Abstract Background Existing methods for preparing influenza vaccines pose the greatest challenge against highly pandemic avian influenza H7N9 outbreak in the poultry and humans. Exploring a new strategy for manufacturing and delivering a safe and effective H7N9 vaccine is needed urgently. Results An alternative approach is to develop an influenza H7N9 oral vaccine based on yeast display technology in a timely manner. Hemagglutinin (HA) of A/Anhui/1/2013 (AH-H7N9) is used as a model antigen and characterized its expression on the surface of Saccharomyces cerevisiae (S.cerevisiae) EBY 100. Mice administrated orally with S.cerevisiae EBY100/pYD5-HA produced significant titers of IgG antibody as well as significant amounts of cytokines IFN-γ and IL-4. Importantly, S.cerevisiae EBY100/pYD5-HA could provide effective immune protection against homologous A/Anhui/1/2013 (AH-H7N9) virus challenge. Conclusions Our findings suggest that platform based on yeast surface technology provides an alternative approach to prepare a promising influenza H7N9 oral vaccine candidate that can significantly shorten the preparedness period and result in effective protection against influenza A pandemic.

Soft material robots are developing rapidly benefited from their inherent flexibility, adaptability and safety compared to rigid-bodied robots. However, most soft robots are unable to offer high force/strength due to the low rigidity of soft materials that they are composed of. Absence of position feedback is another problem for soft robots. In this research, we aim to address these two challenges in a novel designed soft smart robotic hand. The design of this soft hand is also delicately considered to make it 3D printable, which shortens the design cycle and reduces the fabrication time. This hand consists of five fingers and a palm, all of which can be actuated independently. The finger is designed with two chambers: one air-tight active chamber which can be actuated by compressed air, one passive chamber filled with loosely arranged particles. During bending actuation, particles inside the passive chamber are squeezed by pressurized air, which causes passive jamming. As a result, the stiffness of the finger is strengthened during bending, which endows the hand with larger force output and load-holding capability. Furthermore, position feedback modules made of conductive elastomers are integrated and co-printed with the finger during fabrication. In this research, a hand prototype is manufactured and several experiments regarding to its characteristics and performance are conducted for evaluation. From experimental results, the soft hand achieves maximum holding weight of 1.452kg with particles and 0.8425kg without particles at same actuation pressure of 450kPa.

Streaming media has been largely used by millions of users every day. The number of customers and programs, e.g., TV series, movies, and various shows, are still growing fast. However, the demand for video transcoding for various personal terminal devices results in the shortage of computing resources and the prolongation of processing delay in centralized video transcoding systems. To solve this issue, we propose a blockchain, especially, smart contract based scheme that can achieve decentralized and on-demand crowdsourcing for video transcoding, which remarkably mitigates the transcoding overhead. Specifically, our scheme consists of four key components such as employers, workers, task allocation, and payment. An employer initializes the smart contract, releases the task, and initiates the smart contract. Workers bid for the task, and the successful bidder will obtain the task and execute the task. The task allocation mechanism and the payment mechanism can guarantee the profits of both and encourage both as well. Moreover, the smart contract consists of the bidding contract and the task execution contract. The extensive analysis of our proposed scheme justified the feasibility, security for defending against typical threats, applicability in realistic situations, and portability for most multimedia such as videos and audios.

BackgroundThe latest guidance on chronic fatigue syndrome (CFS) recommends exercise therapy. Tai Chi, an exercise method in traditional Chinese medicine, is reportedly helpful for CFS. However, the mechanism remains unclear. The present longitudinal study aimed to detect the influence of Tai Chi on functional brain connectivity in CFS.MethodsThe study recruited 20 CFS patients and 20 healthy controls to receive eight sessions of Tai Chi exercise over a period of one month. Before the Tai Chi exercise, an abnormal functional brain connectivity for recognizing CFS was generated by a linear support vector model. The prediction ability of the structure was validated with a random forest classification under a permutation test. Then, the functional connections (FCs) of the structure were analyzed in the large-scale brain network after Tai Chi exercise while taking the changes in the Fatigue Scale-14, Pittsburgh Sleep Quality Index (PSQI), and the 36-item short-form health survey (SF-36) as clinical effectiveness evaluation. The registration number is ChiCTR2000032577 in the Chinese Clinical Trial Registry.Results1) The score of the Fatigue Scale-14 decreased significantly in the CFS patients, and the scores of the PSQI and SF-36 changed significantly both in CFS patients and healthy controls. 2) Sixty FCs were considered significant to discriminate CFS (P = 0.000, best accuracy 90%), with 80.5% ± 9% average accuracy. 3) The FCs that were majorly related to the left frontoparietal network (FPN) and default mode network (DMN) significantly increased (P = 0.0032 and P = 0.001) in CFS patients after Tai Chi exercise. 4) The change of FCs in the left FPN and DMN were positively correlated (r = 0.40, P = 0.012).ConclusionThese results demonstrated that the 60 FCs we found using machine learning could be neural biomarkers to discriminate between CFS patients and healthy controls. Tai Chi exercise may improve CFS patients' fatigue syndrome, sleep quality, and body health statement by strengthening the functional connectivity of the left FPN and DMN under these FCs. The findings promote our understanding of Tai Chi exercise's value in treating CFS.

Although hematopoietic stem cell transplantation (HCT) is the only curative treatment for acute myeloid leukemia (AML), it is associated with significant treatment related morbidity and mortality. There is great need for predictive biomarkers associated with overall survival (OS) and clinical outcomes. We hypothesized that circulating metabolic, inflammatory, and immune molecules have potential as predictive biomarkers for AML patients who receive HCT treatment. This retrospective study was designed with an exploratory approach to comprehensively characterize immune, inflammatory, and metabolomic biomarkers. We identified patients with AML who underwent HCT and had existing baseline plasma samples. Using those samples (n = 34), we studied 65 blood based metabolomic and 61 immune/inflammatory related biomarkers, comparing patients with either long-term OS (≥ 3 years) or short-term OS (OS ≤ 1 years). We also compared the immune/inflammatory response and metabolomic biomarkers in younger vs. older AML patients (≤30 years vs. ≥ 55 years old). In addition, the biomarker profiles were analyzed for their association with clinical outcomes, namely OS, chronic graft versus host disease (cGVHD), acute graft versus host disease (aGVHD), infection and relapse. Several baseline biomarkers were elevated in older versus younger patients, and baseline levels were lower for three markers (IL13, SAA, CRP) in patients with OS ≥ 3 years. We also identified immune/inflammatory response markers associated with aGVHD (IL-9, Eotaxin-3), cGVHD (Flt-1), infection (D-dimer), or relapse (IL-17D, bFGF, Eotaxin-3). Evaluation of metabolic markers demonstrated higher baseline levels of medium- and long-chain acylcarnitines (AC) in older patients, association with aGVHD (lactate, long-chain AC), and cGVHD (medium-chain AC). These differentially expressed profiles merit further evaluation as predictive biomarkers.

IntroductionIn order to study the role of the microbiome in hematopoietic stem cell transplantation (HCT), researchers collect stool samples from patients at various time points throughout HCT. However, stool collection requires active subject participation and may be limited by patient reluctance to handling stool.MethodsWe performed a prospective study on the impact of financial incentives on stool collection rates. The intervention group consisted of allogeneic HCT patients from 05/2017-05/2018 who were compensated with a $10 gas gift card for each stool sample. The intervention group was compared to a historical control group of allogeneic HCT patients from 11/2016-05/2017 who provided stool samples before the incentive was implemented. To control for possible changes in collections over time, we also compared a contemporaneous control group of autologous HCT patients from 05/2017-05/2018 with a historical control group of autologous HCT patients from 11/2016-05/2017; neither autologous HCT group was compensated. The collection rate was defined as the number of samples provided divided by the number of time points we attempted to obtain stool.ResultsThere were 35 allogeneic HCT patients in the intervention group, 19 allogeneic HCT patients in the historical control group, 142 autologous HCT patients in the contemporaneous control group (that did not receive a financial incentive), and 75 autologous HCT patients in the historical control group. Allogeneic HCT patients in the intervention group had significantly higher average overall collection rates when compared to the historical control group allogeneic HCT patients (80% vs 37%, pConclusionOur results demonstrate that a modest incentive can significantly increase collection rates. These results may help to inform the design of future studies involving stool collection.

Activation of NOD-like receptor (NLR) signaling pathway can promote downstream cytokine and proinflammatory cytokines release, and inflammation induced by excess nutrients leads to renal metabolic injury. How the NLRs influence metabolic progress and then lead to the renal injury remains poorly investigated. Compared with rodents, minipigs are more similar to humans and are more ideal animal models for human disease research. In this study, we established a diabetic minipig model through a high-sugar and high-fat diet combined with streptozotocin (STZ) injection. Blood biological markers and renal pathological markers, expression of NLRP subfamily members (NLRP1 and NLRP3) and their downstream cytokines (precursors of IL-1β and IL-18 and mature forms of IL-1β and IL-18), expression of NLRC subfamily members (NLRC1, NLRC2, and NLRC5) and their downstream nuclear factor-κB (NF-κB) signaling pathway molecules (IKKβ, IκBα, and NF-κB p65), and inflammatory cytokines (TNF-α and interleukin-6 (IL-6)) were systematically evaluated. The expression of NLRP3 and its downstream cytokine signaling molecules, the precursors of IL-1β and IL-18, and the mature forms of IL-1β and IL-18 was significantly upregulated. The expression levels of NLRC1, NLRC2, and NLRC5 and activation of the downstream NF-κB pathway molecules phospho-IKKβ, phospho-IκBα, NF-κB p65, and phospho-NF-κB p65 were significantly increased. The TNF-α and IL-6 levels were significantly increased in diabetic pig kidneys. The TGF-β/Smad signaling molecules, TGF-β and P-SMAD2/3, were also increased. These results suggested that the metabolic inflammation activated by NLRs might play an important role in diabetic renal injuries.

High-purity titanium has been subjected to dynamic compression with a strain rate of 103 s−1 to activate {112-2} and {112-4} contraction twins. Electron backscatter diffraction (EBSD), transmission electron microscopy (TEM), and high-resolution TEM (HRTEM) were performed to observe the morphologies and twin boundaries of the contraction twins. The results show that {112-2} twins are the predominant twinning mode, as well as the formation of {112-4} twins due to the change in local stress state at the intersection region of {112-2} twin variants or {112-2} twin and grain boundary. The TEM and HRTEM observations reveal that (0001)‖(1122-) facets and (0001)‖(1121-) facets formed along the {112-2} and {112-4} twin boundaries, respectively. According to the theory of interfacial defects, the propagation of the {112-2} twin boundary was discussed with (b3, 3h{112-2}) and (b1, h{112-2}) twinning disconnections, as well as the growth process of the {112-4} twin boundary.

In this study, we investigated the phonon blockade effect in a parametrically driven and dissipative acoustic cavity at finite temperature. With the approximated analytical results based on the steady-state density-matrix master equation, we found that a quantum-interference-induced phonon blockade exists at finite temperature. We found a crossover between the quantum and thermal regimes on the curve of the second-order correlation function of the acoustic mode as the temperature increases. This phenomenon implies an asymmetry about the quantum and classic regimes. We also numerically simulated the single-phonon emission using the Monte Carlo wave function method. The results showed that a wide and deep dip around the zero time delay exists on the curve of the time-delayed second-order correlation function, which implies the possibility of observing a strong phonon blockade with pulse driving. Our study outlines a potential candidate for a efficient single-phonon source and applications in quantum information and phononic quantum networks.

The contribution percentage of inherent soil productivity (CPISP) refers to the ratio of crop yields under no-fertilization versus under conventional fertilization with the same field management. CPISP is a comprehensive measure of soil fertility. This study used 1 086 on-farm trials (from 1984–2013) and 27 long-term field experiments (from 1979–2013) to quantify changes in CPISP. Here, we present CPISP3 values, which reflect the CPISP states during the first three years after site establishment, for a series of sites at different locations in China collected in 1984–1990 (the 1980s), 1996–2000 (the 1990s), and 2004–2013 (the 2000s). The results showed that the average CPISP3 value for three crops (wheat, rice, and maize) was 53.8%. Historically, the CPISP3 in the 1990s (57.5%) was much higher than those in the 1980s (50.3%), and the 2000s (52.0%) (P≤0.05). Long-term no-fertilization caused CPISP levels to gradually decline and then stabilize; for example, in a mono-cropping system with irrigation, the CPISP values in Northwest and Northeast China declined by 4.5 and 4.0%, respectively, each year for the first ten years, but subsequently, the CPISP values stabilized. In contrast, the CPISP for upland crops in double-cropping systems continued to decrease at a rate of 1.1% per year. The CPISP for upland-paddy cropping decreased very slowly (0.07% per year), whereas the CPISP for paddy cropping decreased sharply (3.1% per year, on average) for the first two years and then remained steady during the following years. Therefore, upland crops in double-cropping systems consume the most inherent soil productivity, whereas paddy fields are favourable for maintaining a high level of CPISP. Overall, our results demonstrate a need to further improve China's CPISP3 values to meet growing productivity demands.

Abstract Background The ideal bearing surface for patients of avascular necrosis (AVN) undergoing total hip arthroplasty (THA) remains controversial. The purpose of this study is to evaluate the clinical outcomes, health-related quality of life (HRQL), and wear of the bearing surface between ceramic-on-ceramic (CoC) and ceramic-on-highly cross-linked polyethylene (CoXPE) THA for patients of AVN after midterm follow-up. Methods We performed a retrospective case-control analysis of 93 CoC and 77 CoXPE consecutive THAs for patients of AVN. The cases were followed at a minimum 5 years follow-up (average 7 years). Harris hip score (HHS) score and bearing-related complications were assessed. The health-related quality of life (HRQL) was assessed with the Short Form 36 (SF-36). Plain radiographs and computed tomography (CT) were used for radiographic evaluation. Results Both the CoC group and CoXPE group showed statistically significant improvements in HHS scores with no difference between the two bearing surfaces. There was no significant difference as for SF-36 at the latest follow-up between two groups, except for significant higher scores in the dimensions of general health in the CoC group (75.7 vs 64.7, P = 0.032). No radiographic evidence of osteolysis and loosening was present at the final follow-up. The mean wear rate of the CoC was 0.0096 mm/year and the CoXPE was 0.047 mm/year after evaluation with reconstructed CT. Conclusions CoC THAs acts as well as CoXPE THAs for patients with femoral head avascular necrosis after midterm follow-up. CoC bearing can significantly decrease the wearing rate than CoXPE bearing.

Aiming at the enhancement of the lightweight formability potential of aluminum alloy, the bulging and tensile properties of a 5052 Aluminum alloy sheet were tested on a microcomputer controlled sheet metal forming tester and tensile testing machine. The effects of different blank holder force, punch velocity and lubrication conditions were investigated on bulging properties by the experimental analysis. The cupping values (Erichsen Cupping Index: IE) of sheets with a thickness of 1.2 mm at room temperature were obtained under different process parameters. Meanwhile, the anisotropic property of the material was analyzed in different rolling directions. The results show that the sheet cupping values increase with the increase of blank holder force and punch velocity, and the stress state was changed due to the changing of the blank holder force and strain rate. Moreover, the use of lubricating conditions with a lower coefficient of friction allows the sheet to exhibit a larger cupping value. The effect of rolling direction on the anisotropy of 5052 aluminum alloy sheet is distinct, which means in the aluminum alloy sheet forming process the anisotropy factor should be carefully considered.

The ability of spoof surface plasmon polaritons (SSPPs) to confine electromagnetic fields in a subwavelength regime enables the design of miniaturized antennas. However, the impedance matching scheme for miniaturized spoof plasmonic antennas has not been studied systematically. In this paper, we propose a general method in the antenna design based on SSPPs, providing a feasible solution to impedance matching at the feeding point of miniaturized spoof plasmonic antennas. To verify the method, a prototype of a planar spoof plasmonic dipole antenna is simulated, fabricated and measured, of which the dipole arm length is reduced by 35.2% as compared with the traditional dipole antenna. A peak gain level of 4.29 dBi and the radiation efficiency of about 94.5% were measured at 6 GHz. This general method can be extended to solve the impedance matching problem in the design of other spoof plasmonic devices.

Background: Breast cancer is the most common cancer in women and the leading cause of female cancer deaths worldwide. Obesity causes chronic inflammation and is a risk factor for post-menopausal breast cancer and poor prognosis. Obesity triggers increased infiltration of macrophages into adipose tissue, yet little research has focused on the effects of macrophages in early stages of breast tumor development in obese patients. In this study, the effects of pro-inflammatory macrophages on breast cancer–adipocyte crosstalk were investigated. Methods: An innovative human cell co-culture system was built and used to model the paracrine interactions among adipocytes, macrophages, and breast cancer cells and how they facilitate tumor progression. The effects on cancer cells were examined using cell counts and migration assays. Quantitative reverse-transcription polymerase chain reaction was used to measure the expression levels of several cytokines and proteases to analyze adipocyte cancer association. Results: Macrophage-conditioned media intensified the effects of breast cancer–adipocyte crosstalk. Adipocytes became delipidated and increased production of pro-inflammatory cytokines, even in the absence of cancer cells, although the expression levels were highest with all three cell components. As a result, co-cultured breast cancer cells became more aggressive, with increased proliferation and migration compared to adipocyte–breast cancer co-cultures treated with unconditioned media. Conclusions: A novel co-culture model was built to evaluate the crosstalk among human macrophages, adipocytes, and breast cancer cells. We found that macrophages may contribute to adipocyte inflammation and cancer association and thus promote breast cancer progression.

The investigation for etiology of ischemic stroke in young adults remains a diagnostic challenge. Hyoid bone–related carotid injury is a rare and under-recognized cause of ischemic stroke, without established guidelines. We describe a case of recurrent ischemic stroke in a young patient presumably attributed to an impingement of the carotid artery by an elongated hyoid bone, and present other cases reported in the literature. Based on the imaging study as well as the lack of other findings, we attributed recurrent neurovascular events to the repetitive mechanical stimulation by the elongated hyoid bone that caused a vessel wall injury with subsequent thrombus and embolus. Given repeated recurrence under antiplatelet treatment, anticoagulation was added. The following 2-year follow-up showed no new neurologic events or any other complaints. Among the young, a broad spectrum of possibilities should be considered and we call attention to this infrequent etiology of ischemic stroke.

A 57-year-old woman was diagnosed with IDH-wildtype (IDHwt) astrocytoma (World Health Organization grade II) with the molecular characteristics of glioblastoma. She underwent concurrent radiotherapy and chemotherapy according to the Stupp protocol in combination with a multi-target antiangiogenic drug and additional intrathecal chemotherapy using methotrexate. During treatment, the patient’s tumor showed rapid progression. The chemotherapy with temozolomide was stopped and replaced with radiotherapy combined with tumor treating fields (TTF), the poly (adenosine diphosphate-ribose) polymerase (PARP) inhibitor niraparib, and anlotinib. After the radiotherapy was completed, the symptoms of increased intracranial pressure and epilepsy were well controlled. Considering the patient’s tolerance to the treatment, the combined therapy of TTF and anlotinib was continued, and osimertinib, a third-generation epidermal growth factor receptor–tyrosine kinase inhibitor with good permeability of the blood–brain barrier, was added. The patient was regularly followed up and had no obvious adverse drug reactions. Head magnetic resonance imaging (plain scan + enhanced scan) suggested that the lesions were stable. For rapidly progressing glioblastomas or histological grade II/III IDHwt astrocytomas, the combination of TTF and a PARP inhibitor during radiotherapy may have a synergistic effect on tumor control and is well tolerated by patients.

For direct formic acid fuel cells (DFAFC), palladium (Pd)-based alloy catalysts with competitive morphology and elemental composition are essential to boost the performance of the formic acid oxidation reaction (FAOR) in the anode zone. Herein, we design and synthesize RuPdx alloy nano-network structures (ANs) via the facile wet-chemical reduction of Pd-Ru cyanogel (Pdx [Ru(CN)6]y·aH2O) as an effective electrocatalyst for the FAOR. The formation of Pd-Ru cyanogel depends on the facile coordination of K2PdCl4 and K3 [Ru(CN)6]. The unique structure of cyanogel ensures the presentation of a three-dimensional mesoporous morphology and the homogeneity of the elemental components. The as-prepared RuPd3 ANs exhibit good electrocatalytic activity and stability for the FAOR. Notably, the RuPd3 ANs achieve a mass-specific activity of 2068.4 mA mg−1 in FAOR, which shows an improvement of approximately 16.9 times compared to Pd black. Such a competitive FAOR performance of RuPd3 ANs can be attributed to the advantages of structure and composition, which facilitate the exposure of more active sites, accelerate mass/electron transfer rates, and promote gas escape from the catalyst layer, as well as enhance chemical stability.

Soft materials such as biological tissues are prone to deformation and generate different stable structures under external stimulation. This property is widely used to create tunable patterns, and the tuning of the wrinkling patterns can be applied to the control of elastic waves. In this paper, the wrinkling modes of film/substrate systems with different geometric dimensions and material parameters were studied. It is verified by numerical simulation that the elastic wave band gaps corresponding to the two wrinkling modes can be effectively superposed in one system, and the experimental samples with two wrinkling modes coexisting in one system were prepared by parameter optimization and a moisture-curing process. A vibration test showed that the hybrid system could effectively suppress the propagation of elastic waves. Combined with engineering needs, the wrinkling system under different loading conditions was studied, which provides a design guide for widening and regulating the elastic wave band gap.

The number ratio of carbon-rich to oxygen-rich asymptotic giant branch (AGB) stars (the so-called C/M ratio) is closely related to the evolution environment of the host galaxy. This work studies the C/M ratio in 14 galaxies within the Local Group with the most complete and clean sample of member stars identified in our previous works. The borderlines between carbon-rich AGB and oxygen-rich AGB stars as well as red supergiants are defined by Gaussian mixture model fitting to the number density in the (J−K)/K diagram for the member stars of the LMC and M33, and then applied to the other galaxies by shifting the difference in the position of tip red giant branch (TRGB). The C/M ratios are obtained after precise and consistent categorization. Although for galaxies with larger distance modulo there is greater uncertainty, the C/M ratio is clearly found to decrease with the color index (J−K)0 of TRGB as the indicator of metallicity, which agrees with previous studies and can be explained by the fact that carbon stars are more easily formed in a metal-poor environment. Furthermore, the C/M ratio within M33 is found to increase with galactocentric distance, which coincides with this scenario and the galactic chemical evolution model. On the other hand, the C/M ratio within M31 is found to decrease with galactocentric radius, which deserves further study.

Root restriction is a physical and ecological cultivation mode which restricts plant roots into a limited container to regulate vegetative and reproduction growth by reshaping root architecture. However, little is known about related molecular mechanisms. To uncover the root-related regulatory network of endogenous RNAs under root restriction cultivation (referred to RR), transcriptome-wide analyses of mRNAs, long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs) involved in root development were performed. During root development, RR treatment had a positive effect on root weight, typically, young roots were significantly higher than conventional cultivation (referred to NR) treatment, suggesting that root architecture reconstruction under RR was attributed to the vigorous induction into lateral roots. Furthermore, a total of 26,588 mRNAs, 1971 lncRNAs, and 2615 circRNAs were identified in root of annual “Muscat Hamburg” grapevine by the transcriptomic analyses. The expression profile of mRNAs, lncRNAs and circRNA were further confirmed by the quantitative real-time PCR (RT-qPCR). Gene ontology enrichment analysis showed that a majority of the differentially expressed mRNAs, lncRNAs and circRNAs were enriched into the categories of cellular process, metabolic process, cell part, binding, and catalytic activity. In addition, the regulatory network of endogenous RNAs was then constructed by the prediction of lncRNA-miRNA-mRNA and circRNA-miRNA-mRNA network, implying that these RNAs play significant regulatory roles for root architecture shaping in response to root restriction. Our results, for the first time, the regulatory network of competitive endogenous RNAs (ceRNAs) functions of lncRNA and circRNA was integrated, and a basis for studying the potential functions of non-coding RNAs (ncRNAs) during root development of grapevine was provided.

Central nervous system (CNS) trauma activates a persistent repair response that leads to fibrotic scar formation within the lesion. This scarring is similar to other organ fibrosis in many ways; however, the unique features of the CNS differentiate it from other organs. In this review, we discuss fibrotic scar formation in CNS trauma, including the cellular origins of fibroblasts, the mechanism of fibrotic scar formation following an injury, as well as the implication of the fibrotic scar in CNS tissue remodeling and regeneration. While discussing the shared features of CNS fibrotic scar and fibrosis outside the CNS, we highlight their differences and discuss therapeutic targets that may enhance regeneration in the CNS.

With the rapid development of high-resolution synthetic aperture radar (SAR) systems, the technique that utilizes multiple two-dimensional (2-D) SAR images with different view angles to extract three-dimensional (3-D) coordinates of targets has gained wide concern in recent years. Unlike the traditional multi-channel SAR utilized for 3-D coordinate extraction, the single-channel curvilinear SAR (CLSAR) has the advantages of large variation of view angle, requiring fewer acquisition data, and lower device cost. However, due to the complex aerodynamic configuration and flight characteristics, important issues should be considered, including the mathematical model establishment, imaging geometry analysis, and high-precision extraction model design. In this paper, to address these challenges, a 3-D vector model of CLSAR was presented and the imaging geometries under different view angles were analyzed. Then, a novel 3-D coordinate extraction approach based on radargrammetry was proposed, in which the unique property of the SAR system, called cylindrical symmetry, was utilized to establish a novel extraction model. Compared with the conventional approach, the proposed one has fewer constraints on the trajectory of radar platform, requires fewer model parameters, and can obtain higher extraction accuracy without the assistance of extra ground control points (GCPs). Numerical results using simulated data demonstrated the effectiveness of the proposed approach.

Online state-of-charge (SOC) estimation for lithium-ion batteries is one of the most important tasks of the battery management system in ensuring its operation safety and reliability. Due to the advantages of learning the long-term dependencies in between the sequential data, recurrent neural networks (RNNs) have been developed and have shown their superiority over SOC estimation. However, only time-series measurements (e.g., voltage and current) are taken as inputs in these RNNs. Considering that the mapping relationship between the SOC and the time-series measurements evolves along with the battery degradation, there still remains a challenge for RNNs to estimate the SOC accurately throughout the battery’s lifetime. In this paper, a dual-input neural network combining gated recurring unit (GRU) layers and fully connected layers (acronymized as a DIGF network) is developed to overcome the above-mentioned challenge. Its most important characteristic is the adoption of the state of health (SOH) of the battery as the network input, in addition to time-series measurements. According to the experimental data from a batch of LiCoO2 batteries, it is validated that the proposed DIGF network is capable of providing more accurate SOC estimations throughout the battery’s lifetime compared to the existing RNN counterparts. Moreover, it also shows greater robustness against different initial SOCs, making it more applicable for online SOC estimations in practical situations. Based on these verification results, it is concluded that the proposed DIGF network is feasible for estimating the battery’s SOC accurately throughout the battery’s lifetime against varying initial SOCs.

BackgroundDrug-resistance is a major obstacle to the treatment of breast cancer. Circular RNA (circRNA) circ-MMP11 has been reported to be promoting the progression of breast cancer. This study is designed to explore the role and mechanism of circ-MMP11 in lapatinib resistance in breast cancer.MethodsCirc-MMP11, microRNA-153-3p (miR-153-3p), and Anillin (ANLN) levels were detected by real-time quantitative polymerase chain reaction (RT-qPCR). Cell viability, number of colonies, apoptosis, migration, and invasion were detected by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT), colony formation, flow cytometry, and transwell assays, respectively. Exosomes were exerted and detected by differential centrifugation and a transmission electron microscope. The protein levels of CD63, CD9, and ANLN were assessed by western blot assay. The binding relationship between miR-153-3p and circ-MMP11 or ANLN was predicted by circinteractome or starbase, and then verified by a dual-luciferase reporter assay and RNA pull-down assay. The biological role of circ-MMP11 on breast cancer tumor growth and drug resistance was detected by the xenograft tumor model in vivo.ResultsCirc-MMP11 and ANLN were highly expressed, and miR-153-3p was decreased in LR breast cancer tissues and cells. Circ-MMP11 could be transported by exosomes. Furthermore, circ-MMP11 knockdown promoted lapatinib sensitivity by repressing cell viability, colony number, migration, invasion, and boosting apoptosis in LR breast cancer cells. Circ-MMP11 deficiency improved the drug sensitivity of breast cancer in vivo. Mechanically, circ-MMP11 could regulate ANLN expression through sponging miR-153-3p.ConclusionCirc-MMP11 could be transferred by exosomes in breast cancer cells. And circ-MMP11 functioned as a sponge of miR-153-3p to regulate ANLN expression, thereby promoting lapatinib resistance in breast cancer cells, providing therapeutic targets for the treatment of breast cancer.

The problems of data abnormalities and missing data are puzzling the traditional multi-modal heterogeneous big data clustering. In order to solve this issue, a multi-view heterogeneous big data clustering algorithm based on improved Kmeans clustering is established in this paper. At first, for the big data which involve heterogeneous data, based on multi view data analyzing, we propose an advanced Kmeans algorithm on the base of multi view heterogeneous system to determine the similarity detection metrics. Then, a BP neural network method is used to predict the missing attribute values, complete the missing data and restore the big data structure in heterogeneous state. Last, we ulteriorly propose a data denoising algorithm to denoise the abnormal data. Based on the above methods, we construct a framework namely BPK-means to resolve the problems of data abnormalities and missing data. Our solution approach is evaluated through rigorous performance evaluation study. Compared with the original algorithm, both theoretical verification and experimental results show that the accuracy of the proposed method is greatly improved.

Nucleotide oligomerization domain-like receptors (NLRs), a class of pattern recognition receptors, participate in the host’s first line of defense against invading pathogenic microorganisms. NLR family caspase recruitment domain containing 5 (NLRC5) is the largest member of the NLR family and has been shown to play an important role in inflammatory processes, angiogenesis, immunity, and apoptosis by regulating the nuclear factor-κB, type I interferon, and inflammasome signaling pathways, as well as the expression of major histocompatibility complex I genes. Recent studies have found that NLRC5 is also associated with neuronal development and central nervous system (CNS) diseases, such as CNS infection, cerebral ischemia/reperfusion injury, glioma, multiple sclerosis, and epilepsy. This review summarizes the research progress in the structure, expression, and biological characteristics of NLRC5 and its relationship with the CNS.

The role of microbial β-diversity in soil ecosystem function is not well-studied. Here, the authors use genetic data to show that microbial α-diversity levels may have impacts on stochastic/deterministic assembly processes and functions of soil microbiome.

Objective: Salvanic Acid B (Sal B) was proved to show significant effect against fibrosis and myocardial injury. This study aimed to investigate the protective effects and the mechanisms of Sal B on myocardial fibrosis. Methods: The mice were randomly assigned to five groups: control group, model group, positive group, low-dose group, high-dose group. Hematoxylin-Eosin (HE) staining and Masson staining were used to assess the myocardial physiological changes and measure the myocardial fibrosis area. Expression of transforming growth factor-beta (TGF-β), drosophila mothers against decapentaplegic (Smad)2, Smad3 and Smad7 were analyzed by immunohistochemistry and real-time PCR. On the other hand, mouse cardiac fibroblasts (CFs) cells were co-treated with 20 ng/mL TGF-β1 and different concentrations of Sal B (5, 10, and 20 ng/mL) for 24 h. The cells morphology changes were assessed under a microscope, and the protein expressions induced by TGF-β1 were detected by Western blot. The joint planning of spare parts and maintenance workers in a multiple echelon inventory system with lateral and cross-echelon transshipment can reduce the total cost of the system. However, partial pooling is not extensively considered in the joint optimization problem of maintenance resources. In this paper, the objective is to determine the optimal inventory of spare parts and the number of maintenance workers to minimize the total cost of the system considering partial pooling. First, a greedy heuristic is used to obtain the initial inventory. Then, the cat swarm optimization algorithm is formulated to produce nearly optimal results, which can solve larger instances with a faster computation time. Furthermore, a maintenance system with 4 local warehouses, 3 central warehouses and 1 plant serving 5 machine groups is analyzed, in which each machine consists of 5 key components that can breakdown independently. The results verify the effectiveness of the proposed optimization approach. Finally, the total cost for different resource provision scenarios, failure rates and system parameters are discussed.

Space solar cell components always suffered from the complex and hostile space environment. The reliability of space solar cell components is important to ensure the mission success of space satellites. However, the current research mostly focuses on solar cells' degradation mechanisms. There is less research based on the performance parameters of space solar cell component's degradation to analyze its reliability. Therefore, a reliability analysis method based on the method of modeling the performance degradation of space solar cell components is proposed. By combining general stress-strength interference model and Gamma process, the reliability model of space solar cell components is established. The accelerated degradation test based on the equivalent particle dose is performed to simulate the radiation influence and verify the proposed method. Compared with the displacement damage dose method, the proposed method can further develop the reliability analysis based on performance degradation analysis. Moreover, the cover glass and interconnection are also taken into account together to form a completed component analysis. Through analyzing the cover glass and interconnection experiment results, the cover glass can affect the component reliability; whereas, the interconnection has little influence at the particle radiation condition. Based on the proposed method, the reliability is easier to calculate with the degradation of output power.

In this paper, linearity improved Doherty power amplifier (DPA) is proposed for linearity improvement by modifying the input capacitor value of peaking amplifier. The non-foster circuits are substituted for the conventional λ/4 transmission line in front of peaking amplifier, which the 90° phase shifter and impedance matching can be performed by the proposed structure. Furthermore, the input capacitor value of peaking amplifier FET can be modified through modifying bias voltage and match resistance of non-foster circuits. Therefore, the AM-PM distortion of peak amplifier is suppressed, and the linearity of total DPA is improved. Compared to a conventional DPA, the third-order inter-modulation distortion of proposed DPA is reduced by 12 dB without cutting efficiency.

A complex human-machine system (CHMS) consists of heterogeneous components with extensive human-machine interactions. CHMSs are typical multistate systems with the ability to adapt to disturbances such as machine failures. These characteristics must be considered comprehensively to accurately evaluate the reliability and performance of a CHMS. However, the existing literature scarcely considers both the reliability and performance simultaneously. In this paper, we propose an agent-based approach to model and evaluate a CHMS. First, a general agent-based modeling framework for a CHMS is generated by analyzing the structure and operations of a CHMS. Then, a dual-clock mechanism is introduced to describe the behaviors of the machine failures and human errors. Two environmental disturbance modeling methods are proposed based on the state transitions of the agent and random events. The methods to model the repair and reconfiguration behaviors are presented based on the contract network. A Monte Carlo-based method is developed to evaluate the reliability and performance of the CHMS simultaneously. Finally, a deck scheduling process for an aircraft carrier is used as a case study to verify the approach. The results show that the reliability and performance of a CHMS can be effectively evaluated.

Magnesium alloys play an important role in lightweight structures, which are extensively used in different industries due to their excellent mechanical and physical properties. In this paper, the formability of the AZ31B magnesium alloy sheet was studied by using tensile tests at different temperatures (from 25 to 250 °C) and strain rates (from 0.017 s−1 to 0.34 s−1). The results showed that the material behaves with positive temperature sensitivity when forming at a temperature lower than 200 °C. The effect of the strain rates on the formability of AZ31B was larger at high temperatures. The metallography of AZ31B at different temperatures and strain rates was observed by OM. The results showed that the partially recrystallized structure was exhibited at a temperature of 150 °C. With the increase in temperature, the approximate complete recrystallization was exhibited at a temperature of 250 °C. The fracture morphology of AZ31B was observed at different strain rates and temperatures by SEM. Additionally, the main fracture pattern was quasi-cleavage at room temperature. However, with the increase in temperature, the fracture pattern was transited from a quasi-cleavage pattern to a ductile fracture pattern. The ductile fracture pattern was the main fracture pattern at a temperature of 250 °C.

In this paper, a multi-functional soft robot module that can be used to constitute a variety of soft robots is proposed. The body of the soft robot module made of rubber is in the shape of a long strip, with cylindrical chambers at both the top end and bottom end of the module for the function of actuators and sensors. The soft robot module is driven by supercoiled polymer artificial muscle (SCPAM) strings, which are made from conductive nylon sewing threads. Artificial muscle strings are embedded in the chambers of the module to control its deformation. In addition, SCPAM strings are also used for the robot module’s sensing based on the linear relationship between the string’s length and their resistance. The bending deformation of the robot is measured by the continuous change of the sensor’s resistance during the deformation of the module. Prototypes of an inchworm-like crawling robot and a soft robotic gripper are made, whose crawling ability and grasping ability are tested, respectively. We envision that the proposed proprioceptive soft robot module could potentially be used in other robotic applications, such as continuum robotic arm or underwater robot.

For unmanned aerial vehicle (UAV) synthetic aperture radar (SAR) imaging, motion errors cannot be obtained accurately when high precision motion sensors are not equipped on the platform. This means that traditional data-based motion compensation (MOCO) cannot be directly implemented due to large measurement errors. In addition, classic autofocusing techniques, such as phase gradient autofocus (PGA) or map-drift algorithm (MDA), do not perform well with spatially variant errors, greatly affecting the imaging qualities, especially for high-resolution and large-swath cases. In this study, an improved spatially variant MOCO approach based on an MDA is developed to effectively eliminate the spatially variant errors. Based on the coarse and precise MDA chirp rate error estimation, motion errors are optimally acquired by the random sample consensus (RANSAC) iteration. Two-dimensional (2D) mapping is used to decouple the spatially variant residual errors into two linear independent dimensions so that the chirp-z transform (CZT) can be performed for echo data correction. Unlike traditional approaches, the spatially variant components can be compensated without any measured motion information, which indicates that the proposed approach can be applied to the common UAV SAR system with significant measurement errors. Simulations and real data experiments were used to evaluate the performance of the proposed method. The simulation results show that the proposed algorithm is able to effectively minimize spatially variant errors and generate much better imaging results.

The research into curvilinear flight synthetic aperture radar (CF-SAR) is the inevitable result of the comprehensive practicality of SAR. The flight path of the SAR platform in real applications, which is highly nonlinear or curvy due to three-dimensional velocity and acceleration, cannot be described by the traditional uniform linear motion model. New mathematical models, signal characteristics, imaging algorithms, and system design criteria must be proposed and investigated for CF-SAR. This paper provides a comprehensive overview of CF-SAR. Firstly, the basic concept, unified model, and general signal characteristics of CF-SAR are defined, derived, and analyzed, respectively. Additionally, the advantages and drawbacks of current methodologies are reviewed. Discussions on the CF-SAR’s applications are presented from the perspective of typical platforms, new configurations, and advanced technologies, which are suitable means to fulfill the increasing user requirements. Finally, the challenges faced by CF-SAR are summarized, and some future trends for the study of CF-SAR are explored. Hopefully, this paper will serve as a reference for SAR researchers/engineers and stimulate the future development and actual application of CF-SAR.

With a new generation of bike sharing services emerging, the development of dockless bike sharing services results in considerable socioeconomic and environmental benefits but also creates new issues, such as inappropriate parking behaviors and bike imbalances. To solve the inappropriate parking problem, electric fences have been introduced to guide users to park bikes in designated zones. Considering the role of electric fences in restricting user parking behaviors, an electric fence-based intelligent scheduling method for rebalancing dockless bike sharing systems is proposed in this paper. As a dynamic method that considers the real-time usage of bike sharing systems, an electric fence adjusts its capacity based on real-time information, which guides users to return bikes to electric fences with greater urgency. Because existing approaches require prespecified models and are unable to consider all the intricacies in the dynamic optimization problem, a model-free intelligent scheduling approach based on deep Q-learning that can adapt to the changing distributions of customer arrivals, available bikes, bike locations, and user travel times is used to solve the problem. Finally, a case study involving Beihang University is employed, which shows that the method performs well in rebalancing the bike sharing system and improving the mean utilization (MU) and customer satisfaction (CS).

Cellulose has attracted considerable attention as the most promising potential candidate raw material for the production of bio-based polymeric materials. In the last decade, significant progress has been made in the production of biopolymers based on different cellulose forms. In this study, cellulose was obtained in an innovative and environmentally friendly way, using boxwood powder. Crude cellulose was obtained by treating Buxus powder with an ethanol–acetic acid–water mixture. Refined cellulose was then obtained by treatment with an acidic sodium hypochlorite solution and alkaline hydrogen peroxide solution. The novel chemistry of cellulose prepared by this method promises to be not only green, but also highly desirable, because of its lower emissions and low cost. It is crucial for the future of the global polymer industry. The refined cellulose was subjected to a high-temperature hydrothermal treatment under two temperatures and time conditions, with temperature gradients of 120, 140, and 160 °C, and time gradients of 1, 2, and 3 h. The samples were subjected to infrared and thermogravimetric analyses. The cellulose undergoes dehydration and thermal degradation reactions during the heat treatment process, and the thermal stability of the residual is enhanced, compared with that of virgin cellulose. Between 120 and 140 °C, the hydroxyl and hypomethyl groups on the surface of cellulose are shed. Groups in the amorphous region of the polymer are the first to be shed. The dehydration reaction reduces the number of free hydroxyl groups on the surface of the cellulose molecules. The dehydration reaction was accelerated by an increase in temperature. Between 140 and 160 °C, the β-(1,4)-glycosidic bond begins to slowly break and some furans are generated. The structure of cellulose undergoes reorganization during thermal treatment. The thermal stability of the modified material is greater than that of untreated cellulose.

Accurate life prediction and reliability evaluation of lithium-ion batteries are of great significance for predictive maintenance. In the whole life cycle of a battery, the accurate description of the dynamic and stochastic characteristics of life has always been a key problem. In this paper, the concept of the digital twin is introduced, and a digital twin for reliability based on remaining useful cycle life prediction is proposed for lithium-ion batteries. The capacity degradation model, stochastic degradation model, life prediction, and reliability evaluation model are established to describe the randomness of battery degradation and the dispersion of the life of multiple cells. Based on the Bayesian algorithm, an adaptive evolution method for the model of the digital twin is proposed to improve prediction accuracy, followed by experimental verification. Finally, the life prediction, reliability evaluation, and predictive maintenance of the battery based on the digital twin are implemented. The results show the digital twin for reliability has good accuracy in the whole life cycle. The error can be controlled at about 5% with the adaptive evolution algorithm. For battery L1 and L6 in this case, predictive maintenance costs are expected to decrease by 62.0% and 52.5%, respectively.

Current diagnostic standards for PJI rely on inflammatory markers that are typically elevated in autoimmune diseases, thus making the diagnosis of PJI in patients with rheumatoid arthritis and joint replacement particularly complicated. There is a paucity of data on differentiating PJI from rheumatoid arthritis in patients with previous arthroplasty. In this study, we retrospectively analyzed the cases of 17 patients with rheumatoid arthritis and 121 patients without rheumatoid disease who underwent surgical intervention due to microbiology-positive PJI of the hip or knee joint. We assessed clinical patient characteristics, laboratory parameters, and prosthesis survival rates in patients with and without rheumatoid arthritis and acute or chronic PJI. ROC analysis was conducted for the analyzed parameters. In patients with chronic PJI, peripheral blood CRP (p = 0.05, AUC = 0.71), synovial WBC count (p = 0.02, AUC = 0.78), synovial monocyte cell count (p = 0.04, AUC = 0.75), and synovial PMN cell count (p = 0.02, AUC = 0.80) were significantly elevated in patients with rheumatoid arthritis showing acceptable to excellent discrimination. All analyzed parameters showed no significant differences and poor discrimination for patients with acute PJI. Median prosthesis survival time was significantly shorter in patients with rheumatoid arthritis (p = 0.05). In conclusion, routinely used laboratory markers have limited utility in distinguishing acute PJI in rheumatoid patients. In cases with suspected chronic PJI but low levels of serum CRP and synovial cell markers, physicians should consider the possibility of activated autoimmune arthritis.

Lupus nephritis (LN) is a common complication of systemic lupus erythematosus (SLE) and a major risk factor for morbidity and mortality. The abundant cell-free nucleic (DNA/RNA) in SLE patients, especially dsDNA, is a key substance in the pathogenesis of SLE and LN. The deposition of DNA/RNA-immune complexes (DNA/RNA-ICs) in the glomerulus causes a series of inflammatory reactions that lead to resident renal cell disturbance and eventually renal fibrosis. Cell-free DNA/RNA is the most effective inducer of type I interferons (IFN-I). Resident renal cells (rather than infiltrating immune cells) are the main source of IFN-I in the kidney. IFN-I in turn damages resident renal cells. Not only are resident renal cells victims, but also participants in this immunity war. However, the mechanism for generation of IFN-I in resident renal cells and the pathological mechanism of IFN-I promoting renal fibrosis have not been fully elucidated. This paper reviews the latest epidemiology of LN and its development process, discusses the mechanism for generation of IFN-I in resident renal cells and the role of IFN-I in the pathogenesis of LN, and may open a new perspective for the treatment of LN.

Neutrophils are short-lived cells of the innate immune system and the first line of defense at the site of an infection and tissue injury. Pattern recognition receptors on neutrophils recognize pathogen-associated molecular patterns or danger-associated molecular patterns, which recruit them to the destined site. Neutrophils are professional phagocytes with efficient granular constituents that aid in the neutralization of pathogens. In addition to phagocytosis and degranulation, neutrophils are proficient in creating neutrophil extracellular traps (NETs) that immobilize pathogens to prevent their spread. Because of the cytotoxicity of the associated granular proteins within NETs, the microbes can be directly killed once immobilized by the NETs. The role of neutrophils in infection is well studied; however, there is less emphasis placed on the role of neutrophils in tissue injury, such as traumatic spinal cord injury. Upon the initial mechanical injury, the innate immune system is activated in response to the molecules produced by the resident cells of the injured spinal cord initiating the inflammatory cascade. This review provides an overview of the essential role of neutrophils and explores the contribution of neutrophils to the pathologic changes in the injured spinal cord.

Xuebijing (XBJ), the only drug approved for the sepsis and multiple organ dysfunction, and its protective effects against acute liver injury (ALI) and its mechanism. The aim of this study was to evaluate the protective effect of XBJ on cecal ligation and perforation (CLP)-induced mouse ALI model and LPS-induced RAW264.7 cell ALI model. Mice were pretreated with XBJ before the CLP model was established, and serum and liver tissues were collected at the end of the experiment to assess the levels of inflammatory factors and liver injury. Results showed that XBJ pretreatment reduced liver/body weight, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities in serum, and inhibited levels of pro-inflammatory factors in serum. Cells were treatment with XBJ and modeled by LPS modeling increased cell viability in the XBJ-treated group compared to the model group and XBJ also decreased serum pro-inflammatory factors in a dose-dependent manner. Western blot detected that XBJ also up-regulated the phosphorylated levels of glycogen synthase kinase-3β (p-GSK-3β) and cAMP-response element-binding protein (p-CREB) and down-regulated the phosphorylated level of nuclear factor kappa-B (p-NF-κB) in liver and cell. After overexpression of GSK-3β in cells, the mechanism was further investigated using CO-IP analysis. The binding of p-NF-κB and p-CREB to CREB-binding protein (CBP) was increased and decreased, respectively, indicating that GSK-3β regulated inflammation by regulating the binding of p-NF-κB and p-CREB to CBP. The present studies suggested that the hepatoprotective effect of XBJ may be through up-regulation of GSK-3β (Ser9) and increasing the binding of p-CREB to CBP, thereby alleviating the inflammatory response.

Although the increased expression of members of the chondroitin sulfate proteoglycan family, such as neuron-glial antigen 2 (NG2), have been well documented after an injury to the spinal cord, a complete picture as to the cellular origins and function of this NG2 expression has yet to be made. Using a spinal cord injury (SCI) mouse model, we describe that some infiltrated bone marrow-derived macrophages (BMDMΦ) are early contributors to NG2/CSPG4 expression and secretion after SCI. We demonstrate for the first time that a lesion-related form of cellular debris generated from damaged myelin sheaths can increase NG2/CSPG4 expression in BMDMΦ, which then exhibit enhanced proliferation and decreased phagocytic capacity. These results suggest that BMDMΦ may play a much more nuanced role in secondary spinal cord injury than previously thought, including acting as early contributors to the NG2 component of the glial scar.

Phyllodes tumor (PT) is a special type of breast tumors, including three types: malignant, borderline, and benign. Most of these tumors form unilateral disease and can rapidly increase in size. The occurrence of axillary lymph node metastasis is rare. Tumor-associated hypoglycemia can be divided into non-islet cell tumor and insulinoma. In non-islet cell tumor hypoglycemia (NICTH), a considerable high molecular weight form of insulin like growth factor 2 (IGF-2) is formed, which abnormally binds to insulin receptors in the tissues and causes hypoglycemia. Breast phyllodes tumors with NICTH are rare and first reported in 1983. Surgical resection is the main treatment and hypoglycemia symptoms usually resolve after surgery. Nevertheless, prior to surgery, intravenous glucose infusion is used to maintain blood glucose levels. A female patient presented with a rapidly growing breast mass and was diagnosed with a phyllodes tumor with NICTH at our hospital in August 2020; she was successfully treated through surgical resection. We reviewed the relevant literature to investigate and analyze the relationship between NICTH and phyllodes tumors, as well as optimize its diagnosis and treatment.

The evolutionarily conserved TRanscript-EXport (TREX) complex plays central roles during mRNP (messenger ribonucleoprotein) maturation and export from the nucleus to the cytoplasm. In yeast, TREX is composed of the THO sub-complex (Tho2, Hpr1, Tex1, Mft1, and Thp2), the DEAD box ATPase Sub2, and Yra1. Here we present a 3.7 Å cryo-EM structure of the yeast THO•Sub2 complex. The structure reveals the intimate assembly of THO revolving around its largest subunit Tho2. THO stabilizes a semi-open conformation of the Sub2 ATPase via interactions with Tho2. We show that THO interacts with the serine–arginine (SR)-like protein Gbp2 through both the RS domain and RRM domains of Gbp2. Cross-linking mass spectrometry analysis supports the extensive interactions between THO and Gbp2, further revealing that RRM domains of Gbp2 are in close proximity to the C-terminal domain of Tho2. We propose that THO serves as a landing pad to configure Gbp2 to facilitate its loading onto mRNP.

Peptides composed of a short chain of amino acids can play significant roles in plant growth, development, and stress responses. Most of these functional peptides are derived by either processing precursor proteins or direct translation of small open reading frames present in the genome and sometimes located in the untranslated region sequence of a messenger RNA. Generally, canonical peptides serve as local signal molecules mediating short- or long-distance intercellular communication. Also, they are commonly used as ligands perceived by an associated receptor, triggering cellular signaling transduction. In recent years, increasing pieces of evidence from studies in both plants and animals have revealed that peptides are also encoded by RNAs currently defined as non-coding RNAs (ncRNAs), including long ncRNAs, circular RNAs, and primary microRNAs. Primary microRNAs (miRNAs) have been reported to encode regulatory peptides in Arabidopsis, grapevine, soybean, and Medicago, called miRNA-encoded peptides (miPEPs). Remarkably, overexpression or exogenous applications of miPEPs specifically increase the expression level of their corresponding miRNAs by enhancing the transcription of the MIRNA (MIR) genes. Here, we first outline the current knowledge regarding the coding of putative ncRNAs. Notably, we review in detail the limited studies available regarding the translation of miPEPs and their relevant regulatory mechanisms. Furthermore, we discuss the potential cellular and molecular mechanisms in which miPEPs might be involved in plants and raise problems that needed to be solved.

Grafting cultivation is implemented worldwide mainly to resist abiotic and biotic stresses and is an effective method to improve watermelon production. However, grafting may affect fruit development and quality. In our experiment, pumpkin-grafted (PG) watermelon fruits developed slower and the ripening period was extended compared to self-grafted (SG) fruits. We found that the concentrations of abscisic acid (ABA) among endogenous phytohormones were dramatically reduced by pumpkin grafting. In order to understand these changes at the gene expression level, we performed a comprehensive analysis of the fruit flesh transcriptomes between PG and SG during fruit development and ripening. A total of 1,675 and 4,102 differentially expressed genes (DEGs) were identified between PG and SG. Further functional enrichment analysis revealed that these DEGs were associated with carbohydrate biosynthesis, phytohormone signaling transmission, and cell wall metabolism categories. ABA centric phytohormone signaling and fruit quality-related genes including ABA receptor, PP2C proteins, AP2-EREBP transcription factors, sucrose transporter, and carotenoid isomerase were co-expressed with fruit ripening. These results provide the valuable resource for understanding the mechanism of pumpkin grafting effect on watermelon fruit ripening and quality development.
