Tripartite motif protein 21 (TRIM21) is an E3 ubiquitin ligase and cytosolic antibody receptor of the TRIM family. Previous reports have indicated that TRIM21 plays an important role during viral infection. This study aimed at examining the role of TRIM21 in the replication of porcine epidemic diarrhea virus (PEDV) and showed that TRIM21 inhibits PEDV proliferation by targeting and degrading the nucleocapsid (N) protein through the proteasomal pathway. Furthermore, the endogenous expression of TRIM21 was found to be downregulated by PEDV infection in Vero and LLC-PK1 cells. Overexpression of TRIM21 inhibited PEDV replication, whereas knockdown of TRIM21 increased viral titers and N protein levels. TRIM21 was found to interact and colocalize with the N protein, and the TRIM21-mediated antiviral effect was dependent on its ubiquitin ligase activity, which engages in polyubiquitination and degradation of the N protein in a proteasome-dependent manner. Taken together, these findings provide information about the role of TRIM21 in PEDV proliferation and increase our understanding of host-virus interactions. Amines are important for new particle formation and subsequent growth in the atmosphere. Consequently, the processes involved are receiving more attention in recent years. Here, we conduct a field observation in order to investigate the atmospheric particulate amines at a background site in the Yangtze River Delta (YRD) during the summer of 2018. Four amines in PM2.5, i.e., methylamine (MA), dimethylamine (DMA), diethylamine (DEA), and trimethylamine (TMA), were collected, twice daily and analyzed. During the campaign, our measurements found the concentrations of MA, DMA, DEA, and TMA of 15.0 ± 15.0, 6.3 ± 6.9, 20.4 ± 30.1, and 4.0 ± 5.9 ng m−3, respectively, and the four amines correlated well with each other. The concentration of amines appear to be independent of whether they were collected during the day or night. Both MA and DMA exhibited a bimodal size distribution that had peaks at 0.67 and 1.1 µm, suggesting amines preferably distribute on submicron particles. Boundary layer height (BLH), relative humidity, and pH of aerosols were found have a negative relationship with amines, while aerosol liquid water content (ALWC) was found to have a positive relationship with amines. The PMF (positive matrix factorization) source apportionment results showed that the main source of amines in Chongming Island was of anthropogenic origin such as industrial and biomass emission, followed by marine sources including sea salt and marine biogenic sources. Given that the YRD region is still suffering from complex atmospheric pollution and that the knowledge on aerosol amines is still limited, more field studies are in urgent need for a better understanding of the pollution characteristics of amines.

Background: Fanconi Anemia (FA) is a rare genetic disease characterized by bone marrow failure and cancer susceptibility. FA gene mutations are also widespread somatically in non-FA cancer patients. While the FA pathway participates in DNA repair, it is also linked to R-loop metabolism and regulation. R-loops are co-transcriptional RNA:DNA hybrids that form as a result of replication-transcription collisions that are enhanced due to DNA damage, but their aberrant formation drives genomic instability and is an endogenous source of genotoxicity. However, the interplay between FA proteins, DNA repair, and R-loops remains mechanistically ill-defined. Disturbance of mRNA processing, export, and splicing has also been linked to R-loop-mediated genomic instability. The splicing factor SRSF1 participates in mRNA export through the Nuclear Export Factor 1 (NXF1). Aberrant splicing factors expression has been linked to R-loops, cancer, and myelodysplastic syndrome (MDS), which clinically resembles FA. In the present study, we study the coupling of the DNA damage response to the prevention of pathogenic R-loops via mRNA export regulation through the FA pathway and SRSF1. Methods: Cells: FA-D2 mutant, corrected, and ubiquitin-dead mutant; HeLa. siRNA transfections to reduce SRSF1 and FANCD2 protein levels. Cell survival assays against DNA damaging agents. In vitro immunoprecipitation and ubiquitination assays using purified SRSF1 and ID2. DART assay, S9.6 immunofluorescence, and S9.6 slot-blot to measure R loops in cells. Immunoprecipitation of mRNPs used to detect mature mRNA-bound ribonucleoproteins. RNA FISH to measure mRNA export. Results: In this study, we provide strong evidence that the FA pathway coordinates the prevention of R-loop formation and subsequent genomic instability through its interaction with splicing factors via regulation of mRNA export. First, we demonstrate that knockdown of SRSF1, which is known to result in increased R-loops, also is associated with diminished activation of the FA pathway. Second, the central FA complex ID2 binds to SRSF1 in cells and in recombinant protein assays. Further, we demonstrate that SRSF1 activates the FA pathway by binding ID2 and stimulates its monoubiquitination in an RNA-dependent fashion. This activity is associated with co-localization in cells at transcriptional-DNA damage inducible R loops. In turn, FANCD2 monoubiquitination proves crucial for the assembly of the SRSF1-NXF1 nuclear export complex and mRNA export. Importantly, cancer-associated SRSF1 mutants fail to interact with FANCD2, leading to inefficient FANCD2 monoubiquitination, decreased mRNA export and R-loops accumulation. Conclusion: Our findings uncover a novel mechanism of FANCD2 and SRSF1 in the prevention of R-loop formation via coupling of mRNA export to the DNA damage response, thus preventing genomic instability. 159. Polysaccharides from Sargassum fusiforme after UV/H2O2 degradation effectively ameliorate dextran sulfate sodium-induced colitis

In this study, degraded polysaccharides from Sargassum fusiforme (PSF-T2) were prepared by UV/H2O2 treatment for 2 h, and its effects on ameliorating dextran sulfate sodium-induced colitis were evaluated using a mouse model. Results showed that PSF-T2 relieved colitis symptoms, characterized by increasing the colon length and body weight, decreasing disease activity index and relieving colon damage. In addition, PSF-T2 decreased the secretion and expression of IL-1β, IL-6 and TNF-α, and increased the expression of MUC-2, ZO-1 and occludin. Besides, PSF-T2 promoted the production of short-chain fatty acids and modulated gut microbiota composition (increasing the abundance of Lactobacillaceae, Lachnospiraceae, Oscillospiraceae and Desulfovibrionaceae, and decreasing Bacteroidaceae and Erysipelotrichaceae). These results suggested that polysaccharides from Sargassum fusiforme after UV/H2O2 degradation could ameliorate colitis by decreasing inflammation, protecting the intestinal barrier and modulating gut microbiota. It can provide a theoretical basis for the preparation of bioactive polysaccharides by free radical degradation.

Fig wasp has always been thought the species-specific pollinator for their host fig (Moraceae, Ficus) and constitute a model system with its host to study co-evolution and co-speciation. The availability of a high-quality genome will help to further reveal the mechanisms underlying these characteristics. Here, we present a high-quality chromosome-level genome for Valisa javana developed by a combination of PacBio long-read and Illumina short-read. The assembled genome size is 296.34 Mb from 13 contigs with a contig N50 length of 26.76 kb. Comparative genomic analysis revealed expanded and positively selected genes related to biological features that aid fig wasps living in syconium of its highly specific host. Protein-coding genes associated with chemosensory, detoxification and venom genes were identified. Several differentially expressed genes in transcriptome data of V. javana between odor-stimulated samples and the controls have been identified in some olfactory signal transduction pathways, e.g. olfactory transduction, cAMP, cGMP-PKG, Calcim, Ras and Rap1. This study provides a valuable genomic resource for a fig wasp, and sheds insight into further revealing the mechanisms underlying their adaptive traits to their hosts in different places and co-speciation with their host.

Introduction: Anterior cruciate ligament rupture is a common clinical sports injury, and its incidence has an increasing tendency. The understanding by nurses of the best practices in the postoperative period of various surgeries has contributed to the early return of patients to their activities. Therefore, there is an urgent demand for optimizing the hospital protocol for the postoperative stage. Objective: Optimize perioperative nursing care and postoperative functional exercises in sports ligament injuries. Methods: Thirty patients (18 males, age 29.2±8.7 years) with knee cruciate ligament injury admitted to Sanya University Hospital from January to December were selected. Three to five days after admission, based on a regular physical and chemical examination and cruciate ligament reconstruction surgery under epidural anesthesia. Prevention care for postoperative complications, monitoring of vital signs and alternating position on the stretcher, mobilizations, and pain care were detailed. According to the Lysholm and Tegner activity scales, evaluation was made in the functional loss and level of recovery caused by the knee ligament injury. Results: The hospital stay in this group was 12 to 43 days, with a mean of (25.4±8.6) days. After two months of follow-up, 11 cases showed excellent functional recovery, 17 cases of functional recovery were good, the excellent and good index was 93.3%. After six months of follow-up, all patients in this group returned to work and normal life. Conclusion: Optimized treatment with functional training in the post-surgical period, good care, and adapted exercises are of great importance for the brief recovery of a sports function. RESUMO Introdução: A ruptura do ligamento cruzado anterior é uma lesão clínica esportiva comum e sua incidência tem uma tendência crescente. A compreensão pelas enfermeiras sobre as melhores práticas no pós-operatório de várias cirurgias tem colaborado para o retorno precoce dos pacientes às suas atividades. Urge a necessidade de uma otimização no protocolo hospitalar para esse pós-cirúrgico. Objetivo: Otimizar as condutas de enfermagem perioperatória e os exercícios funcionais pós-operatórios das lesões ligamentares esportivas. Métodos: Trinta pacientes (18 homens, idade de 29.2±8.7 anos) com lesão do ligamento cruzado do joelho internados no hospital da Universidade Sanya entre janeiro a dezembro foram selecionados. Decorridos 3 a 5 dias após a admissão, com base em exames físicos e químicos regulares, e cirurgia de reconstrução do ligamento cruzado sob anestesia peridural. Foram detalhados os cuidados de prevenção para complicações no pós-operatório, monitoramento de sinais vitais e alternância de posição na maca, mobilizações e cuidados com a dor. A debilidade funcional e o nível de recuperação causados pela lesão ligamentar do joelho foram avaliados de acordo com a escala de pontuação de Lysholm e a escala de atividade de Tegner. Resultados: A permanência hospitalar nesse grupo foi de 12 a 43 dias, com uma média de (25,4±8,6) dias. Após 2 meses de acompanhamento, 11 casos mostraram excelente recuperação funcional, 17 casos de recuperação funcional foram bons, o índice excelente e bom foi de 93,3%. Depois de 6 meses de acompanhamento, todos os pacientes desse grupo voltaram ao trabalho e à vida normal. Conclusão: O tratamento otimizado com treinamento funcional no pós-cirúrgico, bons cuidados e exercícios adaptados são de grande importância para a breve recuperação da função esportiva. Urge la necesidad de una optimización en el protocolo hospitalario para este postoperatorio. Objetivo: Optimizar los cuidados de enfermería perioperatorios y los ejercicios funcionales postoperatorios en las lesiones ligamentosas deportivas. Métodos: Se seleccionaron treinta pacientes (18 varones, con una edad de 29,2±8,7 años) con lesión del ligamento cruzado de la rodilla ingresados en el Hospital Universitario de Sanya de enero a diciembre. De tres a cinco días después del ingreso, según el examen físico y químico regular, y la cirugía de reconstrucción del ligamento cruzado bajo anestesia epidural. Se detallaron los cuidados de prevención de las complicaciones postoperatorias, la vigilancia de las constantes vitales y la alternancia de posición en la camilla, las movilizaciones y los cuidados del dolor. La debilidad funcional y el nivel de recuperación causados por la lesión del ligamento de la rodilla se evaluaron según la escala de puntuación de Lysholm y la escala de actividad de Tegner. Resultados: La estancia hospitalaria en este grupo fue de 12 a 43 días, con una media de (25,4±8,6) días. Tras 2 meses de seguimiento, 11 casos mostraron una excelente recuperación funcional, 17 casos de recuperación funcional fueron buenos, el índice de excelente y bueno fue del 93,3%. Tras 6 meses de seguimiento, todos los pacientes de este grupo volvieron al trabajo y a la vida normal. Conclusión: El tratamiento optimizado con entrenamiento funcional en el periodo postoperatorio, los buenos cuidados y los ejercicios adaptados son de gran importancia para la breve recuperación de la función deportiva. To evaluate vessel density (VD) and structural features in Posner-Schlossman syndrome (PSS) patients' eyes using optical coherence tomography angiography (OCTA).An observational study was conducted among 23 affected eyes (group A), 23 contralateral unaffected eyes (group B), and 23 control eyes (group C).The macular superficial, macular deep, and optic nerve head (ONH) VD of group A were 45.26 ± 5.80, 46.78 ± 6.96, and 46.10 ± 4.22, respectively. The macular superficial VD, macular deep VD, and retinal thickness of group A were significantly lower than those of group C (allOCTA can detect the fundus change in the macular and ONH regions of the PSS's affected eyes.

The mechanisms of chemoreception in fig wasps (Hymenoptera, Agaonidae) are of primary importance in their co-evolutionary relationship with the fig trees they pollinate. We used transcriptome sequences of 25 fig wasps in six genera that allowed a comparative approach to the evolution of key molecular components of fig wasp chemoreception: their odorant (OR) and gustatory (GR) receptor genes. In total, we identified 311 ORs and 47 GRs, with each species recording from 5 to 30 OR genes and 1-4 GR genes. 304 OR genes clustered into 18 orthologous groups known to be sensitive to cuticular hydrocarbons (CHC), pheromones, acids, alcohols and a variety of floral scents such as cineole, Linalool, and Heptanone. 45 GR genes clustered into 4 orthologous groups that contain sweet, bitter, CO

Mesenchymal stromal cells (MSCs) have become a promising treatment for inflammation-related diseases, and their therapeutic efficacy mainly depends on crosstalk between MSCs and inflammation. However, methods to improve the immunosuppressive efficiency of MSCs in different diseases still need to be developed. In this study, we investigated whether preconditioning MSCs with a disease-related inflammatory cytokine could increase their immunosuppressive properties and improve therapeutic efficacy. In a contact hypersensitivity (CHS) mouse model, inflammatory profile screening revealed that among all tested cytokines, monocyte chemotactic protein-1 (MCP-1) exhibited the most significantly increased level in the local microenvironment. As expected, MSCs preconditioned with MCP-1 (P-MSCs) exhibited an enhanced ability to downregulate proinflammatory cytokine secretion, induce regulatory T cells, inhibit T cell proliferation, and polarize M2-type macrophages. In vivo experiments showed that P-MSCs alleviated ear swelling and local proinflammatory cytokine production more effectively than control MSCs. Mechanistically, MCP-1 could significantly activate the signal transducer and activator of transcription 3 (STAT3) signaling pathway and induce the expression of cyclooxygenase-2 (COX2) and prostaglandin E2 (PGE2) in MSCs. STAT3 inhibitor reversed the MCP-1-mediated enhancing of their immunosuppressive ability. Collectively, our findings demonstrate that CHS-related MCP-1 preconditioning enhanced the immunomodulatory effects of MSCs and improved their therapeutic efficacy in CHS. Enhancing the immunosuppressive efficacy of MSCs by preconditioning with certain disease-related inflammatory cytokines may provide a new strategy for MSC-based therapies for inflammatory diseases.

Norfloxacin (NOF) is an environmentally harmful and ubiquitous aquatic pollutant with extensive production and application. In this study, a novel composition named carbon-based composite photocatalytic material of zinc oxide and zinc sulphide (ZnO/ZnS@BC) was successfully obtained by the impregnation-roasting method to remove NOF under UV-light. Scanning electron microscopy, X-ray photoelectron spectroscopy, transmission electron microscopy and energy dispersive spectrometer characterised the composition. ZnO/ZnS was successfully decorated on the surface of biochar (BC). The pH, the ZnSO4/PS ratio, and ions and quenchers, were investigated. High removal efficiency was obtained with a pH of 7 and a ZnSO4/PS ratio of 1:1, and the removal ratio of NOF reached 95% within three hours; the adsorption and degradation ratios reached 46% and 49%, respectively. Fe2+ promoted the degradation of NOF, whereas other ions inhibited it, with NO3− showing the strongest inhibitory effect. Three reactive species (tert-butanol, quinone, and ammonium oxala) were identified in the catalytic system. The decreasing order of the contribution of each reactive species was: O2− > ·OH− > h+. Additionally, a recycling experiment demonstrated the stability of the catalyst; the catalytic degradation ratio of NOF reached 78% after five successive runs. Therefore, ZnO/ZnS@BC possessed strong adsorption capacity and high ultraviolet photocatalysis ability.

Induction of estrus is an effective management tool for increasing the pregnancy rate in ewes, and there is a long estrus interval after lambing in spring in Small-tail Han ewe. In this study, norgestrel releasing intra-vaginal devices (PRID) and PRID + pregnant mare serum gonadotrophin (PMSG) were used for estrus induction in Small-tail Han ewe, and expression of follicle-stimulating hormone receptor (FSHR), luteinizing hormone receptor (LHR), estrogen receptor (ER) and progesterone receptor (PGR) mRNA in the ovarial stroma, oviduct ampulla and endometrium was analyzed by quantitative real-time PCR. The results showed that the estrus-induction with PRID and PRID + PMSG almost had the same effects on increasing the percentage of animals in estrus and conception rate of the ewes, and the estrus-induction had no significant effect on the expression of LHR, ER and PGR mRNA in the oviduct ampulla, ovarial stroma and endometrium, and FSHR in the ovarial stroma and endometrium. MXenes, two-dimensional transition metal carbides, exhibit excellent properties in many fields such as energy storage. The first discovered, Ti3C2, has been widely used in lithium ion batteries. However, thermal properties and delamination of few-layer MXenes have been insufficiently explored. In this work, few-layer Ti3C2 nanosheets were prepared under ultra-high pressure and by low temperature exfoliation. Furthermore, a flexible cellulose nanofiber (CNF)/Ti3C2 composite film was fabricated by simple vacuum-assisted filtration. The heat spreader of Ti3C2/CNF paper exhibits in-plane and out-of-plane thermal conductivities of 11.57 and 1.57 W m−1 K−1 with 50 wt% loading of filler, respectively. Moreover, the synthesized Ti3C2/CNF composite films presented outstanding flexibility because the films can be folded to any shape, which enables Ti3C2/CNF composite films to be potentially applied in flexible electronic devices for heat dissipation.

170. Comprehensive humoral and cellular immune responses to SARS-CoV-2 variants in diverse Chinese populations: A benefit perspective of national vaccination

The emerging SARS-CoV-2 variants have made great challenges to current vaccine and pandemic control strategies. B.1.1.529 (Omicron), which was classified as a variant of concern (VOC) by the World Health Organization on November 26th, 2021, has quickly become the dominant circulating variant and causing waves of infections. It is urgent to understand the current immune status of the general population given that pre-existing immunity has been established by national vaccination or exposure to past variants. Using sera from 85 individuals (including 21 convalescents of natural infection, 15 cases suffered a breakthrough infection after vaccination, and 49 vaccinated participants without infection history), we showed that the cross-neutralizing activity against VOCs such as Omicron can be detected in 53 (62.4%) cases, although less potent than against the Wuhan-1 strain (WT), with a 3.9-fold reduction in geometric mean neutralizing titer (GMT) (130.7, 95% CI 88.4-193.3 vs 506, 355.8-719.7, respectively). Subgroup analysis revealed significantly enhanced neutralizing activity against WT and VOCs in Delta convalescent sera. The neutralizing antibodies against Omicron were detectable in 75% of convalescents and 44.9% of healthy donors (p = 0.006), with a GMT of 289.5, 180.9-463.3 and 42.6, 31.3-59, respectively. However, the protective effect against VOCs was weaker in young convalescents (aged < 18y), with a detectable rate of 50% and a GMT of 46.4 against Omicron, similar to vaccinees. The pan-sarbecovirus neutralizing activities were not observed in vaccinated SARS-CoV-1 survivors. A booster dose significantly increased the breadth and magnitude of neutralization against WT and VOCs to different degrees than full vaccination. In addition, we showed that COVID-19 inactivated vaccines can elicit Omicron-specific T cell responses. The positive rates of ELISpot reactions were 26.7% (4/15) and 43.8% (7/16) in the full vaccination group and the booster vaccination group, respectively. The neutralizing antibody titers declined while T-cell responses remain robust over 6 months. These findings will inform the optimization of public health vaccination and intervention strategies to protect diverse populations against SARS-CoV-2 variants.

The detection of circulating tumor cells (CTCs) still faces a huge challenge partially because of low abundance of CTCs (1-10 cells/mL). In this work, a plasmonic titled fiber Bragg grating biosensor is proposed for detection of breast cancer cells. The biosensor is made by an 18° TFBG with a 50 nm-thick gold nanofilm coating over the surface of the fiber, further immobilized with a specific antibody against GPR30, which is a membrane receptor expressed in many breast cancers, serving as bait. In vitro tests have confirmed that the proposed biosensor can detect breast cancer cells in concentration of 5 cells/mL within 20 minutes and has good linearity in the range of 5–1000 cells/mL, which has met the requirement of CTC detection in real conditions. Furthermore, theoretical analysis based on the experimental results shows that the limit of detection can even reach single-cell level. Our proposed biosensor has a simple structure, is easy to manufacture, is of small size, and has a good performance, making it a good choice for real-time, label-free, and milliliter-volume detection of cancer cells in future.

172. Tributyltin inhibits development of pubertal Leydig cells in rats

Tributyltin (TBT) is extensively applied as an antifouling mediator, and it is well-known as an endocrine disrupting chemical. However, it remains elusive whether TBT can affect the development of pubertal Leydig cells (LCs), as a part of its endocrine mechanism of action. In this study, male Sprague Dawley rats (35-day-old) orally received TBT (0, 1, or 5 mg/kg) for 24 days. Immature LCs (ILCs) were separated from 35-day-old rats, and the cells were exposed to TBT at various concentrations (0, 0.05, 0.5, 1, or 5 μM) for 24 h. In vivo TBT treatment decreased the number of LCs and inhibited androgen production (2.92 ± 0.44, 1.16 ± 0.29, and 0.67 ± 0.10 ng/ml after treatment with TBT at 0, 1, and 5 mg/kg, respectively). In vitro TBT treatment reduced androgen production, cell viability, and cell cycle progression, while increased the levels of reactive oxygen species (ROS) along with subsequent apoptosis, particularly at the concentration of 5 μM. According to in vitro and in vivo findings, TBT downregulated the expression levels of genes that could control steroidogenesis (Hsd17b3, Scarb1, Hsd3b1, and Star), and decreased protein levels due to potential reduction of NR5A1 (Nr5a1). In summary, TBT inhibited cholesterol transport and activities of certain steroidogenic enzymes, thereby leading to the reduction of androgen production.

177. Soil quality and C-S-R fungal communities in monoculture plantations after conversion from subtropical forest

- Abstract
Intensive clear cutting of natural forests and conversion to monoculture plantations are ongoing worldwide, leading to degradation of soil quality and microbial functions. Here, we compared soil quality index (SQI) and fungal community in a natural forest (Forest) and four 5-year-old monoculture plantations, including Camellia oleifera (Oil), Amygdalus persica (Peach), Myrica rubra (Berry) and Cunninghamia lanceolata (Fir), in a subtropical region of China. After conversion, soil pH rose up to 0.31, but organic carbon, total nitrogen, sucrase, acid protease, glutaminase and phosphatase activities decreased by 83%, 59%, 43%, 31%, 64%, 66% and 77%, respectively, in the plantations. Correspondingly, the SQI dropped by 65%. High-throughput sequencing of the ITS1 region demonstrated an increase in α-diversity and a striking difference in β-diversity following conversion. Changes in the dominant fungal taxa following forest conversion to plantations was interpreted by Grime’s C-S-R life history framework. Conversion increased the fungal groups with stress-tolerant (S) and ruderal (R) strategies - mainly copiotrophic saprophytes, such as Ascomycota and Zygomycota, but decreased the fungal groups with competitor (C) strategies - mainly oligotrophic saprophytes and mycorrhizal fungi, such as Basidiomycota. Genera affiliated to those phyla including Pseudophialophora, Rhytisma increased, but Russula decreased. Redundancy analysis and structural equation modeling indicated that the diversity and composition of fungal communities changed with soil quality degradation, which were mainly driven by increased soil pH, decreased available carbon and nutrients (N, P), and related enzymes activities.

Mesenchymal stem cells (MSCs) have attracted great interest for cell therapy and tissue regeneration due to their self-renewal capacity, multipotency and potent immunomodulatory effects on immune cells. However, heterogeneity of MSCs has become a prominent obstacle to limit their translation into practice, as cells from different tissue sources or each individual have great differences in their transcriptomic signatures, differentiation potential and biological functions. Therefore, there is an urgent need for consensus standard for the quality control and technical specifications of MSCs. 'Human Mesenchymal Stem Cells' is the latest set of guidelines on hMSC in China, jointly drafted and agreed upon by experts from the Chinese Society for Stem Cell Research. This standard specifies the technical requirements, test methods, test regulations, instructions for use, labelling requirements, packaging requirements, storage requirements, transportation requirements and waste disposal requirements for hMSC, which is applicable to the quality control for hMSC. It was originally released by the China Society for Cell Biology on 9 January 2021. We hope that publication of these guidelines will facilitate institutional establishment, acceptance and execution of proper protocols, and accelerate the international standardization of hMSC for clinical development and therapeutic applications.

This study was aimed to characterize Gracilaria lemaneiformis polysaccharides and evaluate their protective effects on Lipopolysaccharide-induced injury in IEC-6 cells. The G. lemaneiformis polysaccharide was degraded by UV/H2O2 treatment and purified to three fractions named GLP-1.0 M, GLP-1.4 M and GLP-1.6 M. The purified fractions were mainly composed of galactose, glucose and xylose. The structural analysis showed that GLP-1.6 M was a typical sulfated red alga polysaccharide containing the linear backbone of β-(1 → 3)- and α-(1 → 4)-linked galactosyl residues, anhydro-galactose units. In the Lipopolysaccharide-induced IEC-6 cells model, GLP-1.6 M exerted the strongest in vitro anti-inflammatory activity by inhibiting the release and expressions of tumor necrosis factor-α, interleukin-6 and interleukin-1β by 89.93%, 67.82% and 38.06%, respectively. Meanwhile, GLP-1.6 M enhanced the intestinal barrier function via up-regulating the expressions of tight junctions and mucin. Purpose To explore the utility of the recovery time (RT) after temporal contrast adaptation in primary open-angle glaucoma (POAG) visual function analysis, especially in severe and end-stage glaucoma, by the Erlanger Flicker Test (EFT). Methods This study included 80 POAG eyes (45 subjects) and 20 normal eyes (20 subjects). POAG eyes were divided into 5 groups. The diagnostic efficacy of the EFT was assessed, and the RT of POAG eyes at different stages was compared. The EFT results were compared with glaucomatous structure and function test results. A nomogram was developed to predict disease progression by the RT and structural indicators. Results In the normal eyes, as the test contrast increased, the RT gradually decreased. The EFT test–retest reproducibility was good, with intraclass correlation coefficient values of 0.6 (P P P ≤ 0.001). The receiver operating curve indicated that RT12% showed the best overall area under the curve (0.863). We included RT25% and average optic nerve fiber layer thickness in constructing the nomogram. POAG eyes were further divided into 8 stages. According to the probability distribution, this model showed good performance for visual function analysis in advanced glaucoma. Conclusions Combined with traditional glaucomatous structural and functional parameters, the EFT can be used in the diagnosis and visual function analysis of POAG, especially for severe and end-stage glaucoma. It could be a potential test for disease staging in severe and end-stage glaucoma.

Background Steroid-resistant (SR) acute graft-versus-host disease (aGVHD) lacks standard second-line treatment. Mesenchymal stromal cells (MSCs) have potential efficacy in SR aGVHD. We aimed to assess the efficacy and safety of MSCs combined with basiliximab and calcineurin inhibitor as second-line therapy for SR aGVHD. Methods A randomized phase 3 trial involved 203 SR aGVHD patients at nine centers in China (September 2014–March 2019). Participants were randomized at a 1:1 ratio to receive second-line therapy with (n = 101) or without (n = 102) MSCs. The primary endpoint was the overall response (OR) at day 28. Secondary and safety endpoints included durable OR at day 56, failure-free survival, overall survival (OS), chronic GVHD (cGVHD), infection, hematological toxicity and relapse. Results Of 203 patients, 198 (97.5%; mean age, 30.1 years; 40.4% women) completed the study. The OR at day 28 was higher in the MSC group than the control group (82.8% [82 patients] vs. 70.7% [70]; odds ratio, 2.00; 95% confidence interval [CI], 1.01–3.94; P = 0.043). The durable OR at day 56 was also higher in the MSC group (78.8% [78 patients] vs. 64.6% [64]; odds ratio, 2.02; 95% CI, 1.08–3.83; P = 0.027). The median failure-free survival was longer in the MSC group compared with control (11.3 months vs. 6.0 months; hazard ratio (HR) 0.68; 95% CI, 0.48–0.95, P = 0.024). The 2-year cumulative incidence of cGVHD was 39.5% (95% CI, 29.3–49.4%) and 62.7% (51.4–72.1%) in the MSC and control groups (HR 0.55, 95% CI, 0.36–0.84; P = 0.005). Within 180 days after study treatments, the most common grade 3 and 4 adverse events were infections (65 [65.7%] in the MSC group vs. 78 [78.8%] in the control group) and hematological toxicity (37 [37.4%] vs. 53 [53.5%]). The 3-year cumulative incidence of tumor relapse was 10.1% (95% CI, 5.2–17.1) and 13.5% (7.5–21.2%) in the MSC and control groups, respectively (HR 0.75, 95% CI, 0.34–1.67, P = 0.610). Conclusions MSCs plus second-line treatments increase the efficacy of SR aGVHD, decrease drug toxicity of second-line drugs and cGVHD without increasing relapse, and are well-tolerated. Porcine epidemic diarrhea virus (PEDV) is a globally distributed alphacoronavirus that has reemerged lately, resulting in large economic losses. During viral infection, type I interferon (IFN-I) plays a vital role in the antiviral innate immunity. However, PEDV has evolved strategies to limit IFN-I production. To suppress virus replication, the host must activate IFN-stimulated genes and some host restriction factors to circumvent viral replication. This study observed that PEDV infection induced early growth response gene 1 (EGR1) expression in PEDV-permissive cells. EGR1 overexpression remarkably suppressed PEDV replication. In contrast, depletion of EGR1 led to a significant increase in viral replication. EGR1 suppressed PEDV replication by directly binding to the IFN-regulated antiviral (IRAV) promoter and upregulating IRAV expression. A detailed analysis revealed that IRAV interacts and colocalizes with the PEDV nucleocapsid (N) protein, inducing N protein degradation via the E3 ubiquitin ligase MARCH8 to catalyze N protein ubiquitination. Knockdown of endogenous MARCH8 significantly reversed IRAV-mediated N protein degradation. The collective findings demonstrate a new mechanism of EGR1-mediated viral restriction, in which EGR1 upregulates the expression of IRAV to degrade PEDV N protein through MARCH8. IMPORTANCE PEDV is a highly contagious enteric coronavirus that has rapidly emerged worldwide and has caused severe economic losses. No currently available drugs or vaccines can effectively control PEDV. PEDV has evolved many strategies to limit IFN-I production. We identified EGR1 as a novel host restriction factor and demonstrated that EGR1 suppresses PEDV replication by directly binding to the IRAV promoter and upregulating the expression of IRAV, which interacts with and degrades the PEDV N protein via the E3 ubiquitin ligase MARCH8 to catalyze nucleocapsid protein ubiquitination, which adds another layer of complexity to the innate antiviral immunity of this newly identified restriction factor. A better understanding of the innate immune response to PEDV infection will aid the development of novel therapeutic targets and more effective vaccines against virus infection.

Influenza a virus exploits host machinery to benefit its replication in host cells. Knowledge of host factors reveals the complicated interaction and provides potential targets for antiviral treatment. Here, instead of the traditional proteomic analysis, we employed a 4D label free proteomic method to identify cellular factors in A549 cells treated with avian H9N2 virus. We observed that 425 proteins were upregulated and 502 proteins were downregulated. Western blotting and quantitative real-time PCR results showed that the zinc-finger CCHC-type containing protein 3 (ZCCHC3) levels were markedly induced by H9N2 infection. Transient expression assay showed that ZCCHC3 expression decreased NP protein levels and viral titers, whereas knockdown of ZCCHC3 enhanced viral growth. Specifically, ZCCHC3 promoted the expression of IFN-β, leading to the increased transcription of IFN downstream antiviral factors. Surprisingly, viral NS1 protein was able to antagonize the antiviral effect of ZCCHC3 by downregulating IFN-β. Eventually, we observed that chicken finger CCCH-type containing protein 3, named ZC3H3, could also suppress the replication of H9N2 virus and the coronavirus-infectious bronchitis virus (IBV) in DF-1 cells. Together, our results showed the cellular proteomic response to H9N2 infection and identified ZCCHC3 as a novel antiviral factor against H9N2 infection, contributing to the understanding of host-virus interaction.

Mesenchymal stromal cells (MSCs) have been considered a promising alternative for treatment of acute respiratory distress syndrome (ARDS). However, there is significant heterogeneity in their therapeutic efficacy, largely owing to the incomplete understanding of the mechanisms underlying the therapeutic activities of MSCs. Here, we hypothesize that the cholinergic anti-inflammatory pathway (CAP), which is recognized as a neuroimmunological pathway, may be involved in the therapeutic mechanisms by which MSCs mitigate ARDS. Using lipopolysaccharide (LPS) and bacterial lung inflammation models, we found that inflammatory cell infiltration and Evans blue leakage were reduced and that the expression levels of choline acetyltransferase (ChAT) and vesicular acetylcholine transporter (VAChT) in lung tissue were significantly increased 6 hours after MSC infusion. When the vagus nerve was blocked or α7 nicotinic acetylcholine (ACh) receptor (α7nAChR)-knockout mice were used, the therapeutic effects of MSCs were significantly reduced, suggesting that the CAP may play an important role in the effects of MSCs in ARDS treatment. Our results further showed that MSC-derived prostaglandin E2 (PGE2) likely promoted ACh synthesis and release. Additionally, based on the efficacy of nAChR and α7nAChR agonists, we found that lobeline, the nicotinic cholinergic receptor excitation stimulant, may attenuate pulmonary inflammation and alleviate respiratory symptoms of ARDS patients in a clinical study (ChiCTR2100047403). In summary, we reveal a previously unrecognized MSC-mediated mechanism of CAP activation as the means by which MSCs alleviate ARDS-like syndrome, providing insight into the clinical translation of MSCs or CAP-related strategies for the treatment of patients with ARDS.

The blood-brain barrier (BBB) prevents antibodies from penetrating the CNS and limits conventional antibody-based approaches to brain tumors. We now show that ENT2, a transporter that regulates nucleoside flux at the BBB, may offer an unexpected path to circumventing this barrier to allow targeting of brain tumors with an anti-DNA autoantibody. Deoxymab-1 (DX1) is a DNA-damaging autoantibody that localizes to tumors and is synthetically lethal to cancer cells with defects in the DNA damage response. We found that DX1 penetrated brain endothelial cells and crossed the BBB, and mechanistic studies identify ENT2 as the key transporter. In efficacy studies, DX1 crosses the BBB to suppress orthotopic glioblastoma and breast cancer brain metastases. ENT2-linked transport of autoantibodies across the BBB has potential to be exploited in brain tumor immunotherapy, and its discovery raises hypotheses on actionable mechanisms of CNS penetration by neurotoxic autoantibodies in CNS lupus.

Photoaging is a complex and multistage process triggered mainly by ultraviolet (UV) radiation due to exposure to sunlight. Photoaging induces DNA damage and oxidative stress that initiate an inflammatory response and an increase of matrix metalloproteinases (MMPs) expression, which results in cumulative changes in skin appearance, structure, and functions, and eventually causes skin carcinogenesis. Dietary polysaccharides from bio-resources have been utilized as functional ingredients in healthy food, cosmetics, and drug due to their good bioactivities. However, a systematic introduction to their effects and underlying mechanisms in anti-photoaging is limited. This review discusses the damage and pathogenesis of UV-induced photoaging and summarizes the up-to-date advances in research on the anti-photoaging activity of non-starch polysaccharides from natural edible resources considering the influence of oxidative stress, DNA damage, MMPs regulation, inflammation, and melanogenesis, primarily focusing on the cellular and molecular mechanisms. This paper will help to understand the anti-photoaging functions of dietary non-starch polysaccharides from natural resources and further application in drug and functional food.

Enhanced understanding of flow structure in braided rivers is essential for river regulation, flood control, and infrastructure safety across the river. It has been revealed that the basic morphological element of braided rivers is confluence-bifurcation units. However, flow structure in these units has so far remained poorly understood with previous studies having focused mainly on single confluences/bifurcations. Here, the flow structure in a laboratory-scale confluence-bifurcation unit is numerically investigated based on the FLOW-3D® software platform. Two discharges are considered, with the central bars submerged or exposed respectively when the discharge is high or low. The results show that flow convergence and divergence in the confluence-bifurcation unit are relatively weak when the central bars are submerged. Based on comparisons with a single confluence/bifurcation, it is found that the effects of the upstream central bar on the flow structure in the confluence-bifurcation unit reign over those of the downstream central bar. Concurrently, the high-velocity zone in the confluence-bifurcation unit is less concentrated than that in a single confluence while being more concentrated than that observed in a single bifurcation. The present work unravels the flow structure in a confluence-bifurcation unit and provides a unique basis for further investigating morphodynamics in braided rivers.Highlights 3D flow structure in a confluence-bifurcation unit (CBU) is numerically investigated.Flow convergence/divergence in CBU is relatively weak when central bars are submerged.Effects of the upstream central bar on CBU flow reign over those of the downstream central bar.The high-velocity zone is less/more concentrated in the CBU than in a single confluence/bifurcation when the central bars are exposed.

Summary: Background: Bronchiolitis obliterans syndrome (BOS) after allo-HSCT is a devastating complication with limited therapeutic options. We aimed to assess the efficacy and safety of mesenchymal stem cells (MSCs) in BOS after allo-HSCT. Methods: This multicenter prospective cohort study enrolled 81 allo-HSCT recipients whose BOS were diagnosed within 6 months. The choice of prednisone and azithromycin combined with or without MSCs was based on patient preferences (MSC n = 49, non-MSC n = 32). The primary endpoint was response rate at 3 months, defined as the proportion of patients achieving FEV1 improvement or steroid sparing. The trial was registered at ClinicalTrials.gov (NCT02543073). Findings: Response rate was 35/49 patients (71%, 95% CI 59 to 84%) and 14/32 (44%, 27 to 61%) in MSC and non-MSC group, respectively (p = 0.013). The addition of MSCs was associated with a better difference for change in FEV1 rate of decline, compared to non-MSC group (53 mL/months, 2 to 103; p = 0.040). The 3-year overall survival post-diagnosis was 70.6% (55.9 to 85.3%) and 58.2% (36.1 to 78.5%) in MSC and non-MSC group, respectively (p = 0.21). Clinical improvement was accompanied by a significant increase of interleukin (IL)-10-producing CD5+B cells. There was no statistical difference in the rates of infections and leukemia relapse between the two groups. MSCs were well-tolerated with no serious adverse events. Interpretation: MSCs offer an effective and safe therapeutic option for BOS after allo-HSCT. Our study strengthens evidence for clinical use of MSC therapy in BOS. These data also provide novel insight into potential biological mechanisms of MSC treatment and support further investigation in larger randomized controlled trials. Beak and feather disease virus (BFDV) is an infectious agent responsible for feather degeneration and beak deformation in birds. In March 2017, an epidemic of psittacine beak and feather disease (PBFD) struck a farm in Fuzhou in the Fujian Province of southeast China, resulting in the death of 51 parrots. In this study, the disease was diagnosed and the pathogen was identified by PCR and whole genome sequencing. A distinct BFDV strain was identified and named as the FZ strain. This BFDV strain caused severe disease symptoms and pathological changes characteristic of typical PBFD in parrots, for example, loss of feathers and deformities of the beak and claws, and severe pathological changes in multiple organs of the infected birds. Phylogenetic analysis showed that the FZ strain was more closely related to the strain circulating in New Caledonia than the strains previously reported in China. Nucleotide homology between the FZ strain and other 43 strains of BFDV ranged from 80.0% to 92.0%. Blind passage experiment showed that this strain had limited replication capability in SPF Chicken Embryos and DF-1 Cells. Furthermore, the capsid (Cap) gene of this FZ strain was cloned into the pGEX-4T-1 expression vector to prepare the polyclonal anti-Cap antibody. Western blotting analysis using the anti-Cap antibody further confirmed that the diseased parrots were infected with BFDV. In this study, a PBFD and its pathogen was identified for the first time in Fujian Province of China, suggesting that future surveillance of BFDV should be performed. Rationale: Mesenchymal stem cells (MSCs) have been demonstrated to ameliorate inflammatory bowel disease by their actions on multiple immune cells, especially on regulatory B cells (Breg cells). However, the phenotypes and functions of human MSCs (hMSCs)-treated Breg cell subsets are not yet clear. Methods: Purified B cells were cocultured with MSCs and the phenotypes and immunomodulatory functions of the B cells were analyzed by FACS and proliferation assays in vitro. Also, a trinitrobenzenesulfonic acid-induced mouse colitis model was employed to detect the function of MSC-treated Breg cells in vivo. Results: We demonstrated that coculturing with hMSCs significantly enhanced the immunomodulatory activity of B cells by up-regulating IL-10 expression. We then identified that a novel regulatory B cell population characterized by CD23 and CD43 phenotypic markers could be induced by hMSCs. The CD23+CD43+ Breg cells substantially inhibited the inflammatory cytokine secretion and proliferation of T cells through an IL-10-dependent pathway. More significantly, intraperitoneal injection of hMSCs ameliorated the clinical and histopathological severity in the mouse experimental colitis model, accompanied by an increase in the number of CD23+CD43+ Breg cells. The adoptive transfer of CD23+CD43+ B cells effectively alleviated murine colitis, as compared with the CD23-CD43- B cells. Treatment with CD23+CD43+ B cells, and not hMSCs, substantially improved the symptoms of colitis in B cell-depleted mice. Conclusion: the novel CD23+CD43+ Breg cell subset appears to be involved in the immunomodulatory function of hMSCs and sheds new light on elucidating the therapeutic mechanism of hMSCs for the treatment of inflammation-related diseases.

We have proposed and experimentally demonstrated a plasmonic tilted fiber Bragg grating based biosensor for in-situ detection of the small biomolecule S-adenosyl-L-homocysteine (AdoHcy). The sensor can also monitor the molecule's interaction with the protein Set7 in real time. The biosensor is made by coating a 50-nm-thick gold film over an 18° tilted fiber Bragg grating. A spectrally dense comb of backward-propagating cladding resonance modes with Q-factor as high as 104 is excited due to the tilted grating. The spectral overlap between the excited cladding modes and the broader absorption feature of the surface plasmon makes this device a unique tool for high-accuracy measurement of small shifts of plasmon resonance wavelength. Surface biomolecular functionalization of the gold film enables us to not only detect the corresponding biomolecule but also study its interaction with other biomolecules. Our experimental results demonstrate that our biosensor can detect AdoHcy at concentration as low as 1 nM. Furthermore, we have successfully real-time monitored interaction between the protein Set7 and AdoHcy at different concentrations. Our measurements are consistent with the biological theory that the association rate of a pair of biomolecules depends on their concentrations.

197. Transverse Offset Based Single-Multi-Single Mode Fiber Structure for Vector Rotation Sensing

In this paper, we develop an alternative all-fiber interferometer, which is based on a commercial graded-index multimode fiber mismatch spliced between two single-mode fibers. Compared to the conventional single-multi-single mode fiber structure with all of the fibers perfectly aligned, theoretical analysis shows that more power is coupled into the LP11 mode while less power is coupled into the LP01 mode, resulting in a power balance between these two modes and a higher interference extinction ratio. Experiments have been carried out to evaluate the performance of the proposed structure, and the experimental results show that the extinction ratio in the transmission spectra can be obtained with a value higher than 15 dB. Moreover, the results indicate that the output power of the proposed scheme is polarization dependent. Based on this polarization-dependent behavior, we propose a Michelson interferometer scheme which can be used as a vector rotation sensor providing a sensitivity of approximately 0.0077 nm/degree in the rotation range of 0 to ±180° with a linearity higher than 99%. Our sensor has the advantages of low-cost, simple manufacture and ease of implementation, making it widely used in engineering applications such as shape sensing and structural health monitoring.

In this study, a polysaccharide was extracted from Physalis pubescens L. (named PP). Its antihyperglycemic and antihyperlipidemic activities were evaluated in STZ-induced diabetic mice. Results showed that PP was determined to be composed of rhamnose (Rha), arabinose (Ara), fucose (Fuc), xylose (Xyl), mannose (Man), glucose (Glc) and galactose (Gal) with molar percentages of 4.65%, 17.34%, 1.43%, 6.24%, 5.52%, 45.5%, and 19.31%, respectively. The average molecular weight (Mw) was found to be 20.0 kDa. It had a strong α-glucosidase inhibitory activity in vitro. PP treatment could enhance the oral glucose tolerance, and increase the levels of SOD, GSH, CAT, vitamin C, vitamin E, HDL-c, C-peptide, GCK and hepatic glycogen in diabetic mice. Besides, PP treatment could also decrease the levels of MDA, TG, TC, LDL-c, BUN and G-6-Pase. The regulating effects were stronger in high dose PP treatment than those in the low and medium dose treatments. In short, PP played an important role in protecting STZ-induced diabetic mice, and the effect was closely related to its activities in antioxidation and regulating glucose and lipid metabolism.

With the rapid growth of remote sensing image data, it has become necessary to effectively and efficiently retrieve images from a big image database for managing and exploiting such data. This paper presents a novel method for content-based remote sensing image retrieval (CBRSIR) that re-ranks the initial retrieval result using two image-to-class distances, which are the similarity between an image and an image class. One is the image-to-training-class distance between an image and each image class of the training dataset, which uses the information included in the train samples with known-label. It is used to calculate the weight of the image classes and estimating the class of an image. The other is the image-to-query-class distance between an image and the query class that is obtained using the k-nearest neighbor (k NN) method. The proposed method first obtains the initial retrieval results via sorting the distances of the CNN feature between the query image and each retrieved image in ascending order. Then, the image-to-query-class distance is calculated according to the initial result, and the weight of each class is calculated according to the class probability of images obtained by CNN and the image-to-training-class distances. Finally, the initial result is re-ranked to get the final retrieval result via the weighted image-to-class distances. The performance of the proposed method is tested on UCMD and PatternNet databases. Experimental results show the effectiveness of the proposed re-ranking scheme in image retrieval in comparison to other state-of-the-art techniques.

Pulse lavage (PL) debridement and ultrasound are both known to be the treatment of biofilm-related periprosthetic joint infection (PJI). However, the efficacy of these in combination is unknown in eradicating biofilm from the orthopaedic metal implant surface. This study was conducted to understand the efficacy of PL and ultrasound in combination in eradicating bacterial biofilms on titanium alloy in vitro. Biofilms of Staphylococcus aureus strains were grown on titanium alloy coupons for 24 h. Then, the coupons were taken to each treatment group: (i) debrided with PL, (ii) exposed to ultrasound, or (iii) exposed to both. An untreated biofilm was set as a control group. Viable plate count and confocal microscopy using live/dead staining was used to measure the amount of biofilm. Viable plate count showed an approximate two-log reduction in CFU/cm2 in PL alone, from an initial cell count on the mental surface of approximately 109 CFU/cm2. The ultrasound caused an approximate seven-log reduction, and the combination group eradicated viable biofilm bacteria completely. Confocal imaging corroborated the CFU data. Our results indicate that PL and ultrasound both are remarkably in eradicating biofilm, and the combination of PL and ultrasound is more effective than alone in reducing biofilm.
