Abstract

Euphorhelipanes A (1) and B (2), two Euphorbiaditerpenoids with a new 4-(5,5-dimethylheptan-2-yl)-2,7-dimethylbicyclo[4.3.0]nonane skeleton, were isolated from a 95% ethanol extract of the whole plants of Euphorbia helioscopia. Their structures were elucidated by spectroscopic data analysis, quantum chemical calculations, and single-crystal X-ray diffraction data. Compounds 1and 2represent the first examples of Euphorbiaditerpenoids with a 5/6 fused carbon ring system, and their plausible biosynthetic pathways originating from jatrophanes are proposed. Compounds 1and 2showed a triglyceride-lowering effect in oleic-acid-stimulated HuH7 cells at concentrations of 1–50 μM.

190. Discovery of 29-O-acyl-toosendanin-based derivatives as potent anti-cancer agents.

Abstract

Structural modification of natural products is an effective strategy to discover potent lead compounds with improved medicinal performance. Toosendanin (TSN), a natural limonoid with diverse pharmacological properties, was selected as the starting material for structural modification to obtain more active anti-cancer agents in the current study. A library containing 25 structurally diverse derivatives (including 12 new ones) were constructed on the basis of the structure-guided modification of TSN. Subsequent cytotoxic assay of this library discovered that compounds 14 , 18 , and 25 showed more significant antiproliferative activity than the precursor TSN in MDA-MB-468 cell model and so as compounds 14 , 17 – 19 , 21 , and 25 in Hela cell model. Among them, the new derivative 29- O -(6-chloronicotinoyl)-toosendanin (25) exhibited the most potent antiproliferative activity (IC 50 s 0.05–0.06 μM), being more active than TSN (IC 50 s 0.14–0.24 μM) and even the first-line drug adriamycin (IC 50 s ∼ 0.07 μM) in both tested cancer cell lines. The SARs study uncovered that the hemiacetal group, the 14,15-epoxy ring, 1-OH, 7-OH, 3-OAc, and 12-OAc were viewed as the essential active groups and the 29-OH was the critically active modification position of TSN for the enhancement of cytotoxicity. The discovering of 25 from TSN-based derivatives might serve as a lead compound for anti-cancer chemotherapy, which may shed light on rationally design TSN-based derivatives for obtaining more potent anti-tumor agents. Abstract

Five new cassane diterpenoids, caesalmins I-M ( 1- 5), and 23 known analogs were isolated from the seeds of Caesalpinia minax. Their structures were elucidated by spectroscopic methods and comparison with reported data. The antioxidant properties of 1- 28 were determined by the method of oxygen radical absorbance capacity of fluorescein (ORAC-FL), and 14 compounds exhibited good antioxidant activities with ORAC-FL values of 2.24-4.89 Trolox equivalents. Abstract

Acid condensation rate is an important factor denoting the acid corrosion, and the reduction of the acid condensation can significantly relieve the acid corrosive effect on the wall surface and improve the security of the equipments. In this study, the characteristics of both heat transfer and acid condensation of the finned tube in heat exchanger were numerically studied. In the numerical model, we simulated the acid condensation by considering the vapor-liquid equilibrium effect and multi-component diffusion effect. Based on the H-type finned oval tube, we proposed three novel types of fins to both enhance the heat transfer and reduce the acid condensation. The parametric effects of gas temperature, acid vapor concentration, water vapor concentration, and Reynolds number were investigated on different fin structures. The results show that the tube bank with the new structured fins can improve the performance on both heat transfer and acid anti-condensation. [ABSTRACT FROM AUTHOR]

Eleven known compounds were isolated from the whole plant of Hedyotis cryptantha .On the basis of spectroscopic data, their structures were identified as asperuloside (1), asperulosidic acid (2), asperulosidic acid methyl ester (3), asperulosidic acid ethyl ester (4 ), 3,4-dihydro-3-methoxy asperuloside (5), kaempferol (6), kaempferol 3,7-di-O -β-D-glucoside (7), kaempferol 3-O -β-D-galactopyranosyl- (1 → 3) -β-D-galactopyranoside (8), ursonic acid (9), stigmasterol (10), and β-sitosterol (11) .All of these compounds were isolated from this plant for the first time, while compounds 7 and 8 were found from the genus Hedyotis for the first time. Thirteen new Euphorbiaditerpenoids, euphylonanes A–M (1–13), and eight known ones were isolated from the whole plants of Euphorbia hylonoma. Compounds 1and 2are two rearranged ingenanes bearing a rare 6/6/7/3-fused ring system. Compound 3represents the first example of a 9,10-epoxy tigliane, while 4–21are typical ingenanes varying with substituents. Structures were elucidated using a combination of spectroscopic, computational, and chemical methods. Most ingenanes exerted a significant antiadipogenic effect in 3T3-L1 adipocytes, among which 4was the most active with an EC50value of 0.60 ± 0.27 μM. Mechanistic study revealed that 4inhibited the adipogenesis and lipogenesis in adipocytes via activation of the AMPK signaling pathway.
