Agricultural activities are in the Netherlands a major source of ammonia (NH3), nitrogen oxide (NO), nitrous oxide (N2O), methane (CH4) and particulate matter (PM10 and PM2.5). The emissions in 2016 were calculated using the National Emission Model for Agriculture (NEMA). Some figures in the time series 1990-2015 were revised. The method calculates the NH3 emission from livestock manure based on the total ammonia nitrogen (TAN) content in manure. In 2016 NH3 emissions from livestock manure, fertilizer and other sources in agriculture, from hobby farms, private parties and manure disposal in nature areas amounted to 116.8 million kg NH3, 1.3 million kg more than in 2015. Nitrogen excretion increased due to expansion of the dairy herd, but because of a larger share of low emission housing and more manure exports outside agriculture, the increase in NH3 emission remained limited. N2O emissions in 2016 were 21.1 million kg at virtually the same level as in 2015(21.2). NO emissions in 2016 totaled 22.8 million kg compared to 22.6 million kg in 2015. CH4 emissions increased from 496 to512 million kg due to the expansion of the dairy herd. Emissions of particulate matter PM10 and PM2.5, 6.5 and 0.6 million kg respectively, did not change compared to 2015. NH3 emissions from livestock manure in the Netherlands dropped by almost two thirds since 1990, mainly as a result of lower nitrogen excretion rates by livestock and low emission manure application. Emissions of N2O and NO also decreased over the same period, but less strongly (38% and 31% respectively), due to higher emissions from manure injection into the soil and the shift from poultry housing systems with liquid manure towards solid manure systems. CH4 emissions reduced by 13% between 1990 and 2016 caused by a decrease in livestock numbers and increased feed efficiency of dairy cattle. Landbouwkundige activiteiten zijn in Nederland een belangrijke bron van ammoniak (NH3), stikstofoxide (NO), lachgas (N2O),methaan (CH4) en fijnstof (PM10 en PM2,5). De emissies in 2016 zijn berekend met het National Emission Model for Agriculture(NEMA). Tegelijk zijn enkele cijfers in de reeks 1990-2015 aangepast op basis van nieuwe inzichten. De rekenmethodiek gaatbij de berekening van de NH3-emissie uit dierlijke mest uit van de hoeveelheid totaal ammoniakaal stikstof (TAN) in de mest. In2016 bedroeg de NH3-emissie uit dierlijke mest, kunstmest en overige bronnen in de landbouw, bij hobbybedrijven,particulieren en bij mestafzet in natuurterreinen 116,8 miljoen kg NH3, 1,3 miljoen kg meer dan in 2015. De stikstofuitscheidingnam toe vanwege uitbreiding van de melkveestapel, maar door meer emissiearme huisvesting en een grotere mestafzet buitende landbouw bleef de toename van de NH3-emissie beperkt. De N2O-emissie lag in 2016 met 21,1 miljoen kg vrijwel ophetzelfde niveau als in 2015 (21,2). De NO-emissie bedroeg in 2016 22,8 miljoen kg tegen 22,6 miljoen kg in 2015. De CH4-emissie nam door de groei van de melkveestapel toe van 496 tot 512 miljoen kg. De emissies van fijnstof PM10 en PM2,5,respectievelijk 6,5 en 0,6 miljoen kg, veranderden niet ten opzichte van 2015. Sinds 1990 is de ammoniakemissie uit dierlijkemest en kunstmest met tweederde gedaald, vooral door een lagere stikstofexcretie en door emissiearme mesttoediening.Emissies van N2O en NO daalden in dezelfde periode eveneens, maar minder sterk (38% respectievelijk 31%) omdat door hetin de bodem brengen van mest deze emissies hoger zijn geworden vergeleken met bovengrondse mesttoediening en door deomschakeling van stalsystemen met dunne naar vaste mest bij pluimvee. Tussen 1990 en 2016 daalde de emissie van CH4 met13% door een afname van de dieraantallen en hogere efficiënties van melkvee.

- Abstract
RIVM rapport:Total greenhouse gas (GHG) emissions in the Netherlands in 2016 increased by approximately 0.2%, compared with 2015 emissions. This increase was mainly the result of increased natural gasconsumption for space heating. On the other hand, the emission of electricity production has declined. In 2016, total GHG emissions (including indirect CO2 emissions and excluding emissions from Land use, land use change and forestry (LULUCF)) in the Netherlands amounted to 195.2 Tg CO2 eq. This is approximately 12.4% below the emissions in the base year2 (222.9 Tg CO2 eq.). CO2 emissions in 2016 were still above the level in the base year (+1.6%). This increase was offset by the reduction since 1990 in emissions of methane, nitrous oxide and fluorinated gases (CH4, N2O and F-gases). This report documents the Netherlands' annual submission for 2018 of its GHG emissions inventory in accordance with the 2006 IPCC Guidelines for National Greenhouse Gas Inventories (IPCC, 2006) provided by the United Nations Framework Convention on Climate Change (UNFCCC), the Kyoto Protocol (KP) and the European Union's Greenhouse Gas Monitoring Mechanism. The report includes explanations of observed trends in emissions; an assessment of the sources with the highest contribution to total national emissions (key sources) and the uncertainty in their emissions; an itemization of methods, data sources and emission factors (EFs) applied; and a description of the quality assurance system and the verification activities performed on the data., In 2016 is de totale uitstoot van broeikasgassen van Nederland met ongeveer 0,2 procent gestegen ten opzichte van de uitstoot in 2015. Deze stijging komt vooral doordat er meer aardgas is verbruikt om ruimtes te verwarmen. Daarentegen staat dat de uitstoot van de electriciteitsproductie is afgenomen. De totale uitstoot van broeikasgassen naar de lucht wordt uitgedrukt in CO2-equivalenten en bedroeg in 2016 195,2 miljard kilogram. Ten opzichte van het zogeheten Kyoto-basisjaar (222,9 miljard kilogram CO2-equivalenten) is dit een afname van ongeveer 12,4 procent. Dit basisjaar, dat afhankelijk van het broeikasgas 1990 of 1995 is, dient voor het Kyoto Protocol als referentiejaar voor de uitstoot van broeikasgassen. De emissie van CO2 lag in 2014 voor het eerst onder het niveau van het basisjaar 1990. Sindsdien is de CO2-uitstoot toegenomen met circa 5 procent en komt hij boven het niveau van het basisjaar 1990 (+1,6 procent) te liggen. Deze toename werd voor de totale emissie van broeikasgassen ruim gecompenseerd door de lagere emissies van methaan, distikstofoxide en gefluoreerde gassen (CH4, N2O en F-gassen). Dit blijkt uit een inventarisatie van broeikasgasemissies die het RIVM jaarlijks op verzoek van het ministerie van Economische Zaken en Klimaat (EZK) opstelt. Met deze inventarisatie voldoet Nederland aan de nationale rapportageverplichtingen voor 2018 van het Klimaatverdrag van de Verenigde Naties (UNFCCC), van het Kyoto Protocol en van het Bewakingsmechanisme Broeikasgassen van de Europese Unie. De inventarisatie bevat verder trendanalyses voor de uitstoot van broeikasgassen in de periode 1990-2016, een analyse van belangrijkste emissiebronnen ('sleutelbronnen'), evenals de onzekerheid in hun emissies. Daarnaast zijn in de inventarisatie de gebruikte berekeningsmethoden beschreven, evenals databronnen en gebruikte emissiefactoren. - Abstract
The National Emission Model for Agriculture (NEMA) is used to calculate emissions to air from agricultural activities in the Netherlands on a national scale. Emissions of ammonia (NH3) and other N-compounds (NOx and N2O) are calculated from animal housing, manure storage, manure application and grazing using a Total Ammoniacal Nitrogen (TAN) flow model. Furthermore, emissions from application of inorganic N fertilizer, compost and sewage sludge, cultivation of organic soils, crop residues, and ripening of crops are calculated. NEMA is also used to estimate emissions of methane (CH4) from enteric fermentation and manure management, particulate matter (PM) from manure management and agricultural soils and carbon dioxide (CO2) from liming. Emissions are calculated in accordance with international guidance criteria and reported in an annual Informative Inventory Report (IIR; for air pollutants) and National Inventory Report (NIR; for greenhouse gases). This methodology report describes the outline and backgrounds of the emission calculations with NEMA.

- Abstract
Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10-8), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20 % of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

Recent Arctic studies suggest that sea ice decline and permafrost thawing will affect phytoplankton dynamics and stimulate heterotrophic communities. However, in what way the plankton composition will change as the warming proceeds remains elusive. Here we investigate the chemical signature of the plankton-dominated fraction of particulate organic matter (POM) collected along the Siberian Shelf. POM (> 10 µm) samples were analysed using molecular biomarkers (CuO oxidation and IP25) and dual-carbon isotopes (δ13C and Δ14C). In addition, surface water chemical properties were integrated with the POM (> 10 µm) dataset to understand the link between plankton composition and environmental conditions. δ13C and Δ14C exhibited a large variability in the POM (> 10 µm) distribution while the content of terrestrial biomarkers in the POM was negligible. In the Laptev Sea (LS), δ13C and Δ14C of POM (> 10 µm) suggested a heterotrophic environment in which dissolved organic carbon (DOC) from the Lena River was the primary source of metabolisable carbon. Within the Lena plume, terrestrial DOC probably became part of the food web via bacteria uptake and subsequently transferred to relatively other heterotrophic communities (e.g. dinoflagellates). Moving eastwards toward the sea-ice-dominated East Siberian Sea (ESS), the system became progressively more autotrophic. Comparison between δ13C of POM (> 10 µm) samples and CO2aq concentrations revealed that the carbon isotope fractionation increased moving towards the easternmost and most productive stations. In a warming scenario characterised by enhanced terrestrial DOC release (thawing permafrost) and progressive sea ice decline, heterotrophic conditions might persist in the LS while the nutrient-rich Pacific inflow will likely stimulate greater primary productivity in the ESS. The contrasting trophic conditions will result in a sharp gradient in δ13C between the LS and ESS, similar to what is documented in our semi-synoptic study.

In 2013 is de totale broeikasgasemissie van Nederland met ongeveer 0,2 procent gedaald ten opzichte van de emissie in 2012. Deze daling komt vooral door de afname van brandstofgebruik in de transportsector en de petrochemische industrie. De totale broeikasgasemissie in 2013 bedraagt 195,8 teragram (megaton of miljard kilogram) CO2 equivalenten. Ten opzichte van het basisjaar (221,1 Tg CO2 equivalenten) is dit een afname van ongeveer 11,5 procent. Beide getallen zijn exclusief de emissies afkomstig uit het soort landgebruik en de verandering daarin, zoals natuurontwikkeling of ontbossing (land use, land use change and forestry, LULUCF). Dit blijkt uit een inventarisatie van broeikasgasemissies die het RIVM op verzoek van het ministerie van Infrastructuur en Milieu (IenM) heeft opgesteld. Met deze inventarisatie voldoet Nederland aan de nationale rapportageverplichtingen voor 2015 van het Klimaatverdrag van de Verenigde Naties (UNFCCC)4 en van het Bewakingsmechanisme Broeikasgassen van de Europese Unie. De emissiecijfers zijn in absolute zin gewijzigd ten opzichte van eerdere rapportages omdat de nu gerapporteerde cijfers berekend zijn conform de nieuwste UNFCCC 2006 Guidelines. De inventarisatie bevat verder trendanalyses voor de emissies van broeikasgassen in de periode 1990-2013, een analyse van belangrijkste emissiebronnen (sleutelbronnen) evenals de onzekerheid in hun emissies. Daarnaast biedt de inventarisatie documentatie van de gebruikte berekeningsmethoden, databronnen en toegepaste emissiefactoren. Ten slotte bevat het een overzicht van het kwaliteitssysteem en de validatie van de emissiecijfers door de Nederlandse Emissieregistratie.

Betere informatie over ammoniakemissies De berekende ammoniakemissies in Nederland blijven dalen. Wel blijkt voor de periode 1990-2013 dat de totale emissie van ammoniak hoger is dan eerder gerapporteerd. De ammoniakemissies kunnen dankzij betere waarnemingen nauwkeuriger worden berekend en een aantal nieuwe bronnen zijn toegevoegd. De emissies van stikstofoxiden, zwaveldioxiden en niet methaan vluchtige organische stoffen blijven licht dalen. RIVM verzamelt en rapporteert de emissiecijfers samen met partnerinstituten in het project Emissieregistratie. Nieuwe onderzoeksresultaten Op basis van nieuw onderzoek konden bepaalde emissiefactoren voor ammoniak nauwkeuriger worden berekend. Zo is geconstateerd dat er per varken meer ammoniak wordt uitgestoten dan eerder verondersteld. Daarnaast blijkt de mest die over het land wordt verspreid meer stikstof te bevatten, waardoor er meer ammoniak vrijkomt. Verder is berekend dat de bijdrage van wegverkeer aan de ammoniakuitstoot in Nederland hoger is dan eerder gerapporteerd. Eén van de bronnen die volgens internationale voorschriften (EMEP Guidebook 2013) nu is meegenomen in de berekeningen, is de uitstoot van ammoniak door gewassen terwijl ze rijpen ('gewasafrijping') en na de oogst ('gewasresten'). Hetzelfde geldt voor de ammoniak die vrijkomt bij het gebruik van compost (zoals van gft-afval) en het slib van rioolwaterzuiveringsinstallaties. Het RIVM verzamelt en analyseert de cijfers. Behalve bovengenoemde stoffen gaat het om de uitstoot van koolmonoxide, fijn stof, zware metalen en persistente organische stoffen. De uitstoot van al deze stoffen is tussen 1990 en 2013 gedaald. Dit komt vooral door schonere auto's en brandstoffen en door emissiebeperkende maatregelen in de industrie.

Agricultural activities are in the Netherlands a major source of ammonia (NH3), nitrogen oxide (NO), nitrous oxide (N2O),methane (CH4) and particulate matter (PM10 and PM2.5). The emissions in 2014 were calculated using the National EmissionModel for Agriculture (NEMA). At the same time some figures in the time series 1990-2013 were revised. The method calculatesthe ammonia emission from livestock manure on the basis of the total ammonia nitrogen (TAN) content in manure. Ammoniaemissions from livestock manure, fertilizers and other sources in 2014 were 121 million kg, which was almost 4 million kghigher than in 2013, mainly due to expansion of the dairy herd and a higher N-content of roughage. N2O emissions increasedfrom 19.1 million kg in 2013 to 19.4 million kg in 2014. NO emission increased slightly from 16.9 to 17.2 million kg. Methaneemissions increased from 499 to 503 million kg. Emissions of particulate matter increased slightly from 6.3 to 6.4 million kgPM10 as a result of higher poultry numbers. Emission of PM2.5 in both years was 0.6 million kg. Ammonia emissions fromlivestock manure in the Netherlands dropped by almost two thirds since 1990, mainly as a result of lower nitrogen excretionrates by livestock and low-emission manure application. Nitrous oxide and nitrogen oxide also fell over the same period, butless steeply (by about 40%), due to higher emissions from manure injection into the soil and to the shift from poultry housingsystems based on liquid manure to solid manure systems. De emissie vanfijnstof nam licht toe van 6,3 miljoen kg PM10 tot 6,4 miljoen kg, door een toename van het aantal stuks pluimvee. De emissievan PM2,5 bedroeg in beide jaren 0,6 miljoen kg. Sinds 1990 is de ammoniakemissie uit dierlijke mest en kunstmest mettweederde gedaald, vooral door een lagere stikstofuitscheiding door landbouwhuisdieren en emissiearme mesttoediening.Emissies van lachgas en stikstofoxide daalden in dezelfde periode eveneens, maar minder sterk (ca. 40%) omdat doorondergronds toedienen van mest de emissies hoger zijn geworden en door de omschakeling van stalsystemen met dunne naarvaste mest bij pluimvee. Tussen 1990 en 2014 daalde de emissie van methaan met 16% door een afname in de dieraantallenen een hogere voeropname en productiviteit van melkvee.

Total greenhouse gas (GHG) emissions from the Netherlands in 2015 increased by approximately 4%, compared with 2014 emissions. This increase was mainly the result of the increased electricity production in coal fired plants compared to 2014. Furthermore fuel combustion in all sectors was increased as the winter of 2015 was less mild as the one in 2014. In 2015, total GHG emissions (including indirect CO2 emissions and excluding emissions from Land use, land use change and forestry (LULUCF)) in the Netherlands amounted to 195.2 Tg CO2 eq. This is approximately 12.5% below the emissions in the base year2 (223.1 Tg CO2 eq). CO2 emission have increased above the level in the base year 1990 in 2015 (+ 1.5%). This increase was offset by the reduction in the emissions since 1990 of methane, nitrous oxide and fluorinated gases (CH4, N2O and F-gases). This report documents the Netherlands’ 2017 annual submission of its greenhouse gas emissions inventory in accordance with the 2006 IPCC Guidelines for National Greenhouse Gas Inventories (IPCC, 2006) provided by the United Nations Framework Convention on Climate Change (UNFCCC), the Kyoto Protocol and the European Union’s Greenhouse Gas Monitoring Mechanism. The report includes explanations of observed trends in emissions; an assessment of the sources with the highest contribution to the national emissions (key sources) and the uncertainty in their emissions; an itemization of methods, data sources and emission factors (EFs) applied; and a description of the quality assurance system and the verification activities performed on the data.

Arctic amplification of climate warming is intensifying the thaw and coastal erosion of the widespread and carbon-rich Siberian Ice Complex Deposits (ICD). Despite the potential for altering long-term carbon dynamics in the Arctic, the susceptibility of organic carbon (OC) to degradation as the ICD thaw is poorly characterised. This study identifies signs of OC degradation in three Siberian ICD regimes of coastal erosion through elemental, isotopic and molecular analyses. The degree of erosion appears to determine the extent of degradation. The moisture-limited and beach-protected ICD bluff near Buor-Khaya Cape, characterised by thermokarst mounds (baydzherakhs), represents a dormant regime with limited ongoing degradation. Conversely, the more exposed ICD scarps on eroding riverbanks (Olenek Channel, Lena Delta) and coastal slopes (Muostakh Island) showed more pronounced signs of ongoing OC decay. Different parameters suggest that degradation can partially explain the shift of the OC signature with 14C age in the thawing ICD. Exposure time, degree of erosion, slope gradient and moisture conditions appear to be key factors determining the degradation propensity of OC in exposed ICD. These field results document the lability of OC in ICD upon thaw and illustrate the potential for transferring old OC into the rapidly cycling atmosphere-biosphere carbon pools.

International audience; The study of ecological services (ESs) is fast becoming a cornerstone of mainstream ecology, largely because they provide a useful means of linking functioning to societal benefits in complex systems by connecting different organizational levels. In order to identify the main challenges facing current and future ES research, we analyzed the effects of the publication of the Millennium Ecosystem Assessment (MEA, 2005) on different disciplines. Within a set of topics framed around concepts embedded within the MEA, each co-author identified five key research challenges and, where feasible, suggested possible solutions. Concepts included those related to specific service types (i.e. provisioning, supporting, regulating, cultural, aesthetic services) as well as more synthetic issues spanning the natural and social sciences, which often linked a wide range of disciplines, as was the case for the application of network theory. By merging similar responses, and removing some of the narrower suggestions from our sample pool, we distilled the key challenges into a smaller subset. We review some of the historical context to the MEA and identify some of the broader scientific and philosophical issues that still permeate discourse in this field. Finally, we consider where the greatest advances are most likely to be made in the next decade and beyond.

RIVM rapport:Total greenhouse gas (GHG) emissions from the Netherlands in 2015 increased by approximately 4%, compared with 2014 emissions. This increase was mainly the result of the increased electricity production in coal fired plants compared to 2014. Furthermore fuel combustion in all sectors was increased as the winter of 2015 was less mild as the one in 2014. In 2015, total GHG emissions (including indirect CO2 emissions and excluding emissions from Land use, land use change and forestry (LULUCF)) in the Netherlands amounted to 195.2 Tg CO2 eq. This is approximately 12.5% below the emissions in the base year2 (223.1 Tg CO2 eq). CO2 emission have increased above the level in the base year 1990 in 2015 (+ 1.5%). This increase was offset by the reduction in the emissions since 1990 of methane, nitrous oxide and fluorinated gases (CH4, N2O and F-gases). This report documents the Netherlands' 2017 annual submission of its greenhouse gas emissions inventory in accordance with the 2006 IPCC Guidelines for National Greenhouse Gas Inventories (IPCC, 2006) provided by the United Nations Framework Convention on Climate Change (UNFCCC), the Kyoto Protocol and the European Union's Greenhouse Gas Monitoring Mechanism. The report includes explanations of observed trends in emissions; an assessment of the sources with the highest contribution to the national emissions (key sources) and the uncertainty in their emissions; an itemization of methods, data sources and emission factors (EFs) applied; and a description of the quality assurance system and the verification activities performed on the data., In 2015 is de totale uitstoot van broeikasgassen van Nederland met ongeveer 4 procent gestegen ten opzichte van de emissie in 2014. Deze stijging komt vooral doordat er meer elektriciteit is geproduceerd in de door steenkool gestookte elektriciteitscentrales. Daarnaast is er meer brandstof gebruikt voor ruimteverwarming dan in 2014 als gevolg van de koudere winter. Totaal van alle broeikasgassen gedaald De totale emissie van broeikasgassen naar de lucht wordt uitgedrukt in CO2-equivalenten en bedroeg in 2015 195,2 miljard kilogram (megaton of teragram). Ten opzichte van het zogeheten Kyoto-basisjaar (223,1 miljard kilogram CO2-equivalenten) is dit een afname van ongeveer 12,5 procent. Dit basisjaar, dat afhankelijk van het broeikasgas 1990 of 1995 is, dient voor het Kyoto-protocol als referentiejaar voor de uitstoot van broeikasgassen. CO2 emissie gestegen tot boven niveau 1990 De emissie van CO2 lag in 2014 voor het eerst onder het niveau van het basisjaar 1990. In 2015 is de CO2-uitstoot toegenomen met 4,5% en komt daarmee weer boven het niveau van het basisjaar 1990 (+1,5 procent) te liggen. Deze toename werd voor de totale emissie van broeikasgassen ruim gecompenseerd door de lagere emissies van methaan, distikstofoxide en gefluoreerde gassen (CH4, N2O en F-gassen). Dit blijkt uit een inventarisatie van broeikasgasemissies die het RIVM jaarlijks op verzoek van het ministerie van Infrastructuur en Milieu (IenM) opstelt. Met deze inventarisatie voldoet Nederland aan de nationale rapportageverplichtingen voor 2017 van het Klimaatverdrag van de Verenigde Naties (UNFCCC), van het Kyoto Protocol en van het Bewakingsmechanisme Broeikasgassen van de Europese Unie. De inventarisatie bevat verder trendanalyses voor de emissies van broeikasgassen in de periode 1990-2015, een analyse van belangrijkste emissiebronnen ('sleutelbronnen'), evenals de onzekerheid in hun emissies. - Abstract
Few genome-wide association studies (GWAS) account for environmental exposures, like smoking, potentially impacting the overall trait variance when investigating the genetic contribution to obesity-related traits. Here, we use GWAS data from 51,080 current smokers and 190,178 nonsmokers (87% European descent) to identify loci influencing BMI and central adiposity, measured as waist circumference and waist-to-hip ratio both adjusted for BMI. We identify 23 novel genetic loci, and 9 loci with convincing evidence of gene-smoking interaction (GxSMK) on obesity-related traits. We show consistent direction of effect for all identified loci and significance for 18 novel and for 5 interaction loci in an independent study sample. These loci highlight novel biological functions, including response to oxidative stress, addictive behaviour, and regulatory functions emphasizing the importance of accounting for environment in genetic analyses. Our results suggest that tobacco smoking may alter the genetic susceptibility to overall adiposity and body fat distribution.

© The Author 2017. Published by Oxford University Press. All rights reserved. Pre-pregnancy maternal obesity is associated with adverse offspring outcomes at birth and later in life. Individual studies have shown that epigenetic modifications such as DNA methylation could contribute. Within the Pregnancy and Childhood Epigenetics (PACE) Consortium, we meta-analysed the association between pre-pregnancy maternal BMI and methylation at over 450,000 sites in newborn blood DNA, across 19 cohorts (9,340 mother-newborn pairs). We attempted to infer causality by comparing the effects of maternal versus paternal BMI and incorporating genetic variation. In four additional cohorts (1,817 mother-child pairs), we meta-analysed the association between maternal BMI at the start of pregnancy and blood methylation in adolescents. In newborns, maternal BMI was associated with small ( < 0.2% per BMI unit (1 kg/m 2 ), P < 1.06 × 10 -7 ) methylation variation at 9,044 sites throughout the genome. Adjustment for estimated cell proportions greatly attenuated the number of significant CpGs to 104, including 86 sites common to the unadjusted model. At 72/86 sites, the direction of the association was the same in newborns and adolescents, suggesting persistence of signals. However, we found evidence for a6causal intrauterine effect of maternal BMI on newborn methylation at just 8/86 sites. In conclusion, this well-powered analysis identified robust associations between maternal adiposity and variations in newborn blood DNA methylation, but these small effects may be better explained by genetic or lifestyle factors than a causal intrauterine mechanism. This highlights the need for large-scale collaborative approaches and the application of causal inference techniques in epigenetic epidemiology.

To the Editor: Complex diseases, including asthma, have genetic and environmental origins. Genome-wide association studies have identified multiple genes for the development of asthma, yet they only explain a limited proportion of asthma heritability. Interactions between genetic predisposition and exposure to passive smoking might explain in part the hidden heritability of childhood asthma. However, to date, this approach has not been reported for the discovery of interactions between genes and tobacco smoke exposure. We performed a genome-wide interaction study (GWIS) on childhood asthma to identify genes that interact with 2 well-known environmental risk factors for childhood-onset asthma: in utero and childhood tobacco smoke exposure. We meta-analyzed interaction results from 9 studies participating in the GABRIEL consortium1 including more than 6,000 subjects of European descent. We replicated our findings in 4 independent studies including more than 13,000 subjects. Childhood-onset asthma was defined as asthma diagnosed by a doctor before the age of 16 years, which is consistent with the definition in the GABRIEL consortium.1 In utero tobacco smoke exposure was defined as “exposure to maternal tobacco smoking at any time during pregnancy.” Childhood tobacco smoke exposure was defined as “exposure to passive tobacco smoking at any time from birth until 16 years of age.” Details on the number of subjects, the design of the individual studies, and outcome and exposure definitions are provided in Tables E1 to E4 in this article's Online Repository at www.jacionline.org. The effects of in utero tobacco smoke exposure and childhood tobacco smoke exposure were analyzed separately. All individual studies were analyzed by using a logistic regression model containing the genetic effect, the effect of tobacco smoke exposure, and an interaction term indicating the interaction between the genetic effect and tobacco smoke exposure. Further methodological considerations on GWISs and details on the statistical analyses are described in this article's Online Repository at www.jacionline.org. For in utero tobacco smoke exposure, the discovery genome-wide meta-analysis consisted of 2,654 cases and 3,073 control subjects derived from 7 studies (see Table E1). Overall, in utero tobacco smoke exposure increased the risk of childhood-onset asthma (see Fig E1 in this article's Online Repository at www.jacionline.org). A total of 536,705 single nucleotide polymorphisms (SNPs) were included in the interaction meta-analysis. Fig E2 in this article's Online Repository at www.jacionline.org shows the Manhattan plot. We identified 27 SNPs in the discovery sample with a P value of less than 10−4 based on the fixed effect model (Table I and see Table E5 in this article's Online Repository at www.jacionline.org). Findings did not reach genome-wide significance but were consistent over all studies included, and no significant heterogeneity across studies was present (P value Q-statistic < .05). Four of these SNPs on chromosome 10 were in high linkage disequilibrium with each other in the discovery meta-analysis (r2 = 0.82-0.96). The most prominent marker was located on chromosome 18 near EPB41L3 (Forest plot, see Fig E3 in this article's Online Repository at www.jacionline.org). Table E6 in this article's Online Repository at www.jacionline.org shows the associations in exposed and nonexposed subjects. EPB41L3 belongs to the protein 4.1 family of membrane-associated proteins, is involved in cell-cell junctions,2 and might play a role in apoptosis.3 The literature shows that in utero tobacco smoke exposure affects the expression of genes involved in biological processes, such as cell proliferation and apoptosis, and influences lung development of the child in general.4 Our data suggest that this effect of in utero smoke exposure might potentially occur through mechanisms involving EPB41L3 (see the additional text in this article's Online Repository). Table I Results of the GWIS of in utero tobacco smoke exposure and childhood-onset asthma For childhood tobacco smoke exposure, the discovery genome-wide meta-analysis consisted of 3,048 cases and 3,509 control subjects derived from 9 studies (see Table E1). Overall, childhood tobacco smoke exposure increased the risk of childhood-onset asthma (see Fig E1). A total of 538,233 SNPs were included in the interaction meta-analysis. We identified 35 SNPs in the discovery sample with a P value of less than 10−4 based on the fixed effect model. Four of these SNPs were excluded because they showed heterogeneity, and the P value of the random effect was greater than 10−4. Findings did not reach genome-wide significance. Table II and Table E7 (see this article's Online Repository at www.jacionline.org) the results for the top SNPs. Seven SNPs on chromosome 5 (except rs2312164) were in high linkage disequilibrium with each other in the discovery studies (r2 = 0.83-1.00). Table II Results of the GWIS on childhood tobacco smoke exposure and childhood-onset asthma The most prominent marker was located on chromosome 6 in PACRG (parkin coregulated gene; Forest plot, see Fig E5 in this article's Online Repository at www.jacionline.org). Table E8 in this article's Online Repository at www.jacionline.org shows the associations in exposed and nonexposed subjects. PACRG is located next to and has an overlapping promoter region with parkin 2 (PARK2).5 The gene has been associated with leprosy and parkinsonian diseases and has an important role in motile cilia function and cilia morphogenesis.2,6 PACRG is relatively highly expressed in the trachea and nasal mucosa. Ciliary dysfunction might impair mucus clearance from the airways and has been shown to affect asthma severity. Our data suggest that changes in ciliary function particularly affect the development of asthma in children exposed to passive tobacco smoke. The genes that have been reported previously to interact with tobacco smoke exposure with respect to asthma development (ie, TNF,7 GSTP1,7 and ADAM338) were not among our most significant hits. This can be explained by the fact that the genetic variants in these candidate gene studies have a strong main effect on asthma development. Bouzigon et al9 showed a more pronounced effect of the 17q21 region on the development of early-onset asthma in children with early-life tobacco smoke exposure than in those without. The genetic effect of these markers in our GWIS showed a similar direction, but the interaction was not significant. This study on childhood asthma is the first hypothesis-free GWIS specifically aiming to identify SNPs that interact with tobacco smoke exposure in disease development. We found suggestive evidence for an interaction between rs8094633 on chromosome 18 near EPB41L3 and in utero tobacco smoke exposure and an interaction between rs1575472 on chromosome 6 in PACRG and childhood tobacco smoke exposure. The SNPs found have not been identified previously in general genome-wide association studies on childhood asthma. Interestingly, the SNPs interacting with in utero and childhood tobacco smoke exposure were different and were not involved in the same pathway (see Fig E6 in this article's Online Repository at www.jacionline.org). Interactions between these SNPs and tobacco smoke exposure in utero and in childhood might explain part of the missing heritability of asthma. Future research needs to confirm these findings and further unravel the biological pathways.
