A new original "cryostabilization" technique (Dore, 1992; Dore et al, 2004) was applied to different animal tissues and organs. In this study, we apply this technique also to vegetal tissues (ripening and ripe fleshy fruits, buds and embryos). The proposed method preserves good tissue morphology and intracellular substances, and avoids loss of epicuticular waxes; moreover it maintains in situ some enzymatic activities. Being some samples very difficult to handle the original protocol had to be modified. The correct orientation in embedding of samples is an important requirement which is not easy to achieve when deadling with small fragments of tissues.

The Large High Altitude Air Shower Observatory (LHAASO) project is a new generation multi-component instrument, to be built at 4410 meters of altitude in the Sichuan province of China, with the aim to study with unprecedented sensitivity the spec trum, the composition and the anisotropy of cosmic rays in the energy range between 10$^{12}$ and 10$^{18}$ eV, as well as to act simultaneously as a wide aperture (one stereoradiant), continuously-operated gamma ray telescope in the energy range between 10$^{11}$ and $10^{15}$ eV. The experiment will be able of continuously surveying the TeV sky for steady and transient sources from 100 GeV to 1 PeV, t hus opening for the first time the 100-1000 TeV range to the direct observations of the high energy cosmic ray sources. In addition, the different observables (electronic, muonic and Cherenkov/fluorescence components) that will be measured in LHAASO will allow to investigate origin, acceleration and propagation of the radiation through a measurement of energy spec trum, elemental composition and anisotropy with unprecedented resolution. The remarkable sensitivity of LHAASO in cosmic rays physics and gamma astronomy would play a key-role in the comprehensive general program to explore the High Energy Universe. LHAASO will allow important studies of fundamental physics (such as indirect dark matter search, Lorentz invariance violation, quantum gravity) and solar and heliospheric physics. In this document we introduce the concept of LHAASO and the main science goals, providing an overview of the project.

The ARGO-YBJ experiment, a full coverage extensive air shower (EAS) detector located at high altitude (4300 m a.s.l.) in Tibet, China, has smoothly taken data, with very high stability, since November 2007 to the beginning of 2013. The array consisted of a carpet of about 7000 m2 Resistive Plate Chambers (RPCs) operated in streamer mode and equipped with both digital and analog readout, providing the measurement of particle densities up to few particles per cm2. The unique detector features (full coverage, readout granularity, wide dynamic range, ..) and location (very high altitude) allowed a detailed study of the lateral density profile of charged particles at ground very close to the shower axis and its description by a proper lateral distribution function (LDF). In particular, the information collected in the first 10 m from the shower axis have been shown to provide a very effective tool for the determination of the shower development stage ("age") in the energy range 50 TeV - 10 PeV. The sensitivity of the age parameter to the mass composition of primary Cosmic Rays is also discussed.

Se analizó la composición cuantitativa del macrozoobentos de los ríos Potrero y Chorrillo a lo largo de 7 estaciones de muestreo. Se registraron 59 taxa de macroinvertebrados bentónicos.Los taxa más representativos fueron: Ephemeroptera, Diptera, Coleoptera, Trichoptera y Oligochaeta. El análisis de agrupamiento entre las estaciones de muestreo evidenció dostramos: el primero agrupó a las estaciones anteriores al dique y elsegundo a las estaciones posteriores.La composición faunística presentó cambios significativos a lo largo del perfil longitudinal. El tramo postembalse tuvo una disminución en la diversidad, la riqueza taxonómica y la abundancia relativa. En las dos estaciones posteriores a los embalses la comunidad estaba marcada por la dominancia de muy pocas especies. La perturbación del dique Potrero crea un tramo regulado con características particulares ya que permanece aislado por largos períodos.

Abstract Background Rare genetic variation in the human population is a major source of pathophysiological variability and has been implicated in a host of complex phenotypes and diseases. Finding disease-related genes harboring disparate functional rare variants requires sequencing of many individuals across many genomic regions and comparing against unaffected cohorts. However, despite persistent declines in sequencing costs, population-based rare variant detection across large genomic target regions remains cost prohibitive for most investigators. In addition, DNA samples are often precious and hybridization methods typically require large amounts of input DNA. Pooled sample DNA sequencing is a cost and time-efficient strategy for surveying populations of individuals for rare variants. We set out to 1) create a scalable, multiplexing method for custom capture with or without individual DNA indexing that was amenable to low amounts of input DNA and 2) expand the functionality of the SPLINTER algorithm for calling substitutions, insertions and deletions across either candidate genes or the entire exome by integrating the variant calling algorithm with the dynamic programming aligner, Novoalign. Results We report methodology for pooled hybridization capture with pre-enrichment, indexed multiplexing of up to 48 individuals or non-indexed pooled sequencing of up to 92 individuals with as little as 70 ng of DNA per person. Modified solid phase reversible immobilization bead purification strategies enable no sample transfers from sonication in 96-well plates through adapter ligation, resulting in 50% less library preparation reagent consumption. Custom Y-shaped adapters containing novel 7 base pair index sequences with a Hamming distance of ≥2 were directly ligated onto fragmented source DNA eliminating the need for PCR to incorporate indexes, and was followed by a custom blocking strategy using a single oligonucleotide regardless of index sequence. These results were obtained aligning raw reads against the entire genome using Novoalign followed by variant calling of non-indexed pools using SPLINTER or SAMtools for indexed samples. With these pipelines, we find sensitivity and specificity of 99.4% and 99.7% for pooled exome sequencing. Sensitivity, and to a lesser degree specificity, proved to be a function of coverage. For rare variants (≤2% minor allele frequency), we achieved sensitivity and specificity of ≥94.9% and ≥99.99% for custom capture of 2.5 Mb in multiplexed libraries of 22–48 individuals with only ≥5-fold coverage/chromosome, but these parameters improved to ≥98.7 and 100% with 20-fold coverage/chromosome. Conclusions This highly scalable methodology enables accurate rare variant detection, with or without individual DNA sample indexing, while reducing the amount of required source DNA and total costs through less hybridization reagent consumption, multi-sample sonication in a standard PCR plate, multiplexed pre-enrichment pooling with a single hybridization and lesser sequencing coverage required to obtain high sensitivity.

In this paper we report on the observations of TeV gamma ray sources performed by the air shower de- tector ARGO-YBJ. The objects studied in this work are the blazar Markarian 421 and the extended galactic source MGROJ1908+06, monitored during 2 years of operation. Mrk421 has been detected by ARGO-YBJ with a statisti- cal significance of 11 standard deviations. The observed TeV emission was highly variable, showing large enhance- ments of the flux during active periods. The study of the spectral behaviour during flares revealed a positive correla- tion of the hardness with the flux, as already reported in the past by the Whipple telescope, suggesting that this is a long term property of the source. ARGO-YBJ observed a strong correlation between TeV gamma rays and the X-ray flux measured by RXTM/ASM and SWIFT/BAT during the whole period, with a time lag compatible with zero, supporting the one-zone SSC model to describe the emission mechanism. MGROJ1908+06 has been detected by ARGO-YBJ with 5 standard deviation of significance. From our data the source appears extended and the measured extension is ext=0.48 +0.26 0.28 , in agreement with a previous HESS obser- vation. The average flux is in marginal agreement with that reported by MILAGRO, but significantly higher than that ob- tained by HESS, suggesting a possible flux variability. The large field of view ( 2 sr) and the high duty cycle allow a continuous monitoring of the sky in the declination band from 0 to 70 . In this paper we report on the ob- servations of two bright gamma ray sources, performed dur- ing 2 years: the blazar Mrk421 and the extended galactic source MGROJ1908+06.

The detector is made of a single layer of RPCs operated in streamer mode, fully. instrumenting a central carpet of about 5700 m2, then a guard ring extends the partially instrumented area to. about 11,000 m2. The large field of view (∼ 2 sr) and the high duty cycle (≥ 85%) allow a continuous monitoring. of the sky in the declination band from -10. ◦. to 70. ◦. ; the detector operates with an energy threshold of a few. hundred GeV. Cosmic ray antiprotons provide an important probe for the study of cosmic-ray propagation in the interstellar space and to investigate the existence of Galactic dark matter. Cosmic rays are hampered by the Moon, therefore a deficit of cosmic rays in its direction is expected (the so-called Moon shadow). The Earth–Moon system acts as a magnetic spectrometer. In fact, due to the geomagnetic field the center of the Moon shifts westward by an amount depending on the primary cosmic ray energy. Paths of primary antiprotons are therefore deflected in an opposite sense in their way to the Earth. This effect allows, in principle, the search of antiparticles in the opposite direction of the observed Moon shadow. The ARGO-YBJ experiment, in stable data taking since November 2007 with an energy threshold of a few 100s of GeV, is observing the Moon shadow with high statistical significance. Using about 1 year data, an upper limit of the p ¯ / p flux ratio in the few-TeV energy region is set to a few percent with a confidence level of 90%.

We perform simulations for future Cherenkov Telescope Array (CTA) observations of RX~J1713.7$-$3946, a young supernova remnant (SNR) and one of the brightest sources ever discovered in very-high-energy (VHE) gamma rays. Special attention is paid to explore possible spatial (anti-)correlations of gamma rays with emission at other wavelengths, in particular X-rays and CO/H{\sc i} emission. We present a series of simulated images of RX J1713.7$-$3946 for CTA based on a set of observationally motivated models for the gamma-ray emission. In these models, VHE gamma rays produced by high-energy electrons are assumed to trace the non-thermal X-ray emission observed by {\it XMM-Newton}, whereas those originating from relativistic protons delineate the local gas distributions. The local atomic and molecular gas distributions are deduced by the NANTEN team from CO and H{\sc i} observations. Our primary goal is to show how one can distinguish the emission mechanism(s) of the gamma rays (i.e., hadronic vs leptonic, or a mixture of the two) through information provided by their spatial distribution, spectra, and time variation. This work is the first attempt to quantitatively evaluate the capabilities of CTA to achieve various proposed scientific goals by observing this important cosmic particle accelerator., Comment: 19 pages, 5 figures. In these models, VHE gamma rays produced by high-energy electrons are assumed to trace the nonthermal X-ray emission observed by XMM-Newton, whereas those originating from relativistic protons delineate the local gas distributions. The local atomic and molecular gas distributions are deduced by the NANTEN team from CO and H i observations. Our primary goal is to show how one can distinguish the emission mechanism(s) of the gamma rays (i.e., hadronic versus leptonic, or a mixture of the two) through information provided by their spatial distribution, spectra, and time variation. This work is the first attempt to quantitatively evaluate the capabilities of CTA to achieve various proposed scientific goals by observing this important cosmic particle accelerator., Published by IOP, Chicago, Ill. ARGO-YBJ is a full coverage layer of Resistive Plate Counters (RPCs) covering an area of about 5.800 m 2 to be installed at the YangBaJing Laboratory (Tibet, China) at 4.300 m a.s.l . The DAQ system is based on a Central Station which receives the detector information from distributed readout electronics. Data from each RPC are acquired from a Receiver Card which reads out and digitizes the space and time information from 10 pick-up pads and gives out the pad multiplicity for trigger purposes. The Receiver Cards are hosted in Local Stations which Group 12 RPCs. On trigger occurrence, each Local Station sends the collected data to the Central Station. Both the detector and the electronics allow 1 ns accuracy in time measurements. In this work a detailed description of the Local Station and the Trigger System are presented.
