Abstract

The reserve of primordial follicles, which serves all oocytes for the female reproductive lifespan, is established a few days after birth in mice. During this process, more than half of the oocytes are primarily eliminated by apoptosis. Autophagy, the conserved intracellular process maintaining cellular homeostasis, serves as a protective mechanism for oocyte survival. In the current study, we speculate a new role for autophagy during primordial folliculogenesis. Active autophagy was observed in perinatal ovaries from 16.5 days post coitus to 3 days post parturition. The inhibition of autophagy by 3-methyladenine (3-MA) increased the number of cyst oocytes and delayed follicle formation in vivo and in organ cultures. Furthermore, the reactive oxygen species (ROS) level was elevated in ovaries treated with 3-MA, while N-acetylcysteine, an oxidant, alleviated the inhibitory effect of 3-MA on primordial folliculogenesis. Additionally, the expression of growth differentiation factor 9 and transforming growth factor β1, which regulates follicle activation, was decreased after 3-MA treatment. These data suggest that the physiological level of autophagy in perinatal ovaries regulates germ cell cyst breakdown and primordial follicle assembly by ROS clearance and exerts extensive effects on further follicular development.

Abstract

Despite being used as an anti‐leukemic drug, the poor solubility of 6‐mercaptopurine (6‐MP) limits its use in topical and parenteral applications. Dendrimers are commonly used as drug carriers to improve their solubility in aqueous solution. In this work, the interactions between 6‐MP and the amine‐terminated poly(amidoamine) dendrimers (PAMAM−NH2) were investigated by various NMR technology. The chemical shift titrations disclosed that the 6‐MP interacted with the surface of PAMAM−NH2 mainly through electrostatics. The determination of diffusion coefficient and relaxation measurements further confirmed the presence of interactions in 6‐MP/PAMAM−NH2 complexes. In addition, the encapsulation of 6‐MP within the cavity of PAMAM−NH2 was revealed through nuclear Overhauser effect spectroscopy and Saturation Transfer Double Difference analysis. Finally, the binding strength (H‐8 is 100% and H‐2 is 70%) of 6‐MP to PAMAM−NH2 was quantitatively expressed using epitope maps. Abstract

The wide application of melamine–formaldehyde resins (MFRs) in the food industry aggravates plastic pollution problems. Recycling of the waste MFRs remains a big challenge due to its compact structure. Herein, a high MFR degradation ratio of ca. 84 wt% was achieved under mild conditions (180 °C) in a methanesulfonic acid (MSA)–tetrahydrofuran (THF)–H2O system with cyanuric acid as the main degradation product. The SEM results showed that the degradation of MFRs followed different reaction mechanisms when H2O and THF were applied as solvents, respectively. The combination of the two solvents showed an obvious synergistic effect for the degradation of MFRs. The degradation experimental results with melamine as the model intermediate indicated that the degradation of MFRs proceeded via the selective cleavage of the sp2C–sp3N and sp3C–sp3N bonds to recover valuable C and N alternated-aromatic ring (CNAR) units, followed by their transformation into a valuable chemical of cyanuric acid. 181. Elevated Insulin Levels Compromise Mouse Decidualization with Decreases on Uterine Endometrial Apoptosis in Early-Stage Pregnancy

Abstract

Objective: Compromised reproduction was a serious problem in women with hyperinsulinism and insulin resistance. However, the mechanism is not clear. Women with hyperinsulinism and insulin resistance have reduced fertility, but the underlying mechanism is still not known. Aberrant endometrial decidualization in early pregnancy has been linked to pregnancy complications, including infertility and recurrent miscarriage. Methods: C57BL/6J mice were randomly divided into normal control group and high insulin-exposed group. High insulin-exposed group were given a subcutaneous injection of human recombinant insulin. After insulin treatment, mice were sacrificed in early pregnancy. Serum and uterine tissues were collected for analysis. ELISA were used to evaluate changes in blood glucose and various hormones. Western blotting, immunohistochemical staining and quantitative measurement were used to evaluate the uterine decidualization and endometrial apoptosis in early pregnancy mice. For in vitro studies, decidualization was induced in mESCs and T-HESCs from control and high-insulin-exposed group. Western blotting was used to examine the protein expressions of apoptosis and PI3K/Akt signalling pathway related proteins. Results: Serum levels of insulin, E2, P4, LH, FSH and blood glucose were significantly altered in mice treated with insulin. Endometrial decidualization in early pregnancy was impaired by high insulin levels. Furthermore, endometrial apoptosis which is essential for decidualization was also markedly decreased. Suppression of insulin signaling by blocking insulin receptor reverses these alterations. Importantly, activated PI3K/Akt was found to have associated with decreased apoptosis during compromised decidualization by high insulin levels in vitro and in vivo. Conclusion: High insulin levels suppress endometrial apoptosis by PI3K/Akt signaling pathway, which further impairs decidualization in early pregnancy. 182. Probing the interactions of GlcNH2 with boric acid via NMR spectroscopy.

Abstract

The essential role of boronic esters in controlling both the direction and selectivity of chemical reactions as well as their significant function in catalytic activity have been demonstrated for industrially important processes. The specific interaction analyses of the monosaccharide GlcNH2 with boric acid are of interest since monosaccharides serve as model systems for the more sophisticated carbohydrate molecules. The interaction of GlcNH2 with boric acid was systematically investigated by numerous NMR techniques. A 1 : 1 chelate boron complex coordinated at the cis-1,2 position of GlcNH2 was identified as the major species in DMSO-d6 solution via1H and 13C INEPT DOSY NMR spectroscopy. This specific boron nitrogen coordination mechanism was further supported by the 1H–15N HSQC spectra. Variations in the spin–lattice relaxation times (T1) of the 13C1 nucleus also provided quantitative data regarding this non-covalent interactions. This is an application of 1H, 13C INEPT DOSY, 1H–15N HSQC, and relaxation methods to study such aggregations in solutions. 199. The Relationship between Alu I Polymorphisms in the Calcitonin Receptor Gene and Fluorosis Endemic to Chongqing, China

Abstract

Objective This study explored the association between an Alu I polymorphism at position 1,377 of the calcitonin receptor (CTR) gene and endemic fluorosis. Subjects and Methods A case-control study of 321 participants was conducted in regions with high fluorosis rates (Wushan and Fengjie counties) and those without high fluorosis rates (Yubei Qu county; termed nonfluorosis areas) in Chongqing, China. The participants were divided into three groups: the fluorosis group (FG) from areas with high fluoride exposure (121), the nonfluorosis group (NFG) from areas with high fluoride exposure (130), and a control group (CG) from areas with no excessive fluoride exposure (70). An Alu I polymorphism in the CTR gene was genotyped using polymerase chain reaction-restriction fragment length polymorphism analysis. Results The genotype distributions within each group were as follows: CC 60.33% (73/121), CT 30.58% (37/121) and TT 9.09% (11/121) for the FG; CC 74.62% (97/130), CT 21.54% (28/130) and TT 3.85% (5/130) for the NFG, and CC 68.57% (48/70), CT 31.43% (22/70) and TT 0% (0/70) for the CG. Significant differences in Alu I genotypes were observed among the groups (χ2 = 12.317, υ = 4, p = 0.015). Allele frequencies of CTR genotypes differed significantly among the groups (χ2 = 8.859, υ = 2, p = 0.012): C 75.62% (183/242) and T 24.38% (59/242) for the FG, C 85.38% (222/260) and T 14.62% (38/260) for the NFG, and C 84.29% (118/140) and T 15.71% (22/140) for the CG. Conclusion An association between fluorosis and the Alu I polymorphism in the CTR gene was observed in fluoride-exposed populations.

200. Ternary deep eutectic solvents catalyzed d-glucosamine self-condensation to deoxyfructosazine: NMR study

Abstract

Ternary deep eutectic solvents (TDESs) comprising choline chloride (ChCl), glycerol and l-arginine were synthesized as catalysts and solvents for the conversion of d-glucosamine (GlcNH2) into deoxyfructosazine (DOF). The interactions between these three components in the prepared TDESs were studied by 1H-, 35Cl-NMR spectra and 1H diffusion-ordered spectroscopy (DOSY) measurements. The chemical shift changes of active hydrogen in the 1H-NMR spectra of TDES system and widening of signals in the 35Cl-NMR spectra confirmed the hydrogen bonding interaction between the components, which was further supported by the decrease of diffusion coefficients (D) of the TDES components according to 1H DOSY measurements. The influences of reaction temperature and l-arginine content in the TDESs on the yield of DOF were also studied. The experimental results have shown that when the molar ratio of ChCl, glycerol, and l-arginine was 1:2:0.1, DOF was the major product with a yield of 22.6% at 90 °C for 120 min. The chemical shift titration indicated that the carboxyl group of l-arginine in the TDES is the catalytical active site, so the mechanism of the catalytic reaction between GlcNH2and the TDES was proposed. Moreover, a reaction intermediate, dihydrofructosazine, was identified in the self-condensation reaction of GlcNH2by an in situ1H NMR technique.
