Infants born to mothers with obesity have a greater risk for childhood obesity and metabolic diseases; however, the underlying biological mechanisms remain poorly understood. We used a Japanese macaque model to investigate whether maternal obesity combined with a Western-style diet (WSD) impairs offspring muscle insulin action. Adult females were fed a control or WSD prior to and during pregnancy through lactation, and offspring subsequently weaned to a control or WSD. Muscle glucose uptake and signaling were measured ex vivo in fetal (n = 5-8/group) and juvenile (n = 8/group) offspring. In vivo signaling was evaluated after an insulin bolus just prior to weaning (n = 4-5/group). Maternal WSD reduced insulin-stimulated glucose uptake and impaired insulin signaling at the level of Akt phosphorylation in fetal muscle. In juvenile offspring, insulin-stimulated glucose uptake was similarly reduced by both maternal and postweaning WSD and corresponded to modest reductions in insulin-stimulated Akt phosphorylation relative to controls. We conclude that maternal WSD leads to a persistent decrease in offspring muscle insulin-stimulated glucose uptake even in the absence of increased offspring adiposity or markers of systemic insulin resistance. Switching offspring to a healthy diet did not reverse the effects of maternal WSD on muscle insulin action, suggesting earlier interventions may be warranted.

The duration and nature of immunity generated in response to SARS-CoV-2 infection is unknown. Many public health responses and modeled scenarios for COVID-19 outbreaks caused by SARSCoV-2 assume that infection results in an immune response that protects individuals from future infections or illness for some amount of time. The timescale of protection is a critical determinant of the future impact of the pathogen. The presence or absence of protective immunity due to infection or vaccination (when available) will affect future transmission and illness severity. The dynamics of immunity and nature of protection are relevant to discussions surrounding therapeutic use of convalescent sera as well as efforts to identify individuals with protective immunity. Here, we review the scientific literature on antibody immunity to coronaviruses, including SARS-CoV-2 as well as the related SARS-CoV-1, MERS-CoV and human endemic coronaviruses (HCoVs). We reviewed 1281 abstracts and identified 322 manuscripts relevant to 5 areas of focus: 1) antibody kinetics, 2) correlates of protection, 3) immunopathogenesis, 4) antigenic diversity and cross-reactivity, and 5) population seroprevalence. While studies of SARS-CoV-2 are necessary to determine immune responses to it, evidence from other coronaviruses can provide clues and guide future research.

Background and Aims: The preponderance of microscopic colitis (MC) in females may be associated with postmenopausal increased risk of atherosclerosis. The aim of this study was to describe the prevalence of atherosclerotic diseases in adults with MC. Methods: Retrospective observational study of patients with a diagnosis of MC or incomplete MC at our institution from 2008 to 2018. We performed a chart review and extracted data on demographics, comorbidities, medications, diagnosis, imaging, and endoscopy. Data were analyzed descriptively. Logistic regression was used to estimate the unadjusted effects of different variables on MC. Results: Of 269 patients, 265 had a MC diagnosis; 236 (89.06%) had collagenous colitis or lymphocytic colitis; and 29 (10.94%) were diagnosed with incomplete MC. Majority were female (79.55%), ≥ 65 years (59.11%), and white (88.81%). Majority had the following risk factors for atherosclerosis, smoking (52.04%), hypertension (58.21%), and hyperlipidemia (59.5%). The prevalence of coronary artery disease (CAD), peripheral artery disease (PAD), and cerebrovascular disease (CVD) was 16.79% (12.32, 21.27), 8.58% (5.23–11.94), and 7.49% (4.33–10.65), respectively. There was no difference in the prevalence of CAD and CVD after adjusting for sex. Females with MC had reduced odds of PAD compared with males. Conclusion: The prevalence of CAD, PAD, and CVD was 16.79%, 8.58%, and 7.49%, respectively. Similar to the general population, smoking, hypertension, and hyperlipidemia are risk factors for atherosclerosis in MC.

Intelligent transportation systems have recently become a promising technology for future industry to provide safe, green, and automated driving. In this regard, the Third Generation Partnership Project (3GPP) has proposed to utilize cellular communication to enable direct communication between vehicles with and without cellular infrastructure assistance. 3GPP has introduced the Sensing-Based Semi-Persistent Scheduling (S-SPS) technique when coverage of the cellular system is absent. S-SPS faces resource collision and performance degradation problems when channel load increases in the network. This paper suggests a decentralized congestion control and transmission power control mechanism (TPC-DCC) with an adaptive threshold for the received signal as a combination method to decrease channel load in the network. Furthermore, this work introduces a novel channel load adjustment. The new adjusting algorithm is based on a constant difference between the upper and lower boundaries of channel load at each level to handle channel overload and provide more flexibility using DCC mechanisms. The interactions of the proposed algorithm with S-SPS and the Extended-Estimation Reservation Resource Allocation (E-ERRA) algorithms that the authors previously proposed are investigated. The results indicate that system performance can be substantially improved when the transmission power and reception threshold are adaptively adjusted to the proposed channel load adjustment. The results of E-ERRA with the proposed channel load adjustment method show promising results compared to S-SPS.

Abstract Malaria due to the Plasmodium falciparum parasite remains a threat to human health despite eradication efforts and the development of anti-malarial treatments, such as artemisinin combination therapies. Human movement and migration have been linked to the propagation of malaria on national scales, highlighting the need for the incorporation of human movement in modeling efforts. Spatially couped individual-based models have been used to study how anti-malarial resistance evolves and spreads in response to drug policy changes; however, as the spatial scale of the model increases, the challenges associated with modeling of movement also increase. In this paper we discuss the development, calibration, and validation of a movement model in the context of a national-scale, spatial, individual-based model used to study the evolution of drug resistance in the malaria parasite.

In cattle, prenatal transportation stress has been associated with differential methylation of genes related to metabolism, but the effects of prenatal transportation stress on skeletal muscle mitochondria and oxidative stress have not been investigated. We tested the hypothesis that prenatally stressed calves would exhibit increased skeletal muscle mitochondrial function resulting in greater oxidative stress than calves from non-stressed dams. Serum and longissimus thoracis muscle samples were collected from yearling Brahman calves whose mothers were stressed by transportation at five time points during gestation [i.e., prenatally stressed (PNS); eight bulls and six heifers] and control calves (CON; four bulls and six heifers). Serum was evaluated for concentration of the stress hormone, cortisol and for a marker of muscle perturbation, creatine kinase activity. Muscle samples were analyzed for concentration of a by-product of lipid peroxidation, malondialdehyde, and activity of the antioxidants, superoxide dismutase and glutathione peroxidase. Additionally, muscle mitochondrial volume density and function were estimated by citrate synthase and cytochrome c oxidase activities, respectively. Data were analyzed using mixed linear models with sex, treatment, and the sex × treatment interaction as fixed effects. No investigated variable differed between CON and PNS calves (p ≥ 0.3). These data suggest that prenatal transportation stress does not have an impact on skeletal muscle mitochondrial metabolism or markers of stress or muscle damage in Brahman yearling calves at rest. However, previously reported negative impacts of prenatal stress on inflammatory responses suggest that PNS calves may be differentially equipped to handle an acute stressor. Future research should investigate the energetic and inflammatory implications of acute stressors in animals subjected to prenatal stress.

Summary: Lineage switching can induce therapy resistance in cancer. Yet, how lineage fidelity is maintained and how it can be lost remain poorly understood. Here, we have used CRISPR-Cas9-based genetic screening to demonstrate that loss of SMARCB1, a member of the SWI/SNF chromatin remodeling complex, can confer an advantage to clear cell renal cell carcinoma (ccRCC) cells upon inhibition of the renal lineage factor PAX8. Lineage factor inhibition-resistant ccRCC cells formed tumors with morphological features, but not molecular markers, of neuroendocrine differentiation. SMARCB1 inactivation led to large-scale loss of kidney-specific epigenetic programs and restoration of proliferative capacity through the adoption of new dependencies on factors that represent rare essential genes across different cancers. We further developed an analytical approach to systematically characterize lineage fidelity using large-scale CRISPR-Cas9 data. An understanding of the rules that govern lineage switching could aid the development of more durable lineage factor-targeted and other cancer therapies.

The Educative Teacher Performance Assessment (edTPA) is a widely implemented performance assessment in the United States used to indicate the readiness of pre-service teachers to enter the classroom and the level of pre-service teacher preparation supported by individual colleges of education. The purpose of this study is to investigate the psychometric qualities (i.e., validity and reliability) of the edTPA specifically in the context of pre-service music education candidates. The study was guided by the following questions: 'What is the overall psychometric quality (i.e., validity and reliability) of the edTPA rubric items in the context of pre-service music educators?' (Research Question 1); 'How do the rubric criteria fit the measurement model and how do they vary in difficulty?' (Research Question 2); and (c) 'Do the category response structures for each criterion empirically cooperate to provide meaningful measures?' (Research Question 3). Results indicated overall adequate model-data fit but demonstrated measurement optimization concerns attributed to the category response structures. Implications for pre-service music education training are discussed.

BACKGROUND: Southern Province, Zambia has experienced a dramatic decline in Plasmodium falciparum malaria transmission in the past decade and is targeted for elimination. Zambias National Malaria Elimination Program recommends reactive case detection (RCD) within 140 m of index households to enhance surveillance and eliminate remaining transmission foci. METHODS: To evaluate whether RCD captures local transmission, we genotyped 26 microsatellites from 106 samples collected from index (n = 27) and secondary (n = 79) cases detected through RCD in the Macha Hospital catchment area between January 2015 and April 2016. RESULTS: Participants from the same RCD event harbored more genetically related parasites than those from different RCD events, suggesting that RCD captures, at least in part, infections related through local transmission. Related parasites clustered in space and time, up to at least 250 m from index households. Spatial analysis identified a putative focal transmission hotspot. CONCLUSIONS: The current RCD strategy detects focal transmission events, although programmatic guidelines to screen within 140 m of index households may fail to capture all secondary cases. This study highlights the utility of parasite genetic data in assessing programmatic interventions, and similar approaches may be useful to malaria elimination programs seeking to tailor intervention strategies to the underlying transmission epidemiology.

Local and cross-border importation remain major challenges to malaria elimination and are difficult to measure using traditional surveillance data. To address this challenge, we systematically collected parasite genetic data and travel history from thousands of malaria cases across northeastern Namibia and estimated human mobility from mobile phone data. We observed strong fine-scale spatial structure in local parasite populations, providing positive evidence that the majority of cases were due to local transmission. This result was largely consistent with estimates from mobile phone and travel history data. However, genetic data identified more detailed and extensive evidence of parasite connectivity over hundreds of kilometers than the other data, within Namibia and across the Angolan and Zambian borders. Our results provide a framework for incorporating genetic data into malaria surveillance and provide evidence that both strengthening of local interventions and regional coordination are likely necessary to eliminate malaria in this region of Southern Africa.

PurposeTo evaluate the safety, recommended phase II dose (RP2D) and efficacy of pexidartinib, a colony stimulating factor receptor 1 (CSF-1R) inhibitor, in combination with weekly paclitaxel in patients with advanced solid tumors.Patients and methodsIn part 1 of this phase Ib study, 24 patients with advanced solid tumors received escalating doses of pexidartinib with weekly paclitaxel (80 mg/m2). Pexidartinib was administered at 600 mg/day in cohort 1. For subsequent cohorts, the dose was increased by ⩽50% using a standard 3+3 design. In part 2, 30 patients with metastatic solid tumors were enrolled to examine safety, tolerability and efficacy of the RP2D. Pharmacokinetics and biomarkers were also assessed.ResultsA total of 51 patients reported ≥1 adverse event(s) (AEs) that were at least possibly related to either study drug. Grade 3-4 AEs, including anemia (26%), neutropenia (22%), lymphopenia (19%), fatigue (15%), and hypertension (11%), were recorded in 38 patients (70%). In part 1, no maximum tolerated dose was achieved and 1600 mg/day was determined to be the RP2D. Of 38 patients evaluable for efficacy, 1 (3%) had complete response, 5 (13%) partial response, 13 (34%) stable disease, and 17 (45%) progressive disease. No drug-drug interactions were found. Plasma CSF-1 levels increased 1.6- to 53-fold, and CD14dim/CD16+ monocyte levels decreased by 57-100%.ConclusionsThe combination of pexidartinib and paclitaxel was generally well tolerated. RP2D for pexidartinib was 1600 mg/day. Pexidartinib blocked CSF-1R signaling, indicating potential for mitigating macrophage tumor infiltration.
