156. Characteristics of Heavy Metals Contamination in Lotus Root in the Dongting Lake Area, China

Abstract

Heavy metal contamination in soils in the Dongting Lake areas has evoked widespread concerns about the excessive heavy metals in aquatic product. Based on the national standards of food contaminant limits and the method of comprehensive pollution index, heavy metals of Cd, Pb, Cu, Zn, Mn in lotus root were clarified through field investigation in the Dongting Lake area. Results showed that lotus root in the Dongting Lake area was contaminated seriously by heavy metals. Cd and Pb were two main pollutants and the single pollution indices were 5.70 and 8.35 respectively. According to the comprehensive pollution index of heavy metals, lotus root in Yueyanglou District and Yuanjiang City were classified into medium pollution and Junshan District, Huarong County, Nan County, and Datong District were classified into heavy pollution. Principal component analysis showed that planting areas of lotus root were clumped and medium and heavy pollution areas were separated significantly. Habitat contamination by heavy metals and decreasing area of lotus ponds were two main factors for excessive heavy metals in lotus root. Thus, some measurements, such as habit restoration, were proposed for local government to decrease heavy metals in planting areas and to promote the healthy development of lotus root industry in the Dongting Lake area.

Abstract

The cause of reduced plasma pyridoxal-5 prime-phosphate (PLP) concentration and the validity of plasma PLP concentration as a vitamin B-6 status indicator during pregnancy are not well understood. In this study, pregnant and nonpregnant women consumed a controlled diet for 5 d to exclude dietary intake as a factor in altered vitamin B-6 metabolic utilization. Plasma PLP and pyridoxal (PL) concentrations were significantly lower and higher, respectively, and plasma PL+PLP concentration was significantly lower during pregnancy. When plasma PLP and PL concentrations were normalized based on plasma albumin concentration, the differences were less marked but were still significant. The erythrocyte aspartate aminotransferase activity coefficient was significantly greater in pregnant women which is a further indication of reduced vitamin B-6 concentrations in the blood. No difference was observed in the urinary excretion of 4-pyridoxic acid between the pregnant and nonpregnant women. Results of this study demonstrate that vitamin B-6 status indices are lower during pregnancy without differences in the urinary excretion of 4-pyridoxic acid.

164. All-transretinoic acid plus low-dose rituximab vs low-dose rituximab in corticosteroid-resistant or relapsed ITP

Abstract

The study aimed to compare the efficacy and safety of all-transretinoic acid (ATRA) plus low-dose rituximab (LD-RTX) with LD-RTX monotherapy in corticosteroid-resistant or relapsed immune thrombocytopenia (ITP) patients. Recruited patients were randomized at a ratio of 2:1 into 2 groups: 112 patients received LD-RTX plus ATRA, and 56 patients received LD-RTX monotherapy. Overall response (OR), defined as achieving a platelet count of ≥30 × 109/L confirmed on ≥2 separate occasions (≥7 days apart), at least a doubling of the baseline platelet count without any other ITP-specific treatment, and the absence of bleeding within 1 year after enrollment, was observed in more patients in the LD-RTX plus ATRA group (80%) than in the LD-RTX monotherapy group (59%) (between-group difference, 0.22; 95% CI, 0.07-0.36). Sustained response (SR), defined as maintenance of a platelet count >30 × 109/L, an absence of bleeding, and no requirement for any other ITP-specific treatment for 6 consecutive months after achievement of OR during 1 year following enrollment, was achieved by 68 (61%) patients in the combination group and 23 (41%) patients in the monotherapy group (between-group difference, 0.20; 95% CI, 0.04-0.35). The 2 most common adverse events (AEs) for the combination group were dry skin and headache or dizziness. Our findings demonstrated that ATRA plus LD-RTX significantly increased the overall and sustained response, indicating a promising treatment option for corticosteroid-resistant or relapsed adult ITP. Abstract

Field plot trials were carried out with cabbage, brassica chinensis and corn involving 3 different fertilization schemes including non-fertilizer, chemical fertilizer, chemical fertilizer with organic manure to investigate the effects of the fertilization schemes on farmland quality as well as vegetable yield. The results showed that the fertilization scheme of organic manure caused accumulation of arsenic, lead and chromium, and the accumulation increased with organic manure amount;pH of alkaline soil was decreased by organic manure;the chemical fertilizer with organic manure significantly increased the content of soil organic matter, available nitrogen, available phosphorus and available potassium, as well as the yield of cabbage, brassica chinensis and corn, which indicated the increasing of soil fertility and vegetable field were based on fertilizer inputs.

196. Ethanol extract of Verbena officinalis alleviates MCAO-induced ischaemic stroke by inhibiting IL17A pathway-regulated neuroinflammation.

Abstract

The prevention and treatment of ischaemic stroke is a worldwide challenge, and effective clinical treatment strategies are lacking. Studies have demonstrated the efficacy of Verbena officinalis in managing cerebrovascular disorders. However, the neuroprotective bioactive components and mechanisms remain unclear. To investigate the pharmacological combinatorial components and mechanism underlying the anti-ischemic stroke effect of the ethanol extract of Verbena officinalis (VO Ex). The components of VO Ex were identified by HPLC. A middle cerebral artery occlusion (MCAO) induced brain injury model was used to assess the therapeutic effect of VO Ex. The activity of the chemical components of VO Ex was evaluated using a primary astrocyte injury model induced by oxygen-glucose deprivation/reperfusion (OGD/R). RNA sequencing was used to reveal the potential targets of VO Ex against cerebral ischemia–reperfusion injury (CIRI), and the results were verified by qRT-PCR and western blotting. The key components and target binding ability were predicted by molecular docking. Finally, the mechanism of combinatorial components was verified by experiments. The HPLC results indicated that the main ingredients of VO Ex were hastatoside, verbenalin, acteoside, luteolin, apigenin and hispidulin. In vivo experiments showed that VO Ex improved MCAO-induced acute cerebral ischemic injury. Transcriptomic data and biological experiments suggested that VO Ex exerted therapeutic effects through IL17A signalling pathways. The in vitro experiments indicated that verbenalin, acteoside, luteolin, apigenin and hispidulin exhibited neuroprotective activities. The novel formula of VALAH, derived from the aforementioned active ingredients, exhibited superior efficacy compared to each individual component. Molecular docking and mechanistic studies have confirmed that VALAH functions in the treatment of ischaemic stroke by suppressing the activation of the IL17A signalling pathway. This work is the first to reveal that VO Ex effectively inhibits the IL17A signaling pathway and mitigates neuroinflammation following ischemic stroke. Moreover, we identified the novel formula VALAH as the bioactive combinatorial components for VO Ex. Further research suggests that the activity of VALAH is associated with IL17A-mediated regulation of neuroinflammation. Abstract

A modified imidazole curing agent, EMI-g-BGE, was encapsulated for one-package of diglycidyl ether of bisphenol A (DGEBA) epoxy resin system. Polyetherimide (PEI) was used as the wall material, and the emulsion solvent evaporation method was used to form the microcapsules. The morphology and particle size distribution of microcapsules were evaluated by scanning electron microscopy (SEM), mastersizer analyzer. Microcapsules exhibited spherical shapes and the mean particle size was about 745 nm. The curing kinetic of DGEBA/microcapsules curing agent was studied by nonisothermal differential scanning calorimetry (DSC) technique at different heating rates. Dynamic DSC scans indicated the microcapsule was an effective curing agent of epoxy resin. The apparent activation energy Ea was 88.03 kJ/mol calculated through Kissinger method, more than DGEBA/EMI-g-BGE system. This microcapsule of EMI-g-BGE exhibited a long shelf life, and the curing did not occur in this epoxy-microcapsule resin system for more than 3months at room temperature. The kinetic parameters were determined by Málek method and a two-parameter ( m, n) autocatalytic model (Šesták–Berggren equation) was found to be the most adequate selected kinetic model, which showed the encapsulation of the curing agent EMI-g-BGE did not change the cure reaction mechanism of the epoxy resin system. From the experimental data, the nonisothermal DSC curves show the results being in accordant with those theoretically calculated.

Abstract

For the low accuracy of image reconstruction in Electrical Capacitance Tomography(ECT),a image reconstruction method for the ECT was proposed based on the particle filter algorithm.Firstly,the principle of image reconstruction of the ECT was analyzed.Then,the search process of the optimal solution for image reconstruction of the ECT was described as a system state estimation process,and a state space model was established.Furthermore,to obtain the minimum variance estimation of image reconstruction,the image reconstruction result of Linear Back Projection(LBP) algorithm was taken as the initial state,and the optimal weights of random samples obtained from the state space were calculated by the measured information.Finally,the simulation experiments with five different flow regimes were performed.The experiment results show that the average image error of reconstruction results by proposed method is 42.93%,and the average correlation coefficient with the original image is 0.813 9,which is much better than corresponding indicators obtained by LBP algorithm,Landweber iterative algorithm and IMNSNOF algorithm.In conclution,the image reconstruction method with high efficiency and accuracy can provide a new way for ECT research.
