Wprowadzenie: Pacjenci z wrodzoną niedoczynnością tarczycy (congenital hypothyroidism - CH) mają większe niż w populacji ogólnej ryzyko wystąpienia innych pozatarczycowych, wrodzonych nieprawidłowości. Nieprawidłowości te dotyczą głównie narządów i układów, w których prawidłowy rozwój i funkcjonowanie zaangażowane są geny odpowiedzialne również za prawidłowy przebieg organogenezy i syntezy hormonów gruczołu tarczowego. Cel pracy: Ocena częstości występowania innych wad towarzyszących CH, z uwzględnieniem możliwych przyczyn tej zależności i roli badań genetycznych. Materiał i metody: Do badania zostały włączone 54 noworodki objęte badaniem przesiewowym w kierunku CH w latach 2010-2017 w województwie małopolskim. Przeprowadzono dokładną retrospektywną analizę dokumentacji medycznej tych pacjentów. Wyniki: W grupie badanej u 20 z 54 pacjentów (37%) stwierdzono obecność współtowarzyszących CH, wrodzonych nieprawidłowości innych narządów. U 10 (18,5%) potwierdzono obecność wady serca, u 5 (9,25%) stwierdzono nieprawidłowe objawy ze strony układu oddechowego, u 7 (12,96%) stwierdzono nieprawidłowe objawy ze strony układu pokarmowego, u 5 (9,25%) rozpoznano nieprawidłowości ze strony układu moczowo-płciowego, u 3 (5,55%) stwierdzono nieprawidłowości ze strony układu nerwowego, u 6 (11,1%) rozpoznano nieprawidłowości ze strony układu mięśniowo-szkieletowego. Wnioski: Analiza uzyskanych danych i aktualnego piśmiennictwa wskazują, że największą rolę w powstawaniu pozatarczycowych nieprawidłowości u pacjentów z CH mogą mieć czynniki genetyczne. Zasadna wydaje się identyfikacja mutacji, która doprowadziła do CH. Znając mutacje mogące odpowiadać za wady współtowarzyszące CH, można by objąć pacjentów z CH odpowiednimi badaniami skriningowymi, mającymi na celu ich wczesne zdiagnozowanie, a w efekcie - odpowiednie, natychmiastowe leczenie. Introduction: Patients with congenital hypothyroidism (CH) can have an increased risk of occurrence of extrathyroidal defects compared to the general population, which could influence their development. The abnormalities occur mainly in organ systems whose development and function is dependent on genes that are also responsible for proper organogenesis of the thyroid gland and thyroid hormone synthesis. Aim of the study: The aim of the study was to evaluate the frequency of extrathyroidal defects in CH patients, taking into consideration the cause of this co-occurrence and the role of genetic tests. Material and methods: The study included 54 newborns with positive screening test for CH based on elevated TSH level, in the years 2010-2017, from South-Eastern Poland. The data was retrieved retrospectively from patients’ medical records. Results: Twenty of 54 newborns with CH (37%) had congenital defects of other organs. In 10 (18.5%) cardiac defects were found, in 5 (9.25%) abnormal symptoms of the respiratory system, 7 (12.96%) had abnormalities of the gastrointestinal system, five (9.25%) had genitourinary abnormalities, 3 (5.55%) had abnormalities of the nervous system, and 6 (11.1%) had musculoskeletal abnormalities. Conclusions: The analysis of our data and current literature suggests that genetic factors play the most important role in the development of extrathyroidal abnormalities in newborns with CH. Identifying the mutation causing CH, the potential defects that can accompany newborns with CH, screening could be offered for these patients in order to obtain an earlier diagnosis and implement early and appropriate treatment.

Wprowadzenie: Zespół Cushinga zależny od kortykotropiny (adrenocorticotropic hormone - ACTH), zwany również chorobą Cushinga (Cushing’s disease - CD), jest rzadkim schorzeniem u dzieci i młodzieży, którego objawy różnią się od obserwowanych u osób dorosłych. Cel pracy: Ocena procesu diagnostycznego i wyników terapii CD u dzieci i młodzieży. Materiał i metody: Do pracy włączono 4 kolejne pacjentki w wieku 7-15 lat z CD rozpoznaną na podstawie wyników badań hormonalnych i badania histopatologicznego usuniętego gruczolaka przysadki po przeprowadzeniu retrospektywnej analizy dokumentacji medycznej pacjentów leczonych w naszym ośrodku w latach 2012-2018. Wyniki: Mediana czasu od początku objawów klinicznych do rozpoznania CD w prezentowanej grupie pacjentów wynosiła 1,5 roku. Wszystkie pacjentki miały objawy CD typowe dla wieku rozwojowego: gwałtowny przyrost masy ciała z zahamowaniem tempa wzrastania, zaburzenia zachowania, dojrzewania bądź miesiączkowania. Diagnostyka pacjentów była przeprowadzona zgodnie z obowiązującym standardem, najwięcej trudności diagnostycznych sprawiła postać cykliczna CD rozpoznana u jednej z pacjentek. Wszystkie pacjentki przebyły przezklinową całkowitą resekcję gruczolaka przysadki. Po zabiegu wszystkie wymagały przejściowej substytucji glikokortykoidów w czasie, który wyraźnie korelował z długością czasu od wystąpienia objawów do ustalenia rozpoznania. U wszystkich uzyskano normalizację wyników hormonalnych i ustąpienie objawów klinicznych po leczeniu neurochirurgicznym, jednakże wszystkie pacjentki nadal są otyłe. W badaniach MR przysadki u 3 pacjentek nie stwierdza się odrostu masy guza, u najstarszej jest jego podejrzenie. Wnioski: W przypadku współwystępowania zahamowania procesu wzrastania (niekoniecznie niskorosłości) z przyrostem masy ciała (niekoniecznie otyłości), cech androgenizacji i zaburzeń zachowania konieczne jest przeprowadzenie diagnostyki w kierunku CD. Otyłość może być stałym powikłaniem CD rozpoznanej w dzieciństwie mimo jej skutecznego leczenia neurochirurgicznego. Introduction: Adrenocorticotropic hormone-dependent Cushing syndrome, known as Cushing disease (CD), is a rare disease in paediatric patients, the signs and symptoms of which differ from those seen in adult patients. Aim of the study: The objective of the study was to present the diagnostics and treatment results of CD in children and adolescents. Material and methods: We included four consequent patients in the study, 7-15 years old, with CD confirmed by laboratory tests and finally by histology examinations after surgery. The data were retrospectively retrieved from the medical records of all the patients from the years 2012-2018. Results: The median time from the onset of clinical symptoms to CD diagnosis was 1.5 years. All patients presented with rapid weight gain accompanying growth retardation, and behavioural, puberty, or menstruation disorders. The diagnostic process was carried out in accordance with the current standard. Diagnostic difficulties regarded the cyclic CD. All patients underwent transsphenoidal complete resection of the pituitary adenoma, and they all required transient glucocorticoid substitution for a period that correlated with the time from onset of symptoms to diagnosis. All subjects achieved normalisation of hormonal results and resolution of clinical symptoms, but all are still obese. Currently the patients are in remission, but there is a suspicion of the regrowth of the tumour mass in the eldest patient. Conclusions: In each case of the coexistence of growth velocity inhibition (not necessarily short stature) with the weight gain (not necessarily obesity) and the appearance of hyperandrogenism and depressive-anxiety disorders, biochemical diagnostics of CS should be performed. Obesity could be a chronic complication of CD in childhood, even after effective neurosurgery treatment.

Introduction: The aim of our study was to determine the relationship between neuropeptide B (NPB), neuropeptide W (NPW), nutritional and antioxidant status and selected fat- and bone-derived factors in type 1 diabetes mellitus (T1DM) treated using pens (T1DM pen group) or insulin pumps (T1DM pump group) in order to investigate the potential role of NPB and NPW in the clinical outcomes of T1DM. Material and methods: Fifty-eight patients with T1DM and twenty-five healthy controls (CONTR) participated in the study. Assessments of NPB, NPW, total antioxidant status (TAS), leptin, adiponectin, osteocalcin, and free soluble receptor activator for nuclear factor κB (free sRANKL) were conducted. Results: NPB, NPW, leptin, and TAS were lower (by 33%, p < 0.013; 34%, p < 0.008; 290%, p < 0.00004; 21%, p < 0.05; respectively), while adiponectin was by 51% higher (p < 0.006) in T1DM vs. CONTR, while osteocalcin and free sRANKL levels were similar in both groups. NPW was lower in the T1DM pen group both vs. the T1DM pump group (36% lower, p < 0.0009) and vs. the CONTR group (35% lower, p < 0.002). In the T1DM pen group, but not in the T1DM pump group or the CONTR group, the Cole index and TAS levels explain (besides NPB) the variation in NPW values. ROC curves showed that serum levels of leptin, adiponectin, NPB and NPW (but not osteocalcin or free sRANKL) were predictive indicators for T1DM. Conclusions: Measurements of NPB and NPW, besides leptin and adiponectin, are worth considering in the detailed prognosis of nutritional status in T1DM, primarily in the T1DM pen-treated population.

No licensed vaccine is currently available for prevention of Fasciola hepatica infections. However, considering the alarming increase in drug resistance, there is an urgent need for a safe and fully effective vaccine against fasciolosis. Here, we tested if cathepsins L (FhCL3-1, FhCL3-2) and B (FhCB3) secreted by juvenile liver flukes are viable vaccine targets when delivered alone or in combination in a rat model. Since control over the early immune response is crucial for parasite's establishment in its host, it was hypothesised that targeting fluke juvenile stages may prove beneficial. Moreover, it was assumed that selected antigens will act in a cumulative manner to interfere with liver fluke migration and thereby will reduce F. hepatica infection. Recombinant FhCL3-1 and FhCL3-2 delivered alone reduced liver fluke burdens by 47 % and 63 %, respectively. A trivalent vaccine containing rFhCL3-1/CL3-2/CB3 did not increase the protective vaccine efficacy compared to the rFhCL3-2 vaccinated group (53 %), although, reductions in liver fluke wet weight (statistically significant) and liver damage score were most pronounced. Further, the highest IgG1 and IgG2a levels were seen in rFhCL3-2 vaccinated rats, the group for which the highest reduction in worm burden was demonstrated. Moreover, IgG1 and IgG2a levels in vaccinated rats were significantly elevated compared to those reported for control groups up to 4 week post-infection. While the mechanism of protection remains unknown, it appears that it depends on vaccine-induced antibodies directed against cathepsins. The obtained results imply that F. hepatica juvenile-specific cathepsins are promising vaccine candidates that induce responses that successfully target early migratory liver fluke stages. Now, the challenge is to evaluate these juvenile-specific cathepsins for use in livestock.

The paper presents an engine control algorithm, based on an artificial neural network (ANN), which ensures a quick and smooth inertia phase of an upshift in a dual-clutch transmission (DCT). The main emphasis is placed on 3D characteristics, which can be directly implemented in the ECU, to control the inertia phase, when the engine speed decreases to reach the target clutch speed. They are developed based on an ANN model which approximates data obtained during engine test bed measurements in dynamic states. Moreover, to provide an overall gear change algorithm, the dual-clutch assembly activation mechanism was also analysed.

This paper presents the idea of increasing the effectiveness of slag decopperisation in an electric furnace in the "Głogów II" Copper Smelter by replacing the currently added CaCO3with a less energy-intensive technological additive. As a result of this conversion, one may expect improved parameters of the process, including process time or power consumption per cycle. The incentives to optimize the process are the benefits of increasing copper production in the company and the growing global demand for this metal. The paper also describes other factors that may have a significant impact on the optimization of the copper production process. Based on the literature analysis, a solution has been developed that improves the copper production process. The benefits of using a new technology additive primarily include increased share of copper in the alloy, reduced production costs, reduced amount of power consumed per cycle and reduced time it takes to melt. At the conclusion of the paper, the issues raised are highlighted, stressing that mastering the slag slurry process in electric furnaces requires continuous improvement.

This paper presents the possibility of using the numerical methods to analyze the work of hydraulic systems on the example of a cooling system of a power boiler auxiliary devices. The variety of conditions at which hydraulic system that operated in specific engineering subsystems requires an individualized approach to the model solutions that have been developed for these systems modernizing. A mathematical model of a series-parallel propagation for the cooling water was derived and iterative methods were used to solve the system of nonlinear equations. The results of numerical calculations made it possible to analyze different variants of a modernization of the studied system and to indicate its critical elements. An economic analysis of different options allows an investor to choose an optimal variant of a reconstruction of the installation.

Introduction: Chronic abdominal pain in children is a very frequent and sometimes challenging diagnostic issue. Differential diagnosis in that cases is difficult and often connected with numerous, time-consuming, expensive, and frequently stressful diagnostic studies. The aim of the study was to establish whether fecal calprotectin concentration (FCC) and TNF-alpha may be useful in children with chronic abdominal pain to differentiate between inflammatory bowel disease (IBD), other inflammatory gastrointestinal disorders, and functional gastrointestinal disorders. Methods. The study included patients (median age 13 years), who were assigned to functional gastrointestinal disorders group (n=33); inflammatory gastrointestinal disorders other than IBD (n=71), children with IBD (n=37) and 22 healthy children served as a control group. The concertation of FCC and TNF-alpha in stool samples was measured using ELISA. Results: In healthy children and in children with functional disorders FCCs were below 100 μg/g. In patients with IBD FCCs and TNF-alpha were markedly elevated as compare to children with functional gastrointestinal disorders, however using ROC discrimination of IBD patients was significantly better using FCC than TNF-alpha. Conclusion: FCC is better test for differentiation between IBD, other inflammatory gastrointestinal disorders, and functional gastrointestinal disorders as compare to TNF-alpha concentration in stool. FCC as screening test in patients with chronic abdominal pain should allow to diminish unnecessary diagnostic in cases of functional gastrointestinal disorders.

Anti-Muellerian hormone (AMH) as only possible marker in the assessment of ovarian function and reserve after hematopoietic stem cell transplantation (HSCT) in prepubertal girls, young females with composed hypogonadism and females receiving hormonal replacement therapy

The aim of this study was to assess the immune response and the protective efficacy elicited by the vaccination with the recombinant Fasciola hepatica myosin regulatory light chain (FhrMRLC) in Adjuplex® adjuvant against the infection with F. hepatica in rats. Four groups of 15 animals each were used for the study, one group was immunized with the recombinant F. hepatica MRLC in Adjuplex® adjuvant and the other groups remained as adjuvant, positive and negative control groups. The parasitological study showed that a statistically significant reduction of 65.1% and 82.1% in fluke burden and fecal egg count, respectively, was detected in vaccinated animals. In addition, vaccination with FhrMRLC induced a well-defined humoral and cellular immune response characterized by a significant production of specific IgG and IL-2, IL-12, TNF-α and IFN-γ; which confirms the immunogenic capacity of the FhrMRLC.

This paper presents the results of research on the effect of hydrated calcium sulfate (phosphogypsum) on the mechanical properties of composite based on aluminum powder. The phosphogypsum used is a by-product obtained in the phosphate fertilizers industry, e.g., at the Gdanskie Zaklady Fosforowe “Fosfory.” The amount of phosphogypsum produced in Poland must be reduced due to the high costs of its storage and environmental protection. One of the ways to dispose of it is to use it as a filler in composite rods based on aluminum powder. The amount of phosphogypsum added was 2% and 5% by weight in relation to the aluminum powder’s weight. The filled composite was subjected to strength tests to show whether phosphogypsum improves the mechanical properties of aluminum rods. Based on the results obtained, it was revealed that phosphogypsum used as a filler causes similar changes in the mechanical properties as mineral fillers described in the literature. Based on analyses of market potential, it can be easily assumed that there is a significant demand for the results of research on the use of phosphogypsum. The end of the article summarizes the discussed issue, emphasizing that solutions are sought for the use of phosphogypsum.

Monoclonal antibodies have a wide range of applications in basic and applied research as well as in the medical and pharmaceutical industries. Phage display antibody libraries offer an alternative to hybridoma technology for the generation of monoclonal antibodies and can be applied to high-throughput screening and facilitate the generation of novel antibodies. Despite their utility in several fields of research there has been limited application of antibody libraries in the study of trematode parasites. Fasciola hepatica causes considerable loss to the agriculture sector and is also a human pathogen. The parasite's excretory/secretory material contains numerous molecules that facilitate its invasion and survival within the mammalian host, including cathepsin B and L proteases. F. hepatica cathepsin B2 is expressed during the initial weeks of infection and has suspected roles in immune evasion and as a digestive enzyme in the parasite's gut; it is considered a good target for vaccination or therapeutic inhibitors. In this study, we produced a single-chain variable fragment (scFv) phage display library from naive mice. The library was used to identify several scFv that can bind to antigens from adult F. hepatica homogenate, and a scFv that can bind to F. hepatica cathepsin B2. The results highlight the potential applicability of such a library to facilitate the study of F. hepatica and other parasites. This is the first report of the application of a naive phage display antibody library to the study of F. hepatica.

Introduction: Amiodarone is an important antiarrhythmic drug used in paediatric practice, mainly in children with complex congenital cardiac diseases and/or severe arrhythmias. One of the side effects of amiodarone therapy is thyroid dysfunction, which is observed in about 20% of patients. The thyroid dysfunction may present with various forms: from subclinical changes in hormone levels to amiodaroneinduced thyrotoxicosis (AIT) and amiodarone-induced hypothyroidism (AIH). Material and methods: We reported six patients in the age range from two weeks to 14 years, with complex congenital cardiac diseases and severe arrhythmias, who developed amiodarone-induced thyroid dysfunctions: thyrotoxicosis or hypothyroidism or both together. The clinical signs and symptoms of all thyroid dysfunctions were atypical, most patients presented with an aggravation of heart insufficiency. Our patients with thyrotoxicosis were treated with combined therapy including thionamides and corticosteroids due to the presentation of mixed-identified type of AIT. Results: Currently, five patients (one patient’s status is unknown) are in biochemical and clinical euthyreosis; however, in one of them it was impossible to discharge amiodarone treatment. Three of them are still treated with levothyroxine, and two do not need thyroid treatment. Conclusions: Amiodarone-induced thyroid dysfunction is usually atypical; therefore, monitoring of thyroid status before, during, and after amiodarone is demanded. AIH could significantly influence the development of the child, while AIT could significantly deteriorate the clinical status of children with complex cardiac diseases. Early and proper diagnose of AIT and AIH allows the introduction of immediate and appropriate treatment considering the cardiac condition of the young patient.

The article presents the results of ultrasonic flow measurements performed after the hydraulic elbow. Ultrasonic flowmeter with applied head set in accordance with the Z-type was used to carry out the measurements. The results of flow measurements after the hydraulic elbow were referenced to measurements made on a straight section of the pipeline before the elbow, where the flow was stabilized, and the velocity profile was symmetrical with respect to the pipe-line axis. Measurements, with the maintaining a constant volumetric stream flow, were made for 12 different angles of the flowmeter head settings in 16 distances from the hydraulic elbow. The results of the measurements were compared with the velocity values obtained from the flow simulation performed in the ANSYS CFX program. On the basis of the comparison of the measurement results with the simulation results, and also based on the analysis of the velocity profiles, it was found that at the appropriate angle of the head setting, measurements can be made using the ultrasound method at a distance smaller than the one described in the standards. The optimal location of the measurement can be selected on the basis of a computer flow simulation, which is a representation of geometry and measurement conditions. This action scheme can be used in the flow measurements, which are carried out after the obstacle which is disturbing the flow, in the pipelines with large diameters (for example power plants, electrical power and heating plant, chemical industry) where finding a straight section with a length of 15–20 pipeline diameters is problematic.

The subject of the article concerns the measurement of flow streams with disturbed velocity distribution by the means of elbow flowmeter. The velocity disturbances were forced by a different setting of the aperture located at the inlet to the installation. In the article, flow coefficients for the elbow flowmeter for exemplary disturbances of velocity distribution were determined and deviations in relation to the flow without disturbances were calculated. For the exemplary disturbance, numerical calculations were performed, the results of which were compared with the actual results of the experiment.

Recent studies have shown that sclerostin, which is a negative regulator of bone formation, could play an important role in the crosstalk between bone and glucose metabolism. The role of sclerostin and its link with glucose homeostasis in type 1 diabetes mellitus (T1D) has not been yet studied extensively in children. The aim of this study was to assess sclerostin and its relationship between other bone and fat related factors as well as glucose metabolism in children and adolescents with T1D in comparison to their healthy peers.Forty patients with T1D, 18 girls, mean age 12.3 ± 4.7 yrs, and 28 healthy as controls (13.1 ± 4.2 yrs), sex and Tanner stage-matched were included into the study. Fasting blood samples for measurement of sclerostin, osteocalcin (OC), leptin, adiponectin, vitamin D, fasting glucose, lipid profile, HbA1c, and C-peptide were taken at 8.00 AM.Sclerostin levels were significantly higher in patients with T1D than in the control group (p = 0.04) without significant differences between genders. Pearson correlation coefficients revealed a positive association between serum sclerostin levels and leptin OC (r = 0.59, p 0.001) and a negative correlation between serum sclerostin levels and leptin (r = -0.32, p = 0.02) in all of the subjects and no significant correlations between sclerostin and adiponectin, 25(OH)D3, nor lipids. In the group of T1D patients a strong positive association between serum sclerostin levels and OC (r = 0.62, p 0.001), and a negative association between serum sclerostin levels and HbA1c and leptin levels (r = -0.33, p = 0.04; r = -0.33, p = 0.03, respectively) were found. These associations were significant also after adjusting the analysis to the age, SDS-BMI and Tanner staging. In the healthy group after adjustment to age, SDS-BMI and Tanner stage, a negative correlation between sclerostin and C-peptide (r = -0.79, p = 0.02) was found.Our data suggest a possible relationship between sclerostin and glucose metabolism in children and adolescents with T1D. It would be worth to investigate if an increase in sclerostin levels could present as a potential cause of the reduction of bone formation in T1D. Both bone-derived OC as well as fat-derived leptin seems to possibly modulate the participation of sclerostin in metabolic regulation in T1D.

Purpose To present the outcomes of ultrasound (US) follow-ups in children with autoimmune thyroid disease who did not have a thyroid nodule on admission but developed papillary thyroid carcinoma (PTC) and to characterize the parenchymal changes in the thyroid gland prior to the development of PTC. Methods A retrospective thyroid US scan review of 327 patients diagnosed with AIT was performed. Forty patients (40/327, 12.2%) presented nodular AIT variant with a normoechogenic background. Eleven patients (11/327, 3.4%, 11/40, 27.5%) presenting this variant were diagnosed with PTC (nine females—mean age 15.3 years; two males aged 11 and 13 years). In five of 11 patients, the suspicious nodule that was later confirmed to be PTC was detected on the initial US at presentation. For the remaining six females (6/11) who developed PTC during the follow-up, we retrospectively analysed their US thyroid scans and these patients were selected for analysis in this study. Results On admission, the US evaluation revealed an enlarged normoechogenic thyroid gland in three patients and a hypoechogenic thyroid gland with fibrosis as indicated by irregular, chaotic hyperechogenic layers in three patients. No thyroid nodules were identified. Ultrasound monitoring revealed increasing echogenicity of the thyroid parenchyma during the follow-up. PTC developed in a mean time of 4.6 years (1 9/12–7 4/12 years) since referral to the outpatient thyroid clinic and 2.9 years (6/12–6 9/12) since the last nodule-free US thyroid scan. Conclusions Sonographic follow-up assessments warrant further exploration as a strategy to determine PTC susceptibility in the paediatric population.

Two extrudates made of AlSi11 alloy have been produced in the co-extrusion process: solid billet and machining chips from the rolling process. The microstructure was observed, mechanical properties were determined on the basis of the 1-axial stretch test, and the fatigue tests were performed to determine the fatigue resistance of the AlSi11 alloy.

The aim of this study was to determine if there are quantitative differences in Candida fungi between pediatric patients with Crohn’s disease (before and after exclusive enteral nutrition (EEN), and the biologic therapy with anti-tumor necrosis factor alpha – (IFX)), and healthy controls. DNA was isolated from fecal samples and PCR was used to determine the number of fungal cells. Both therapeutic interventions resulted in a statistically significant decrease in Pediatric Crohn’s Disease Activity Index. The numbers of Candida decreased during both therapeutic intervention but the difference was statistically significant for the IFX intervention only (p = 0.045). Moreover, fungi population in both study groups declined during intervention when compared to the control group but the difference was significant before treatment only in the IFX group (p = 0.013). The total distribution of Candida with both IFX and EEN as well as in the control group differed significantly (p = 0.01) before treatment only. No correlation between the numbers of Candida and disease activity as well as the following biochemical parameters: serum iron concentration, protein or glucose level were found. It cannot be ruled out that, in combination with genetic and immunological disorders, fungi can contribute to the initiation of the disease process and perpetuation of active inflammation.

Oral vaccination with edible vaccines is one of the most promising approaches in modern vaccinology. Edible vaccines are an alternative to conventional vaccines, which are typically delivered by injection. Here, freeze-dried transgenic lettuce expressing the cysteine proteinase of the trematode Fasciola hepatica (CPFhVV) was used to orally vaccinate cattle and sheep against fasciolosis, which is the most important trematode disease due to the parasites global distribution, wide spectrum of host species and significant economic losses of farmers. In the study, goals such as reducing the intensity of infection, liver damage and F. hepatica fecundity were achieved. Moreover, we demonstrated that the host sex influenced the outcome of infection following vaccination, with female calves and male lambs showing better protection than their counterparts. Since differences occurred following vaccination and infection, different immunization strategies should be considered for different sexes and host species when developing new control methods. The results of the present study highlight the potential of oral vaccination with plant-made and plant-delivered vaccines for F. hepatica infection control. Hookworms are intestinal nematodes that infect up to 740 million people, mostly in tropical and subtropical regions. Adult worms suck blood from damaged vessels in the gut mucosa, digesting hemoglobin using aspartic-, cysteine- and metalloproteases. Targeting aspartic hemoglobinases using drugs or vaccines is therefore a promising approach to ancylostomiasis control. Based on homology to metalloproteases from other hookworm species, we cloned the Ancylostoma ceylanicum metalloprotease 7 cDNA (Ace-mep-7). The corresponding Ace-MEP-7 protein has a predicted molecular mass of 98.8 kDa. The homology to metallopeptidases from other hookworm species and its predicted transmembrane region support the hypothesis that Ace-MEP-7 may be involved in hemoglobin digestion in the hookworm gastrointestinal tract, especially that our analyses show expression of Ace-mep-7 in the adult stage of the parasite. Immunization of Syrian golden hamsters with Ace-mep-7 cDNA resulted in 50% (p < 0.01) intestinal worm burden reduction. Additionally 78% (p < 0.05) egg count reduction in both sexes was observed. These results suggest that immunization with Ace-mep-7 may contribute to reduction in egg count released into the environment during the A. ceylanicum infection.

Summary The article presents an example of thermal-flow analysis of the bottom ash cooler cooperating with the circulating fluidized bed boiler. There is presented a mathematical model of series-parallel hydraulic system supplying the ash cooler in cooling water. The numerical calculations indicate an influence of changes of the pipeline geometrical parameters on the cooling water flow rate in the system. Paper discusses the methodology of the studies and presents examples of the results of thermal balance calculations based on the results of measurements. The numerical results of the thermal-flow analysis in comparison with the measurements on the object indicate that the presented approach could be used as a diagnostic tool investigating the technical state of the bottom ash cooler.
