As a recognized endocrine disruptor in the environment targeting estrogen receptors (ERs), Bisphenol A (BPA) and its bisphenol S (BPS) analogs are involved in the development of breast cancer. Epigenetic modifications are crucial in many biological processes, and DNA hydroxymethylation (DNAhm) coupled with histone methylation is implicated in epigenetic machinery covering cancer occurrence. Our previous study indicated that BPA/BPS induces breast cancer cell (BCC) proliferation with enhanced estrogenic transcriptional activity and causes the change of DNAhm depending on ten-eleven translocation 2 (TET2) dioxygenase. Herein, we investigated the interplay of KDM2A-mediated histone demethylation with ER-dependent estrogenic activity (EA) and identified their function in DNAhm catalyzed by TET2 for ER-positive (ER+) BCC proliferation induced by BPA/BPS. We found that BPA/BPS-treated ER+ BCCs presented increased KDM2A mRNA and protein levels but reduced TET2 and genomic DNAhm. Furthermore, KDM2A promoted H3K36me2 loss and suppressed TET2-dependent DNAhm by reducing its chromatin binding during BPA/BPS-induced cell proliferation. Results of Co-IP & ChIP assays suggested the direct interplay of KDM2A with ERα in multiple manners. KDM2A reduced the lysine methylation of ERα protein to increase its phosphorylated activation. On the other hand, ERα did not affect KDM2A expression, while KDM2A protein levels decreased after ERα deletion, indicating that ERα binding might maintain KDM2A protein stability. In conclusion, a potential feedback circuit of KDM2A/ERα-TET2-DNAhm was identified among ER+ BCCs with significant effects on regulating BPA/BPS-induced cell proliferation. These insights advanced the understanding of the relationship between histone methylation, DNAhm, and cancer cell proliferation with EA attributed to BPA/BPS exposure in the environment.

The Heilongjiang-Amur River Basin is one of the largest and most complex aquatic systems in Asia, comprising diverse wetland resources. The wetland vegetation in mid-high latitude areas has high natural value and is sensitive to climate changes. In this study, we investigated the wetland vegetation cover changes and associated responses to climate change in the Heilongjiang-Amur River Basin from 2000 to 2018 based on the growing season (May to September) climate and LAI data. Our results indicated that the wetland LAI increased at 0.014 m2·m-2/yr across Heilongjiang-Amur River Basin with the regional climate showed wetting and warming trends. On a regional scale, wetland vegetation in China and Russia had positive partial correlation with solar radiation and minimum air temperature, with precipitation showing a slight lag effect. In contrast, wetland vegetation in Mongolia had positive partial correlation with precipitation. These correlations were further investigated at different climate intervals. We found the precipitation is positively correlated with LAI in the warm regions while is negatively correlated with LAI in the wet regions, indicating an increase in precipitation is beneficial for the growth of wetland vegetation in heat sufficient areas, and when precipitation exceeds a certain threshold, it will hinder the growth of wetland vegetation. In the cold regions, we found solar radiation and minimum air temperature are positively correlated with LAI, suggesting SR and minimum air temperature instead of mean air temperature and maximum air temperature play more important roles in affecting the wetland vegetation growth in the heat limited areas. The LAI was found to be negatively correlated with maximum air temperature in the arid areas, indicating excessive temperature would inhibit the wetland vegetation growth when the water is limited. Our investigation can provide a scientific foundation for the trilateral region in wetland ecosystem protection and is beneficial for a more comprehensive understanding of the responses of wetlands in the middle and high latitudes to climate change.

Succinate is a vital signaling metabolite produced by the host and gut microbiota. Succinate has been shown to regulate host metabolic homeostasis and inhibit obesity-associated inflammation in macrophages by engaging its cognate receptor, SUCNR1. However, the contribution of the succinate-SUCNR1 axis to intestinal barrier dysfunction in obesity remains unclear. In the present study, we explored the effects of succinate-SUCNR1 signaling on high-fat diet (HFD)-induced intestinal barrier dysfunction. Using a SUCNR1-deficient mouse model under HFD feeding conditions, we identified the effects of succinate-SUCNR1 axis on obesity-associated intestinal barrier impairment. Our results showed that HFD administration decreased goblet cell numbers and mucus production, promoted intestinal pro-inflammatory responses, induced gut microbiota composition imbalance, increased intestinal permeability, and caused mucosal barrier dysfunction. Dietary succinate supplementation was sufficient to activate a type 2 immune response, trigger the differentiation of barrier-promoting goblet cells, suppress intestinal inflammation, restore HFD-induced mucosal barrier impairment and intestinal dysbiosis, and eventually exert anti-obesity effects. However, SUNNR1-deficient mice failed to improve the intestinal barrier function and metabolic phenotype in HFD mice. Our data indicate the protective role of the succinate-SUCNR1 axis in HFD-induced intestinal barrier dysfunction.

Functional magnetic resonance imaging (fMRI) during naturalistic viewing (NV) provides exciting opportunities for studying brain functions in more ecologically valid settings. Understanding individual differences in brain functions during NV and their behavioural relevance has recently become an important goal. However, methods specifically designed for this purpose remain limited. Here, we propose a topography-based predictive framework (TOPF) to fill this methodological gap. TOPF identifies individual-specific evoked activity topographies in a data-driven manner and examines their behavioural relevance using a machine learning-based predictive framework. We validate TOPF on both NV and task-based fMRI data from multiple conditions. Our results show that TOPF effectively and stably captures individual differences in evoked brain activity and successfully predicts phenotypes across cognition, emotion and personality on unseen subjects from their activity topographies. Moreover, TOPF compares favourably with functional connectivity-based approaches in prediction performance, with the identified predictive brain regions being neurobiologically interpretable. Crucially, we highlight the importance of examining individual evoked brain activity topographies in advancing our understanding of the brain-behaviour relationship. We believe that the TOPF approach provides a simple but powerful tool for understanding brain-behaviour relationships on an individual level with a strong potential for clinical applications.

Background: Ginseng consumption has been associated with various health outcomes. However, there are no review articles summarizing these reports.Methods: PubMed, Embase, the Cochrane Library of Systematic Reviews, Scopus, CNKI and Wanfang databases were searched from inception to 31 July 2022. The Assessment of Multiple Systematic Reviews-2 (AMSTAR-2) and Grading of Recommendations Assessment, Development and Evaluation (GRADE) systems were used to assess the methodological quality and quality of evidence in each meta-analysis, and the results were summarized in a narrative form.Results: Nineteen meta-analyses that met the eligibility criteria were identified from among 1,233 papers. The overall methodological quality was relatively poor, with only five studies being low-quality, and 14 critically low-quality. When compared with control treatments (mainly placebo), ginseng was beneficial for improving fatigue and physical function, sexual function, menopausal symptoms, metabolic indicators, inflammatory markers, unstable angina and respiratory diseases. Adverse events included gastrointestinal symptoms and potential bleeding; however, no serious adverse events were reported.Conclusion: This umbrella review suggests that ginseng intake has beneficial therapeutic effects for diverse diseases. However, the methodological quality of studies needs to be improved considerably. In addition, it is imperative to establish the clinical efficacy of ginseng through high-quality randomized controlled trials.

Objectives Homocysteine (Hcy) level has been widely identified as a risk factor associated with adverse outcomes in patients with lower extremity atherosclerotic disease (LEAD). However, there are still some knowledge gaps in research on the association between Hcy level and downstream adverse outcomes, such as length of stay (LOS). This study aims to explore whether and to what extent Hcy level is associated with LOS in patients with LEAD.Design Retrospective cohort study.Setting China.Participants, primary and secondary outcomes We conducted a retrospective cohort study of 748 patients from inpatients with LEAD between January 2014 and November 2021 at the First Hospital of China Medical University in China. We used a slew of generalised linear models to evaluate the association between Hcy level and LOS.Results The patients’ median age was 68 years and 631 (84.36%) were males. A dose–response curve with an inflection point at 22.63 µmol/L was observed between Hcy level and LOS after the adjustment of potential confounders. Tissue engineering (TE) cornea is one of the most potential alternatives to the shortage of corneal donors in cornea transplantation. Sodium alginate (SA) hydrogel is commonly used as scaffold in TE. Herein, we present an approach to construct a composite hydrogel, which with SA fiber skeleton structure for shape retention and gelatin surface modification for water retention. The light transmittance, water retention rate, and swelling rate of hydrogels were characterized, and the tensile mechanical properties were also investigated. Keratinocytes were treated with material extract liquor and the results showed that the gelatin modified SA hydrogel has good cytocompatibility. Furthermore, human corneal stromal fibroblasts (HCSFs) from the lenticules were implanted on the surface of gels, and the SA-gelatin hydrogel significantly improved the adhesion and spreading of HCSFs. Finally, we discussed the improvement and application prospect of the composite hydrogel as cornea equivalents.

Objectives: Jian-Pi-Yin decoction (JPY), a prescription derived from the traditional Chinese medicine Shen-Ling-Bai-Zhu-San, has shown good clinical efficacy in the treatment of diarrhea caused by lactose intolerance. However, the mechanism of action of JPY in the treatment of diarrhea is not fully understood. Design: In this study, a rat diarrhea model was induced by high lactose feeding combined with standing on a small platform to investigate the ameliorating effect of JPY on hyper lactose-induced diarrhea in rats and its possible mechanism. Methods: The rat model of hyper lactose diarrhea was given high, medium, and low doses of JPY and the positive control drug Smida by gavage for 1 week. At the same time, NA+-H+ exchanger 3 (NHE3) inhibitor Tenapanor was administered orally for 3 weeks. Body weight, food intake, water intake, grip strength, and severity of diarrhea symptoms were measured in rats throughout the study. The serum, colon, and jejunum tissues of the model and drug-treated rats were collected for histopathological examination and analysis of relevant indicators. Results: JPY significantly alleviated the symptoms of fatigue, diet reduction and diarrhea in the model group. Glucagon-like peptide-1 (GLP-1) and cyclic adenosine monophosphate (cAMP) expression were also down-regulated after JPY treatment. JPY can significantly promote NHE3 in intestinal tissues of rats with diarrhea, and the mechanism is related to the decrease of GLP-1, inhibition of cAMP/PKA pathway activation, an increase of ubiquitin-specific protease 7 (USP7) and USP10 expression, and decrease of NHE3 ubiquitination and phosphorylation. Conclusion: JPY can reduce the expression of GLP-1, reduce the ubiquitination and phosphorylation of NHE3, regulate the expression of NHE3, at least partly improve ion transport in the intestinal epithelium, and improve the imbalance of electrolyte absorption, thus significantly reducing the diarrhea symptoms of rats with high lactose combined with small platform standing. Innovation: In this study, we explored the mechanism of intestinal GLP-1 activation of cAMP/PKA signaling pathway from multiple dimensions, and increased its expression by reducing phosphorylation and ubiquitination of NHE3, thereby treating chronic diarrhea associated with lactose intolerance.

Abstract Background Kawasaki disease (KD) is an acute febrile illness of unknown etiology and predictors for intravenous immunoglobulin (IVIG) resistance have been widely explored in recent decades. Neutrophil to lymphocyte platelet ratio (NLPR) was reported to be associated with the outcomes in many diseases. However, its relationship with IVIG resistance has not be explored. Methods The medical data of patients diagnosed with KD in Children’s Hospital of Soochow University between January 2019 and December 2020 were retrospectively reviewed and analyzed. Patients were trisected into three groups based on NLPR. Logistics regression was used to analyze the association between NLPR and IVIG resistance. Restricted cubic spine was used to exhibit the relationship. Sensitivity analysis and subgroup analysis were also carried out. Results A total of 803 patients were included in the present study (61.8% males; median age: 24 months). IVIG resistance occurred in 74 (9.2%) patients. Multivariable-adjusted analyses revealed higher NLPR (odds ratio [95% confidence interval]: 1.12 [1.00-1.24]) was an independent predictor of IVIG resistance, which was strengthened by sensitivity analyses. The association of NLPR and IVIG resistance was not modified by age, sex, CALs, or days of IVIG initiation ≤ 4. Conclusion NLPR may be a valuable prognostic marker in KD patients with IVIG resistance.

Abstract Periodontal tissue is a highly dynamic and frequently stimulated area where homeostasis is easily destroyed, leading to proinflammatory periodontal diseases. Bacteria–bacteria and cell–bacteria interactions play pivotal roles in periodontal homeostasis and disease progression. Several reviews have comprehensively summarized the roles of bacteria and stem cells in periodontal homeostasis. However, they did not describe the roles of extracellular vesicles (EVs) from bacteria and cells. As communication mediators evolutionarily conserved from bacteria to eukaryotic cells, EVs secreted by bacteria or cells can mediate interactions between bacteria and their hosts, thereby offering great promise for the maintenance of periodontal homeostasis. This review offers an overview of EV biogenesis, the effects of EVs on periodontal homeostasis, and recent advances in EV-based periodontal regenerative strategies. Specifically, we document the pathogenic roles of bacteria-derived EVs (BEVs) in periodontal dyshomeostasis, focusing on plaque biofilm formation, immune evasion, inflammatory pathway activation and tissue destruction. Moreover, we summarize recent advancements in cell-derived EVs (CEVs) in periodontal homeostasis, emphasizing the multifunctional biological effects of CEVs on periodontal tissue regeneration. Finally, we discuss future challenges and practical perspectives for the clinical translation of EV-based therapies for periodontitis. Graphical Abstract

Abstract Real-time Doppler estimation of sonar signals is of great significance to underwater communication and detection. The Hyperbolic-Frequency-Modulated (HFM) waveform-based speed spectrum estimation is one of the modern methods for underwater Doppler estimation, and this paper presents a detailed analyses of the derivation and the performance of this method. The analyzing results show the robustness of the method on multipath channels and multi-highlight targets and provide the main lobe width of the speed spectrum. Moreover, the paper examines the spectral properties of the HFM signal from the frequency-domain Doppler invariance property, which could provide insights for better understanding and utilization of this Doppler invariant waveform.

AbstractFlowering transition is important in plant growth and development, and the SUPPRESSOR OF OVEREXPRESSION OF CO 1 (SOC1) gene belongs to the MADS-box gene family, which is an integrator involved in the regulation of flower formation in plants. Current studies have shown that the structural domain of SOC1 is relatively conserved in a variety of plants. To further investigate the function of the SOC1 homologous gene in pak choi, the full-length coding sequence of pak choi BrcSOC1 was cloned, and the structure, subcellular localization and spatiotemporal expression pattern of BrcSOC1 were analyzed in this study. The tissue-specific expression results showed that BrcSOC1 was expressed in roots, leaves, bolts, flowers and pods. The expression results at different developmental stages showed that BrcSOC1 was expressed at the highest level at floral bud differentiation stage 5. When BrcSOC1 was overexpressed in Arabidopsis thaliana, the flowering time in all transgenic plants was significantly earlier than in the wild type, and the expression analysis indicated that BrcSOC1 positively regulated the expression of the downstream genes AGAMOUS-LIKE 24 (AGL24) and LEAFY (LFY). These results provide an important reference for further research on the mechanism of BrcSOC1 in the flowering pathway of pak choi.

Background: Hydrogen sulfide (H2S) is a small reducing gas molecule with various biological functions such as anti-oxidative, anti-apoptotic and anti-inflammatory activities. In this study, we investigated the therapeutic effects of exogenous H2S in the experimental models of retinal photodamage in vivo and in vitro.Methods: Rats with open eyelids were pretreated with H2S (80~120 μmol/kg) for 10 days and then continuously exposed to blue light (435~445nm, 11.2W/m2) for 8 h to establish in vivo experimental model. ARPE-19 cells were pretreated with H2S and then exposed to blue light to establish in vitro experimental model.Results: In vivo experiments, H2S significantly ameliorated blue light-induced retinal oxidative stress, apoptosis and degeneration. Moreover, H2S inhibited the activation of blue light-induced endoplasmic reticulum (ER) stress CHOP apoptotic signaling. In vitro experiments, H2S improved blue light-induced oxidative stress and oxidative damage. H2S inhibited ROS-mediated activation of ER stress CHOP apoptotic signaling. H2S alleviated blue light-induced apoptosis and increases cell viability. The ER stress inhibitor 4-PBA alleviated blue light-induced apoptosis and increases cell viability.Conclusion: Taken together, these results indicate that H2S can inhibit ROS-mediated ER stress-CHOP apoptosis signal, thereby alleviating blue light-triggered retinal apoptosis and degeneration.

Objectives: Lycium barbarum polysaccharide (LBP) is a natural polysaccharide extracted from Lycium barbarum that has anti-inflammatory, anti-apoptotic and anti-aging effects, and plays a role in the prevention and treatment of various diseases. In this study, we investigated the therapeutic effect of LBP on particulate matter 2.5 (PM2.5)-induced skin damage. Methods: Cell viability was analyzed by MTT and LDH assays. Apoptosis was analyzed by Annexin V-FITC/PI staining. Oxidative stress/damage were assessed by intracellular ROS levels, MDA content and SOD activity. The intracellular protein expression was analyzed by Western blot. Mitochondrial damage was assayed by mitochondrial membrane potential with JC-1 probe. LC3-GFP adenovirus was transfected into HaCaT cells to analyze intracellular autophagosome levels. Results: In PM2.5-treated HaCaT cells, LBP pretreatment reduced PM2.5-induced cytotoxicity, ameliorated cell morphology and reduced cell apoptosis. LBP also inhibited the expression levels of GRP78 and CHOP, reduced the conversion of LC3I to LC3II, inhibited Bax protein and activated Bcl-2 protein. Furthermore, LBP inhibited PM2.5-induced mitochondrial autophagy (mitophagy) and mitochondrial damage. PM2.5-induced autophagy was regulated by endoplasmic reticulum (ER) stress. Conclusion: LBP protects skin cells from PM2.5-induced cytotoxicity by regulating the oxidative stress-ER stress-autophagy-apoptosis signaling axis, revealing that LBP has a great potential for the skin protection.

Insufficient osseointegration and biofilm-associated bacterial infection are important challenges for clinical application of titanium (Ti)-based implants. Here, we constructed mesoporous polydopamine (MPDA) nanoparticles (NPs) loaded with luteolin (LUT, a quorum sensing inhibitor), which were further coated with the shell of calcium phosphate (CaP) to construct MPDA-LUT@CaP nanosystem. Then, MPDA-LUT@CaP NPs were immobilized on the surface of Ti implants. Under acidic environment of bacterial biofilm-infection, the CaP shell of MPDA-LUT@CaP NPs was rapidly degraded and released LUT, Ca2+ and PO43− from the surface of Ti implant. LUT could effectively inhibit and disperse biofilm. Furthermore, under near-infrared irradiation (NIR), the thermotherapy induced by the photothermal conversion effect of MPDA destroyed the integrity of the bacterial membrane, and synergistically led to protein leakage and a decrease in ATP levels. Combined with photothermal therapy (PTT) and quorum-sensing-inhibition strategy, the surface-functionalized Ti substrate had an antibacterial rate of over 95.59% against Staphylococcus aureus and the elimination rate of the formed biofilm was as high as 90.3%, so as to achieve low temperature and efficient treatment of bacterial biofilm infection. More importantly, the modified Ti implant accelerated the growth of cell and the healing process of bone tissue due to the released Ca2+ and PO43−. In summary, this work combined PTT with quorum-sensing-inhibition strategy provides a new idea for surface functionalization of implant for achieving effective antibacterial and osseointegration capabilities.

Context Polyphyllin II (PPII) is a steroidal saponin isolated from Rhizoma Paridis. It exhibits significant antitumor activity such as anti-proliferation and pro-apoptosis in lung cancer.Objective To explore whether PPII induce autophagy and the relationship between autophagy and apoptosis in non-small cell lung cancer (NSCLC) cells.Materials and methods The effects of PPII (0, 1, 5, and 10 μM) were elucidated by CCK8 assay, colony formation test, TUNEL staining, MDC method, and mRFP-GFP-LC3 lentivirus transfection in A549 and H1299 cells for 24 h. DMSO-treated cells were selected as control. The protein expression of autophagy (LC3-II, p62), apoptosis (Bcl-2, Bax, caspase-3) and p-mTOR was detected by Western blotting. We explored the relationship between autophagy and apoptosis by autophagy inhibitor CQ (10 μM) and 3-MA (5 mM).Results PPII (0, 1, 5, and 10 μM) inhibited the proliferation and induced apoptosis. The IC50 values of A549 and H1299 cells were 8.26 ± 0.03 and 2.86 ± 0.83 μM. We found that PPII could induce autophagy. AbstractPak choi (Brassica rapa ssp. chinensis Makino) is a typical seed vernalization vegetable. Because premature bolting in spring causes significant economic losses, it is very important to clarify the flower formation mechanism to prevent this. Cytokinins are important plant hormones involved in regulating plant growth and development. To understand the relationship between cytokinin metabolism and flowering of pak choi, in this study, we determined the cytokinin trans-zeatin content in shoot apices of pak choi at different developmental stages by enzyme-linked immunosorbent assay. The results showed that cytokinin levels increased significantly after low temperature treatment at 4 °C, and continued to increase with vegetative growth after transplanting, reaching a peak at the critical period which is immediately prior to flower buds’ differentiation (S0), then decreased thereafter. The levels for low temperature treatment were consistently higher than controls. To explore the molecular mechanism underpinning the cytokinin changes, expression of homologous genes encoding cytokinin metabolic enzymes was analysed by transcriptome sequencing. The expression levels of nine genes (Bra004037, Bra023701, Bra002204, Bra014968, Bra028326, Bra028182, Bra034022, Bra009143 and Bra005869) were consistent with the changes in cytokinin content. The correlation between differentially expressed genes and cytokinin content in different developmental stages of apexes was analysed, and six closely related genes (Bra023701, Bra002204, Bra014968, Bra028182, Bra005869 and Bra009143) were identified. The results help to illuminate the molecular mechanisms controlling flowering of pak choi.

The flexible DC grid has the capability to transport and distribute electric energy in multiterminal power system, and avoid the adverse impact on AC power grid in reactive power consumption and current harmonic. Unfortunately, because of the discharge of submodule capacitor facing the short-circuit fault of the overhead line, The overcurrent problem caused by the fault restricts the further development of the DC grid. At present, the research on short-circuit fault of flexible DC grid is mainly based on computer software simulation without mathematical expression to describe failure process quantitatively. This paper proposes a short-circuit fault current calculation method describing flexible DC grid fault process. Firstly, the linearized equivalent model of the MMC converter station is obtained by considering the discharge characteristics of the submodule capacitors. Secondly, considering the DC grid space topology, the parsed expression of overcurrent in short circuit fault is derived. Finally, the analysis results of the paper are compared with the software simulation results of the PSCAD/EMTDC platform to verify the validity and accuracy of the overcurrent algorithm.

To address the problem of heavy metal contamination of lead smelting slag such as As, Zn, Pb and Cd in the Sishuizhuang area of Gejiu City, Yunnan Province, oily sludge pyrolysis residue was used to stabilize them to reduce the leaching toxicity of these heavy metals. The pyrolysis residue is a carbonaceous composite containing a large amount of nano-FeS, Fe1-xS, CaS, etc., which has the potential to stabilize the heavy metals in lead slag. It was shown that heavy metals As, Zn, Pb and Cd in lead slag could be efficiently and synchronously adsorbed on the surface of pyrolysis residue, reducing the leaching toxicity of these heavy metals in lead slag. When the weight ratio of pyrolysis residueis is 4% and liquid-solid ratio is 10:100, the leaching concentrations of As, Zn, Pb and Cd in the lead slag can be reduced from 0.7202, 3.5120, 0.3800 and 0.0456 mg/L to 0.0714, 0.1668, 0.0262 and 0.0038 mg/L, respectively, which are lower than the surface water environmental quality standards (GB 3838—2002 ) in the Ⅳ level limit value. The reduction of leaching concentration of As, Zn, Pb and Cd in lead slag is due to the reaction of As in lead slag with Ca2+ and Fe2+ on the surface of pyrolysis slag to produce stable calcium arsenate and ferrous arsenate in situ, and the reaction of Zn2+, Pb2+ and Cd2+ in lead slag with S2- on the surface of pyrolysis slag to produce stable ZnS, PbS and CdS in situ, which improves the stability of As, Zn, Pb and Cd in lead slag.

Short-chain fatty acids (SCFA) can regulate appetite by stimulating the secretion of satiety hormones. However, the impact of short-chain fatty acid propionate on the release of gut satiety hormones and appetite regulation in pigs is not completely understood. In this study, 16 pigs were infused with saline or sodium propionate through a fistula in the caecum during a 28-day experimental period. We characterized the effects of propionate administration on peptide YY (PYY) and glucagon-like peptide 1 (GLP-1) secretion from colonic tissue, and investigated the role of propionate infusion on the expression of appetite-related genes in the colon and hypothalamus. Abstract The positive regulation of bone-forming osteoblast activity and the negative feedback regulation of osteoclastic activity are equally important in strategies to achieve successful alveolar bone regeneration. Here, a molybdenum (Mo)-containing bioactive glass ceramic scaffold with solid-strut-packed structures (Mo-scaffold) was printed, and its ability to regulate pro-osteogenic and anti-osteoclastogenic cellular responses was evaluated in vitro and in vivo. We found that extracts derived from Mo-scaffold (Mo-extracts) strongly stimulated osteogenic differentiation of bone marrow mesenchymal stem cells and inhibited differentiation of osteoclast progenitors. The identified comodulatory effect was further demonstrated to arise from Mo ions in the Mo-extract, wherein Mo ions suppressed osteoclastic differentiation by scavenging reactive oxygen species (ROS) and inhibiting mitochondrial biogenesis in osteoclasts. Consistent with the in vitro findings, the Mo-scaffold was found to significantly promote osteoblast-mediated bone formation and inhibit osteoclast-mediated bone resorption throughout the bone healing process, leading to enhanced bone regeneration. In combination with our previous finding that Mo ions participate in material-mediated immunomodulation, this study offers the new insight that Mo ions facilitate bone repair by comodulating the balance between bone formation and resorption. Our findings suggest that Mo ions are multifunctional cellular modulators that can potentially be used in biomaterial design and bone tissue engineering.

Genome-scale metabolomics analysis is increasingly used for pathway and function discovery in the post-genomics era. The great potential offered by developed mass spectrometry (MS)-based technologies has been hindered, since only a small portion of detected metabolites were identifiable so far. To address the critical issue of low identification coverage in metabolomics, we adopted a deep metabolomics analysis strategy by integrating advanced algorithms and expanded reference databases. The experimental reference spectra and in silico reference spectra were adopted to facilitate the structural annotation. To further characterize the structure of metabolites, two approaches were incorporated into our strategy, i.e., structural motif search combined with neutral loss scanning and metabolite association network. Untargeted metabolomics analysis was performed on 150 rice cultivars using ultra-performance liquid chromatography coupled with quadrupole-Orbitrap MS. Consequently, a total of 1939 out of 4491 metabolite features in the MS/MS spectral tag (MS2T) library were annotated, representing an extension of annotation coverage by an order of magnitude in rice. The differential accumulation patterns of flavonoids between indica and japonica cultivars were revealed, especially O-sulfated flavonoids. A series of closely-related flavonolignans were characterized, adding further evidence for the crucial role of tricin-oligolignols in lignification. Our study provides an important protocol for exploring phytochemical diversity in other plant species.

Abstract The transformer architecture has been the dominant framework for today's image captioning tasks because of its superior performance. However, existing methods based on transformer often lack the integrated use of multi‐level semantic information and are weak in maintaining the relevance of captions to the image. In this paper, a semantic‐meshed and content‐guided transformer network is introduced for image captioning to solve these problems. The semantic‐meshed mechanism allows the model to generate words by selecting semantic information of multiple interaction levels adaptively through attention‐based reconstruction. And the content‐guided module guides the words generation by using attribute features that represent the image content, which aims to keep the generated caption consistent with the main content of the image. Experiments on dataset on the MSCOCO captioning dataset are conducted to validate the authors’ model and achieve superior results compared to other state‐of‐the‐art method approaches.

Abstract For patients with platinum-resistant lung adenocarcinoma (LUAD), the exploration of new effective drug candidates is urgently needed. Fibroblast growth factor receptors (FGFRs) have been identified as promising targets for LUAD therapy. The purpose of this study was to determine the exact role of the irreversible FGFR1-4 inhibitor FIIN-2 in LUAD and to clarify its underlying molecular mechanisms. Our results demonstrated that FIIN-2 significantly inhibited the proliferation, colony formation, and migration of A549 and A549/DDP cells but induced the mitochondria-mediated apoptosis of these cells. Meanwhile, FIIN-2 increased the autophagy flux of A549 and A549/DDP cells by inhibiting the mammalian target of rapamycin (mTOR) and further activating the class III PI3K complex pathway. More importantly, in vivo and in vitro experiments showed that autophagy inhibitors could enhance the cytotoxicity of FIIN-2 on A549 and A549/DDP cells, confirming that FIIN-2 induced protective autophagy. These findings indicated that FIIN-2 is a potential drug candidate for LUAD treatment, and its use in combination with autophagy inhibitors might be an efficient treatment strategy, especially for patients with cisplatin resistance.

In this research, a new series of β duplex phase Ti50Zr25Nb25-xAgx (x = 0, 1, 3 and 5 at. %) alloys were prepared by rapid solidification process. The microstructure, mechanical properties, antibacterial properties, ion release behavior and biocompatibility of the alloys were systematically investigated. The alloys were composed of β main phase and β minor phase, therefor the alloys exhibited a very low Young's modulus of 60-61 GPa, which can effectively reduce the stress shielding effect. The alloys possessed high strength and hardness, which is correlated to the grain refinement strengthening caused by rapid solidification process and solid solution strengthening caused by the addition of Ag. The release of Ag ion increased with the increasing of Ag content. The antibacterial properties also showed the same trend and the Ag5 alloy exhibited 99.1% antibacterial rate. Meanwhile, the alloys possessed good biocompatibilities obtained through cell live and dead staining experiments and cell proliferation experiments.

Abstract Background High glucose-induced damage to the osteogenic differentiation of human periodontal ligament stem cells (PDLSCs) has long been a challenge to periodontal regeneration for diabetic individuals. Metformin is an anti-hyperglycemic drug that exhibits abundant biological activities associated with cell metabolism and downstream tissue regeneration. However, how metformin combats damage to PDLSC osteogenic differentiation under high glucose and the underlying mechanisms remain unknown. Methods Osteogenic differentiation of PDLSCs was assessed by alkaline phosphatase (ALP) staining, ALP activity, Alizarin Red staining and quantitative assay, quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot analysis. RNA-seq analysis was performed to screen target genes of metformin, and the effects of target genes were confirmed using lentivirus transfection. Western blot analysis was also used to detect the protein level of underlying signaling pathways. Results We found that osteogenic differentiation of PDLSCs under high glucose was decreased, and metformin addition enhanced this capacity of differentiation. Furthermore, the results of RNA-seq analysis showed that natriuretic peptide receptor 3 (NPR3) was upregulated in PDLSCs under high glucose and downregulated after metformin addition. When the underlying pathways involved were investigated, we found that upregulation of NPR3 can compromise the metformin-enhanced PDLSC osteogenic differentiation and activate the MAPK pathway (especially the p38 MAPK and Erk1/2 pathway), and that inhibition of the NPR3-mediated p38 MAPK or Erk1/2 pathway enhanced the osteogenic differentiation of PDLSCs under high glucose. Conclusions The present study suggests that metformin may enhance the osteogenic differentiation of PDLSCs under high glucose via downregulation of NPR3 and inhibition of its downstream MAPK pathway. This is the first report identifying the involvement of NPR3-mediated MAPK pathway in the metformin-enhanced osteogenic differentiation, indicating that NPR3 antagonists, such as metformin, may be feasible therapeutics for periodontal tissue regeneration in diabetic individuals.

Hydropower provides important support for achieving dual-carbon goal, but problems such as reservoir resettlement conflicts are common in developing countries, which have become a bottleneck of sustainable hydropower development. Fair distribution of benefits is the key to solve this dilemma. While the concept of benefit sharing as the fundamental guarantee for the sustainable livelihood of landless resettlers is gradually reaching consensus, the amount of benefit distribution has always been an outstanding issue. In order to quantify the amount of benefit sharing scientifically and reasonably, this paper constructs a benefit sharing model of hydropower development from the perspective of stakeholder input, and carries out a case analysis. The results indicate that the main stakeholders of hydropower projects include governments, development enterprises and reservoir resettlers. Their inputs are water or environmental resources, capital, and land resources, which produce hydropower resources. The net present value method can be used to construct the benefit sharing model. In this model, sharing amount of resettlers and other major stakeholders is the product of the sum of the present value of the net cash flow of hydropower projects and the proportion of their respective input. In view of many uncertainties, the grey prediction method and Monte Carlo simulation method are used to solve the model. The case of the Xiluodu hydropower station in China proves the rationality and operability of the model. The study also shows with reference to the model’s calculation results, a shared development fund can be established by drawing money from electricity generation income to ensure long-term benefits for resettlers, while considering the reasonable benefit of other stakeholders. The two are connected. It is expected to achieve a new pattern of mutual benefit and multi-win outcome, which are characterized by stable power station revenue, prosperity and stability of the reservoir area, and increased benefit and common wealth of resettlers. This paper may provide a convenient and novel analysis tool for benefit sharing systems.

Diabetes mellitus is an established risk factor for periodontal disease that can aggravate the severity of periodontal inflammation and accelerate periodontal destruction. The chronic high glucose condition is a hallmark of diabetes-related pathogenesis, and has been demonstrated to impair the osteogenic differentiation of periodontal ligament stem cells (PDLSCs), leading to delayed recovery of periodontal defects in diabetic patients. Reactive oxygen species (ROS) are small molecules that can influence cell fate determination and the direction of cell differentiation. Although excessive accumulation of ROS has been found to be associated with high glucose-induced cell damage, the underlying mechanisms remain unclear. Nicotinamide adenine dinucleotide phosphate (NADPH) is an important electron donor and functions as a critical ROS scavenger in antioxidant systems. It has been identified as a key mediator of various biological processes, including energy metabolism and cell differentiation. However, whether NADPH is involved in the dysregulation of ROS and further compromise of PDLSC osteogenic differentiation under high glucose conditions is still not known. In the present study, we found that PDLSCs incubated under high glucose conditions showed impaired osteogenic differentiation, excessive ROS accumulation and increased NADPH production. Furthermore, after inhibiting the synthesis of NADPH, the osteogenic differentiation of PDLSCs was significantly enhanced, accompanied by reduced cellular ROS accumulation. Our findings demonstrated the crucial role of NADPH in regulating cellular osteogenic differentiation under high glucose conditions and suggested a new target for rescuing high glucose-induced cell dysfunction and promoting tissue regeneration in the future.

Naturalistic viewing (NV) is currently considered a promising paradigm for studying individual differences in functional brain organization. While whole brain functional connectivity (FC) under NV has been relatively well characterized, so far little work has been done on a network level.Here, we extend current knowledge by characterizing the influence of NV on FC in fourteen meta-analytically derived brain networks considering three different movie stimuli in comparison to resting-state (RS). We show that NV increases identifiability of individuals over RS based on functional connectivity in certain, but not all networks. Furthermore, movie stimuli including a narrative appear more distinct from RS. In addition, we assess individual variability in network FC by comparing within- and between-subject similarity during NV and RS. We show that NV can evoke individually distinct NFC patterns by increasing inter-subject variability while retaining within-subject similarity. Crucially, our results highlight that this effect is not observable across all networks, but rather dependent on the network-stimulus combination. Our results confirm that NV can improve the detection of individual differences over RS and underline the importance of selecting the appropriate combination of movie and cognitive network for the research question at hand.

Polarization three-dimensional (3D) imaging technology has received extensive attention in recent years because of its advantages of high accuracy, long detection distance, simplicity, and low cost. The ambiguity in the normal obtained by the polarization characteristics of the target’s specular or diffuse reflected light limits the development of polarization 3D imaging technology. Over the past few decades, many shape from polarization techniques have been proposed to address the ambiguity issues, i.e., high-precision normal acquisition. Meanwhile, some polarization 3D imaging techniques attempt to extend experimental objects to complex specific targets and scenarios through a learning-based approach. Additionally, other problems and related solutions in polarization 3D imaging technology are also investigated. In this paper, the fundamental principles behind these technologies will be elucidated, experimental results will be presented to demonstrate the capabilities and limitations of these popular technologies, and finally, our perspectives on the remaining challenges of the polarization 3D imaging technology will be presented.

BackgroundSexually transmitted diseases (STDs) are a serious issue worldwide. With the popularity of the internet, online health information-seeking behavior (OHISB) has been widely adopted to improve health and prevent disease. ObjectiveThis study aimed to investigate the short-term and long-term effects of different types of OHISBs on STDs, including syphilis, gonorrhea, and AIDS due to HIV, based on the Baidu index. MethodsMultisource big data were collected, including case numbers of STDs, search queries based on the Baidu index, provincial total population, male-female ratio, the proportion of the population older than 65 years, gross regional domestic product (GRDP), and health institution number data in 2011-2018 in mainland China. We categorized OHISBs into 4 types: concept, symptoms, treatment, and prevention. Before and after controlling for socioeconomic and medical conditions, we applied multiple linear regression to analyze associations between the Baidu search index (BSI) and Baidu search rate (BSR) and STD case numbers. In addition, we compared the effects of 4 types of OHISBs and performed time lag cross-correlation analyses to investigate the long-term effect of OHISB. ResultsThe distributions of both STD case numbers and OHISBs presented variability. For case number, syphilis, and gonorrhea, cases were mainly distributed in southeastern and northwestern areas of China, while HIV/AIDS cases were mostly distributed in southwestern areas. For the search query, the eastern region had the highest BSI and BSR, while the western region had the lowest ones. Scanning transmission electron microscope and temperature-dependent photoluminescence were used to study the effects of AlGaN interlayer (IL) following InGaN quantum wells (QWs) on the structural and optical properties of InGaN green laser diodes (LDs) grown on c-plane GaN substrate. It is found that AlGaN IL improves InGaN QW interface sharpness, whereas it does not suppress the localization effect, and moreover, it leads to increasing nonradiative recombination centers in the active region of green InGaN LD. We believe AlGaN IL has advantages and disadvantages to green InGaN LD structures. The advantage is to suppress indium (In) surface segregation, but the disadvantage is that it covers In-rich clusters, which evolve into dark spots during the growth of p-type layers of green LD structures.

ObjectiveThe aim of this study is to compare the efficacy and safety of complete multi-level vs. iliac-only revascularization for the treatment of concomitant iliac and superficial femoral artery (SFA) occlusive disease.MethodsA total of 139 consecutive adult patients with severe stenosis and occlusive iliac and SFA disease with Rutherford categories 2–5 underwent multi-level (n = 71) and iliac-only (n = 68) revascularization at the Department of Intervention Vascular Surgery, Peking University Third Hospital, and Aerospace Center Hospital, between March 2015 and June 2017. Improvement in Rutherford class, perioperative major adverse events, the length of stay, survival rate, and limb salvage rate were assessed. The neutrophil–lymphocyte ratio and platelet–lymphocyte ratio were compared between the two groups.ResultsAt 48 months, improvement in the Rutherford category was observed in the two groups with no significant difference (P = 0.809). Additionally, the two groups were similar concerning the primary patency (84.0% vs. 79.1%, P = 0.717) and limb salvage rate (93.1% vs. 91.3%, P = 0.781). A higher proportion of the perioperative major adverse events (33.8% vs. 27.9%, P = 0.455), the all-cause mortality (11.3% vs. 8.8%, P = 0.632), and the average length of hospital stay [7.0 (6.0, 11.0) vs. 7.0 (5.0, 8.0), P = 0.037] were seen in the multi-level group compared with the iliac-only group.ConclusionFor concomitant iliac and superficial femoral artery occlusive disease, iliac-only revascularization has favorable efficacy and safety outcomes compared with complete multi-level revascularization in selected patients with patent profunda femoris artery and at least one healthy outflow tract of the infrapopliteal artery.

BackgroundPrevious studies have shown that systemic inflammation indicators could predict the survival outcomes of patients with malignant tumors receiving various treatments. Radiotherapy, as a crucial treatment modality, effectively alleviates discomfort in patients with bone metastasis (BM) and greatly improves the quality of life for them. This study aimed to investigate the prognostic value of systemic inflammation index in hepatocellular carcinoma (HCC) patients with BM treated with radiotherapy.MethodsWe retrospectively analyzed clinical data collected from HCC patients with BM who received radiotherapy in our institution between January 2017 and December 2021. The pre-treatment neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and systemic immune-inflammation index (SII) were derived to determine their relationship with overall survival (OS) and progression-free survival (PFS), using the Kaplan-Meier survival curves. The optimal cut-off value of the systemic inflammation indicators for predicting prognosis was assessed by receiver operating characteristic (ROC) curves. Univariate and multivariate analyses were performed to ultimately evaluate the factors associated with survival.ResultsThe study included 239 patients with a median 14-month follow-up. The median OS was 18 months (95% confidence interval [CI] = 12.0-24.0) and the median PFS was 8.5 months (95% CI = 6.5-9.5). The optimal cut-off values for the patients were determined by ROC curve analysis as follows: SII =395.05, NLR=5.43 and PLR = 108.23. The area under the receiver operating characteristic curve values for SII, NLR and PLR in disease control prediction were 0.750, 0.665 and 0.676, respectively. Elevated systemic immune-inflammation index (SII>395.05) and higher NLR (NLR>5.43) were independently associated with poor OS and PFS. In multivariate analysis, Child-Pugh class (P = 0.038), intrahepatic tumor controlled (P = 0.019), SII (P = 0.001) and NLR (P = 0.007) were independent prognostic factors of OS and Child-Pugh class (P = 0.042), SII (P < 0.001) and NLR (P = 0.002) were independently correlated with PFS.ConclusionNLR and SII were associated with poor prognosis in HCC patients with BM receiving radiotherapy and might be considered reliable and independent prognostic biomarkers for HCC patients with BM.

IntroductionMetabolic reprogramming from glycolysis to the mitochondrial tricarboxylic acid (TCA) cycle and oxidative phosphorylation may mediate macrophage polarization from the pro-inflammatory M1 to the anti-inflammatory M2 phenotype. We hypothesized that changes in cardiac macrophage glucose metabolism would reflect polarization status after myocardial infarction (MI), ranging from the early inflammatory phase to the later wound healing phase.MethodsMI was induced by permanent ligation of the left coronary artery in adult male C57BL/6J mice for 1 (D1), 3 (D3), or 7 (D7) days. Infarct macrophages were subjected to metabolic flux analysis or gene expression analysis. Monocyte versus resident cardiac macrophage metabolism was assessed using mice lacking the Ccr2 gene (CCR2 KO).ResultsBy flow cytometry and RT-PCR, D1 macrophages exhibited an M1 phenotype while D7 macrophages exhibited an M2 phenotype. Macrophage glycolysis (extracellular acidification rate) was increased at D1 and D3, returning to basal levels at D7. Glucose oxidation (oxygen consumption rate) was decreased at D3, returning to basal levels at D7. At D1, glycolytic genes were elevated (Gapdh, Ldha, Pkm2), while TCA cycle genes were elevated at D3 (Idh1 and Idh2) and D7 (Pdha1, Idh1/2, Sdha/b). Surprisingly, Slc2a1 and Hk1/2 were increased at D7, as well as pentose phosphate pathway (PPP) genes (G6pdx, G6pd2, Pgd, Rpia, Taldo1), indicating increased PPP activity. Macrophages from CCR2 KO mice showed decreased glycolysis and increased glucose oxidation at D3, and decreases in Ldha and Pkm2 expression. Administration of dichloroacetate, a pyruvate dehydrogenase kinase inhibitor, robustly decreased pyruvate dehydrogenase phosphorylation in the non-infarcted remote zone, but did not affect macrophage phenotype or metabolism in the infarct zone.DiscussionOur results indicate that changes in glucose metabolism and the PPP underlie macrophage polarization following MI, and that metabolic reprogramming is a key feature of monocyte-derived but not resident macrophages.

Frequency Domain Spectroscopy (FDS) is widely used to estimate oil–paper insulation. The high moisture and long aging change the FDS characteristics of the insulating oil–paper, so normalization processing is required. Still, the choice of single activation energy needs further research. This paper studies the FDS characteristic of oil–paper insulation samples with different moisture and aging degrees and improves the traditional temperature correction method. The enhanced Arrhenius model uses the segmented activation energy to make the normalized FDS curve coincide better with the target curve, and the error is reduced. In addition, to verify the method’s effectiveness, this paper proposes an iterative correction process. It corrects the tan δ-f curve of the bushing with an aging time of 800 h based on segmented activation energy, and the overall normalization effect is improved.

Plutella xylostella granulovirus (PlxyGV) biopesticide is an effective tool to control the long-term damage of Plutella xylostella (Linnaeus) to cruciferous vegetables. In China, PlxyGV can be produced on a large scale using host insects, and its products have been registered in 2008. In experiments and biopesticide production, the routine enumeration method of PlxyGV virus particles is to use the Petroff-Hausser counting chamber in dark field microscope. However, the accuracy and repeatability of granulovirus (GV) counting are affected due to the small particle size of GV occlusion bodies (OBs), the limitations of optical microscope, the judgment of different operators, host impurities, the addition of biological products. This limits the convenience of its production, product quality, trading and field application. Here we use PlxyGV as an example, the method based on Real-time fluorescence quantitative PCR (qPCR) was optimized from two aspects of sample treatment and specific primers design, which improved the repeatability and accuracy of absolute quantitative OBs of GV. This study provides basic information for accurate quantitative PlxyGV by qPCR method.

The wetting deformation of coarse-grained materials can seriously affect the safety of earth and rock dams during initial water storage. The wetting model formulas are expressed in various forms and have complex parameters. Only a small amount of test data is fitted by mathematical statistics, and the universality of the obtained wetting model is unknown. Duncan-Chang E-B constitutive model cannot accurately reflect the wetting deformation characteristics of coarse-grained materials. Through the double-line wetting test of coarse-grained materials, the wetting model proposed by predecessors was verified and analyzed. Based on the indoor wetting test data, the parameters of each wetting model were fitted to analyze the accuracy of each wetting model in describing the wetting deformation characteristics. According to the P-Z model in the elastic-plastic theory and the wetting model formula, the P-Z wetting model is established, and the BP artificial neural network is introduced to establish the artificial neural network wetting deformation prediction model based on the P-Z model. The results show that the relationship between wetting axial strain and wetting stress level is best expressed by the exponential function. The relationship between wetting volumetric strain and wetting stress level is best described by Cheng’s linear function. The relative errors between the predicted and experimental values of the proposed neural network prediction model are all within 6%. The relationship between wetting axial strain and wetting stress level is exponential function, and the relationship between wetting volumetric strain and wetting stress level is linear function. The P-Z wetting model proposed in this research can better reflect the wetting deformation characteristics of coarse-grained materials under complex stress paths. The artificial neural network prediction model based on P-Z wetting model is more reliable and accurate, which can meet the prediction requirements of actual engineering for wetting deformation of coarse-grained materials.

The molecular complexes of Re(4,4′-dcbpy)(CO)3Cl (dcbpy = dicarboxylicacid-2,2’-bipyridyl) and [Ru(dcbpy)3]2+ are co-assembled into UiO-66 scaffolds as structural imperfects for CO2 photocatalytic reduction (named as Re-Ru@U). The prepared catalysts are characterized by XRD, Fourier-Transform infrared (FTIR) spectra, X-ray photoelectron spectra (XPS) and N2 adsorption–desorption isotherms. The intact structure of molecular complexes within the matrix are monitored by 1H nuclear magnetic resonance (NMR) spectra through a totally digesting catalyst. The optical properties are studied via absorption and photoluminescence spectra, and the single-electron reduction in Re and Ru complexes is detected by electron paramagnetic resonance (EPR) spectra. An excellent photocatalytic performance is obtained with steady and sustained CO evolution and a turnover number (TON) value of 15 (11 h). The CO activity irradiating by single wavelength presents the absorption-intensity-dependent changing tendency, where the absorption intensity is superposed by Re and Ru complexes. The two radicals related to Re and Ru, respectively, are simultaneously detected in the Re-Ru@U catalyst. It is suggested that the ReC2 component serves as both a photosensitizer and a catalyst, and the RuC2 component works as an additional photosensitizer to supply the second electron for CO2 reduction. The co-assembling of dual metals Re and Ru in the matrix promotes the electron transfer from the reductive Ru centres to one-electron-reduced Re centres and accounts for the superior activity of CO evolution. Our results demonstrate a strategy to develop the multimetallic catalysts via facile assembling into MOF scaffolds to promote photocatalytic performance.

Background: Macrophages and monocytes orchestrate inflammatory processes in the lungs. However, their role in the pathogenesis of chronic obstructive pulmonary disease (COPD), an inflammatory condition, is not well known. Here, we determined the characteristics of these cells in lungs of COPD patients and identified novel therapeutic targets. Methods: We analyzed the RNA sequencing (scRNA-seq) data of explanted human lung tissue from COPD (n = 18) and control (n = 28) lungs and found 16 transcriptionally distinct groups of macrophages and monocytes. We performed pathway and gene enrichment analyses to determine the characteristics of macrophages and monocytes from COPD (versus control) lungs and to identify the therapeutic targets, which were then validated using data from a randomized controlled trial of COPD patients (DISARM). Results: In the alveolar macrophages, 176 genes were differentially expressed (83 up- and 93 downregulated; Padj < 0.05, |log2FC| > 0.5) and were enriched in downstream biological processes predicted to cause poor lipid uptake and impaired cell activation, movement, and angiogenesis in COPD versus control lungs. Classical monocytes from COPD lungs harbored a differential gene set predicted to cause the activation, mobilization, and recruitment of cells and a hyperinflammatory response to influenza. In silico, the corticosteroid fluticasone propionate was one of the top compounds predicted to modulate the abnormal transcriptional profiles of these cells. In vivo, a fluticasone–salmeterol combination significantly modulated the gene expression profiles of bronchoalveolar lavage cells of COPD patients (p < 0.05). Conclusions: COPD lungs harbor transcriptionally distinct lung macrophages and monocytes, reflective of a dysfunctional and hyperinflammatory state. Inhaled corticosteroids and other compounds can modulate the transcriptomic profile of these cells in patients with COPD.

Lycium barbarum, a homology of medicine and food, contains many active ingredients including polysaccharides, polyphenol, betaine, and carotenoids, which has health benefits and economic value. The bioactive components in Lycium barbarum exhibit the effects of antioxidation, immune regulation, hypoglycemic effects, and vision improvement. Recently, the development of nutrition and health products of Lycium barbarum has been paid more and more attention with the increase in health awareness. A variety of nutrients and bioactive components in wolfberry can be retained or increased using modern fermentation technology. Through fermentation, the products have better flavor and health function, which better meet the needs of market diversification. The main products related to wolfberry fermentation include wolfberry fruit wine, wolfberry fruit vinegar, and lactic acid fermented beverage. In this review, the mainly bioactive components of Lycium barbarum and its deep-processing products of fermentation were summarized and compared. It will provide reference for the research and development of fermented and healthy products of Lycium barbarum.

Zn-BTC (H3BTC refers to 1, 3, 5-benzoic acid) MOF was used as a self-template and a zinc source to prepare ZnS/NiS2 with a layered heterogeneous structure as a promising electrode material using cation exchange and solid-phase vulcanization processes. The synergistic effect of the two metal sulfides enhances the application of ZnS/NiS2. And the high specific surface area and abundant active sites further promote the mass/charge transfer and redox reaction kinetics. In the three-electrode system, the specific capacitance was as high as 1547 F/g at a current density of 1 A/g, along with satisfactory rate capability (1214 F/g at 6 A/g) and cycling performance. Coupled with activated carbon (AC), the prepared hybrid device (ZnS/NiS2 as the positive electrode and AC as the negative electrode) (ZnS/NiS2/AC) can be operated under a potential window of 1.6 V and provides a high energy density of 26.3 Wh/kg at a power density of 794 W/kg. Notably, the assembled ZnS/NiS2//AC showed little capacity degradation after 5000 charge/discharge cycles.

A 6500 V SiC trench MOSFET with integrated unipolar diode (UD-MOS) is proposed to improve reverse conduction characteristics, suppress bipolar degradation, and reduce switching loss. An N type base region under the trench dummy gate provides a low barrier path to suppress hole injection during the reverse conduction operation. The reverse conduction voltage VON is reduced to 1.11 V, and the reverse recovery charge (QRR) is reduced to 1.22 μC/cm2. The gate-to-drain capacitance (CGD) and gate-to-source capacitance (CGS) of the UD-MOS are also reduced to improve switching loss due to the thick oxide layer between the trench gate and dummy gate. The proposed device exhibits an excellent loss-related figure of merit (FOM). It provides a high-voltage SiC MOSFET prototype with potential performance advantages for voltage source converter-based high voltage direct current applications.

The extracellular polymeric substance (EPS) plays a key factor in biofilm formation. However, the research on the importance of each EPS fraction is mainly concentrated in the activated sludge field. In this study, biofilms were cultivated under different hydrodynamic conditions in indoor flumes, and the important regulatory effects of dissolved EPS (SB-EPS), loosely bound EPS (LB-EPS), and tightly bound EPS (TB-EPS) on biofilm formation were investigated. The results indicated that the ratios of soluble EPS (S-EPS), loosely bound EPS (LB-EPS), and tightly bound EPS (TB-EPS) were 27:74:108 in the turbulent flow, 38:48:71 in the transitional flow, and 89:51:51 in the laminar flow. Regarding proportion, TB-EPS and LB-EPS were secreted more in the turbulent flow, while S-EPS was secreted slightly more in the laminar flow. S-EPS lacks the structural strength provided by bound EPS. Under the special bonding effects of LB-EPS and TB-EPS, many microcolonies join to form biofilms. The polysaccharide content in the EPS of biofilms remained dominant under all conditions. Polysaccharides are the core of biofilm formation, which enhance bacterial aggregation and make biofilm dense. Through the mutual verification of the results in the microscopic and macroscopic fields, the mechanism of biofilm formation was further elucidated, especially, in Stage IV, due to the special bonding effects of LB-EPS and TB-EPS, many colonies adhere to the mature biofilm. Further studies are required to investigate the extracellular polysaccharides and proteins in EPS along with their properties in biofilms.

The leaf inclination angle is an important agronomic rice trait and an indicator of ideal plant architecture, yield and revenue. Based on 120 recombinant inbred lines (RILs) obtained from crossing of the F1 generation of the indica rice cultivar Huazhan as the male parent and the japonica rice cultivar Nekken2 as the female parent, followed by inbreeding for 12 generations, the leaf inclination angle of the first, second and third leaves from the top of the plants were analyzed. At the same time, quantitative trait locus (QTL) mapping of the leaf inclination angle was performed using encrypted genetic maps constructed for this population. A total of 33 QTLs were detected, including two related to the first leaf inclination angle (FLIA), nine related to the second leaf inclination angle (SLIA) (highest Lod value of 5.94), and 22 related to the third leaf inclination angle (TLIA) (highest Lod value of 8.53). At the same time, candidate genes analysis were conducted on the detected QTLs’ intervals, and a total of 15 candidate genes were screened. The expression levels of candidate genes were detected by RT-qPCR, we found that LOC_Os03g46920, LOC_Os03g52630, LOC_Os04g24328, LOC_Os08g25380, LOC_Os09g23200, LOC_Os09g32080, LOC_Os09g35940, LOC_Os09g37330 and LOC_Os09g37495 displayed extremely significant differences in expression between the parents. The results showed that these genes may be the cause of the difference in leaf inclination. The present study provided substantial foundation for the further validation of the function of leaf inclination angle genes and molecular breeding practices.

The majority of the microbial diversity in nature has not been recovered through cultivation. Enrichment is a classical technique widely used in the selective cultivation of specific taxa. Whether enrichment is suitable for cultivation studies that aim to recover large numbers of species remains little explored. To address this issue, we evaluated the potential of enrichment pretreatment in the cultivation of bacteria from marine sediments. Upon obtaining and classifying a total of 943 pure cultures from chitin and cellulose enrichment pretreatment systems and a control system, our results showed that species obtained using enrichment pretreatment differed greatly from those without enrichment. Multiple enrichment media and different enrichment times increased the number of cultivated species in a sample. Amplicon sequencing showed that the increased relative abundance during pretreatment contributed greatly to bacterial cultivation. The testing of degradation abilities against chitin and cellulose and the whole-genome sequencing of representative strains suggested that microorganism–microorganism interactions play roles in the expanded diversity of cultivated bacteria. This study provides new insights into the abilities of enrichment in exploring cultivable diversity and mining microbial resources.

The 1.55 μm waveband integrated external cavity tunable diode lasers have excellent merits such as their small volume, low cost, low power consumption, wide tuning range, narrow linewidth, large side mode suppression ratio, and high output power. These merits have attracted many applications for the lasers, such as in wavelength division multiplexing, passive optical networks, mobile backhaul, and spectral sensing technology. In this paper, firstly, the basic structure and principle of integrated external cavity tunable diode lasers are introduced, and then two main integrated structures of 1.55 μm waveband external cavity tunable diode lasers are reviewed and compared in detail, namely the hybrid integrated structure and monolithic integrated structure of 1.55 μm waveband integrated external cavity tunable diode lasers. Finally, the research progress in 1.55 μm waveband integrated external cavity tunable diode lasers in the last decade are summarised, and the advantages and disadvantages of 1.55 μm waveband integrated external cavity tunable diode lasers are analysed. The results show that, with the transformation of optical communication into more complex modulation formats, it is necessary to integrate miniature 1.55 μm waveband external cavity tunable diode lasers. Low-cost integrated 1.55 μm waveband external cavity tunable diode lasers are expected to be used in the next generation of optical transceivers in small-factor modules.
