Abstract

We prepared a new rare-earth compound, Eu9MgS2B20O41 (EMSBO), and characterized its structural and physical properties. EMSBO consists of triple-Kagome-layer slabs separated by nonmagnetic ions and groups. Within each slab, intervalence charge transfer has been found to occur between the Eu2+ and Eu3+ Kagome layers, a new channel for quantum fluctuation of magnetic moments. The measured magnetic susceptibilities and the specific heat capacity exhibit very similar features characteristic of quantum spin liquid behaviors observed in other materials.

Abstract

The critical interactions at the dimer interface of kappa-bungarotoxin was analyzed to determine the significance of dimer interface on the activity of kappa-neurotoxins. Analysis of mutated kappa-bungarotoxin residues such as R54A, T60A, Q58A and L57A indicated the presence of beta-sheet conformations that are similar to native kappa-bungarotoxin. Furthermore, the folding process was disrupted in kappa-bungarotoxin mutants due to the trapping of the partially folded polypeptide in the form of disulfide-linked aggregates.

Abstract

Breastfeeding is the most complete nutritional method of feeding infants, but several impediments affect the decision to breastfeed, including questions of drug safety for medications needed during lactation. Despite recent FDA guidance, few labels provide clear dosing advice during lactation. Physiologically based pharmacokinetic modeling (PBPK) is well suited to mechanistically explore pharmacokinetics and dosing paradigms to fill gaps in the absence of extensive clinical studies and complement existing real‐world data. For lactation‐focused PBPK (Lact‐PBPK) models, information on system parameters (e.g., expression of drug transporters in mammary epithelial cells) is sparse. The breast cancer resistance protein (BCRP) is expressed on the apical side of mammary epithelial cells where it actively transports drugs/substrates into milk (reported milk: plasma ratios range from 2 to 20). A critical review of BCRP and its role in lactation was conducted. Longitudinal changes in BCRP mRNA expression have been identified in women with a maximum reached around 5 months postpartum. Limited data are available on the ontogeny of BCRP in infant intestine; however, data indicate lower BCRP abundance in infants compared to adults. Current status of incorporation of drug transporter information in Lact‐PBPK models to predict active secretion of drugs into breast milk and consequential exposure of breast‐fed infants is discussed. In addition, this review highlights novel clinical tools for evaluation of BCRP activity, namely a potential non‐invasive BCRP biomarker (riboflavin) and liquid biopsy that could be used to quantitatively elucidate the role of this transporter without the need for administration of drugs and to inform Lact‐PBPK models. Abstract

Wetlands and ponds are frequently used to remove nitrate from effluents or runoffs. However, the efficiency of this approach is limited. Based on the assumption that introducing vertical mixing to water column plus carbon addition would benefit the diffusion across the sediment-water interface, we conducted simulation experiments to identify a method for enhancing nitrate removal. The results suggested that the sediment-water interface has a great potential for nitrate removal, and the potential can be activated after several days of acclimation. Adding additional carbon plus mixing significantly increases the nitrate removal capacity, and the removal of total nitrogen (TN) and nitrate-nitrogen (NO3(-)-N) is well fitted to a first-order reaction model. Adding Hydrilla verticillata debris as a carbon source increased nitrate removal, whereas adding Eichhornia crassipe decreased it. Adding ethanol plus mixing greatly improved the removal performance, with the removal rate of NO3(-)-N and TN reaching 15.0-16.5 g m(-2) d(-1). The feasibility of this enhancement method was further confirmed with a wetland microcosm, and the NO3(-)-N removal rate maintained at 10.0-12.0 g m(-2) d(-1) at a hydraulic loading rate of 0.5 m d(-1).

Abstract

TRIM13, a member of the TRIM family, is a RING domain containing E3 ubiquitin ligase which plays critical roles in diverse cellular processes including cell death, cancer and antiviral immunity. However, its expression and molecular mechanism on renal cell carcinoma (RCC) have not been characterized. This study explored the clinical significance and biological function of TRIM13 in human RCC. Western blot analyses and Immunohistochemical were performed in RCC tissues. The clinical relevance of TRIM13 in RCC was evaluated by immunohistochemical staining using tissue microarray. The role of TRIM13 in migration was studied in renal cell carcinoma cell lines of 786-O through knocking down TRIM13 with siRNA and over-expression of TRIM13. The regulation of TRIM13 on migration and invasion were determined by wound-healing and transwell assays. Western blot analyses showed that TRIM13 expression was dramatically decreased in RCC tissues compared with adjacent non-tumorous tissues. Up-regulation of TRIM13 in 786-O cells resulted in decreased NF-kB, MMP-9 and p-AKT levels and the capability for migration and invasion. In contrast, the ectopic expression of TRIM13 decreased the migration and invasion ability of 786-O cells. These findings indicate that TRIM13 decreases RCC metastasis and invasion may serve as a candidate RCC prognostic marker and a potential therapeutic target. [ABSTRACT FROM AUTHOR]

Abstract

A 7-port microstrip grid array is firstly presented in this paper, showing sufficient design flexibility to configure the large microstrip grid array into one or two unbalanced antennas as well as one or two balanced antennas. As an example, we designed and fabricated the 7-port microstrip grid array of 145 radiating elements in RT/duroid 5880 substrate with a size of 100×100×0.381 mm3 for 24-GHz applications. The measured results show that the 7-port microstrip grid array has achieved excellent performances: 1.57% impedance bandwidth, 4.51% gain bandwidth, and 25.15-dBi gain at 24.12 GHz as an unbalanced antenna, 2.11% impedance bandwidth, 4.80% gain bandwidth, and 25.36-dBi gain at 24.09 GHz as a balanced antenna, and 3.06% impedance bandwidth, 5.42% gain bandwidth, and 25.82-dBi gain at 24.73 GHz as two balanced antenna. The proposed structure can also operate as MIMO antennas with envelope correlations between radiation patterns of the two antenna elements lower than −30 dB over the 10-dB input impedance bandwidth of the proposed structure.

Abstract

In this paper, a microstrip grid array antenna is designed and compared to bandwidth-enhanced patch array antenna. The proposed microstrip grid array antenna has 145 radiating elements which are on a single-layer electrically-thin RT/duroid 5880 substrate. By adding two stubs near the feed point, the impedance matching of large microstrip grid array antenna is achieved without affecting its radiation characteristics. Long sides of the meshes far from the feed point are removed to decrease the sidelobe level. The comparison between proposed microstrip grid array antenna and patch array antenna indicates that the microstrip grid array antenna has higher peak realized gain, higher aperture efficiency, and lower cross-polarization level.

Abstract

Sex chromosomes evolved from autosomes many times across the eukaryote phylogeny. Several models have been proposed to explain this transition, some involving male and female sterility mutations linked in a region of suppressed recombination between X and Y (or Z/W, U/V) chromosomes. Comparative and experimental analysis of a reference genome assembly for a double haploid YY male garden asparagus (Asparagus officinalis L.) individual implicates separate but linked genes as responsible for sex determination. Dioecy has evolved recently within Asparagus and sex chromosomes are cytogenetically identical with the Y, harboring a megabase segment that is missing from the X. We show that deletion of this entire region results in a male-to-female conversion, whereas loss of a single suppressor of female development drives male-to-hermaphrodite conversion. A single copy anther-specific gene with a male sterile Arabidopsis knockout phenotype is also in the Y-specific region, supporting a two-gene model for sex chromosome evolution.

194. Rapid and visual RPA-CRISPR/Cas12a detection for Staphylococcus pseudintermediusand methicillin-resistant S. pseudintermediusin clinical samples of dogs and cats

Abstract

Staphylococcus pseudintermediuscan cause severe infections of the skin, ear and other tissues in dogs and cats. Methicillin-resistant S. pseudintermedius(MRSP) has recently become more prevalent, posing a severe threat to companion animals and public health. Therefore, rapid and accurate diagnosis of S. pseudintermediusand MRSP infections in dogs and cats is essential for timely controlling infections. The development of CRISPR/Cas technology offers an innovative solution for rapid diagnosis. Here, we established an assay combining recombinant polymerase amplification (RPA) and CRISPR/Cas12a. By separately detecting the spsJgene, the specific gene of S. pseudintermedius, and the mecAgene, the methicillin resistance gene, this method allows for the direct detection of methicillin-susceptible S. pseudintermedius(MSSP) and MRSP in clinical samples at 37 °C for a total of 40 min, The results can be directly visualized by the naked eye under blue light. The limits of detection of the RPA-CRISPR/Cas12a assay were 103copies per reaction for the spsJgene and 104copies per reaction for the mecAgene. The RPA-CRISPR/Cas12a detection successfully detected and differentiated clinical isolates of MSSP and MRSP without cross-reactivity with other tested bacteria species. The evaluation of the detection performance of RPA-CRISPR/Cas12a with 47 clinical samples (without culture) from dogs and cats showed that the results of detection were 100% consistent with those of clinical culture and colony sequencing, which was more sensitive than PCR. RPA-CRISPR/Cas12a assay can quickly and sensitively detect S. pseudintermediusand MRSP in clinical samples without expensive instruments, making it suitable for small veterinary clinics.
