161. Identification and characterization of plant growth-promoting properties of bacterial endophytes from selected Zingiberaceae plants.

Abstract

Aims: This study aims to isolate, characterize and screen the plant growth-promoting bacteria from Zingiberaceae plants. Plant promoting activities such as indole-3-acetic acid (IAA), phosphate solubilization, zinc solubilization and nitrogen-fixing capabilities are determined, and the IAA production of selected isolates are optimized. Methodology and results: Endophytic bacteria were isolated from the plant samples by surface sterilization on nutrient agar (NA) plates and incubated at 30 °C for 2-3 days. The bacteria were identified based on their phenotypic characteristics and 16S rRNA gene sequence analyses. All isolates were identified as genera Bacillus, Lysinibacillus, Kerstersia, Klebsiella and Brucella. The isolates exhibited phosphate solubilization (1.5 ± 0.75-37.5 ± 8.75 Solubilization Index, SI), zinc solubilization (2.5 ± 0-60 ± 1.5 SI) and IAA production (0.1 ± 0.2-115.7 ± 1.6 µg/mL), while 3 isolates possessed nitrogen-fixing capabilities. Five isolates (PHAS-2, PWS-2, PWR-2, PHBS-2 and SCG-2) were selected for IAA optimization. Isolate PWR-2 produced the maximum IAA at 447.7 ± 0 µg/mL when tryptophan concentration was maintained at 1.0%. Conclusion, significance and impact of study: Genera of bacteria included Bacillus, Lysinibacillus, Kerstersia, Klebsiella and Brucella were successfully isolated from Zingiberaceae plants. All the isolates showed the capability to produce IAA, while some isolates exhibited phosphate solubilization and zinc solubilization, and a few possessed nitrogen-fixing capabilities. Abstract

Strain KK1-2T is the type strain of marine Streptomyces chomphonensis. Since the discovery of this Streptomyces species, the secondary metabolite of the type strain has not been characterized. Based on genome sequence analysis, the biosynthetic gene cluster of piericidin A1 was detected in the genome of strain KK1-2T. The isolation of secondary metabolites and spectroscopy data confirm the chemical structure of piericidin A1 produced by the type strain. This study is the first report of the genome analysis and the secondary metabolite produced by the type strain of S. chumphonensis. [ABSTRACT FROM AUTHOR]

Abstract

DESCRIPTION OF STREPTOMYCES KRUNGCHINGENSIS SP. NOV. Streptomyces krungchingensis (krung.ching.en′ sis. N.L. masc. adj. krungchingensis pertaining to Krung Ching Waterfall National Park in Nakhon Si Thammarat Province of Thailand, where the type strain was isolated). Aerobic, mesophilic, filamentous, Gram-stain-positive actinomycete that produces extensively branched substrate and aerial mycelia. Flexuous spore chains are observed on aerial mycelia. The spore surface is smooth. White to light-greenish grey aerial mass is produced on ISP2, ISP3, ISP4 and nutrient agar but absent on ISP5, ISP6 and ISP7 media. The substrate mycelia appear greyish yellow on ISP2, ISP7 and nutrient agar, light olive brown on ISP3 and ISP4, brilliant yellow on ISP5 and greyish olive green on ISP6. Yellow to brown pigment is observed on most tested media except ISP5. Reduces nitrate to nitrite and hydrolyses starch but does not hydrolyse milk. Liquefaction of gelatin is weakly positive. Utilizes cellobiose, fructose, myo -inositol, D- mannitol, D- mannose, raffinose, ribose and sucrose as sole carbon sources, weakly utilizes salicin and D- xylose but does not utilize L- arabinose, cellulose, melezitose or D- sorbitol. Shows enzyme activities of acid phosphatase, alkaline phosphatase, esterase (C4), b-galactosidase, leucine arylamidase and N -acetyl-b- glucosaminidase, shows weak activities of esterase lipase (C8), b- glucosidase, naphthol-AS-BI-phosphohydrolase and valine arylamidase but does not show activities of a- chymotrypsin, cystine arylamidase, a- fucosidase, a- galactosidase, a- glucosidase, b- glucuronidase, lipase (C14), a- mannosidase or trypsin. Growth occurs at pH 5–9, at 15–37 Ǫ C and with 0–6 % (w/ v) NaCl. The cell-wall peptidoglycan contains LL- diaminopimelic acid. Ribose and glucose are observed as whole-cell sugars. The menaquinones are MK-9(H 8), MK-9(H 6) and MK-9 (H 4). The N -acyl type of muramic acid is acetyl. Phospholipids are diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannoside, two unknown lipids and an unidentified aminolipid. The major cellular fatty acids are anteiso-C 15: 0, C 16: 0, iso-C 16: 0, iso-C 15: 0 and iso-C 14: 0. The type strain, KC-035 T (= NBRC 110087 T = KCTC 29 503 T = TISTR 2402 T), was isolated from soil collected from Krung Ching Waterfall National Park, Nakhon Si Thammarat Province, Thailand. The DNA G+C content of the type strain is 72 mol%.

Abstract

Objective: This research aimed to investigate the effect of cell lysis methods on a quantity of coenzyme Q10 (CoQ10) produced by Methylobacterium strains. Methods: Methylobacterium organophilum NBRC 15689T and Methylobacterium strain LRY1-08 were used to produce of CoQ10. The 4 methods of cell lysis treatment were used to disrupt the cell wall of bacteria and to increase the release of CoQ10 which was followed by extraction with isopropanol and hexane (3:5). Content of CoQ10 was analyzed with HPLC method. Results: The results showed that the cell lysis treatment using methanol and 0.3% sodium chloride (10:1) containing 1% Triton X-100 with sonication and then extraction with isopropanol and hexane (3:5) were found to yield the highest CoQ10 content for both Methylobacterium strains. Methelobacterium organophilum NBRC 15689T produced significantly higher CoQ10 content than Methylobacterium strain LRY1-08 which was consistent with its higher dry cell weight. Conclusion: The results indicated that this cell lysis method could be used to extract CoQ10 from plasma membrane of the bacteria. Methylobacterium organophilum NBRC 15689T was a promising strain for CoQ10 production source which should be further optimized factors affecting CoQ10 production. [ABSTRACT FROM AUTHOR]
