Abstract

Mutations in ABCA1 uniformly decrease plasma HDL-cholesterol (HDL-C) and reduce cholesterol efflux, yet different mutations in ABCA1 result in different phenotypic effects in heterozygotes. For example, truncation mutations result in significantly lower HDL-C and apoliprotein A-I (apoA-I) levels in heterozygotes compared with nontruncation mutations, suggesting that truncation mutations may negatively affect the wild-type allele. To specifically test this hypothesis, we examined ABCA1 protein expression in response to 9-cis-retinoic acid (9-cis-RA) and 22-R-hydroxycholesterol (22-R-OH-Chol) in a collection of human fibroblasts representing eight different mutations and observed that truncation mutations blunted the response to oxysterol stimulation and dominantly suppressed induction of the remaining full-length allele to 5–10% of wild-type levels. mRNA levels between truncation and nontruncation mutations were comparable, suggesting that ABCA1 expression was suppressed at the protein level. Dominant negative activity of truncated ABCA1 was recapitulated in an in vitro model using transfected Cos-7 cells. Our results suggest that the severe reduction of HDL-C in patients with truncation mutations may be at least partly explained by dominant negative suppression of expression and activity of the remaining full-length ABCA1 allele.

Abstract

Salt-affected soils are extensively present and constitute about 7% of total land surface. However, our knowledge about greenhouse gas (GHG) turnover between the atmosphere and the saline soils is very limited. In order to evaluate the potential of GHG consumption in saline soils, we measured gas fluxes of carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) from the soil surface to the atmosphere under saline-alkaline mudflat and various community types in the Yellow River Delta in China. In general, the emissions of GHG of different ecosystems showed a unique peak diurnal pattern, with the peak at 13:00 h and the lowest value at 07:00 h. The CH4and N2O emission of different ecosystems followed the order: saline-alkaline mudflat>T. chinensis>S. salsa>P. australis, while the CO2emission followed the order:T. chinensis>P. australis>S. salsa> saline-alkaline mudflat. On the whole, saline-alkaline mudflat and different vegetations acted as CO2and N2O source in spring, while saline-alkaline mudflat andP. australiscommunities acted as CH4source and CH4sink, respectively. However,T. chinensisandS. salsacommunities acted as CH4sink before 12:00 h and CH4source after 12:00 h. Although measurements of the CO2, CH4and N2O fluxes were taken simultaneously, CH4and N2O fluxes were strongly correlated with soil moisture, temperature and electrical conductivity, while no significant correlation was found between CO2flux with above environmental factors. These results probably suggest that factors other than soil temperature, moisture and salinity exerted a larger impact on fluxes than on CH4and N2O release and/or that there were not enough samples for CO2flux measurements because of its higher spatial and temporal variability. Abstract

The genomic organisation of the seven cultivated Vigna species, V. unguiculata, V. subterranea, V. angularis, V. umbellata, V. radiata, V. mungo and V. aconitifolia, was determined using sequential combined PI and DAPI ( CPD) staining and dual-colour fluorescence in situ hybridisation ( FISH) with 5S and 45S rDNA probes. For phylogenetic analyses, comparative genomic in situ hybridisation ( cGISH) onto somatic chromosomes and sequence analysis of the internal transcribed spacer ( ITS) of 45S rDNA were used. Quantitative karyotypes were established using chromosome measurements, fluorochrome bands and rDNA FISH signals. All species had symmetrical karyotypes composed of only metacentric or metacentric and submetacentric chromosomes. Distinct heterochromatin differentiation was revealed by CPD staining and DAPI counterstaining after FISH. The rDNA sites among all species differed in their number, location and size. cGISH of V. umbellata genomic DNA to the chromosomes of all species produced strong signals in all centromeric regions of V. umbellata and V. angularis, weak signals in all pericentromeric regions of V. aconitifolia, and CPD-banded proximal regions of V. mungo var. mungo. Molecular phylogenetic trees showed that V. angularis and V. umbellata were the closest relatives, and V. mungo and V. aconitifolia were relatively closely related; these species formed a group that was separated from another group comprising V. radiata, V. unguiculata ssp. sesquipedalis and V. subterranea. This result was consistent with the phylogenetic relationships inferred from the heterochromatin and cGISH patterns; thus, fluorochrome banding and cGISH are efficient tools for the phylogenetic analysis of Vigna species.
