151. 3-Bromopyruvate potentiates TRAIL-induced apoptosis in human colon cancer cells through a reactive oxygen species- and caspase-dependent mitochondrial pathway

Abstract

Introduction Colorectal cancer (CRC) is the third most common cancer and the fourth leading cause of cancer-related deaths throughout the world (Favoriti et al. 2016). CRC is a disease of [...], Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL) is a promising anticancer cytokine with minimal toxicity towards normal cells. Nevertheless, most primary cancers are often intrinsically TRAIL-resistant or can acquire resistance after TRAIL therapy. This study aimed to investigate the inhibitory effect of co-treatment of 3-bromopyruvate (3-BP) as a potent anticancer agent with TRAIL on colon cancer cells (HT-29). The results of present study indicated that combined treatment with 3-BP and TRAIL inhibited the proliferation of HT-29 cells to a greater extent (88.4%) compared with 3-BP (54%) or TRAIL (11%) treatment alone. In contrast, the combination of 3-BP and TRAIL had no significant inhibitory effect on the proliferation of normal cells (HEK-293) (8.4%). At a cellular mechanistic level, the present study showed that 3-BP sensitized human colon cancer cells to TRAIL-induced apoptosis via reactive oxygen species generation, upregulation of Bax, downregulation of Bcl-2 and survivin, release of cytochrome c into the cytosol, and activation of caspase-3. In normal cells, 3-BP, TRAIL, or combination of both had no significant effect on the reactive oxygen species levels, release of cytochrome c, and caspase-3 activity. Therefore, the combination of 3-BP and TRAIL can be a promising therapeutic strategy for treatment of colon cancer. Key words: 3-BP, TRAIL, colorectal cancer, TRAIL resistance, apoptosis. Le ligand inducteur de l'apoptose apparente au facteur de necrose tumorale (ou TRAIL pour << tumor necrosis factor-related apoptosis-inducing ligand >>) constitue une cytokine anticancereuse prometteuse dotee d'une toxicite minime envers les cellules normales. Neanmoins, la plupart des cancers primitifs sont souvent intrinsequement resistants au TRAIL ou peuvent acquerir une resistance apres un traitement par celui-ci. Ces travaux visaient a etudier l'effet inhibiteur de l'administration concomitante de 3-bromopyruvate (3-BP), en tant que puissant anticancereux, avec le TRAIL contre les cellules du cancer du colon (HT-29). Les resultats de la presente etude ont montre que l'administration de l'association du 3-BP avec TRAIL permettait d'inhiber la proliferation de cellules HT-29 dans une plus grande mesure (88,4%) que le 3-BP ou TRAIL administres seuls (54 et 11%, respectivement). En revanche, l'association du 3-BP avec TRAIL n'a eu aucun effet inhibiteur notable sur la proliferation de cellules normales (HEK-293) (8,4%). Sur le plan des modes d'action cellulaires, la presente etude a montre que le 3-BP entrainait une sensibilisation des cellules du cancer du colon a l'apoptose provoquee par TRAIL par l'intermediaire de la production de derives reactifs de l'oxygene, de la regulation a la hausse de Bax, de la regulation a la baisse de Bcl-2 et de la survivine, de la liberation du cytochrome c dans le cytosol ainsi que de l'activation de la caspase 3. Dans les cellules normales, le 3-BP, TRAIL ou l'association des deux n'ont eu aucun effet notable sur les taux de derives reactifs de l'oxygene, la liberation de cytochrome c ni l'activite de la caspase 3. Par consequent, l'association de 3-BP avec TRAIL pourrait constituer une strategie therapeutique prometteuse pour le traitement du cancer du colon. Abstract

Kallistatin (KL) is a member of the serine proteinase inhibitor (serpin) family regulating oxidative stress, vascular relaxation, inflammation, angiogenesis, cell proliferation, and invasion. The heparin-binding site of Kallistatin has an important role in the interaction with LRP6 leading to the blockade of the Wnt signaling pathway. In this study, we aimed to explore the structural basis of the Kallistatin-LRP6E1E4 complex using in silico approaches and evaluating the anti-proliferative, apoptotic, and cell cycle arrest activities of Kallistatin in colon cancer lines. The molecular docking showed Kallistatin could bind to the LRP6E3E4 much stronger than LRP6E1E2. The Kallistatin-LRP6E1E2 and Kallistatin-LRP6E3E4 complexes were stable during Molecular Dynamics (MD) simulation. The Molecular Mechanics/Poisson-Boltzmann Surface Area (MM/PBSA) showed that the Kallistatin-LRP6E3E4 has a higher binding affinity compared to Kallistatin-LRP6E1E2. Kallistatin induced higher cytotoxicity and apoptosis in HCT116 compared to the SW480 cell line. This protein-induced cell-cycle arrest in both cell lines at the G1 phase. The B-catenin, cyclin D1, and c-Myc expression levels were decreased in response to treatment with Kallistatin in both cell lines while the LRP6 expression level was decreased in the HCT116 cell line. Kallistatin has a greater effect on the HCT116 cell line compared to the SW480 cell line. Kallistatin can be used as a cytotoxic and apoptotic-inducing agent in colorectal cancer cell lines. Abstract

Occult hepatitis B (OBI) is a major challenging clinical entity characterized by the absence of hepatitis B surface antigen (HBsAg). The persistence of OBI may progress to fibrosis, cirrhosis, and hepatocellular carcinoma. This study was aimed to investigate the prevalence of OBI among HD patients. In the present cross-sectional study, 89 sera samples of hemodialysis individuals were tested for HBsAg and HBc-IgG by Enzyme-linked Immunosorbent Assay (ELISA). In addition, the HBV DNA has tested in sera and peripheral blood mononuclear cell (PBMC) samples by nested-PCR. Out of 89 patients, 51(57.3%) were males, and 38 (42.7%) females. The ages ranged from 24 to 90 years (with a mean of 57.5±1.37 years). All the sera samples had normal levels of Aspartate Aminotransferase (AST) and Alanine Transaminase (ALT) but had high levels of Creatinine (Cr) (6.9±2.17) and Blood Urea Nitrogen (BUN) (61.83±2.03). 2/89 (2.2%) sera samples were positive for both HBsAg and HBc-IgG test; in addition, HBV DNA was detected in both sera and their PBMC samples. The sera of 15/89 (16.85%) were only positive for the HBc-IgG test, including 10/51 (19.6%) males and 5/38 (13.2%) females (P=0.5). The high 16.85% prevalence OBI has been found among HD patients. To manage OBI infection, screening of HBV DNA should be implemented for HD patients by sensitive molecular means such as nested-PCR and real-time PCR.

Abstract

Background: We detected eight trematodes in the small intestine of a road-killed jackal (Canis aureus) from Hamidiyeh District near the city of Ahvaz, Khuzestan Province in 2010. Methods: Three worms were stained with carmine acid, mounted in Canada balsam on glass slides and examined under a light microscope at 1000X magnification. PCR and sequencing of a partial ITS2 sequence were used to approve the diagnosis. Results: The flukes measured ≈1 mm in length with an elongated ovoid shape resembling the members of heterophyid, and only one testis, characteristics of the genus Haplorchis. Sequencing of a 481-bp fragment of the ITS2 locus from the worms revealed 97%-98% identity with the similar sequences of the H. taichui flukes previously identified in the fish, cat, and humans from Thailand, China, and Vietnam. Conclusion: Further studies with the application of reliable molecular tools to diagnose trematode infections in wildlife and humans can bring more insight into the epidemiology of fish-borne flukes including H. taichui in this area.

Abstract

The increasing incidences of cancer at the global scale have recently resulted in the invention of various biotechnology approaches among which the oncolytic virotherapy is a new strategy for the treatment of multiple tumors. Herpes simplex virus (HSV) based vectors are one of the most studied oncolytic agents, worldwide. Moreover, syngeneic animal models are the principal parts of the oncolytic virotherapies investigation. The effects of a dual fluorescent γ34.5 deleted vector-HSV-GR- on three mouse tumor cell lines were studied in this work. We previously generated a dual fluorescent labeled oncolytic HSV-HSV-GR- (both copies of γ34.5 were inactivated by insertion of two distinct fluorescent dyes, GFP and mCherry) in our laboratory; subsequently, they were used as oncolytic viruses. The three 4T1, TC-1, and CT26 cell lines were infected with HSV-GR. The infection efficacy and the elimination potency of HSV-GR were analyzed by photomicrography and flow cytometry methods. HSV-GR showed a significant efficiency to infect the cell lines examined. Flow cytometry analyses demonstrated that HSV-GR infected 89.3%, 86.1%, and 92.4% of 4T1, TC-1, and CT26 cells, respectively. Moreover, propidium iodide (PI) staining of infected cells indicated that HSV-GR could kill 27.9%, 21.2%, and 21.3% of 4T1, TC-1, and CT26 cells, respectively. Interestingly, HSV-GR infected cells were capable of expressing both GFP and mCherry at the same time. The promising effects of the oncolytic virus HSV-GR in the mouse syngeneic tumor cell system have shed more light on the therapeutic potential of this anti-cancer approach.

167. Prevalence of hepatitis C virus among HIV-infected patients

Abstract

Background and Objectives: Hepatitis C virus and Human Immunodeficiency Virus (HIV) share the same rate of transmission. HIV/HCV co-infected individuals may result in faster progression of liver fibrosis and highly increase the risk of cirrhosis, hepatocellular carcinoma development. Thus this study was conducted to determine co-infection of HCV genotypes in positive HIV patients in Ahvaz city, Iran. Materials and Methods: The sera samples were collected from confirmed 78 infected HIV, 67 (85.89%) males and 11 (14.1%) females. All sera samples were tested for HCV Ab using ELISA test. The HCV Ab positive samples were tested for detection of 5' untranslated (UTR) and core regions of HCV genome using nested RT-PCR. The PCR products of 5UTR and core regions were sequenced to determine HCV genotypes. Results: Among the 78 infected HIV, 25 (32.05%) cases including 20 (25.64%) males and 5 (6.41%) females were positive for HCV Ab (p=0.316). 53 (67.94%) of HIV patients were negative for HCV Ab. Among 25 positive HCV Ab, 19 (24.35%) cases including 15 (19.23%) males and 4 (5.12%) females were positive for HCV RNA (p=0.447). The PCR products of 5 positive samples were randomly sequenced. The results of sequences and alignments showed that the detected HCV genotypes were three 3a and two 1a. The occurrence of genotype HCV 1a was found in one male injecting drug user Injecting Drug User (IDU) and one female. The HCV 3a genotype was detected in the three males IDU. Conclusion: The results of this survey indicated that 32.05% of HIV patients were positive for HCV Ab, among them 24.35% were positive HCV RNA. HCV genotype 3a was dominant and detected in the three males IDU. Regarding the consequences of HIV/HCV co-infection, it is suggested that HCV RNA detection should be regularly checked in individuals infected with HIV.

Abstract

The coat color of dromedary is usually uniform and varies from black to white, although dark- to light-brown colors are the most common phenotypes. This project was designed to gain knowledge on novel color-related variants using genotyping-by-sequencing (GBS). The association between the SNPs and coat color was tested using MLM (mixed linear models) with kinship matrix. Three GWAS models including white color vs. non-white color, black vs. non-black color, and light-brown vs. dark-brown color were performed. There were no distinct genetic clusters detected based on the color phenotypes. However, admixture occurred among all individuals of the four different coat color groups. We identified nine significant SNPs associated with white color after Bonferroni correction, located close to ANKRD26, GNB1, TSPYL4, TEKT5, DEXI, CIITA, TVP23B, CLEC16A, TMPRSS13, FXYD6, MPZL3, ANKRD26, HFM1, CDC7, TGFBR3, and HACE1 genes in neighboring flanking regions. The 13 significant SNPs associated with black color and the candidate genes were: CAPN7, CHRM4, CIITA, CLEC16A, COL4A4, COL6A6, CREB3L1, DEXI, DGKZ, DGKZ, EAF1, HDLBP, INPP5F, MCMBP, MDK, SEC23IP, SNAI1, TBX15, TEKT5, TMEM189, trpS, TSPYL4, TVP23B, and UBE2V1. The SNAI1 gene interacted with MCIR, ASIP and KIT genes. These genes play a key role in the melanin biosynthetic and pigmentation biological process and melanogenesis biological pathway. Further research using a larger sample size and pedigree data will allow confirmation of associated SNPs and the identified candidate genes.

Abstract

Background and Objective: Prostate cancer is considered as one of the most common malignancies in males. Despite the decreasing trend during the past decade, the incidence continues to rise in adolescents worldwide. Recently, new emerging technologies beside TRUS-guided prostate biopsy were introduced and showed high potential in the diagnosis of prostate cancer. The current study aimed at investigating the role of blood cell count and its prognostic value on pathologic findings.Methods:The current prospective analysis was conducted on patients with abnormality from January 2013 to January 2015. A total of 168 CBC tests including absolute Neutrophil and lymphocyte counts were performed on males at the Department of Urology, Razi University Hospital (Guilan, Iran). Of these, all males had both CBCs and free:total (F/T) prostate-specific antigen (PSA) ratio.Results: Average age of patients was 63.5±7. It was observed that Neutrophil count and Neutrophil percentage was not significantly different among the groups with different pathologies. In comparison to biopsy findings none of the cell count had suitable specificity and sensitivity. In the multivariate analysis to predict malignancy, only age (B=0.1, S.E=0.04, EXP [B]) =1.1 and sig=0.00) and PSA level (B=0.1, S.E=0.04, EXP [B]) =1.1 and sig=0.00) were significant.Conclusions: In comparison to biopsy findings, none of the cell counts (absolute or percent) had suitable specificity and sensitivity, and the cell count and percentage before biopsy were not different among the groups with different pathologies. Powerful and long-term studies are required to evaluate prognostic values of cell count on adenocarcinoma.

Abstract

Viruses are one of the most important concerns for human health, and overcoming viral infections is a worldwide challenge. However, researchers have been trying to manipulate viral genomes to overcome various disorders, including cancer, for vaccine development purposes. CRISPR (clustered regularly interspaced short palindromic repeats) is becoming one of the most functional and widely used tools for RNA and DNA manipulation in multiple organisms. This approach has provided an unprecedented opportunity for creating simple, inexpensive, specific, targeted, accurate, and practical manipulations of viruses, such as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), human immunodeficiency virus-1 (HIV-1), and vaccinia virus. Furthermore, this method can be used to make an effective and precise diagnosis of viral infections. Nevertheless, a valid and scientifically designed CRISPR system is critical to make more effective and accurate changes in viruses. In this review, we have focused on the best and the most effective ways to design sgRNA, gene knock-in(s), and gene knock-out(s) for virus-targeted manipulation. Furthermore, we have emphasized the application of CRISPR technology in virus diagnosis and in finding significant genes involved in virus-host interactions.

Abstract

Objective: This study aims to assess the effectiveness of web-based cognitive-behavioral therapy (CBT) in enhancing pain self-efficacy among patients diagnosed with rheumatoid arthritis (RA), a chronic condition characterized by significant pain and functional limitations. Methods and Materials: Employing a quasi-experimental design, the study recruited adult RA patients aged 30-60 years from specialized clinics in Mashhad, Iran. Participants were randomly assigned to either the intervention group, which received web-based CBT sessions focused on managing pain and improving self-efficacy, or the control group, which received no such intervention. The intervention consisted of eight sessions designed to educate patients on pain management, cognitive restructuring, and coping strategies. Data on pain self-efficacy were collected at three points: pre-intervention, immediately post-intervention, and at a follow-up session using the Pain Self-Efficacy Questionnaire (PSEQ). Data were analyzed with analysis of variance with repeated measurements and post-hoc tests using SPSS-22. Findings: Statistical analysis revealed significant improvements in pain self-efficacy scores in the intervention group compared to the control group at post-test and follow-up stages (p<0.001). Conclusion: Web-based cognitive-behavioral therapy significantly improves pain self-efficacy in patients with rheumatoid arthritis, providing a feasible and effective approach to managing chronic pain and enhancing patients' beliefs in their ability to control pain. Abstract

Absorption of toluene vapors from atmospheric air through surface absorption of adsorbents based on gas phase extraction was obtained. In experimental study, through pilot simulator, toluene vapors absorption and absorbents efficiency in the dynamic and static system was examined by nano graphene and graphene oxide (NG and GO).The effect of different parameters on toluene absorption by NG and GO such as: absorption capacity, temperature desorption, contact time and repeatability, the amount of sorbents and recovery adsorption were investigated. According to discovered findings, capacity and efficiency of absorption by absorbents depend to kind of absorbent, mass of absorbent, toluene density, temperature, air flow rate and contact time. The toluene concentration in air was determined by gas chromatography spectrometer with flame ionization detector (GC-FID). The mean of absorption recovery (AR) and the absorption capacity (AC) for nano graphene absorbent was higher than graphene oxide, respectively (ARNG: 97.3% > ARGO: 23.5%; ACNG: 285.2 mg/g > ACNGO: 67.3 mg/g)

189. Ecosystem Sustainability Assessment Using IUCN (Case Study: Dorook Basin)

Abstract

Sustainable Sustainability assessment using IUCN is a technical tool to understand and measure sustainable development. This technique allows comparing the existing conditions of the region with its ideal state, namely, full human development and a healthy and productive ecosystem. This approach simultaneously examines ecosystem sustainability and human sustainability. In this study, Dorook basin sustainability was assessed using IUCN method by 7 criteria and 17 indicators in the human section and 8 criteria and 18 indicators in the ecosystem section. In this study, Dorook basin was divided into two sub-basin called Aghcheshmeh and Ghaleghafeh. The measurements were done in each sub-basin separately. The results of analysis indicated that relation to sustainable human criterion and indicators, land use changes is the lowest score criterion. The criterion is moderate in education, skill and poverty in both sub-basins. By increasing youth employment, it can reduce dependency and improve the standard of poverty. The food security and nutrition standards are acceptable in both sub-basin. The criterion of justice and equality is poor and acceptable conditions in sub-basins of Aghcheshmeh and Ghaleghafeh, respectively. The final score of the Ghaleghafeh sub-basin is equal to 44 and the final score of the Aghcheshmeh sub-basin is 61. Generally the scores of both sub-basins indicate that sub-basins are in the medium range in terms of sustainability. The final score of the Dorook basin is 52, which is within the medium range. Investigating the indices showed that the area in terms of land use and erosion in comparison with other indicators is in a very bad situation and this index has the most negative effect in basin. Extended Abstract 1-Introduction According to FAO studies, about twenty million hectares of forest have been planted from 1995 to 1980 in developed countries, while two hundred million hectares of forest has been reduced in developing countries during this period. In contrast to other approaches, the IUCN method of Sustainability Assessment gives equal treatment to people and the ecosystem. It involves stakeholders in determining for themselves what sustainability means in their context. It uses, as its framework, a hierarchy of elements and objectives to translate the concept of sustainable development into concrete targets and measurable indicators. Thus making clear to all those involved what features everyone agrees contribute to measuring sustainability. The elements are designed to be tailored to local conditions and needs. The IUCN Sustainability Assessment Method is a user-focused process and thus can be used at international, regional, national, district or local scales. The IUCN Sustainability Assessment Method, in its full version, describes a process with seven stages. It is presented as a cycle because the assessment process is designed to allow updates that can show changes over time. According to the IUCN method widely used in different countries, in this study a set of measurements was determined according to the IUCN sustainability assessment method and organized based on issues and concerns related to forests. 2-Materials and Methods Dorook Catchment is the political section of Golestan province, Minoodasht city, and central section. There are 13 Villages in the basin. This catchment is the most important in Golestan province because of deforestation and loss of forest. A full Sustainability Assessment follows all seven stages in the assessment cycle. The general purpose of such assessment is to engage in reflection about sustainability, collect data for indicators, combine indicators and undertake analysis based on the indicators and use the data and process to assess priorities for action. Many indicator models use the Pressure-State-(Impact)-Response model to identify and organize indicators. Pressure three key types of indicators one covering the element's state, one showing pressures and one showing the way society is responding to the problem. Sustainability assessment primarily uses 'state' indicators, because in most cases, they are the most reliable measure of an element. Pressure indicators are useful in instances where measuring the pressure that one area is having on another is preferable – particularly cross border pollution, long-range transport of air or water pollutants and impacts on the global atmosphere. The use of response indicators is not encouraged in sustainability assessments, as it is often hard to link the state of an element with the responses society is undertaking to alleviate the problem. Three main components of ecosystems have been used including forests; water environment and climate were a total of 18 indicators and eight criteria. 7 criteria and 17 indicators were measured in human wellbeing. 3-Results and Discussion The results of analysis indicated that the relation to sustainable human criterion and indicators, land use changes is the lowest score criterion. The scores of Ghaleghafi and Aghcheshme are 30 and 0 respectively, indicating that the rate of land degradation is high. Changing of forest to agricultural land is the most important role in reducing stability in Dorook watershed. Education and skills criterion is medium in two sub-basin. Poverty criterion is medium in both sub-basin. In order to improve the conditions situation in the farms provide area youth by increasing employment rates, reducing dependency ratio. Food supply and food safety is in acceptable condition in both sub-basin. Justice and equality Criteria is in poor condition and acceptable in Aqcheshme and Ghaleghafi respectively. Score of soil conservation criteria in Aghcheshme sub-basin is 0 and in Ghaleghafi sub-basin is 28 that seems reasonable according to land use changes made in the following basins. Water status map shows that both sub-basin in good condition and is acceptable and In general, water ecosystem are in a much better situation than forest and human ecosystems. Weather and climate criteria in Aghcheshme sub-basins is in acceptable condition and in Ghaleghafi sub-basins is in the middle. 4-Conclusion Sustainable development indicators are expressing such interaction model between society and the environment. In general, Dorook basin ecosystems in Ghaleghafi sub-basins socio-economic performance and water criteria is in stable condition and in Aghcheshme sub-basins conservation and soil conservation are in unstable condition. The entire watershed is in the middle range and change conditions to improve sustainability need to promote appropriate policies.

According to a recent survey, it is known this RNAs have vital role in regulation cellular pathways at transcriptional, posttranscriptional and epigenetic levels. These noncoding genes are often aberrantly expressed in a variety of human cancers. However, the biological functions of most ncRNAs remain largely in doubt. In this review, we proved that a remarkable part of the genetic etiology of cancer is imposed by noncoding regulatory sequences. The purpose of this review is aimed to give an outlook of using of noncoding RNA as diagnostic markers and therapeutic targets. These observations emphasized that the recognition of coding genes and Research continued evolution and function of non-coding RNAs for a comprehensive understanding human complex diseases like cancer are essential.

Abstract

BACKGROUND: Recent evidence has shown a positive correlation between obesity and viral infections with a particular emphasis on the human adenovirus-36 (Ad-36). Ad-36 is the first human virus that may increase adiposity in animals, and it is considered as a possible risk factor for obesity in humans; however, the results were not consistent across all the studies. The present study was conducted to examine the influence of Ad-36 infection on obesity in a rat model. METHODS: Eight-week-old male Wistar rats weighing 170-240 gram (g), were randomly divided into two groups, infection group (48 rats) and a control group (12 rats). The rats in the infection group were infected with human Ad-36. All rats were given free access to a normal chow diet and water. They were weighed weekly. RESULTS: The mean ± standard deviation (SD) body weights were 229.0 ± 25.9 g and 232.3 ± 16.6 g in the infection and control groups, respectively at the time of infection. The mean ± SD body weight of the infection group (304.0 ± 39.0 g) was higher than the control group (301.0 ± 36.5 g) at 12 weeks post-infection (P = 0.82). Although two groups had approximately same food intakes, the mean change in body weight was greater in the infection group than the control group (75.8 ± 27.9 g vs. 70.8 ± 24.5 g) but it was not significant (P = 0.57). CONCLUSION:We did not find a statistically significant association between weight gain and Ad-36 infection in the rat model. It seems that longer follow-up duration is needed to develop a significant weight gain in the infected rats. Rats can be used as a good animal model for further investigations about Ad-36-induced obesity, provided not to rely merely on weight measurements. Evaluating body composition or histopathological assessments are suggested.

Abstract

This equivalence, randomized, clinical trial aimed to compare the postoperative pain of root canal therapy (RCT) with pulpotomy with mineral trioxide aggregate (PMTA) or calcium-enriched mixture (PCEM) in permanent mature teeth. In seven academic centers, 550 cariously exposed pulps were included and randomly allocated into PMTA (n = 188), PCEM (n = 194), or RCT (n = 168) arms. Preoperative “Pain Intensity” (PI) on Numerical Rating Scale and postoperative PIs until day 7 were recorded. Patients’ demographic and pre-/intra-/postoperative factors/conditions were recorded/analysed. The arms were homogeneous in terms of demographics. The mean preoperative PIs were similar (P=0.998), the mean sum PIs recorded during 10 postoperative intervals were comparable (P=0.939), and the trend/changes in pain relief were parallel (P=0.821) in all study arms. The incidences of preoperative moderate-severe pain in RCT, PMTA, and PCEM arms were 56.5%, 55.7%, and 56.7%, which after 24 hours considerably decreased to 13.1%, 10.6%, and 12.9%, respectively (P=0.578). 197. In Silico Characterization of Epitopes from Human Rotavirus VP7 Genotype G9 Design for Vaccine Development

Abstract

Acute gastroenteritis caused by Rotavirus remains the leading cause of child mortality worldwide. Rotavirus genotype G9 circulates in humans throughout the world. Antibodies against the outer glycoproteins VP7 and VP4 Rotavirus capsid are the main neutralization antibodies in the vaccine assessment. This study aimed to select an epitope to evoke T and B cells' response, as a favorable candidate for vaccine development using in Silico evaluation. In the present study, Rotavirus genotypes were determined in 100 stools specimens collected from children with acute diarrhea. The results showed predominant G genotype, G9 (38.5%) followed by G2 (22.9%), G1 (16.5%), G12 (11.4%), G4 (6.4%), and G3 (4.3%). The G9 was dominant in this study and other regions of Iran; thus, this study was conducted to select an epitope from Rotavirus genotype G9 as a promising epitope candidate for future vaccine development. For this reason, several works including a complete sequence of VP7 G9, phylogenetic analysis, Prediction of Protein Structure, Physicochemical Properties of Protein and Epitope prediction were carried out. The outcomes of this study revealed that the complete sequence of VP7 (G9) was comprised of 1062 nt with 326 amino acids (accession number MH824633). The selected epitope contained amino acid sequence of STLCLYYPTEASTQIGDTEWKN with the best score for T and B cells response. Based on data of computational biology, the selected epitope can optimistically have considered as an epitope candidate for rotavirus vaccine development.

Abstract

Background and Objectives: Adenoviral keratoconjunctivitis is an extremely frequent ophthalmological disease caused by various serological subtypes of human adenovirus (HAdV) worldwide. Adenoviruses serotypes 8, 11, 19, 37 frequently cause epidemic keratoconjunctivitis (EKC). This study was conducted to evaluate the frequency of adenovirus serotypes in patients with EKC in Ahvaz , Iran. Materials and Methods: Eighty- eight ocular swabs were collected from patients with EKC. The specimens were analyzed for detection of adenovirus by standard PCR. The PCR products were further sequenced and analyzed to determine the serotypes. Results: The study population consisted of 49/88 (55.7%) males and 39/88 (44.3%) females. Among them 25 (51.02%) males and 22 (56.41%) females were positive for HAdV serotype 8 (p= 0.488). Overall forty-seven (53.4%) samples were positive for AdV serotype 8 while forty- one patients (46.59%) were negative for the adenovirus serotypes. Conclusion: The results of this study revealed predominanance of HAdV 8 with high prevalence of 53.4% among patients with Keratoconjunctivitis. Forty- one patients (46.59%) were negative for adenovirus. Still, the role for other related viruses such as enteroviruses need to be investigated in patients with EKC.
