic time frames. The purpose was to re-evaluate the role of PRI as an indicator of seasonally shifting pigment (chlorophyll, carotenoid and anthocyanin) contents, and hence photosynthetic activity, across a range of tree and crop species. We studied natural vegetation - three tree species (maple, chestnut and beech) and two managed irrigated and rainfed crop species (maize and soybean), contrasting in photosynthetic pathway and leaf structure, and having wide variation of pigment content and composition. In anthocyanin-free leaves, PRI related to both total chlorophyll (Chl) and carotenoid (Car) contents, however, much closer relationships were found between PRI and Car to Chl ratio (Car/Chl). The sensitivity of PRI to Car/Chl varied widely in tree species with the degree of secondary carotenoids. In crop leaves where the Car vs. Chl relationship was very close, the slopes of PRI vs. Car/Chl relationships for maize and soybean were almost identical. PRI vs. Car/Chl relationships for leaves of different tree species formed a significant, uniform relationship with PRI. Two crops also formed a significant, uniform PRI vs. Car/Chl relationship with a slope half the value found for trees. In anthocyanin-containing leaves, PRI did not clearly relate to any pigment content because reflectance values at both PRI wavebands are affected by anthocyanin content. The findings of a strong link between leaf level PRI and Car/Chl over seasonal and ontogenetic time spans supports recent findings calling for a more careful evaluation of the relationship between PRI and either LUE or photosynthetic activity. In particular, studies that contrast short-term (e.g. diurnal) vs. long-term (e.g. seasonal) pigment, PRI, and photosynthetic responses in contrasting vegetation types are needed. The goal of this study was to explore the relationships between stand-level photochemical reflectance index (PRI) and canopy structure/pigment pools, as well as light use efficiency (LUE) of photosynthetically active vegetation focusing on seasonal or ontogenetic time frames. PRI was originally designed as a means of assessing the xanthophyll cycle and LUE over short (e.g. diurnal) time frames, and few studies have explored the drivers of PRI over longer, seasonal time frames, particularly in crops having different photosynthetic pathways or canopy structures. Consequently, our purpose was to understand and quantify the drivers of PRI responses over seasonal time scales for two crops, maize (C 4 ) and soybean (C 3 ), contrasting in photosynthetic pathway, leaf structure and canopy architecture. In both crops, PRI was very closely related to green LAI (R 2 > 0.90) and stand chlorophyll (Chl) content (R 2 > 0.93). The slopes of the relationships in different phenological stages, vegetative and reproductive, were substantially different (3-fold smaller in the vegetative stage). The main cause of this disparity was the high PRI value of soil/residue background. While PRI was a sensitive indicator of the changes in stand green LAI and stand Chl content over the full growing season, it was not sensitive to LUE; LUE explained below 12% of PRI variation in maize and 19% in soybean. Unlike leaf-level PRI, stand-level PRI was not clearly related to the Car/Chl ratio, presumably because the large changes in canopy structure (affecting stand Chl and green LAI) had a dominant influence on PRI over this time frame. The strong relationship between PRI and stand Chl content as well as between PRI and Chl-related vegetation index over a growing cycle allowed us to subtract the stand Chl content effect from measured PRI to reveal the component of PRI most likely related to periods of stress. However, for accurate subtraction of the Chl effect from long-term PRI records, thoughtful study of uncertainties related to “natural” variation of PRI-stand Chl relationships, and stand Chl content estimation for different varieties of the same species and for different species is required. The findings of a strong link between stand-level PRI and stand green LAI and Chl content and the lack of a clear relationship between PRI and LUE over seasonal and ontogenetic time spans suggest the need for a more careful evaluation of the relationship between PRI and either LUE or photosynthetic activity. In particular, studies that contrast short-term (e.g. diurnal) vs. long-term (e.g. seasonal) pigment, PRI, and photosynthetic responses in contrasting vegetation types are needed to clarify the different mechanisms involved at different temporal and spatial scales. These findings have important implications for attempts to monitor photosynthetic phenology from remote sensing, many of which have relied on PRI as an indicator of photosynthetic activity. Melatonin plays an important role in tolerance to multiple stresses in plants. Recent studies have shown that melatonin relieves photoinhibition in plants under cold stress; however, the mechanisms are not fully understood. Non-photochemical quenching (NPQ) is a key process thermally dissipating excess light energy that plants employ as a protective mechanism to prevent the over reduction of photosystem II. Here, we report the effects of exogenous melatonin on NPQ and mitigation of photoinhibition in tomato seedlings exposed to moderate light during chilling. In response to moderate light during chilling, the maximum quantum yield (Fv/Fm) and the effective photochemical efficiency (F0v/F0m) of PSII were both substantially reduced, showing severe photoinhibition in tomato seedlings, whereas exogenous application of melatonin effectively alleviated the photoinhibition. Further experiment showed that melatonin accelerated the induction of NPQ in response to moderate light and maintained higher level of NPQ upon longer exposure to light during chilling. Consistent with the increased NPQ was the elevated de-epoxidation state of xanthophyll pigments in melatonin-pretreated seedlings exposed to light during chilling. Enzyme activity assay showed that violaxanthin deepoxidase (VDE), which catalyzes the de-epoxidation reaction in the xanthophyll cycle, was activated by light and the activity was further enhanced by application of melatonin. Further analysis revealed that melatonin induced the expression of VDE gene in tomato seedlings under moderate light and chilling conditions. Ascorbic acid is an essential cofactor of VDE and the level of it was found to be increased in melatonin-pretreated seedlings. Feeding tomato seedlings with dithiothreitol, an inhibitor of VDE, blocked the effects of melatonin on the de-epoxidation state of xanthophyll pigments and the induction of NPQ. Collectively, these results suggest that exogenous melatonin mitigates photoinhibition by accelerating NPQ through the stimulation of VDE activity and the enhancement of de-epoxidation state of xanthophyll pigments. Micromonas strains of small prasinophyte green algae are found throughout the world's oceans, exploiting widely different niches. We grew arctic and temperate strains of Micromonas and compared their susceptibilities to photoinactivation of Photosystem II, their counteracting Photosystem II repair capacities, their Photosystem II content, and their induction and relaxation of non-photochemical quenching. In the arctic strain Micromonas NCMA 2099, the cellular content of active Photosystem II represents only about 50 % of total Photosystem II protein, as a slow rate constant for clearance of PsbA protein limits instantaneous repair. In contrast, the temperate strain NCMA 1646 shows a faster clearance of PsbA protein which allows it to maintain active Photosystem II content equivalent to total Photosystem II protein. Under growth at 2 °C, the arctic Micromonas maintains a constitutive induction of xanthophyll deepoxidation, shown by second-derivative whole-cell spectra, which supports strong induction of non-photochemical quenching under low to moderate light, even if xanthophyll cycling is blocked. This non-photochemical quenching, however, relaxes during subsequent darkness with kinetics nearly comparable to the temperate Micromonas NCMA 1646, thereby limiting the opportunity cost of sustained downregulation of PSII function after a decrease in light. Eustigmatophyte algae represent an interesting model system for the study of the regulation of the excitation energy flow due to their use of violaxanthin both as a major light-harvesting pigment and as the basis of xanthophyll cycle. Fluorescence induction kinetics was studied in an oleaginous marine alga Nannochloropsis oceanica. Nonphotochemical fluorescence quenching was analyzed in detail with respect to the state of the cellular xanthophyll pool. Two components of nonphotochemical fluorescence quenching (NPQ), both dependent on the presence of zeaxanthin, were clearly resolved, denoted as slow and fast NPQ based on kinetics of their formation. The slow component was shown to be in direct proportion to the amount of zeaxanthin, while the fast NPQ component was transiently induced in the presence of membrane potential on subsecond timescales. The applicability of these observations to other eustigmatophyte species is demonstrated by measurements of other representatives of this algal group, both marine and freshwater. Plants are permanently exposed to rapidly changing environments, therefore it is evident that they had to evolve mechanisms enabling them to dynamically adapt to such fluctuations. Here we study how plants can be trained to enhance their photoprotection and elaborate on the concept of the short-term illumination memory in Arabidopsis thaliana . By monitoring fluorescence emission dynamics we systematically observe the extent of non-photochemical quenching (NPQ) after previous light exposure to recognise and quantify the memory effect. We propose a simplified mathematical model of photosynthesis that includes the key components required for NPQ activation, which allows us to quantify the contribution to photoprotection by those components. Due to its reduced complexity, our model can be easily applied to study similar behavioural changes in other species, which we demonstrate by adapting it to the shadow-tolerant plant Epipremnum aureum . Our results indicate that a basic mechanism of short-term light memory is preserved. The slow component, accumulation of zeaxanthin, accounts for the amount of memory remaining after relaxation in darkness, while the fast one, antenna protonation, increases quenching efficiency. With our combined theoretical and experimental approach we provide a unifying framework describing common principles of key photoprotective mechanisms across species in general, mathematical terms. Photooxidative damage to the needle leaves of evergreen trees results from the absorption of excess excitation energy. Efficient dissipation of this energy is essential to prevent photodamage. In this study, we determined the fluorescence transients, absorption spectra, chlorophyll contents, chlorophyll a/ b ratios, and relative membrane permeabilities of needle leaves of Pinus koraiensis, Pinus tabulaeformis, and Pinus armandi in both cold winter and summer. We observed a dramatic decrease in the maximum fluorescence ( F ) and substantial absorption of light energy in winter leaves of all three species. The F decline was not correlated with a decrease in light absorption or with changes in chlorophyll content and chlorophyll a/ b ratio. The results suggested that the winter leaves dissipated a large amount of excess energy as heat. Because the cold winter leaves had lost normal physiological function, the heat dissipation depended solely on changes in the photosystem II supercomplex rather than the xanthophyll cycle. Information on plant responses to combined ozone and cadmium stresses are scarce and limited to herbaceous species. In this research, two poplar clones (I-214 and Eridano), differently sensitive to O3, were grown for 5 weeks in pots supplied with 0, 53.5, and 160.5 mg(Cd) kg-1 (soil d.m.) and then exposed to 15-d O3 fumigation (0.06 mm3 dm-3, 5 h a day). The effects of the two stressors, alone or in combination, on Cd, Ca, Fe, and Zn accumulation in above-nad below-ground organs, photosynthesis, leaf pigments, and accumulation of H2O2 and NO were investigated. Cadmium induced a reduction in stomatal conductance and a significant accumulation of H2O2 and NO in both clones nad negatively affected the carotenoid content in I-214. Ozone, on the other hand, counteracted Cd accumulation in the above-ground organs and significantly increased the xanthophyll de-epoxidation state indicating photoinhibition in O3-treated plants. Surprisingly, O3 alone or in combination with Cd decreased H2O2 accumulation in I-214. The NO production was generally stimulated by Cd, whereas it decreased following O3 exposure in I-214. The overall data indicate that Cd and O3 induced clone specific responses. Moreover, when they were applied in combination, antagonistic rather than synergistic effects were observed.

Phenotypic plasticity in response to environmental variation occurs at all organizational levels and across temporal scales within plants. However, the magnitude and functional significance of this plasticity is little explored in perennial species. We examined the influence of different light regimes and nitrogen (N) availability on the morphological and physiological plasticity of coffee seedlings (Coffea arabica L.). Potted plants were grown under full sunlight and shade (50%) and were fertilized with Hoagland’s solutions containing 0, 16 or 23mM N. Most leaf traits responded to light with a classic full sunlight vs. shade dichotomy [e.g., compared with those grown under full sunlight, 50% leaves had a thinner palisade mesophyll and a lower leaf mass per area (LMA) for improved light capture]. The outer periclinal cell walls in both epidermises exhibited thick epicuticular wax and three distinct layers. Chloroplasts of the mesophyll cells were densely occupied by thylakoids and starch grains. These characteristics were observed most clearly in plants supplemented by nitrogen or in those grown in shade conditions. Large starch granules were observed, but no membrane injuries were observed in either treatment. The plasticity index was high for the physiological traits that are associated with photoprotection and the maintenance of a positive carbon balance under shade but was low for most morpho-anatomical features.
