Objective. To construct the eukaryotic expression vector hypoxia-inducible factor 1α-[pcDNA.sub.3,1] and to investigate its transfective efficiency into mesenchymal stem cells (MSCs) in vitro and the expression of HIF-1α gene in MSCs. Methods. mRNA of Wistar Rats' myocardial cells was extracted, and cDNA was synthesized with Reverse Transcription Kit, HIF-1α was amplified by polymerase chain reaction (PCR), and constructed into pcDNA31. Transfected HIF-1α-[pcDNA.sub.3,1] into MSCs by liposome mediated method. The expression of HIF-1a in the cells was detected by Western Blot Analysis and ELISA. Results. Eukaryotic expression vector HIF-1α-[pcDNA.sub.3,1] was constructed successfully. Analyzed by flow cytometer, The MSCs' surfaces mark were CD44+, SH3(CD73)+, CD34-,CD45- and the CD44+ cells and SH3(CD73)+ cells were 94.7% and 97.3%, respectively, showing the high purity of the cultured MSCs. After inducing, the cultured MSCs can differentiate into osteoblasts and adipocytes successfully. In HIF-1α gene transfected MSCs, the expression of HIF-1α mRNA and HIF-1α protein were both increased obviously. Conclusion. HIF-1α was cloned successfully. Open heart surgery supported by cardiopulmonary bypass is associated with heart and lung ischemia-reperfusion injury (IRI). Limb remote ischemic preconditioning (RIPC) reduces injury caused by ischemia-reperfusion in multiple distant organs. We conducted a prospective clinical trial (randomized and controlled) to test the feasibility and safety of limb RIPC, as well as its protective effects against myocardial and pulmonary IRI for infants undergoing repair of simple congenital heart defects. Infants undergoing repair of ventricular septal defects were enrolled in our study and randomly assigned to one of two treatment groups: limb RIPC or control. RIPC was induced twice (24 h and 1 h preoperatively) via three 5-min cycles of ischemia and reperfusion on the left upper arm using a blood pressure cuff. Lung compliance, respiratory index (RI), and cardiac inotropic score (IS) were calculated for each patient. Serum concentrations of the following factors were measured perioperatively: interleukin (IL)-6, IL-8, IL-10, and tumor necrosis factor (TNF)-alpha; lactate dehydrogenase (LDH), creatine kinase (CK), and its isoenzyme (CK-MB), and troponin I (TnI); malondialdehyde (MDA) and superoxide dismutase (SOD). The expression of heat shock protein 70 (HSP 70) in cardiomyocytes was analyzed by Western blot. Surgical outcomes, including limb movement and sensory function, were recorded in detail. Sixty infants weighting less than 7 kg were studied, with 30 patients in the RIPC group and 30 in the control group. Within 6 months of discharge from the hospital, no limb disability, sensory disturbance, or other surgical complications were found in any patient. Compared with the control group, patients in the RIPC group had higher Cs and Cd, along with lower RI and IS at various postoperative phases. At the beginning of the operation, serum concentrations of IL-6, IL-8, IL-10, TNF-alpha, LDH, CK, and TnI were higher in the RIPC group than the control group. Postoperatively, release of cytokines and leakage of heart enzymes were attenuated in the RIPC group; serum concentrations of cytokines and heart enzymes were lower in the RIPC group at some, but not all, postoperative time points. Furthermore, the RIPC group had lower coronary sinus venous concentrations of MDA and higher concentrations of SOD. Similarly, the expression of HSP 70 was upregulated in cardiomyocytes from the RIPC group. Limb RIPC can be applied safely and easily in infants, can attenuate systemic inflammatory response syndrome, and can increase systemic tolerance to IRI, imparting a protective effect against myocardial and pulmonary IRI. The expression of HSP 70 has an important role in the mechanism of action for RIPC.

A geometric active contour model without re-initialization that can be used for grey and color image segmentation is presented in this paper. It combines directional information about edge location based on Cumani operator as a part of driving force, with the improved geodesic active contours containing Bays error based statistical region information. Moreover, an extra term that penalizes the deviation of the level set function from a signed distance function is also included in the model, thus the costly re-initialization procedure can be completely eliminated and all these measures are integrated in a unified frame. Experimental results on real grey and color images have shown that our model can precisely extract contours of images and its performance is much better and faster than the geodesic-aided C-V (GACV) model.

The Late Triassic to Paleogene (T3-E) basin occupies an area of 143100 km2, being the sixth area of the whole of SE China; the total area of synchronous granitoid is about 127300 km2; it provides a key for understanding the tectonic evolution of South China. From a new 1:1500000 geological map of the Mesozoic-Cenozoic basins of SE China, combined with analysis of geometrical and petrological features, some new insights of basin tectonics are obtained. Advances include petrotectonic assemblages, basin classification of geodynamics, geometric features, relations of basin and range. According to basin-forming geodynamical mechanisms, the Mesozoic-Cenozoic basin of SE China can be divided into three types, namely: 1) para-foreland basin formed from Late Triassic to Early Jurassic (T3-J1) under compressional conditions; 2) rift basins formed during the Middle Jurassic (J2) under a strongly extensional setting; and 3) a faulted depression formed during Early Cretaceous to Paleogene (K1-E) under back-arc extension action. From the rock assemblages of the basin, the faulted depression can be subdivided into a volcanic-sedimentary type formed mainly during the Early Cretaceous (K1) and a red-bed type formed from Late Cretaceous to Paleogene (K2-E). Statistical data suggest that the area of all para-foreland basins (T3-J1) is 15120 km2, one of rift basins (J2) occupies 4640 km2, and all faulted depressions equal to 124330 km2 including the K2-E red-bed basins of 37850 km2. The Early Mesozoic (T3-J1) basin and granite were mostly co-generated under a post-collision compression background, while the basins from Middle Jurassic to Paleogene (J2-E) were mainly constrained by regional extensional tectonics. Three geological and geographical zones were surveyed, namely: 1) the Wuyishan separating zone of paleogeography and climate from Middle Jurassic to Tertiary; 2) the Middle Jurassic rift zone; and 3) the Ganjiang separating zone of Late Mesozoic volcanism. Three types of basin-granite relationships have been identified, including compressional (a few), strike-slip (a few), and extensional (common). A three-stage geodynamical evolution of the SE-China basin is mooted: an Early Mesozoic basin-granite framework; a transitional Middle Jurassic tectonic regime; intracontinental extension and red-bed faulted depressions since the Late Cretaceous.

Background In China, the optimal management of individuals living with chronic HBV infection (CHB) remains an unmet need. The EVOLVE Study was a 5-year prospective, longitudinal, observational study that compared the clinical outcomes in treatment-naive CHB patients receiving entecavir (ETV) or lamivudine (LAM)-based therapies.Methods Males or females aged ≥18 years, diagnosed with CHB regardless of cirrhosis or hepatitis B e antigen (HBeAg) status were enrolled from tier 2 city hospitals (between 2012–2014). The choice of initial therapy and subsequent treatment modifications was at the discretion of treating physicians. Key outcomes included treatment modifications, virological response (HBV DNA <300 copies/ml) and HBV disease progression.Results Of the 3,408 patients enrolled, 1,807 and 628 received ETV and LAM-based therapy, respectively. The mean age was 39.5 years, 74% were male and 22.9% had cirrhosis. The rate of treatment modification was higher in the LAM-based versus ETV group (25.9% versus 13.7%); viral breakthrough was the most common reason in the LAM-based group versus financial reasons in the ETV group. At week 240, the virological response rate was 73% in both treatment groups. Compared with LAM-based therapy, ETV was associated with a significantly lower incidence of viral breakthrough (12.6% versus 2.1%) and genotypic resistance (10.1% versus 1.2%; P<0.0001 for both); significantly lower risk of HBV disease progression (14.0% versus 10.7%; P=0.0113); and lower rates of progression to decompensated cirrhosis (9.6% versus 6.4%) and hepatocellular carcinoma (1.9% versus 0.8%).Conclusions This real-world, longitudinal study demonstrated a significantly lower risk of HBV-related disease progression, viral breakthrough and resistance with ETV versus LAM-based therapy. The results indicate that the Weishan granite is a multistage batholith formed during the late Indosinian-early Yanshanian time. The intruded time of the late Indosinian granite is 211.0±1.6Ma and 215.7±1.9Ma (two samples), whereas that of the early Yanshanian granite is 187.4±3.5Ma and 184.5±5.1Ma (two samples). In combination with other geochronological data for Indosinian rocks of South China and the adjacent region, it is inferred that the late Indosinian granites of South China (especially Hunan Province) are probably formed under extension regime as a consequence of post-collision stress relaxation, which is a spontaneous response to intracontinental thickening attributed to the collision and extrusion of two Indosinian seams, namely Qinling-Dabie and Song Ma. Moreover, it is also deduced that the early Yanshanian granites of Hunan Province could not be directly related with the subduction of Paleo-Pacific plate towards Eurasian continent, and they are most likely derivation of the mid-or lower-crustal materials because of decompressional melting under the continuous extension setting.

Characteristics of progressive ductile deformation and remnant igneous fabrics indicate that the foliated diorites in the middle sector of the Jiangshao fracture belt in Zhuji County, Zhejiang, are mylonites deformed from diorites. The mylonitization occurred about 353–370 Ma ago under a condition of 425–475°C and 3.8–4.6 kb. The relation between the deformation and metamorphism was discussed based on petrographic evidence and mineral chemistry and assemblage. It is suggested that the deformation was responsible for the development of low-grade greenschist-facies metamorphism of mylonitic diorites by introducing fluid phase to accelerate metamorphic reaction and diflusion.

Metamorphic xenoliths within the Nushan alkali basalt of northeastern Anhui (NEA), China, are from the middle-lower crust. They could be divided into two end-members: basic and acid. Intermediate xenoliths are scarcely found. Basic two-pyroxene granulites (pyriclasites) were formed at 720–810° C and 7–8 kb. Petrological and geochemical studies indicate that the primary magma of the protoliths of basic granulites was derived from the metasomatized upper mantle, while the parental magma of the acid end-member was probably produced by partial melting of the basic rocks. The protoliths of charnockites and grey gneisses represent respectively the early and late crystallization products of the granitic magma. The Nushan granulites are much different in many aspects from the granulites exposed in the northern part of North China, which implies the inhomogeneity regarding to the early evolution of the North China terrane.

The Jiangshan-Shaoxing fracture belt (JSFB) is a Late Proterozoic geosuture due to island arc-continent collision in South China. The Cathaysian Block (CT), lying on the southeast side of JSFB, is composed of green schist-amphibolite complexes in the form of a series of tectonic flakes. On the northwest side of JSFB, which is located in the border area of Zhejiang, Jiangxi and Anhhi provinces ( abbreviated as ZJP-JXP-AHP ), are distributed an ophiolite suite and other rocks, constituting the Jiangnan ancient island arc (JN) on the southeast margin of the Yangtze Block (Y Z). The collision between JN and CT at R∼ 0.9Ga ago led to the folding of JN, followed by the intrusion (∼ 0.9–0.8Ga ago) of many dioritic and ultramafic stitching plutons along the fracture belt. As a result, the basic Precambrian tectonic framework of southeastern China was shaped.
