Abstract

Biocrust-mediated in situ bioremediation could be an alternative strategy to mitigate metal(loid) pollution in aquatic habitats. To better understand the roles of biocrusts in regulating the fate of metal(loid)s, we examined the morphology, composition and structure of biological aqua crusts (BAC) developed in the mine drainage of a representative Pb/Zn tailing pond, and tested their effectiveness for immobilizing typical metal(loid)s. Unlike terrestrial biocrusts, BAC results from an assembly of compounds produced by the strong microbial activity and mineral compounds present in the aquatic environment. The BAC exhibited a unique flexible, spongy and porous structure with a specific surface area of 12-22 m2 g-1, and was able to effectively concentrate various metal(loid)s (e.g. Cd, 0.26-0.60 g kg-1; Pb, 0.52-0.66 g kg-1; As, 10.4-24.3 g kg-1). The concentrations of metal(loid)s (e.g. Cd and As) in the BAC were even three to seven times higher than those in the source tailings, and more than 98% of immobilized metal(loid)s were present as the highly stable non-EDTA-exchangeable fraction. Adsorption on the well distributed micro-particles of the clay minerals (e.g. kaolinite) and the organic matters (2.0-2.7 wt.%) were found to be the major mechanisms for BAC to bind metal cations, whereas adsorption and coprecipitation on Fe/Mn oxide (e.g. FeOOH), was proposed to be the dominant pathway for accumulating metal(loid)s, especially As. The decrease in aqueous concentrations of the metal(loid)s along the drainage could be attributed in part to the scavenging effects of the BAC. These findings therefore provide new insights into the possible and efficient strategy for metal(loid) removal from water bodies, and highlighted the important role of BAC as a nature-based solution to benefit the bioremediation of mining area.

Abstract

Quantification of tracer kinetics using dynamic positron emission tomography (PET) provides important information for understanding the physiological and biochemical processes in humans and animals. A common procedure is to reconstruct a sequence of dynamic images first, and then apply kinetic analysis to the time activity curve of a region of interest derived from the reconstructed images. Obviously, the choice of image reconstruction method and its parameters affect the accuracy of the time activity curve and hence the estimated kinetic parameters. This paper analyzes the effects of penalized likelihood image reconstruction on tracer kinetic parameter estimation. Approximate theoretical expressions are derived to study the bias, variance, and ensemble mean squared error of the estimated kinetic parameters. Computer simulations show that these formulae predict correctly the changes of these statistics as functions of the regularization parameter. It is found that the choice of the regularization parameter has a significant impact on kinetic parameter estimation, indicating proper selection of image reconstruction parameters is important for dynamic PET. A practical method has been developed to use the theoretical formulae to guide the selection of the regularization parameter in dynamic PET image reconstruction. Abstract

Organic acids play an important role in metal tolerance, uptake, and translocation in hyperaccumulators. Phytolacca americana is a rare earth element (REE) hyperaccumulator, but the underlying mechanisms on REE tolerance and accumulation mediated by organic acids are poorly understood. Here, we reported for the first time the strategy of P. americana to enhance REE tolerance and accumulation through organic acids from root external secretion to internal biosynthesis. Different from the exclusion of heavy metal by organic acid in the typical plants, the results showed that oxalate secretion (0.3–0.6 μmol h−1 g−1 root DW) induced by yttrium (Y) could not prevent Y from entering the roots, resulting in excess Y uptake by P. americana. Yttrium stress also stimulated the accumulation of malate and citrate by 1.4- and 2.0-folds in the root cortex. Exogenous malate and citrate promoted the redistribution of Y from the root cell walls to the shoot by 30% and 21%, respectively. Based on comparative transcriptome analysis, 6-fold up-regulation was observed in PaNIP1;2, whose homology AtNIP1;2 is responsible for the transport of Al-malate in Arabidopsis. These results suggested that the promoted formation of Y-malate complexes within the roots potentially accelerated the transport of Y from P. americana roots to shoots through PaNIP1;2. Our study revealed the potential mechanism of organic acids in the external exclusion and internal detoxification and translocation of REE in P. americana roots, which provided a basis for improving the efficiency of REE phytoextraction.

Abstract

Positron emission tomography (PET) is actively used in a diverse range of applications in oncology, cardiology, and neurology. The use of PET in the clinical setting focuses on static (single time frame) imaging at a specific time-point post radiotracer injection and is typically considered as semi-quantitative; e.g. standardized uptake value (SUV) measures. In contrast, dynamic PET imaging requires increased acquisition times but has the advantage that it measures the full spatiotemporal distribution of a radiotracer and, in combination with tracer kinetic modeling, enables the generation of multiparametric images that more directly quantify underlying biological parameters of interest, such as blood flow, glucose metabolism, and receptor binding. Parametric images have the potential for improved detection and for more accurate and earlier therapeutic response assessment. Parametric imaging with dynamic PET has witnessed extensive research in the past four decades. In this paper, we provide an overview of past and present activities and discuss emerging opportunities in the field of parametric imaging for the future.

Abstract

Purpose: To compare the safety and efficacy of left versus right internal jugular vein access for portal vein puncture during transjugular intrahepatic portosystemic shunt (TIPS) creation in patients with a small liver and short vertical puncture distance.Materials and Methods: The vertical distance from the hepatic vein orifice to the puncture point of the portal vein was measured by CT and DSA. A distance ≤ 30 mm is defined as a short vertical puncture distance. After 1:1 propensity score matching (PSM), 29 patients of left internal jugular vein-TIPS (LIJ-TIPS) and 29 patients of right internal jugular vein-TIPS (RIJ-TIPS) were included. The number of needle punctures, fluoroscopy time, and radiation dose during the puncture process were statistically analyzed.Results: There was no significant difference in the average vertical puncture distances on CT or DSA between LIJ-TIPS and RIJ-TIPS (19.10 ± 0.60 mm vs. 19.30 ± 0.60 mm, P = 0.840; 22.02 ± 0.69 mm vs. 22.23 ± 0.64 mm, P = 0.822, respectively). The average number of needle punctures, fluoroscopy time, and radiation dose in LIJ-TIPS were significantly lower than those in RIJ-TIPS (2.07 ± 0.20 vs. 4.10 ± 0.24, P < 0.001; 78.45 ± 12.80 s vs. 201.16 ± 23.71 s, P < 0.001; 31.55 ± 7.04 mGy vs. 136.69 ± 16.38 mGy, P < 0.001, respectively). Within three punctures, the technical success rate in LIJ-TIPS was significantly higher than that in RIJ-TIPS (86.2 vs. 27.6%, P < 0.001). The incidence of hemoperitoneum in LIJ-TIPS was significantly lower than that in RIJ-TIPS (0% vs. 13.8%, P = 0.038).Conclusion: The left internal jugular vein could be used as primary access for TIPS creation in patients with a small liver and short vertical puncture distance. Abstract

Background Serum phospholipase A2 receptor (PLA2R) antibody (SAb) and glomerular deposits of PLA2R antigen (GAg) have been tested widely in idiopathic membranous nephropathy (MN). Recently, we noticed a special form of PLA2R-associated MN with positive circulating PLA2R antibody but negative PLA2R deposits in the glomeruli by immunofluorescence on frozen tissue (IF-F). The significance of this form of PLA2R-associated MN is yet to be elucidated. This study aimed to explore the clinicopathological features of these PLA2R-associated MN patients. Methods This study enrolled 229 biopsy-proven PLA2R-associated MN patients with SAb+. SAb was measured by enzyme-linked immunosorbent assay, and GAg was detected by IF-F. These patients were divided into SAb+/GAg+ and SAb+/GAg− groups. Clinicopathological characteristics of SAb+/GAg+ and SAb+/GAg− PLA2R-associated MN patients were compared. PLA2R antigens of 19 SAb+/GAg− PLA2R-associated MN patients were verified by immunohistochemistry on paraffin tissue (IHC-P). Results Among 229 SAb+ PLA2R-associated MN patients, 210 (91.70%) were GAg+ and 19 (8.3%) were GAg−. These 19 SAb+/GAg− PLA2R-associated MN patients presented positive PLA2R deposits by IHC-P. Compared with SAb+/GAg+ PLA2R-associated MN patients, SAb+/GAg- PLA2R-associated MN patients had higher levels of serum PLA2R antibody (P = 0.004), increased proteinuria (P = 0.008), lower serum albumin (P = 0.019), more prominent chronic pathological lesions in terms of glomerulosclerosis score (P = 0.025), interstitial fibrosis score (P = 0.016), tubular atrophy score (P = 0.010) and total renal chronicity score (P = 0.010), and were more likely to be accompanied by focal segmental glomerulosclerosis (P = 0.014). Higher SAb level was associated with the total renal chronicity score (odds ratio per 100 RU/mL, 1.16; 95% confidence interval 1.01–1.33; P = 0.033). Conclusions PLA2R-associated MN patients with seropositive PLA2R antibody but negative PLA2R deposits in the glomeruli by IF-F have higher levels of SAb and worse clinicopathological manifestations compared with their double-positive counterparts.
