MultiLure traps were deployed in a Hawaiian orchard to compare the attraction of economically important fruit flies and nontarget insects to the three-component BioLure and torula yeast food lures. Either water or a 20% propylene glycol solution was used to dissolve the torula yeast or as capture fluid in BioLure traps. Torula yeast in water was more attractive than BioLure for male and female Bactrocera cucurbitae (Coquillett) and Bactrocera dorsalis (Hendel) and as attractive for Ceratitis capitata (Wiedemann), and the addition of propylene glycol significantly inhibited the attractiveness of torula yeast. The known synergistic effect of propylene glycol with BioLure, resulting in increased captures of Anastrepha flies, was not observed with Bactrocera. Nontarget Drosophilidae, Neriidae, Phoridae, Calliphoridae, Sarcophagidae, and Muscidae were more strongly attracted to BioLure, and both lures collected Chloropidae equally. As with fruit flies, propylene glycol in torula yeast significantly decreased nontarget captures. The results therefore suggest that torula yeast in water is a more effective attractant than BioLure for pest Bactrocera while minimizing nontarget captures.

Studies were conducted in Hawaii to measure attraction of male melon fly, Bactrocera cucurbitae (Coquillett) (Diptera: Tephritidae), to SPLAT-Cue-Lure (C-L) and SPLAT-Melo-Lure (M-L) (raspberry ketone formate). Direct field comparisons of SPLAT-C-L and SPLAT-M-L at low (5%) and high (20%) concentrations indicated few differences in attraction over a 15-wk period. Subsequently, only SPLAT-Spinosad-C-L (5%) was compared with Min-U-Gel C-L with naled (standard used in California) in weathering studies. Treatments were weathered for 1, 2, 4, and 8 wk in Riverside, CA, and shipped to Hawaii for attraction/toxicity tests under field and semifield conditions by using released males of controlled ages, and for feeding tests in the laboratory. In terms of attraction, SPLAT-Spinosad-C-L compared favorably to, or outperformed the current standard of Min-U-Gel-C-L with naled. In terms of toxicity, the cumulative 24-h mortality did not differ between the two insecticide-containing C-L treatments in field cage studies after 8 wk. However, in feeding studies in which individual males were exposed for 5 min to the different C-L treatments after 4 wk of weathering, SPLAT-Spinosad-C-L demonstrated reduced mortality compared with the Min-U-Gel-C-L with naled, suggesting reduced persistence of the spinosad material. Spinosad has low contact toxicity and when mixed with SPLAT and C-L offers a reduced risk alternative for control of B. cucurbitae and related C-L-responding species, without many of the negative effects to humans and nontargets of broad-spectrum contact poisons such as naled.

Background: Chemosterilisation with lufenuron bait stations is a recently developed technique that is being implemented for Ceratitis capitata Wiedemann control. The aim of this work was to evaluate the chemosterilising effect of lufenuron against four economically important Latin American fruit flies species: Anastrepha ludens (Loew.), A. obliqua Macquart, A. serpentina Wiedemann and A. striata Schiner (Diptera: Tephritidae) in order to design a similar strategy for their control., Results: Sexually mature adults were treated by ingestion with concentrations ranging from 0.1 to 30.0 mg g(-1) of lufenuron in the diet. In addition, conspecific crosses with only one of the sexes being treated (30.0 mg g(-1)) were performed in order to appraise the contribution of each sex to the sterilising effect. In all cases, fecundity was not affected by the treatments, as opposed to fertility where all Anastrepha species studied were significantly affected, although to different extents. The conspecific crosses showed that treated males of A. ludens, A. obliqua and A. serpentina were not able to transmit the sterility to their respective untreated females. Only in the case of A. striata did crossing treated males with untreated females significantly reduced egg hatch., Conclusion: Although further investigations are required, the present results demonstrate that the use of lufenuron for controlling A. striata could be potentially viable.

Apple maggot, Rhagoletis pomonella (Walsh) (Diptera: Tephritidae), is a key pest in apple (Malus spp.) production areas located in the northeastern and midwestern United States and the eastern provinces of Canada. The development of Surround WP has offered a new approach for controlling apple maggot and other tephritid species, because this material is considered to be compatible with advanced integrated pest management and organic production systems. We conducted studies aimed at identifying the behavioral and biological effects of this material on apple maggots. Specifically, we examined the effect of Surround WP on the visual ecology of adult flies under field conditions, on tactile responses of flies in semifield trials, and on fly mortality in laboratory-based-bioassays. We demonstrated that an even coating of white particles over a fruit-mimicking sphere surface reduced visual attractiveness. We also found that spotty-coated fruit-mimicking spheres (meant to mimic ripe fruit bearing an uneven coating of Surround WP) were perceived by flies as not having the ideal round silhouette shape stimulus. Surround WP served as a tactile deterrent; the residence time of females introduced on to treated fruit was much shorter compared with untreated fruit. Surround WP also had a toxic effect on both adult apple maggot and Rhagoletis suavis (Loew); flies exposed to and forced to stand on Surround-treated surfaces died in <2 d in all trials. The combined effectiveness of Surround WP is based on a reduction in the attractiveness of fruit-based visual cues, an increase in the likelihood of flies leaving treated surfaces due to tactile deterrence, and a potential for increased mortality due to exposure to Surround WP particles.

Western cherry fruit fly, Rhagoletis indifferens Curran (Diptera: Tephritidae),is a major pest of sweet cherry, Prunus avium (L.) L., in the Pacific Northwest of the United States. Spinosad bait is applied weekly to kill flies before they develop eggs, but its effects on oviposition by reproductively mature flies are unknown. In this study, the main objective was to identify insecticide bait treatments that can prevent oviposition after being ingested by reproductively mature R. indifferens. First, flies were fed liquid bait. Of flies fed spinosad bait, 20% oviposited and all died within 1 d. Of flies fed acetamiprid + sucrose, 72% oviposited after 1 wk, and all recovered from paralysis within 1 d. Of flies fed spinosad bait + acetamiprid, 7% oviposited and most died within 1 d. None of the flies fed spinosad bait + thiamethoxam oviposited and all died within 1 d. Of flies fed thiamethoxam + sucrose, 2% oviposited and most died within 1 d. None of the flies fed spinosad + thiamethoxam + sucrose oviposited and all died within 1 d. Of flies fed spinosad + sucrose, 11% oviposited and almost all died within 1 d. Next, flies were fed 2-d-old dried baits. No flies fed dried spinosad + thiamethoxam + sucrose and thiamethoxam + sucrose oviposited. On the basis of absolute numbers of eggs laid by flies fed liquid and dried treatments, spinosad + thiamethoxam + sucrose may be the most effective of the seven insecticide baits tested for preventing oviposition by reproductively mature R. indifferens.

Background: Among target pests of the insecticide spinosad is the olive fruit fly, Bactrocera oleae (Rossi) (Diptera: Tephritidae). In Cyprus, spinosad has been sporadically used since its registration in 2002, whereas in Greece its use has been very limited since its registration in 2004, particularly in biological olive cultivars in Crete. By contrast, in California it has been the only insecticide used against the olive fruit fly since its registration in 2004. This study aimed at examining the resistance status of the olive fruit fly to spinosad., Results: Populations from California, Greece and Cyprus, plus a laboratory population, were tested. Bioassays were performed by oral or topical application of different concentrations of the insecticide. Cypriot populations demonstrated no resistance as compared with that of the laboratory population. Among the Greek populations, only one from Crete demonstrated a fourfold increase in resistance, whereas five populations from California demonstrated a 9-13-fold increase., Conclusion: The observed resistance increase was associated with spinosad applications in the respective areas. These values are relatively low and do not yet pose a serious control problem in the field. However, the observed variation documents that spinosad tolerance has increased in areas where the insecticide has been more extensively used.

Release-recapture studies were conducted with both feral and sterile females of the Caribbean fruit fly, Anastrepha suspensa (Loew) (Diptera: Tephritidae), to determine sampling range for a liquid protein bait (torula yeast/borax) and for a two-component synthetic lure (ammonium acetate and putrescine). Tests were done in a guava, Psidium guajava L., grove and involved releasing flies at a central point and recording the numbers captured after 7 h and 1, 2, 3, and 6 d in an array of 25 Multilure traps located 9-46 m from the release point. In all tests, highest rate of recapture occurred within the first day of release, so estimations of sampling range were based on a 24-h period. Trap distances were grouped into four categories (<10, 10-20, 20-30, and >30 m from release point) and relative trapping efficiency (percentage of capture) was determined for each distance group. Effective sampling range was defined as the maximum distance at which relative trapping efficiency was > or = 25%. This corresponded to the area in which 90% of the recaptures occured. Contour analysis was also performed to document spatial distribution of fly dispersal. In tests with sterile flies, immature females dispersed farther and were recovered in higher numbers than mature females, regardless of attractant, and recapture of both cohorts was higher with torula yeast. For mature feral flies, range of the synthetic lure was determined to be 30 m. With sterile females, effective range of both attractants was 20 m. Contour maps indicated that wind direction had a strong influence on the active space of attractants, as reflected by distribution of captured flies.

Methyl eugenol (ME) and cue-lure (C-L) traps with solid lure dispensers were deployed in areas with low and high populations of oriental fruit fly, Bactrocera dorsalis (Hendel), and melon fly, Bactrocera cucurbitae (Coquillett), respectively. In low-density areas, standard Jackson traps or Hawaii Fruit Fly Areawide Pest Management (AWPM) traps with FT Mallet ME wafers impregnated with dimethyl dichloro-vinyl phosphate (DDVP) or AWPM traps with Scentry ME cones and vapor tape performed equally as well as standard Jackson traps with liquid ME/C-L and naled. Standard Jackson traps or AWPM traps with FT Mallet C-L wafers impregnated with DDVP or AWPM traps with Scentry C-L plugs with vapor tape performed equally as well as standard Jackson traps with a lure-naled solution. In high density areas, captures with traps containing FT Mallet wafers (ME and C-L) outperformed AWPM traps with Scentry cones and plugs (ME and C-L) with DDVP insecticidal strips over a 6-mo period. Captures of B. dorsalis and B. cucurbitae with wafers containing both ME and raspberry ketone (FT Mallet MC) were equivalent to those containing separate lures. From a worker safety and convenience standpoint, FT Mallet ME and C-L wafers with DDVP or Scentry plugs, with or without DDVP vapor tape, are more convenient and safer to handle than standard liquid insecticide formulations used for monitoring and male annihilation programs in Hawaii, and for detections traps used on the U.S. mainland. Furthermore, the FT Mallet MC wafer might be used in a single trap in place of two separate traps for detection of both ME and C-L responding fruit flies.

The sterile insect technique may be implemented to control populations of the oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), when environmental concerns preclude widespread use of chemical attractants or toxicants. The goal of the present study was to evaluate whether the mating competitiveness of sterile B. dorsalis males could be increased via pre-release feeding on methyl eugenol. Males of the oriental fruit fly are strongly attracted to this plant-borne compound, which they ingest and use in the synthesis of the sex pheromone. Previous studies conducted in the laboratory and small field-cages have shown that males given methyl eugenol produce a more attractive pheromone for females and have a higher mating success rate than males denied methyl eugenol. Here, levels of egg sterility were compared following the release of wild-like flies and either methyl eugenol-fed (treated) or methyl eugenol-deprived (control) sterile males in large field enclosures at four over flooding ratios ranging from 5:1 to 60:1 (sterile: wild-like males). Treated sterile males were fed methyl eugenol for 1-4 h (depending on the over flooding ratio tested) 3 d prior to release. Eggs were dissected from introduced fruits (apples), incubated in the laboratory, and scored for hatch rate. The effect of methyl eugenol was most pronounced at lower over flooding ratios. At the 5:1 and 10:1 over flooding ratios, the level of egg sterility observed for treated, sterile males was significantly greater than that observed for control, sterile males. In addition, the incidence of egg sterility reported for treated sterile males at these lower over flooding ratios was similar to that noted for treated or control sterile males at the 30:1 or 60:1 over flooding ratios. This latter result, in particular, suggests that pre-release feeding on methyl eugenol allows for a reduction in the number of sterile flies that are produced and released, thus increasing the cost-effectiveness of the sterile insect technique.

Bait stations represent an environmentally friendly attract-and-kill approach to fruit fly population suppression. Recently a novel, visually attractive, rain-fast bait station was developed in Hawaii for potential use against multiple species of pestiferous fruit flies. Here, we compared the efficacy of GF-120 NF Naturalyte Fruit Fly Bait applied either as foliar sprays or onto bait stations in reducing female oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), population density and level of fruit infestation in commercial papaya orchards in Hawaii. Trapping and infestation data were used as indicators of the effectiveness of the two bait application methods. For the first 10 weeks of the study, captures of female B. dorsalis in monitoring traps were significantly greater in control plots than in plots treated with foliar sprays or bait stations. Six weeks after the first bait spray, incidence of infestation (i.e. number of fruit with one or more B. dorsalis larvae) of quarter to half-ripe papaya fruit was reduced by 71.4% and 63.1% for plots with bait stations and foliar sprays, respectively, as compared to control plots. Twelve weeks after first spray, incidence of infestation was reduced by only 54.5% and 45.4% for plots with bait stations and foliar sprays, respectively, as compared to control plots. About 42% less GF-120 was used in orchard plots with bait stations compared to those subject to foliar sprays. The impact of field sanitation on the outcome is also discussed. The results indicate that bait stations can provide a simple, efficient, and economical method of applying insecticidal baits to control fruit flies and a safer alternative to foliar sprays.

The Bactrocera dorsalis (Hendel) is one of the most economically damaging pests and generally is controlled by insecticides. This research was done by evaluate the impact of rhizomes of Alpinia galanga extract on population of B. dorsalis. The rhizome of A. galanga was extracted by Soxhlet's apparatus using hexane, dichloromethane, ethyl acetate and 95% ethanol as separated solvent system. Various concentrations of all crude extracts were trailed by Topical spray application with adult B. dorsalis, completely randomized designs, 5 replicates. The results showed hexane crude extract gave the highest control efficiency to adult B. dorsalis which the 24 hours-LC50 values was 4,866.06 +/- 184.52 ppm (hexane), 24,156.66 +/- 880.33 ppm (dichloromethane), 16,744.73 +/- 641.27 ppm (ethyl acetate) and 6,337.54 +/- 145.42 ppm (95% ethanol). Thus, this extract may an alternative way for control this insect pest in the future.

We determined the efficacy of three potential oviposition deterrents in reducing fruit infestation by Anastrepha obliqua in tropical plum and mango orchards. These were: (1) Extracts of feces of Mexican fruit fly, Anastrepha ludens, known to contain the A. ludens host marking pheromone (HMP) and (2) two fully synthetic simplified analogues of the naturally occurring compound, which we have named desmethyl A. ludens HMP (DM-HMP) and Anastrephamide. Two applications of feces extracts 2 or 3 wk before fruit color break reduced A. obliqua infestation in plums by 94.1, 75.9, and 72% when measured 8, 14, and 25 d, respectively, after application. The natural A. ludens-HMP containing extract retained its effectiveness despite considerable rainfall (112.5 mm) and high A. obliqua populations. The synthetic desmethyl HMP derivative (DM-HMP) also reduced infestation in plums by 53.3 and 58.7% when measured, 18 and 26 d, respectively, after application. Finally, applications of Anastrephamide resulted in fruit loss cut by half and an 80% reduction in numbers of fly larvae per fruit. Our results confirm previous findings indicating that there is interspecific cross-recognition of the HMP in two of the most pestiferous Anastrepha species and open the door for the development of a highly selective, biorational Anastrepha management scheme.

Bactrocera cucurbitae (Coquillett), also known as melon fruit fly, is one of the major insect pests of cucurbits in several parts of Asia, Africa and Pacific. In the present investigation, effect of lectins from two sources i.e. Arisaema intermedium Blume and Arisaema wallichianum Hook f. (Family-Araceae) has been studied on the development of second instar larvae of melon fruit fly. The lectins were incorporated separately in artificial diet at a concentration of 10 to 160 microg ml(-1) and fed adlibitum to the second instar larvae. Both the lectins were found to prolong the development period and significantly inhibited the pupation and emergence in a dose dependent manner. Total development period was found to be prolonged by 3.5 and 2.3 days in case of larvae fed on artificial diet containing A. intermedium (AIL) and A. wallichianum (AWL), respectively. LC50 values calculated on the basis of adult emergence came out to be 32.8 and 29 microg ml(-1) for AIL and AWL, respectively. Both the lectins tested, were found to increase the activity of esterases as larvae proceeded from 24 to 72 hr of treatment. The activity of acid phosphatase decreased significantly in larvae reared on diet containing LC50 of AIL, while in case of AWL significant decrease was observed only at 72 hr of treatment. Alkaline phosphatase activity decreased significantly on treatment with both of these lectins. These results showed that AIL and AWL have promising anti-insect potential. So, lectin gene/s from either of these species can be cloned and subsequently can be employed to develop transgenics to control melon fruit flies specifically and insect pests in general. This approach could be used as a part of Integrated pest management (IPM) strategies.

Field-based experiments were used to determine the lethal activity of insecticides on apple maggot, Rhagoletis pomonella (Walsh) (Diptera: Tephritidae), eggs and larvae in apple (Malis spp.) fruit. The organophosphates azinphosmethyl and phosmet and the neonicotinoids thiacloprid, acetamiprid, clothianidin, and thiamethoxam showed significant curative activity on the apple maggot postinfestation, when applied topically to apple fruit 24 h postharvest. Of the compounds tested, only phosmet showed significant curative activity on apple maggot when applied 14 d postharvest. The pyrethroid esfenvalerate, oxadiazine indoxacarb, anthranilic diamide chlorantraniliprole, and spinosyns spinosad and spinetoram did not show activity on apple maggot eggs and larvae in fruit. Residue profiles showed that for most compounds, the majority (>85%) of residues were found in the skin and the outside 2 mm of the apple flesh. For the neonicotinoid compounds, however, substantial portions of residues were found to penetrate in and beyond the outer flesh regions, and the total flesh residue recoveries were generally greater than those from the skin. Residues of azinphosmethyl and phosmet were detected in all three flesh regions, with the largest proportions recovered from the skin.

A signature of eclosion hormone (EH) action in insect ecdysis is elevation of cGMP in Inka cells, leading to massive release of ecdysis triggering hormone (ETH) and ecdysis initiation. Although this aspect of EH-induced signal transduction is well known, the receptor mediating this process has not been identified. Here, we describe a receptor guanylyl cyclase BdmGC-1 and its isoform BdmGC-1B in the Oriental fruit fly Bactrocera dorsalis that are activated by EH. The B form exhibits the conserved domains and putative N-glycosylation sites found in BdmGC-1, but possesses an additional 46-amino acid insertion in the extracellular domain and lacks the C-terminal tail of BdmGC-1. Combined immunolabeling and in situ hybridization reveal that BdmGC-1 is expressed in Inka cells. Heterologous expression of BdmGC-1 in HEK cells leads to robust increases in cGMP following exposure to low picomolar concentrations of EH. The B-isoform responds only to higher EH concentrations, suggesting different physiological roles of these cyclases. We propose that BdmGC-1 and BdmGC-1B are high- and low-affinity EH receptors, respectively.

At present, the most effective synthetic lures for pest Anastrepha fruit flies are multicomponent blends that include ammonia and the diamine synergist putrescine (1,4-diaminobutane). Both chemicals generally have been regarded as protein cues that result in female-biased attraction. Using electroantennography (EAG) and flight tunnel bioassays, this study evaluated response of the Caribbean fruit fly, Anastrepha suspensa (Loew) to vapors released from commercial lure formulations of ammonium bicarbonate and putrescine. Over a range of doses tested, EAG response to ammonium bicarbonate was equivalent for both sexes, but female response was significantly greater than male response to putrescine and to a 1:1 mixture of ammonium bicarbonate and putrescine. Amplitude of EAG response to the mixture was approximately equal to the summation of responses to the individual substrates. Using a fixed dose of substrate, EAG measurements from females 1-14 d old indicated that antennal sensitivity to both lures varied according to physiological state of the fly. Peak response to ammonium bicarbonate was recorded from immature females, peak response to putrescine from sexually mature females. In bioassays, more females were captured with ammonium bicarbonate plus putrescine than with ammonium bicarbonate alone. This difference was not observed in males, resulting in a higher female to male ratio in captures with ammonium bicarbonate plus putreseine (3:1) versus ammonium bicarbonate alone (1:1). Results suggest that separate olfactory receptors are involved in detection of the two semiochemicals, and that the putrescine component is primarily responsible for the female-biased attraction.

Present study was undertaken to investigate the influence of D-galactose binding lectin from Erythrina indica Lam. on the eggs and second instar larvae (64-72 hr) of melon fruit fly, Bactrocera cucurbitae (Coquillett). The lectin from E. indica seeds was extracted and purified by affinity chromatography using asilofetuin linked porous amino activated silica beads. The effects of various concentrations (0, 125, 250, 500 and 1000 microg ml(-1)) of lectin were studied on freshly laid eggs (0-8 hr) of B. cucurbitae which showed non-significant reduction in percent hatching of eggs. However, the treatment of second instar larvae (64-72 hr) with various test concentrations (0, 25, 50, 100 and 200 microg ml(-1)) of lectin significantly reduced the percent pupation and percent emergence of B. cucurbitae depicting a negative correlation with the lectin concentration. The LC50 (81 microg ml(-1)) treatment significantly decreased the pupal weight. Moreover, the treatment of larvae had also induced a significant increase in the remaining development duration. The activity of three hydrolase enzymes (esterases, acid and alkaline phosphatases), one oxidoreductase (catalase) and one group transfer enzyme (glutathione S-transferases) was assayed in second instar larvae under the influence of LC50 concentration of lectin for three exposure intervals (24, 48 and 72 hr). It significantly suppressed the activity of all the enzymes after all the three exposure intervals except for esterases which increased significantly.

A fruit fly bait to attract and kill adult fruit flies, GF-120, was tested in cages to determine effects of pretreatment diet and bait aging before use on cumulative mortality rates of Mexican fruit fly, Anastrepha ludens (Loew) (Diptera: Tephritidae). Protein-starved and protein-fed, 9-d-old flies both experienced varying overall cumulative mortality at 4, 8, 24, and 48 h. Pretreatment diet had no significant effect on mortality. Overall mortality rates were below 10% for 4 h, 39-43% at 8 h, but mortality in all treatments increased to 89-93% by 24 h, and 99% by 48 h. In a second experiment, GF-120 baits were either freshly prepared or aged for 24 h. Subtreatments consisted of protein-fed and protein-starved flies. The 24-h-aged bait killed significantly more flies at 4 and 8 h than the freshly prepared bait. Protein-starved flies had significantly higher mortality at 4 h and marginally higher mortality at 8 h than protein-fed flies. At 24 and 48 h, there were no significant differences among treatments, and overall morality rose to 99-100% by 48 h. These results may explain differences noted in previous publications in which fruit fly mortality to GF-120 was reported as unusually low as well as reports of bait ineffectiveness for protein-fed flies. The overall impact of any initial repellency of GF-120 seems negligible as judged by overall cumulative mortality at later evaluation times.

The West Indian fruit fly, Anastrepha obliqua (Macquart), is considered one of the most important pests of several fruit crops in Latin America. Studies on the sensorial ecology of A. obliqua may contribute to the improvement of a trapping system for monitoring this fruit fly species. In this study, we investigated the responses of both sexes of A. obliqua to different visual cues and to the combination of visual and chemical cues in field cages tests. Both sexes were more attracted to lime-green, orange, and yellow spheres than to red, black, or white spheres. Flies were more attracted to three-dimensional models than to flat models. A. obliqua was more attracted to 8-, 10-, and 12-cm-diameter spheres than to 4- and 6-cm-diameter spheres. Flies were more attracted to lime-green spheres baited with mombin fruit odor, Spondias mombin L., than to spheres offering either visual or cues alone.

Background: Development of attract-and-kill bait stations for pest fruit flies has been limited by the water solubility of sugar needed as a feeding stimulant and by the volatility of chemical attractants. A wax-based matrix was developed that provides the longevity needed for field use and is biodegradable., Results: Laboratory bioassays with the Caribbean fruit fly, Anastrepha suspensa (Loew), confirmed the efficacy of bait stations containing avermectin, methomyl, spinosad and phloxine B. Field cage studies demonstrated that significant mortality occurred with either 1% (w/v) spinosad or 1% (w/v) methomyl bait stations versus pesticide-free bait stations. Bait stations were exposed to environmental conditions by placing them in trees at the ARS station in Miami, Florida, between tests. There was no loss in efficacy, in spite of exposure to over 360 mm of rainfall over the 56 days of the study, indicating that the bait stations could provide population suppression for at least 1-2 months when used in subtropical environments., Conclusion: A long-lasting, female-targeted fruit fly bait station, such as the one developed herein, could provide a cost-effective option for fruit fly population suppression that would be an important tool in tephritid pest management and control. Additional studies are needed to demonstrate efficacy against wild fruit fly populations and determine deployment strategies.

Decreasing pesticide use in olive groves is central to controlling pathogens and pests such as Bactrocera oleae. This has led to the development of mass trapping devices which not only minimize pesticide use but, with improved efficacy of attractants, also decrease costs associated with pest control and ensures that the quality of olive oil is safe for human consumption. This study was undertaken to test a new device which utilizes reduced quantities of both insecticide (lambda-cyalothrin) as well as the female olive fly pheromone (1,7-dioxaspiro-(5.5)-undecane). The new device was tested against an older device manufactured by the same company. The use of plastic polymers as substrate for encapsulating the pheromone allowed for a slower pheromone release, prolonging the efficacy and duration and thus reducing costs. The density of adult populations was monitored using yellow chromotropic traps that were checked every ten days and the degree of olive infestation, as determined by preimago stages, was assessed by analyzing 100 drupes per plot. Infestation analyses were performed every ten days. The control plot had the lowest density of adults and the highest drupe infestation rate. The new devices were more effective than the older devices in both attracting adults and controlling infestation of drupes. Moreover, the new devices containing reduced amounts of pheromone and insecticide were cheaper and exhibited longer functional efficacy. In addition to the slower release of attractants, the plastic polymers used in these newer devices were also more resistant to mechanical and weather degradations. Results demonstrate that mass trapping can indeed be an effective means of controlling B. oleae via eco-sustainable olive farming.

The efficacy of GF-120 NF Naturalyte Fruit Fly Bait in combination with field sanitation was assessed as a control for female oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), in papaya (Carica papaya L.) orchards in Hawaii. Three different bait spray regimes were evaluated: every row (high use of the bait), every fifth row (moderate use), and every 10th row (low use). Orchard plots in which no bait was applied served as controls. For five of the seven biweekly periods that followed the first bait spray, trapping data revealed significantly fewer female B. dorsalis captured in plots subject to high and moderate bait use than in control plots. Differences in incidence of infestation among treatments were detected only by the third (12 wk after first spray) fruit sampling with significantly fewer infested one-fourth to one-half ripe papaya fruit in plots subject to high and moderate bait use than in control plots. Parasitism rates by Fopius arisanus (Sonan) (Hymenoptera: Braconidae) were not negatively affected by bait application. Results indicate that foliar applications of GF-120 NF Naturalyte Fruit Fly Bait either to all rows (every other tree), or to every fifth row (every tree) in combination with good sanitation can effectively reduce infestation by B. dorsalis in papaya orchards in Hawaii.

In regions with a humid summer climate, the use of water-soluble bait to control apple maggot is often limited by rainfall. We studied increasing the rainfastness of GF-120 fruit fly bait by adding paraffin wax emulsion. First, we verified that adding 10% wax to a mixture containing 16.7% GF-120 did not reduce the mortality of female apple maggot compared with GF-120 without wax. In addition, we determined that fly mortality caused by GF-120 plus wax subjected to simulated rain was similar to that caused by GF-120 without wax not subjected to rain. Other assays showed that higher fly mortality resulted from increasing the proportion of wax from 10 to 15%, and lower mortality resulted from decreasing GF-120 from 16.7 to 10 or 5%. The availability of spinosad on or near droplets of a mixture consisting of 5, 10, or 15% GF-120 and 15% wax was determined before and after the droplets were subjected to three 15-min periods of simulated rain. We found an initial steep decline in dislodgeable spinosad and smaller decreases after subsequent periods of rain. In a small-plot field trial, fruit infestation by apple maggot in plots treated with a mixture consisting of 16.7% GF-120 and 19.2% wax was less than in plots treated with 16.7% GF-120 without wax but not less than in control plots. Overall, we found that adding paraffin wax emulsion to GF-120 increased rainfastness in laboratory bioassays, and specifically that it retained the active ingredient spinosad. However, our field data suggest that optimal rainfastness requires the development of mixtures with > 19.2% wax, which may preclude application using standard spray equipment.

The Ethiopian fruit fly, Dacus ciliatus, is an oligophagous pest of cucurbit crops, particularly melons, cucumbers, and marrows (summer squash). The present study aimed to identify host attractants for D. ciliatus and was guided by a behavioral bioassay and an electrophysiological assay. We tested volatile compounds from the fruits of a host plant, ripe and unripe Galia melon, Cucumis melo var. reticulates. Both sexes were attracted to melon volatiles. Those of ripe melon were preferred. Gas chromatography-electroantennographic detection analysis of the behaviorally active ripe melon volatiles consistently showed that 14 compounds elicited similar antennal responses from both sexes. Twelve compounds were identified by gas chromatography-mass spectrometry (GC-MS) using GC-MS libraries, retention indices (RI), and authentic standards. The electrophysiological activities of the compounds that were present at sufficient levels for identification, benzyl acetate, hexanyl acetate, (Z)-3-hexenyl acetate, (Z)-3-octenyl acetate, octanyl acetate, (Z)-3-decenyl acetate, and (E)-beta-farnesene, were evaluated at six different dosage levels by using electroantennography (EAG). Benzyl and hexanyl acetates elicited dose responses only in males, while other tested compounds elicited dose responses in both sexes. The strongest responses were observed for doses between 100 ng and 10 microg. The dose response, in terms of attractiveness to synthetic compounds within the active range (as determined by EAG), also was evaluated in the behavioral bioassay. Synthetic acetates were attractive to both sexes when tested individually. Significant attraction was observed when individual compounds were applied in the bioassay arena at doses of 0.5-1 microg/dispenser. Blends of compounds in equal proportions also were attractive to the insects. The most attractive blend was a mixture of four or five identified acetates. The addition of an equal proportion of (E)-beta-farnesene to this mixture had a deterrent effect.

Effectiveness of GF-120 (Dow Chemical) Fruit Fly Bait containing the insecticide spinosad in controlling mango-infesting fruit flies (Diptera: Tephritidae) was assessed by comparing treated orchards with untreated orchards. Twelve mango, Mangifera indica L., plantations located in six villages (two similar orchards per village: one orchard treated and orchard untreated) scattered in the Borgou department (northern Benin) were monitored weekly with fly traps, and the fruit was sampled twice for larval infestation at the beginning and in the middle of May in both 2006 and 2007. The two main mango fruit fly pests are Ceratitis cosyra (Walker) and Bactrocera invadens Drew, Tsuruta & White, an invasive species that recently spread throughout West Africa. In both the 2006 and 2007 seasons, C. cosyra had the earliest peak of abundance, and the difference between treated and untreated orchards, in terms of mean number of flies trapped per week and per trap, was significant only in 2007. B. invadens populations quickly increased with the onset of the rains, from mid-May onward, with no significant difference between treated and untreated orchards. In 2006 and 2007, the larval infestation by B. invadens was significantly lower in plots treated with GF-120 than in untreated control plots. GF-120 provided an 81% reduction in the number of pupae per kilogram of fruit after weekly applications for 7 wk in 2006 and an 89% reduction after 10 wk of weekly applications in 2007. The possibility of integrating GF120 bait sprays in an integrated pest management package is discussed in relation to market requirements.

Methyl eugenol (ME) and cue-lure (C-L) traps to detect tephritid flies on the U.S. mainland were tested with and without insecticides under Hawaiian weather conditions against small populations of oriental fruit fly, Bactrocera dorsalis (Hendel) and melon fly, Bactrocera cucurbitae (Coquillett), respectively. In comparative tests, standard Jackson traps with naled and the Hawaii fruit fly areawide pest management (AWPM) trap with 2,2-dichorovinyl dimethyl phosphate (DDVP) insecticidal strips outperformed traps without an insecticide. Addition of the reduced risk insecticide spinosad did not increase trap capture significantly compared with Jackson traps without an insecticide. Captures in AWPM traps with DDVP compared favorably with those for the Jackson trap with liquid naled (the Florida standard). In subsequent tests, captures with solid Farma Tech wafer dispensers with ME or C-L and DDVP placed inside Jackson and AWPM traps were equal to those for a Jackson trap with naled, currently used for detection of ME and C-L responding fruit flies in Florida. Farma Tech ME and C-L wafers with DDVP would be more convenient and safer to handle than current liquid insecticide formulations (e.g., naled) used for detection programs in Florida.

The Caribbean fruit fly, Anastrepha suspensa (Loew), like many polyphagous tephritids, exhibits a lek polygyny mating system, and juvenile hormone levels and adult diet are known to have important positive effects on male sexual success. Among the potential components of this success are male lek tenure and female response to the sexual signals of lekking males. Male A. suspensa where submitted to four different treatments: (M(+)P(+)) application of juvenile hormone analog, methoprene (M) and sugar and hydrolyzed yeast as adult food; (M(+)P(-)) application of M and sugar as adult food; (M(-)P(+)) no application of M and sugar and hydrolyzed yeast as adult food; and (M(-)P(-)) no application of M and sugar as adult food. M(+)P(+) males initiated and participated more in aggregations, mated more frequently, and occupied the lek centers more often. They also had fewer unsuccessful mounting attempts than males in all the other treatments. M(+)P(+) males also emitted pheromones and acoustically signaled more often and attracted more females than males in other treatments. Male sexual performance was improved due to methoprene, protein supply, and the interaction of methoprene and protein for most of the parameters. Since the success of the sterile insect technique (SIT), a commonly employed technique to control pest tephritids, requires the release of males that can form leks, engage in agonistic sexual interactions, and attract females, these positive effects of protein and methoprene may substantially improve SIT programs.

Field-cage experiments were performed to determine the effectiveness of MultiLure traps (Better World MFG Inc., Fresno, CA) baited with NuLure (Miller Chemical and Fertilizer Corp., Hanover, PA) or BioLure (Suterra LLC, Inc., Bend, OR) in capturing individually marked Mexican fruit fly, Anastrepha ludens (Loew), and West Indian fruit fly, Anastrepha obliqua (Macquart) (Diptera: Tephritidae), of both sexes. Experimental treatments involved wild and laboratory-reared flies of varying ages (2-4 and 15-18 d) and dietary histories (sugar only, open fruit, open fruit plus chicken feces, and hydrolyzed protein mixed with sugar). Data were divided into two parts: total captures over a 24-h period and trap visits/landings, entrances into interior of trap ,and effective captures (i.e., drowning in liquid bait or water) over a 5-h detailed observation period (0600-1100 hours). The response to the two baits varied by fly species, gender, physiological state, age, and strain. Importantly, there were several highly significant interactions among these factors, underlining the complex nature of the response. The two baits differed in attractiveness for A. obliqua but not A. ludens. The effect of strain (wild versus laboratory flies) was significant for A. ludens but not A. obliqua. For effect of dietary history, adults of both species, irrespective of sex, were significantly less responsive to both baits when fed on a mixture of protein and sugar when compared with adults fed the other diets. Finally, we confirmed previous observations indicating that McPhail-type traps are quite inefficient. Considering the total 24-h fly tenure in the cage, and independent of bait treatment and fly type (i.e., strain, adult diet, gender and age), of a total of 2,880 A. obliqua and 2,880 A. ludens adults released into the field cages over the entire study (15 replicates), only 564 (19.6%) and 174 (6%) individuals, respectively, were effectively caught. When only considering the 5-h detailed observation period and independent of bait treatment and fly type, of a total of 785 marked flies that landed on traps (519 females and 266 males, respectively), only 10.3% (144 females and 59 males) and 20.8% (25 females and 18 males) A. obliqua and A. ludens individuals, respectively, ended up being effectively captured. We discuss the practical implications of these findings with respect to developing new baits and designing new traps and to the interpretation of capture results in the field.

Background: Chlorantraniliprole formulated as a 350 g kg(-1) WG (Altacor 35WG) for management of apple maggot Rhagoletis pomonella (Walsh), blueberry maggot R. mendax Curran and cherry fruit fly R. cingulata (Loew) (Diptera: Tephritidae) was evaluated in laboratory assays and field trials., Results: A tarsal contact toxicity bioassay showed that a surface residue of 500 mg L(-1) of chlorantraniliprole caused significantly higher mortality of male and female flies of all species compared with a control. Male apple maggot and blueberry maggot mortality was significantly higher than that for females, but there was similar mortality of male and female cherry fruit flies. An ingestion toxicity bioassay showed that 500 mg L(-1) of chlorantraniliprole in diet caused significantly higher mortality of male and female flies of all species than the control, but there were no significant differences among the sexes. Delayed egglaying by females that had ingested chlorantraniliprole was found, but there were no significant sublethal effects on either the number of eggs laid or the egg hatch. Field trials with apple maggot and cherry fruit fly showed that protection of fruit by chlorantraniliprole was comparable with that of standard broad-spectrum insecticides., Conclusions: The present data indicate that chlorantraniliprole has suppressant activity against Rhagoletis fruit flies, preventing fruit infestation primarily through direct lethal effects.

Ovarian sizes (length and width) were measured in young females of Anastrepha fraterculus (Wiedemann) (Diptera, Tephritidae) subjected or not to the inhibitor alpha-difluormethylornithine (alpha-DFMO). The most effective concentration of alpha-DMFO used was 50 mM and the ovarian measurements (length and width) of the treated females were smaller than those of females not treated with alpha-DMFO. These data may suggest some relationship between ornithine decarboxylase (ODC) and sexual maturation in A. fraterculus.

Based on the observation of the development and reproduction of experimental populations of alpharmethrin-resistant and susceptible Bactrocera dorsalis strains at (28 +/- 2) degrees C, the life tables of the populations were established, and the differences in the relative ecological fitness of the two strains was analyzed. The results showed that comparing with alpharmethrin-susceptible B. dorsalis strain, the resistant one had longer durations of development stages, average generation, and previposition, its fecundity was not stable and lower, and the hatch of eggs and the emergence of adults were depressed. The fecundity of the susceptible and resistant strains was 324.0 and 240.7, respectively. Based on the life tables, the population trend index (I) of resistant strain was 79.10, while that of susceptible strain was 116.97. The net reproduction rate (R0) and the intrinsic rate of increase (rm) of resistant strain were 1414.00 and 7.9463, while those of susceptible strain were 2184.00 and 8.3809, respectively. The relation fitness of resistant strain was only 0.6474. All of the results revealed that the resistant strain was at an obvious inferior position of reproduction and development.

Diazinon has been used extensively in the past as part of California eradication programs for tephritid fruit flies (Diptera: Tephritidae) as a soil drench, but it is being phased out for this purpose in the United States. Therefore, in this study, the toxicity of Platinum, Force, Admire, Regent, and Warrior was estimated after application to sand and soil as drenches for control of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann); melon fly, Bactrocera cucurbitae (Coquillett); and oriental fruit fly, Bactrocera dorsalis (Hendel), in Hawaii. Susceptibility of each species differed. In sand, the order of toxicity at LC50 based on the 95% confidence limit overlap approach for C. capitata from most toxic to least toxic was diazinon > Force = Warrior > Admire = Platinum > Regent. The order of toxicity for B. dorsalis was diazinon > Platinum = Warrior = Force > Regent = Admire. The order of toxicity for B. cucurbitae was Warrior = diazinon > Force = Regent = Platinum = Admire. Based on the dose ratio method, Warrior was not significantly different at LC50 than diazinon for B. cucurbitae only. All other insecticides were significantly different from diazinon at LC50. Studies in sand were followed by an evaluation of specific concentrations of Warrior and Force in soil collected from two sites on the island of Kauai. Average concentrations that caused at least 95% mortality in soil in all three fruit fly species were 121 g active ingredient (AI)/ha for Force and 363 g (AI)/ha for Warrior compared with 182 g (AI)/ha for diazinon. These results indicate that Force and Warrior could be used as soil treatments for control of tephritid fruit flies.

Background: Six sweet cherry (Prunus avium L.) cultivars were tested with GF-120 with spinosad (0.2 g L(-1) spinosad bait) or without it (blank bait) to understand leaf phytotoxicity observed in the field., Results: Spinosad bait and blank bait did not differ significantly with respect to damage observed. Leaf damage was found almost exclusively at the abaxial (lower) surfaces with the doses (0, 17, 20, 25 or 40%) and cultivars tested. The effects of the blank bait on abaxial surfaces increased from 24 to 168 h, and with dose, in terms of the proportion of droplets (0.00, 0.42, 0.52, 0.75 or 0.94) and area (0.0, 18.7, 23.5, 40.5 or 91.6 mm) burned. In addition, chlorophyll was reduced with increasing dose on abaxial surfaces (SPAD = 44.6, 36.1, 34.1, 31.0, 21.5), but not on adaxial (upper) surfaces (SPAD = 44.6, 44.2, 44.0, 44.8, 44.4). The chlorophyll level in undamaged leaves (adaxial surfaces) differed by cultivar. Cherry leaves were less damaged by a 20% bait application in June (0.26) than in July (0.46) and August (0.50). Incidental insect leaf feeding at bait locations occurred at a low rate and was highest on abaxial bait surfaces., Conclusions: Applying GF-120 to the adaxial leaf surface, or at doses of

alpha-Ionone, alpha-ionol, and their mixtures with phenolic volatiles act as potential male lures for the solanaceous fruit fly Bactrocera latifrons (Hendel). However, the attractiveness of these compounds is not as strong as that of other well-known tephritid male lures, such as methyl eugenol for Bactrocera dorsalis. Isophorone and isophorol, which have a partial skeletal structure of alpha-ionone/alpha-ionol (i.e., trimethylcyclohexene), were attractive to B. latifrons males, and their mixtures with alpha-ionol exhibited stronger activity than any of the individual compounds. We also tested 3-oxo-alpha-ionone, 3-oxo-alpha-ionol, 3-hydroxy-alpha-ionone, and 3-hydroxy-alpha-ionol, hybrid compounds between isophorone/isophorol and alpha-ionone/alpha-ionol. 3-Oxo-alpha-ionone and 3-oxo-alpha-ionol were active both as attractants and phagostimulants for males. The results suggest that the introduction of an oxygen atom at the 3-position of the alpha-ionone/alpha-ionol molecule optimizes the specific chemosensory responses in B. latifrons males.

The lectin from tubers of cobra lily, Arisaema curvatum Kunth was purified by affinity chromatography using asialofetuin-linked amino activated porous silica beads. The concentration dependent effect of lectin was studied on second instar larvae (64-72 hr) of Bactrocera cucurbitae (Coq.). The treatment not only resulted in a significant reduction in the percentage pupation and emergence of the adults from treated larvae but it also prolonged the remaining larval development period. A very low LC50 value, 39 mgl(-1) of lectin was obtained on the basis of adult emergence using probit analysis. The activity of three hydrolase enzymes (esterases, acid and alkaline phosphatases), one oxidoreductase (catalase) and one group transfer enzyme (GSTs: Glutathione S-transferases) was assayed in second instar larvae under the influence of the LC50 of lectin at increasing exposure intervals (0, 24, 48 and 72 hr). The Arisaema curvatum lectin significantly decreased the activity of all the enzymes except for esterases, where the activity increased as compared to control at all exposure intervals. The decrease in pupation and emergence as well as significant suppression in the activities of two hydrolases, one oxidoreductase and one GST enzyme in treated larvae of B. cucurbitae indicated that this lectin has anti-metabolic effect on the melon fruit fly larvae.

A current trapping system for Anastrepha fruit flies uses a two-component food-based synthetic attractant consisting of ammonium acetate and putrescine (1,4-diaminobutane). Development of more effective monitoring programs may be realized through identification of additional attractant chemicals. This study examined response of the Caribbean fruit fly, Anastrepha suspensa (Loew), to putrescine and four homologous terminal diamines, differing only in carbon chain length. Using a fixed dose of each diamine substrate, electroantennogram (EAG) responses from mature females to putrescine and cadaverine (1,5-diaminopentane) were not significantly different from each other but were significantly greater than responses to longer chain diamines. Over a range of doses tested, mean female EAG response was greater than male response to both putrescine and cadaverine. In an initial field test, capture of female flies in traps baited with ammonium acetate and either putrescine or cadaverine was higher than in traps baited with ammonium acetate and any of the other diamines. In a subsequent field test, traps baited with putrescine, cadaverine, or 1,6-diaminohexane in combination with ammonium acetate captured more female flies than traps baited with ammonium acetate alone. A significantly greater synergistic effect on female captures was observed with either putrescine or cadaverine than with 1,6-diaminohexane. Thus, of the diamines evaluated, cadaverine elicited both antennal and behavioral responses comparable to that of putrescine and will be studied further as a potential attractant for pest Anastrepha species.

Background: The most important pest of olive orchards worldwide is the olive fruit fly Bactrocera oleae (Gmelin). Its control in Greece has been based on organophosphates (OPs), but their intense use has led to the development of resistance. A test previously developed to monitor the trait may not be as robust as originally thought. The pyrethroid alpha-cypermethrin has recently been registered for bait sprays, as an alternative to OPs., Results: The susceptibility of 20 B. oleae populations to alpha-cypermethrin was examined. Variation was observed in their response, with LD(50) ranging from 0.14 to 3.28 ng insect(-1) and resistance factors from 2.3 to 54.7. Resistance mechanisms were investigated. Cytochrome P450 monoxygenase activities showed an association with resistance. Sequences in the domain IIS4-IIS6 of the B. oleae para-type sodium channel were also analysed, but no resistance-associated mutations were identified. Finally, a novel diagnostic assay able to reliably monitor the frequency of the iAChE G488S resistance mutation was developed., Conclusion: This is the first attempt to evaluate the efficacy of alpha-cypermethrin against B. oleae from Greece. Data showed that it can be used effectively, but also highlighted the importance of continuous monitoring. The IIS4-IIS6 sodium channel region is the default area in which to look for resistance mutations if target-site resistance to pyrethroids arises. The application of the novel iAChE molecular diagnostic may facilitate the introduction of pyrethroids alongside OPs currently in use.

Background: Fruit flies (Diptera: Tephritidae) represent a major biosecurity threat to the horticulture sector of New Zealand, which is entirely free of these invasive pests. A nationwide surveillance programme is conducted to ensure any incursion is detected as early as possible. A review of the lure dispensers used is reported here., Results: Lure dispenser emission trials found that the currently used lure plugs release lure more slowly under New Zealand subtropical to temperate climates than wafer dispensers. Subsequent trapping experiments at high altitude in Hawaii (as a mimic of New Zealand meteorological and expected fruit fly ecological conditions) compared Lynfield traps baited with the existing lure plug dispensers and newer wafer dispensers. Catches of wild Oriental fruit flies, Bactrocera dorsalis (Hendel), were 9.5-fold higher with methyl eugenol wafers than with the plugs. Recaptures of sterile melon flies, Bactrocera cucurbitae (Coquillet), were 2.6-fold higher with cuelure wafers than with the plugs. Recaptures of sterile Mediterranean fruit flies, Ceratitis capitata Weid., were not significantly higher with trimedlure wafers than with the plugs., Conclusions: Release rate and trapping experiments found new lure dispensers differed in release rate characteristics from existing dispensers under temperate and subtropical conditions, and indicated some potential for improvement in surveillance efficacy.

Specialized Pheromone and Lure Application Technology (SPLAT) methyl eugenol (ME) and cue-lure (C-L) "attract-and-kill" sprayable formulations containing spinosad were compared with other formulations under Hawaiian weather conditions against oriental fruit fly, Bactrocera dorsalis (Hendel), and melon fly, Bactrocera cucurbitae (Coquillett) (Diptera: Tephritidae), respectively. Field tests were conducted with three different dispensers (Min-U-Gel, Acti-Gel, and SPLAT) and two different insecticides (naled and spinosad). SPLAT ME with spinosad was equal in performance to the standard Min-U-Gel ME with naled formulation up to 12 wk. SPLAT C-L with spinosad was equal in performance to the standard Min-U-Gel C-L with naled formulation during weeks 7 to12, but not during weeks 1-6. In subsequent comparative trials, SPLAT ME + spinosad compared favorably with the current standard of Min-U-Gel ME + naled for up to 6 wk, and it was superior from weeks 7 to 12 in two separate tests conducted in a papaya (Carica papaya L.) orchard and a guava (Psidium guajava L.) orchard, respectively. In outdoor paired weathering tests (fresh versus weathered), C-L dispensers (SPLAT + spinosad, SPLAT + naled, and Min-U-Gel + naled) were effective up to 70 d. Weathered ME dispensers with SPLAT + spinosad compared favorably with SPLAT + naled and Min-U-Gel + naled, and they were equal to fresh dispensers for 21-28 d, depending on location. Our current studies indicate that SPLAT ME and SPLAT C-L sprayable attract-and-kill dispensers containing spinosad are a promising substitute for current liquid organophosphate insecticide formulations used for areawide suppression of B. dorsalis and B. cucurbitae in Hawaii.

Laboratory bioassays and field trials were conducted to evaluate an "attract-and-kill" system using methyl eugenol (ME) with neonicotinoid insecticides against male oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae). In laboratory bioassays, mortality of male flies resulting from the conventional toxicant, naled was 98.3-100% at 24 through 72 h after treatment, whereas the neonicotinoid insecticides imidacloprid and acetamiprid caused only approximately 60-80% at 24 through 72 h after treatment. In the assays of residual effect, naled was persistent up to 96 wk, whereas imidacloprid or acetamiprid was persistent up to 150 wk, resulting in 38.9 or 61.2% male mortality, respectively. Imidacloprid, in particular, caused a delayed lethal effect on flies. In another experiment, male mortality within 28 wk from clothianidin, another neonicotinoid insecticide, was approximately 80% after exposure for 24 h, suggesting a delayed lethal effect similar to those treated with imidacloprid, and mortality was up to 91.8%, if observed, 72 h after treatment. In field trials, attractiveness was similar between ME alone and ME incorporated with naled or neonicotinoids, indicating that addition of these insecticides to ME in traps is not repellent to B. dorsalis males. Using an improved wick-typed trap with longer attractiveness for simulating field application, addition of imidacloprid or acetamiprid maintained 40.1 or 64.3% male mortality, respectively, when assayed once every 2 wk from traps placed in orchards for 42 wk without changing the poison, whereas incorporation with naled resulted in as high as 98.1% after 34 wk and approximately 80% at 42 wk, indicating that persistence is increased compared with sugarcane fiberboard blocks for carrying poison attractants. This study also suggests that neonicotinoid insecticides could be used as an alternative for broad-spectrum insecticides as toxicants in fly traps.

Survival of freezing not only requires organisms to tolerate ice formation within their body, but also depends on the rapid redistribution of water and cryoprotective compounds between intra- and extracellular compartments. Aquaporins are transmembrane proteins that serve as the major pathway through which water and small uncharged solutes (e.g. glycerol) enter and leave the cell. Consequently, we examined freeze-tolerant larvae of the goldenrod gall fly, Eurosta solidaginis, to determine whether aquaporins are present and if their presence promotes freeze tolerance of specific tissues. Immunoblotting with mammalian anti-AQP2, -AQP3 and -AQP4 revealed corresponding aquaporin homologues in E. solidaginis, whose patterns of expression varied depending on acclimation temperature and desiccation treatment. To examine the role of aquaporins in freeze tolerance, we froze fat body, midgut and salivary gland tissues in the presence and absence of mercuric chloride, an aquaporin inhibitor. Survival of fat body and midgut cells was significantly reduced when mercuric chloride was present. In contrast, survival of the salivary gland did not decrease when it was frozen with mercuric chloride. Overall, this study supports our hypothesis that naturally occurring aquaporins in E. solidaginis are regulated during desiccation and promote cell survival during freezing.

This study assessed the effects of Surround (kaolin) on several behavioral parameters of female blueberry maggot, Rhagoletis mendax Curran (Diptera: Tephritidae). First fruit visited, walking, cleaning, and oviposition behavior were quantified in two-choice and no-choice assays where females encountered Surround-treated and untreated fruit of highbush blueberries (Vaccinium spp.). In two-choice assays, females had a propensity (68%) to first visit untreated blueberries. In two-choice and no-choice tests, number of walking bouts and duration of walking bouts were significantly shorter on the Surround-treated than on untreated fruit. Few oviposition attempts on fruit were observed, irrespective of treatments and assays. Chromameter measurements showed significant modification of the blueberry skin color parameters lightness, hue, and saturation between untreated fruit compared with fruit dipped once or twice in a suspension of Surround. Oviposition trials with field-treated fruit showed that blueberries treated with Surround had fewer oviposition scars than the control, and this was more pronounced with weekly applications of Surround. Uneven coating of the fruit by Surround in field applications may have resulted in higher acceptance rates by flies than in blueberries individually dipped and used in laboratory trials.

A homologue of the doublesex gene (Bddsx) has been cloned from the oriental fruit fly, Bactrocera dorsalis (Hendel). Northern analysis indicates a differential expression of Bddsx in male and female flies, as reported for other dsx genes. A structural conservation of DNA binding domain/oligomerization domain 1 and oligomerization domain 2 suggests that the doublesex protein (BdDSX) of this fruit fly serves as a transcriptional factor for downstream sex-specific gene expression. The putative transformer/transformer-2 protein binding sequence in female-specific transcript suggests that a preserved alternative splicing process found in other flies mediates the synthesis of Bddsx transcript. RNA interference (RNAi) data from adult abdominal dsRNA injection assays indicate that female-specific dsx dsRNA reduces specifically its own transcript, inhibits selectively expression of the yolk protein gene (Bdyp1), and delays ovary development. The number of matured eggs is significant reduced after RNAi treatment, but the sex ratio of offspring is not biased. Moreover, 27% of female progeny with RNAi show deformed ovipositor, but male flies are not affected. Although this is a transient treatment, the specific Bddsx(f) interference offers a promising and novel approach to oriental fruit fly control in the future.

Neem leaf dust and a commercial formulation of neem were tested on adult Bactrocera cucurbitae (Coquillett) and Bactrocera dorsalis (Hendel) to determine their effects on the longevity, fecundity and ovarian development. Different combinations of neem leaf dust and a commercial formulation of a neem compound incorporated with sugar solution and adult rearing diets were tested. The Laboratory tests showed that ingestion of neem can significantly reduced the longevity and fertility of both the fly species. Significantly fewer pupae were collected from adults fed on laboratory rearing diet and nimbicidine as water source. Effect of neem treatment on the pupation and subsequent adult emergence of late-instar larvae was negligible. Microscopic observation indicated that the decreased fecundity was due to the block of ovarian development. Experimental results confirmed that neem can be effectively used as a safe alternative insecticide for the control of Bactrocera species.

Several attempts for evaluating environmental impact of active agents allowed in organic olive farming against Bactrocera oleae have been made, but very few studies were performed contemporaneously on more than one of them. Insects were chosen as indicators because they are known to react very quickly to environmental perturbations, mainly at the community level. In fact, the coenosis is the functional unit interacting with biotic and abiotic environmental parameters. Seven taxa, known for their sensitivity to habitat alterations, were sampled and grouped in functional groups: predators and parasitoids, phytophagouses and pollinators. The coenotic balance between these two functional groups was analyzed. The study was carried out in an organic olive orchard in the municipality of Terranova da Sibari, Cosenza, Southern Italy. The tested active agents (Azadirachtin, Rotenone, Copper Oxychloride) were sprayed twice (end of September and middle October). During the treatments the population dynamics of all the taxa were knocked-down. No one tested compound seems to be harmless to the entomocoenosis, particularly on phytophagouses and pollinators. In truly organic farming it is necessary to provide natural refuge areas to beneficial insects (i.e. pest antagonists) in which no active agents are sprayed and alternative preys could be found.

Field-trapping evaluations of the new male attractant, formic acid 4-(3-oxobutyl) phenyl ester (raspberry ketone formate [RKF]) were conducted in Hawaii with wild populations of melon flies, Bactrocera cucurbitae Coquillett (Diptera: Tephritidae), to determine its activity in the field and to evaluate new plastic matrix formulations. All tests were compared with the standard melon fly attractant 4-(4-acetoxyphenyl) -2-butanone (cuelure [CL]), which is the attractant of choice for detection programs aimed at melon fly and other cuelure-responding Bactrocera fruit flies. Results of these tests over a range of doses on cotton wicks showed that at a 1-g dose raspberry ketone formate was 1.5-2 times more attractive compared with cuelure for up to 11 wk in the field. Lower doses applied on cotton wicks were less active, presumably due to hydrolysis of RKF to raspberry ketone. Raspberry ketone formate embedded in a plastic plug formulation also was field tested, and it was shown to be more attractive to male melon fly compared with cuelure. The use of this new attractant in control and detection programs is discussed.

Feral Mexican fruit flies, Anastrepha ludens (Loew) (Diptera: Tephritidae), were trapped in a citrus orchard in Mexico by using two types of synthetic food-odor lures, the AFF lure (Anastrepha fruit fly lure, APTIV, Inc., Portland, OR) and the BioLure (two-component MFF lure, Suterra LLC, Inc., Bend, OR). In Multilure traps (Better World Manufacturing, Inc., Miami, FL) containing water, BioLures captured about the same numbers of flies as AFF lures. In Multilure traps containing antifreeze solution, BioLures captured 2 and 5 times more flies than AFF lures in two experiments. BioLures, and AFF lures did not differ in attractiveness when used on sticky traps (Intercept trap, APTIV, Inc.; and sticky cylinder trap). Multilure traps captured >4 times as many flies as sticky traps with the exception that captures of females did not differ between Multilure and sticky traps baited with AFF lures. The percentage of females captured in Multilure traps was greater when traps were baited with BioLures compared with AFF lures, but the reverse was true for sticky traps. Sticky cylinder traps captured a higher percentage of females than Multilure traps. The most effective trap/lure combination was the Multilure trap baited with BioLure and antifreeze. In comparison with tests of these two lures in Texas, results were similar for Multilure traps, but they differed for sticky cylinder traps in that AFF lures were consistently more attractive than BioLures in Texas, but not in Mexico.

Attraction of oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), and nontarget insects to preservative fluids ethylene glycol antifreeze, propylene glycol antifreeze, or mineral oil in bucket traps that contained captured decaying male oriental fruit flies, a male lure (methyl eugenol), and a toxicant (DDVP vapor insecticidal strip) were compared with dry control traps. Significantly (P < 0.05) greater numbers of B. dorsalis were captured in propylene glycol antifreeze traps than in other attractant trap types. Among attractant trap types with lowest negative impacts on nontarget insects, control traps captured significantly lower numbers of three species and one morphospecies of scavenger flies, one species of plant-feeding fly, and one species each of sweet-and lipid-feeding ants. Mineral oil traps captured significantly lower numbers of two species of scavengers flies and one morphospecies of plant-feeding fly, and one species of sweet-feeding ant. Because of the fragile nature of endemic Hawaiian insect fauna, the propylene glycol antifreeze bucket trap is best suited for use in environments (e.g., non-native habitats) where endemic species are known to be absent and mineral oil traps are more suited for minimizing insect captures in environmentally sensitive habitats.

After pharmacophagy of methyl eugenol (ME), males of Bactrocera carambolae (Diptera: Tephritidae) produced (E)-coniferyl alcohol (CF) along with its endogenously synthesized pheromonal compounds. CF was shown to be released into the air by the ME-fed males only during the courtship period at dusk and attracted significantly more males and females than the ME-deprived males in wind tunnel assays. However, earlier onset of sexual attraction and a higher mating success were observed only in the wind tunnel and field cage assays on the third day posttreatment of ME. Field cage observations on the male-to-male interaction indicated that the ME-deprived males did not exhibit aggregation behavior, but that ME feeding promoted aggregation behavior in B. carambolae. Field cage observations revealed that the ME-deprived males were not only attracted to the ME-fed males, but also appeared to feed on their anal secretions. The secretions were subsequently confirmed to contain CF along with endogenously produced pheromonal compounds. Results obtained for B. carambolae were compared to those previously obtained from its sibling species, Bactrocera dorsalis, and are discussed in light of species advancement in fruit fly-plant relationships.

Wheat germ oil was added to a larval liquid diet for rearing Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) to optimize fruit fly quality. Effects of various concentrations of wheat germ oil at 0.04, 0.07, 0.15, 0.30, and 0.66% and their possible mode of action were evaluated. Results suggest that addition of wheat germ oil does not affect pupal weight, larval developmental period, adult emergence, mating ability, or peak time for egg production. But there was a significant increase in pupal recovery, percentage of adult fliers, egg production, or egg hatch for larvae fed the diet with wheat germ oil compared with those reared on the liquid diet without wheat germ oil. The increase in egg hatch and fliers was dose dependent. Therefore, addition of wheat germ oil to fruit fly rearing diet is a novel way to improve fruit fly quality, especially in egg hatch, fliers, egg production, and pupal recovery.
