Objective A detailed analysis of the morphology of distal humeral articulation can help in the creation of anatomic prostheses of hemiarthroplasty. This study used statistical shape modeling to evaluate the 3D morphology of the distal humerus in healthy Chinese individuals and to investigate the proper articular morphology differences. Methods A statistical shape model (SSM) of the distal humerus was created using CT scans of 106 survey‐confirmed nonpathologic elbows. In addition, the articular components of each principal component (PC) were selected and fitted on the mean mode. The Euclidean point‐to‐mesh distance of articular modes was calculated as a measurement the proper change in the morphology of the articulation. Results The first seven PCs jointly accounted for 80.9% of the total variation (44.4%, 12.2%, 7.9%, 5.9%, 4.1%, 3.4% and 3%, respectively). In the mean model, the distance between the medial and lateral epicondyles was 57.4 mm, the width of the articulation was 42.1 mm, and the angle of the transepicondylar line (TEL) and C line was 4.8°. The articular surface differences of the first PC were significant (RMS: 1.43 mm in the −3 SD model and 2.38 mm in the +3 SD model), whereas under other conditions, the differences were not remarkable despite the maximum deformation not exceeding 1 mm. Conclusion A novel method (SSM) was used to evaluate the 3D morphology of the distal humerus in healthy Chinese individuals and investigate the proper articular shape differences. We found the proper shape of articular surface basically transformed into one variation pattern which was relevant to the bone size, even though the morphology of distal humerus possessed complicated variation modes. The findings of this study can be helpful to design the next generation of elbow hemiarthroplasty in the future.

Five species of torrenticolid mites (Acari, Hydrachnidia), collected in the Anzihe and Qingliangfeng national nature reserves, R. P. China, are identified. Three species are described as new to science: Torrenticola pseudosiamis Gu & Guo, sp. nov., T. anziensis Gu & Guo, sp. nov., and Monatractides sichuanensis Gu & Guo, sp. nov. The other two species, M. macrocorpis Gu & Guo, 2019, M. xiaoxiensis Gu & Guo, 2019, are newly reported from Zhejiang Province. Descriptions and illustrations of these species are included. Abstract Background Gastrointestinal stromal tumor (GIST) is a rare type of cancer that occurs in the gastrointestinal tract. The majority of GIST cases carry oncogenic forms of KIT, the receptor for stem cell factor (SCF). Small molecule kinase inhibitor imatinib is effective in prolonging the survival of GIST patients by targeting KIT. However, drug resistance often develops during the therapeutic treatment. Here, we produced a SCF-emtansine drug conjugate (SCF-DM1) with favorable drug efficacy towards GIST cells. Methods Recombinant human SCF (rhSCF) was expressed in E. coli cells and further purified with Ni–NTA Sepharose and Phenyl Sepharose. It was then conjugated with DM1, and the conjugated product SCF-DM1 was evaluated using in vitro cell-based assays and in vivo xenograft mouse model. Background Antimicrobial resistance is a global health threat. Antibiotics are commonly prescribed for children with uncomplicated lower respiratory tract infections, but there is little randomised evidence to support the effectiveness of antibiotics in treating these infections, either overall or relating to key clinical subgroups in which antibiotic prescribing is common (chest signs; fever; physician rating of unwell; sputum/rattly chest; shortness of breath). Objectives To estimate the clinical effectiveness and cost-effectiveness of amoxicillin for uncomplicated lower respiratory tract infections in children both overall and in clinical subgroups. Design Placebo-controlled trial with qualitative, observational and cost-effectiveness studies. Setting UK general practices. Participants Children aged 1–12 years with acute uncomplicated lower respiratory tract infections. Outcomes The primary outcome was the duration in days of symptoms rated moderately bad or worse (measured using a validated diary). Secondary outcomes were symptom severity on days 2–4 (0 = no problem to 6 = as bad as it could be); symptom duration until very little/no problem; reconsultations for new or worsening symptoms; complications; side effects; and resource use. Methods Children were randomised to receive 50 mg/kg/day of oral amoxicillin in divided doses for 7 days, or placebo using pre-prepared packs, using computer-generated random numbers by an independent statistician. Children who were not randomised could participate in a parallel observational study. Semistructured telephone interviews explored the views of 16 parents and 14 clinicians, and the data were analysed using thematic analysis. Throat swabs were analysed using multiplex polymerase chain reaction. Results A total of 432 children were randomised (antibiotics, n = 221; placebo, n = 211). The primary analysis imputed missing data for 115 children. The duration of moderately bad symptoms was similar in the antibiotic and placebo groups overall (median of 5 and 6 days, respectively; hazard ratio 1.13, 95% confidence interval 0.90 to 1.42), with similar results for subgroups, and when including antibiotic prescription data from the 326 children in the observational study. Reconsultations for new or worsening symptoms (29.7% and 38.2%, respectively; risk ratio 0.80, 95% confidence interval 0.58 to 1.05), illness progression requiring hospital assessment or admission (2.4% vs. 2.0%) and side effects (38% vs. 34%) were similar in the two groups. Complete-case (n = 317) and per-protocol (n = 185) analyses were similar, and the presence of bacteria did not mediate antibiotic effectiveness. NHS costs per child were slightly higher (antibiotics, £29; placebo, £26), with no difference in non-NHS costs (antibiotics, £33; placebo, £33). A model predicting complications (with seven variables: baseline severity, difference in respiratory rate from normal for age, duration of prior illness, oxygen saturation, sputum/rattly chest, passing urine less often, and diarrhoea) had good discrimination (bootstrapped area under the receiver operator curve 0.83) and calibration. Parents found it difficult to interpret symptoms and signs, used the sounds of the child’s cough to judge the severity of illness, and commonly consulted to receive a clinical examination and reassurance. Parents acknowledged that antibiotics should be used only when ‘necessary’, and clinicians noted a reduction in parents’ expectations for antibiotics. Limitations The study was underpowered to detect small benefits in key subgroups. Conclusion Amoxicillin for uncomplicated lower respiratory tract infections in children is unlikely to be clinically effective or to reduce health or societal costs. Parents need better access to information, as well as clear communication about the self-management of their child’s illness and safety-netting. Future work The data can be incorporated in the Cochrane review and individual patient data meta-analysis. Trial registration This trial is registered as ISRCTN79914298. Plain language summary Background Children are commonly prescribed antibiotics for chest infections, but such infections are becoming resistant to antibiotics, and it is not clear if antibiotics work in treating them. Methods A total of 432 children who saw their general practitioner with a chest infection were given either an antibiotic (amoxicillin) or a placebo (no antibiotic) for 7 days. Symptom diaries documented the infection’s duration and its side effects. Children not in the placebo study were able to participate in another study that documented the same outcomes (an ‘observational study’). We interviewed parents, doctors and nurses about their observations and concerns. Our patient and public involvement and engagement work with parents indicated that a 3-day symptom reduction was required to justify giving antibiotics. Results After seeing the doctor, parents whose children received antibiotics rated infective symptoms as moderately bad or worse for 5 days, and parents whose children received the placebo rated these for 6 days. Side effects and complications were similar in the two groups. Findings were similar when including the results of the observational study, and for children in whose chest the doctor could hear wheeze or rattles; who had fever; who were rated by the doctor as more unwell, who were short of breath, or who had had bacteria detected in the throat. The costs to the NHS per child were similar (antibiotics, £29; placebo, £26), and the wider costs to society were the same (antibiotics, £33; placebo, £33). Parents found it difficult to interpret their child’s symptoms, and commonly used the sound of the cough to judge severity. Parents commonly consulted to receive an examination and reassurance, and accepted that antibiotics should be used only when ‘necessary’. Clinicians noted a reduction in parents’ expectations for antibiotics. Conclusion Amoxicillin for chest infections in children is unlikely to be effective. General practitioners should support parents to self-manage at home and give clear communication about when and how to seek medical help if they continue to be concerned. Design This was a placebo-controlled trial with qualitative research and health economic analysis, and a parallel observational cohort. Setting UK general practices. Participants Participants were children aged between 6 months and 12 years presenting to primary care with an acute lower respiratory tract infection, defined as one in which an acute cough is the predominant symptom and judged by the general practitioner (GP) to be infective in origin, lasting < 21 days, and with other symptoms or signs localising to the lower respiratory tract (shortness of breath, sputum, pain), and in whom pneumonia was not suspected clinically. Outcomes The primary outcome was the duration in days of symptoms rated moderately bad or worse (measured using a validated diary). The secondary outcomes were symptom severity on days 2–4 (0 = no problem to 6 = as bad as it could be); symptom duration until very little/no problem; reconsultations for new or worsening symptoms; progression of illness sufficient to require hospital assessment; side effects; and resource use. Ethics The protocol was approved by the South West – Central Bristol Research Ethics Committee (reference 15/SW/0300). Methods Children were randomised to receive 50 mg/kg/day oral amoxicillin in divided doses for 7 days, or placebo, using pre-prepared packs randomised by an independent statistician using computer-generated random numbers. Children whom clinicians were unwilling to randomise or parents who were unwilling for their child to be randomised were invited to participate in an observational study in which the same data as in the trial were collected. The revised target sample size (agreed with the Trial Steering Committee, the Data Monitoring and Ethics Committee and the funder) to detect an important clinical difference of 3 days in symptoms duration was 298 participants for 80% power and 398 participants for 90% power. Semistructured interviews were used to explore the views of management and the decisions to participate in the trial. Parents were purposefully sampled by whether they took part in the trial or the observational study, and by practice. Clinicians who recruited participants into the study were also invited to take part in a telephone interview. The interviews were analysed using thematic analysis. Throat swabs were analysed for the presence of bacteria and viruses by multiplex polymerase chain reaction. Statistical analysis Cox regression was used for the primary outcome and for total symptom duration, adjusting for age, baseline symptom severity, prior duration of illness and comorbidity. Linear regression was used for symptom severity, and logistic regression was used for reconsultation, progression of illness and side effects, adjusting for the same baseline covariates as in the primary analysis. Analysis was by intention to treat, as randomised regardless of non-adherence or protocol deviations. Multiple imputation was used as the primary analysis, comprising all variables from the analysis model and any predictors of missingness, and using 100 imputations. Prespecified subgroup analyses were carried out on chest signs, sputum/rattly chest, history of fever, physician rating of unwell, shortness of breath, oxygen saturation below 95%, and STARWAVe clinical prediction rule for hospitalisation. For the observational data set, stratification by propensity scores was used to control for confounding by indication, and the data were merged with the trial data set to facilitate more powerful analyses. A logistic regression model was built to predict the progression of illness, and discrimination was assessed using estimates of area under the receiver operator curve that were bootstrapped for internal validation. Health economic analysis Both cost-effectiveness (in GBP per unit of primary outcome) and cost per quality-adjusted life-year (QALY) were estimated. The base case took an NHS perspective, but some non-NHS costs were also included (remedies and time off work). Resource use data were collected by a notes review in primary care supplemented by the diary. Unit costs of primary care consultation, community services, outpatient visits and accident and emergency attendances were costed based on the Personal Social Services Research Unit. National reference costs were used to cost hospital stay based on corresponding diagnostic categories. Medications were priced based on the British National Formulary. All costs were based on 2019 prices. QALYs were based on the EQ-5D-Y (EuroQol-5 Dimensions Youth), collected weekly, and on the recommended national tariff. Trial results A total of 432 children were randomised (antibiotics, n = 221; placebo, n = 211). The duration of moderately bad symptoms was similar in the two groups [median 5 vs. 6 days, respectively; hazard ratio (HR) 1.13, 95% confidence interval (CI) 0.90 to 1.42]. Return with new or worsening symptoms (29.7% vs. 38.2%; risk ratio 0.80, 95% CI 0.58 to 1.05), progression of illness requiring hospital assessment (2.4% vs 2.0%) and side effects (38% vs. 34%) were also similar in the two groups. A small difference in mean symptom severity on days 2–4 (1.8 vs. 2.1 points; difference 0.28 points, 95% CI 0.04 to 0.51) is unlikely to be clinically meaningful. No differences were seen for the primary outcome in the five prespecified clinical subgroups in which antibiotic prescribing is common: chest signs subgroup (antibiotics 6 days vs. placebo 6 days; HR 0.97, 95% CI 0.65 to 1.43), sputum/rattly chest (5 vs. 7 days; 1.16, 95% CI 0.83 to 1.64), fever (5 vs. 6 days; 1.23, 95% CI 0.88 to 1.73), physician rating of unwell (5 vs. 6 days; 1.25, 95% CI 0.85 to 1.83) and shortness of breath (5 vs. 6 days; 1.13, 95% CI 0.72 to 1.77). There was also no evidence that the presence of bacteria in the throat swab mediated antibiotic effectiveness. Estimates from complete cases (n = 317) were very similar, as were estimates from a per-protocol analysis for children taking 11 or more of the of 15 doses in the first 5 days. NHS costs per child were slightly higher with antibiotics (antibiotic, £29; placebo, £26) and non-NHS costs were the same (antibiotics, £33; placebo, £33), but QALY data were too incomplete for robust imputation. The incremental cost per QALY (incremental cost-effectiveness ratio) was £30,851 (95% CI –£73,639 to £109,429) based on estimates from the means of complete cases and £6417 (95% CI –£12,240 to £20,535) based on the estimates using imputed data. Observational study A total of 326 children were recruited to the observational study. The estimate of benefit of antibiotics for the primary outcome was similar to that in the trial (HR 1.16, 95% CI 0.95 to 1.41). A prognostic model to predict the progression of illness consisting of seven variables (baseline severity, difference in respiratory rate from normal for age, duration of prior illness, oxygen saturation, sputum/rattly chest, passing urine less often and diarrhoea) had good discrimination (bootstrapped area under the receiver operator curve 0.85) and calibration, and a three-item model (respiratory rate, oxygen saturation, sputum/rattly chest) also performed well (area under the receiver operator curve 0.81). Qualitative results Thirty semistructured telephone interviews were conducted with 16 parents and 14 clinicians. Parents found it difficult to interpret the symptoms and signs, and commonly used the sounds of the cough to judge severity, which highlights the need to provide better information to support parents. Many parents said that the main reason for consulting was to receive a clinical examination and reassurance regarding illness severity. Parents acknowledged that antibiotics should be used only when ‘necessary’, and many of the clinicians also noted a shift in parents’ expectations about antibiotics and that they were satisfied with a clinical assessment, reassurance and advice. Decisions to take part in the trial were influenced by the perceived risks associated with taking a placebo compared with immediate antibiotics, and with taking antibiotics unnecessarily. Clear communication about the self-management of their child’s illness and ‘safety-netting’ (information on the natural course of the illness and advice about when it might be necessary to reconsult) were identified as important when implementing ‘no antibiotic’ prescribing strategies to reassure parents and to support prescribing decisions. Limitations The study was underpowered to detect small benefits in the key clinical subgroups. The trial included children who were more unwell than those in recent large generalisable cohorts, which suggests that, if anything, the benefit of antibiotics has been overestimated. Given the very large numbers of missing data, the imputed estimates in the economic analysis must be viewed with caution. If the costs of AMR were included, then these estimates of cost-effectiveness would worsen. Conclusions Implications for clinical care Amoxicillin for uncomplicated chest infections in children makes little difference to symptom burden or to health or societal costs. Better access to information is needed to support parents’ decision-making, as is clear clinician communication about the self-management of their child’s illness and safety-netting. A prognostic score using variables that can be collected very easily during consultations can be used to identify children who are at low risk of illness progression. Implications for future research •The data can be incorporated in a Cochrane review and an individual patient data meta-analysis. •Further work on the incremental QALY gain from antibiotics is needed, assessing a range of models and their implications when imputing missing QALY data, and better evidence is needed about how to incorporate AMR resource implications in modelling. •The prognostic score should be externally validated and could be developed as an app with automated outputs, and thereafter used as a tool to reduce antibiotic prescribing for antimicrobial stewardship interventions. ObjectiveTo explore the effectiveness of combined immunotherapy (IT) and stereotactic radiosurgery (SRS) and address the gap between evidence-based clinical practice and academic knowledge of optimal timing of IT relative to SRS. In addition, to meet the unmet need for an up-to-date prognostic assessment model in the era of IT.MethodsThe data of 86 non-small cell lung cancer brain metastasis (NSCLCBM) patients treated with SRS to 268 brain metastases (BMs) were retrospectively extracted from our hospital database. The Kaplan–Meier analysis was employed for overall survival (OS) and a log-rank test for comparison between groups. Cox proportional hazards regression models were used to identify the significant prognostic factors. The prognostic nomogram was established utilizing the rms package of R software.ResultsIT was found to be associated with improved OS (from BM diagnosis: HR 0.363, 95% CI 0.199 - 0.661, P < 0.001; from SRS: HR 0.472, 95% CI 0.260 - 0.857, P = 0.014). Individuals who received IT in combination with SRS had better OS than those who didn’t (from the day of BM diagnosis: 16.8 vs. 8.4 months, P = 0.006; from the day of SRS: 12 vs. 7 months, P = 0.037). Peri-SRS timing of IT administration was a significant prognostic factor for OS (from BM diagnosis: HR 0.132, 95% CI 0.034 - 0.517, P = 0.004; from SRS: HR 0.14, 95% CI 0.044 - 0.450, P = 0.001). Initiating IT after SRS led to superior OS than concurrent or before (from BM diagnosis: 26.5 vs. 14.1 vs. 7.1 months; from SRS: 21.4 vs. 9.9 vs. 4.1 months, respectively). Additionally, we build a nomogram incorporating IT, cumulative intracranial tumor volume (CITV), and recursive partitioning analysis (RPA), demonstrating a remarkable prognosis prediction performance for SRS-treated NSCLCBM patients.ConclusionPeri-SRS IT is a promising approach in treating NSCLCBM, as improved OS was observed without significantly increasing adverse events. Receipt of IT post-SRS was associated with superior OS than those who received IT concurrently or before. Incorporating IT and CITV into the RPA index could augment its prognosis assessment value for SRS-treated NSCLCBM patients, predominantly in the wild-type.

Ferromanganese (Fe-Mn) polymetallic nodules are significant marine mineral resources containing various metal elements of substantial economic and scientific research value. Previous studies have primarily focused on the mineralogy and geochemistry of the nodules, while research on their nano-mineralogy is still lacking. In this study, we conducted scanning electron microscopy (SEM), X-ray powder diffraction (XRD), transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FT-IR), Brunauer–Emmett–Teller (BET) porous structure gas adsorption/desorption, and specific surface area analysis to examine the nano-mineralogy and mineralization of the polymetallic nodules from the interbasin of seamounts in the western Pacific Ocean (IBSWP). The results indicate that the growth profiles of the IBSWP polymetallic nodules exhibit microstructural features such as laminated, stromatolithic, columnar, and mottled structures. The mineral compositions are primarily composed of Fe-Mn minerals and detritus, including quartz; minerals from the feldspar group; and minerals from the clay group. The Fe-Mn phase minerals are relatively poorly crystallized. The Mn-phase minerals contain vernadite and small amounts of todorokite, buserite, and birnessite, while the Fe-phase minerals are mainly comprised of amorphous FeOOH. The main ore-forming minerals consist of nano-minerals, and the nanostructures of the polymetallic nodules endow them with unusually large specific surface areas and pore volumes, resulting in strong adsorption properties. The unique nano-properties and surface/interface adsorption effects of Fe-Mn minerals play a crucial role in controlling the enrichment of ore-forming elements.

(1) Background: Prosthetic rehabilitation is essential for upper limb amputees to regain their ability to work. However, the abandonment rate of prosthetics is higher than 50% due to the high cost of rehabilitation. Virtual technology shows potential for improving the availability and cost-effectiveness of prosthetic rehabilitation. This article systematically reviews the application of virtual technology for the prosthetic rehabilitation of upper limb amputees. (2) Methods: We followed PRISMA review guidance, STROBE, and CASP to evaluate the included articles. Finally, 17 articles were screened from 22,609 articles. (3) Results: This study reviews the possible benefits of using virtual technology from four aspects: usability, flexibility, psychological affinity, and long-term affordability. Three significant challenges are also discussed: realism, closed-loop control, and multi-modality integration. (4) Conclusions: Virtual technology allows for flexible and configurable control rehabilitation, both during hospital admissions and after discharge, at a relatively low cost. The technology shows promise in addressing the critical barrier of current prosthetic training issues, potentially improving the practical availability of prosthesis techniques for upper limb amputees.

The rice–crayfish farming model has been rapidly developed and become an economically viable method to supply food in China in recent years. However, its environmental and economic sustainability has not been thoroughly investigated. This study uses a survey in 2016 and a field experiment in 2017 in Qianjiang, Hubei Province, China to assess the relative economics of concurrent rice–wheat (RW), rice–crayfish (RC), and crayfish monoculture (CM) models in waterlogged land areas. The field survey indicated that the RC model had a higher benefit–cost ratio (3.5:1) than the RW (2.0:1) and CM (3.1:1) models and the RC model protected farmers’ enthusiasm for grain production facing unfavourable weather conditions. The field experiment aimed to explore nitrogen management strategies in RC fields. In the experiment, four levels of nitrogen concentration gradient - 0 kg N ha−1 (0 N), 75 kg N ha−1 (75 N), 150 kg N ha−1 (150 N) and 225 kg N ha−1 (225 N), were set in a 2-year-old rice–crayfish (RC2) field, an 8-year-old rice–crayfish (RC8) field, and a RW field as a control. The field experiment results suggested that the peak rice yield in RW, RC2, and RC8 occurred when 225 N, 150 N and 75 N were used, respectively. In RC2 and RC8, however, residual feed-nitrogen that was not used by crayfish was utilized by rice plants. Thus, an optimal amount of nitrogen in RC fields was proposed to improve the nitrogen use efficiency and reduce environmental pollution by nitrogen fertilizer. Farmers use less nitrogen but have higher net income in RC than in RW and CM. It is necessary to sustainably develop integrated farming technologies (i.e., proper field configurations for rice fields) to effectively sustain rice production. The results also showed that the RC farming model was a viable diversification option for rice farmers in waterlogged land.

Various biochar samples were used for exploring the Fe(III)/biochar interactions and transformation of reactive oxygen species during co-activation of H2O2. The enhanced •OH production is positively related to the biochars' capacity of reducing Fe(III) into Fe(II), which is then linked with their redox-active moieties and adsorption of Fe(III). Besides the •OH produced sustainably from Fenton reaction between H2O2 and regenerated Fe(II), a novel route involving dissolved O2 is proposed to enhance •OH production in the co-activated system. Consequently, faster and more efficient oxidation of refractory pollutants was achieved by co-activation of H2O2 with Fe(III) and biochar together.

Fe-Mn polymetallic crusts and nodules from the South China Sea (SCS) consist of submarine ferromanganese (Fe-Mn) oxide precipitates, and represent important marine mineral resource with substantial economic and scientific research value. Previous studies on the SCS polymetallic crusts and nodules were mainly focused on their bulk mineralogy and geochemistry, whilst research on their nanomineralogy is still lacking. In this study, transmission electron microscopy (TEM), Raman spectroscopic mapping, and in-situ micro X-ray diffraction (XRD) analysis were conducted on the nano-mineralogy of the SCS polymetallic crusts and nodules. It is found that the SCS polymetallic crusts and nodules consist mainly of layered/columnar/mottled nano-phase Fe-Mn minerals and detritus such as quartz, feldspar, and clays. Also, an independent Ti mineral phase has been documented, and the mineralogical analysis reveals the transformation from vernadite to birnessite and todorokite. Titanium forms colloidal minerals in seawater and precipitates into the crusts and nodules with other colloids, such as FeOOH and Si-Al. Vernadite and birnessite can be transformed to todorokite with stable structure under sub-oxic conditions. Therefore, the SCS polymetallic crusts and nodules were formed in a short period of sub-oxic environment and diagenetic process, and the transformation can influence the enrichment of Ni and other metals during the crust/nodule growth.

BackgroundFms-like tyrosine kinase 3 (FLT3) gene mutations occur in approximately 30% of all patients with acute myeloid leukemia (AML). Internal tandem duplication (ITD) in the juxtamembrane domain and point mutations within the tyrosine kinase domain (TKD) are two distinct types of FLT3 mutations. FLT3-ITD has been determined as an independent poor prognostic factor, but the prognostic impact of potentially metabolically related FLT3-TKD remains controversial. Hence, we performed a meta-analysis to investigate the prognostic significance of FLT3-TKD in patients with AML.MethodsA systematic retrieval of studies on FLT3-TKD in patients with AML was performed in PubMed, Embase, and Chinese National Knowledge Infrastructure databases on 30 September 2020. Hazard ratio (HR) and its 95% confidence intervals (95% CIs) were used to determine the effect size. Meta-regression model and subgroup analysis were used for heterogeneity analysis. Begg’s and Egger’s tests were performed to detect potential publication bias. The sensitivity analysis was performed to evaluate the stability of findings in meta-analysis.ResultsTwenty prospective cohort studies (n = 10,970) on the prognostic effect of FLT3-TKD in AML were included: 9,744 subjects with FLT3-WT and 1,226 subjects with FLT3-TKD. We found that FLT3-TKD revealed no significant effect on disease-free survival (DFS) (HR = 1.12, 95% CI: 0.90–1.41) and overall survival (OS) (HR = 0.98, 95% CI: 0.76–1.27) in general. However, meta-regressions demonstrated that patient source contributed to the high heterogeneity observed in the prognosis of FLT3-TKD in AML. To be specific, FLT3-TKD represented a beneficial prognosis of DFS (HR = 0.56, 95% CI: 0.37–0.85) and OS (HR = 0.63, 95% CI: 0.42–0.95) for Asians, whereas it represented an adverse prognosis of DFS for Caucasians with AML (HR = 1.34, 95% CI: 1.07–1.67).ConclusionFLT3-TKD revealed no significant effects on DFS and OS of patients with AML, which is consistent with the controversial status nowadays. Patient source (Asians or Caucasians) can be partially explained the different effects of FLT3-TKD in the prognosis of patients with AML.

Abstract Magnetic skyrmions are promising information carriers for dense and energy‐efficient information storage owing to their small size, low driving‐current density, and topological stability. Electrical detection of skyrmions is a crucial requirement to drive skyrmion devices toward applications. The use of a magnetic tunnel junction (MTJ) is commonly suggested for this purpose as MTJs are key spintronic devices for large‐scale commercialization that can convert magnetic textures into electrical signals. To date, however, it has been challenging to realize skyrmions in MTJs due to incompatibility between standard skyrmion materials and highly efficient MTJ electrodes. Here, a material stack combining magnetic multilayers, which host 100 nm scale skyrmions, with a perpendicularly magnetized MTJ, is reported. The devices are designed so that the skyrmions in the multilayer are imprinted into the MTJ's free layer via magnetostatic interactions. The electrical response of a single skyrmion is successfully identified by employing simultaneous imaging of the magnetic texture and the electrical measurement of the MTJ resistance. The results are an important step toward all‐electrical detection of skyrmions.

Gain-of-function mutations of SHP2, especially D61Y and E76K, lead to the development of neoplasms in hematopoietic cells. Previously, we found that SHP2-D61Y and -E76K confer HCD-57 cells cytokine-independent survival and proliferation via activation of MAPK pathway. Metabolic reprogramming is likely to be involved in leukemogenesis led by mutant SHP2. However, detailed pathways or key genes of altered metabolisms are unknown in leukemia cells expressing mutant SHP2. In this study, we performed transcriptome analysis to identify dysregulated metabolic pathways and key genes using HCD-57 transformed by mutant SHP2. A total of 2443 and 2273 significant differentially expressed genes (DEGs) were identified in HCD-57 expressing SHP2-D61Y and -E76K compared with parental cells as the control, respectively. Gene ontology (GO) and Reactome enrichment analysis showed that a large proportion of DEGs were involved in the metabolism process. Kyoto Encyclopedia of Gene and Genome (KEGG) pathway enrichment analysis showed that DEGs were the mostly enriched in glutathione metabolism and biosynthesis of amino acids in metabolic pathways. Gene Set Enrichment Analysis (GSEA) revealed that the expression of mutant SHP2 led to a significant activation of biosynthesis of amino acids pathway in HCD-57 expressing mutant SHP2 compared with the control. Particularly, we found that ASNS, PHGDH, PSAT1, and SHMT2 involved in the biosynthesis of asparagine, serine, and glycine were remarkably up-regulated. Together, these transcriptome profiling data provided new insights into the metabolic mechanisms underlying mutant SHP2-driven leukemogenesis.

Premenstrual syndrome (PMS), a recurrent and moderate disorder that occurs during the luteal phase of the menstrual cycle and quickly resolves after menstruation, is characterized by somatic and emotional discomfort that can be severe enough to impair daily activities. Current therapeutic drugs for PMS such as selective serotonin reuptake inhibitors are not very satisfying. As a critical pineal hormone, melatonin has increasingly been suggested to modulate PMS symptoms. In this review, we update the latest progress on PMS-induced sleep disturbance, mood changes, and cognitive impairment and provide possible pathways by which melatonin attenuates these symptoms. Moreover, we focus on the role of melatonin in PMS molecular mechanisms. Herein, we show that melatonin can regulate ovarian estrogen and progesterone, of which cyclic fluctuations contribute to PMS pathogenesis. Melatonin also modulates gamma-aminobutyric acid and the brain-derived neurotrophic factor system in PMS. Interpreting the role of melatonin in PMS is not only informative to clarify PMS etiology but also instructive to melatonin and its receptor agonist application to promote female health. As a safe interaction, melatonin treatment can be effective in alleviating symptoms of PMS. However, symptoms such as sleep disturbance, depressive mood, cognitive impairment are not specific and can be easily misdiagnosed. Connections between melatonin receptor, ovarian steroid dysfunction, and PMS are not consistent among past studies. Before final conclusions are drawn, more well-organized and rigorous studies are recommended.

Abstract Chronic lymphocytic leukemia (CLL) is characterized by the accumulation of neoplastic B lymphocytes with high levels of Wnt5a in the plasma. Currently, the cell source of Wnt5a remains controversial. The receptor of Wnt5a is ROR1, whose expression is associated with disease progression and resistance to venetoclax, a BCL-2 inhibitor approved for the treatment of CLL. In this study, we found that the levels of Wnt5a in the plasma of CLL patients were positively correlated with absolute monocyte counts, but not lymphocyte counts. We cultured monocyte-derived nurse-like cells (NLCs) from patients with CLL, and detected Wnt5a expressed in NLCs. Flow cytometry and transwell assays showed that the antibody neutralizing Wnt5a inhibited the enhanced survival and migration in CLL cells co-cultured with NLCs. Furthermore, we performed a drug screening with CLL cells cultured with or without NLCs with a library containing 133 FDA-approved oncology drugs by using high-throughput flow cytometry. We observed a significant resistance to venetoclax in CLL cells co-cultured with NLCs. Immunoblot revealed the activation of NF-κB with enhanced expression of MCL-1 and BCL-XL in CLL cells co-cultured with NLCs. Neutralizing Wnt5a or blocking NF-κB pathway significantly decreased the expression of MCL-1 and BCL-XL, which leads to enhanced sensitivity to venetoclax in CLL cells co-cultured with NLCs. In conclusion, our data showed that NLCs could be one of the sources of Wnt5a detected in patients with CLL, and Wnt5a-induced NF-κB activation in the CLL microenvironment results in resistance to venetoclax in CLL cells.

Radiotherapy-associated secondary cancer is an important issue for the treatment of breast cancer (BCa). This study aimed to investigate the molecular mechanism and genetic risk factors for radiation-associated secondary diseases in BCa.

The electroencephalogram (EEG), for measuring the electrophysiological activity of the brain, has been widely applied in automatic detection of epilepsy seizures. Various EEG-based seizure detection algorithms have already yielded high sensitivity, but training those algorithms requires a large amount of labelled data. Data labelling is often done with a lot of human efforts, which is very time-consuming. In this study, we propose a hybrid system integrating an unsupervised learning (UL) module and a supervised learning (SL) module, where the UL module can significantly reduce the workload of data labelling. For preliminary seizure screening, UL synthesizes amplitude-integrated EEG (aEEG) extraction, isolation forest-based anomaly detection, adaptive segmentation, and silhouette coefficient-based anomaly detection evaluation. The UL module serves to quickly locate the determinate subjects (seizure segments and seizure-free segments) and the indeterminate subjects (potential seizure candidates). Afterwards, more robust seizure detection for the indeterminate subjects is performed by the SL using an EasyEnsemble algorithm. EasyEnsemble, as a class-imbalance learning method, can potentially decrease the generalization error of the seizure-free segments. The proposed method can significantly reduce the workload of data labelling while guaranteeing satisfactory performance. The proposed seizure detection system is evaluated using the Children’s Hospital Boston-Massachusetts Institute of Technology (CHB-MIT) scalp EEG dataset, and it achieves a mean accuracy of 92.62%, a mean sensitivity of 95.55%, and a mean specificity of 92.57%. To the best of our knowledge, this is the first epilepsy seizure detection study employing the integration of both the UL and the SL modules, achieving a competitive performance superior or similar to that of the state-of-the-art methods.

With the widespread use of mobile and sensing devices, and the popularity of online map-based services, such as navigation services, the volume of spatio-temporal data is growing rapidly. Conventional big data technologies in existing distributed systems cannot effectively process spatio-temporal big data with temporal continuity and spatial proximity. How to construct an effective index for the application requirements of spatio-temporal data in a distributed environment has become one of the hotspots of spatio-temporal big data research. Many spatio-temporal indexing methods have been proposed to support efficient query processing of spatio-temporal data. In this article, the various spatio-temporal big data indexing methods proposed by domestic and foreign researchers from 2010 to 2020 are classified and summarized according to the distributed environment and application background, and the hot issues that need to be paid attention to in the future are proposed according to the changes in application requirements

This study aimed to investigate the effects of dietary tannic acid (TAN) on the gas production, growth performance, antioxidant capacity, rumen microflora, and fermentation function of beef cattle through in vitro and in vivo experiments. TAN was evaluated at 0.15% (dry matter basis, DM) in the in vitro experiment and 0.20% (DM basis) in the animal feeding experiment. The in vitro results revealed that compared with control (CON, basal diet without TAN), the addition of TAN significantly increased the cumulative gas production and asymptotic gas production per 0.20 g dry matter substrate (p < 0.01), with a tendency to reduce methane concentration after 96 h of fermentation (p = 0.10). Furthermore, TAN supplementation significantly suppressed the relative abundance of Methanosphaera and Methanobacteriaceae in the fermentation fluid (LDA > 2.50, p < 0.05). The in vivo experiment showed that compared with CON, the dietary TAN significantly improved average daily gain (+0.15 kg/d), dressing percent (+1.30%), net meat percentage (+1.60%), and serum glucose concentration (+23.35%) of beef cattle (p < 0.05), while it also significantly reduced hepatic malondialdehyde contents by 25.69% (p = 0.02). Moreover, the TAN group showed significantly higher alpha diversity (p < 0.05) and increased relative abundance of Ruminococcus and Saccharomonas (LDA > 2.50, p < 0.05), while the relative abundance of Prevotellaceae in rumen microbial community was significantly decreased (p < 0.05) as compared to that of the CON group. In conclusion, the dietary supplementation of TAN could improve the growth and slaughter performance and health status of beef cattle, and these favorable effects might be attributed to its ability to alleviate liver lipid peroxidation, enhance glucose metabolism, and promote a balanced rumen microbiota for optimal fermentation.

This study explored the effects of drinking heated water in the cold seasons on the serum metabolism, rumen microbial fermentation, and metabolome of beef cattle. Twelve fattening cattle (642 ± 14.6 kg) aged 21 to 22 months were randomly and equally divided into two groups based on body weight: one receiving room-temperature water (RTW; average 4.39 ± 2.55 °C) and the other heated water (HW; average 26.3 ± 1.70 °C). The HW group displayed a significant decrease in serum glucose (p < 0.01) and non-esterified fatty acid (p < 0.01), but increases in insulin (p = 0.04) and high-density lipoprotein (p = 0.03). The rumen fermentation parameters of the HW group showed substantial elevations in acetate (p = 0.04), propionate (p < 0.01), isobutyrate (p = 0.02), and total volatile fatty acids (p < 0.01). Distinct bacterial composition differences were found between RTW and HW at the operational taxonomic unit (OTU) level (R = 0.20, p = 0.01). Compared to RTW, the HW mainly had a higher relative abundance of Firmicutes (p = 0.07) at the phylum level and had a lower abundance of Prevotella (p < 0.01), norank_f_p-215-o5 (p = 0.03), and a higher abundance of NK4A214_group (p = 0.01) and Lachnospiraceae_NK3A20_group (p = 0.05) at the genus level. In addition, NK4A214_group and Lachnospiraceae_NK3A20_group were significantly positively correlated with the rumen propionate and isovalerate (r > 0.63, p < 0.05). Prevotella was negatively correlated with rumen propionate and total volatile fatty acids (r = −0.61, p < 0.05). In terms of the main differential metabolites, compared to the RTW group, the expression of Cynaroside A, N-acetyl-L-glutamic acid, N-acetyl-L-glutamate-5-semialdehyde, and Pantothenic acid was significantly upregulated in HW. The differentially regulated metabolic pathways were primarily enriched in nitrogen metabolism, arginine biosynthesis, and linoleic acid metabolism. Prevotella was significantly positively correlated with suberic acid and [6]-Gingerdiol 3,5-diacetate (r > 0.59, p < 0.05) and was negatively correlated with Pantothenic acid and isoleucyl-aspartate (r < −0.65, p < 0.05). NK4A214_group was positively correlated with L-Methionine and glycylproline (r > 0.57, p < 0.05). Overall, our research demonstrates the important relationship between drinking water temperature and metabolic and physiological responses in beef cattle. Heating drinking water during cold seasons plays a pivotal role in modulating internal energy processes. These findings underscore the potential benefits of using heated water as a strategic approach to optimize energy utilization in beef cattle during the cold seasons.

Mammals exhibit circadian rhythms in their behavior and physiological activities to adapt to the diurnal changes of the environment. Improper feeding methods can disrupt the natural habits of animals and harm animal health. This study investigated the effects of feeding amount and feeding time on growing rabbits in northern China during spring. A total of 432 healthy 35-day-old weaned rabbits with similar body weight were randomly assigned to four groups: whole day diet-unrestricted feeding (WUF), whole day diet-restricted feeding (WRF), nighttime diet-unrestricted feeding (NUF), and nighttime diet-restricted feeding (NRF). The results showed that nighttime diet-unrestricted feeding improved performance, circadian rhythm of behavior, and body temperature, while reducing the risk of diarrhea and death. WRF group increased daytime body temperature but had no significant difference in feed conversion rate. The study suggests that nighttime diet-unrestricted feeding in spring can improve the growth and welfare of rabbits in northern China. Our study underscores the pivotal role of feeding timing in enhancing animal health. Future investigations should delve into the underlying mechanisms and expand the application of this strategy across seasons and regions to improve rabbit husbandry practices.

The Yong’an-Meixian Late Paleozoic Hercynian depression, located in western Fujian-eastern Guangdong, is an important metallogenic belt in China. The Yushui copper-polymetallic massive sulfide deposit from the middle part of the depression, with extremely high copper grades, has attracted considerable attention and research interest from geologists for years. In most previous research, the ore-forming material source and metallogenic process were inferred from indirect evidence (i.e., using H-O-C-S isotopic systematics as geochemical tracers). In this paper, the ore-forming process of the Yushui deposit has been studied directly by using nontraditional stable (Fe–Cu) isotopes for the first time, providing new evidence for the genesis of this deposit. The results show that there is a relatively negative Fe-Cu isotopic composition in the Yushui deposit, with δ56Fe values ranging from −0.519 to −1.063‰ and δ65Cu values ranging from −1.539 to −1.609‰, respectively. The fractionation of Fe isotopes is primarily attributed to hydrothermal leaching of the basement strata by ore-forming fluids, along with rapid precipitation of sulfides during the ore-forming process. On the other hand, the fractionation of Cu isotopes is probably controlled by the relatively low temperature of ore formation, sulfide precipitation and the involvement of organic matter in mineralization. Combining our findings with previous studies, the ore-forming materials of the Yushui deposit are likely derived from the basement ore-bearing strata (pre-Devonian strata) through leaching by hydrothermal fluids. Moreover, some of the ore bodies might have been locally overprinted by late-stage hydrothermal reworking and alteration.

The research aimed to investigate the suitable drinking water temperature in winter and its effect on the growth performance, antioxidant capacity, and rumen fermentation function of beef cattle. A total of 40 beef cattle (640 ± 19.2 kg) were randomly divided into five treatments with eight cattle in each treatment raised in one pen according to initial body weight. Each treatment differed only in the temperature of drinking water, including the room-temperature water and four different heat water groups named RTW, HW_1, HW_2, HW_3, and HW_4. The measured water temperatures were 4.39 ± 2.546 °C, 10.6 ± 1.29 °C, 18.6 ± 1.52 °C, 26.3 ± 1.70 °C, and 32.5 ± 2.62 °C, respectively. The average daily gain (ADG) showed a significant linear increase during d 0 to 60 and a quadratic increase during d 31 to 60 with rising water temperature (p < 0.05), and the highest ADG of 1.1911 kg/d was calculated at a water temperature of 23.98 °C (R2 = 0.898). The average rectal temperature on d 30 (p = 0.01) and neutral detergent fiber digestibility (p < 0.01) increased linearly with increasing water temperature. Additionally, HW_2 reduced serum triiodothyronine, thyroxine, and malondialdehyde (p < 0.05), and increased serum total antioxidant capacity (p < 0.05) compared with RTW. Compared with HW_2, RTW had unfavorable effects on ruminal propionate, total volatile fatty acids, and cellulase concentrations (p < 0.05), and lower relative mRNA expression levels of claudin-4 (p < 0.01), occludin (p = 0.02), and zonula occludens-1 (p = 0.01) in the ruminal epithelium. Furthermore, RTW had a higher abundance of Prevotella (p = 0.04), Succinivibrionaceae_UCG-002 (p = 0.03), and Lachnospiraceae_UCG-004 (p = 0.03), and a lower abundance of Bifidobacteriaceae (p < 0.01) and Marinilabiliaceae (p = 0.05) in rumen compared to HW_2. Taken together, heated drinking water in cold climates could positively impact the growth performance, nutrient digestibility, antioxidant capacity, and rumen fermentation function of beef cattle. The optimal water temperature for maximizing ADG was calculated to be 23.98 °C under our conditions. Ruminal propionate and its producing bacteria including Prevotella, Succinivibrionaceae, and Lachnospiraceae might be important regulators of rumen fermentation of beef cattle drinking RTW under cold conditions.

In this study, we analyzed the clay mineralogy and geochemistry of surface and drill core samples from the northeastern Beibu Gulf in order to unravel the sediment provenance of, and factors controlling, the sedimentary environment. The main clay mineral assemblage in the surface sediment samples included kaolinite (27–72%), smectite (4–51%), illite (7–20%), and chlorite (8–17%). The study area comprises three major clay distribution zones (from the northeastern coastal area to central Beibu Gulf basin), i.e., the kaolinite-dominated, kaolinite–smectite, and smectite-dominated zones. The zoning of the clay mineralogy and major and trace elements indicated the mixing of coarse terrigenous sediments with distal fine sediments. Early Holocene sea level rise was documented in core B15-1, which had three sedimentary units (divided into Units 1–3 from top to bottom), as revealed by the changes in the dominant clay minerals and geochemical/oxide ratios (SiO2/Al2O3, Rb/Sr, Sr/Ba, and Ti/Ca) in Unit 2. Unit 1 and Unit 3 were likely deposited in the continental and marine environments, respectively. The low sedimentation rate and hydrodynamic disturbance may have influenced the deposition process more than climatic fluctuations.

Coal gasification slag is a kind of solid coal chemical waste which mainly contains aluminosilicate minerals.The gasification slag modified by mechanical grinding and chemical excitation can be used as active admixture of composite Portland cement, which can effectively reduce the carbon footprint in the production and preparation of cement building materials.In order to clarify the working performance of modified gasification slag cement fresh paste, the compatibility of gasification slag-cement binary system with superplasticizer was evaluated by studying the fluidity, ζ-potential and particle size distribution of gasification slag-cement composite paste.The results show that the coal gas slag modified by diethyl alcohol monoisopropanolamine is a kind of mesoporous material, and it has good workability when the content is not more than 30 %.Polycarboxylic acid superplasticizer is beneficial to the dispersibility and fluidity of modified gas slag cement paste, and the fresh coal gas slag cement paste mixed with polycarboxylic acid superplasticizer shows modified Bingham fluid characteristics.The experimental results have high theoretical reference value for studying the workability of gas slag-cement binary system and its compatibility with superplasticizer.

ObjectiveIn order to reduce the risk of export, the current status of the maximum residue limits (MRLs) of pesticides in garlic at home and abroad were studied. And on this basis, the suggestions for improving the standards of pesticide residues of China in garlic were given.MethodsCurrent and valid standards of pesticide MRLs in garlic from European Union (EU), International Codex Alimentarius Commission (CAC), Japan, South Korea and China were collected, compared and analyzed.ResultsThere are 129 pesticide MRLs related to garlic in China, and the EU, Japan, South Korea and CAC standards have 510, 252, 173 and 23 pesticide MRLs, respectively. There are 93, 67, 56 and 18 pesticides in the EU, Japan, South Korea and CAC standards that are the same as those in China, respectively. Among them, 49, 19, 22 and 4 pesticides MRLs are lower than China, respectively.ConclusionDue to the different dietary structures and registered pesticides in different countries, China and the EU, CAC, Japan and South Korea have significant differences in the total number of pestcides and their MRLs. It is recommended to improve relevant standards in China based on the actual garlic production and dietary needs in China.

ObjectiveTo detect the content of Cadmium (Cd) in crab sold in Beijing from 2018 to 2020 and carry out health risk assessment.MethodsRandom sampling method was adopted, common crab sold in Beijing were sampled. The content of Cd was determined by atomic absorption spectroscopy to evaluate the health risk.ResultsA total of 83 crab samples were collected. The average level of Cd in crabs was 0.647 mg/kg. Among them, the Cd content in freshwater crabs ranged from 0.043-0.620 mg/kg with the average level of 0.183 mg/kg. Whereas, the concentration of Cd in seawater crabs ranged from 0.033-3.200 mg/kg with the average level of 1.070 mg/kg. Moreover, the difference of the over-limit ratio was significant during three year. The THQ value and total THQ value of heavy metals in crab ingested by children and adults decreased in turn. The THQ values of Cd were less than 1.ConclusionAlthough the results indicated that there were certain amounts of Cd residues in the crabs sold in Beijing during 2018-2020, the dietary risk was relatively low because of the low intake.

Current rectification in the THz regime using geometric diodes has been explained previously by exotic transport properties. Here, an alternative mechanism based on asymmetric bias-induced barrier lowering is presented.

Abstract Achieving a high‐gain omnidirectional patch antenna with low profile is a challenging task. On the basis of improving antennas by metamaterials, a broadband near‐zero‐index metamaterial (NZIM) is designed to enhance the horizontal gain of the omnidirectional patch antenna. The zero‐index metamaterial is loaded on the substrate of three antennas with different working bands (4.99–5.50 GHz). The results show that the horizontal gain of antennas is enhanced by more than 0.6 dB at various frequencies. The roundness of antennas is also optimised. Therefore, using NZIM can increase the horizontal gain with a low profile (λ/30) structure size. The low profile, high‐gain omnidirectional patch antenna has great potential in the conformal field.

Members of the SNF1-related protein kinase 2 (SnRK2) family are plant-specific serine or threonine kinases that play a pivotal role in the response of plants to abiotic stresses. Members of this plant-specific kinase family have included a critical regulator (SnRK2) of abscisic acid (ABA) response in plants. Plant organ development is governed substantially by the interaction of the SnRK2 and the phytohormone abscisic acid (ABA). Recent research has revealed a synergistic link between SnRK2 and ABA signaling in a plant’s response to stress such as drought and shoot growth. SnRK2 kinases play a dual role in the control of SnRK1 and the development of a plant. The dual role of SnRK2 kinases promotes plant growth under optimal conditions and in the absence of ABA while inhibiting the growth of plants in response to ABA. In this review, we have uncovered the roles of ABA-activated SnRK2 kinases in plants, as well as their physiological mechanisms.
