Abstract

The divalent anion sodium symporter (DASS) family (SLC13) play critical roles in metabolic homeostasis, influencing many processes including fatty acid synthesis, insulin resistance, and adiposity. DASS transporters catalyse the Na+-driven concentrative uptake of Krebs cycle intermediates and sulfate into cells; disrupting their function can protect against age-related metabolic diseases and can extend lifespan. An inward-facing crystal structure and an outward-facing model of a bacterial DASS family member, VcINDY from Vibrio cholerae, predict an elevator-like transport mechanism involving a large rigid body movement of the substrate binding site. How substrate binding influences the conformational state of VcINDY is currently unknown. Here, we probe the interaction between substrate binding and protein conformation by monitoring substrate-induced solvent accessibility changes of broadly distributed positions in VcINDY using a site-specific alkylation strategy. Our findings reveal that accessibility to all positions tested are modulated by the presence of substrates, with the majority becoming less accessible in the presence of saturating concentrations of both Na+ and succinate. We also observe separable effects of Na+ and succinate binding at several positions suggesting distinct effects of the two substrates. Furthermore, accessibility changes to a solely succinate-sensitive position suggests that substrate binding is a low affinity, ordered process. Mapping these accessibility changes onto the structures of VcINDY suggests that Na+ binding drives the transporter into an as-yet-unidentified conformational state, involving rearrangement of the substrate binding site-associated re-entrant hairpin loops. These findings provide insight into the mechanism of VcINDY, which is currently the only structural-characterised representative of the entire DASS family.

Abstract

Cover crops seeded after wheat (Triticum aestivum L.) provide agronomic, environmental and soil health services. Long-term cover crop trials were established at Ridgetown and Elora, ON, in 2017. But in the short-tern, cover crop populations, C:N ratios, N uptake and biomass in November, and subsequent corn (Zea mays L.) grain yields, were examined among combinations of five cover crop treatments, two nitrogen treatments before cover crop seeding, and two tillage systems in 2017 and 2018. In general, mixtures of cover crop species did not produce more biomass than monocultures at 3 of 4 site-years. N fertilizer applied after wheat harvest increased biomass of non-legumes and tended to reduce C:N ratios in November. Cover crops did not increase grain corn yields when adequately fertilized with N, but where N was limited, yields were improved after red clover (Trifolium pratense L.) and decreased after oat (Avena sativa L.). Cover crop performance can be maximized in the short-term, but longer-term studies are needed.

Abstract

This dissertation explores the life and work of the writer Wallace Stegner (1909-1993) and his intellectual efforts to create, clarify, and defend the contours of a “geography of hope” in the American West. Chapter 1 begins with Stegner’s move to California in 1945. In the context of his developing regional vision as expressed in early articles and books, it traces Stegner’s attempts to build a range of institutions in California as well as his first writings that either adopted the state as its subject or used it as a setting for fictional work. Chapter 2 explores a research project that Stegner undertook with funding from the Wenner-Gren Foundation and as a member of the Stanford Center for Advanced Studies in Behavioral Sciences (CASBS) in the mid-1950s. This project reflected Stegner’s understanding of western history and his own past in more comprehensive terms and the chapter traces his growing commitment to fiction as a resource for addressing the questions he thought most important to explore in his historical moment. The subsequent three chapters include close readings of three novels: All the Little Live Things (1967), Angle of Repose (1971), and The Spectator Bird (1976). The novels are united by location, themes, and a first-person narrator who opens the book with ruminations and something of a thesis. Two of these three books won major awards, so they also marked the summit of Stegner’s national recognition as a writer of American fiction. Each of the readings reveals ways that Stegner and his readers addressed the cultural changes of the 1960s, adding nuance to the historiography of an era that has more often been marked by an emphasis on polarization. The conclusion is focused on Greensboro, Vermont, the place where Stegner chose to have his ashes spread after his death. Greensboro served as a realistic but still at times utopian foil for Stegner’s exploration of the western “formless non-communities” that are the focus points of the previous chapters

Abstract

Objectives/Hypothesis: The literature on outcomes of end‐to‐side (ETS) anastomoses for microvascular reconstruction of the head and neck is limited. This series reviews ETS in free tissue transfer (FTT) across multiple institutions to better understand their usage and associated outcomes. Study Design: Retrospective review of 2482 consecutive patients across three tertiary institutions. Methods: Adult patients (> 18) who received a FTT from 2006 to 2019 were included. Results: Two hundred and twenty‐one FTT were identified as requiring at least one ETS anastomosis. These ETS cases had a failure rate of 11.2% in comparison to 3.8% in a cohort of end‐to‐end (ETE) cases (P <.001). ETS cases were significantly more likely to have a prior neck dissection (P <.001), suggesting the ETS method was utilized in select circumstances. A second ETS anastomosis improved survival of the FTT (P =.006), as did utilization of a coupler over suture (P =.002). Failure due to venous thrombosis was significantly more common with one ETS anastomosis instead of two ETS anastomoses (P =.042). Conclusions: ETS is effective but is often used as a secondary technique when ETE is not feasible; as such, in this series, ETS was associated with higher failure. A second anastomosis and the use of the coupler for completing the anastomoses were associated with lower rates of failure.
