Abstract

A new signal readout method for position-sensitive multi-output detectors, such as those in high-energy spectroscopy measurement and nuclear imaging, was developed by combining the charge division circuit, summing circuit and charge-to-time conversion (QTC) circuit. The 64 outputs of a Hamamatsu H8500 position-sensitive photomultiplier tube were processed, and three digital pulses were generated. The widths of digital pulses were determined using the time-to-digital converter in an field programmable gate array. The energy and position information of incident γ-rays is estimated based on the proportionality between the width of digital pulses and input charge created by γ-photons. A prototype was built using discrete components and tested, and the energy and position resolutions were improved compared with that obtained with standard ADCs. This method greatly simplifies the front-end electronics and the digital interface. It enables a compact electronics system and an easy integration into an ASIC.

Abstract

The pH-sensitive polyacrylic acid (PAA) hydrogels were synthesized by gamma-ray irradiation at an ambient temperature. The influences of dose, monomer concentration, cross-linking agent content, pH, and ionic strength on the swelling ratio ( SR ) of the PAA hydrogels were investigated in detail. The results show that the SR of the hy-drogel decreases with an increase in the dose, monomer concentration, and cross-linking agent content. In alkaline solution, the SR of the hydrogels is much higher than that in acid solution. Also, the ionic strength can influence the SR of the hydrogels. The more the concentration, the lower the SR .

Abstract

In this study, hydrogels of P(NIPA-co-NVP)/clay were synthesized by γ-ray radiation. Different thermo-sensitive hydrogels were made under different experimental conditions such as dose, monomer ratio, and content of clay. X-ray diffraction shows that the layer distance between Na-clays is changed from 1.6 to 2.7 nm because Na-clay pieces can be intercalated or exfoliated by HTMAB, and that between P(NIPA-co-NVP)/clay pieces is 3.4 nm. The swelling property tests show that the LCST of P(NIPA-co-NVP) is higher than PNIPA. With the increase of NVP content, LCST is higher. As the ratio of NIPA/NVP is 95/5, hydrogel shows the best swelling property and LCST is 37°C. LCST of P(NTPA-co-NVP)/clay hydrogel is not changed, but the strength and swelling properties are better.

Abstract

A fast neutron detection system based on a scintillating plastic fiber array and multiplexer was designed to measure the spectrum of fast neutrons ranged 10 MeV-100 MeV. With the method of nuclear recoil, the energy of incident neutron was determined by measuring the recoil proton track and deposited energy in scintillating plastic fibers. The detection system was composed of a scintillating plastic fiber array, 6 position sensitive photomultiplier tubes, and a high-density readout electronics based on the multiplexer. The scintillating plastic fiber array was made as a staggered structure with two kinds of fibers in different sizes (0.5 mm-square fiber and 3 mm-square fiber). The structure provided a wider detection energy range and better detection efficiency than arrays made with uniform plastic fibers. A dedicated digital electronics system was well designed to control the whole readout system to provide 384-channel signal processing. The detector had a 48 mm × 48 mm effective detection area and a mechanical size of 34 cm × 34 cm × 27 cm. In the simulation of the detector model performance, the system gave an energy resolution of 23%-35% for neutrons ranged 10 MeV-100 MeV. Experimental results showed that the detector had a good energy linearity and energy resolutions were, respectively, 35.82% at 14.817 MeV, 36.84% at 21.264 MeV, 35.90% at 23.069 MeV, and 32.90% at 24.220 MeV. The optimized prototype model had potential in increasing fast neutron detection performance.

Abstract

Abstract: Recent findings demonstrate that leptin plays a significant role in chondrocyte and osteoblast differentiation. However, the mechanisms by which leptin acts on cartilage endplate (CEP) cells to give rise to calcification are still unclear. The aim of this study was to evaluate the effects of leptin that induced mineralization of CEP cells in vitro and in vivo. We constructed a rat model of lumbar disc degeneration and determined that leptin was highly expressed in the presence of CEP calcification. Rat CEP cells treated with or without leptin were used for in vitro analysis using RT‐PCR and Western blotting to examine the expression of osteocalcin (OCN) and runt‐related transcription factor 2 (Runx2). Both OCN and Runx2 expression levels were significantly increased in a dose‐ and time‐dependent manner. Leptin activated ERK1/2 and STAT3 phosphorylation in a time‐dependent manner. Inhibition of phosphorylated ERK1/2 using targeted siRNA suppressed leptin‐induced OCN and Runx2 expression and blocked the formation of mineralized nodules in CEP cells. We further demonstrated that exogenous leptin induced matrix mineralization of CEP cells in vivo. We suggest that leptin promotes the osteoblastic differentiation of CEP cells via the MAPK/ERK signal transduction pathway and may be used to investigate the mechanisms of disc degeneration. Abstract

Bioluminescence tomography (BLT) provides fundamental insight into biological processes in vivo. To fully realize its potential, it is important to develop image reconstruction algorithms that accurately visualize and quantify the bioluminescence signals taking advantage of limited boundary measurements. In this study, a new 2‐step reconstruction method for BLT is developed by taking advantage of the sparse a priori information of the light emission using multispectral measurements. The first step infers a wavelength‐dependent prior by using all multi‐wavelength measurements. The second step reconstructs the source distribution based on this developed prior. Simulation, phantom and in vivo results were performed to assess and compare the accuracy and the computational efficiency of this algorithm with conventional sparsity‐promoting BLT reconstruction algorithms, and results indicate that the position errors are reduced from a few millimeters down to submillimeter, and reconstruction time is reduced by 3 orders of magnitude in most cases, to just under a few seconds. The recovery of single objects and multiple (2 and 3) small objects is simulated, and the recovery of images of a mouse phantom and an experimental animal with an existing luminescent source in the abdomen is demonstrated. Abstract

An instrument used for measuring multiple scintillators' light output and energy resolution was developed. The instrument consisted of a light sensor array which was composed of 64 discrete SiPMs (Silicon Photomultipliers), a corresponding individual channel readout electronics system, and a data processing algorithm. A Teflon grid and a large interval between adjacent SiPMs were employed to eliminate the optical cross talk among scintillators. The scintillators' light output was obtained by comparing with a reference sample with known light output. Given the SiPM temperature dependency and the difference among each SiPM, a temperature offset correction algorithm and a non-uniformity correction algorithm were added to the instrument. A positioning algorithm, based on nine points, was designed to evaluate the performance of a scintillator array. Tests were performed to evaluate the instrument's performance. The uniformity of 64 channels for light output measurement was better than 98%, the stability was better than 98% when temperature varied from 15 °C to 40 °C, and the nonlinearity under 511 keV was better than 2%. This instrument was capable of selecting scintillators and evaluating the packaging technology of scintillator arrays with high efficiency and accuracy.

Abstract

Apolipoprotein A-1 (Apo A-I) Milano, a naturally occurring Arg173to Cys mutant of Apo A-1, has been shown to reduce atherosclerosis in animal models and in a small phase 2 human trial. We have shown the superior atheroprotective effects of Apo A-I Milano (Apo A-IM) gene compared to wild-type Apo A-I gene using transplantation of retrovirally transduced bone marrow in Apo A-I/Apo E null mice. In this study, we compared the effect of dietary lipid lowering versus lipid lowering plus Apo A-IM gene transfer using recombinant adeno-associated virus (rAAV) 8 as vectors on atherosclerosis regression in Apo A-I/Apo E null mice. All mice were fed a high-cholesterol diet from age of 6 weeks until week 20, and at 20 weeks, 10 mice were euthanized to determine the extent of atherosclerosis. After 20 weeks, an additional 20 mice were placed on either a low-cholesterol diet plus empty rAAV (n = 10) to serve as controls or low-cholesterol diet plus 1 single intravenous injection of 1.2 × 10(12)vector genomes of adeno-associated virus (AAV) 8 vectors expressing Apo A-IM (n = 10). At the 40 week time point, intravenous AAV8 Apo A-IM recipients showed a significant regression of atherosclerosis in the whole aorta (P< .01), aortic sinuses (P< .05), and brachiocephalic arteries (P< .05) compared to 20-week-old mice, whereas low-cholesterol diet plus empty vector control group showed no significant regression in lesion size. Immunostaining showed that compared to the 20-week-old mice, there was a significantly reduced macrophage content in the brachiocephalic (P< .05) and aortic sinus plaques (P< .05) of AAV8 Apo A-IM recipients. These data show that although dietary-mediated cholesterol lowering halts progression of atherosclerosis, it does not induce regression, whereas combination of low-cholesterol diet and AAV8 mediated Apo A-I Milano gene therapy induces rapid and significant regression of atherosclerosis in mice. 197. Use of green fluorescent protein as molecular marker for tagging Bacillus brevis in soil under the control of a novel constitutive promoter F1.

Abstract

A constitutive expression vector pHY300-F1gfp was constructed to test the function of promoter F1 subcloned from a rice epiphyte Bacillus brevis strain DX01. The DX01 cells harboring plasmid pHY300-F1gfp were detected to produce bright green fluorescence. Subsequently, the gfp-tagged B. brevis strain was released into the soil and its survival was investigated by PCR and the detection of green fluorescence. The spatial location of in situ gfp-tagged bacterial cells on the root surface of rice seedlings was visualized. All these results indicated that green fluorescent protein is an ideal molecular marker for the detection of the activities of promoter F1, and it is also a reliable probe to monitor specific B. brevis bacteria in the environment. The DX01 cells harboring the plasmid pHY300-F1gfp were shown to produce bright green fluorescence. The results were confirmed by Western blot analysis and fluorescence-activated cell sorting. Expression of the F1 promoter was constitutive. To improve the activity of F1, insertion mutagenesis of F1 based on in vitro transposition reaction was performed. 199. Vertically oriented TiO2 nanotube arrays with different anodization times for enhanced boiling heat transfer

Abstract

Abstract: Pool boiling of saturated water on a plain Ti surface and surfaces covered with vertically-oriented TiO2 nanotube arrays (NTAs) has been studied. The technique of potentiostatic anodization using non-aqueous electrolytes was adopted to fabricate three types of TiO2 NTAs distinguished by their anodization time. Compared to the bare Ti surface, the incipient boiling wall superheat on the TiO2 NTAs was decreased by 11 K. Both the critical heat flux and heat transfer coefficient of pool boiling on the TiO2 NTAs were higher than those from boiling on a bare Ti surface. The measured maximum critical heat flux and heat transfer coefficient values were 186.7 W/cm2 and 6.22 W/cm2K, respectively. Different performances for the enhancement of heat transfer by the three types of TiO2 NTAs were attributed to the different degrees of deformation in the nanostructure during boiling. Long-term performance of the nanomaterial-coated surfaces for enhanced pool boiling showed degradation of the TiO2 NTAs prepared with an anodization time of 3 hours.

200. Small supernumerary marker chromosome originating from chromosome 10 associated with an apparently normal phenotype

Abstract

Small supernumerary marker chromosomes sSMC originating from chromosome 10 are rare. Only seven cases have been documented, and among those three cases were diagnosed prenatally. We reported on another prenatal diagnosis of a de novo mosaic sSMC in an apparently normal female fetus whose mother had conceived with assisted reproductive technology ART procedures. Gbanding analysis of amniotic cells was performed. Spectral karyotyping SKY and fluorescence in situ hybridization FISH studies with chromosome 10specific alphoid satellite DNA probe were used to identify the chromosome 10 origin of the sSMC. Further FISH study with telomeric sequence probes showed that the sSMC lacked a hybridization signal, suggesting that the marker could be a ring chromosome. FISH studies using BAC clone probes specific for the regions within 10p11.2, 10q11.1, and 10q11.2 showed that the short arm breakpoint was located between 29.8 and 30.7 Mb from the 10p telomere, and that the long arm breakpoint was located less than 43.6 Mb from the 10p telomere. The karyotype of the fetus was 47,XX,mar. ish der10SKY CEP 10, CTD2130I7, RP1189J23−46,XX. Oligonucleotide microarraybased copy number variations CNV analysis was also performed and showed a 6.7 Mb duplication from 10p11.2 to 10q11.2 36.2–42.9 Mb with Affymetrix SNParray 6.0 genotype: arr cgh. 10p11.2q11.2CN519687 → CN541524 X 3. At the 1year followup, the baby did not have any findings of the trisomy 10p syndrome. This observation provided further credence to the concept that additional chromosome material of proximal 10p11.2 may not contribute to the trisomy 10p syndrome phenotype. © 2009 WileyLiss, Inc.
