Abstract

Organic reactions in water have attracted great attention due to their advantages such as unique reaction performance and environmental friendliness. Organic reactions as well as polymerizations in aqueous media have been extensively investigated, and so far, there has been a massive amount of reporting about polymerizations in water. However, reports about click polymerization in water have been rare. Herein, click polymerization of activated alkyne and aromatic amine in aqueous media is developed. The “on water” effect facilitates polymerization in aqueous media better than in organic solvents, and its mechanism is deciphered through experimental data and theoretical calculations. Water participates in the reaction and reduces the energy barrier to some extent. Besides, polymerization makes it possible for aromatic amine with low reactivity to be linked. By using this strategy, polymers with high molecular weights can be obtained in high yields (up to 95.4%). They show good thermal stability and high refractivity. They can be photodegraded. The polymers with tetraphenylethylene moieties show aggregation-induced emission and can be used as materials for generating photopatterns and visualizing agents for specific staining of lysosome in living cells.

Abstract

Multifunctional nanoaggregates are widely used in cancer phototheranostics. However, it is challenging to construct their multifunctionality with a single component, and deliver them rapidly and efficiently without complex modifications. Herein, a NIR-absorbing small molecule named TBT-2(TP-DPA) is designed and certify its theranostic potentials. Then, their nanoaggregates, which are simply encapsulated by DSPE-PEG, demonstrate a photothermal efficiency of 51% while keeping a high photoluminescence quantum yield in the NIR region. Moreover, the nanoaggregates can be excited and delivered by an 808 nm pulse laser to solid tumors within only 40 min. The delivery efficiency and theranostic efficacy are better than that of the traditional enhanced permeability and retention (EPR) effect (generally longer than 24 hours). This platform is first termed as the photoinduced thermoacoustic (PTA) process, and confirm its application requires both NIR-responsive materials and pulse laser irradiation. This study not only inspires the design of multifunctional nanoaggregates, but also offers a feasible approach to their fast delivery. The platform reported here provides a promising prospect to boost the development of multifunctional theranostic drugs and maximize the efficacy of used medicines for their clinical applications.

178. Robust Data-Driven Structural Impact Localization With Multisensor Real-Time Monitoring

Abstract

Transient impact loads can easily cause structural damage and destruction, especially for composite structures that are susceptible to collisions. An accurate impact source localization method is crucial for evaluating the potential structure damage location. The impact response signals, on the other hand, contain varying degrees of high- and low-frequency noise, and the structure is not limited to a single dimension. In this article, the robust localization method with empirical mode decomposition combined wavelet denoising (EMD-WD) and multi-output support vector regression (MSVR) is proposed. Following the achievement of intrinsic mode function (IMF) of distinct frequency bands, the last order IMF is abandoned to filter the low-frequency disturbance. Following that, the discrete wavelet transform is used to treat high-frequency noise. Then, multidimensional features of impact responses, including time domain (TT), frequency domain (FF), time–frequency domain (TF), and distance domain (DD), are extracted. Finally, MSVR trained with multidimensional feature vectors is used to predict various impact location coordinate values. The results on plate structure and cylinder composite monitoring data prove that the proposed method can realize acceptable localization performance on both the 2-D and 3-D structures compared with other methods, with certain abilities to resist noise interference.

Abstract

Background: Hepatocellular carcinoma (HCC) is a highly heterogeneous cancer with major challenges in both prevention and therapy. Metformin, adenosine monophosphate‐activated protein kinase (AMPK) activator, has been suggested to reduce the incidence of HCC when used for patients with diabetes in preclinical and clinical studies. However, the possible effects of metformin and their mechanisms of action in non‐diabetic HCC have not been adequately investigated. Methods: Fah−/− mice were used to construct a liver‐injury‐induced non‐diabetic HCC model for exploring hepatocarcinogenesis and therapeutic potential of metformin. Changes in relevant tumour and biochemical indicators were measured. Bulk and single‐cell RNA‐sequencing analyses were performed to validate the crucial role of proinflammatory/pro‐tumour CD8+ T cells. In vitro and in vivo experiments were performed to confirm Cyp26a1‐related antitumour mechanisms of metformin. Results: RNA‐sequencing analysis showed that chronic liver injury led to significant changes in AMPK‐, glucose‐ and retinol metabolism‐related pathways in Fah−/− mice. Metformin prevented the formation of non‐diabetic HCC in Fah−/− mice with chronic liver injury. Cyp26a1 ddexpression in hepatocytes was significantly suppressed after metformin treatment. Moreover, downregulation of Cyp26a1 occurred in conjunction with increased levels of all‐trans‐retinoic acid (atRA), which is involved in the activation of metformin‐suppressed hepatocarcinogenesis in Fah−/− mice. In contrast, both CD8+ T‐cell infiltration and proinflammatory/pro‐tumour cytokines in the liver were significantly upregulated in Fah−/− mice during chronic liver injury, which was notably reversed by either metformin or atRA treatment. Regarding mechanisms, metformin regulated the decrease in Cyp26a1 enzyme expression and increased atRA expression via the AMPK/STAT3/Gadd45β/JNK/c‐Jun pathway. Conclusions: Metformin inhibits non‐diabetic HCC by upregulating atRA levels and downregulating CD8+ T cells. This is the first reporting that the traditional drug metformin regulates the metabolite atRA via the Cyp26a1‐involved pathway. Abstract

The use of functional nanoparticles as peroxidase-like (POD-like) catalyst has recently become a focus of research in cancer therapy. Phthalocyanine is a macrocyclic conjugated metal ligand, which is expected to achieve a high POD-like catalytic activity, generating free radicals and inhibiting the proliferation of cancer cells. In this paper, we synthesized phthalocyanine nanocrystals with different structures through noncovalent self-assembly confined within micro-emulsion droplets, and manganese phthalocyanine (MnPc) possessing a metal-N-C active center was used as the building block. These nano-assemblies exhibit shape-dependent POD-like catalytic activities, because the emulsifier and MnPc co-mixed assembly reduced the close packing between MnPc molecules and exposed more active sites. The assembly had a water-dispersed nanostructure, which is conducive to accumulation at tumor sites through the enhanced permeability and retention effect (EPR). Because of a highly efficient microenvironmental response, the assembly showed higher catalytic activity only emerged under the acidic tumor-like microenvironment, but caused less damage to normal tissues in biomedical applications. In vivo and in vitro catalytic therapy tests showed excellent anti-tumor effects. This work explored a new way for the application of metal-organic macromolecules such as MnPc as nanozymes for catalytic tumor therapy.
