Ion mobility (IM) is an important analytical technique for determining ion collision cross section (CCS) values in the gas-phase and gaining insight into molecular structures and conformations. However, limited instrument resolving powers for IM may restrict adequate characterization of conformationally similar ions, such as structural isomers, and reduce the accuracy of IM-based CCS calculations. Recently, we introduced an automated technique for extracting pure IM and collision-induced dissociation (CID) mass spectra of IM overlapping species using chemometric deconvolution of post-IM/CID mass spectrometry (MS) data [J. Am. Soc. Mass Spectrom., 2014, 25, 1810-1819]. Here we extend those capabilities to demonstrate how extracted IM profiles can be used to calculate accurate CCS values of peptide isomer ions which are not fully resolved by IM. We show that CCS values obtained from deconvoluted IM spectra match with CCS values measured from the individually analyzed corresponding peptides on uniform field IM instrumentation. We introduce an approach that utilizes experimentally determined IM arrival time (AT) shift factors to compensate for ion acceleration variations during post-IM/CID and significantly improve the accuracy of the calculated CCS values. Also, we discuss details of this IM deconvolution approach and compare empirical CCS values from traveling wave (TW)IM-MS and drift tube (DT)IM-MS with theoretically calculated CCS values using the projected superposition approximation (PSA). For example, experimentally measured deconvoluted TWIM-MS mean CCS values for doubly-protonated RYGGFM, RMFGYG, MFRYGG, and FRMYGG peptide isomers were 288.8 Å(2), 295.1 Å(2), 296.8 Å(2), and 300.1 Å(2); all four of these CCS values were within 1.5% of independently measured DTIM-MS values.

In Sub-Saharan Africa rapid urban growth combined with rising poverty is creating diverse urban environments, the nature of which are not adequately captured by a simple urban-rural dichotomy. This paper proposes an alternative classification scheme for urban mapping based on a gradient approach for the southern portion of the West African country of Ghana. Landsat Enhanced Thematic Mapper Plus (ETM+) and European Remote Sensing Satellite-2 (ERS-2) synthetic aperture radar (SAR) imagery are used to generate a pattern based definition of the urban context. Spectral mixture analysis (SMA) is used to classify a Landsat scene into Built, Vegetation and Other land covers. Landscape metrics are estimated for Built and Vegetation land covers for a 450 meter uniform grid covering the study area. A measure of texture is extracted from the SAR imagery and classified as Built/Non-built. SMA based measures of Built and Vegetation fragmentation are combined with SAR texture based Built/Non-built maps through a decision tree classifier to generate a nine class urban context map capturing the transition from unsettled land at one end of the gradient to the compact urban core at the other end. Training and testing of the decision tree classifier was done using very high spatial resolution reference imagery from Google Earth. An overall classification agreement of 77% was determined for the nine-class urban context map, with user's accuracy (commission errors) being lower than producer's accuracy (omission errors). Nine urban contexts were classified and then compared with data from the 2000 Census of Ghana. Results suggest that the urban classes appropriately differentiate areas along the urban gradient.

Context: Care home residents in the UK care homes account for almost 1.5 million emergency bed days each year. Avoidance of unnecessary hospital transfers benefits residents, and reduces demand on the healthcare systems but is difficult to implement. Objective: We synthesized evidence on interventions that influence unplanned hospital admissions or attendances by care home residents. Study design: This is a Systematic Review of Randomised Controlled Trials. Dataset: PUBMED, MEDLINE, EMBASE, ISI Web of Science, CINAHL and the Cochrane Library were searched from 2012 to 2021, building on a review published in 2013. Population studied: Care home residents over the age of sixty-five years old. Intervention: Interventions that influence (decrease or increase) acute hospital admissions or attendances from care home residents. Outcome measures: Hospitalization, Emergency Department Attendance, and Rehospitalization. Results Thirty-eight randomized studies were included in this review. A narrative synthesis was conducted and the weight of evidence was described with vote counting. Advance care planning and goals of care setting appear to be effective at reducing hospitalizations from care homes. Other effective interventions, in order of increasing risk of bias, were: nurse practitioner/specialist input, palliative care intervention, influenza vaccination, and enhancing access to intravenous therapies in care homes. Factors that affect hospitalization and emergency department attendances of care home residents are complex. This review supports the already established use of advanced care planning and influenza vaccination to reduce unscheduled hospital attendances. It is likely that more than one intervention will be needed to impact on healthcare utilization across the care home population. Abstract Background Measles is a vaccine preventable, highly transmissible viral infection that affects mostly children under five years. It has been ear marked for elimination and Nigeria adopted the measles elimination strategies of the World Health Organization (WHO) African region to reduce cases and deaths. This study was done to determine trends in measles cases in Bayelsa state, to describe cases in terms of person and place, identify gaps in the case-based surveillance data collection system and identify risk factors for measles infection. Methods We carried out a secondary data analysis of measles case-based surveillance data for the period of January 2014 to December 2018 obtained in Microsoft Excel from the State Ministry of Health. Cases were defined according to WHO standard case definitions. We calculated frequencies, proportions, estimated odds ratios (OR), 95% confidence intervals (CI) and multivariate analysis. Results A total of 449 cases of measles were reported. There were 245(54.6%) males and the most affected age group was 1–4 years with 288(64.1%) cases. Of all cases, 289(9.35%) were confirmed and 70 (48.27%) had received at least one dose of measles vaccine. There was an all-year transmission with increased cases in the 4th quarter of the year. Yenegoa local government area had the highest number of cases. Timeliness of specimen reaching the laboratory and the proportion of specimens received at the laboratory with results sent to the national level timely were below WHO recommended 80% respectively. Predictors of measles infection were, age less than 5 years (AOR: 0.57, 95% CI: 0.36–0.91) and residing in an urban area (AOR: 1.55, 95% CI:1.02–2.34). Conclusions Measles infection occurred all-year round, with children less than 5 years being more affected. Measles case-based surveillance system showed high levels of case investigation with poor data quality and poor but improving indicators. Being less than 5 years was protective of measles while living in urban areas increased risk for infection. We recommended to the state government to prioritize immunization activities in the urban centers, start campaigns by the 4th quarter and continue to support measles surveillance activities and the federal government to strengthen regional laboratory capacities.

Temporal trajectories of apparent vegetation abundance based on the multi-decadal Landsat image series provide valuable information on the postfire recovery of chaparral shrublands, which tend to mature within one decade. Signals of change in fractional shrub cover (FSC) extracted from time-sequential Normalized Difference Vegetation Index (NDVI) data can be systematically biased due to spatial variation in shrub type, soil substrate, or illumination differences associated with topography. We evaluate the effects of these variables in Landsat-derived metrics of FSC and postfire recovery, based upon three chaparral sites in southern California which contain shrub community ecotones, complex terrain, and soil variations. Detailed validations of prefire and postfire FSC are based on high spatial resolution ortho-imagery; cross-stratified random sampling is used for variable control. We find that differences in the composition and structure of shrubs (inferred from ortho-imagery) can substantially influence FSC-NDVI relations and impact recovery metrics. Differences in soil type have a moderate effect on the FSC-NDVI relation in one of the study sites, while no substantial effects were observed due to variation of terrain illumination among the study sites. Arithmetic difference recovery metrics – based on NDVI values that were not normalized with unburned control plots – correlate in a moderate but significant manner with a change in FSC (R2 values range 0.47–0.59 at two sites). Similar regression coefficients resulted from using Landsat visible reflectance data alone. The lowest correlations to FSC resulted from Soil-Adjusted Vegetation Index (SAVI) and are attributed to the effects of the soil-adjustment factor in sparsely vegetated areas. The Normalized Burn Ratio and Normalized Burn Ratio 2 showed a moderate correlation to FSC. This study confirms the utility of Landsat NDVI data for postfire recovery evaluation and implies a need for stratified analysis of postfire recovery in some chaparral landscapes.

Conservation actions focused on flagship species are effective at raising funds and awareness. Here, McGowan et al. show that prioritizing areas for conservation based on the presence of flagship species results in the selection of areas with ~ 79-89% of the total species that would be selected by maximizing biodiversity representation only.

A hallmark of Listeria (L.) monocytogenes pathogenesis is bacterial escape from maturing entry vacuoles, which is required for rapid bacterial replication in the host cell cytoplasm and cell-to-cell spread. The bacterial transcriptional activator PrfA controls expression of key virulence factors that enable exploitation of this intracellular niche. The transcriptional activity of PrfA within infected host cells is controlled by allosteric coactivation. Inhibitory occupation of the coactivator site has been shown to impair PrfA functions, but consequences of PrfA inhibition for L. monocytogenes infection and pathogenesis are unknown. Here we report the crystal structure of PrfA with a small molecule inhibitor occupying the coactivator site at 2.0 Å resolution. Using molecular imaging and infection studies in macrophages, we demonstrate that PrfA inhibition prevents the vacuolar escape of L. monocytogenes and enables extensive bacterial replication inside spacious vacuoles. In contrast to previously described spacious Listeria-containing vacuoles, which have been implicated in supporting chronic infection, PrfA inhibition facilitated progressive clearance of intracellular L. monocytogenes from spacious vacuoles through lysosomal degradation. Thus, inhibitory occupation of the PrfA coactivator site facilitates formation of a transient intravacuolar L. monocytogenes replication niche that licenses macrophages to effectively eliminate intracellular bacteria. Our findings encourage further exploration of PrfA as a potential target for antimicrobials and highlight that intra-vacuolar residence of L. monocytogenes in macrophages is not inevitably tied to bacterial persistence. Author summary Listeria monocytogenes is a bacterial pathogen that causes severe, often fatal disease in vulnerable individuals. Listeria monocytogenes infects host cells, where it needs to escape from its entry vacuole to replicate inside the host cell cytoplasm and spread to neighboring cells. Inability of Listeria to escape from vacuoles has been associated with long-term bacterial survival inside infected immune cells and chronic infection. We discovered that inhibition of the master regulator of the bacterial factors that orchestrate the intracellular lifestyle of Listeria monocytogenes traps the bacteria inside their entry vacuoles and facilitates extensive bacterial replication in spacious vacuoles. Yet, the majority of infected host immune cells survive the infection and ultimately clear the bacteria. Thus, our data demonstrate that intra-vacuolar existence of Listeria monocytogenes in immune cells is not inevitably tied to bacterial persistence and chronic infection. We provide high resolution structural insights into how an effective inhibitory molecule interacts with the master regulator of Listeria virulence, which may be exploited for rational drug design.

Wildfire burn severity has important implications for postfire vegetation recovery and boundary-layer climate. We used a collection of Moderate Resolution Imaging Spectroradiometer (MODIS) datasets to investigate the impact of burn severity (relative differenced Normalized Burn Ratio, RdNBR) on vegetation recovery (Enhanced Vegetation Index, EVI), albedo change, and land surface temperature in seven California ecoregions, including: Southern California Mountains (SCM), Southern California Coast (SCC), Central California Foothills (CCF), Klamath (K), Cascades (C), Eastern Cascades (EC), and Sierra Nevada (SN). A statewide MODIS-derived RdNBR dataset was used to analyze the impact of burn severity on the five-year postfire early-summer averages of each biophysical variable between the years 2003-2020. We found that prefire EVI values were largest, and prefire albedo and temperature were lowest in the K, C, EC, and SN ecoregions. Furthermore, the largest changes between prefire and first-year postfire biophysical response tended to occur in the moderate and high burn severity classes across all ecoregions. First-year postfire albedo decreased in the K, C, EC, and SN but increased in the SCM, SCC, and CCF ecoregions. The greatest decreases, but most rapid recovery, of EVI occurred after high severity fires in all ecoregions. After five-years post-fire, EVI and land surface temperature did not return to prefire levels in any burn severity class in any ecoregion.

There is great diversity in social behavior across the animal kingdom. Understanding the factors responsible for this diversity can help inform theory about how sociality evolves and is maintained. The Australian Tree Skink (Egernia striolata) exhibits inter- and intra-population variability in sociality and is therefore a good system for informing models of social evolution. Here, we conducted a multi-year study of a Tree Skink population to describe intra-population variation in the social organization and mating system of this species. Skinks aggregated in small groups of 2–5 individuals, and these aggregations were typically associated with shared shelter sites (crevices and hollows within rocks and trees). Aggregations were typically made up of one or more adult females and, often, one male and/or juvenile(s). Social network and spatial overlap analyses showed that social associations were strongly biased toward kin. Tree skinks also exhibited high site fidelity regardless of age or sex. There were high levels of genetic monogamy observed with most females (87%) and males (68%) only breeding with a single partner. Our results indicate that Tree Skinks reside in small family groups and are monogamous, which corresponds with existing research across populations. Similar to previous work, our study area consisted of discrete habitat patches (i.e., rock outcrops, trees, or both), which likely limits offspring dispersal and promotes social tolerance between parents and their offspring. Our study clearly demonstrates that there is intra-population variability in Tree Skink social behavior, but it also provides evidence that there is a high degree of inter-population consistency in sociality across their geographic range. We also highlight promising possible avenues for future research, specifically discussing the importance of studying the nature and extent of Tree Skink parental care and quantifying the fitness outcomes of kin-based sociality in this species, which are topics that will further our understanding of the mechanisms underlying variation in vertebrate social behavior.

Urban expansion is a form of land cover and land use change (LCLUC) that occurs globally, and population growth can be a driver of and be driven by LCLUC. Determining the cause–effect relationship is challenging because the temporal resolution of population data is limited by decadal censuses for most countries. The purpose of this study is to explore the relationship and relative timing between population change and land use change based on a case study of northern Taiwan from 1990 to 2015. A unique dataset on population was acquired from annually-updated governmental-based population registers maintained at the district level, and land-use expansion data (Residential, Employment, and Transportation Corridor categories) were derived from dense time series of Landsat imagery. Linear regression was applied to understand the general relationship between population and land use and their changes. The strongest relationships were found between population and areal extent of Residential land use, and between population change and Residential areal change. Lagged correlation analysis was implemented for identifying the time lag between population growth and land use change. Most districts exhibited Residential and Employment expansion prior to population growth, especially for districts in the periphery of metropolitan areas. Conversely, the core of metropolitan areas exhibited population growth prior to Residential and Employment expansion. Residential and Employment expansion were deemed to be drivers of population change, so population change was modeled with ordinary least square and geographically weighted regression with Residential and Employment expansion in both synchronized and time lag manners. Estimated population growth was found to be the most accurate when geographic differences and time lags from urban land use expansion were both incorporated.

Objectives To identify and map evidence about the consequences of unpaid caring for all carers of older people, and effective interventions to support this carer population.Design A rapid review of systematic reviews, focused on the consequences for carers of unpaid caring for older people, and interventions to support this heterogeneous group of carers. Reviews of carers of all ages were eligible, with any outcome measures relating to carers’ health, and social and financial well-being. Searches were conducted in MEDLINE, PsycInfo and Epistemonikos (January 2000 to January 2020). Records were screened, and included systematic reviews were quality appraised. Summary data were extracted and a narrative synthesis produced.Results Twelve systematic reviews reporting evidence about the consequences of caring for carers (n=6) and assessing the effectiveness of carer interventions (n=6) were included. The review evidence typically focused on mental health outcomes, with little information identified about carers’ physical, social and financial well-being. Clear estimates of the prevalence and severity of carer outcomes, and how these differ between carers and non-carers, were absent. A range of interventions were identified, but there was no strong evidence of effectiveness. In some studies, the choice of outcome measure may underestimate the full impact of an intervention.Conclusions Current evidence fails to fully quantify the impacts that caring for older people has on carers’ health and well-being. Information on social patterning of the consequences of caring is absent. Systematic measurement of a broad range of outcomes, with comparison to the general population, is needed to better understand the true consequences of caring. Classification of unpaid caring as a social determinant of health could be an effective lever to bring greater focus and support to this population. Further work is needed to develop and identify suitable interventions in order to support evidence-based policymaking and practice.

Objectives To investigate whether National Early Warning Scores (NEWS/NEWS2) could contribute to COVID-19 surveillance in care homes.Setting 460 care home units using the same software package to collect data on residents, from 46 local authority areas in England.Participants 6464 care home residents with at least one NEWS recording.Exposure measure 29 656 anonymised person-level NEWS from 29 December 2019 to 20 May 2020 with component physiological measures: systolic blood pressure, respiratory rate, pulse rate, temperature and oxygen saturation. Baseline values for each measure calculated using 80th and 20th centile scores before March 2020.Outcome measure Cross-correlation comparison of time series with Office for National Statistics weekly reported registered deaths of care home residents where COVID-19 was the underlying cause of death, and all other deaths (excluding COVID-19) up to 10 May 2020.Results Deaths due to COVID-19 were registered from 23 March 2020 in the local authority areas represented in the study. Between 23 March 2020 and 10 May 2020, there were 5753 deaths (1532 involving COVID-19 and 4221 other causes). We observed a rise in the proportion of above-baseline NEWS beginning 16 March 2020, followed 2 weeks later by an increase in registered deaths (cross-correlation of r=0.82, p

Student achievement of the curriculum is the school's reason for being. This research, however, suggests that the behavior of school leaders is not effective in accomplishing four critical outcomes of curriculum development: (1) the creation of well-written documents that use a common framework; (2) curriculum that is aligned with state and national standards; (3) an understanding and teaching of the curriculum by teachers; and (4) all students successfully learning the curriculum. Leaders must orchestrate multiple changes that provide opportunities for teachers to work in teams, focus resources effectively on implementing the curriculum, and establish accountability for results.
