This work is part of a larger project on the study of algal biodiversity of a lotic system in La Pampa province. In this first stage we analyze the algal flora of samples collected during the summer season and autumn of 2010. The study area is located in the floodplain of the Colorado River, the natural boundary between the provinces of La Pampa and Río Negro at its intersection with Provincial Route 11 (Department Lihuel Calel). The methodology used in the collection followed the standard parameters for lotic environments. The samples were fixed with 4% formaldehyde and deposited in the herbarium of the Faculty of Agronomy (UNLPam) under the symbol SRFA. The preliminary results of the algal flora of summer and autumn shows 64 taxa, of which 26.5% corresponds to Cyanophyceae, 36% to Chlorophyceae and Bacillariophyceae, and 1.5% to Dinophyceae. During the summer the class Chlorophyceae species contributed 40% and fall did the Bacillariophyceae with 43.7%. The 37.5% of the registered species were common to both seasons.

Astaxanthin esters are a major form of astaxanthin found in nature. However, the exact mechanisms of the biosynthesis and storage of astaxanthin esters were previously unknown. We found that Schizochytrium sp. synthesized both astaxanthin and docosahexaenoic acid (DHA)-enriched lipids. The major type of astaxanthin produced was free astaxanthin along with astaxanthin-DHA monoester and other esterified forms. DHA accounted for 41.0% of the total fatty acids from astaxanthin monoesters. These compounds were deposited mainly in lipid droplets. The biosynthesis of the astaxanthin esters was mainly carried out by a novel diacylglycerol acyltransferase ScDGAT2-1, while ScDGAT2-2 was involved only in the production of triacylglycerol. We also identified astaxanthin ester synthases from the astaxanthin-producing algae Haematococcus pluvialis and Chromochloris zofingiensis , as well as a thraustochytrid Hondaea fermentalgiana with an unknown carotenoid profile. This investigation enlightens the application of thraustochytrids for the production of both DHA and astaxanthin and provides enzyme resources for the biosynthesis of astaxanthin esters in the engineered microbes.

The microalga Raphidocelis subcapitata was isolated from the Nitelva River (Norway) and subsequently deposited in the collection of the Norwegian Institute of Water Research as "Selenastrum capricornutum Printz". This freshwater microalga, also known as Pseudokirchneriella subcapitata, acquired much of its notoriety due to its high sensitivity to different chemical species, which makes it recommended by different international organizations for the assessment of ecotoxicity. However, outside this scope, R. subcapitata continues to be little explored. This review aims to shed light on a microalga that, despite its popularity, continues to be an "illustrious" unknown in many ways. Therefore, R. subcapitata taxonomy, phylogeny, shape, size/biovolume, cell ultra-structure, and reproduction are reviewed. The nutritional and cultural conditions, chronological aging, and maintenance and preservation of the alga are summarized and critically discussed. Applications of R. subcapitata, such as its use in aquatic toxicology (ecotoxicity assessment and elucidation of adverse toxic outcome pathways) are presented. Furthermore, the latest advances in the use of this alga in biotechnology, namely in the bioremediation of effluents and the production of value-added biomolecules and biofuels, are highlighted. To end, a perspective regarding the future exploitation of R. subcapitata potentialities, in a modern concept of biorefinery, is outlined. Untreated or poorly treated wastewater still represents environmental issues world-widely. Wastewater, especially saline wastewater treatment, is still primarily associated with high costs from physical and chemical processes, as high salinity hinders biological treatment. One favourable way is to find the suitable biological pathways and organisms to improve the biological treatment efficiency. In this context, halophilic microorganisms could be strong candidates to address the economics and effectiveness of the saline wastewater treatment process. Dunaliella salina is a photoautotrophic microalga that grows in saline environments. It is known for producing marketable bio-compounds such as carotenoids, lipids, and proteins. A biological treatment based on D. salina cultivation offers the opportunity to treat saline wastewater, reducing the threat of possible eutrophication from inappropriate discharge. At the same time, D. salina cultivation could yield compounds of industrial relevance to turn saline wastewater treatment into a profitable and sustainable process. Most research on D. salina has primarily focused on bioproduct generation, leaving thorough reviews of its application in wastewater treatment inadequate. This paper discusses the future challenges and opportunities of using D. salina to treat wastewater from different sources. In this study, two strains of symbiotic bacteria (SOB-1 and SOB-2) were isolated from Scenedesmus obliquus, and various algal-bacterial mutualistic systems were established under acetochlor (ACT) stress conditions. Following exposure to varying ACT concentrations from 2.0 to 25.0 μg/L, the capacity for co-cultured bacteria to degrade ACT was enhanced in 7 days by up to 226.9% (SOB-1) and 193.0% (SOB-2), compared with axenic algae, although bacteria exposed to higher ACT concentrations exacerbated algal metabolic stress, oxidative states, apoptosis and cellular lysis. ACT reduced carbohydrates in the phycosphere by up to 31.5%; compensatory nutrient plunder and structural damage by bacteria were the potential exploitation pathways determined based on the inhibition of bacterial infection using a glucanase inhibitor. The ACT-induced reduction in algal antimicrobial substances, including fatty acids and phenolics (by up to 58.1% and 56.6%, respectively), also facilitated bacterial exploitation of algae. ACT-dependent interspecific interaction coefficients between algae and bacteria generated from long-term symbiosis cultures implied that bacteria moved from mutualism (0 and 2.0 μg/L ACT) to exploitation (7.9 and 25.0 μg/L ACT). The population dynamic model under incremental ACT-concentration scenarios inferred that theoretical systematic extinction may occur in algal-bacterial systems earlier than in axenic algae. The Asian tiger mosquito, Aedes albopictus, is a highly invasive and aggressive species capable of transmitting a large number of etiological agents of medical and veterinary importance, posing a high risk for the transmission of emerging viruses between animals and humans. In this work, we evaluated the mosquitocidal activity of Neochloris aquatica against A. albopictus throughout its development and analyzed whether this effect was potentiated when the microalga was cultivated under stress conditions due to nutrient deprivation. Our results suggest that N. aquatica produces metabolites that have negative effects on these insects, including larval mortality, interruption of pupal development, and incomplete emergence of adults when fed on microalgae in the larval stages. When microalgae were cultured under stress conditions, an increase in molting defects was recorded, and the number of healthy adults emerged drastically decreased. Histological studies revealed severe signs of total disintegration of different tissues and organs in the thorax and abdomen regions. The muscles and fat bodies in the midgut and foregut were severely distorted. In particular, larval intestinal tissue damage included vacuolization of the cytoplasm, destruction of brush border microvilli, and dilation of the intercellular space, which are distinctive morphological characteristics of apoptotic cells. Astaxanthin accumulation in Haematococcus pluvialis typically occurs alongside the formation of secondary cell wall (SCW), hindering astaxanthin extraction and bio-accessibility. A potential solution lies in cultivating astaxanthin-rich motile cells lacking SCW. This study explored the influence and underlying mechanism of nitrogen-deprivation (ND) on SCW formation and established a connection between pyrimidine metabolism and SCW development. Then, various pyrimidine and ND combinations were examined to cultivate astaxanthin-rich motile cells. The results indicated that, compared to the nitrogen-replete group, the combination of uridine and ND increased the proportion of motile cells by 25-33 times, achieving 95 %, and enhanced astaxanthin yield by 26.52 %. Moreover, the efficiency of astaxanthin extraction from intact, wet motile cells was 91 % - 95 %, which was 5.6-9.0 times that from non-motile cells. Microalgal lipids are precursors to the production of biodiesel, as well as a source of valuable dietary components in the biotechnological industries. So, this study aimed to assess the effects of nutritional (nitrogen, and phosphorus) starvations and salinity stress (NaCl) on the biomass, lipid content, fatty acids profile, and predicted biodiesel properties of green microalga Monoraphidium braunii. The results showed that biomass, biomass productivity, and photosynthetic pigment contents (Chl. a, b, and carotenoids) of M. braunii were markedly decreased by nitrogen and phosphorus depletion and recorded the maximum values in cultures treated with full of N and P concentrations (control, 100%). These parameters were considerably increased at the low salinity level (up to 150 mM NaCl), while an increasing salinity level (up to 250 mM NaCl) reduces the biomass, its productivity, and pigment contents. Nutritional limitations and salt stress (NaCl) resulted in significantly enhanced accumulation of lipid and productivity of M. braunii, which represented more than twofold of the control. Furthermore, these conditions have enhanced the profile of fatty acid and biodiesel quality-related parameters. Microalgae play a crucial role in global carbon cycling as they convert carbon dioxide into various valuable macromolecules. Among them, Haematococcus pluvialis ( H. pluvialis ) is the richest natural source of astaxanthin (AXT), which is a valuable antioxidant, anti-inflammatory, and antiapoptosis agent. These benefits make AXT highly commercially valuable in pharmaceuticals, cosmetics, and nutritional industries. However, intrinsic genetic characteristics and extrinsic cultivation conditions influence biomass gains, leading to low productivity and extraction as the main techno-economic bottlenecks in this industry. Thus, detecting AXT in H. pluvialis is essential to determine the influence of multiple parameters on biocompound accumulation, enabling optimization of cultivation and enrichment of AXT-rich H. pluvialis cells. This work developed an opto-acousto-fluidic microplatform for detection, analysis, and sorting of microalgae. Via label-free monitoring and extraction of sample-induced ultrasonic signals, a photoacoustic microscopic system was proposed to provide a full-field visualization of AXT's content and distribution inside H. pluvialis cells. When employed as on-chip image-based flow cytometry, our microplatform can also offer high-throughput measurements of intracellular AXT in real time, which demonstrates similar results to conventional spectrophotometry methods and further reveals the heterogeneity of AXT content at the single-cell level. In addition, a solenoid valve-pump dual-mode cell sorter was integrated for effective sorting of cells with a maximum working frequency of 0.77 Hz, reducing the fluid response time by 50% in rising and 40-fold in recovery. The H. pluvialis cells which have more AXT accumulation (>30 μm in diameter) were 4.38-fold enriched with almost no dead empty and small green cells. According to the results, automated and reliable photoacoustics-activated cell sorting (PA-ACS) can screen AXT-rich cells and remove impurities at the terminal stage of cultivation, thereby increasing the effectiveness and purity of AXT extraction. The proposed system can be further adopted to enrich strains and mutants for the production of biofuels or other rare organic substances such as β-carotene and lutein.

Microalgae not only play a vital role in the ecosystem but also hold promising commercial applications. Conventional methods of detecting and monitoring microalgae rely on field sampling followed by transportation to the laboratory for manual analysis, which is both time-consuming and laborious. Although machine learning (ML) algorithms have been introduced for microalgae detection in the laboratory, no integrated platform approach has yet emerged to enable real-time, on-site sampling and analysing. To solve this problem, here, we develop an automated and intelligent microfluidic platform (AIMP) that can offer automated system control, intelligent data analysis, and user interaction, providing an economical and portable solution to alleviate the drawbacks of conventional methods for microalgae detection and monitoring. We demonstrate the feasibility of the AIMP by detecting and classifying four microalgal species ( Cosmarium , Closterium , Micrasterias , and Haematococcus Pluvialis ) that exhibit varying sizes (from a few to hundreds of microns) and morphologies. The trained microalgae species detection network (MSDN, based on YOLOv5 architecture) achieves a high overall mean average precision at 0.5 intersection-over-union (mAP@0.5) of 92.8%. Furthermore, the versatility of the AIMP is demonstrated by long-term monitoring of astaxanthin production from Haematococcus Pluvialis over a period of 30 days. The AIMP achieved 97.5% accuracy in the detection of Haematococcus Pluvialis and 96.3% in further classification based on astaxanthin accumulation. This study opens up a new path towards microalgae detection and monitoring using portable intelligent devices, providing new ideas to accelerate progress in the ecological studies and commercial exploitation of microalgae.

Layered double hydroxides (LDHs), regarding their physical and structural properties, have different and wide applications industry and their increasing use may raise ecological and human health concerns. However, the potential toxicity mechanisms of LDHs in different organisms are still unclear. In the present work, after synthesizing of ZnFe-SO 4 LDH and studying of its characterization by XRD, FT-IR, SEM, EDX-mapping, TEM and Raman, its toxicity in Tetradesmus obliquus was evaluated. According to experimental results, the growth of the algae and content of photosynthetic pigments were significantly decreased after treatment with 100 mg/L of ZnFe-SO 4 LDH. The high dose exposure to the LDH also inhibited the activity of SOD and POD enzymes, possibly due to the LDH- catalyzed reactive oxygen species production. In addition, lipid peroxidation and the content of phenolic compounds, as no-enzymatic antioxidants were increased by enhancement of the LDH concentration. The rise of phenol, flavonoids and MDA contents could be regarded as some manifestations and responses to the toxic effects of the contaminant in the algae cells. Sea lettuce ( Ulva ) is a genus of green macroalgae present along all the coasts of the world's oceans. It represents about 100 species with diverse habitats. Inter- and intra-species natural variation is very large, both in terms of growth characteristics and biomass biochemical composition. As a result, Ulva biomass has a wide range of applications and strain selection can achieve significant increases in yield(s). Establishing solid, long term and cost-effective methodologies for the conservation of Ulva genetic diversity is then required to safeguard and reuse selected strains. Here, we report a cryopreservation-based protocol for the long-term preservation of foliose Ulva strains. Strains from seven different Ulva species were cryopreserved for 15 and/or 120 days in liquid nitrogen, and of the 3 replicates cryopreserved, at least one survived, allowing us to successfully recover all strains. Astaxanthin is a red-colored secondary metabolite with excellent antioxidant properties, typically finds application as foods, feed, cosmetics, nutraceuticals, and medications. Astaxanthin is usually produced synthetically using chemicals and costs less as compared to the natural astaxanthin obtained from fish, shrimps, and microorganisms. Over the decades, astaxanthin has been naturally synthesized from Haematococcus pluvialis in commercial scales and remains exceptional, attributed to its higher bioactive properties as compared to synthetic astaxanthin. However, the production cost of algal astaxanthin is still high due to several bottlenecks prevailing in the upstream and downstream processes. To that end, the present study intends to review the recent trends and advancements in astaxanthin production from microalgae. The structure of astaxanthin, sources, production strategies of microalgal astaxanthin, and factors influencing the synthesis of microalgal astaxanthin were discussed while detailing the pathway involved in astaxanthin biosynthesis. Aquatic organisms are exposed to several compounds that occur in mixtures in the environment. Thus, it is important to investigate their impacts on organisms because these combined effects can be potentiated. Cobalt (Co) and nickel (Ni) are metals that occur in the environment and are used in human activities. To the best of our knowledge, there are no studies that investigated the combined effects of these metals on a freshwater Chlorophyceae. Therefore, this study analyzed the isolated and combined effects of Co and Ni in cell density, physiological and morphological parameters, reactive oxygen species (ROS), carbohydrates and photosynthetic parameters of the microalga Raphidocelis subcapitata. Data showed that Co affected the cell density from 0.25 mg Co L -1 ; the fluorescence of chlorophyll a (Chl a) (0.10 mg Co L -1 ); ROS production (0.50 mg Co L -1 ), total carbohydrates and efficiency of the oxygen evolving complex (OEC) at all tested concentrations; and the maximum quantum yield (Φ M ) from 0.50 mg Co L -1 . Ni exposure decreased ROS and cell density (0.35 mg Ni L -1 ); altered Chl a fluorescence and carbohydrates at all tested concentrations; and did not alter photosynthetic parameters. Regarding the Co-Ni mixtures, our data best fitted the concentration addition (CA) model and dose-ratio dependent (DR) deviation in which synergism was observed at low doses of Co and high doses of Ni and antagonism occurred at high doses of Co and low doses of Ni. The Tunuyán and Mendoza River Basins (Province of Mendoza, Argentina) have been selected as a representative semiarid region to test the applicability of an integrated water quality evaluation. To detect spatio-temporal variations of anthropic contamination, physicochemical and bacteriological parameters, as well as three ecotoxicological assays, were assessed in reference sites for 3 years. Bioassays based on the nematode Caenorhabditis elegans, the vascular plant Lactuca sativa, and the algae Pseudokirchneriella subcapitata were performed and toxicological categories were established. Our results showed that water quality, as well as water toxicity, deteriorates as both river systems run through urban areas. Interestingly, monitoring sites with good physicochemical and bacteriological qualities but with toxicity were identified, illustrating that traditional water quality studies do not predict potential toxic effects on living organisms. In addition, a multivariate statistical analysis was performed to detect clusters of monitoring sites according to the water quality status. Crocin is one of the most valuable components of the Chinese medicinal plant Crocus sativus and is widely used in the food, cosmetics, and pharmaceutical industries. Traditional planting of C. sativus is unable to fulfill the increasing demand for crocin in the global market, however, such that researchers have turned their attention to the heterologous production of crocin in a variety of hosts. At present, there are reports of successful heterologous production of crocin in Escherichia coli , Saccharomyces cerevisiae , microalgae, and plants that do not naturally produce crocin. Of these, the microalga Dunaliella salina , which produces high levels of β -carotene, the substrate for crocin biosynthesis, is worthy of attention. This article describes the biosynthesis of crocin, compares the features of each heterologous host, and clarifies the requirements for efficient production of crocin in microalgae.

This research examines the biological treatment of undiluted vegetable waste digestate conducted in a bubble column photobioreactor. Initially, the bioreactor containing 3N-BBM medium was inoculated with Microglena sp., Tetradesmus obliquus, and Desmodesmus subspicatus mixture with a density of 1.0 × 10 4 cells/mL and the consortium was cultivated for 30 days. Then, the bioreactor was semi-continuously fed with liquid digestate with hydraulic retention time (HRT) of 30 days, and the treatment process was continued for the next 15 weeks. The change in the microalgal and cyanobacterial species domination was measured in regular intervals using cell counting with droplet method on a microscope slide. At the end of the experiment, Desmonostoc sp. cyanobacteria (identified with 16S ribosomal RNA genetical analysis) as well as Tetradesmus obliquus green algae along with Rhodanobacteraceae and Planococcaceae bacteria (determined with V3-V4 16sRNA metagenomic studies) dominated the microbial community in the photobioreactor. The present study shows the characterization of the bacterial communities associated with different systems during the cultivation of the microalga Tetradesmus obliquus. For that, sequential cultivation was performed in three different systems: (1) Photobioreactor bench-scale; (2) flat-panel photobioreactor; and (3) thin-layer cascade. Cultures were monitored daily for growth parameters and biomass samples were collected for characterization of bacterial communities using metagenomic. A total of 195,177 reads were produced, resulting in the identification of 72 OTUs. In the grouping of bacterial communities, 3 phyla, 6 classes, 28 families, and 35 taxa were found. The bacteria Brevundimonas and Porphyrobacter had a higher relative abundance compared with other taxa found. These taxa were present in all cultivation systems forming a possible core community. The effect of salicylic acid (SA), an endogenous plant growth regulator, on adaptation of the green microalga, Dunaliella salina to N starvation was investigated through the study of enzymatic antioxidant system and biochemical changes. Algal cells in the exponential growth phase were exposed to N deficiency with 100 μM of SA. N starvation significantly decreased cell number, chlorophyll a, and hydrogen peroxide and while highly increased levels of fresh weight, soluble sugars, starch, proteins, free amino acids, and the activity of antioxidant enzymes. N-starved cells treated with SA enhanced cell number and hydrogen peroxide content, but accumulated lower amounts of metabolites and enzymatic activities compared to untreated cultures. However, the levels of fresh weight, chlorophyll, β-carotene, and soluble proteins remained roughly unchanged relative to N starvation alone. Proteolytic activity was well correlated with accumulation of amino acids in control and other treatments. The results suggest that exogenous SA treatment can enhance adaptation to N starvation by establishing the enzymatic balance to adjust levels of metabolites and directing them to the growth processes. With a unique cell structure and clear genetic background, Chlamydomonas reinhardtii has become a model species for fundamental research on microalgae and lower plants. Genetic transformations in the nucleus and chloroplast and mitochondrial genomes of C. reinhardtii have been successfully developed. However, research of C. reinhardtii has mainly focused on physiology and molecular genetics and few studies investigating cultivation methods have been performed. Small scales, slow growth, and low yields from conventional photoautotrophic and mixotrophic cultures severely limit the further development and application of C. reinhardtii. This study sought to optimize and scale up the heterotrophic cultivation process for C. reinhardtii. Critical parameters in 500-mL shake flasks were optimized as follows: liquid volume of 150 mL, initial inoculation of 0.08 g L−1, growth temperature of 30 °C, and shaking speed of 150 rpm. In addition, this study confirmed that, although C. reinhardtii can assimilate acetic acid for heterotrophic growth (optimum initial concentration of 80 mM), neither glucose nor glycerol can be the only carbon source. Nitrate, ammonia, and urea can be used as nitrogen sources for heterotrophic culture, while the optimal carbon and nitrogen ratio for heterotrophic culture was 40:1. Under optimal conditions, the C. reinhardtii biomass increased from 0.457 to 1.32 g L−1. Furthermore, for the first time, heterotrophic cultures of C. reinhardtii were scaled up to 5-L and 50-L fermenters, in which the cell density reached 25.44 g L−1 after 237 h, which is 2.82-fold higher than previously reported. Abstract Microalgae have been widely used in ecotoxicological studies in order to evaluate the impacts of heavy metals in aquatic ecosystems. However, there are few studies that analyze the effects of metals in an integrative way on photosynthetic apparatus of freshwater microalgae in the generation of reactive oxygen species (ROS) and biochemical composition. Therefore, this study aimed to assess cadmium (Cd) and lead (Pb) toxicity using synchronously physiological and biochemical endpoints, specially detecting lipidic classes for the very first time during Cd and Pb-exposure to Raphidocelis subcapitata. Here we show that analyzing the algae growth, the IC50–72 h for Cd was 0.04 µM and for Pb was 0.78 µM. In general, the Cd affected the biochemical parameters more, leading to an increase in total lipid content (7.2-fold), total carbohydrates (3.5-fold) and ROS production (3.7-fold). The higher production of lipids and carbohydrates during Cd-exposure probably acted as a defense mechanism, helping to reduce the extent of damage caused by the metal in the photosynthetic apparatus. For Pb, the physiological parameters were more sensitive, which resulted in changes of chlorophyll a synthesis and a reduction of both efficiency of oxygen-evolving complex and quantum yields. Besides that, we observed changes in the lipid class composition during Cd and Pb-exposure, suggesting these analyses as great biomarkers to assess metal toxicity mechanisms in ecological risk assessments. Thereby, here we demonstrate the importance of using multiple endpoints in ecotoxicological studies in order to obtain a better understanding of the mechanisms of metal toxicity to R. subcapitata. Abstract Carotenoid sources in shrimp diets have shown to be effective for improving survival, growth, reproductive capacity, stress resistance, and also for diminishing disease. Dunaliella sp. is known to have high levels of β-carotenes, which works as pro-vitamin A, enhancing the immune response in shrimp. However, the administration of Dunaliella sp. in shrimp diet needs to be evaluated to determine the appropriate dose and frequency of administration needed to optimize performance in cultured white shrimp. Diets with three different concentrations of Dunaliella sp. flour (1.5, 2 and 3%) were tested, and each one was administered at three different time frequencies: daily, and at 3- and 7-days intervals. Shrimp fed for 20 days were then infected with Vibrio parahaemolyticus (1 × 106 CFU/mL). Hemolymph parameters including protein, glucose, lactate, cholesterol and triglycerides were analyzed to evaluate shrimp stress status. Additionally, L. vannamei innate non-specific immune response was examined by evaluating the activity of prophenoloxidase (proPO), phenoloxidase (PO) and superoxide dismutase (SOD) in hemolymph; shrimp survival was also recorded. Survival after infection with V. parahaemolyticus was higher for shrimp fed with diets consisting of 2% Dunaliella sp. administered every 3 and 7 days. Shrimp fed a diet consisting of 2% or 3% Dunaliella sp. administered every third day showed positive physiological and immune responses to infection. A decrease in lipid oxidation in plasma triglycerides was observed at 48 h post inoculation in shrimp fed at all diets regimes due to Dunaliella sp. antioxidant action. Warming and copper (Cu) toxicity are two key abiotic stressors that strongly affect cell growth, photosynthetic rate, and metabolism in microalgae. In this study, a freshwater chlorophyte, Scenedesmus quadricauda, was exposed to various concentrations of copper sulfate (300, 600, and 1000 μM nominal concentrations of CuSO4·5H2O) at 25 and 35 °C. The changes in cell density, photosynthetic parameters, in vivo absorption spectra, reactive oxygen species (ROS) levels, and metabolic profile were analyzed. The effects of copper toxicity on the physiology and biochemistry of microalgae were highly dependent on water temperature. The interactive effects of both stressors induced significant impact on the photosynthetic parameters such as maximum quantum yield (Fv/Fm), saturation irradiance (Ek), and non-photochemical quenching (NPQ). Temperature induced significant impact on cell density, Ek and NPQ, while the Cu toxicity significantly affected the Fv/Fm and NPQ. Changes in the in vivo absorption spectra and high levels of reactive oxygen species (ROS) were observed across different treatments. Overall, S. quadricauda adapted to the two abiotic stresses via NPQ and metabolic restructuring. Key metabolites including glycine, proline, hexadecanoic acid, propanoic acid, octadecanoic acid, galactose, lactose, and sucrose were involved in the microalgal response. The feasibility of utilizing crude glycerol as carbon source for heterotrophic growth of the green microalgae Chlorella pyrenoidosa and Coccomyxa subellipsoidea C-169 was investigated. The highest biomass concentration of C. pyrenoidosa (6.25 g L−1) and C. subellipsoidea C-169 (7.62 g L−1) was achieved in basal medium containing 5 and 10 g L−1crude glycerol. Compared to pure glycerol and glucose, the algal cells grown in crude glycerol media obtained a higher intracellular protein content, while the microalgal lipid consists of a large amount of unsaturated fatty acids. The most abundant fatty acids in C. pyrenoidosa and C. subellipsoidea C-169 were linolenic acid (C18:3) and palmitic acid (16:0). The cetane number (49.0) of both investigated algal species corresponds to the requirements of the Standard B100. Our results indicate that crude glycerol could provide a promising alternative feedstock for heterotrophic growth of these two microalgae. [ABSTRACT FROM AUTHOR]

We sampled the first mitogenomes from a novel lineage of the Chlorophyceae that is sister to Sphaeropleales. Although the Jenufa perforata (27,180 bp) and Jenufa minuta (41,488 bp) mitogenomes differ markedly in size, gene order and intron content, their repertoires of conserved genes are essentially identical. Their rRNA genes display the same fragmentation patterns as their counterparts in Sphaeropleales. Their tRNA gene complements include two ancestral genes shared with Oedogoniales and Chaetophorales (Chlorophyceae) as well as a derived gene for a tRNA species decoding UGA as tryptophan. Human gut microbiota plays an important role in several metabolic processes and human diseases. Various dietary factors, including complex carbohydrates, such as polysaccharides, provide abundant nutrients and substrates for microbial metabolism in the gut, affecting the members and their functionality. Nowadays, the main sources of complex carbohydrates destined for human consumption are terrestrial plants. However, fresh water is an increasingly scarce commodity and world agricultural productivity is in a persistent decline, thus demanding the exploration of other sources of complex carbohydrates. As an interesting option, marine seaweeds show rapid growth and do not require arable land, fresh water or fertilizers. The present review offers an objective perspective of the current knowledge surrounding the impacts of seaweeds and their derived polysaccharides on the human microbiome and the profound need for more in-depth investigations into this topic. Animal experiments and in vitro colonic-simulating trials investigating the effects of seaweed ingestion on human gut microbiota are discussed.
