Abstract

Microbial metabolism directly participates in soil nutrient recycling and ecosystem stability. Although the rhizosphere soil is one of the most active habitats, information on how microorganisms acquire nutrients based on ecoenzymatic stoichiometry is lacking, especially for major food crops under field conditions. Thus, this study aimed to explore the soil nutrient properties and microbial biomass concentrations as well as extracellular enzymatic activities and focused on carbon (C), nitrogen (N), and phosphorus (P) cycling of different crop types, including potato (Solanaceae), maize (cereal), soybean (Leguminosae), and buckwheat (Polygonaceae) in the arid area of northern Shaanxi, China. Microorganisms in the rhizosphere soil of four crops were subjected to relative C and N limitation tests based on ecoenzymatic activities. Among the crops, potato and soybean exhibited the maximum limitations. Linear regression analysis of soil nutrients, microorganisms (stoichiometric homeostasis), and threshold elemental ratio synthetically supported microbial metabolic limitations. This finding coincided with the highest and lowest microbial carbon use efficiencies in soybean (0.58) and potato (0.51), illustrating the distinguishing physiological responses of microbes. Partial least squares path modeling further confirmed that the soil microbial nutrient limitations were significantly regulated by soil nutrient properties and extracellular enzyme activities. Taken together, soil microbial metabolism in agricultural ecosystems was co‐limited by relative C and N regardless of the main food crop families in this region in China (northern Loess Plateau). These observations clarified the nutrient cycling driven by soil elemental stoichiometry at the root–soil interface in arid and oligotrophic ecosystems. [ABSTRACT FROM AUTHOR]

164. Extraction, identification and bioactivities of polyphenols from green tea dust.

Abstract

Green tea dust was used as raw material in this study. The polyphenols in the green tea dust were extracted by a solid-liquid ratio of 1:20 g/mL, extraction temperature of 85 °C, ethanol concentration of 60% and extraction time of 60 min, and solvents with different polar were employed to extract and separate and the bioactive components in the green tea dust into 4 fractions. The mass concentration of total phenols contained in the different fractions was determined, and the bioactivates of different fractions were evaluated through free radical scavenging assay, and their inhibitory activities on pancreatic lipase and α-amylase were also studied. The results showed that the ethyl acetate extracted fraction had the highest total polyphenol content (15.66 mg/mL) and the strongest bioactivities among all the fractions. Its IC50 for the free radical scavenging rate of DPPH and ABTS were 0.05 mg/mL and 0.15 mg/mL, respectively, and inhibitory activities on pancreatic lipase and α-amylase were IC50 0.89 mg/mL and 0.39 mg/mL, respectively. The polyphenols in the ethyl acetate extracted phase were further characterized and quantified by HPLC and MS. A total of 15 polyphenolic compounds were identified by MS and was first identified of phlorizin (5.44 mg/g) in green tea dust. Quantitative analysis of the compounds with high contents by HPLC revealed that epigallocatechin, catechin and chlorogenic acid were present at high contents of 72.03 mg/g, 48.62 mg/g and 41.58 mg/g respectively. Abstract

A high ferroelectric Curie temperature TC, a large piezoelectric coefficient d33 and resistivity at high temperatures are three virtues that must be considered for piezoelectric materials with potential applications in high temperature sensors. In this paper, the effect of the B2O3 sintering aid on the microstructural and electrical properties of CaBi2Nb2O9 is comprehensively investigated. X-ray and neutron powder diffraction combined with Rietveld refinement demonstrate that a trace amount of B3+ can be incorporated into the Nb-sites in Nb–O octahedra resulting in an increase in the lattice distortion. The addition of B2O3 induces the preferential growth of grains along the c-axis, as confirmed by the corresponding (001) pole figure. The enhanced distortion degree of the Nb–O octahedra and microstructural grain orientation give rise to improvements in the ferroelectric and piezoelectric properties of CaBi2Nb2O9. The polycrystalline sample with the addition of 0.25 wt% B2O3 can realize a large d33 of 20.4 pC N−1 and an ultra-high electrical resistivity of 1.4 × 107 Ω cm (at 600 °C), while still retaining a high TC of 958 °C. These properties are comparable to those of most piezoelectric single crystals used at high temperature. Abstract

Dextranase specifically hydrolyzes dextran and is used to produce functional isomalto-saccharide prebiotics. Moreover, dextranase is used as an additive in mouthwash to remove dental plaque. We cloned and expressed the dextranase gene of the marine bacterium Bacillus aquimaris S5. The length of the BaDex gene was 1788 bp, encoding 573 amino acids. Using bioinformatics to predict and analyze the amino acid sequence of BaDex, we found the isoelectric point and instability coefficient to be 4.55 and 29.22, respectively. The average hydrophilicity (GRAVY) was −0.662. The secondary structure of BaDex consisted of 145 alpha helices, accounting for 25.31% of the protein; 126 extended strands, accounting for 21.99%; and 282 random coils, accounting for 49.21%. The 3D structure of the BaDex protein was predicted and simulated using SWISS-MODEL, and BaDex was classified as a Glycoside Hydrolase Family 66 protein. The optimal temperature and pH for BaDex activity were 40°C and 6.0, respectively. The hydrolysates had excellent antioxidant activity, and 8 U/mL of BaDex could remove 80% of dental plaque in MBRC experiment. This recombinant protein thus has great promise for applications in the food and pharmaceutical industries.

Abstract

The immobilization technology provides a potential pathway for enzyme recycling. Here, we evaluated the potential of using dextranase immobilized onto hydroxyapatite nanoparticles as a promising inorganic material. The optimal immobilization temperature, reaction time, and pH were determined to be 25 &deg, C, 120 min, and pH 5, respectively. Dextranase could be loaded at 359.7 U/g. The immobilized dextranase was characterized by field emission gun-scanning electron microscope (FEG-SEM), X-ray diffraction (XRD), and Fourier-transformed infrared spectroscopy (FT-IR). The hydrolysis capacity of the immobilized enzyme was maintained at 71% at the 30th time of use. According to the constant temperature acceleration experiment, it was estimated that the immobilized dextranase could be stored for 99 days at 20 &deg, C, indicating that the immobilized enzyme had good storage properties. Sodium chloride and sodium acetic did not desorb the immobilized dextranase. In contrast, dextranase was desorbed by sodium fluoride and sodium citrate. The hydrolysates were 79% oligosaccharides. The immobilized dextranase could significantly and thoroughly remove the dental plaque biofilm. Thus, immobilized dextranase has broad potential application in diverse fields in the future.

Abstract

Vaterite, the least stable phase among three anhydrous polymorphs of calcium carbonate (CaCO3), was prepared via the reaction between the ethanol solution of calcium chloride (CaCl2) and the aqueous solution of sodium carbonate (Na2CO3), which is named ethanol-calcium method. The effects of aging times and reaction temperatures on the formation of vaterite were investigated. The polymorphs and morphologies were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM), respectively, and Fourier transform infrared spectroscopy (FT-IR) was used to verify the existence of vaterite. XRD results indicate that the amount of vaterite decreases from 90.4% to 81.4% as increasing in aging times from 0 min to 42 h and decreases from 85.8% to 70.2% as increasing in reaction temperatures from 0 °C to 60 °C. SEM results show that vaterite and calcite as-prepared are their typical morphologies of spherical and rhombohedral, respectively. This research extends the route to prepare the metastable vaterite and provides new insights into its controllable synthesis.

The anti-tumor effects and potential mechanism of Lip E7/CpG were assessed by assays of tumor growth inhibition, immune cells, in vivo cytotoxic T lymphocyte (CTL) responses and cytokines, chemokines, CD31, Ki67 and p53 expression in the TME. In addition, toxicity of Lip E7/CpG to major organs was evaluated.Results: Lip E7/CpG had a diameter of 122.21± 8.37 nm and remained stable at 4°C for 7 days.
