Abstract

A CW,LD pumped Nd∶YVO4,532 nm green laser with high stability output power of 144 mW was reported.The laser stability was achieved by double closed-loop feedback system,which provided the high accuracy temperature control of LD and KTP crystal by thermoelectric cooler(TEC) and feedback control of laser power.The accuracy of temperature control reached the level of ±0.1 ℃ in the experiment.Meanwhile,the real-time feedback of LD output power was based on the error variance of nonlinearity curve by Si-photo detecting variables with LD current.Threshold mode PI control algorithm could reduce the stable error and their output signals were used to adjust LD pump current source as so to gain the output power stability.The long-term stability was excellent with ±1% variation in the output power.

166. Isolation and Identification of Representative Fungi from Shaoxing Rice Wine Wheat Qu Using a Polyphasic Approach of Culture-Based and Molecular-Based Methods

Abstract

Ribosomal intergenic spacer analysis (RISA) and traditional culture-based methods were employed to examine the fungal community of Shaoxing rice wine wheat Qu. Results showed that RISA profiles of total DNA exhibited nine distinguishable bands. By comparison, the RISA fingerprints recovered from enrichment media gave variable patterns containing fewer bands. Bands corresponding to five fungal species detected by RISA of total DNA were also observed in RISA fingerprints from enrichment cultures. A total of twenty-four pure cultures were isolated with media of potato dextrose agar (PDA), Czapek (CPK) and wheat Qu extraction (WQE). A total of eight, eighteen and eleven fungal species and 7.9 × 105 cfu/g Qu, 3.0 × 105 cfu/g Qu, and 3.9 × 106 cfu/g Qu, were isolated using PDA, CPK and WQE media, respectively. Predominance of each representative species did vary with the culture medium. Comparison of ITS sequences of excised RISA bands and recovered pure isolates with those in GenBank database revealed that the representative fungal species in wheat Qu were Absidia corymbifera, Saccharomyces cerevisiae, Rhizopus oryzae, Rhizomucor pusillus, Aspergillus oryzae, Candida tropicalis, Emericella nidulans, Clavispora lusitaniae, Aspergillus fumigatus, Aspergillus niger, Rhizopus microsporus, Pichia guilliermondii and Pichia anomala. Species of Saccharomyces cerevisiae, Candida tropicalis and Rhizopus oryzae detected in the original samples by analysis of RISA were not recovered in any of the three media. In addition, some species recovered from the enrichments were not detected by RISA analysis. Results obtained with culture-based or molecular-based methods clearly confirmed the importance of developing an integrated approach to gain a better understanding of the microbial community in wheat Qu.

Abstract

Abstract: Interleukin (IL)‐33/ST2 pathway plays crucial roles in tumour growth and metastasis. The aim of this study was to investigate the association of two functional polymorphisms (IL‐33 rs7025417 and ST2 rs3821204) with osteosarcoma (OS) risk. The rs7025417 and rs3821204 were genotyped by Taqman assay. IL‐33mRNA and protein levels were measured by real‐time PCR or enzyme‐linked immunosorbent assay. The luciferase activity was measured by a dual luciferase reporter gene assay. The allele‐specific transcription factor binding for rs7025417 was examined by ChIP‐seq. The IL‐33 rs7025417 CC genotype was significantly associated with a decreased risk of OS (CC vs TT: OR = 0.59, 95% CI, 0.41‐0.85; recessive model: OR = 0.68, 95% CI, 0.49‐0.94; C vs T: OR = 0.76, 95% CI, 0.63‐0.91). Combined analysis showed that the IL‐33 rs7025417CT/CC‐ST2 rs3821204CG/CC and the IL‐33 rs7025417CT/CC‐ST2 rs3821204GG genotypes also had a decreased risk of OS. IL‐33mRNA and protein levels in OS patients were significantly higher than controls. Patients with the rs7025417 CC genotype exhibited lower levels of IL‐33 (P = .03). The rs7025417 C allele presented a lower transcriptional activity by disrupting the binding site to c‐Myb (P < .01). Moreover, the rs3821204 G/C influences the transcriptional activity and ST2mRNA expression by altering the binding site of miR‐202‐3p. These findings suggest that the rs7025417 and rs3821204 may have a combined effect to protect against the development of OS by decreasing the expression levels of IL‐33 or ST2. Abstract

Abstract: The current study evaluated antioxidant activity of Dracocephalum rupestre Hance, characterized by the polyphenolic compounds in the ethyl acetate fraction (EAF) from D. rupestre and investigated the protective mechanisms of EAF in carbon tetrachloride (CCl4)‐induced hepatic injury. EAF showed the largest antioxidant capacity as demonstrated by DPPH, ABTS, OH, and FRAP assays (p ≤ .05). Using LC–MS, two polyphenolics, for example, rosmarinic acid, and eriodictyol were identified in EAF. Total phenol content of D. rupestre was correlated significantly with FRAP value (r = .999, p ≤ .001). The results also showed that EAF decreased serum ALT and AST activities compared with model group, as well as the histological findings. In addition, EAF remarkably decreased MDA and LDH levels in liver. This is the first time to reveal the hepatoprotective effect of EAF from D. rupestre, which may be developed as a new drug for treatment of liver injury in future. Practical applications: Dracocephalum rupestre Hance is known as Chinese herbal medicine with pharmacological functions of antioxidant, anti‐inflammatory and has been mainly used in the treatment of damp‐heat, headache, fever, jaundice, hepatitis, liver toxicity, and other diseases. In order to detect the potential application of this plant, the antioxidant and hepatoprotective activities of D. rupestre were investigated. It was found that the ethyl acetate fraction (EAF) from D. rupestre displayed strong antioxidant activity. We also demonstrate that EAF could lessen CCl4‐induced acute liver injury in mice. Abstract

Ontology plays an important part on Semantic Web, Information Retrieval, and Intelligent Information Integration etc. Ontology learning gets widely studied due to many problems in totally manual ontology construction. Term extraction influences many respects of ontology learning as it's the basis of ontology learning hierarchical structure. This paper mines topics of the corpus based on Latent Dirichlet Allocation (LDA) which uses Variational Inference and Expectation-Maximization (EM) Algorithm to estimate model parameters. With the help of irrelevant vocabulary, the paper provides better experimental results which show that the distribution of topics on terms reveals latent semantic features of the corpus and relevance among words.

Abstract

Response surface methodology (RSM) was used to study the effect of liquid: solid ratio, cellulase concentration and reaction time on the extraction of polysaccharide from Fructus tribuli. Regression analysis was performed on the data obtained. The most relevant variable was solid: liquid ratio. The coefficient determination (R2) was good for the second-order model. The optimal condition of polysaccharides extraction from Fructus tribuli was found, these were a liquid: solid ratio of 24.36:1 (v/w), a cellulase concentration of 1.78% and a reaction time of 2.84h. By means of additional experiments, the adequacy of this model was confirmed.

Abstract

To explore the mechanism of fatty liver formation induced by high non-protein energy diets in grass carp ( Ctenopharyngodon idella), basal diet and high-energy diets were fed to juvenile grass carp for 9 weeks. The experimental groups fed on high-energy diets which included a high-lipid diet (H- LIP), a high-carbohydrate diet (H- CHO) and a high-lipid and carbohydrate diet (H- CL). The control group fed on basal diet. Growth performance, liver fat accumulation, serum biochemical indexes and the expression levels of lipid metabolism-related genes ( SREBP-1, PPARγ, FAS, ACC1, and LPL) and mi RNAs (miR-33, miR-122, and miR-370) were examined at the end of the feeding trial. There were no significant differences in growth rate and feed efficiency among the four groups. However, significant increase in mesenteric and liver fat contents, and lipid droplets in the liver was induced by high-lipid and high-carbohydrate diets. There were significant differences in serum biochemical indicators such as AST/ ALT, GLB, TG and TP, and liver fatty acid composition between the control and experimental groups. The expression levels of SREBP-1, PPARγ, FAS, ACC1 and LPL were upregulated, while CPT-1 was downregulated with the high-energy treatments. Additionally, the expression levels of miR-33, miR-122 and miR-370 in the liver were higher in the three high-energy treatments than those in the control ( P < 0.05). The results suggest that modifications of lipid metabolism-related genes and mi RNAs may be involved in fatty liver formation induced by high non-protein energy diets in grass carp. Abstract

This paper introduces a method for automatic composition of Semantic Web services. First, according to OWL-S ServiceProfile, we define formal Web service model for meaningful matchmaking through the use of composition rules. These rules compare the syntactic and semantic features of Web services to determine whether two services are composable. Then, Petri net is used to model for composite service and its precedence relationship, also the relationship between composite service and T invariant is built. Therefore, the composition of Web services can be achieved by solving the Tinvariant.
