155. Efficacy and safety evaluation of PSOX, DOF and SOX regimens as neoadjuvant chemotherapy for advanced gastric cancer.

Abstract

Aim: To explore the efficacy and safety of paclitaxel+oxaliplatin+S-1 (PSOX), docetaxel+oxaliplatin+fluorouracil (DOF) and oxaliplatin+S-1 (SOX) regimens as neoadjuvant chemotherapy for advanced gastric cancer (GC). Methods: A retrospective analysis was used in 306 patients with GC who underwent neoadjuvant chemotherapy, consisting of 102 from the PSOX group, 100 from the DOF group and 104 from the SOX group. Results: The total effective rates and disease control rates for the PSOX, DOF and SOX groups were 31.4, 18 and 16.3% and 96.1, 94 and 92.3%, respectively. The highest total effective rate and disease control rate were found in the PSOX groups. Moreover, no difference among the PSOX, DOF and SOX groups on the incidence of adverse events was observed (p > 0.05). Conclusion: The PSOX regimen is an alternative neoadjuvant chemotherapy regimen for GC patients. [ABSTRACT FROM AUTHOR]

Abstract

ObjectiveEarly detection of a tumour remains an unmet medical need, and approaches with high sensitivity and specificity are urgently required. Mass cytometry time-of-flight (CyTOF) is a powerful technique to profile immune cells and could be applied to tumour detection. We attempted to establish diagnostic models for hepatocellular carcinoma (HCC) and pancreatic ductal adenocarcinoma (PDAC).DesignWe performed CyTOF analysis for 2348 participants from 15 centres, including 1131 participants with hepatic diseases, 584 participants with pancreatic diseases and 633 healthy volunteers. Diagnostic models were constructed through random forest algorithm and validated in subgroups.ResultsWe determined the disturbance of systemic immunity caused by HCC and PDAC, and calculated a peripheral blood immune score (PBIScore) based on the constructed model. The PBIScore exhibited good performance in detecting HCC and PDAC, with both sensitivity and specificity being around 80% in the validation cohorts. We further established an integrated PBIScore (iPBIScore) by combining PBIScore and alpha-fetoprotein or carbohydrate antigen 19-9. The iPBIScore for HCC had an area under the curve (AUC) of 0.99, 0.97 and 0.96 in training, internal validation and external validation cohorts, respectively. Similarly, the iPBIScore for PDAC showed an AUC of 0.99, 0.98 and 0.97 in the training, internal validation and external validation cohorts, respectively. In early-stage and tumour-marker-negative patients, our iPBIScore-based models also showed an AUC of 0.95–0.96 and 0.81–0.92, respectively.ConclusionOur study proved that the alterations of peripheral immune cell subsets could assist tumour detection, and provide a ready-to-use detection model for HCC and PDAC.

Crocin is a kind of bioactive constituent found in the stigmas of saffron, which has shown various pharmacological activities.Methods: In this study, we investigated the inhibitory effect of crocin on gastric cancer AGS cells proliferation and explored the underlying mechanism. A series of methods were used including cell counting kit assay, gene microarray analysis, qRT-PCR, Celigo image cytometry, cell clone formation assay, Western blot, and cell xenograft growth in vivo.Results: The results indicated that crocin inhibited AGS cells proliferation and promoted cell apoptosis. Further studies suggested that crocin decreased a series of genes expression, among which TPM4 gene downregulation inhibited the tumor cells proliferation and tumor growth in mice, and overexpression of TPM4 gene abolishes the inhibitory effect of crocin. Further study using microarray analysis suggested that knocking down of TPM4 altered genes related to the proliferation and apoptosis of cells.Discussion: Crocin could inhibit the gastric cancer cells AGS cells proliferation by regulating TPM4 gene expression, and TPM4 may be a promising therapeutic target for GC treatment.Keywords: gastric cancer, crocin, TPM4, HER2

Abstract

Ultrafine full-vulcanized polybutadiene rubber (UFBR) in particle sizes of ca. 50-100 nm has been used for modifying mechanical and processing performances of polypropylene (PP), and PP-g-maleic anhydride (PP-MA) has been used as a compatibilizer for enhancing the interfacial adhesion between the two components. The results show that PP/UFBR possesses rheological behaviors such as highly branched PP when UFBR content in blends reaches 10 wt%, while in contrast, the much low content of UFBR combining small amount of PP-MA endows the material with rheological characteristics of high melt strength materials like highly branched PP. The mechanism accounting for the rheological behaviors of binary blends and effectiveness of PP-MA on PP/UFBR blends has been proposed. POLYM. ENG. Abstract

Gastric cancer is the fourth most common cause of cancer death globally and the second most common in Asia. Many studies suggest that Crocin has the potential for gastric cancer antineoplastic combined chemotherapy protocols. Here we investigated genomic changes related to the inhibitory effect of Crocin, and elucidated the molecular mechanism of this inhibition in gastric carcinoma cells. We found that, compared with the control group, 216 significantly upregulated and 301 significantly downregulated genes were identified in Crocin-treated AGS cells. Many of these differentially expressed genes in AGS cells are involved in Nrf2-mediated oxidative stress response, p53 signaling, and integrin signaling, which suggested the mechanism of Crocin functions in therapy of gastric cancer. In summary, our study indicates that Crocin has the potential for gastric cancer adjuvant treatment through reducing cell oxidative stress levels.

Abstract

Monitoring mitochondrial derived copper(II) in live cells is highly demanded, but accurately detecting is unmet due to the interference with cytoplasmic copper(II). Herein, we have reported the design, synthesis and characterization of photocontrollable fluorogenic probe, M Cu -3, which is equipped with a photo-labile group (nitrobenzyl group) and mitochondria targeting unit (triphenylphosphonium salt). This novel probe showed an intense fluorescence enhancement in response to copper(II) without interference from other metal cations in the biological condition (pH 6–9). The detection limit is 1.7 × 10 −7 mol/L in HEPES buffer. The confocal fluorescence imaging results demonstrated M Cu -3 can visualize mitochondrial copper(II) in live mammalian cells. The clear advantage of our photocontrollable method is successful to avoid the influence of cytoplasmic copper(II) during mitochondria specific detection.

197. Research progress on LED quality regulating cell growth and target chemicals accumulation in microalgae.

Abstract

Light is the key factor affecting photoautotrophic growth of microalgae. The huge advantages of light quality supplied by light-emitting diodes (LEDs) (LQSLs) in promoting microalgal growth and target chemicals accumulation have been gaining great attention from science and the relevant industries. Moreover, the microalgal cultivation process based on LQSLs is an emerging technology for high-efficient production of microalgae-based chemicals. This review aims to provide an overview of the absorption spectra and photoreceptors in microalgae, the effects of different LQSLs on microalgae growth and target chemicals accumulation, and the cultivation strategies for high-efficient production of target chemicals by harnessing LQSLs. The path forward for further economical and high-efficient production of microalgal target chemicals using LQSLs with respect to 4 different opportunities is also discussed, including novel LEDs development to enhance the conversion efficiencies of electricity-to-light and light-to-biomass, high- cell-density cultivation of microalgae using LQSLs technology for carbon neutrality, next-generation photobioreactors design based on internal LEDs illumination for industrial application, and employing different LQSLs as signals for promoting the production of light-dependent chemicals in microalgae under heterotrophic/mixotrophic cultivation.
