Abstract

Clinical, physiological and environmental conditions of decreased PaO2 increase carbohydrate metabolism. Decreased PaO2 increases limb glucose uptake decreases the percentage of lactate obtained from blood glucose. Tissues other than skeletal muscle increase blood lactate and pyruvate when PaO2 decreases. A decrease in free fatty acid levels shifts the mechanism toward increased metabolic carbohydrates.

Abstract

Recession in insulin levels during exercise enhances glucose production by stimulating the actions of another hormone, glucagon. The fall in insulin below minimum threshold level increases the hepatic glucose production even when glucagon is suppressed. The effect of insulin on glucose utilization and stimulation of glucagon action is not linear. The decrease in insulin does not affect muscle glucose uptake and metabolism.

Abstract

A study of the influence of chronic selective insulin deficiency on gluconeogenesis and glycogenolysis during acute physiological hypercortisolemia in conscious dogs which are fasted overnight reveals that deficiency of insulin influences glycogenolysis more significantly than gluconeogenesis during acute hypercortisolemia. Increase in glycogenolysis during insulin deficiency plays an important role in worsening the hyperglycemia.

Abstract

Study of the effects of a single sitting of exercise on hexokinase II (HK II) mRNA and HK II activity in male rats revealed an increase in skeletal muscle HK II mRNA in response to exercise. Total HK activity increased greatly in soleus and gastroconmius/plantaris after 8 and 24 hours of exercise. Heat treated homogenates showed increased HK I activity and exercise did not influence heat stable HK activity.

Abstract

The hepatic nerves can modulate hepatic glycogenolysis and glycogenesis and thus might be expected to be involved in the response of the animal to the transition from the fed to the food-deprived state. Therefore the arterial concentrations and net hepatic balance of glucose and its metabolites, as well as the hepatic glycogen concentrations, were compared in hepatic-innervated and -denervated dogs 18, 24 and 42 h after their usual daily meal. Arterial concentrations of glucose, alanine, lactate and glycerol; net hepatic balances of glucose, alanine and glycerol; and glycogen concentrations were similar in hepatic-innervated and -denervated dogs at each time investigated. Net hepatic balances of lactate (with negative values indicating uptake) in hepatic-innervated and -denervated dogs, respectively, were: 18 h, 4.1 +/- 4.3 vs. -4.3 +/- 3.6 microns mol center dot kg to the -1 center dot min to the -1; 24 h, 4.8 +/- 3.6 vs. -6.7 +/- 1.7 micron mol center dot kg to the -1 center dot min to the -1 (P < 0.05); 42 h, -7.0 +/- 2.0 vs. -6.8 +/- 1.0 micron mol center dot kg to the -1 center dot min to the -1. Based on changes in net hepatic lactate balance, the denervated liver responds more rapidly to food deprivation than the innervated liver, but the metabolic state of the liver appears similar by 42 h after a meal.

Abstract

Catheterized, depancreatized (PX) and normal (C) dogs were studied to estimate insulin-deficient gluconeogenesis regulation. Hepatic alanine uptake, arterial levels, hepatic concentrations and net gut output were unaltered during rest in C and PX. During exercise, all factors increased in PX, with only a fall in alanine concentration in C. Lactate induced gluconeogenesis was lesser in C than in PX at rest due to the enhanced efficiency of intrahepatic gluconeogenesis. Hepatic load increase and larger output of gut lactate and alanine also accounted for enhanced gluconeogenesis in the diabetic state.

Abstract

A study of the effects of chronically elevated plasma cortisol level on glycogenolysis and gluconeogenesis is presented. The study protocol included the use of dyes to monitor glucose production and continuous hydrocortisone infusions of ovenight-fasted dogs. The results showed that cortisol promotes hepatic gluconeogenesis by enhancing substrate delivery to the liver, raises plasma insulin levels to inhibit intrahepatic gluconeogenesis and enhances glucose cycling through glycogen. It also inhibits non-hepatic glucose utilization. These effects indicate a role for cortisol in stress response.

Abstract

As the knowledge base and importance of mitochondrial physiology to human health expands, the necessity for harmonizing the terminology concerning mitochondrial respiratory states and rates has become increasingly apparent. The chemiosmotic theory establishes the mechanism of energy transformation and coupling in oxidative phosphorylation. The unifying concept of the protonmotive force provides the framework for developing a consistent theoretical foundation of mitochondrial physiology and bioenergetics. We follow guidelines of the International Union of Pure and Applied Chemistry (IUPAC) on terminology in physical chemistry, extended by considerations of open systems and thermodynamics of irreversible processes. The concept-driven constructive terminology incorporates the meaning of each quantity and aligns concepts and symbols with the nomenclature of classical bioenergetics. We endeavour to provide a balanced view of mitochondrial respiratory control and a critical discussion on reporting data of mitochondrial respiration in terms of metabolic flows and fluxes. Uniform standards for evaluation of respiratory states and rates will ultimately contribute to reproducibility between laboratories and thus support the development of databases of mitochondrial respiratory function in species, tissues, and cells. Clarity of concept and consistency of nomenclature facilitate effective transdisciplinary communication, education, and ultimately further discovery.

Despite this, ischemia-induced death of transplanted cells remains a major hurdle to the efficacy of treatment. ‘Superhealer’ MRL/MpJ mesenchymal stem cells (MRL-MSCs) have been reported to exhibit increased engraftment resulting in reduced infarct size and enhanced contractile function. This study determines whether intrinsic differences in mitochondrial oxidative phosphorylation (OXPHOS) assist in explaining the enhanced cellular survival and engraftment of MRL-MSCs. Findings Compared to wild type MSCs (WT-MSCs), mitochondria from intact MRL-MSCs exhibited an increase in routine respiration and maximal electron transport capacity by 2.0- and 3.5-fold, respectively. When routine oxygen utilization is expressed as a portion of maximal cellular oxygen flux, the MRL-MSCs have a greater spare respiratory capcity. Additionally, glutamate/malate succinate-supported oxygen consumption in permeabilized cells was elevated approximately 1.25- and 1.4-fold in the MRL-MSCs, respectively. Conclusion The results from intact and permeabilized MSCs indicate MRL-MSCs exhibit a greater reliance on and capacity for aerobic metabolism. The greater capacity for oxidative metabolism may provide a protective effect by increasing ATP synthesis per unit substrate and prevent glycolysis-mediated acidosis and subsequent cell death upon transplantation into the glucose-and oxygen-deprived environment of the infarcted heart.

Abstract

To test whether hepatic insulin action and the response to an insulin-induced decrement in blood glucose are enhanced in the immediate postexercise state as they are during exercise, dogs had sampling (artery, portal vein, and hepatic vein) catheters and flow probes (portal vein and hepatic artery) implanted 16 days before a study. After 150 min of moderate treadmill exercise or rest, dogs were studied during a 150-min hyperinsulinemic (1 mU * [kg.sup.-1] * [min.sup.-1]) euglycemic (n = 5 exercised and n = 9 sedentary) or hypoglycemic (65 mg/dl; n = 8 exercised and n = 9 sedentary) clamp. Net hepatic glucose output (NHGO) and endogenous glucose appearance ([R.sub.a]) and utilization ([R.sub.d]) were assessed with arteriovenous and isotopic ([3-[sup.3]H]glucose) methods. Results show that, immediately after prolonged, moderate exercise, in relation to sedentary controls: 1) the glucose infusion rate required to maintain euglycemia, but not hypoglycemia, was higher; 2) [R.sub.d] was greater under euglycemic, but not hypoglycemic conditions; 3) NHGO, but not [R.sub.a], was suppressed more by a hyperinsulinemic euglycemic clamp, suggesting that hepatic glucose uptake was increased; 4) a decrement in glucose completely reversed the enhanced suppression of NHGO by insulin that followed exercise; and 5) arterial glucagon and cortisol were transiently higher in the presence of a decrement in glucose. In summary, an increase in insulin action that was readily evident under euglycemic conditions after exercise was abolished by moderate hypoglycemia. The means by which the glucoregulatory system is able to overcome the increase in insulin action during moderate hypoglycemia is related not to an increase in [R.sub.a] but to a reduction in insulin-stimulated [R.sub.d]. The primary site of this reduction is the liver. liver; metabolism; oxidation; glucose; lactate; nonesterified fatty acids
