Objective To investigate the protective effects of FK506 combined with low temperature mechanical perfusion on the reduction of ischemia reperfusion injury of rat liver transplants. Methods SD rats were divided into HTK group and FK506 + HTK group. Orthotopic liver transplantation was performed 3 hours after machine perfusion of 150 mL HTK (HTK group) or FK506+150 mL HTK (FK506 + HTK group). Blood samples were collected at 3 and 24 hours after transplantation in both groups, and serum samples were centrifuged for liver function test. Twenty-four hours after transplantation, liver tissues were collected and fixed with formalin for HE staining. Results Serum ALT levels in FK506+HTK group were significantly lower than that in HTK group (P＜0.05). Inflammatory infiltration in FK506+HTK group was less than that in the HTK group, and the liver structure was intact. Conclusions FK506 added into perfusate for machine perfusion of the donor liver can effectively reduce liver function damage and inflammation damage within 3 hours after transplantation, and has a certain protective effect on the liver.

The coupling relationship between polymer gel system and reservoir pores is an important basis for scientific formulation of tertiary oil recovery programs. The polymer gel profile control agent can significantly modify the fluid migration path inside the reservoir. In this study, artificial cores were used to carry out indoor water flooding experiments after polymer gel injection that considering formation permeability, polymer concentration and polymer injection rates. Based on the accurate records of the breakthrough pressure and injection volume during water flooding, as well as a large number of observations of the microscopic morphology of the cores after the polymer gel profile control and displacement, a set of criteria for determining reservoir-matched polymer gel profile control agents is proposed. The experimental results show that a complex spatial network structure is formed after the gelation of the polymer gel system. As the polymer concentration increases, the network structure of the gel system becomes denser. At the same time, the unit network structure is becoming smaller, and the order of the grid chain structure becomes clearer. Under the condition of low polymer concentrations, the backbone structures of the gel are obvious, but there are a lot of fine pores. When the polymer concentration is greater than 1,500 mg/L and the injection rate is less than 0.15 ml/min, the water flooding breakthrough pressure is greater than 0.2 MPa, the plugging rate is greater than 95%, and a good plugging effect is obtained.

Objective To screen the differential proteins related to aging in human urine and to find out the functional changes of proteins related to aging. Methods Fourteen healthy men and 15 healthy women were enrolled and divided them into three groups according to their ages (20-30, 40-50 and over 60 years old). The differential proteins were screened by label-free quantification and 2DLC-MS/MS. Results In male group, there were 100 proteins differentially expressed in three groups, 66 were over-expressed and 34 were under-expressed. In female group, there were 94 proteins differentially expressed in three groups, 79 were overexpressed and 15 were under-expressed. There were 12 differential expressed proteins related to aging in both male and female groups. These protein pathways mainly focused on coagulation system, protein ubiquitination pathway, LXR/RXR activation and lactose degradation III, etc. Conclusions Proteins related to aging can be found in urine, which may be used as biomarkers of aging and providing basis for subsequent discovery of disease biomarkers.
