Postoperative delirium is common especially in the elderly and is associated with high rates of morbidity and mortality. Non-pharmacological multicomponent interventions are effective in reducing the incidence and to a degree the duration of postoperative delirium and are recommended in international guidelines on postoperative delirium as first line intervention for management of delirium. Non-pharmacological management of postoperative delirium consists of strategies for risk stratification, risk reduction by non-pharmacological bundle interventions, early recognition of delirium by screening protocols and immediate therapy of underlying causes of delirium and continuation of non-pharmacological bundles. Non-pharmacological bundle interventions address common perioperative risk factors. Bundles comprise strategies for oxygenation, mobilization, hydration and nutrition, sensory and cognitive stimulation, reorientation, modifications of environmental factors such as design aspects and noise reduction, adequate analgesia, management of agitation and anxiety, protecting circadian rhythms for example by adequate light exposure during daytime, family involvement and timely reduction of unnecessary catheters and anticholinergic drugs. Introduction: Postoperative delirium (POD) is seen in approximately 15% of elderly patients and is related to poorer outcomes. In 2017, the Federal Joint Committee (Gemeinsamer Bundesausschuss) introduced a 'quality contract' (QC) as a new instrument to improve healthcare in Germany. One of the four areas for improvement of in-patient care is the 'Prevention of POD in the care of elderly patients' (QC-POD), as a means to reduce the risk of developing POD and its complications.The Institute for Quality Assurance and Transparency in Health Care identified gaps in the in-patient care of elderly patients related to the prevention, screening and treatment of POD, as required by consensus-based and evidence-based delirium guidelines. This paper introduces the QC-POD protocol, which aims to implement these guidelines into the clinical routine. There is an urgent need for well-structured, standardised and interdisciplinary pathways that enable the reliable screening and treatment of POD. Along with effective preventive measures, these concepts have a considerable potential to improve the care of elderly patients., Methods and Analysis: The QC-POD study is a non-randomised, pre-post, monocentric, prospective trial with an interventional concept following a baseline control period. γ-Alumina with incorporated metal oxide species (including Fe, Cu, Zn, Bi, and Ga) was synthesized by liquid-assisted grinding-mechanochemical synthesis, applying boehmite as the alumina precursor and suitable metal salts. Various contents of metal elements (5 wt.%, 10 wt.%, and 20 wt.%) were used to tune the composition of the resulting hybrid materials. The different milling time was tested to find the most suitable procedure that allowed the preparation of porous alumina incorporated with selected metal oxide species. The block copolymer, Pluronic P123, was used as a pore-generating agent. Commercial γ-alumina (S BET = 96 m 2 ·g -1 ), and the sample fabricated after two hours of initial grinding of boehmite (S BET = 266 m 2 ·g -1 ), were used as references. Analysis of another sample of γ-alumina prepared within 3 h of one-pot milling revealed a higher surface area (S BET = 320 m 2 ·g -1 ) that did not increase with a further increase in the milling time. So, three hours of grinding time were set as optimal for this material. The synthesized samples were characterized by low-temperature N 2 sorption, TGA/DTG, XRD, TEM, EDX, elemental mapping, and XRF techniques. The higher loading of metal oxide into the alumina structure was confirmed by the higher intensity of the XRF peaks. Samples synthesized with the lowest metal oxide content (5 wt.%) were tested for selective catalytic reduction of NO with NH 3 (NH 3 -SCR). Among all tested samples, besides pristine Al 2 O 3 and alumina incorporated with gallium oxide, the increase in reaction temperature accelerated the NO conversion. The highest NO conversion rate was observed for Fe 2 O 3 -incorporated alumina (70%) at 450 °C and CuO-incorporated alumina (71%) at 300 °C. The CO 2 capture was also studied for synthesized samples and the sample of alumina with incorporated Bi 2 O 3 (10 wt.%) gave the best result (1.16 mmol·g -1 ) at 25 °C, while alumina alone could adsorb only 0.85 mmol·g -1 of CO 2 . Furthermore, the synthesized samples were tested for antimicrobial properties and found to be quite active against Gram-negative bacteria, P. aeruginosa (PA). Background: Measuring pain intensity in patients unable to communicate is a challenge on general wards. Observation-based pain scoring tools have been used for patients with dementia or in critically ill intensive care unit (ICU) patients. However, there is no established or validated assessment tool for non-ICU patients without dementia related cognitive deficiencies who cannot communicate. The "Behavioural Pain Scale Non-Intubated" (BPS-NI) and the "Pain Assessment In Advanced Dementia-German" (PAINAD-G) are potential tools to fill this gap., Methods: This study was performed with verbal non-ICU patients on general wards at Charité Berlin. Two assessors independently rated pain intensity using the BPS-NI and the PAINAD-G along with patients' self-ratings on the Numeric Rating Scale (NRS). The interrater-reliability of BPS-NI and PAINAD-G was calculated and ROC-analyses were performed to identify cut-off values for medium and intense pain for each score. Effectiveness was calculated using percentage agreement. In total, 126 patients were included into analysis., Results: The BPS-NI showed substantial congruence in interrater-reliability (Cohens-Kappa 0.71), whereas the PAINAD-G showed moderate congruence (Kappa 0.48). Based on ROC-analyses, for medium pain levels a cut-off 4 (BPS-NI) or 2 (PAINAD-G) and for severe pain levels cut-off 5 (BPS-NI) and 3 (PAINAD-G) would result in good accordance with self-reported NRS for pain., Conclusion: The BPS-NI shows a good validity in measuring pain intensity in patients on general wards and may possibly be used for patients unable to communicate. This study concerns the fabrication of CTAB- and N,N-dimethyltetradecylamine-grafted zirconia and evaluation of their ability to adsorb vanadium ions. The effectiveness of ZrO 2t> functionalization and the different nature of the modifiers used were confirmed by differences in the porosity (ZrO 2 : S BET = 347 m 2 /g; ZrO 2 -CTAB: S BET = 375 m 2 /g, ZrO 2 -NH + : S BET = 155 m 2 /g), types of functional groups, and isoelectric points (the ZrO 2 and CTAB-modified samples have IEPs = 3.8 and 3.9, ZrO 2 -NH + has IEP = 7.1) of the prepared adsorbents. The designed materials were tested in batch adsorption experiments involving the removal of vanadium ions from model wastewaters at various process parameters, among which pH proved to be the most important. Based on equilibrium and kinetic evaluations, it was proved that the sorption of V(V) ions followed pseudo-second-order and intraparticle diffusion models, and the data were better fitted to the Langmuir model, suggesting the following order of the sorbents in terms of favorability for V(V) ion adsorption: ZrO 2 -NH +  > ZrO 2 > ZrO 2 -CTAB. The estimated maximum monolayer capacity of ZrO 2 -NH + for V(V) (87.72 mg/g) was the highest among the tested materials. Additionally, it was confirmed that adsorption of V(V) ions onto synthesized materials is a heterogeneous, exothermic, and spontaneous reaction, as evidenced by the calculated values of thermodynamic parameters. The key goals included the transfer of experimental findings obtained using model solutions to the adsorption of V(V) ions from solutions arising from the leaching process of spent catalysts. The highest adsorption efficiencies of 70.8% and 47.5% were recorded for the ZrO 2 -NH + material in acidic solution; this may be related to the protonization of -NH + groups, which favors the sorption of V(V) ions. Novel alumina-based materials enriched with vanadia and lanthana were successfully synthesized via in situ modification using a mechanochemical method, and were applied in ammonia-induced selective catalytic reduction of nitrogen oxides (SCR process). The synthesis was optimized in terms of the ball milling time (3 or 5 h), vanadium content (0.5, 1 or 2 wt% in the final product), and lanthanum content (0.5 or 1 wt% in the final product). Vanadium (V) oxide was immobilized on an alumina support to provide catalytic activity, while lanthana was introduced to increase the affinity of nitrogen oxides and create more active adsorption sites. Mechanochemical synthesis successfully produced mesoporous materials with a large specific surface area of 279-337 m 2 /g and a wide electrokinetic potential range from 60 to (- 40) mV. Catalytic tests showed that the incorporation of vanadia resulted in a very large improvement in catalytic performance compared with pristine alumina, increasing its efficiency from 14 to 63% at 400 °C. The best SCR performance, a 75% nitrogen oxide conversion rate at a temperature of 450 °C, was obtained for alumina enriched with 2 and 0.5 wt% of vanadium and lanthanum, respectively, which may be considered as a promising result., (© 2022. This paper discusses the properties of metal oxide/biochar systems for use in wastewater treatment. Titanium, zinc, and iron compounds are most often combined with biochar; therefore, combinations of their oxides with biochar are the focus of this review. The first part of this paper presents the most important information about biochar, including its advantages, disadvantages, and possible modification, emphasizing the incorporation of inorganic oxides into its structure. In the next four sections, systems of biochar combined with TiO 2 , ZnO, Fe 3 O 4 , and other metal oxides are discussed in detail. In the next to last section probable degradation mechanisms are discussed. Literature studies revealed that the dispersion of a metal oxide in a carbonaceous matrix causes the creation or enhancement of surface properties and catalytic or, in some cases, magnetic activity. Addition of metallic species into biochars increases their weight, facilitating their separation by enabling the sedimentation process and thus facilitating the recovery of the materials from the water medium after the purification process. Purpose: To follow the development of the microsporidian Spraguea americanus within the nervous tissue of Lophius. An attempt to determine when and how the infection begins., Methods: Acquiring different age groups of Lophius and recovering the infected sites, particularly the supramedullary neuron fibers and preparing them for microscopy., Results: The youngest juvenile Lophius recovered were 140 mm long with established infections. These infections consisted of meronts and sporoblasts but no spores. The evidence indicates these infections began a month or so earlier., Conclusions: Early stages of S. americanus development occur only in juvenile Lophius and not present in older fish. The prediction is infections of all Spraguea species begin early in the life of benthic juvenile Lophius. The high incidence of infection among these fish is an indicator that the location where the infection begins is likely rich in infective spores., (© 2022. Purpose: Most eukaryotic cells have a plasma membrane with glycoproteins on the outer leaflet and cytoplasmic proteins on the inner leaflet. However, the microsporidians examined have a peculiar outer membrane with cytoplasmic proteins on the surface. His study was to determine if this is true and identify the presence of key cytoplasmic proteins on the exterior of the sporoplasm and spore stages., Methods: Specific probes including GFP-labeled genes, antibodies, and electron microscopy were applied to studies on the sporoplasm and spore stages of different microsporidian species., Results: Cytoplasmic membrane proteins cadherin, intermediate filaments, dynactin p150Glued and tubulin were identified on the outer leaflet of the exterior membrane of the sporoplasm or spore stages of Spraguea americanus, Anncaliia algerae, Ameson michaelis and Thelohania sp., Conclusion: The invasive sporoplasm stage of microsporidians acquires an outer membrane from a cytoplasmic organelle and it has been shown that the outside surface of this membrane bears cytoplasmic proteins. This membrane retains these cytoplasmic proteins even on the spore stage. For an intracellular parasite that locates directly within the cytoplasm of a host cell, the presence of these cytoplasmic proteins on the surface of the parasite appears to have a significant effect on the host's response., (© 2021. Stable fluid bodies, such as the ocean and atmosphere, are composed of a series of increasingly dense layers, defined by density stratification interfaces in which the medium properties (e.g., temperature, salinity) change. The intensity of the stratification between the layers influences the internal mixing dynamics and entrainment, facilitating the transport of dissolved constituents within the fluid medium. Acoustic systems offer the means for high resolution observations of these interfaces, which allow for continuous data collection over broad spatial scales. Here, a one-dimensional acoustic scattering model is presented for predicting acoustic backscatter from stratification interfaces, which is widely applicable to the acoustic water column data collected with ship-mounted sonars. Model predictions based on hydrographic profiles suggest that in many oceanic cases, the density gradient perturbations can be disregarded, and sound speed perturbations alone drive the majority of the acoustic scattering. A frequency-dependent scattering intensity based on the sharpness of the stratification interface is predicted by the model, suggesting a path to remote estimations of the physical medium properties through broadband acoustic inversion. more...

The wastewater pollution of tanneries is of high concern. The investigation of technologies to minimize the consumption of chemicals in the leather production process can reduce the environmental burden. We focus on the reduction of ammonium salts in the leather production process. Salt-free deliming of animal hides with compressed carbon dioxide as deliming agent is performed for the first time in a technical scale 20-L drum. As a result, CO 2 -deliming at 30 bar and 30 °C is two times faster than conventional deliming. In addition, the deliming efficiency is slightly improved. The initial calcium (Ca) content of the hides of 8 g/kg reaches the lowest value of 2 g/kg after a process time of 3 h. However, a process time of 60 min is sufficient to reach an elimination of 50 wt% of the initial lime. The resulting Ca-content of 4 g/kg after 60 min CO 2 -deliming at 30 bar is comparable with the Ca-content of conventional delimed hide. We clarify that the ampholytic character of the collagen itself enables a buffering of the pH-value at pH-7. The stable pH-value supports the selection of specific bating enzymes that decompose non-collagen proteins. No buffering salts contaminate the wastewater. The high-pressure CO 2 -deliming process has high potential to reduce wastewater emissions, save costs for chemicals, and process time in industrial beamhouse applications. more...

On their spore surfaces, Microsporidia often develop a canopy of filaments with characteristics of intermediate filaments (IF), as we demonstrated in previous studies on Thelohania sp., Ameson michaelis, and Spraguea lophii. Genomic studies indicate that among invertebrates, lamins that may localize in the cytoplasm or nucleus, are the only known IF type. These IFs can bind to the substrate containing cell adhesion molecules (CAMs) cadherins, associated with β and γ catenins. The objects of this study were to determine whether microsporidia have CAMs with the attached IFs on their envelopes and to find out if these proteins are provided by the host. An examination was made for localization of lamins and CAMs on the spores of the mentioned above species and Anncaliia algerae, plus in the host animals. Then, we determined whether the spores of A. michaelis and A. algerae could bind vertebrate nuclear lamin onto the spore surface. We also tested transgenic Drosophila melanogaster stocks bearing cadherin-GFP to see whether developing A. algerae parasites in these hosts could acquire host CAMs. The tests were positive for all these experiments. Due to the brittle nature of poly(lactic acid) many attempts have been made to flexibilize this polyester for applications such as thin films and foils. However, due to complex phase behavior, many drawbacks for plasticizer and blend components are described. To overcome miscibility, post crystallization and migration issues a principle of click-chemistry was employed to change the molecular characteristics from external to internal plasticization by fixation of a plastisizing unit with help of a stereocomplex crystallization. Hydroxyl terminated polycaprolactone oligomers were used as a macroinitiator for the ring opening polymerization of d-lactide, resulting in blockcopolymers with plasticizing unit polycaprolactone and compatibilizing poly(d-lactic acid)-blocks. The generated block copolymers were blended with a poly(l-lactic acid)-matrix and formed so called stereocomplex crystals. In comparison to unbound polycaprolactone the polycaprolactone blocks show a lower migration tendency regarding a solution test in toluene. Besides that, trapping the plasticizing units via stereocomplex also improves the efficiency of the plasticizer. In comparison to polymer blends with the same amount of non-bonded polycaprolactone oligomers of the same molecular weight, block copolymers with poly(d-lactic acid) and polycaprolactone can shift the glass transition temperature to lower values. This effect can be explained by the modulated crystallization of the polycaprolactone-blocks trapped into the matrix, so that a higher effective amount can interact with the poly(l-lactic acid)-matrix. more...

Representatives of the genus Anncaliia are known as natural parasites of dipteran and coleopteran insects, amphipod crustaceans, but also humans, primarily with immunodeficiency. Anncaliia algerae-caused fatal myositis is considered as an emergent infectious disease in humans. A. (=Nosema, Brachiola) algerae, the best studied species of the genus, demonstrates the broadest among microsporidia range of natural and experimental hosts, but it has never been propagated in Drosophila. We present ultrastructural analysis of development of A. algerae in visceral muscles and adipocytes of Drosophila melanogaster 2 wk after per oral experimental infection. We observed typical to Anncaliia spp. features of ultrastructure and cell pathology including spore morphology, characteristic extensions of the plasma membrane, and presence of "ridges" and appendages of tubular material at proliferative stages. Anncaliia algerae development in D. melanogaster was particularly similar to one of A. algerae and A.(Brachiola) vesicularum in humans with acute myositis. Given D. melanogaster is currently the most established genetic model, with a fully sequenced genome and easily available transgenic forms and genomic markers, a novel host-parasite system might provide new genetic tools to investigate host-pathogen interactions of A. algerae, as well to test antimicrosporidia drugs., (© 2019 International Society of Protistologists.) more...

Scientific development has increased the awareness of water pollutant forms and has reawakened the need for its effective purification. Oxyanions are created by a variety of redox-sensitive metals and metalloids. These species are harmful to living matter due to their toxicity, nondegradibility, and mobility in aquatic environments. Among a variety of water treatment techniques, adsorption is one of the simplest, cheapest, and most effective. Since metal-oxide-based adsorbents poses a variety of functional groups onto their surface, they were widely applied in ions sorption. In this paper adsorption of harmful oxyanions by metal oxide-based materials according to literature survey was studied. Characteristic of oxyanions originating from As, V, B, W and Mo, their probable adsorption mechanisms and comparison of their sorption affinity for metal-oxide-based materials such as iron oxides, aluminum oxides, titanium dioxide, manganium dioxide, and various oxide minerals and their combinations are presented in this paper. more...

Although there is enough heat contained in inflowing warm Atlantic Ocean water to melt all Arctic sea ice within a few years, a cold halocline limits upward heat transport from the Atlantic water. The amount of heat that penetrates the halocline to reach the sea ice is not well known, but vertical heat transport through the halocline layer can significantly increase in the presence of double diffusive convection. Such convection can occur when salinity and temperature gradients share the same sign, often resulting in the formation of thermohaline staircases. Staircase structures in the Arctic Ocean have been previously identified and the associated double diffusive convection has been suggested to influence the Arctic Ocean in general and the fate of the Arctic sea ice cover in particular. A central challenge to understanding the role of double diffusive convection in vertical heat transport is one of observation. Here, we use broadband echo sounders to characterize Arctic thermohaline staircases at their full vertical and horizontal resolution over large spatial areas (100 s of kms). In doing so, we offer new insight into the mechanism of thermohaline staircase evolution and scale, and hence fluxes, with implications for understanding ocean mixing processes and ocean-sea ice interactions. more...

The electrochemical reduction of carbon dioxide is usually studied in aqueous solutions under ambient conditions. However, the main disadvantages of this method are high hydrogen evolution and low faradaic efficiencies of carbon-based products. Supercritical CO 2 (scCO 2 ) can be used as a solvent itself to suppresses hydrogen evolution and tune the carbon-based product yield; however, it has received little attention for this purpose. Therefore, the focus of this study was on the electrochemical reduction of scCO 2 . The conductivity of scCO 2 was increased through the addition of supporting electrolyte and a cosolvent (acetonitrile). Furthermore, the addition of protic solutions of different pH to scCO 2 was investigated. 1 m H 2 SO 4 , trifluoroethanol, H 2 O, KOH, and CsHCO 3 solutions were used to determine the effect on current density, faradaic efficiency, and selectivity of the scCO 2 reduction. The reduction of scCO 2 to methanol and ethanol are reported for the first time. However, methane and ethylene were not observed. Bioelectrochemical power-to-gas (BEP2G) is considered a potentially convenient way of storing renewable surplus electricity in the form of methane. In methane-producing bioelectrochemical systems (BESs), carbon dioxide and electrical energy are converted into methane, using electrodes that supply either electrons or hydrogen to methanogenic archaea. This review summarizes the performance of methane-producing BESs in relation to cathode potential, electrode materials, operational strategies, and inoculum. Analysis and estimation of energy input and production rates show that BEP2G may become an attractive alternative for thermochemical methanation, and biochemical methanogenesis. Magnesium imide (MgNH) was produced by monitoring the decomposition process of magnesium amide with in situ neutron diffraction. Significant changes in the structure of magnesium amide are detected during heat treatment and eventually result in the formation of crystalline MgNH. A model for the crystal structure of magnesium imide (MgNH) is presented for the first time. Remarkably, magnesium imide offers unique structural features similar to the cyclosilicate class and can be described as a porous solid formed by a sequence of linked chains of face sharing Mg(6)N(6) hexagonal prism clusters. more...

The improving factors brought by LiBH(4) were studied experimentally. Although a minimum in activation energy was observed in the middle stage of hydrogen desorption for the samples with and without LiBH(4) doping, a reduction in the activation energy was manifest in the overall conversion range due to the LiBH(4) introduction. In contrast to the pure 2LiH-Mg(NH(2))(2), the system component Mg(NH(2))(2) crystallized in the presence of LiBH(4) at about 150 °C in situ from the post-milled amorphous mixture prior to hydrogen desorption. We believe the formation of the N-atom matrix by the crystalline Mg(NH(2))(2) is relevant to the subsequent dehydrogenation as it shares a similar sublattice structure with the desorption product. The formation and disappearance of Li(4)(BH(4))(NH(2))(3) in the dehydrogenation step witnessed the capturing and releasing of LiNH(2) that is generated from the dehydrogenation of 2LiH-Mg(NH(2))(2), which alters not only the kinetics but also thermodynamics in favor of an improved hydrogen sorption performance. Neutron diffraction data from in situ deuteration and dedeuteration of Li(3)N are presented under different pressure regimes, whereby reaction pathways differing from the widely reported stoichiometric pathway of Li(3)N + 2D(2)<--> Li(2)ND + LiD + D(2)<--> LiND(2) + 2LiD are observed. At sufficiently high pressures, where the deuterium chemical potential is comparable with the heat of amide formation, the reaction appears to be driven straight to the amide plus deuteride phase mixture. At lower pressures, a cubic phase exhibiting a concentration-dependent variation in lattice parameter is observed. In dedeuteration, two sets of reflections from cubic structures with distinct lattice parameters are observed, both of which exhibit a continual decrease in cell volume. In spite of high prices for fossil raw materials the production of biomass-based products is rarely economically successful today. Depending on the location feedstock prices are currently so high that products from renewable resources are not marketable when produced in existing process chains. Apart from the higher feedstock costs one reason is that at present no optimized production systems exist in contrast to the chemical and petrochemical industry where these systems have been established over the last decades. If we succeed in developing production systems modelled on those of petroleum refineries where we can provide a flexible coupled production of energy, fuels, materials and chemicals chances are good to enable a lastingly successful production on the basis of renewable resources. Based on examples of fat-based and sugar-based concepts ideas for platform oriented biorefineries are outlined. more...

We present experimental results on the self-focusing of light beams in a photorefractive, optically active Bi12TiO20 crystal. Using circular polarization of the input HeNe laser beam we succeeded to create a stable self-focused, asymmetric light-beam over several hours. The southern green stink bug, Nezara viridula (L.), and certain of its host plants were examined to determine the prevalence and biological characteristics of an intestinal trypanosomatid. Promastigotes with short (< or = 17.5 microm excluding flagellum) and long forms (> or = 25.0 microm) usually infected < 50% of the bugs before August and > 50% (maximum 95%) during August-October, but prevalence was not host-density dependent. The flagellate was detected in adults and in all nymphal instars, at all sampling sites where at least 10 bugs were captured, and in bugs from all host plants sampled (soybean, red clover, vetch). Of bugs with flagellates, 27% were heavily infected (> 20 flagellates per 160X microscope field). Weights of infected and uninfected adults did not differ. Live flagellates were detected in bug feces and in one stem of red clover. When bugs were fed soybean pods, tomatoes, or snap beans in the laboratory, only once were flagellates detected in plant tissue (snap beans). The flagellate was cultured in modified Medium 199. This flagellate is prevalent in N. viridula populations in Louisiana and apparently does not cause significant pathological effects in N. viridula or its host plants, including soybean. more...

Inability to consistently rear healthy Trichoplusia ni led to a study of its rearing diseases. Four diseases were designated after preliminary research which included electron microscopy: cytoplasmic polyhedrosis (due to cytoplasmic polyhedrosis virus, or CPV), nuclear polyhedrosis (due to nucleopolyhedrovirus, or NPV), "neonate death" syndrome (mortality in first or second instars), and "late-instar" syndrome (death in late instars accompanied by bacterial decomposition). Infectious agents were not detected by electron microscopy in insects with the latter two diseases. Prevalence of CPV and NPV, but not the neonate-death or late-instar diseases, in progeny was significantly associated with pairs of mating adults. In conjunction with egg-surface decontamination, this indicated that both viruses may be transmitted transovarially. Pupae killed by CPV had virions in the cytoplasm of infected cells, but polyhedra were empty, not occluding virions. None of the diseases had a consistent pattern of prevalence associated with the date on which eggs were laid after oviposition began. Prevalence rates of cytoplasmic polyhedrosis, nuclear polyhedrosis, and the late-instar disease were significantly greater at 95-100% relative humidity (RH) than at RH levels of 75% or below. These same three diseases killed significantly more insects in crowded rearing conditions (four or five larvae per cup with 10.2-cm(2) diet surface) than in uncrowded conditions (one to three larvae per cup). Microsporidian spores of Trachipleistophora hominis Hollister, isolated from a human, readily infected larval stages of both Anopheles quadrimaculatus Say sensu lato and Culex quinque-fasciatus Say. Mosquito infections with T. hominis were located, primarily, in abdominal muscles in segment numbers 4 through 6; other spores were found in the hemocoel and proboscis. Nearly 50% of the infected mosquito larvae survived to the adult stage. Spores recovered from adult mosquitoes were inoculated into mice and resulted in significant muscle infection at the site of injection. Surface plaque matrix (PQM) and a tubular arrangement of filaments border Trachipleistophora hominis parasites during growth within host muscle. The PQM at the parasite surface forms a network of processes which can be associated with filamentous tubules. Peroxidase tracer delineated the PQM and showed apparent connections with the tubules. The tubules at the interface of T. hominis-infected cells are structurally similar to the extrasporular tubules of the microsporidian, Ameson michaelis. The extrasporular tubules of A. michaelis and the proteins from T. hominis-infected muscle reacted to keratin antibodies, K8.13, K4 and K13. Conversely, antibodies produced to T. hominis-infected muscle, reacted with the extrasporular tubular proteins of A. michaelis. Toxoplasma gondii, an intracellular protozoan parasite, resides within a host-derived vacuole that is rapidly modified by a parasite-secreted membranous tubular network. In this study we investigated the involvement of heterotrimeric G proteins in the secretory pathway of T. gondii. Aluminum fluoride (AlFn), a specific activator of heterotrimeric G proteins, induced secretion from isolated tachyzoites of T. gondii in vitro, as seen by light optics and electron microscopy. In Western blot analyses, antibodies to G protein alpha subunits reacted with 39-42 kDa proteins from T. gondii isolates. Antibodies to G(o) alpha and Gs alpha coupled to the fluorescent probe fluorescein isothiocyanate localized to the paranuclear region of T. gondii. Gi3 alpha immunoprobes were confined to the cytoplasmic matrix of T. gondii and also labeled the parasitophorous vesicle. Fluorescein isothiocyanate-conjugated GA/1, an antipeptide antisera directed toward the GTP binding site common to G protein alpha subunits, was confined to the lateral cytoplasmic domain of the parasites where secretion is most prominent. We detected no infections with species of Plasmodium, Haemoproteus, Leucocytozoon, Sarcocystis or Trypanosoma in blood smears, liver and spleen impressions, and muscle tissue from 136 redheads (Aythya americana) collected or captured at the Chandeleur Islands, Louisiana (USA), during three winters (1987 to 1990). One bird, a juvenile male, was infected with an unidentified species of microfilaria. Thus, we found no evidence that hematozoa had an effect on redheads during the course of the study. more...

The interaction between the Toxoplasma parasitophorous vacuole and vimentin-type intermediate filaments in Vero cells was investigated via immunofluorescence microscopy. A significant rearrangement of host cell vimentin around the Toxoplasma parasitophorous vacuoles occurs throughout the course of infection. Host cell vimentin associates with the parasitophorous vacuoles within an hour after invasion. This vimentin overcoating of the vacuole is initiated at the host cell nuclear surface. During parasite multiplication, vimentin retains a closely defined association with the cytosolic surface of the parasitophorous vacuole. In addition, the vimentin intermediate filaments originating from the host cell nuclear surface are progressively rearranged around the enlarging parasitophorous compartment. During infections, the order of vimentin cytoskeleton is normal throughout the cell and appears redefined only at the vicinity of the parasitophorous vacuole. Depolymerization of the intermediate filaments was achieved with the phosphatase inhibitors okadaic acid and calyculin A. Disruption of the intermediate filament networks resulted in displacement of the parasitophorous vacuoles from the host cell nuclear surface. The data indicate that host cell vimentin binds to the Toxoplasma parasitophorous vacuoles and that the host intermediate filament network serves to dock the parasite compartment to the host cell nuclear surface. more...

Treatment of Litomosoides carinii infected Mastomys natalensis with diethylcarbamazine (DEC: 500 mg/kg p.o.) was followed by increased occurrence of microfilariae in the bronchi of the host after 40 min and lasting at least until 6 h after treatment. After 4 h, increased levels of larvae were observed in the gut. Only a few microfilariae occurred in the bladder and sputum. Accumulations of microfilariae were found furthermore in the Lnn. hepaticae whereas no changes were observed in the inguinal or jejunal and lung and pleura associated lymph nodes. Increased numbers of microfilariae were found in the peritoneal cavity only after 8 and continuing until at least 48 h after treatment. In contrast, after haloxon treatment (100 mg/kg p.o.) an accumulation of microfilariae was found in the peritoneal cavity only, following a time course similar to that after DEC. more...

The Glugea stephani-induced xenoma in the winter flounder, Pseudopleuronectes americanus, is a large spherical host-parasite complex, up to 4.0 mm in diameter, with the host and parasite components of the xenoma being most active in the peripheral zone. The xenoma has an extensive periodic acid-silver methenamine-positive surface coat covering the plasma membrane. The surface of this membrane is amplified by the presence of numerous folds and fine tubular extensions. The peripheral zone of the xenoma contains many host-cell mitochondria in addition to numerous microsporidan parasites. At the ultrastructural level, the peripheral zone of the host-cell cytoplasm appears normal. Inside the peripheral region of the 0.4-1.0 mm xenoma, the host-cell component largely disintegrates in the presence of microsporidan parasites undergoing sporogenesis. more...

Encephalitozoon cuniculi grow within ever-increasing parasitophorous vacuoles (PV) in peritoneal macrophages. The PV boundary membrane conforms to a rich arrangement of blebs; similar, but free vesicles were observed within the PV space. An iron dextran-concanavalin A marker was used to express visually clustered distributions of Con A receptors on the PV boundary blebs and free vesicles; no marker was observed on other membrane surfaces within the PV. These results, combined with the observation that the PV grows while the host cytoplasm decreases in mass, implicate the PV boundary blebs of interiorizing into vesicles by a pinocytic mechanism. Phagocytic vacuoles, secondary lysosomes and pinocytic vesicles were labeled by incubating infected macrophages in minimum essential medium with ferritin. Ferritin readily accumulated in secondary lysosomes and phagocytic vacuoles; however, ferritin was excluded from parasitophorous vacuoles containing E. cuniculi. Acid phosphatase cytochemical reaction product was observed in lysosomes and phagocytic vacuoles; however, parasitophorous vacuoles with vegetative E. cuniculi were always negative. more...

Reaginic and homocytotropic IgG antibodies in sera using passive cutaneous anaphylaxis (PCA) test and antigen from Litomosoides carinii were followed in Mastomys natalensis, infected with L. carinii, Dipetalonema viteae, Brugia malayi or B. pahangi. Groups of animals with infections of various ages so as to cover a total infection period of up to 300 to 420 days post-infection (p.i.), depending on the species of parasites, were bled at 1- to 3-week intervals over periods of 50-112 days. In addition, intradermal tests were performed on animals infected with L. carinii to detect immediate type hypersensitivity. Reaginic antibodies were usually first detected in the 3rd week after infection. Thereafter, a marked increase of PCA titres was observed in the 4th week p.i. leading to maximum titres 4 weeks after infection with D. viteae and B. pahangi and 6 weeks after B. malayi infection. Mean maximum titres were between 1:40 and 1:160. Following the peak response, titres decreased markedly until the beginning of patency in infections with D. viteae, B. malayi and B. pahangi whereas a constant course was observed at this time in animals infected with L. carinii. A further rise in PCA titres occurred in all infections around the beginning of patency, resulting in maximum reagin levels in L. carinii infections (mean titre 1:80) and moderate titres in the other infections. During early patency there was an inverse relationship between microfilaraemia density and levels of reaginic antibodies. However, in the phase of decreasing parasitaemia in L. carinii infected animals, microfilariae counts and PCA titres were directly correlated. Homocytotropic IgG antibodies showed relatively constant PCA titres of about 1:20 in L. carinii infected Mastomys throughout the observation period. In D. viteae infections they were demonstrated at 30 days p.i., reaching titres of about 1:40. B. malayi infected animals showed a maximum titre of 1:40 40 days p.i.. Thereafter, titres decreased continuously and homocytotropic IgG antibodies were absent at 110 days p.i.. High titres were observed at day 150 but thereafter sera were negative. B. pahangi infected animals showed moderate titres (1:5) 35 days p.i.. Thereafter, antibodies were found at low titres until 115 days p.i.. This study examines the relationship between phagosome acidification and phagosome-lysosome fusion events using phagocytized Glugea hertwigi spores. The incidence of lysosome fusion with Glugea spores in phagosomes of mouse peritoneal macrophages and of Tetrahymena was monitored using colloidal gold and acridine orange as labels for secondary lysosomes. Over 80% of the Glugea phagosomes remained segregated from the labeled compartments in macrophages after 60 min; this inhibition of fusion was still evident after 4 h. In Tetrahymena, Glugea spores also showed a high capacity to block fusion with secondary lysosomes (67%); however, spores coated with cationized ferritin showed an 80% fusion rate with labeled acidic compartments (i.e. lysosomes) after 60 min with both Tetrahymena and macrophages. The pH of phagosome compartments was monitored by measuring the emissions of fluorescein isothiocyanate (FITC)-labeled Glugea ingested by Tetrahymena. Tetrahymena phagosomes with FITC-Glugea did not acidify within the first hour after phagocytosis; however, phagosomes with cationized ferritin-labeled Glugea underwent acidification during this time period. This acidification took place although the capability of the host cells' lysosomes to fuse was blocked by pretreatment with poly-D-glutamic acid. The cationized ferritin bound to Glugea spores was uncoupled from the spore wall prior to fusion with colloidal gold-labeled compartments. Catalase and superoxide dismutase detected in both RH and C strain Toxoplasma gondii tachyzoites were distinctly different in electrophoretic mobility from host cell enzymes. Catalase and superoxide dismutase activity levels were similar in both Toxoplasma strains and showed narrow pH optima around 8.0. Toxoplasma superoxide dismutase was resistant to cyanide but inhibited by azide or peroxide, consistent with an iron-containing enzyme typical of protozoan parasites. These enzymes may play a role in intracellular survival; however, they do not appear to be the basis for differences in virulence to mice. more...

Toxoplasma gondii belongs to a group of highly virulent intracellular parasites that reside in host cell vacuoles which resist typical phagosome-lysosome fusion. Live Toxoplasma replicate prodigiously within modified phagocytic vacuoles formed during invagination of the host plasma membrane. In contrast, heat-killed Toxoplasma or specific antibody (heat-inactivated)-coated live Toxoplasma-containing vacuoles readily undergo lysosome fusion and digestion in normal macrophages. Of newly recognized significance to Toxoplasma survival is the microbicidal effect of phagosome acidification, which reportedly can occur independently of fusion with other acidic vesicles. We report here that modified live Toxoplasma-containing vacuoles fail to acidify in normal macrophages, as indicated by the sensitive pH probe fluorescein. In contrast, when live Toxoplasma are coated with specific antibody (heat-inactivated), they trigger phagosome acidification when entering normal macrophages. A similar acidification is observed when normal phagocytes ingest dead Toxoplasma. Extracellular Toxoplasma are highly susceptible to acidic pH conditions, indicating that the acidification block in the modified vacuoles may be important for intracellular survival. more...
