162. Mapping and effect analysis of QTL for phenology traits in wheat using 55K chip technology.

Abstract

Phenology is an important agronomic trait for common wheat, which has a great impact to explore its genetic mechanism and effect for wheat breeding and application. In this study, 240 recombinant inbred lines (RILs) derived from Yangmai 158 and Hiller were used to identify the phenology traits in 2-4 environments. A total of 52 QTL were detected on chromosomes 2A, 2D, 3D, 4B, 4D, 5A, 5B, 5D, 6A, 6B, 7A, 7B, and 7D by using the constructed high-density genetic map. QJS/BS/HS/FS/MS.nau-5D.2, QJS/BS/HS/FS/MS.nau-2D.1, and QJS/BS/HS/FS/MS.nau-7B.1 were detected for several years, which explained 4.56%-46.86%, 1.32%-33.40%, and 2.37%-13.27% of phenotypic variation, respectively. QBS/HS.nau-2A.3, QHS/FS.nau-5B.2, QFS.nau-2A.5, QBS.nau-6A.2, QJS.nau-4D.2, QJS.nau-6A.3, QBS.nau-2A.2, QBS/HS.nau-6A.1, QFS.nau-7A.2, QMS.nau-3D, QMS.nau-4D.1, and QMS.nau-6B.1 were new QTL. Pyramiding of multiple phenology loci with large effects or repeated is an effective approach to shorten the growth period in different degrees, which could be used to cultivate early-maturing and high-yield wheat varieties. Abstract

(-)-Epigallocatechin-3-gallate (EGCG) is the primary active ingredient in green tea and has been used for cancer prevention in clinical trials. The anti-tumor effects of EGCG stem from its ability to inhibit the activities of many oncoproteins, such as AKT, VEGFR, STAT3, and mutant p53. However, the clinical efficacy of EGCG is unsatisfactory. How to improve the anti-tumor effects of EGCG is an open question. Here we report that EGCG inhibits the tumor suppressive Hippo signaling pathway and activates downstream YAP in colorectal cancer (CRC) cells. Activation of YAP impedes the anti-tumor effects of EGCG. YAP blockade increases the sensitivity of CRC cells to EGCG treatment.

Abstract

AIM: To demonstrate the feasibility of mesenchymal stem cell (MSC)-mediated nano drug delivery, which was characterized by the “Trojan horse”-like transport of hypoxia-inducible factor-1α small interfering RNA (HIF-1α siRNA) between MSCs and retinal pigment epithelial cells (RPE) under hypoxia environment. METHODS: Plasmid and lentivirus targeting the human HIF-1α gene were designed and constructed. HIF-1α siRNA was encapsulated into poly(lactic-co-glycolic acid) nanoparticles (PLGA-NPs) through the water-in-oil-in-water (w/o/w) multiple emulsion technique. The effect of PLGA-NPs uptake on the expression of HIF-1α mRNA was tested in RPE cells by real-time quantitative polymerase chain reaction (qPCR) and additional transfected conditions were used as control, including lentivirus group, nude plasmid group and blank PLGA group. MSCs were transfected with the NPs and the transfection efficacy was evaluated by flow cytometry. Transwell co-culture system of transfected MSCs and RPE cells was constructed under hypoxia environment. The effects of MSC-loaded HIF-1α siRNA PLGA-NPs on proliferation, apoptosis, and migration of RPE cells were then evaluated. The effect of transfected MSCs on HIF-1α expression of RPE cells was analyzed by using qPCR at the time points 24h, 3d, and 7d. RESULTS: The average diameter of PLGA-NPs loaded with HIF siRNA was 314.1 nm and the zeta potential was -0.36 mV. The transfection efficiency of PLGA-NPs was 67.3%±5.2% into MSCs by using flow cytometry. Abstract

Low back pain (LBP) is a very common disorder , and it causes significant pain, impairment of daily activities, and decreases quality of life.. With approximately 84% of adults experiencing an episode of LBP at some point during their lifetime and variable recurrence rates (5% to 60%). It is associated with more global disability than any other condition.Chronic low back pain (CLBP) refers to LBP has persisted for more than 3 months. CLBP carries an enormous economic burden from both direct (e.g. treatment) and indirect (e.g. lost work productivity) costs. In the United States, approximately $100 billion of medical expenses is spent annually for back pain with an additional $50 billion arising from indirect costs due to the lost in productivity and disability benefit payments. CLBP is also associated with impaired quality of life, mobility and daily function as well as social isolation, disability and depression. It is a major health problem leads to more years living with disability than any other musculoskeletal condition.Although financial burden and disability attributed to chronic low back pain is substantially different between countries, the incremental impact of the worldwide health care system is expected to be significant in the coming decades. According to 2017 global statistics, low back pain accounts for 7.3% point prevalence of activity-limiting illnesses, affecting 54 billion people at all ages.
