Abstract

Many agonists evoke events in the cell nucleus through the control of Ca2+ signals. Recent studies using isolated nuclei have indicated that the nuclear envelope is a store for nuclear Ca2+. However, the release of Ca2+ directly from the nuclear envelope in living cells has never been reported. In the present study, we have investigated the changes of Ca2+ signals at the cyto-nucleoplasmic interface of rat C6 glioma cells using confocal microscopy. Digital imaging indicates that fluo-3, a Ca2+-sensitive fluorescent probe, was concentrated in or around the nuclear envelope. Our experiments also revealed that C6 cells at rest produced spontaneous Ca2+ spikes in the absence of chemical stimulation. The amplitude of the repetitive Ca2+ spikes was higher at the nuclear envelope than in the whole cell or cytosol. After image subtraction, circular rims of Ca2+ release and uptake were seen at the outer boundary of the nucleus. When the cells were treated with thapsigargin (2 μM), a specific Ca2+-ATPase inhibitor, a long-lasting Ca2+ release was observed at the nuclear envelope. Moreover, most of the released Ca2+ was directed inwardly to the nucleoplasm with little outward diffusion. Our results thus indicate: (1) that the nuclear envelope is a Ca2+ store that possesses the ability to discharge and sequestrate Ca2+; and (2) the Ca2+-releasing channels are present in the inner nuclear membrane. Abstract

Adenosine 3', 5'-monophosphate (cAMP) was measured in the CSF of 42 patients undergoing radiological investigation, neurosurgical procedures, or investigation of hepatic coma. The concentration of cAMP was significantly higher in ventricular CSF than in lumbar CSF. Premedication with pentobarbitone plus promethazine increased cAMP in lumbar CSF. There was no difference in cAMP concentration in lumbar CSF obtained before or after injection of air or after the administration of diazepam during lumbar pneumoencephalography. Lumbar CSF cAMP concentration was significantly increased in patients in hepatic coma. The concentration of cAMP in the lateral ventricle was not affected by general anaesthesia or by the presence of a complete block of the aqueduct of Sylvius. There was no decrease in lumbar CSF cAMP in patients with a complete stenosis of the aqueduct of Sylvius, partial blocks of CSF flow at the cervical level, or a complete block at the lower thoracic level. The concentration of cisternal CSF cAMP was similar to that of lumbar CSF. These results suggest that (1) there is a ventriculolumbar gradient in the concentration of cAMP but of insufficient magnitude to be detected by mixing of lumbar and ventricular CSF during pneumoencephalography, (2) lumbar CSF cAMP concentration is not dependent on brain as a source of this nucleotide; the source of this nucleotide may be largely derived from the spinal cord, (3) premedication may affect the concentration of cAMP in lumbar CSF cAMP, (4) the formation of cAMP is unimpaired in hepatic coma. 165. Optimal Scheduling of Cooperative Spectrum Sensing in Cognitive Radio Networks

Abstract

In cognitive radio (CR) networks, secondary users can be coordinated to perform spectrum sensing so as to detect primary user activities more accurately. However, more sensing cooperations for a channel may decrease the transmission time of the secondary users, or lose opportunities for exploiting other channels. In this paper, we study this tradeoff by using the theory of oartially observable Markov decision process (POMDP). This formulation leads to an optimal sensing scheduling policy that determines which secondary users sense which channels with what miss detection probability and false alarm probability. A myopic policy with lower complexity yet comparable performance is also proposed. We further analytically study the properties and the solution structure for the myopic and the optimal policies under a simplified system model. Theoretical results reveal that under certain conditions, some simple but robust structures of the value function exist, which lead to an easy way to obtain the solution of POMDP. Moreover, the cooperative sensing scheduling problem embedded in our POMDP, which is generally a hard combinatorial problem, can be analyzed in an efficient way. Numerical and simulation results are provided to illustrate that our design can utilize the spectrum more efficiently for CR users.

167. Applications of Multipore Model for the Mechanism Identification during the Adsorption of Dye on Activated Carbon and Bagasse Pith

Abstract

A solution to the intraparticle diffusion model has been developed on the basis of the pore structure of adsorbents. This global model incorporates the mechanism of external film mass transport, intraparticle macropore surface diffusion, and adsorption at the surface in the micropores limited by a first-order reaction kinetic rate constant. To assess the model, it has been applied to the adsorption of acid and basic dyes onto two adsorbents, active carbon and bagasse pith. The two adsorbents were selected because of their differences:  active carbon has a large BET surface area and microporous structure, whereas bagasse pith has a small surface area and only a spongelike macropore structure. The branched pore model developed and tested is able to distinguish these physical−structural characteristic differences between the two adsorbents using two very different adsorbates.

172. Comparison of screening methods for detection of extended‐spectrum β‐lactamases and their prevalence among Escherichia coliand Klebsiellaspecies in Hong Kong

Abstract

Three tests, the disk diffusion test, the double‐disc synergy test and the inhibitor‐potentiated disc diffusion test, were compared for their abilities to detect production of extended‐spectrum β‐lactamases (ESBL) in 702 Escherichia coliand 472 Klebsiellaspp. strains from four hospitals. Eleven percent E. coliand 13%Klebsiellaspp. were found to produce ESBL. As an indicator of ESBL activity, the sensitivities of the five extended‐spectrum β‐lactams were as follows: cefotaxime (100%), cefpodoxime (99.3%), ceftriaxone (98.6%), aztreonam (93%) and ceftazidime (57.7%) when interpreted using the National Committee for Clinical Laboratory Standards criteria. Their positive predictive values ranged from 67.8–83.8%. Both the inhibitor‐potentiated disc diffusion test and the double‐disc synergy test (at three inter‐disc widths of 20, 25 and 30 mm) were capable of identifying all the ESBL‐producers. However, at a single inter‐disc width of 30 mm, the double‐disc synergy test has limited sensitivity (83.8%). As a second test for confirming ESBL activity in strains with reduced susceptibility to β‐lactams, the inhibitor‐potentiated disc diffusion test is therefore a simple and reliable option.

Abstract

We propose a simple approximate model for a nonsaturated IEEE 802.11 DCF network. Our model is much simpler than others that have appeared in the literature. The main simplification we introduce is that the attempt rate in the nonsaturated setting can be approximated by scaling the attempt rate of the saturated setting with the probability of a packet arrival. We verify our model using ns-2 simulation and show that our MAC access delay result is the most accurate amongst related work, while our collision probability and throughput results achieve comparable accuracy to [1]. Abstract

The release of labelled noradrenaline from rat cerebral cortes slices preloaded with L-[7,8- 3 H]noradrenaline ([ 3 H]NA) was measured in Krebs-Ringer-Bicarbonate media containing either 5.0 (KR) or 50 mM KCl. Experiments were also performed in media containing either 2.5 mM of or no calcium. Total ginsenoside, at concentrations ranged from 0.001 to 50 μg/ml, reduced the high K + -evoked release of [ 3 H]NA in calcium-containing but not in calcium-free media, and this inhibition was concentration-dependent. It, however, failed to affect the release of [ 3 H]NA significantly from undepolarized samples in KR media. These results are in agreement with suggestion that ginsenosides may affect behavior and brain function.

178. Ca 2+is released from the nuclear tubular structure into nucleoplasm in C6 glioma cells after stimulation with phorbol ester

Abstract

It is well established that cellular Ca 2+is an important messenger that controls many nuclear functions but the source of nuclear Ca 2+is far from clear. It has long been thought that Ca 2+is translocated from the cytosol over a long distance to activate the nuclear transcription machinery. However, this model is at best an incomplete one. With the aid of confocal microscopy, we observed tubules extended deep inside the nucleus of C6 cells in agreement with previous studies (Fricker et al. (1997) J. Cell Biol. 136, 531–544). When cells were stimulated with phorbol 12-myristate 13-acetate or phorbol 12,13-diacetate, Ca 2+was released from these tubules. DiOC6(3), a vital marker for intracellular membranes, stained the tubule in the nucleus of the same cell used for Ca 2+imaging. Moreover, results from labelling the cells with rhodamine 123 further indicate that the tubule was formed by a double-membraned invagination with mitochondria inside. Studies with acridine orange showed that chromatin was excluded from the tubules. Taken together, our results demonstrate that the nuclear tubule is a structural entity responsible for the release of Ca 2+into the nucleoplasm after stimulation with phorbol ester.

Abstract

The expression of intercellular adhesion molecule 2 (ICAM-2) in adult tissues is restricted to vascular endothelial cells and megakaryocytes. We have previously shown that the endothelial-specific in vivo activity of the human ICAM-2 promoter is contained within a small (0.33-kilobase (kbp)) 5'-flanking region of the gene. Here we describe the in vitro characterization of this region. The ICAM-2 promoter is TATA-less, and transcription in endothelial cells initiates at four sites. Reporter gene expression directed by the promoter was 125-fold greater than vector alone in bovine aortic endothelial cells but less than 2-fold vector alone in non-endothelial (COS) cells, confirming that specificity in vivo was paralleled in vitro. The addition of 2.7 kbp of 5'-flanking region to the 0.33-kbp fragment had no effect on promoter activity or specificity. The mutation of an Sp1 motif centered at base pair -194 or an eight-base pair palindrome at -268 each reduced promoter activity by 70%. Mutation of GATA motifs at -145 and -53 reduced promoter activity by 78 and 61%, respectively. Specific binding of bovine aortic endothelial cells nuclear proteins to the Sp1 and GATA sites was demonstrated by gel shift analysis. Promoter activity in COS cells was transactivated 3-4-fold by overexpression of GATA-2. The results presented here suggest that transcription from the ICAM-2 promoter in endothelial cells is regulated by the interplay of several positive-acting factors and provide the basis for further analysis of endothelial-specific gene expression.

181. Myelin Basic Protein Is a Zinc-Binding Protein in Brain: Possible Role in Myelin Compaction

Abstract

The zinc-binding proteins (ZnBPs) in porcine brain were characterized by the radioactive zinc-blot technique. Three ZnBPs of molecular weights about 53 kDa, 42 kDa, and 21 kDa were identified. The 53 kDa and 42 kDa ZnBPs were found in all subcellular fractions while the 21 kDa ZnBP was mainly associated with particulate fractions. This 21 kDa ZnBP was identified by internal protein sequence data as the myelin basic protein. Further characterization of its electrophoretic properties and cyanogen bromide cleavage pattern with the authentic protein confirmed its identity. The zinc binding properties of myelin basic protein are metal specific, concentration dependent and pH dependent. The zinc binding property is conferred by the histidine residues since modification of these residues by diethyl-pyrocarbonate would abolish this activity. Furthermore, zinc ion was found to potentiate myelin basic protein-induced phospholipid vesicle aggregation. It is likely that zinc plays an important role in myelin compaction by interacting with myelin basic protein.

Abstract

The inhibition of ascorbate transport by rat adrenal quarters in response to steroidogenesis in vitro was shown to be highly specific with respect to tissue, substrate and steroidogenic agent. The transport system in vitro is capable of net accumulation of ascorbate. The evidence is consistent with the conclusion that ascorbate `depletion' in the adrenal gland is due to a specific block by corticoids of the uptake of ascorbate.

We have investigated the pulsation behavior of InGaAsP semiconductor lasers with a proton-bombarded segment. These lasers emit picosecond (30–70 ps) pulses at gigahertz (0.6–3.0 GHz) rates. An antireflection-coated diode in an external cavity is passively mode locked and multistable; as many as four co-existing states are observed. Interlocking hysteresis loops are observed in the pulsation frequency, pulse width, and output power as functions of the bias current. Ion-beam-assisted etching has been used to fabricate mass-transported InGaAsP/InP buried- heterostructure lasers. These lasers have a novel, deeply etched rectangular mesa that results in reduced current leakage. Both single-stripe lasers and Y-junction-coupled multiple-stripe laser arrays have been demonstrated. The single-stripe lasers have 12 mA cw threshold currents, differential quantum efficiencies of 32%–34% per facet, and smooth single-lobe far-field patterns. The MICs of 17 antimicrobial agents for 181 Streptococcus pneumoniae strains were determined by the E-test. Overall, 69.1% were penicillin resistant (MIC > 0.06 μg/ml). Resistance to ciprofloxacin (MIC > 2 μg/ml), levofloxacin (MIC > 2 μg/ml), or trovafloxacin (MIC > 1 μg/ml) was found in 12.1, 5.5, or 2.2% of the strains, respectively. Copyright © 2019 American Chemical Society. Brønsted bases have recently been reported to catalyze the glucose isomerization to fructose. This contribution describes the rational bottom-up approach to the design and synthesis of heterogeneous systems based on a C60 platform surface engineered with catalytically active amino functionalities tethered to carbon nanomaterials within variable (polar/apolar) chemical microenvironments. We demonstrate how carbocatalysts performance for the glucose-to-fructose isomerization may improve depending on amines' neighboring functional groups and their chemical interplay at the C-nanocarrier surface.
